High water absorption aerogels with three-dimensional porous structures and high specific surface areas have been fabricated from methoxy polyethylene glycol‑polycaprolactone block copolymer (mPEG‑PCL) via a facile approach by electrospinning, mechanical homogeneous shearing and dispersing short fibers in the water, freeze-drying, and heat crosslinking. The three-dimensional nanofiber aerogels (TNAs) exhibit continuous porous structures which are restructured with crisscrossed and dispersed nanofibers. The aerogel displays a low density (36 ± 3 mg/cm 3 ), a high specific surface area (230 m 2 /g), a high pore volume (5.9 cm 3 /g), a high porosity of 85% as well as an average pore diameter (1.12 μm). The as-made mPEG-PCL TNAs deliver a high average water-absorption capacity of 24.11 g/g and a good retention ability owing to the three-dimensional porous structures and large micropores population. The water absorption could still reach approximately 25 g/g after 10 compression cycles of TNAs. The average dehydration weight loss of 3.1% demonstrates that the materials are hardly soluble in the water, and the structural characteristic would not be changed after water adsorption. The antimicrobial activity against the bacteria Staphylococcus aureus and Escherichia coli are assessed that the TNAs are suitable carriers of antimicrobial agents. Moreover, as showed by cytotoxic tests, the TNAs are neglectable toxic, which are suitable for applications in environmental bioengineering. Based on the high water absorption and biocompatibility, the TNAs could be suitable candidates for health care materials and medical products.

Microcarrier cell culture systems provide one of the most promising techniques for cell amplification due to their high surface area-to-volume ratio. In this study, biodegradable polycaprolactone (PCL) microspheres tethered with carboxylated poly(glycidyl methacrylate)–REDV conjugates were developed by a combination of surface-initiated atom transfer radical polymerization (ATRP) and azide–alkyne click chemistry as conducive microcarriers for rapid cell expansion of human umbilical vein endothelial cells (HUVECs). Azido-terminated poly(glycidyl methacrylate) (PGMA-N3) brushes were grafted onto the PCL microspheres by surface-initiated ATRP. Subsequent carboxylation of PGMA-N3 brushes was accomplished by azide–alkyne click reaction with hexynoic acid. REDV peptides were covalently conjugated to the pendent carboxyl groups on the side chain of carboxylated PGMA-COOH brushes via carbodiimide chemistry to enhance the cytocompatibility of the three-dimension (3D) PCL scaffolding system. Success in each functionalization step was ascertained by the measurement of attenuated total reflectance-Fourier transform infrared spectroscopy (ATR-FTIR), X-ray photoelectron spectroscopy (XPS), scanning electron microscopy (SEM) and wet laser particle size analysis. In vitro cell-loading assay of HUVECs demonstrated a significant improvement of cell adhesion and proliferation on the REDV-immobilized PCL microsphere surfaces, and a confluent layer of HUVECs was formed after 7 days of incubation. The highly biocompatible and transportable nature of functionalized PCL microcarriers offers significant potential as a cell expansion platform.

pH/redox dual-sensitive nanoparticles (NPs) based on a poly(ε-caprolactone) (PCL) and polyethylene glycol (PEG) triblock copolymer were developed and investigated aiming at improving the drug release property of PCL in cancer chemotherapy. A redox-sensitive disulfide bond and pH-sensitive benzoic-imine linkers were introduced to the backbone of the amphiphilic block copolymer termed SCHE, which could self-assemble into stable spherical NPs in aqueous solutions with an average size of about 100 nm. Doxorubicin (DOX) as a hydrophobic anticancer drug was loaded into the NPs by a dialysis method. The in vitro drug release results showed that the accumulative release amount of DOX at pH 5.0 with 10 mM glutathione (GSH) was accelerated apparently by more than twice compared to that at pH 7.4 without GSH. After a pre-treatment with GSH, the intracellular fluorescence intensity of Hela cells was enhanced compared to those without the pretreatment, which indicated faster DOX release in cells with higher GSH concentration. In an MTT assay, no obvious toxicity of blank NPs was found. In conclusion, SCHE NPs could serve as a novel colloidal drug delivery system in cancer chemotherapy and the introduced unstable linkers could enhance the drug release rate at tumor sites.

Surface modification can enhance the biological properties of polycaprolactone (PCL) scaffolds. In this study, bionic hydrolysis and biomimetic polydopamine (PDA) coating were investigated, individually and in combination, to enhance the bioactivity of inner cube 3D-printed PCL scaffolds. The modified scaffolds were evaluated for their bioactivity, and the underlying mechanism was studied based on their physiochemical properties. The modifications significantly improved the surface roughness , hydrophilicity, surface adhesion ability, and biological properties of the scaffolds. The hydrolyzed PCL scaffolds exhibited micro-cracks and pits on their surface, with carboxyl and hydroxyl groups present. PDA, in the form of micro-particles with phenolic hydroxyl and amino groups, was coated onto the PDA-coated scaffolds. The combination-treated scaffolds had shallower pits, smaller particles, and lower levels of phenolic hydroxyl and amino groups. PDA coating demonstrated the best properties in promoting the adhesion, proliferation, and osteogenesis of rat bone marrow mesenchymal stem cells. Although the combination-treated scaffolds showed better cell proliferation than the hydrolyzed scaffolds, they did not exhibit a synergistic effect on osteogenic induction, instead weakening the osteogenic induction ability of the PDA-coating and hydrolysis treatments. Therefore, the material’s characteristics and the substantial variation of different chemical reactions must be considered during biological material modification.

Polycaprolactone (PCL) is widely utilized in bone tissue engineering due to its excellent biocompatibility and processability; however, its inherent bioinertness and hydrophobicity significantly restrict its clinical osteogenic efficacy. To overcome these limitations, we incorporated sol–gel synthesized silicate-based bioactive glass (BG) into a PCL matrix and fabricated a series of composite scaffolds with varying BG contents via direct ink writing (DIW) 3D printing. Rheological characterization confirmed that all ink formulations exhibited shear-thinning behavior, with viscosity increasing monotonically with BG content. DSC analysis revealed that BG incorporation progressively reduced the crystallinity of PCL from 51.47% to 36.23%. We systematically evaluated the physicochemical properties, mechanical resilience, and in vitro degradation behavior of these scaffolds. The results indicated that BG incorporation significantly improved the surface hydrophilicity, with the contact angle decreasing from 104.8 ± 2.81° to 69.8 ± 2.91°. Furthermore, as the BG content increased, the porosity and mechanical strength exhibited an initial increase followed by a subsequent decrease, yet all values remained within the range of human cancellous bone. Notably, cellular assays revealed that the introduction of 58SBG enhanced cell–matrix interactions; the PCL/BG scaffolds promoted superior cell attachment and more extensive morphological spreading compared to pure PCL. Among all groups, the PCL/30BG composite scaffold demonstrated the most optimal balance of mechanical integrity and biological response. Consequently, the PCL/30BG scaffold developed in this study exhibits immense potential as a bone graft substitute, providing a promising approach for clinical bone defect repair strategies.

Electrospun nanofiber membranes incorporating ZnO and Ag nanoparticles were developed using a coaxial electrospinning technique to address microbial contamination in urban water bodies. Polycaprolactone (PCL) served as the polymer matrix, and five formulations (S0–S4) with increasing nanoparticle concentrations were synthesized. Morphological analyses revealed uniform fiber formation with optimal distribution of ZnO and Ag at moderate loadings. S3 (3 wt% ZnO, 0.3 wt% AgNO₃) exhibited a mean fiber diameter of 487 nm, pore size of 2.0 µm, and porosity of 82.8 %. ATR-FTIR and XRD analyses confirmed the presence and dispersion of the incorporated nanoparticles without altering PCL’s structural integrity. Wettability improved significantly, with water contact angle decreasing from 114.3° (S0) to 93.4° (S4). Mechanical testing showed that S3 retained tensile strength (4.3 MPa) and ductility (180 % elongation). Antibacterial assays demonstrated a peak inhibition zone of 11.2 mm against Staphylococcus aureus and 10.4 mm against Escherichia coli . Filtration trials using simulated urban wastewater revealed that S3 achieved a log reduction value of 4.4 (corresponding to 99.996 % bacterial removal), reduced turbidity by 91.8 %, and lowered conductivity by 12.3 %. Despite reduced flux with increasing nanoparticle content, S3 maintained a satisfactory water flux of 112.6 L/m 2 ·h. These multifunctional membranes offer a promising route for enhancing microbial safety and improving physical water quality without the drawbacks of traditional disinfection. This study provides insights into nanoparticle–polymer interactions and offers design guidelines for practical, high-performance water purification systems in urban environments.

As gastric ulcers remain a significant clinical challenge, this study presents the development of a flexible microneedle system based on cerium/zinc-doped bioactive glass (CeZnBG) and a PCL/PLA matrix for targeted ulcer therapy. The design leverages the synergistic antioxidant and antibacterial properties of CeZnBG, combined with the localized drug delivery capability of microneedles, to enhance ulcer healing. CeZnBG was synthesized via high-temperature sintering at various temperatures (950 °C, 1050 °C, 1150 °C, and 1250 °C), and subsequently incorporated into PCL/PLA microneedles (PCMN) at doping concentrations of 0.5 %, 1.5 %, and 2.5 % to form composite materials. The structural and physicochemical characteristics of the CeZnBG-PCMN, were investigated using X-ray diffraction (XRD), scanning electron microscopy (SEM), Energy Dispersive X-ray Spectroscopy (EDS), mechanical properties, differential scanning calorimetry (DSC), and X-ray photoelectron spectroscopy (XPS). In vitro assays, including ROS scavenging and cytocompatibility, were used to assess the bioactivity of the composites. The results demonstrated that PCMN containing 1.5 % CeZnBG maintained excellent cytocompatibility (with relative cell viability exceeding 80 %) while exhibiting notable antibacterial performance, indicating promising potential for targeted gastric ulcer repair.

Cardiovascular disease has become the leading cause of death. It is the common goal for researchers worldwide to develop small-diameter vascular grafts (SDVGs) which could meet clinical needs. In this study, PLGA@PCL core-shell structural fibrous SDVGs was fabricated by coaxial electrospinning process, and then the surface heparinization of the vascular material was carried out after H2N-PEG-NH2 fixed on sodium hydroxide-treated electro-spun PCL tubes. Finally, the long-term patency and tissue regeneration of the grafts were evaluated in vivo through the rabbit carotid artery replacement model. The results indicate that the heparin-modified PLGA@PCL core-shell structural fibrous SDVGs achieved long-term patency and the arrangement of collagen and elastin in the neointima was similar to the native vessel in the rabbits after 9 months. After 3 months postoperatively, endothelialization was almost complete, and vascular calcification was also observed. It can be concluded that surface heparinization is a feasible modification method for in situ tissue-engineered vascular grafts, and controlling the occurrence of vascular calcification is another important issue to be solved in the development of SDVGs, and it is also the focus of our next research work.

Due to their high porosity, large specific surface area, and structural similarity with the extracellular matrix (ECM), electrospun nanofiber membranes are often endowed with the antibacterial properties for biomedical applications. The purpose of this study was to synthesize nano-structured Sc2O3-MgO by doping Sc3+, calcining at 600 °C, and then loading it onto the PCL/PVP substrates with electrospinning technology with the aim of developing new efficient antibacterial nanofiber membranes for tissue engineering. A scanning electron microscope (SEM) and energy dispersive X-ray spectrometer (EDS) were used to study the morphology of all formulations and analyze the types and contents of the elements, and an X-ray diffraction (XRD), thermogravimetric analysis (TGA), and Fourier transform attenuated total reflection infrared spectroscopy (ATR-FTIR) were used for further analysis. The experimental results showed that the PCL/PVP (SMCV-2.0) nanofibers loaded with 2.0 wt% Sc2O3-MgO were smooth and homogeneous with an average diameter of 252.6 nm; the antibacterial test indicated that a low load concentration of 2.0 wt% Sc2O3-MgO in PCL/PVP (SMCV-2.0) showed a 100% antibacterial rate againstEscherichia coli(E. coli).Keywords:nano-textured;Sc2O3-MgO;antibacterial;electrospinning

The core–shell structured nanofibers are routinely fabricated by coaxial solution and single-nozzle emulsion electrospinning of two polymers. Herein, the core–shell structured polymeric nanofibers were electrospun from mixed solutions of poly(ether urethane) (PEU) with poly(ε-caprolactone) (PCL) or other biodegradable aliphatic polyesters via a single-nozzle spinneret. For comparison, the mixed solutions electrospinning of poly(carbonate urethane) (PCU) with these polyesters including PCL was also conducted. The morphologies and hierarchical structures of the as-spun nanofibers were characterized by scanning electron microscopy (SEM), transmission electron microscopy (TEM), in vitro lipase degradation, differential scanning calorimeter (DSC), and Fourier-transform infrared (FTIR) analyses. It was shown that the blends of PEU with PCL or other biodegradable aliphatic polyesters were solely electrospun into the core–shell structured nanofibers with PEU as a shell and PCL or other biodegradable aliphatic polyesters as a core. The thickness of core/shell layers from 833/606 to 193.3/54.2 μm was adjustable by varying the feeding mass ratio from 1:3 to 3:1 of PEU to PCL or other biodegradable aliphatic polyesters. In DSC analysis, the Tonset of PEU@PCL core–shell fiber was 1.31 °C higher than that of the PCL fiber, while Tm was approximate. Furthermore, changing the electrospinning solvents of PEU with PCL or other biodegradable aliphatic polyesters retained the formation of core–shell nanofibers. In contrast, the blends of PCU with these polyesters tended to form co-continuous structured nanofibers. The effects of the physicochemical properties of mixed solutions on the charged liquid droplets, whipped jets, and morphology of the electrospun nanofibers were also inspected. The creation of core–shell nanofibers from the blends of PEU with PCL or other biodegradable aliphatic polyesters was most likely due to the interaction between the inherent thermodynamic phase separation of the polymers and their external stretching kinetic phase separation during electrospinning.

The occurrence of acute thrombosis, directly related to platelet aggregation and coagulant system, is a considerable reason for the failure of small-diameter vascular grafts. Heparin is commonly used as a functional molecule for graft modification due to the strong anticoagulant effect. Unfortunately, heparin cannot directly resist the adhesion and aggregation of platelets. Therefore, we have prepared a heparin-aspirin compound by coupling heparin with aspirin, an antiplatelet drug, and covalently grafted it onto the surface of polycaprolactone/polyurethane composite tube. In this way, the graft not only showed a dual function of both anticoagulation and antiplatelet, but also effectively avoided the rapid drug release and excessive toxicity to other organs caused by simple blending the medicine with material matrix. The compound retained the original function of heparin, showing good hydrophilicity and biocompatibility, which could promote the adhesion and proliferation of endothelial cells (ECs) and facilitate the process of tissue regeneration . What's more, the compound showed more effective than heparin in reducing platelet activation and preventing thrombosis. The graft modified by this compound maintained completely unobstructed for one month of implantation, while severe obstruction or stenosis occurred in PCL/PU and PCL/PU-Hep lumen at the first week, verifying the effect of the compound on preventing acute thrombosis. In general, this study proposed a designing method for small-diameter vascular graft which could prevent acute thrombosis and promote intimal construction.

We synthesized amino-modified poly(ε-caprolactone) PCN-b-PEG-b-PCN (PECN) triblock copolymers and studied the contribution of the introduced amino groups to the drug delivery efficiency of PECN nanoparticles (NPs) and their injectable thermosensitive hydrogels. PECN15 with an optimal amino group content was obtained. Firstly, the hydrophobic drug paclitaxel (PTX) was loaded into PECN15 up to 5.91% and formed PTX/PECN NPs 90 nm in size and with a slightly positive charge (7.3 mV). Furthermore, the injectable PTX/PECN NPs aqueous solution (25 wt%) at ambient temperature could undergo fast gelation at 37 °C and sustainedly release PTX/PECN NPs in 10 days. More importantly, compared with our previously reported PECT NPs, the PECN NPs without an increase in toxicity could improve the cell uptake and enhance intracellular drug release by responding to the acidic environment of the endosome. Thus, the PTX/PECN NPs presented a lower IC50 of 3.14 μg mL−1 than that of the PTX/PECT NPs (7.67 μg mL−1) and free PTX (4.65 μg mL−1). Moreover, through peritumoral injection, the PTX/PECNGel showed 94.27% inhibition rate of tumor growth on day 19, higher than PTX/PECTGel (72.28%) and Taxol® (47.03%). Therefore, the PECN NPs hydrogel provided a more effective injectable platform to enhance local cancer chemotherapy, and also provided the possibility of further functionalization by the reactive amino groups.
