Metabolic profiling, antibacterial effects, and preservation mechanisms of Paenibacillus provencensis metabolites in refrigerated Sea Bass (Perca fluviatilis)

Li Liu,Yakun Guo,Weijia Zhang,Fan Bai,Jinlin Wang,Ruichang Gao,Xiaoming Jiang,Yuanhui Zhao,Xinxing Xu

This study proposed to investigate the inhibition mechanism of the Paenibacillus provencensis (PP-2) metabolites on the dominant spoilage bacteria of sea bass and the application in preservation. Shewanella (71.11 %) and Pseudomonas (13.54 %) were the main genera. The Shewanella putrefaciens (L-1) and Pseudomonas fragi (L-4) were isolated and purified based on spoilage factors as dominant spoilage bacteria. PP-2 supernatant metabolites at minimum inhibitory concentration (MIC, 150 mg/mL) inhibited L-1 and L-4 by more than 95 % with significant antioxidant properties. The PP-2 metabolites were detected by LC-MS mainly including organic acids and derivatives, lipids molecules, organic heterocyclic compounds, and organic oxides. According to the transmission electron microscope (TEM) results, Additionally, the metabolites demonstrated antioxidant properties and disrupted bacterial cell membranes, highlighting their potential for controlling seafood spoilage. PP-2 metabolites significantly lowered pH ( p  < 0.05), TBARS values (0.33 ± 0.01 μmol/g), and TVB-N levels (24.06 ± 2.25 mg/100 g) compared to Nisin-treated and control groups after 6–8 d. After 8 d, colony counts in the PP-2 group (7.90 ± 0.01 log CFU/g) remained below the spoilage threshold. PP-2 metabolites also reduced protein oxidation, with carbonyl content at 0.33 ± 0.01 μmol/g and sulfhydryl content at 5.26 ± 0.18 μmol/g. The metabolites preserved textural properties, showing slower declines in hardness, elasticity, and chewiness, and lower juice leakage (9.37 ± 1.27 %). Metabolomic analysis revealed that PP-2 metabolites disrupted several key bacterial metabolic pathways, leading to bacterial death. These results suggest that PP-2 metabolites enhance seafood preservation by targeting bacterial metabolic processes.

Biomimetic hydrogel strategy inspired by sea cucumbers: Integrating antibacterial, osteoimmune, and osteogenic functions for infected bone repair

Xinyi Li,Mengyu Huang,Tianyi Chen,Xinyang Liu,Shuaiqi Gan,Ping Zhang,Song Chen,Ding Bai,Xinyuan Xu,Jianshu Li

Infected bone defects remain a major clinical challenge due to persistent bacterial colonization and chronic inflammation that impair bone regeneration. Chitosan modified with gallic acid (CG) had attracted considerable attention for antibacterial, reactive oxygen species (ROS) scavenging, adhesive and drug-loading capacity. However, its ability to simultaneously balance antibacterial activity, potent immunomodulation, and osteogenesis remains limited. Therefore, inspired by the hierarchical “coelomic matrix–glycoprotein layer–spicule network” of sea cucumbers achieving primary defense, immune regulation, and structural repair, we developed a self-assembling injectable hydrogel (CG/BG/Sa) composed of gallic acid–modified chitosan (CG), sialic acid (Sa), and bioactive glass (BG). The CG matrix mimicked the coelomic matrix, providing a robust antibacterial barrier (>95%), efficient ROS scavenging (91.44%), and sustained Sa release, while exhibiting strong NIR-responsive photothermal conversion that further enhanced bacterial eradication. Released Sa functioned as a glycoprotein-like layer, inducing M2 macrophage polarization (1.62-fold) to remodel the osteoimmune microenvironment and protect stem cells. Meanwhile, BG acted as dispersed spicules, enhancing mechanical stability and releasing Ca 2+ /PO 4 3− ions to promote osteogenic and angiogenic differentiation. In vivo , the photothermal-enhanced antibacterial effect further reduced inflammation and promoted osteogenesis and neovascularization. Mechanistically, CG/BG/Sa upregulated the PPAR signaling pathway while suppressing Toll-like receptor signaling, thereby accelerating bone defect repair (1.80-fold). Collectively, this biomimetic hydrogel integrated antibacterial, immunoregulatory, and osteoinductive activities, offering a promising therapeutic platform for the precise treatment of infectious bone defects.

Different drying methods drive divergent quality profiles in sea buckthorn protein hydrolysates: antioxidant preservation versus aroma complexity

Chen Chen,Pan Zhang,Yuqian Wei,Xiaoyue Liang,Le Wang,Xizhe Fu,Jian Zhang,Yue Zhao

Drying critically impacts protein structure and function. Sea buckthorn peptide industrialization is challenged by preserving structural integrity and bioactivity during drying, as thermal and mechanical stresses alter protein properties and flavor. Systematic comparisons of drying methods for sea buckthorn protein hydrolysates are lacking. This study evaluated vacuum freeze-drying (VFD), Spray-drying (SD), and microwave vacuum drying (MVD) effects on sea buckthorn protein hydrolysates (SBP) structure, antioxidant capacity, and flavor. VFD-SBP demonstrates exceptional antioxidant properties (DPPH and ABTS radical scavenging rates of 72.34 ± 4.32% and 81.56 ± 2.99%), augmented amino acid retention capabilities, and enhanced thermal stability, respectively. SD-SBP possessed richer volatiles with a roasted nutty aroma, while VFD-SBP better preserved heat-sensitive flavors, yielding a pea-like odor. In contrast to prior studies limited to single methods or parameters, this work provides a comprehensive multidimensional comparison, and offers theoretical support for the high-value utilization of sea buckthorn protein hydrolysates in functional foods and nutraceuticals.

LC-MS-based non-targeted metabolomics analysis of sea urchin during frozen storage under different packaging methods

Jia-bo Huang,Yi-hui Tang,Jing Li,Jing-ru Chen,Peng-fei Jiang

Non-targeted metabolomics was employed to investigate key metabolites and metabolic pathways influencing sea urchin quality during frozen storage (−18 °C) under different packaging methods (normal, nitrogen, and vacuum). Comparative analysis revealed significant metabolic variations between frozen and fresh samples. KEGG enrichment analyses demonstrated that the metabolism of the normal packages was more disturbed, while there was a significant accumulation of polyunsaturated fatty acids (linolenic and α-Linolenic acid). Nitrogen packaging partially mitigated oxidation, but polyunsaturated fatty acids (linolenic and eicosapentaenoic acids (EPA)) still accumulated. Vacuum packaging had the best metabolic stability, minimal changes in amino acid metabolism, and effectively inhibited the production of polyunsaturated fatty acids, thereby substantially reducing off-flavor formation. This study provides a crucial theoretical foundation for controlling quality deterioration in frozen-stored sea urchins and offers valuable insights for optimizing commercial transportation and marketing strategies.

Impact of air-impingement jet drying on the quality and flavor of Red Sea bream surimi powder

Quanquan Li,Yue Zhao,Hongwei Luan,Ying Bu,Wenhui Zhu,Xuepeng Li

The effects of air-impingement jet drying (AID) at temperature of 40–70 °C on red sea bream surimi powder were studied. Results showed that water activity was below 0.5 in all groups. Total volatile basic nitrogen (TVB-N) levels were significantly below 30 mg/100 g. The 40 °C treatment group showed the most uniform surface morphology after rehydration, with the highest water-holding capacity (WHC) and gel strength. Fourier transform infrared (FTIR) spectroscopy revealed changes in C–H, C = O, C = C, and CH2 groups. Lactic acid, citric acid, and succinic acid contributed significantly to the taste, while 5′-nucleotides contributed relatively less. Electronic tongue showed that T40 was more bitter and aftertaste, while the electronic nose revealed that T70 had higher levels of inorganic sulfides, nitrogen oxides, short-chain alkanes, and aromatic compounds. Overall, T50 represents the optimal conditions for AID-dried surimi powder. These findings provide a scientific foundation for optimizing the AID drying process.

Xanthine Oxidase Inhibitory Peptides From Sea Cucumber (Stichopus japonicus): Virtual Screening, Molecular Dynamics, and in Vitro Assessment

Siyu Liu,Jiaming Huang,Yuansen Liu,Yu Wang,Wenjie Yu,Yuqing Xia,Chunxing Li,Qin Yang,Rongping Bu,Ping Shi

This study employed a computer aided virtual screening strategy integrating water solubility, toxicity, and xanthine oxidase (XOD) binding affinity to identify novel XOD inhibitory peptides from sea cucumber (Stichopus japonicus) proteins. Two peptides, WGNEEF and GGPGDR, were successfully screened, synthesized, and experimentally validated. Molecular docking revealed that GGPGDR exhibited stronger binding affinity to XOD than WGNEEF. In vitro assays demonstrated that GGPGDR and WGNEEF exhibited significant XOD inhibitory activity, with IC 50 values of 1.09 ± 0.04 mg/mL and 2.52 ± 0.06 mg/mL, respectively. Both peptides also showed notable antioxidant capacity in a concentration dependent manner, and WGNEEF displayed particularly strong radical scavenging activity, achieving scavenging rates of 37.59%, 73.69%, and 99.01% against DPPH, ABTS + , and •OH radicals at 1.2 mg/mL. Cytotoxicity evaluation confirmed acceptable biocompatibility, with LO2 cell viability exceeding 80% for both peptides. Molecular dynamics simulations revealed that the GGPGDR/XOD complex possessed higher structural stability, as reflected by lower RMSD, a more compact conformation, and a more persistent hydrogen bond network. Collectively, these findings provide structural insight into peptide-XOD interactions and support the potential of sea cucumber-derived peptides as functional ingredients for hyperuricemia management. Practical Applications The peptides WGNEEF and GGPGDR, derived from Stichopus japonicus , exhibit xanthine oxidase inhibitory and antioxidant activities, showing potential as natural ingredients in functional foods or dietary supplements for the management of hyperuricemia and gout.

Hygroscopic properties of sodium and potassium salts as related to saline mineral dusts and sea salt aerosols

Huanhuan Zhang,Wenjun Gu,Yong Jie Li,Mingjin Tang

Mineral dust, soil, and sea salt aerosols are among the most abundant primary inorganic aerosols in the atmosphere, and their hygroscopicity affects the hydrological cycle and global climate. We investigated the hygroscopic behaviors of six Na- and K-containing salts commonly found in those primary organic aerosols. Their hygroscopic growths as a function of relative humidity (RH) agree well with thermodynamic model prediction. Temperature dependence of deliquescence RH (DRH) values for five of those salts was also investigated, which are comparable to those in literature within 1%–2% RH, most showing negative dependence on temperature. Hygroscopic growth curves of real-world soil and sea salt samples were also measured. The hygroscopic growths of two more-hydroscopic saline soil samples and of sea salt can be predicted by the thermodynamic model based on the measured water-soluble ionic composition. The substantial amounts of water-soluble ions, including Na + and K + , in saline soil samples imply that even nascent saline soil samples are quite hygroscopic at high-RH (>80%) conditions. For three less-hygroscopic dust samples, however, measurements showed higher water uptake ability than that predicted by the thermodynamic model. The small amount of water taken up by less-hygroscopic dust samples suggests that dust particles might contain thin layers of water even to very low RH. The results of this study provide a comprehensive characterization of the hygroscopicity of Na- and K-containing salts as related to their roles in the hygroscopic behaviors of saline mineral dusts and sea salt aerosols.

A Highly Sensitive 3D-Printed Flexible Sensor for Sensing Small Pressures in Deep-Sea High-Pressure Environment

Siqi Hu,Wuxu Zhang,Shengbin Li,Yuwei Wang,Zhiyi Gao,Xiangling Xia,Huiyun Xiao,Qi Zhang,Dan Xu,Feng Xu,Jinyun Liu,Baoru Bian,Yuanzhao Wu,Yiwei Liu,Jie Shang,Run-Wei Li

The origin of life on Earth is believed to be from the ocean, which offers abundant resources in its depths. However, deep-sea operations are limited due to the lack of underwater robots and rigid grippers with sensitive force sensors. Therefore, it is crucial for deep-sea pressure sensors to be integrated with mechanical hands for manipulation. Here, a flexible stress sensor is presented that can function effectively under high water pressure in the deep ocean. Inspired by biological structures found in the abyssal zone, our sensor is designed with an internal and external pressure balance structure (hollow interlocking spherical structure). The digital light processing (DLP) three-dimensional (3D) printing technology is utilized to construct this complex structure after obtaining optimized structural parameters using finite element simulation. The sensor exhibits linear sensitivity of 0.114 kPa–1 within the range of 0–15 kPa and has an extremely short response time of 32 ms, good dynamic-static load response capability, and excellent resistance cycling stability. It shows high sensitivity of 1.74 kPa–1 even under 30 MPa static water pressures and the theoretical working pressure can exceed 110 MPa, which provides a new solution for deep-sea robots.

Adverse effects of polystyrene nanoplastics on sea cucumber Apostichopus japonicus and their association with gut microbiota dysbiosis

Zelong Zhao,Xuda Wang,Jingwei Jiang,Ying Dong,Yongjia Pan,Xiaoyan Guan,Bai Wang,Shan Gao,Zhong Chen,Zunchun Zhou

The mariculture environment is a sink of microplastics (MPs) due to its enclosed nature and mass use of plastics. Nanoplastics (NPs) are MPs with a diameter <1 μm that have a more toxic effect on aquatic organisms than other MPs. However, little is known about the underlying mechanisms of NP toxicity on mariculture species. Here, we performed a multi-omics investigation to explore gut microbiota dysbiosis and associated health problems induced by NPs in juvenile sea cucumber Apostichopus japonicus , a commercially and ecologically important marine invertebrate. We observed significant differences in gut microbiota composition after 21 days of NP exposure. Ingestion of NPs significantly increased core gut microbes, especially Rhodobacteraceae and Flavobacteriaceae families. Additionally, gut gene expression profiles were altered by NPs, especially those related to neurological diseases and movement disorders. Correlation and network analyses indicated close relationships between transcriptome changes and gut microbiota variation. Furthermore, NPs induced oxidative stress in sea cucumber intestines, which may be associated with intraspecies variation in Rhodobacteraceae in the gut microbiota. The results suggested that NPs were harmful to the health of sea cucumbers, and they highlighted the importance of the gut microbiota in the responses to NP toxicity in marine invertebrates.

Influence of Centrifugation and Transmembrane Treatment on Determination of Polyphenols and Antioxidant Ability for Sea Buckthorn Juice

Dan Wu,Qile Xia,Huilin Huang,Jinhu Tian,Xingqian Ye,Yanbin Wang

When the total phenolic content (TPC) and antioxidant activity of sea buckthorn juice were assayed by spectrophotometry, the reaction solutions were not clarified, so centrifugation or membrane treatment was needed before determination. In order to find a suitable method for determining TPC and antioxidant activity, the effects of centrifugation and nylon membrane treatment on the determination of TPC and antioxidant activity in sea buckthorn juice were studied. TPC was determined by the Folin-Ciocalteau method, and antioxidant activity was determined by DPPH, ABTS, and FRAP assays. For Treatment Method (C): the sample was centrifuged for 10 min at 10,000 rpm and the supernatant was taken for analysis. Method (CF): The sample was centrifuged for 10 min at 4000 rpm, filtered by Nylon 66 filtration membranes with pore size of 0.22 μm, and taken for analysis. Method (F): the sample was filtered by Nylon 66 filtration membranes with pore size of 0.22 μm and taken for analysis. Method (N): after the sample of ultrasonic extract solution reacted completely with the assay system, the reaction solution was filtered by Nylon 66 filtration membranes with pore size of 0.22 μm and colorimetric determination was performed. The results showed that centrifugation or transmembrane treatment could affect the determination of TPC and antioxidant activity of sea buckthorn juice. There was no significant difference (p> 0.05) between methods (CF) and (F), while there was a significant difference (p< 0.05) between methods (C) (F) (N) or (C) (CF) (N). The TPC and antioxidant activity of sea buckthorn juice determined by the four treatment methods showed the same trend with fermentation time, and the TPC and antioxidant activity showed a significant positive correlation (p< 0.05). The highest TPC or antioxidant activity measured by method (N) indicates that method (N) has the least loss of TPC or antioxidant activity, and it is recommended for sample assays. Keywords:DPPH;ABTS;FRAP;centrifugation;transmembrane

Preparation and properties of fucoxanthin-loaded liposomes stabilized by sea cucumber derived cholesterol sulfate instead of cholesterol

Yunling Wang,Jinyue Yang,Yuming Wang,Yaoguang Chang,Changhu Xue,Tiantian Zhang

The preparation of steady-state phospholipid liposomes requires cholesterol as a stabilizer, but excessive intake of cholesterol may increase the risk of cardiovascular disease. The sulfated sterols extracted from sea cucumber, mainly including sulfated 24-methylene cholesterol and cholesterol sulfate, have been reported to have a variety of physiological activities. Sulfated sterols are similar to cholesterol in structure and have the potential to replace cholesterol to prepare novel stable multifunctional liposomes, allowing the liposomes to act as carriers for the delivery of less bioavailable nutrients while allowing sulfated sterols in the lipid bilayer to exert physiologically active effects. This study aimed to prepare a novel multifunctional nanoliposome stabilized with sulfated sterols from sea cucumber instead of cholesterol by ultrasound-assisted thin-film dispersion method. The results showed that stable and uniformly dispersed nanoliposomes could be formed when the substitution ratio of sea cucumber-derived cholesterol sulfate was 100% and the ratio of lecithin to cholesterol sulfate was 3:1. Fucoxanthin encapsulated liposome with egg yolk lecithin/sea cucumber-derived cholesterol sulfate/fucoxanthin mass ratio of 6:2:3 was successfully prepared, with an average particle size of 214 ± 3 nm, polydispersity index (PDI) value of 0.297 ± 0.006, the zeta potential of −57.2 ± 1.10 mV, and the encapsulation efficiency of 85.5 ± 0.8%. The results of digestion and absorption in vitro and in vivo showed that liposomes could significantly improve the bioavailability of fucoxanthin and prolong its residence time in serum. As an efficient multifunctional carrier, this novel liposome has great potential for applications in functional foods and biomedicine .

Structural Characterization and Anticoagulant Activities of a Keratan Sulfate-like Polysaccharide from the Sea Cucumber Holothuria fuscopunctata

Ru Chen,Weili Wang,Ronghua Yin,Ying Pan,Chen Xu,Na Gao,Xiaodong Luo,Jinhua Zhao

A sulfated polysaccharide (AG) was extracted and isolated from the sea cucumberH. fuscopunctata, consisting of GlcNAc, GalNAc, Gal, Fuc and lacking any uronic acid residues. Importantly, several chemical depolymerization methods were used to elucidate the structure of the AG through a bottom-up strategy. A highly sulfated galactose (oAG-1) and two disaccharides labeled with 2,5-anhydro-D-mannose (oAG-2, oAG-3) were obtained from the deaminative depolymerized product along with the structures of the disaccharide derivatives (oAG-4~oAG-6) identified from the free radical depolymerized product, suggesting that the repeating building blocks in a natural AG should comprise the disaccharide β-D-GalS-1,4-D-GlcNAc6S. The possible disaccharide side chains (bAG-1) were obtained with mild acid hydrolysis. Thus, a natural AG may consist of a keratan sulfate-like (KS-like) glycosaminoglycan with diverse modifications, including the sulfation types of the Gal residue and the possible disaccharide branches α-D-GalNAc4S6S-1,2-α/β-L-Fuc3Slinked to the KS-like chain. Additionally, the anticoagulant activities of the AG and its depolymerized products (dAG1-9) were evaluated in vitro using normal human plasma. The AG could prolong activated partial thromboplastin time (APTT) in a dose-dependent manner, and the activity potency was positively related to the chain length. The AG and dAG1-dAG3 could prolong thrombin time (TT), while they had little effect on prothrombin time (PT). The results indicate that the AG could inhibit the intrinsic and common coagulation pathways. Keywords:aminoglycan;structural features;keratan sulfate;depolymerization;oligosaccharides;anticoagulant activity
