Liposome, a promising delivery system with phospholipid bilayer, could improve the water solubility of curcumin, which is a polyphenolic natural product with many bioactivities. However, the instability of liposome limits its development in application. In this study, the chitosan was used to protect the structure stability of phospholipid bilayer of curcumin liposome and control the sustained release rate of curcumin further. The temperature stability and phase transition were studied by differential scanning calorimetry. When curcumin molecule was embedded in phospholipid bilayer, the pre-transition temperature (50.40 °C) and main phase transition temperature (61.47 °C) decreased about 10 °C compared to it without curcumin, and phospholipid bilayer fluidity was increased. The pre-transition temperature was disappeared after chitosan coated on the surface of curcumin liposome. The thermal stability test and X-ray diffraction also indicate that high temperature could change the structure of liposome and chitosan could protect the curcumin from damage and leak. The in vitro release study showed that the cumulative release rate got faster with temperature increased and chitosan could decrease the release rate. These results may guide the potential application of chitosan-coated liposome as a carrier of curcumin in nutraceutical and functional foods.

Dexanabinol (HU-211) is an artificially synthesized cannabinoid derivative that exerts neuroprotective effects through anti-inflammatory and antioxidant effects. Curcumin exhibits antidepressant effects in the treatment of major depressive disorder. To investigate the antidepressant effects of solid lipid nanoparticles loaded with both curcumin and dexanabinol, and the underlying mechanisms associated with this combination, we established wild-type ( CBR1 +/+ ) and cannabinoid receptor 1 (CBR1) knockout ( CBR1 –/– ) mouse models of major depressive disorder, through the intraperitoneal injection of corticosterone, for 3 successive days, followed by treatment with intraperitoneal injections of solid lipid nanoparticles loading with curcumin (20 mg/kg) and dexanabinol (0.85 mg/kg), for 2 successive days. Our results revealed that solid lipid nanoparticle loading with curcumin and dexanabinol increased the mRNA and protein expression levels of the mature neuronal markers neuronal nuclei, mitogen-activated protein 2, and neuron-specific beta-tubulin III, promoted the release of dopamine and norepinephrine, and increased the mRNA expression of CBR1 and the downstream genes Rasgef1c and Egr1 , and simultaneously improved rat locomotor function. However, solid lipid nanoparticles loaded with curcumin and dexanabinol had no antidepressant effects on the CBR1 –/– mouse models of major depressive disorder. This study was approved by the Institutional Ethics Committee of Tongji Hospital of Tongji University, China (approval No. 2017-DW-020) on May 24, 2017.

Perylenediimide-cored (PDI-cored) polymers have been widely applied as fluorescent drug carriers due to their high fluorescence quantum yield and outstanding chemical/photophysical stability. However, most of these delivery systems lack detailed drug delivery specifics, especially when and where the drug is released. In this study, a PDI-based nanocarrier (PPL-B) is constructed and applied to monitor curcumin release with an “off–on” fluorescence switch. The star-shaped drug carrier has an amphiphilic structure with fluorescent PDI as the core and poly(amino acid) as the shell, which is suitable for drug delivery. A model drug curcumin is loaded to PPL-B and quenches its fluorescence significantly through photo-induced electron transfer (PET). Upon curcumin release, the fluorescence of PPL-B achieves an “off–on” switch as a result of PET interruption. In light of this, curcumin release is successfully monitored by CUR@PPL-B at the cellular level. This PDI-based nanocarrier for drug release monitoring with a PET mechanism shows great potential in the drug delivery field.

Oral ulcers are a common oral mucosal disease that seriously affect the quality of life. Traditional drug treatments have shown unsatisfactory efficacy and potential adverse reactions. In this study, curcumin-loaded multifunctional magnesium metal–organic framework-embedded hyaluronic acid-soluble microneedles patches were developed to optimize treatment strategies for oral ulcers. This microneedles patch achieves efficient release of curcumin and Mg 2+ in the ulcer through precisely targeted delivery and controllable release mechanism, significantly regulates inflammation, promotes cell migration and angiogenesis, and accelerates the ulcer healing process. At the same time, the synergistic effect of curcumin and gallic acid effectively alleviated oxidative stress, while the backplate ε-poly- l -lysine and needle tip Mg 2+ jointly constructed an antibacterial barrier to effectively inhibit pathogens. Verification using an oral ulcer rat model showed that the microneedles patch exhibited excellent therapeutic effects. This not only opens up a new avenue for clinical oral treatment but also marks a breakthrough in nanobiomaterials science and drug delivery technology and heralds a broad prospect in the field of oral ulcer treatment in the future. Smart-responsive wound dressings are promising materials for rapid wound healing and infection clearance in a painless and noninvasive method. In this work, we developed a smart wound dressing, which was composed of polyaniline (PANI) and curcumin (Cur) composite nanofiber (PANI-Cur) by electrospinning. The regulation of interfacial electrons occurred through hydrogen bonds and electrostatic interaction between Cur and PANI, resulting in enhanced light absorption ability, lower bandgap, and better separation and transfer of interfacial electrons compared with that of PANI and Cur. The process of separation and transfer of interfacial electrons would be promoted under the assistance of light based on in situ Kelvin probe force microscopy (KPFM). This PANI-Cur hybrid nanofiber showed an excellent sterilization effect against 99.77 % ± 0.10 % of Staphylococcus aureus and 98.45 % ± 0.27 % of Escherichia coli under light irradiation . The PANI-Cur nanofiber exhibited an accelerated wound healing rate when used for repairing mouse skin injuries under light. This work could provide a reference for the preparation of intelligent antibacterial fibers.

To achieve the effective loading and delivery of curcumin, novel disulfide-crosslinked nanoparticles based on modified dextrin were developed for the encapsulation of curcumin. Thiolated dextrin (Dt-SH) was obtained via sodium periodate oxidation and cysteamine grafting. The Dt-SH exhibited a rough, flake-like morphology, was classified as an amorphous material and demonstrated enhanced enzyme resistance. Subsequently, spherical nanoparticles with sizes ranging from 92.52 to 157.12 nm and zeta potentials between +23.59 and + 29.90 mV were self-assembled in an aqueous solution. Thiol modification promoted interconnection and aggregation of the nanoparticles. These nanoparticles exhibited pH-dependent size variations. Taking curcumin as a hydrophobic model, nanoparticles showed intestinal targeted release in vitro . Fluorescence spectroscopy and thermodynamic analysis indicated that curcumin bound to Dt-SH nanoparticles primarily through hydrogen bonding and van der Waals forces, with hydrophobic interactions contributing. These findings supported the potential of thiolated dextrin nanoparticles in the effective delivery of hydrophobic compounds.

The curcumin-malic acid-aspartic acid polymer (PCMA) as a new water treatment agent was prepared by solid phase synthesis of curcumin, malic acid and aspartic acid. The static scale inhibition experiments showed that PCMA can inhibit CaCO 3 and CaSO 4 scale formation by 100.0 % and had excellent scale inhibition effect under various experimental conditions. The mechanism of action of PCMA was obtained by X-ray diffraction, scanning electron microscope and molecular dynamics simulation. Electrochemical test showed that PCMA is an anodic corrosion inhibitor that achieves 93.1 % corrosion inhibition by forming a protective film on the surface of Q235 carbon steel. Besides, fluorescence spectra proved that PCMA has stable fluorescence intensity.

Curcumin is widely recognized for its diverse antitumor properties, ranging from breast cancer to many other types of cancers. However, its role in the tumor microenvironment remains to be elucidated. In this study, we established a 3D tumor spheroids model that can simulate the growth environment of tumor cells and visualized the antitumor metabolic alteration caused by curcumin using mass spectrometry imaging technology. Our results showed that curcumin not only exerts a profound impact on the growth and proliferation of breast cancer cells but in situ multivariate statistical analysis also reveals the significant effect on the overall metabolic profile of tumor spheroids. Meanwhile, our visualization map characterized curcumin metabolic processes of reduction and glucuronidation in tumor spheroids. More importantly, abnormal metabolic pathways related to lipid metabolism and polyamine metabolism were also remodeled at the metabolite and gene levels after curcumin intervention. These insights deepen our comprehension of the regulatory mechanism of curcumin on the tumor metabolic network, furnishing powerful references for antitumor treatment.

A thrombin-responsive curcumin liposome-modified PCL/PLGA vascular graft (uPA-Cur-PDA@PCL/PLGA) was developed. • uPA-Cur-PDA@PCL/PLGA enabled on-demand uPA release for thrombolysis upon thrombin stimulation. • uPA-Cur-PDA@PCL/PLGA scavenged ROS and promoted M2 macrophage polarization via curcumin liposomes. • In vivo, uPA-Cur-PDA@PCL/PLGA maintained 3-month patency, promoted endothelialization, and reduced inflammation. • This strategy offers a promising approach for antithrombosis and vascular regeneration in small-diameter vascular grafts. The failure of small-diameter vascular grafts (SDVGs) is primarily attributed to thrombosis and pathological inflammation, leading to incomplete endothelialization and ultimately poor long-term patency. Current antithrombotic strategies are limited by insufficient durability and the risk of systemic coagulation disturbances associated with continuous anticoagulation. Herein, we developed a thrombin-responsive vascular graft enabling on-demand thrombolysis and modulation of the pathological microenvironment. Curcumin was encapsulated into liposomes to overcome its poor bioavailability, endowing the graft with reactive oxygen species (ROS) scavenging and immunomodulatory effects. Thrombin-responsive curcumin-loaded liposomes (uPA-Cur-Lip) were then constructed by sequentially conjugating a thrombin-cleavable peptide and urokinase-type plasminogen activator (uPA) to the liposomes. Subsequently, uPA-Cur-Lip was immobilized onto electrospun PCL/PLGA vascular grafts via polydopamine (PDA) coating, yielding uPA-Cur-PDA@PCL/PLGA. In vitro studies confirmed sustained curcumin release from the liposomes and thrombin-responsive "on-demand" thrombolytic activity. Furthermore, uPA-Cur-PDA@PCL/PLGA enhanced endothelial cell migration, alleviated oxidative stress, and promoted M2 macrophage polarization. The results of in vivo rat abdominal aorta implantation showed that uPA-Cur-PDA@PCL/PLGA exhibited sustained patency for 3 months without thrombosis, along with improved endothelialization and reduced inflammation. Overall, we developed a thrombin-responsive curcumin liposome-modified SDVG that enables on-demand thrombolysis and modulates the local microenvironment, thereby promoting vascular regeneration and improving long-term graft performance. Background Acute kidney injury (AKI) is a critical clinical syndrome with high morbidity and mortality, primarily driven by mitochondrial oxidative stress and tubular epithelial cell apoptosis. Current antioxidant therapies are limited by poor bioavailability and lack of renal specificity. To address this, we developed a dual-targeting nanomedicine based on ultrasmall chitosan oligosaccharide-functionalized ruthenium-curcumin nanodots (LMWC/Ru-Cur).Methods Ru-Cur coordination polymer nanodots were synthesized and subsequently coated with low-molecular-weight chitosan (LMWC). The nanoparticles were characterized for size, surface charge, stability, and antioxidant capacity. In vitro studies using HK-2 cells assessed cytocompatibility, cellular uptake, and protection against H2O2- or cisplatin-induced injury via measurements of viability, mitochondrial ROS, membrane potential, and apoptosis. In vivo efficacy and biodistribution were evaluated in murine models of ischemia-reperfusion- and cisplatin-induced AKI.Results The resulting LMWC/Ru-Cur nanodots exhibited uniform size (~7.6 nm), good aqueous stability, and potent broad-spectrum radical scavenging ability. They were efficiently internalized by renal tubular cells via megalin receptor-mediated endocytosis, leading to significantly enhanced renal accumulation. Treatment with LMWC/Ru-Cur attenuated oxidative stress, restored mitochondrial function, reduced apoptosis in injured HK-2 cells, and improved renal function (serum creatinine and blood urea nitrogen), histopathology, and inflammatory cytokine levels in both AKI models, outperforming free curcumin or unmodified Ru-Cur. The nanodots also demonstrated favorable short-term biocompatibility and in vivo biosafety. Conclusion LMWC/Ru-Cur nanodots represent a promising targeted nanotherapeutic strategy for AKI, integrating passive glomerular filtration with active receptor-mediated tubular delivery to effectively mitigate oxidative stress and mitochondrial damage, thereby preserving renal function. This work provides a rational design for metal-polyphenol based nanomedicines in the treatment of acute organ injury.

A novel ethyl cellulose-sodium alginate/chitosan/curcumin (EC-SA/CS/Cur) composite microgel system was developed through a one-step emulsion method. Optimized homogenization at 1,300 rpm for 2 min enabled consistent fabrication of EC-SA/CS/Cur microgels with a mean particle size of 2.85 μm. The microstructural morphology, particle size distribution, and chemical composition of the optimized EC-SA/CS/Cur microgels were characterized by microscopy, laser particle size analysis, and Fourier-transform infrared spectroscopy. The microgels exhibited a curcumin encapsulation efficiency of 54.4 ± 6.8% and a drug loading capacity of 0.032 ± 0.004%. The microgels exhibited excellent stability under enzymatic and physiological pH environments while displaying a unique sustained-release profile in ethanol-containing media; under agitated conditions, a cumulative release of 3.2 μg/mL was achieved within 6 h, representing a 28.0% increase in cumulative release compared to static conditions. The minimum inhibitory concentration of EC-SA/CS/Cur microgels was determined as 675 μg/mL; after 3 h of exposure, their antimicrobial efficacy significantly outperformed free curcumin. These results confirm that the EC-SA/CS microgel system acts as an effective delivery vehicle, markedly enhancing curcumin’s antimicrobial performance. This study provides a promising strategy for developing stable, high-efficiency curcumin delivery systems with improved therapeutic potential, particularly for oral/throat protection against alcohol-induced irritation via on-site curcumin release to alleviate local inflammatory damage.

The hydrophobicity and poor oral bioavailability of curcumin limit its application in the food industry. In this study, LP-CNCs/PNFs complexes were prepared by electrostatic interaction and used as stabilizers to prepare curcumin-loaded Pickering emulsions. The interaction between LP-CNCs and PNFs formed stable interface structures, which improved the emulsifying properties of LP-CNCs and enhanced the storage stability and rheological properties of emulsions. LP-CNCs/PNFs-based Pickering emulsions exhibited excellent protective capability for encapsulating curcumin. The stable and thick interface layer was formed on the surface of oil droplets, which provided a physical barrier against UV radiation and thermal resistance and slowed down the degradation of curcumin induced by the external environment. Moreover, during in vitro digestion, Pickering emulsions demonstrated excellent stability, leading to an improvement in curcumin's bioaccessibility from 37.29 % (C4P0) to 73.99 % (C4P4) after digestion. Consequently, double fibrils-based Pickering emulsions provide the theoretical foundation for improving the bioaccessibility of hydrophobic active substances and the development of healthy foods.
