As a transcellular messenger molecule, nitric oxide (NO) is involved in many physiological and pathological processes. The accurate quantitative detection of NO in vivo is particularly important. In this work, a novel electrochemical NO sensor was fabricated via electro-polymerization of ethylenediamine (EDA) on multi-walled carbon nanotubes (MWNTs) film. The sensor combined the porous structure and good conductivity of MWNTs with the good electro-catalytic ability of p-EDA film. For the detection of NO, it has a linear range from 95 nM to 11 μM and the limit of detection reaches 95 nM with excellent reproducibility. So it was successfully applied to monitor NO release from rat liver sample for investigating the inhibition or promotion effect of drugs including oleanolic acid (OA), baicalin and Nω-nitro- l -arginine methyl ester ( l -NAME) on nitric oxide synthase (NOS). The results showed that OA could promote NO release and the amount of NO increases about 1.9 times, but baicalin and l -NAME could inhibit the release of NO and the amount of NO decreased by two-thirds and 37.5%, respectively.

Photodynamic therapy (PDT) is a non-invasive treatment method for tumors by exciting photosensitizers (PS) upon light irradiation to generate cytotoxic reactive oxygen species (ROS). However, the low oxygen concentration near the tumor tissue limits the therapeutic effect of PDT. Herein, we synthesized six chlorin e6 derivatives containing NO-donors to enhance their antitumor activity by synergistic effect of ROS and NO. The results revealed that the new NO-donor containing photosensitizers (PS-NO) exhibited more potent photodynamic activity than chlorin e6, and the introduction of NO donor moieties to chlorin e6 increased the level of NO and ROS in cells. The addition of Ferrostatin-1, a ferroptosis inhibitor, markedly reduced the photodynamic activity of PS-NO as well as the level of NO and ROS in cells. Mechanism studies further showed that PS-NO could reduce intracellular GSH level, inhibit GPX4 activity and promote malondialdehyde (MDA) accumulation upon light irradiation, which suggested the ferroptosis mechanism underlying the PDT effect of PS-NO.

Nitric oxide (NO) gas treatment offers a promising strategy for tumor therapy; however, its practical application is still limited due to its poor efficacy and biotoxicity which were caused by gas leakage during blood delivery. Herein, a nano-platform (CMH-OBN) composed of chlorin e6-melanin-hyaluronic acid nanoparticles (Ce6-MNP-HA, CMH) and oxidized bletilla striata polysaccharide microcapsules (Oxi-BSP) carrying NO donors was prepared for responsive and cascaded release of NO, reactive oxygen species (ROS) and its secondary metabolite reactive nitrogen species (RNS) in tumor sites. Melanin not only endowed CMH with good photothermal properties, but also helped Ce6 to produce a large number of ROS under near-infrared (NIR) irradiation. OBN microcapsules, which were sensitive to ROS, can release NO donors under the stimulation of ROS released by CMH nanoparticles under NIR irradiation and can further release NO in the tumor microenvironment (TME) with high expression of glutathione (GSH). NO could further up-regulate soluble guanylate cyclase-cyclic guanosine monophosphate (sGC-cGMP) signal pathways to relieve hypoxia, thus further enhancing the photodynamic therapy (PDT). Moreover, the cascaded release of ROS and NO could produce RNS with higher lethality, which could sequentially initiate the cellular apoptotic procedure and promote immunotherapy by activating T cells at the tumor sites. More interestingly, the CMH-OBN nano-platform could supply magnetic resonance imaging (MRI) and infrared photothermal imaging guidance for tumor therapy. In conclusion, the development of a CMH-OBN nano-platform provides a satisfactory demonstration by combining NO therapy with photothermal therapy (PTT), PDT and immunotherapy for the treatment of cancer.

Taste receptors are not only expressed in the taste buds, but also in other nongustatory tissues, including the reproductive system. Taste receptors can be activated by various tastants, thereby exerting relatively physiologic functions. The aim of this study was to investigate the effects and potential mechanisms underlying ovarian taste receptor activation on progesterone production using saccharin sodium as the receptor agonist in a pseudopregnant rat model. Taste 1 receptor member 2 (TAS1R2) and taste 2 receptor member 31 (TAS2R31) were demonstrated to be abundantly expressed in the corpora lutea of rats, and intraperitoneal injection of saccharin sodium can activate both of them and initiate their downstream signaling cascades. The activation of these ovarian taste receptors promoted nitric oxide (NO) production via endothelial nitric oxide synthase (eNOS). NO production then increased ovarian cyclic guanosine 3′,5′-monophosphate (cGMP) levels, which, in turn, decreased ovarian cyclic adenosine 3′,5′-monophosphate levels. In addition, the activation of ovarian taste receptors induced apoptosis, possibly through NO and mitogen-activated protein kinase signaling. As a result, the activation of ovarian taste receptors reduced the protein expression of steroidogenesis-related factors, causing the inhibition of ovarian progesterone production. In summary, our data suggest that the activation of ovarian taste receptors inhibits progesterone production in pseudopregnant rats, potentially via NO/cGMP and apoptotic signaling.

Cervical cancer (CC) treatment is limited by chemoresistance, inadequate drug accumulation, and the lack of real-time monitoring. Among the underlying mechanisms, heat shock protein 70 (HSP70) mediates resistance to both chemotherapy and photothermal therapy, making it a rational target for intervention. To address these issues, we developed a novel type of cyclic RGD (cRGD) encapsulated SN38 and IR820 loaded bismuth selenide theranostic nanoplatforms-Bi 2 Se 3 -cRGD@SN38/IR820 nanoparticles (Bi-A@SR NPs) based on a bismuth selenide (Bi 2 Se 3 ) as core decorated with cRGD for tumor targeting, and co-loaded with SN38 and IR820. The RGD peptide contains a guanidino group from its arginine residue that can serve as a donor for nitric oxide (NO) generation, enabling the construct to provide both tumor-homing capability and on-demand NO release. The nanoplatform integrates computed tomography (CT) imaging and NIR fluorescence imaging (NIRF) to verify delivery and guide therapy. Upon 808 nm irradiation, photothermal and photodynamic effects generate hyperthermia and reactive oxygen species (ROS), which trigger on-site NO production from the RGD arginine moiety. This NO suppresses therapy-induced HSP70 upregulation, counteracting stress adaptation and enhancing chemo–phototherapy efficacy. By combining imaging-guided delivery, on-demand NO sensitization, and HSP70 inhibition, this nanotheranostic strategy offers a coordinated approach to improving treatment outcomes in CC.

Fe 2 N@C nanospheres integrate NO release, PTT, and CDT for DFU therapy. • NIR-II-triggered hyperpyrexia amplifies POD-like activity of Fe 2 N@C. • Inorganic Fe 2 N@C acts as a novel NO donor with high release capacity. • Acidic biofilm microenvironment facilitates NO release and CDT activity. • Dynamic therapy possesses great potential on infective DFU therapy. Diabetic foot ulcer (DFU) is one of the most severe complications of diabetes, and its multiple pathological factors render treatment more challenging. We synthesized Fe 2 N@C nanomaterials to integrate synergistic photothermal therapy/chemical dynamic therapy/nitric oxide (PTT/CDT/NO) for DFU therapy. First, Fe 2 O 3 and urea were used as precursors to synthesize Fe 2 N@C nanomaterials via nitridation. The nanomaterial can serve as a new NO donor that releases NO triggered by H + and reactive oxygen species (H 2 O 2 and O 2 ). Furthermore, Fe 2 N@C possess superhigh peroxidase (POD)-like activity (V max = 21.1 Ms. −1 , pH 5.5), catalyzing endogenous H 2 O 2 into hydroxyl radicals ( OH). POD activity can also amplify NO and peroxynitrite (ONOO − ) production. In addition, Fe₂N@C nanomaterials reveal high near-infrared II (NIR-II) light harvest and photothermal conversion efficiency (43.94%). Hyperthermia also synergistically boosts POD-like activity and NO release. Under NIR-II laser irradiation, the combination of PTT/CDT and high NO concentration of can quickly clear bacterial infection. In addition, NO can down-regulate the expression of heat shock proteins (HSPs) to amplify PTT performance. Then, the mild NO supply (without irradiation) further facilitates the reversal of inflammation (M2 macrophage polarization) and tissue regeneration (angiogenesis). This dynamic synergistic therapy promotes fast normal DFU healing. Download: Download high-res image (259KB) Download: Download full-size image

A multifunctional nanocomposite, composed of Ga-loaded Ce-MOF core and NO-responsive microgel shell, is designed to integrate effective drug loading, controlled drug release and multi-target synergy to therapy RA. • The Ga-loaded Ce-MOF core functions dually as a high-capacity drug carrier of natural compound and a nanozyme with superoxide dismutase/catalase-like activities for reactive oxygen species (ROS) elimination, effectively alleviating oxidative stress damage in RA and establishing initial synergistic effects with Ga intrinsic anti-inflammatory activity. • The NO-responsive microgel shell acted as pathological cue-triggered “gatekeeper”, enabling on-demand drug release specifically at inflammatory sites while concurrently scavenging excessive NO to synergetic treat RA. • The nanocomposite significantly inhibits pro-inflammatory cytokines, alleviates cartilage degradation and bone erosion by eliminating excessive RONS, promoting the repolarization of macrophages from M1 to M2 phenotypes, and regulating the MAPK and cGMP/PKG signaling pathways. • This “one-stone-three-birds” strategy that integrating natural drug therapy, stimuli-responsive drug release with synergistic RONS scavenging and immune homeostasis modulation, offers an innovative and efficient approach for treatment of RA and other inflammatory diseases. Rheumatoid arthritis (RA) is a chronic autoimmune disorder characterized by persistent inflammatory injury and progressive destruction of joints. The elimination of excessive reactive oxygen and nitrogen species (RONS) and the restoration of immune homeostasis have emerged as promising therapeutic strategies for RA. Gastrodin (Ga), a bioactive natural constituent derived from Gastrodia elata, possesses inherent antioxidant and anti-inflammatory capacities, whereas its clinical translation and in vivo utility are substantially hampered by inferior environmental stability and uncontrollable drug release behavior. Herein, we fabricated a nitric oxide (NO)-responsive nanocomposite (NCMG) consisted of a Ce-based metal-organic framework (Ce-MOF) nanozyme loaded with Ga and encapsulated by a NO-responsive microgel shell for synergistic RA treatment. The Ce-MOF serves dual functions as a high-efficiency drug carrier and reactive oxygen species (ROS) scavenger, while the outer microgel shell enables precise on-demand Ga release and efficient NO depletion. In vitro experiments and in vivo validation based on adjuvant-induced arthritis (AIA) rat models demonstrated that NCMG markedly inhibits the secretion of pro-inflammatory cytokines, induces macrophage polarization from the pro-inflammatory M1 phenotype to the anti-inflammatory M2 phenotype, and effectively alleviates joint inflammation and bone erosion. Transcriptomic profiling further showed that Ga treatment was associated with downregulation of Il1b and Tnf and alterations in MAPK related gene expression, suggesting the involvement of this pathway in its anti-arthritic activity. This work develops an integrated nanotherapeutic platform combining natural active compounds with multi-component synergistic effects, offering a feasible strategy for RA intervention.

A synergy between FeO X and CuO X was confirmed in FeCuO X /TiO 2 -hBN composites for selective catalytic reduction of NO with NH 3 . Accordingly, the reducibility of composites was modified to promote the NO to NO 2 oxidation but suppress the NH 3 to NO over oxidation, which is of benefit to the NO reduction. Also the optimal operation temperature could be controlled by tuning the Fe:Cu atomic ratio, and Fe 10 Ti 85 BN 5 , Fe 7.5 Cu 2.5 Ti 85 BN 5 and Fe 2.5 Cu 7.5 Ti 85 BN 5 achieved the best NO removal above 90% at 375 °C, 300 °C and 250 °C, respectively. From the NO 2 and NH 3 adsorption behaviors over FeCuO X /TiO 2 -hBN, the reaction followed Langmuir–Hinshelwood mechanism below 275 °C, but followed Eley–Rideal mechanism above 350 °C. The influence of SO 2 on NO removal was found to be temperature dependent, at temperature below 325 °C, slight deactivation was observed, while a promotion was achieved above 350 °C.

Hydrothermal aging tests are important to carry out when evaluating the hydrothermal durability of heterogeneous catalysts in vehicle exhaust emission. Here, we explored the effect of aging on an efficient Mn-based mullite catalyst (SmMn2O5) of NO oxidation. The mullite catalyst was prepared via the hydrothermal method and was subsequently aged in air with a 10% H2O stream at 750 °C for 16 hours. The fresh and aged catalysts were structurally characterized using Powder X-ray diffraction(XRD), Raman, X-ray photoelectron spectroscopy (XPS), scanning electron microscope (SEM), high resolution-transmission electron microscope (HR-TEM), Brunauer–Emmett–Teller (BET) and temperature programmed desorption (TPD). For the performance evaluations, the samples were placed into a U-shape reactor furnace, and NO and NO2 concentrations were in situ recorded with an NOx analyzer. In contrast to fresh mullite, the aged sample showed a 25 °C higher light-off temperature and 11% conversion loss at its maximum conversion temperature of 300 °C. O2-TPD of the aged sample displayed a large decrease of the desorption area, consistent with an ∼3-fold loss of the BET specific surface area. Moreover, HRTEM, XPS and Raman spectroscopy results together indicated that a small portion of the mullite decomposed into perovskite SmMnO3 and Mn2O3, which further reduced the total quantity of Mn active sites. The reduction of the BET surface area and mullite decomposition together caused the decrease of the catalytic performance. We therefore expect maintaining the specific surface area to be important for preventing the loss of catalytic performance during the hydrothermal aging process.

Effective intervention during the early phases of alkali burns is crucial for preventing progressive ocular damage and persistent inflammation, necessitating multifunctional solutions beyond single-component therapies. In this study, we developed a thermosensitive hydrogel based on L-arginine-modified hydroxybutyl chitosan (HBC_Arg). The hydrogel remains liquid at room temperature for easy application and undergoes thermosensitive gelation upon reaching the ocular surface temperature, ensuring prolonged retention and enhanced drug efficacy. The L-arginine modification enables the hydrogel to release nitric oxide (NO), which plays a critical role in modulating immune responses and controlling excessive inflammation. Additionally, EPL@MnO2 nanosheets were encapsulated within the hydrogel for extended-release and enhanced scavenging of reactive oxygen species (ROS), reducing oxidative stress and mitigating alkali burn-induced damage. In a rat ocular alkali burn model, the composite HBC_Arg/MnO2 hydrogel significantly suppressed inflammation, promoted re-epithelialization, enhanced stromal healing, and prevented corneal vascularization and opacity. This multifunctional hydrogel offers a promising advanced therapeutic strategy for treating acute ocular alkali burns, providing potential improvements in visual outcomes and overall quality of life.

Nitric oxide (NO) is a crucial gaseous medium for tumor growth and progression, but it may also cause mitochondrial disorder and DNA damage by drastically increasing its concentration in tumor. Due to its challenging administration and unpredictable release, NO based gas therapy is difficult to eliminate malignant tumor at low safe doses. To address these issues, herein, we develop a multifunctional nanocatalyst called Cu-doped polypyrrole (CuP) as an intelligent nanoplatform ( [email protected] ) to deliver the NO precursor BNN6 and specifically release NO in tumors. Under the aberrant metabolic environment of tumors, [email protected] catalyzes the conversion of antioxidant GSH into GSSG and excess H 2 O 2 into ·OH through Cu + /Cu 2+ cycle, which results in oxidative damage to tumor cells and the concomitant release of cargo BNN6. More importantly, after laser exposure, nanocatalyst CuP can absorb and convert photons into hyperthermia, which in turn, accelerates the aforesaid catalytic efficiency and pyrolyzes BNN6 into NO. Under the synergistic effect of hyperthermia, oxidative damage, and NO burst, almost complete tumor elimination is achieved in vivo with negligible toxicity to body. Such an ingenious combination of NO prodrug and nanocatalytic medicine provides a new insight into the development of NO based therapeutic strategies. Statement of significance A hyperthermia-responsive NO delivery nanoplatform ( [email protected] ) based on Cu-doped polypyrrole was designed and fabricated, in which CuP catalyzed the conversion of H 2 O 2 and GSH into ·OH and GSSG to induce intratumoral oxidative damage. After laser irradiation, hyperthermia ablation and responsive release of NO further coupled with oxidative damage to eliminate malignant tumors. This versatile nanoplatform provides new insights into the combined application of catalytic medicine and gas therapy.
