Biomineralization-inspired porous calcium carbonate microspheres as a controlled release system of herbicides/pesticides

Yaoxun Zeng,Xinyi Li,Fangzheng Chen,Huiling Ye,Keyi Rong,Zhili Ran,Binhong Liu,Zhenxing Pan,Xiuxian Xie,Junze Tang,Xujie Liu,Yan He

Low bioavailability and environmental pollution are the key problems that restrict the sustainable development of traditional herbicides and pesticides. In this study, inspired by biomineralization, we prepared porous calcium carbonate microspheres (PCMs) using polyvinylpyrrolidone (PVP) and carboxymethyl chitosan (CMCs) as particle size and morphology modifiers. PCMs have large specific surface area (87.6 m 2 /g) and mesoporous structure, which allows them to efficiently load herbicides (497±24 mg/g) and pesticides (318±17 mg/g). Additionally, the PCMs exhibit pH-responsive decomposition, enabling responsive release of glyphosate/chlorpyrifos in weak acid environments. In the simulation experiments, we found that the PCMs improved the adhesion ability of glyphosate/chlorpyrifos on the leaf surface and reduced the glyphosate/chlorpyrifos loss rate (1.69-fold reduction for glyphosate; 1.68-fold reduction for chlorpyrifos). Furthermore, glyphosate/chlorpyrifos loaded PCMs (GLY@PCMs/CPF@PCMS) exhibited herbicidal/insecticidal performance with the same efficacy as glyphosate/chlorpyrifos. Therefore, the PCMs provide an effective method for controlling herbicides/pesticides loss, controlled release of herbicides/pesticides, and improving herbicides/pesticides utilization efficiency. As a result, PCMs have the potential to be used as a pesticide/herbicide controlled release system, providing new ideas and theoretical support for the design of multifunctional nanoherbicides/nanopesticides in the future.

Tri-layer structured intelligent hydrogel dressing for controlled-release antibacterial treatment of chronic diabetic wounds

Weikai Zhang,Xiuhong Fu,Hewei Zhang,Shaozhe Zhang,Qingwei Zhang,Chongkai Zhai,Jinyang Zhao,Quanzhong Yang,Shuang Li

Chronic diabetic wounds are notoriously difficult to heal due to a hyperglycemic environment, persistent oxidative stress, and infection, leading to slow healing and frequent complications. In this study, we developed a tri-layer intelligent hydrogel dressing for diabetic wound care. The top layer contains silver nanoparticles (AgNPs) for rapid initial antibacterial action. The middle layer incorporates poly(lactic-co-glycolic acid) (PLGA) microspheres loaded with mangiferin to achieve sustained drug release and promote angiogenesis. The bottom layer is a gelatin methacryloyl (GelMA) hydrogel scaffold that provides structural support. The composite materials' morphology and structure were characterized by scanning and transmission electron microscopy (SEM, TEM). We evaluated the properties of hydrogel contain mechanical properties, degradation and swelling behaviors, drug release profiles, antibacterial and antibiofilm effects, and cell-related biological properties. The results showed that the tri-layer hydrogel exhibited higher mechanical strength and greater degradation stability than a homogeneous GelMA hydrogel. The layered structure of hydrogel effectively delayed the release of mangiferin, resulting in a significantly lower cumulative release over 72 hours compared to a non-layered control. Antibacterial tests demonstrated that the tri-layer hydrogel produced the largest inhibition zones and the highest bacterial-killing rates against Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli), and it markedly inhibited the formation of bacterial biofilms. In vitro, cell studies showed that the tri-layer hydrogel significantly downregulated the pro-inflammatory cytokines IL-6 and TNF-α, upregulated the anti-inflammatory cytokine IL-10, and substantially reduced intracellular reactive oxygen species (ROS) accumulation. In summary, the tri-layer intelligent hydrogel dressing, through its synergistic controlled-release and antibacterial mechanisms, offers an effective multi-factorial intervention strategy for the treatment of chronic diabetic wounds.

Pillar[6]arene@quantum-Dot Membrane for Controlled Release Pesticides Based on Host–Guest Chemistry

Guang Li,Siyun Zhang,Lei Yang,Weiwei Xu,Haonan Qu,Demei Tian,Jia Li,Haibing Li,Xiaobing Zhang

Controlled release of pesticides is highly desirable for improving efficacy and reducing side effects in agricultural applications. Herein, in this work, based on the optical property and host–guest chemistry of quantum dot and pillararene, we develop a new type of fluorescent, current-responsive, pH stimuli-responsive pillar[6]arene@quantum dots mixed-matrix membranes (PQDs-MMMs) for pesticide delivery, which achieves highly selective detection and controlled release for methyl viologen (MV2+). In the experiments of fluorescence spectrum and I–V current, PQDs-MMMs could realize the visual fluorescence detection and current selection for MV2+. Moreover, the controlled release experiments indicated that the PQDs-MMMs could effectively immobilize and stimuli-release the MV2+ depending on the pH regulation, which achieved the pH-stimuli-responsive release of MV2+. Additionally, the herbicidal activity of MV2+-loaded PQDs-MMMs (MV2+@PQDs-MMMs) was tested, and results showed the weed control effects were identified to free MV2+ under pH control. This work provides the great potential of PQDs-MMMs used in agricultural membranes with environmentally friendly and stimuli-responsive release.

Celery cellulose hydrogel as carriers for controlled release of short-chain fatty acid by ultrasound

Ling Yan,Lei Wang,Shengjie Gao,Changhong Liu,Zhengzhu Zhang,Aijin Ma,Lei Zheng

The feasibility of using celery cellulose hydrogels as carriers was explored for controlled release of short-chain fatty acids (SCFAs) triggered by ultrasound. The hydrogels were prepared with the phase inversion method and further characterized using FT-IR, SEM and XRD techniques. At the optimal cellulose concentration (8.33 and 6.25 mg/mL), the hydrogels (F 4 and F 5 ) exhibited the swelling ratio of 185%, and Young’s modulus of the F 4 and F 5 was lower than that of others. The hydrogels were loaded with SCFAs owing to its hydrophilicity and swelling properties, and the maximum loading capacity of SCFAs achieved nearly 80%. Interestingly, the loaded SCFAs within hydrogel carrier could be readily released if an ultrasound trigger is exerted. Our results indicate that the ultrasound-triggered strategy for the SCFAs delivery system could provide a promising basis to achieve on-demand, reproducible, repeated, and tunable dosing of bioactive molecules.

Controlled-release of apatinib for targeted inhibition of osteosarcoma by supramolecular nanovalve-modified mesoporous silica

Xinglong Wang,Gongke Li,Ke Li,Yu Shi,Wenzheng Lin,Chun Pan,Dandan Li,Hao Chen,Jianwei Du,Huihui Wang

Targeted delivery of antitumor drugs has been recognized as a promising therapeutic modality to improve treatment efficacy, reduce the toxic side effects and inhibit tumor recurrence. In this study, based on the high biocompatibility, large specific surface area, and easy surface modification of small-sized hollow mesoporous silica nanoparticles β-cyclodextrin (β-CD)-benzimidazole (BM) supramolecular nanovalve, together with bone-targeted alendronate sodium (ALN) were constructed on the surface of small-sized HMSNs. The drug loading capacity and efficiency of apatinib (Apa) in HMSNs/BM-Apa-CD-PEG-ALN (HACA) were 65% and 25%, respectively. More importantly, HACA nanoparticles can release the antitumor drug Apa efficiently compared with non-targeted HMSNs nanoparticles in the acidic microenvironment of the tumor. In vitro studies showed that HACA nanoparticles exhibited the most potent cytotoxicity in osteosarcoma cells (143B cells) and significantly reduced cell proliferation, migration and invasion. Therefore, the drug-efficient release of antitumor effect of HACA nanoparticles is a promising way to treat osteosarcoma.

Controlled release of silyl ether camptothecin from thiol-ene click chemistry-functionalized mesoporous silica nanoparticles

Yue Yan,Jie Fu,Tianfu Wang,Xiuyang Lu

As efficient drug carriers, stimuli-responsive mesoporous silica nanoparticles are at the forefront of research on drug delivery systems. An acid-responsive system based on silyl ether has been applied to deliver a hybrid prodrug. Thiol-ene click chemistry has been successfully utilized for tethering this prodrug to mesoporous silica nanoparticles . Here, by altering the steric bulk of the substituent on the silicon atom, the release rate of a model drug, camptothecin, was controlled. The synthesized drug delivery system was investigated by analytical methods to confirm the functionalization and conjugation of the mesoporous silica nanoparticles. Herein, trimethyl silyl ether and triethyl silyl ether were selected to regulate the release rate. Under normal plasma conditions (pH 7.4), both types of camptothecin-loaded mesoporous silica nanoparticles (i.e., MSN-Me-CPT and MSN-Et-CPT) did not release the model drug. However, under in vitro acidic conditions (pH 4.0), based on a comparison of the release rates, camptothecin was released from MSN-Me-CPT more rapidly than from MSN-Et-CPT. To determine the biocompatibility of the modified mesoporous silica nanoparticles and the in vivo camptothecin uptake behavior, MTT assays with cancer cells and confocal microscopy observations were conducted, with positive results. These functionalized nanoparticles could be useful in clinical treatments requiring controlled drug release . Statement of Significance As the release rate of drug from drug-carrier plays important role in therapy effects, trimethyl silyl ether (TMS) and triethyl silyl ether (TES) were selected as acid-sensitive silanes to control the release rates of model drugs conjugated from MSNs by thiol-ene click chemistry. The kinetic profiles of TMS and TES materials have been studied. At pH 4.0, the release of camptothecin from MSN-Et-CPT occurred after 2   h, whereas MSN-Me-CPT showed immediate drug release. The results showed that silyl ether could be used to control release rates of drugs from MSNs under acid environment, which could be useful in clinical treatments requiring controlled drug release .

Controlled release of recombinant human cementum protein 1 from electrospun multiphasic scaffold for cementum regeneration

Xiaofeng Chen,Yu Liu,Leiying Miao,Yangyang Wang,Shuangshuang Ren,Xuebin Yang,Yong Hu,Weibin Sun

Periodontitis is a major cause for tooth loss, which affects about 15% of the adult population. Cementum regeneration has been the crux of constructing the periodontal complex. Cementum protein 1 (CEMP1) is a cementum-specific protein that can induce cementogenic differentiation. In this study, poly(ethylene glycol) (PEG)-stabilized amorphous calcium phosphate (ACP) nanoparticles were prepared by wet-chemical method and then loaded with recombinant human CEMP1 (rhCEMP1) for controlled release. An electrospun multiphasic scaffold constituted of poly(ε-caprolactone) (PCL), type I collagen (COL), and rhCEMP1/ACP was fabricated. The effects of rhCEMP1/ACP/PCL/COL scaffold on the attachment proliferation, osteogenic, and cementogenic differentiations of human periodontal ligament cells, (PDLCs) were systematically investigated. A critical size defect rat model was introduced to evaluate the effect of tissue regeneration of the scaffolds in vivo. The results showed that PEG-stabilized ACP nanoparticles formed a core-shell structure with sustained release of rhCEMP1 for up to 4 weeks. rhCEMP1/ACP/PCL/COL scaffold could suppress PDLCs proliferation behavior and upregulate the expression of cementoblastic markers including CEMP1 and cementum attachment protein while downregulating osteoblastic markers including osteocalcin and osteopontin when it was cocultured with PDLCs in vitro for 7 days. Histology analysis of cementum after being implanted with the scaffold in rats for 8 weeks showed that there was cementum-like tissue formation but little bone formation. These results indicated the potential of using electrospun multiphasic scaffolds for controlled release of rhCEMP1 for promoting cementum regeneration in reconstruction of the periodontal complex.

Enhanced food waste anaerobic digestion: An encapsulated metal additive for shear stress-based controlled release

Le Zhang,Jingxin Zhang,Kai-Chee Loh

Eco-friendly delivery carrier of trace metals is very significant to appropriate metal supplementation to improve metal bioavailability and prevent metal precipitation in anaerobic digesters . A novel encapsulated form of dry water (DW) additive was synthesized by the high-speed mixing of nickel ions solution with hydrophobic silica nanoparticles and was characterized using scanning electron microscope (SEM), nuclear magnetic resonance (NMR) and particle size analyzer. Nickel (Ni) was chosen as a model trace element. The encapsulated structure of DW additive and the feasibility of controlled release of Ni ions from DW additive in pure water and anaerobic digesters were verified. The optimal mixing speed for the highest methane yield of digesters with DW additive was determined as 800–1000 rpm. By varying stirring speed, controlled release of trace metal element (Ni) into digester was easily achieved during anaerobic digestion of food waste. The results showed that nickel ions delivery via DW additive significantly enhanced the metal bioavailability by 18.9–42.6% and process stability during 59 days of operation, resulting in an increase of 11.6–31.8% in methane yields compared to the controls. Pyrosequencing analysis indicated that bacterial communities were dominated by phyla Firmicutes , Bacteroidetes , Proteobacteria , Chloroflexi and Spirochaetes while methanogenic genus Methanolinea , Methanoculleus , Methanothrix , Methanomassiliicoccus and Methanospirillum were more abundant in archaea communities. Further, phyla Bacteroidetes , Proteobacteria and Methanoculleus were selectively enriched by the DW additive. Results indicated that it was feasible to apply the method of DW additive supplementation to enhance anaerobic digestion of food waste for biogas production by improving metal bioavailability.

Ultrasound Controlled-Release Hydrogel Promotes Diabetic Wound Healing via Neuroimmune Modulation and Synergistic ROS Scavenging

Mofan Li,Mengxin Wang,Haonan Wang,Yang Sun,Yongyue Zhang,Shuyu Xu,Tianjiao Zhang,Shiti Shama,Xiaolong Liang,Shumin Wang

Diabetic wound (DW) is a diabetes complication characterized by high morbidity and disability rates. Previous therapeutic systems focused on macrophages while neglecting the upstream regulatory factor of neuropeptide-mediated neuroimmune communication. In addition, precise delivery is directly important for the treatment of DW. This study constructed an amphiphilic prodrug molecule MC by covalently conjugating calcitonin gene-related peptide (CGRP) with manganese porphyrin (MnP). MC was then co-assembled with DSPE-PEG-folic acid to form targeted nanoparticles MCF. Subsequently, MCF was loaded into an ultrasound-responsive hydrogel to obtain the MCF@CA system, integrating neuroimmune modulation and reactive oxygen species (ROS) scavenging functions. Upon local administration, ultrasound triggering enables the on-demand release of the nanodrug MCF from MCF@CA. Subsequently, FA targets M1 macrophages, prolonging wound retention time. MnP scavenges ROS, improving fibroblast function and promoting macrophage polarization towards an anti-inflammatory phenotype. This study presents an ultrasound-responsive hydrogel MCF@CA delivering targeted nanoparticles where CGRP regulates the regenerative transition of the immune microenvironment. Animal experiments confirmed that MCF@CA combined with ultrasound significantly promotes DW healing by enhancing collagen deposition, immune modulation, and improving blood supply. Therefore, this study provides an on-demand controlled delivery platform with clear translational potential for diabetic wound therapy.

Hierarchically electrospraying a PLGA@chitosan sphere-in-sphere composite microsphere for multi-drug-controlled release

Liu Zhu,Ye Weilong,Zheng Jingchuan,Wang Qindong,Ma Guowu,Liu Huiying,Wang Xiumei

Sequential administration and controlled release of different drugs are of vital importance for regulating cellular behaviors and tissue regeneration, which usually demands appropriate carriers like microspheres (MS) to control drugs releases. Electrospray has been proven an effective technique to prepare MS with uniform particle size and high drug-loading rate. In this study, we applied electrospray to simply and hierarchically fabricate sphere-in-sphere composite microspheres, with smaller poly(lactic-co-glycolic acid) MS (∼8–10 μm in diameter) embedded in a larger chitosan MS (∼250–300 μm in diameter). The scanning electron microscopy images revealed highly uniform MS that can be accurately controlled by adjusting the nozzle diameter or voltage. Two kinds of model drugs, bovine serum albumin and chlorhexidine acetate, were encapsulated in the microspheres. The fluorescence-labeled rhodamine-fluoresceine isothiocyanate (Rho-FITC) and ultraviolet (UV) spectrophotometry results suggested that loaded drugs got excellent distribution in microspheres, as well as sustained, slow release in vitro. In addition, far-UV circular dichroism and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS) results indicated original secondary structure and molecular weight of drugs after electrospraying. Generally speaking, our research proposed a modified hierarchically electrospraying technique to prepare sphere-in-sphere composite MS with two different drugs loaded, which could be applied in sequential, multi-modality therapy.

Biomass-based multifunctional fertilizer system featuring controlled-release nutrient, water-retention and amelioration of soil

Xinggang Wang,Shaoyu Lü,Chunmei Gao,Xiubin Xu,Yi Wei,Xiao Bai,Chen Feng,Nannan Gao,Mingzhu Liu,Lan Wu

In an effort to enhance the efficiency of fertilizer use and minimize adverse environmental effects, a novel biomass-based, multifunctional controlled-release fertilizer (BMCF) was prepared to improve nutrient use efficiency and enhance crop production systems for more sustainable agriculture practices. The fertilizer design included natural attapulgite as a matrix, co-granulated ammonium zinc phosphate and urea as fertilizer core, cellulose acetate butyrate (CAB) as an inner coating, and a carboxymethyl chitosan-g-poly(acrylic acid)/attapulgite (CMCS-g-PAA/APT) superabsorbent composite as an outer coating. The effect of an APT matrix, CAB inner coating and superabsorbent composite outer coating on nutrient release rate was investigated. The influence of the BMCF on water-holding and water-retention capacity of soil samples was determined. The degradation behavior of the CMCS-g-PAA/APT outer coating in soil solution was evaluated. The experimental results indicated that the product prepared by a simple and economical method can effectively reduce N leaching loss and runoff, improve soil moisture retention capacity, and ameliorate soil acidity and alkalinity.

Long-term and controlled release of chlorhexidine–copper(II) from organically modified montmorillonite (OMMT) nanocomposites

Yue Wu,Ninglin Zhou,Wenhao Li,Hao Gu,Yunting Fan,Jiang Yuan

Drug/metal ion complexes exhibit improved antimicrobial activity and intercalating the above complexes into the interlayer of clay endows a long-term and controlled-release behavior. In this study, chlorhexidine was first complexed with copper (II) ion and then intercalated into the interlayer of MMT to form chlorhexidine–copper (II)/montmorillonite (CHX–Cu/MMT) nanocomposites. The nanocomposites were characterized with Fourier transform infrared (FT-IR), X-ray diffraction (XRD) and thermogravimetric analysis (TGA). A nearly lateral-monolayer arrangement of CHX–Cu was supposed for the intercalation. Release kinetics indicated that the release process satisfied a pseudo-second-order mode. The antibacterial results showed that the CHX–Cu/MMT composites had long-term and controlled-release behavior.
