In this study, spherical shaped chiral mesoporous silica nanoparticles (CMS) was biomimetic synthesized using histidine derivatives (C 16 -L-histidine) as template via the sol–gel reaction and employed as poorly water-soluble drug nimodipine (NMP) carrier. Characteristics of CMS and its application as drug carrier were intensively investigated and compared with MCM41. Then NMP was respectively loaded into CMS and MCM41 with the drug: carrier weight ratio of 2:1. Structural features of NMP before and after drug loading were systemically characterized. The results demonstrated that hydrogen bonds were formed between NMP and carriers during the drug loading process. After drug loading, crystalline state of NMP effectively converted into modification L and amorphous state, and the first form turned out to be easily removed by washing. On the other hand, drug dissolution rate was significantly improved after drug loading, and the best result came from NMP-C3 sample. It was able to release 17.83% of drug within 60 min, which was 6.8-fold higher than the release amount of pure NMP. Undoubtedly, NMP-C3 presented the highest relative bioavailability (386.22%), and the best therapeutic effect. Meanwhile, CMS improved the brain distribution of NMP in vivo.

Zeolitic imidazole framework-8 (ZIF-8) has been demonstrated significant potential as a support material for the fabrication of separation membranes for water treatment applications. However, the separation performance of a ZIF-8-containing membrane is limited by the poor surface and molecular sieving properties of ZIF-8. Herein, a continuous mixed-linker [email protected] ( [email protected] ) layer was synthesized on the amyloid nanofibrils (ANFs)-modified hydrolyzed polyacrylonitrile (HPAN) membrane through a combination of self-assembly and in situ growth methods. The mixed-linker approach facilitated the tuning of surface and molecular sieving properties of ZIF-8 on the membranes, which effectively improved the overall separation and antifouling performance of the [email protected] /ANFs membrane. Compared with the ZIF-8/ANFs membrane, the resultant [email protected] /ANFs membrane exhibits higher water flux (42.6 vs 27.9 L/m 2 ·h·bar) and superior heavy metal ion (Cu 2+ , Ni 2+ , Pb 2+ , and Cd 2+ ) rejection (>98.6 vs 91.2%). Moreover, the [email protected] /ANF membrane also exhibited a higher antibacterial efficiency of 82.9% against Escherichia coli than ANFs/HPAN membrane (16.7%). Therefore, through the mixed-linker approach, considerable modifications of the properties of ZIF-8 were realized, making it a suitable material for the preparation of membranes for wastewater treatment applications.

In this work, carboxylated multi walled carbon nanotubes (CMWCNTs) were firstly prepared and functionalized with poly- L -histidine (PLH), which were then chelated with copper (II) ions to from the nanocomposites of Cu(II)-PLH-CMWCNTs. The nanocomposites could be exploited as an efficient mimic enzyme for sensitive electrochemical detection of salvianic acid A (SAA). Cu(II)-PLH-CMWCNTs owned good charge transfer property and excellent synergetic catalytic effect between the overoxidized imidazole groups and the copper redox-active units. Therefore, highly sensitive electrochemical response to SAA was achieved under optimum experimental conditions. A good linear relationship between differential pulse voltammetry (DPV) peak current and the SAA concentration was established in the range of 0.4–1000 μM. A low detection limit of 0.037 μM and a sensitivity of 0.27 μA μM −1 cm −2 were achieved. The developed biosensor also had advantages of good repeatability, stability and high selectivity, thus, it was successfully applied to the determination of SAA in real samples with satisfactory results, which may have great potential for further exploitation of electroanalysis applications.

Urocanic acid, like histamine, is a degradation product of histidine that may accumulate over the storage of histidine-rich pelagic fish. Recently evidence from the dermatological and related fields has directed the attention of food scientists toward the adverse health effects of urocanic acid, mostly as a possible “missing factor” responsible for scombrotoxicosis besides histamine. However, experimental effort to study the hypothesis remains scarce, especially regarding the formation of urocanic acid in comparison to the well-documented histamine. Here we report a mixed-mode HPLC method for simultaneously analyzing urocanic acid (both the trans - and cis -isomers), histamine, and histidine in chub mackerel ( Scomber japonicus ) fillets and the use of the method to monitor their changes in concentration at typical refrigeration and abuse temperatures. The method features a simple, reliable chromatographic analysis in cation-exchange/reversed-phase/hydrophilic-interaction triple mode of retention, without the need of precolumn derivatization or addition of ion-pairing agents. At both 4 and 20 °C slow but consistent formation of trans -urocanic acid was observed over time with a relatively faster rate at 20 °C. Histamine formation, by contrast, occurred at 20 °C but ceased completely at 4 °C. In addition, we demonstrate the occurrence of trans -to- cis isomerization of UCA in chub mackerel fillets upon direct sunlight exposure: under our conditions, an isomerization yield around 13 % was achieved within hours on a sunny winter day. Overall, this study provides a chemical analysis method and relevant data for exploring the possible role of urocanic acid in the aetiology of scombrotoxicosis.

In this study, we developed a multiphase catalyst, BNNSs/His-Pd, by anchoring palladium nanoparticles onto L-histidine-modified boron nitride nanosheets. BNNSs/His-Pd was shown to be a good 2D heterogeneous catalyst. The enhanced affinity of the modified BNNSs for metal complexes, coupled with their inherent stability, contributes to the catalytic activity of the BNNSs/His-Pd-catalyzed Suzuki coupling reaction. The catalyst showed a broad substrate scope, good catalytic activity, and the convenience of not requiring inert gas protection in the Suzuki–Miyaura cross-coupling reaction. Even after 10 recycling cycles, the product yield remained at 95%, highlighting its significant potential for practical applications. Graphical Stable and efficient BNNSs/His-Pd heterogeneous catalysts were successfully constructed by modifying boron nitride nanosheets with L-histidine. The effect of L-histidine enhanced the dispersion and stability of Pd NPs on BNNSs carriers, combined with the inherent stability of BNNSs which resulted in the improvement of catalytic activity and recycling performance of the catalysts in the Suzuki coupling reaction.

BACKGROUND The Maillard reaction involves the interaction of various amino acids and reducing sugars, resulting in food browning. It often produces appealing aromas and flavors. The complexities of the reaction are such that it can be challenging to identify the often numerous and frequently volatile products formed by it. In the present study, we sought to identify and evaluate an unusual product with anti-oxidant activity arising from a fructose-histidine Maillard reaction model. The anti-oxidant profile of this product was assessed by computational means. RESULTS The fructose-histidine Maillard reaction products (FH-MRPs) were generated by heating a 2:1 mixture of the sugar and the amino acid at 140 °C for 2 h. Chromatographically separable fractions, labelled DM-1 to DM-8, were obtained using silica gel as the stationary phase and dichloromethane/methanol (DCM/MeOH) mixtures as the mobile one. Fraction DM-5 exhibited the highest 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, and further bio-assay guided fractionation led to isolation and identification of 1-(1 H -imidazo[4,5- c ]pyridin-4-yl)ethenone (IMPE) as the active principal, the structure of which was established by nuclear magnetic resonance (NMR) spectroscopic and mass spectral techniques. A mechanism for the formation of IMPE from its precursors is proposed. Density functional theory (DFT) calculations suggest this novel heterocyclic compound exerts its anti-oxidant effects by interacting with DPPH and 2,2'-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radicals. Essentially, IMPE was non-toxic below 300 ug mL −1 , showing a concentration-dependent free radical clearance capacity and reducing power within the 100–1000 μg mL −1 range, and moreover, exhibiting significant Fe 2+ chelating abilities wihin the 50-200 μg mL −1 range. CONCLUSION This study identified the unique FH-MRP, IMPE, and found that it acts as food antioxidant through the chelation of metal ions. © 2024 Society of Chemical Industry.

The detection of Cu 2+ ions and L-histidine (L-His), which are crucial trace elements and amino acids in biological systems, is of paramount importance for environmental health, food safety, and clinical diagnosis. However, conventional detection methods often suffer from drawbacks such as complexity, time-consuming procedures, and high costs. To address these challenges, we have developed an efficient, sensitive, and straightforward detection techniques. In this study, we successfully synthesized red-emitting carbon dots (R-CDs) with a remarkable quantum yield of 26.6 % and devised an R-CDs-based fluorescence sensor that operates via an “on-off-on” mode for the simultaneous detection of Cu 2+ ions and L-His. The sensor's working principle involves the static quenching of R-CDs fluorescence upon the addition of Cu 2+ ions, followed by the restoration of fluorescence through the competitive binding of L-His to Cu 2+ ions. This approach enables the ultrasensitive detection of Cu 2+ ions and L-His, with detection limits as low as 0.58 μM and 0.45 μM, respectively. The sensor has been effectively applied to detect Cu 2+ ions in environmental water samples and L-His in human blood, food additives, and health-care products. It demonstrates excellent selectivity, strong anti-interference ability, and high accuracy, with recovery rates ranging from 95.0 % to 105.0 %. Owing to its simplicity, rapidity, cost-effectiveness, and high sensitivity, the proposed R-CDs-based fluorescence sensor holds great promise as a valuable tool for environmental monitoring, food safety assessment, and clinical diagnosis. Future efforts will focus on further optimizing the sensor's performance and exploring its applications in a broader range of real-world samples, thereby providing substantial support for scientific research and technological advancements in related fields.

L -histidine ionic liquid modified magnetic COF was successfully fabricated. • Covalent modification of supports with IL optimized the enzyme microenvironment. • Introduction of IL enhanced the activity and stability of immobilized lipase. • IL boosted lipase-substrate affinity and catalytic efficiency. • Magnetic responsiveness improved the separation efficiency of immobilized lipase. This study proposes an innovative immobilized lipase construction strategy to address the low activity and poor stability limitations of current biocatalysts for flavor ester synthesis. L -histidine-based ionic liquid (IL) is covalently grafted onto the surface of magnetic covalent organic framework (COF) via Schiff base reaction, and porcine pancreatic lipase (PPL) is further covalently immobilized on the composite carrier through amide bond coupling. The resulting Fe₃O₄@COF-IL-PPL shows 1.6-fold higher catalytic activity and 2.3-fold better thermal stability than free PPL, and 32% higher activity retention after cycles than PPL immobilized on IL-free COF carrier. Its optimal reaction pH and temperature are 7.5 and 50 °C respectively, maintaining 86% of initial activity at 60 °C and 76% activity after 6 h incubation at 50 °C. Under n-hexane reaction system, the isoamyl acetate yield reaches 87.4% under optimal conditions, and the biocatalyst retains 71.4% activity after 10 consecutive isoamyl acetate synthesis cycles. Although the additional preparation steps and higher cost of the support need to be considered for industrial applications, the improved catalytic performance of Fe 3 O 4 @COF-IL-PPL verified the effectiveness of IL surface modification strategy, and the system can be further extended to solvent-free systems to verify its broader applicability.

Construction of magnetic nanotubes and transition metal ion generated by acid etching form its own surface (defined as “self-released” systems) can effectively address the issues of complex operation and poor adsorption performance associated with traditional immobilized metal affinity chromatography (IMAC) materials. Based on the magnetic Ni core and polydopamine (PDA) shell, a core-shell structured Ni@PDA magnetic composite nanotubes and thier acid-etched product (defined as Ni@PDA-T) were developed and synergistically provided excellent magnetic responsiveness, low cell toxixity and more active sites, enabling efficient magnetic separation and surface Ni 2+ self-release. Surface self-released Ni 2+ enabled coordinate capture of histidine-rich proteins without requiring exogenous metal ion supplementation after fixed onto PDA with pH 8.0, simplifying operation and enhancing adsorption efficiency. Given the differences in histidine content among Bovine hemoglobin (BHb), Bovine serum albumin (BSA), and Lysozyme (LYZ), Ni@PDA-T exhibited a maximum adsorption capacity of 4556 mg/g for BHb, which is rich in histidine residues. Density functional theory (DFT) calculations indicate that p-d orbital coordination interactions dominate the His protein's immobilization onto the Ni site and result in superior adsorption behavior for the His protein by DOPAm-Ni 2+ functional group. SDS-PAGE analysis of mixed protein systems and actual bovine blood samples further confirmed the selective adsorption of BHb by Ni@PDA-T. Chelator-free blood purification was achieved through synergistic coordination between self-released Ni 2+ and histidine residues, overcoming the reliance on traditional chelating agents.

Histamine food poisoning (HFP) or scombrotoxic fish poisoning is a major public health concern associated with seafood and seafood products containing copious amounts of free histidine , which may be decarboxylated into scombrotoxic histamine, the main culprit of HFP. Moreover, the deamination product of the histidine, trans -urocanic acid, is assumed to be scombrotoxic by degranulating mast cells to release endogenous histamine after isomerizing to the cis -form, as well as known to have immunosuppressive effects. In recognition of the potential health risks associated with the histidine degradatives, this work aimed to provide a method for the simultaneous determination of histidine, histamine, and urocanic acid isomers in food, based on precolumn dansylation and subsequent reversed-phase HPLC analysis. The dansylation conditions were optimized for yield with the design of experiments approach consisted of a center point-enhanced 2 4 factorial design followed by sequential steepest ascent searches. Satisfactory chromatographic separation of fish sauce was achieved within 9 min by retention modeling of both the analytes and potentially interfering peaks. The method was fully validated with limits of detection between 0.004 and 0.017 mg 100 g −1 fish sauce, and successfully applied to several fish sauce samples, in which the histamine and urocanic acid levels were found low and unlikely of concern, with correspondingly low levels of the precursor histidine.

Nanozyme-based detection methods have gained increasing attention in the field of food safety. However, the effect of the food matrix on the nanozyme activity requires tedious sample pre-treatment for nanozyme-based detection. Therefore, a difunctional histidine-modified magnetic hybrid nanozymes (His-Fe 3 O 4 @Cu) was used as capture probes and signal amplifiers to develop a simple colorimetric aptasensor for Salmonella Typhimurium detection. The prepared His-Fe 3 O 4 @Cu nanozymes demonstrated a 5.0-fold increase in peroxidase-like activity compared to Fe 3 O 4 nanozymes and exhibited superparamagnetism with saturation magnetization values of 63.5 emu/g. By virtue of the strong superparamagnetism of the His-Fe 3 O 4 @Cu nanozymes, aptamer-conjugated His-Fe 3 O 4 @Cu nanozymes first enriched and separated S. Typhimurium from food matrix. Then, aptamer-conjugated His-Fe 3 O 4 @Cu nanozymes employed an etching solution to generate a detection signal via the peroxidase-like activity. In optimal conditions, this proposed colorimetric aptasensor successfully detected S. Typhimurium with a limit of detection (LOD) of 8 CFU/mL in a linear range of 1.2 × 10 1 to 1.2 × 10 6 CFU/mL. Finally, the proposed aptasensor was further validated in three actual samples, with a capture efficiency ranging from 88.6 % to 96.5 % and recoveries between 88.2 % and 104 %. These results revealed the significant potential of this method for S. Typhimurium detection in food samples.
