Nonspecific protein adsorption-resistant materials, the so-called nonfouling materials, are crucial biomaterials in biomedical applications. Up-to-date, little attention was paid to the biodegradability of these materials. In this work, nonfouling zwitterionic copolymerized peptides composed of the N-l-glumatyl-l-lysine dimer (EK) and δ-l-lysinyl-l-glutamic acid dimer (E-K, glutamic acid with the lysine side chain) at various ratios were synthesized to investigate the enzymatic degradation rate. Two types of proteases (trypsin and alkaline protease), which represent a site-specific and less site-specific cleavage protease, respectively, were used to demonstrate the adjustable degradability by tracking the molecular weight (Mw) at different digestion times. Results showed that higher compositions of the E-K dimer lead to slower degradation rates by both proteases and larger fragments after 120 min digestion. With the composition of the E-K dimer over 50%, the degradation of copolymerized peptides by both proteases becomes very slow. This indicated that the bulky lysinyl side chain on E-K can alter the enzymolysis process for adjusting the enzymatic degradability of the newly synthesized zwitterionic copolymerized peptides, which could be promising candidates for biomedical applications in vivo.

The supplementation of dietary limiting amino acids (AA) with crystalline AA makes the use of low-protein diets an option in poultry production. The differing absorption rates of crystalline and protein-bound AA may lead to temporally imbalanced AA in the postabsorptive period. In this study, two experiments were conducted to evaluate the effect of encapsulated L-lysine-HCl (L-Lys-HCl) and DL-methionine (DL-Met) on the laying performance of hens. In exp. 1, a total of 135 forty-seven-wk-old Hy-Line Brown hens were subjected to three dietary treatments for 8 wk: basal diet supplemented with 0.14% L-Lys-HCl and 0.17% DL-Met to satisfy the NRC (1994) total Lys and Met recommendation (control) and basal diet supplemented with encapsulated L-Lys-HCl and DL-Met at the levels of 60% (60CLM, 0.084% L-Lys-HCl and 0.102% DL-Met) or 80% of control (80CLM, 0.112% L-Lys-HCl and 0.136% DL-Met), respectively. In exp. 2, 24 fifty-five-wk-old Hy-Line Brown hens were individually reared in cages and subjected to the same treatments as in exp. 1. The plasma concentrations of free AA and nitrogen metabolites were measured 2, 4, and 6 h after fed. The results showed that dietary AA treatment had no significant influence on body weight (BW), feed intake, laying rate, egg weight, egg mass, or feed efficiency. The expression levels of AA transporters CAT-1, y+LAT1, b0,+AT, B0AT, rBAT, EAAT3, and PepT1 in the duodenum, jejunum, and ileum were not influenced (P > 0.05) by dietary treatment. There was an interaction of dietary AA treatment and time (P < 0.05) and the 80CLM hens exhibited higher concentrations of Lys (P < 0.05) than the controls at 2-h time point. In contrast, plasma Met concentration was not influenced (P > 0.05), while Cys was reduced in the 60CLM hens at every time point. The 80CLM hens had higher taurine concentrations than those receiving the control diet at every postprandial time point. In conclusion, these findings demonstrate that by using encapsulated form, the supplemental levels of synthetic L-Lys-HCl and DL-Met can be effectively reduced by approximately 20% with no negative effect on laying performance. The result suggests that encapsulated Lys and Met may ameliorate the postabsorptive AA balance and contribute to the reduced dietary AA supplemental levels.

Nitrogen-doped ordered mesoporous carbons (NOMCs) were synthesized by single-step hydrothermal self-assembly using F127 as a soft template, hexamine as a formaldehyde source, L-lysine as a polymerization catalyst, and 3-aminophenol as both carbon and nitrogen sources. The microstructure of the NOMCs was characterized by XRD, N2 adsorption/desorption, TEM, FTIR, and XPS. The results indicated that the obtained NOMCs exhibited a large specific surface area, uniform pore size distribution and highly ordered 2-D hexagonal mesostructure (P6mm). Besides, the nitrogen was uniformly doped into the carbon framework in the form of various nitrogen species. The adsorption isotherms of CO2 and N2 were also determined and could be well fitted by a DSL model. The capture capacity of CO2 was affected by both the nitrogen content and mesostructure of the adsorbents. Overall, NOMC-L-0.5 displayed excellent CO2 capture capacity (0 °C, 3.32 mmol g−1; 25 °C, 2.50 mmol g−1), and still demonstrated great regenerability with only 2% loss after several CO2 adsorption/desorption cycles. Moreover, the CO2/N2 selectivity calculated by IAST was as high as 43.2 at 25 °C in a typical composition of flue gas (binary mixtures with 15% CO2). The superior adsorption performance enables NOMCs to be a promising CO2 adsorbent in practical applications.

Traditional packaging materials with single function or weak activity cannot meet the requirements of active packaging. In this study, antimicrobial carboxymethyl cellulose (CMC)-based nanocomposites containing montmorillonite (MMT) and ε-poly-( l -lysine) (ε-PL) were prepared by solution casting method. The morphological, optical, mechanical, barrier, thermal, and antimicrobial properties of the resulted ternary nanocomposites were investigated. The surface morphology of the films was dependent on the contents of the ε-PL used. The incorporation of MMT and ε-PL into CMC matrix improved the UV barrier property, tensile strength, hydrophobicity, and water vapor barrier property. When incorporation of 7.5 wt % ε-PL, the tensile strength increased more than threefold and the water vapor permeability decreased by about 35% compared with pure CMC film, the water contact angle reached up to the highest value of 65.38°. The ε-PL incorporated CMC/MMT films exhibited good antimicrobial activities against Gram-positive ( Staphylococus aureus ) and Gram-negative ( Escherichia coli ) bacteria and fungi ( Botrytis cinerea and Rhizopus oligosporus ). Microbial growth inhibition rate of above 90% was obtained when the content of ε-PL got up to 7.5 wt %. In addition, CMC/MMT/ε-PL7.5 wt % film-forming solution having the appropriate properties was used as coating for the strawberries preservation, the shelf life of strawberries was extended for 2 days. © 2019 Wiley Periodicals, Inc. Development of sensitive, selective, and facile electrochemical analytical approaches to monitor tryptophan in different samples is of high significance. For such approaches, efficient electrodes play the key role. Herein, a NiMn-layered double hydroxide (LDH)@poly- l -lysine (PLL) composite has been prepared through in-situ electro-polymerization and further utilized to monitor tryptophan in medicament and biological systems. The as-synthesized NiMn-LDH@PLL composite is highly stable and features a flower-like morphology. It is found that the electron transfer rate of NiMn-LDH@PLL is greatly improved due to the introduction of PLL. The existing Lewis acid—base interaction between LDH and PLL contributes to the increase of unsaturated metal sites and endows this heterojunction composite with high electrocatalytic activity . Especially, the NiMn-LDH (2:1)@PLL composite displays much improved electrochemical response toward tryptophan sensing. This is due to reasonable balance between the outstanding electrocatalytic ability of nickel and the conductivity improvement caused by manganese species. On the other hand, the coordination promotion between NiMn-LDH and PLL is also conducive to improve the sensitivity of electrochemical response. The fabricated sensor exhibits wide linear response in the concentration ranges of 0.1–40 μM and 40–130 μM for tryptophan detection, along with a low detection limit of 52.7 nM. It further displays a high anti-interference ability for tryptophan monitoring even in the presence of other coexistent amino acids and small-sized biological molecules. This study provides a new way to employ LDHs as brilliant sensing platform to ensemble electrochemical sensors for the monitoring of biomolecules.

Amino acid ionic liquids (AAILs) with l -lysine ( l -Lys) as anion were synthesized and applied as new chiral ligands in Zn(II) complexes for chiral ligand-exchange CE. After effective optimization, baseline enantioseparation of seven pairs of dansylated amino acids was achieved with a buffer of 100.0 mM boric acid, 5.0 mM ammonium acetate, 3.0 mM ZnSO 4 , and 6.0 mM [C 6 mim][ l -Lys] at pH 8.2. To validate the unique behavior of AAILs, a comparative study between the performance of Zn(II)- l -Lys and Zn(II)-[C 6 mim][ l -Lys] systems was conducted. In Zn(II)-[C 6 mim][ l -Lys] system, it has been found that the improved chiral resolution could be obtained and the migration times of the three test samples were markedly prolonged. Then the separation mechanism was further discussed. The role of [C 6 mim][ l -Lys] indicated clearly that the synthesized AAILs could be used as chiral ligands and would have potential utilization in separation science in future.

Nonfouling polypeptides with homogenous alternating charges draw peoples’ attentions for their potential capability in biodegradation. Homogenous glutamic acid (E) and lysine (K) polypeptides were proposed and synthesized before. In this work, a new polypeptide formed by poly(glutamic acid) with lysine side chains (poly(E)-K) was synthesized by facile EDC·HCl/HOBt chemistry and investigated. Results show that these polypeptides also have good nonspecific protein resistance determined by enzyme-linked immunosorbent assay. The lowest nonspecific adsorption of the model proteins, anti-IgG and fibrinogen (Fg), on the self-assembling monolayers (SAMs) surface of poly(E)-K was only 3.3 ± 1.8 and 4.4 ± 1.6%, respectively, when protein adsorption on tissue culture polystyrene surface was set as 100%. And, the relative nonspecific protein adsorption increases when the polypeptide molecular weight increases due to the repression of low density polymer brushes. Moreover, almost no obvious cytotoxicity and hemolytic activity in vitro were detected. This work suggests that polypeptides with various formats of homogenous balanced charges could achieve excellent nonspecific protein resistance, which might be the intrinsic reason for the coexistence of high concentration serum proteins in blood.

Lysine, a renewable resource from biomass fermentation, was simply converted to its corresponding α-hydroxyl acids and then cyclized to give the pure O-carboxyanhydride (OCA) monomer. Ring-opening polymerization of the resulting monomer was carried out using dimethylaminopyridine (DMAP) as a catalyst in CH2Cl2 at room temperature, and gave well-defined lysine-derived polyesters bearing pendant carbobenzyloxy (Cbz)-protected amino groups with number average molecular weight of up to 45 kg mol−1 in narrow polydispersity. 1H NMR, GPC, and MALDI-TOF MS measurements of the products clearly indicated the controlled/living character of the polymerization. Moreover, amino-functionalized polyesters were readily prepared by the removal of the Cbz protecting group, and the integrity of the polyester backbone was confirmed by 1H NMR. These amino-functionalized polyesters showed a tunable glass transition temperature and exhibited excellent cell compatibility, suggesting their potential to be used as novel materials in biomedical applications.

The development of antibiotic-loaded microneedles has been hindered for years by limited excipient options, restricted drug-loading space, poor microneedle formability, and short-term drug retention. Therefore, this study proposes a dissolving microneedle fabricated from the host-defense peptide ε-poly- l -lysine (EPL) as an antibacterial adjuvant system for delivering antibiotics. EPL serves not only as a major matrix material for the microneedle tips, but also as a broad-spectrum antibacterial agent that facilitates the intracellular accumulation of the antibiotic doxycycline (DOX) by increasing bacterial cell membrane permeability. Furthermore, the formation of physically crosslinked networks of EPL affords microneedle tips with improved formability, good mechanical properties, and amorphous nanoparticles (approximately 7.2 nm) of encapsulated DOX. As a result, a high total loading content of both antimicrobials up to 2319.1 μg/patch is achieved for efficient transdermal drug delivery. In a Pseudomonas aeruginosa -induced deep cutaneous infection model, the EPL microneedles demonstrates potent and long-term effects by synergistically enhancing antibiotic activities and prolonging drug retention in infected lesions, resulting in remarkable therapeutic efficacy with 99.91 % (3.04 log) reduction in skin bacterial burden after a single administration. Overall, our study highlights the distinct advantages of EPL microneedles and their potential in clinical antibacterial practice when loaded with amorphous DOX nanoparticles.

Hydrogen peroxide (H 2 O 2 ) was used to modify a natural polymer, sesbania gum (SG), to prepare oxidized sesbania gum (OSG) with the aim of investigating the physicochemical properties, antimicrobial activity of polyethylene oxide (PEO), OSG, and ε-poly(lysine) (ε-PL) composite fibre membranes and their applications in fresh-cut mango preservation. The PEO/OSG/ε-PL composite fibre membranes were successfully prepared via solution blow spinning (SBS) technology. The results of a series of characterizations revealed that ε-PL was successfully loaded into the fibrous membranes, exhibited good biocompatibility, and ε-PL was better encapsulated, with the membranes. The hydrophilicity, thermal stability, mechanical properties, and flexibility of the fibrous membranes were improved, and the water vapour transmission rate was reduced with the ε-PL content. In vitro bacterial inhibition experiments revealed that PEO/OSG/ε-PL fibre membranes with more than 2 wt% added ε-PL effectively inhibited the growth of E. coli and S. aureus. Fruit preservation experiments showed that PEO/OSG/ε-PL fibre membrane could reduce weight loss, inhibit microbial growth, and delay quality deterioration of fresh-cut mango. In addition, the PEO/OSG/ε-PL fibre membrane showed high biodegradability. These results show that the PEO/OSG/ε-PL composite fibre membrane prepared by SBS has application potential as an active packaging material.

Hydrogels have emerged as promising biomaterials due to their excellent performance; however, their biocompatibility, biodegradability, and absorbability still require improvement to support a broader range of medical applications. This paper presents a new biofunctionalized hydrogel based on in situ crosslinking between maleimide-terminated four-arm-poly(ethylene glycol) (4–arm–PEG–Mal) and poly(ε-lysine) (ε–PL). The PEG/ε–PL hydrogels, named LG–n, were rapidly formed via amine/maleimide reaction by mixing 4–arm–PEG–Mal and ε–PL under physiological conditions. The corresponding dry gels (DLG–n) were obtained through a freeze-drying technique.1H NMR, FT–IR, and SEM were utilized to confirm the structures of 4–arm–PEG–Mal and LG–n (or DLG–n), and the effects of solid content on the physicochemical properties of the hydrogels were investigated. Although high solid content could increase the swelling ratio, all LG–n samples exhibited a low equilibrium swelling ratio of less than 30%. LG–7, which contained moderate solid content, exhibited optimal compression properties characterized by a compressive fracture strength of 45.2 kPa and a deformation of 69.5%. Compression cycle tests revealed that LG–n demonstrated good anti-fatigue performance. In vitro degradation studies confirmed the biodegradability of LG–n, with the degradation rate primarily governing the drug (ceftibuten) release efficiency, leading to a sustained release duration of four weeks. Cytotoxicity tests, cell survival morphology observation, live/dead assays, and hemolysis tests indicated that LG–n exhibited excellent cytocompatibility and low hemolysis rates (<5%). Furthermore, the broad-spectrum antibacterial activity of LG–n was verified by an inhibition zone method. In conclusion, the developed LG–n hydrogels hold promising applications in the medical field, particularly as drug sustained-release carriers and wound dressings.

l-tryptophan (l-Trp) plays an important role in human and animal metabolism, and an imbalance or deficiency can cause some chronic diseases. While d-tryptophan (d-Trp) is mainly present in microorganisms and green plants and is rarely found in animals. In this work, a simple and efficient electrochemical sensing platform for the simultaneous detection of tryptophan enantiomers based on an l-lysine-functionalized nickel–zinc bis(dithiolene) metal–organic framework and ferrocene-modified multiwalled carbon nanotubes (Fc-MWCNTs) is presented. The identification effect of l-Lys-Ni Zn-MOF/Fc-MWCNTs/GCE on tryptophan (Trp) enantiomers was studied by differential pulse voltammetry. The synthesized materials have been successfully applied to the detection and analysis of Trp enantiomers, and the enantiomer selection coefficient (IL/ID) detected by the sensor is 1.866, indicating that l-Lys-Ni Zn-MOF/Fc-MWCNT composites have good application potential.
