Phenylboronic acid functionalized magnetic ferroferric oxide nanoparticles and capillary electrophoresis for efficient isolation of flavonoid compounds from leaves of Lonicera japonica Thunb

Yunfeng Yuan,Xiaoge Wang,Jinhua Zhu,Abdelhadi El Jaouhari,Xiuhua Liu,Md. Zaved H. Khan

Flavonoids are bioactive components in natural products, which possess anti-inflammatory, antibacterial, antioxidant, and cardiovascular protective properties. However, due to the complexity and low content of the components in these samples, developing rapid and sensitive methods for the isolation and extraction of flavonoids still remains a challenge in medical and food science. Herein, a 4-formylphenylboronic acid functionalized magnetic Fe 3 O 4 nanomaterial (Fe 3 O 4 @FPBA) was synthesized and applied as a sorbent of magnetic solid-phase extraction (MSPE) to covalently extract flavonoids from leaves of Lonicera japonica Thunb.. The structure, morphology and magnetic properties of Fe 3 O 4 @FPBA particles were characterized by fourier transform infrared spectroscopy (FT-IR), X-ray diffraction (XRD), vibrating sample magnetometer (VSM) and scanning electron microscope (SEM) technologies. Employing luteoloside, luteolin, lonicerin, hyperoside, quercetin and rutin as model compounds, Fe 3 O 4 @FPBA as sorbent, a MSPE coupling with capillary electrophoresis (CE) method was developed and optimized to detect the flavonoids. Adsorption kinetics display that the adsorption of flavonoids by Fe 3 O 4 @FPBA is in line with the Quasi-second-order model, which is controlled by chemisorption mechanism, with the equilibrium adsorption capacity ranging from 3.66 to 6.16 mg/g. The isothermal adsorption model shows that the adsorption is more consistent with Freundlich isotherm equation, and the exponent n is around 1. In addition, the material was applied to the leaves of Lonicera japonica Thunb. extract. Four kinds of flavonoids and three other o-hydroxyl compounds were covalently extracted and magnetically separated. Moreover, the material can still maintain high adsorption properties after recycling 5 times. The material possesses strong magnetism and boric acid ligands, which can realize rapid and high-capacity separation and enrichment of flavonoids in liquid samples. Therefore, the strategy offers an innovative method for the extraction and purification of flavonoids from complex natural plants and also provides a research basis for the discovery of new medicinal compounds based on natural products.

Size-structured flavonoid distribution in Salix babylonica branches modulates Anoplophora glabripennis feeding behavior: A novel mechanistic target for eco-friendly antifeedant development

Zepeng Yang,Huangling Wang,Chen Huang,Cui Wang,Zimeng Geng,Fei Lyu

Asian longhorned beetle ( Anoplophora glabripennis ) has caused significant economic and ecological damage. However, environmentally friendly control strategies remain limited. Elucidating the adult's feeding preference is critical for developing effective and sustainable pest management approaches. Therefore, we firstly compared the feeding preference of adults on the Salix babylonica branch with diameters of 0.5-cm, 2-cm, 4-cm, 8-cm, 16-cm. The results showed that both female and male significantly preferred 2-cm branches over the others. Subsequently, we analyzed metabolites allocation patterns across these branch types, identifying a total of 2216 metabolites. Further analyses showed that 1500 differentially accumulated metabolites (DAMs) in branches with diameters of 0.5-cm, 4-cm, 8-cm, 16-cm compared to 2-cm samples. These DAMs were significantly enriched in flavonoid, flavone and flavonol biosynthesis pathways when comparing the 2-cm branches to others. The contents of 10 DAMs in flavonoid, flavone and flavonol biosynthesis pathways showed a significant negative correlation with feeding consumptions. To evaluate the antifeeding effects of flavonoid-related metabolites, four economically viable and practically flavonoid-related metabolites (+)-taxifolin, naringenin, kaempferol, and phlorizin were selected for feeding bioassays. They significantly inhibited feeding consumption, with (+)-taxifolin, naringenin, and phlorizin reducing by up to 70 %. Dose-response experiments confirmed varying concentrations of these compounds significantly suppressed feeding consumption. Our findings demonstrate that the allocation patterns of flavonoid compounds in branches of different diameters regulate the feeding preferences of A. glabripennis . This study explores a metabolomics-driven approach that centers on size-structured flavonoid reprogramming in S. babylonica branches, aiming to establish a sustainable strategy for disrupting A. glabripennis 's feeding preference.

Rutin, a natural flavonoid glycoside, ameliorates zearalenone induced liver inflammation via inhibiting lipopolysaccharide gut leakage and NF-κB signaling pathway in mice

Yanan Wang,Qianqian Wang,Gaigai Wang,Qiongqiong Zhang,Yongpeng Guo,Xin Su,Yu Tang,Matthew Koci,Jianyun Zhang,Qiugang Ma,Lihong Zhao

Zearalenone (ZEN) poses a potential threat on human and animal health partly through the nuclear factor (NF)-κB signaling pathway. In silico study suggested that rutin effective against TLR4 and NF-κB. A wetting test was designed to evaluate the effect and underlying mechanism of rutin in alleviating ZEN-induced inflammation in animals. Twenty-four female mice were randomly divided into 4 groups: control (basal diet), ZEN group (basal diet + ZEN), rutin group (basic diet + rutin), Z + R group (basal diet + rutin + ZEN). Results showed that rutin effectively alleviated ZEN-induced inflammation and damage of liver and jejunum in mice. Rutin addition reduced the content of lipopolysaccharide (LPS) in serum and liver mainly by improving the intestinal barrier function resulted from the production increase of short-chain fatty acids (SCFA). In sum, this study showed that rutin alleviated ZEN-induced liver inflammation and injury by modulating the gut microbiota, increasing the production of SCFA and improving intestinal barrier function, leading to the decrease of LPS in liver and the inhibition of MyD88 independent NF-κB signaling pathway in mice. Specifically, these findings may provide useful insights into the screening of functional natural compounds and its action mechanism to alleviate ZEN induced liver inflammation.

The p53-inducible gene 3 involved in flavonoid-induced cytotoxicity through the reactive oxygen species-mediated mitochondrial apoptotic pathway in human hepatoma cells

Qiang Zhang,Guangdong Cheng,Hongbin Qiu,Liling Zhu,Zhongjuan Ren,Wei Zhao,Tao Zhang,Lei Liu

Flavonoids have been reported to exhibit prooxidant cytotoxicity against cancer cells, but the underlying mechanism is still poorly understood. Here we investigated the potential mechanism that p53-inducible gene 3 (PIG3), a NADPH:quinone oxidoreductase, mediated the prooxidant cytotoxicity of flavonoids on human hepatoma HepG2 cells. The results showed that flavonoids (apigenin, luteolin, kaempferol, and quercetin) inhibited the growth of HepG2 cells in a dosage- and time-dependent manner, and induced the morphological changes characteristic of apoptosis in HepG2 cells. We also found that expression of PIG3 was increased markedly in HepG2 cells treated with flavonoids at both mRNA and protein levels, which was accompanied by increased intracellular ROS production and a decreased mitochondrial membrane potential (ΔΨm). All these effects were largely reversed through knockdown of the PIG3 gene in HepG2 cells. Western blotting indicated that flavonoids increased cytochrome c release, upregulated the ratio of Bax/Bcl-2, and activated the caspases-9 and -3. Moreover, knockdown of PIG3 could reverse the changes of these apoptotic-related proteins. These results suggest that PIG3 plays an important role in regulating the prooxidant activity and apoptosis-inducing action of flavonoids on HepG2 cells though the ROS-triggered mitochondrial apoptotic pathway.

Structural and digestive characteristics of high amylose maize starch-flavonoid complexes: Role played by the flavonoids in enhancing resistant starch and inhibiting enzymes

Jiaqi Shang,Zhenyuan Wang,Wenfeng Lu,Mi Wang,Jiaheng Zhang

Diabetes and obesity are global health challenges, regulating starch digestion offers a promising strategy. High amylose maize starch (HAMS)-flavonoid complexes were synthesized using five classes of flavonoids with distinct conformations. The structures and digestibility of complexes were characterized to elucidate the interaction mechanisms between flavonoids and HAMS. All complexes exhibited complexing indices exceeding 43%, with the HAMS-catechin complex (HAMS-CAT) achieving the highest index of 67.6%. The degree of short-range molecular order, helical structures, and crystallinity of complexes were significantly influenced by the flavonoids. Notably, the HAMS-CAT exhibited the highest resistant starch content at 51.8%, which was 24.4% higher than that of the control sample. Enzyme inhibition assays revealed that CAT inhibited α-amylase by 77.9%, while naringenin inhibited α-glucosidase by 90.3% at 10 mg/mL. The interaction between flavonoids and HAMS impeded the complexes contact with enzymes, and the released flavonoids inhibited the activities of enzymes to further reduced digestibility. This interaction also protected the antioxidant activity of flavonoids during heating. Among the five flavonoids, CAT exhibits a flexible conformation with five hydroxyl groups and lacks carbonyl group on the C4 chain. Consequently, CAT interacted with HAMS effectively by forming multidentate hydrogen bonds and enhanced the degree of short-range molecular order and crystallinity of HAMS-CAT. This work systematically compares the potential of flavonoids with various conformations in modulating starch structure and digestibility. These findings provide a feasible idea for the application of flavonoids in food ingredients to reduce starch digestibility and offer theoretical support for the industrialization of starch-based healthy foods.

Bioaccessibility and Absorption of Flavonoid C-glycosides from Abrus mollis Using Simulated Digestion, Caco-2 Cell, and In Situ Single-pass Perfusion Models

Wei Shen,Xiaolong Hu,Yajun Niu,Yimeng Lu,Baolin Wang,Hao Wang

Abrus mollis is commonly used as a traditional Chinese medicine for the treatment of liver diseases due to its hepatoprotection and anti-inflammation, but the absorption properties of its main bioactive ingredients remain unclear. Our previous studies verified that the flavonoid C-glycosides, including vicenin-2 (1), isoschaftoside (2), and schaftoside (3), were the major active components in A. mollis for hepatic protection against nonalcoholic fatty liver disease, hepatitis, and hepatic fibrosis. This study investigated the bioaccessibility and transport mechanisms of total flavonoid C-glycoside, as well as vicenin-2 (1), isoschaftoside (2), and schaftoside (3), in A. mollis by simulated digestion and use of the Caco-2 cell model. Moreover, this study attempted to verify their absorption properties by in situ gastrointestinal perfusion in rats. Total flavonoid C-glycoside and 1, 2, and 3 exhibited similar bioaccessibility of 84.58%, 85.13%, 83.05%, and 81.65% respectively after simulated digestion. The transport of total flavonoid C-glycoside in the Caco-2 cell model increased with the concentration, and the transport showed saturation characteristics with the time and concentration of total flavonoid C-glycoside to a certain degree. The Papp values of total flavonoid C-glycoside and the 3 flavonoid C-glycosides were significantly improved by verapamil, probenecid, and EDTA-Na2. Their absorption properties in the gastrointestinal tract were consistent with that found in Caco-2 cells, and superior absorption rates were observed in the duodenum and jejunum. The absorption pattern of total flavonoid C-glycoside may involve multiple transport pathways, including active transport, passive diffusion, and the paracellular pathway. TFC was actively pumped out by P-glycoprotein and multidrug resistance-associated protein. These results revealed that the bioaccessibility and intestinal absorption characteristic of total flavonoid C-glycoside were consistent with the 3 major flavonoids.

Characterization of a Flavonoid 3’/5’/7-O-Methyltransferase from Citrus reticulata and Evaluation of the In Vitro Cytotoxicity of Its Methylated Products

Xiaojuan Liu,Yue Wang,Yezhi Chen,Shuting Xu,Qin Gong,Chenning Zhao,Jinping Cao,Chongde Sun

O-methylation of flavonoids is an important modification reaction that occurs in plants.O-methylation contributes to the structural diversity of flavonoids, which have several biological and pharmacological functions. In this study, anO-methyltransferase gene (CrOMT2) was isolated from the fruit peel ofCitrus reticulata, which encoding a multifunctionalO-methyltransferase and could effectively catalyze the methylation of 3’-, 5’-, and 7-OH of flavonoids with vicinal hydroxyl substitutions. Substrate preference assays indicated that this recombinant enzyme favored polymethoxylated flavones (PMF)-type substrates in vitro, thereby providing biochemical evidence for the potential role of the enzyme in plants. Additionally, the cytotoxicity of the methylated products from the enzymatic catalytic reaction was evaluated in vitro using human gastric cell lines SGC-7901 and BGC-823. The results showed that the in vitro cytotoxicity of the flavonoids with the unsaturated C2-C3 bond was increased after being methylated at position 3’. These combined results provide biochemical insight regarding CrOMT2 in vitro and indicate the in vitro cytotoxicity of the products methylated by its catalytic reaction.Keywords:citrus;flavonoids;O-methyltransferase;bioactivity;cytotoxicity

The flavonoid-rich ethanolic extract from the green cocoon shell of silkworm has excellent antioxidation, glucosidase inhibition, and cell protective effects in vitro

Hai-Yan Wang,Jin-Ge Zhao,Yu-Qing Zhang

The green cocoon shell of a novel variety of silkworm, Bombyx mori, is rich in two types of quercetin and kaempferol flavonoids. The aim of this study was to identify these flavonoids in the ethanolic extract (EE) from green cocoons and develop EE applications in healthy foods. The experimental results indicated that the amount of total amino acids in EE was 27.06%. The flavonoids in EE are presented in quercetin and kaempferol glycosides. The total amount of the two aglycones was 33.42 ± 0.08 mg/g. The IC 50 values of 2,2-diphenyl-1-picrylhydrazyl (DPPH), 1,2’-azino-bis (3-ethylbenzthiazoline-6-sulphonicacid) (ABTS), and hydroxyl radical scavenging abilities were 296.95 ± 13.24 μg/mL, 94.31 ± 9.13 μg/mL, and 9.21 ± 0.15 mg/mL, respectively. The IC 50 values of the inhibitory activities of α-amylase and α-glucosidase were 37.57 ± 6.45 μg/mL and 212.69 ± 22.94 μg/mL, respectively. EE also reduced the level of reactive oxygen species (ROS) and oxidative stress in L02 cells induced by high glucose levels. It also effectively decreased the content of 8-hydroxyl deoxyguanosine (8-OHdG), nuclear factor κB (NF-κB), and tumour necrosis factor alpha (TNF-α) in cells with a good dose effect. These results clearly indicated that the flavonoid-rich EE with excellent antioxidant and glucosidase inhibition abilities significantly reduced the damage to cells caused by oxidative stress and inflammatory reactions. It is suggested that EE might serve as useful functional foods for the treatment of related diseases induced by oxidative stress such as diabetes mellitus.

Constructing natural flavonoid based hyper crosslinked polymer combined to HPLC/MS for accurate determination of phenolic endocrine disruptors in chicken and fish meat

Ziyi Yin,Yanli Dong,Zhi Li,Qianqian Wang,Qiuhua Wu

Development of monitoring methods for phenolic endocrine disrupting chemicals (EDCs) like bisphenol A is imperative, as they disrupt homeostasis of the body's endocrine system, posing serious threats to human health. Herein, a natural flavonoid based hyper cross-linked polymer (STF-HCP1) was synthesized for the first time. STF-HCP1 achieves excellent adsorption of phenolic EDCs (bisphenol A, p -tert-butylphenol, bisphenol B, bisphenol F) through hydrogen bonding and π–π interactions. STF-HCP1 based solid phase extraction was coupled with liquid chromatography-mass spectrometry for sensitive quantification of trace phenolic EDCs in chicken and fish samples. Under optimum conditions, this method showed lower limit of detection (0.58–2.1 ng g −1 for chickens and 0.70–3.2 ng g −1 for fish), satisfactory spiked recovery (91.1–106 %) and high precision (relative standard deviation ≤6.7 %). Beyond offering a viable method for sensitive detection of phenolic EDCs, this study highlights a novel strategy to prepare effective adsorbents using natural raw materials.

Lipidomic Analysis Reveals the Anti-Obesity and Hepatoprotective Effects of Flavonoid Mimetic Components in Adzuki Beans on High-Fat Diet-Induced Obese Mice

Jiayu Zhang,Xiujie Jiang,Qingpeng Xu,Weidong Li,Dongjie Zhang

Obesity and overweight have increasingly posed a serious challenge to public health security. This study systematically evaluated the reversal and regulatory effects of a composite flavonoid component mimicking the composition of adzuki bean flavonoids on high-fat diet (HFD)-induced obesity, related lipid metabolism disorders, and impaired liver function, based on lipid metabolomics and an HFD-induced obese mouse model. The results demonstrated that sustained HFD intake led to significant weight gain, increased adiposity index, dyslipidemia, and altered brown adipose tissue (BAT) cell status in mice, while also exerting adverse effects on hepatic lipid deposition and the lipid metabolic profile associated with liver fibrosis. Intervention with an adzuki bean flavonoid mimic (ABFM) effectively prevented further weight gain and ameliorated abnormal expression of serum lipid and liver function-related indicators. Furthermore, we found that ABFM alleviated HFD-induced liver damage and mitigated the whitening tendency of brown adipose tissue. Lipidomics analysis revealed that ABFM intake significantly improved abnormal hepatic lipid metabolic profiles, notably downregulating the expression levels of diacylglycerol (DG) and phosphatidylglycerol (PG), while markedly ameliorating sphingolipid metabolism disorders and ceramide (Cer) levels, which are highly associated with liver fibrosis. These findings further elucidate the mechanisms by which adzuki bean flavonoid components improve diet-induced obesity and associated liver injury, providing a theoretical basis for exploring safe and effective dietary intervention strategies based on plant flavonoids.

Characterization of a Thermophilic and Acidophilic GH78 α-L-Rhamnosidase from Thermotoga sp. 2812B Capable of Efficiently Hydrolyzing a Variety of Natural Flavonoid Diglycosides

Bin-Chun Li,Weijuan Dong,Bingbing Wu,Yanlong Liu,Na Han,Guo-Bin Ding

α-L-Rhamnosidase can specifically hydrolyze plant natural glycosides and holds significant potential for biocatalytic applications in functional foods, healthy products, and pharmaceutical industries. Herein, a novel thermophilic and acidophilic α-L-rhamnosidase TsRha fromThermotogasp. 2812B belonging to glycoside hydrolase family 78 was identified by genome mining and comprehensively characterized by bioinformatics, computer-aided structural analysis, and biochemical characterization. TsRha possesses a domain architecture comprising one catalytic (α/α)6-barrel domain and four β-sheet domains. TsRha displayed optimal activity at 90 °C and pH 5.0, remarkable thermostability at 80 °C, and considerable tolerance to organic solvents. TsRha exhibited broad substrate selectivity and might efficiently hydrolyze a series of natural flavonoid glycosides with various glycosidic bonds (α-1, α-1, 2, α-1, 6) from different aglycone subgroups (flavanone, flavone, flavonol, and dihydrochalcone). Moreover, it demonstrated high conversion efficiencies toward a variety of natural flavonoid diglycosides rutin, naringin, naringin dihydrochalcone, hesperidin, and troxerutin, achieving ≥99.1% conversion within 20~100 min. The excellent properties including high activity, thermophilicity, acidophilicity, good thermostability, broad substrate spectrum will make the α-L-rhamnosidase TsRha a promising biocatalyst for the efficient production of rare and high-value flavonoid glucosides with improved bioavailability and bioactivity.
