Biochemical characterization and immobilization of a novel pectate lyase ErPL2 for efficient preparation of pectin oligosaccharides

Ling Zheng,Zilong Guo,Yinxiao Xu,Benwei Zhu,Zhong Yao

Pectate lyase (ErPL2) from Echinicola rosea JL3085 showed maximal activity at 45 °C and pH 9.0 with 0.6 mM CaCl 2 . The K m and V max values of ErPL2 for polygalacturonic sodium were 2.098 mmol/L and 0.955 mmol/s, respectively. ErPL2 endolytically degraded pectic substances into oligosaccharides with degree of polymerization (DP) 1–5. To improve the thermostability and pH operation range, recombinant ErPL2 was immobilized onto mesoporous titanium oxide particles (MTOPs). MTOPs have abundant hydroxyl groups on the surface, which is a non-toxicity and good biocompatibility carrier. The residual enzyme activity of immobilized ErPL2 at 40 °C increased remarkably from 11% to 91% compared with free enzyme. The operable pH range was extended from 8–9 to 9–11. Surprisingly, the catalytic efficiency of immobilized ErPL2 was about 19 times higher than free enzyme. To our knowledge this is the first attempt of pectate lyase immobilized on MTOPs and it provides a new option for improving the catalytic performance.

Confirmation and understanding the potential emulsifying characterization of persimmon pectin: From structural to diverse rheological aspects

Yangyang Jia,Ibrahim Khalifa,Meizhu Dang,Yajie Zhang,Lin Zhu,Mengyao Zhao,Kaikai Li,Chunmei Li

In the study, the structure properties of persimmon pectin (PP) affecting the interfacial performance were characterized firstly. Then, the interfacial behavior of PP at the oil-water interface was evaluated by interfacial tension and interfacial shear/dilatational rheology. Further, the emulsifying and emulsion stabilizing capability of PP were explored based on structure elucidation and interfacial characterization. Structural elucidation of PP indicated that structural characteristics, including high-methylated (71.93 ± 1.23%), lower Mw (4.789 × 10 4  g/mol), and abundant hydrophobic substances including acetyl-groups (1.21 ± 0.09%), protein (11.54 ± 0.75 mg/g), and ferulic-acid moieties (2.47 ± 0.13 mg/g), contributed to its rapid adsorption to the oil-water interface. Additionally, PP was rich in both homogalacturonan (62.42 ± 1.12%) and rhamnogalacturonan-I (17.75 ± 0.48%) domains, favoring i t adsorbe d at the oil-water interface. Of all tested emulsifier solutions (1.0%–3.0%, w/v), PP exhibited potent tension-lowering capacity. Additionally, a strong viscoelastic layer of PP (1.0%, w/v) was formed at the oil-water interface based on the interfacial shear (interfacial adsorption and dynamic strain sweep) and interfacial dilatational rheology (interfacial viscoelasticity, frequency and amplitude sweep). Notably, PP exhibited promising emulsifying and emulsion-stabilizing potential, as reflected by small droplet size, creaming thickness (0.68 ± 0.00 mm), and high emulsifying capacity (63.22 ± 1.58%), turbidity (0.37 ± 0.00), etc. By using multi-techniques and multi-scale analysis, the present work effectively correlated the interfacial, structural, droplet characteristics, and bulk emulsion of PP. Results suggested that PP exhibited a promising emulsification potential and could be developed as a novel polysaccharide emulsifier in the food industry.

Gelation behavior and mechanism of Nicandra physalodes (Linn.) Gaertn. seeds pectin induced by Glucono–delta–lactone

Jian Li,Fei Pan,Yurou Yun,Jun Tian,Linyan Zhou

In this study, the physicochemical properties of pectin from Nicandra physalodes (Linn.) Gaertn. seeds (NPGSP) were analysed firstly, and the rheological behavior, microstructure and gelation mechanism of NPGSP gels induced by Glucono–delta–lactone (GDL) were investigated. The hardness of NPGSP gels was increased from 26.27 g to 226.77 g when increasing GDL concentration from 0 % (pH = 4.0) to 1.35 % (pH = 3.0), and the thermal stability was improved. The peak around 1617 cm −1 was decreased as the adsorption peak of the free carboxyl groups was attenuated with addition of GDL. GDL increased the crystalline degree of NPGSP gels, and its microstructure exhibited more smaller spores . Molecular dynamics was performed on systems of pectin and gluconic acid (GDL hydrolysis product), indicating that inter–molecular hydrogen bonds and van der Waals forces were the main interactions to promote gels formation. Overall, NPGSP has the potential commercial value for developing as a thickener in food processing.

Novel pectin-like polysaccharide from Panax notoginseng attenuates renal tubular cells fibrogenesis induced by TGF-β

Chunfan Huang,Xiaoqi Jing,Qianhu Wu,Kan Ding

Renal fibrosis is the final common result of a variety of progressive injuries leading to chronic renal failure. However, there are no effective clinical available drugs for the treatment. Notoginsenoside from Panax notoginseng could ameliorate renal fibrosis. We hypothesized that polysaccharide from this herb might have similar bioactivity. Here, we elucidated structure of a novel pectin-like polysaccharide designed SQD4S2 with a netty antenna backbone of glucogalacturonan substituted by glucoarabinan, glucurogalactan and galactose residues from this herb. Interestingly, SQD4S2 could reverse the morphological changes of human renal tubular HK-2 cells induced by TGF-β. Mechanism study suggested that this bioactivity might associate with N-cadherin (CDH2), Snail (SNAI1), Slug (SNAI2) depression and E-cadherin (CDH1) enhancement. In addition, SQD4S2 could impede critical fibrogenesis associated molecules such as α-SMA, fibronectin , vimentin , COL1A1, COL3A1, FN1 and ACTA2 expression induced by TGF-β in HK-2 cells. Current findings outline a novel leading polysaccharide for against renal fibrosis new drug development.

Self-Assembling pH-Responsive Nanoparticle Platform Based on Pectin–Doxorubicin Conjugates for Codelivery of Anticancer Drugs

Yinghua Tao,Dan Zheng,Jingyang Zhao,Kefeng Liu,Jing Liu,Jiandu Lei,Luying Wang

Pharmaceutical science based on biological nanotechnology is developing rapidly in parallel with the development of nanomaterials and nanotechnology in general. Pectin is a natural polysaccharide obtainable from a wide range of sources. Here, we show that doxorubicin (DOX)-conjugated hydrophilic pectin (PET) comprising an amphiphilic polymer loaded with hydrophobic dihydroartemisinin (DHA) self-assemble into nanoparticles. Importantly, conjugated DOX and DHA could be released quickly in a weakly acidic environment by cleavage of the acid-sensitive acyl hydrazone bond. Confocal microscopy and flow cytometry confirmed that these PET-DOX/DHA nanoparticles efficiently delivered DOX into the nuclei of MCF-7 cells. Significant tumor growth reduction was monitored in a female C57BL/6 mouse model, showing that the PET-DOX/DHA nanoparticle-mediated drug delivery system inhibited tumor growth and may improve therapy. Thus, we have demonstrated that pectin may be useful in the design of materials for biomedical applications.

Konjac glucomannan/pectin/Ca-Mg hydrogel with self-releasing alkalinity to recover phosphate in aqueous solution

Xingyu Wu,Xinyu Zhang,Yumei Xian,Yan Liu,Ling Luo,Lilin Wang,Chengyi Huang,Chao Chen,Jinsong He,Yanzong Zhang

The combination of polysaccharides can obtain stable, degradable, and environmentally friendly hydrogels, which have broad application prospects in adsorbents assembly. With Ca 2+ and Mg 2+ as crosslinkers, a new pectin/Konjac glucomannan/Ca-Mg composite hydrogel was prepared for phosphate adsorption by the alkali-thermal co-reaction method. Since Mg(OH) 2 can create a suitable pH condition for phosphate adsorption by Ca, Ca and Mg synergistically promoted phosphate adsorption and remained stable in the pH range of 4 to 10. FTIR, SEM-EDS, XRD, XPS , and zero potential analysis corroborated that the hydrogel used Ca and Mg as active sites to trap pollutants by electrostatic adsorption and fix phosphate through complexation to form Mg 3 (PO 4 ) 2 ·8H 2 O and CaPO 3 (OH) 2 ·H 2 O. Furthermore, it is unnecessary to separate the recovered phosphate from the hydrogel, and it can be used directly as a fertilizer. By being reused in the soil, it promoted seed germination and seedling growth. This adsorbent has the potential for recovery as a phosphorus-containing organic fertilizer after phosphorus adsorption.

Multibranched flower-like ZnO anchored on pectin/cellulose nanofiber aerogel skeleton for enhanced comprehensive antibacterial capabilities

Weina Wu,Ying Zhou,Jiefeng Pan,Yingying Wu,Gulden Goksen,Ping Shao

In this study, F-ZnO NPs were used as antibacterial agents , mussel bionic dopamine exerted its adhesive action to immobilize F-ZnO NPs on the pectin/CNF aerogel skeleton. Fruit and vegetable antimicrobial mats with safety, long duration of action and high efficiency were prepared and its potential application has been investigated. The results showed that a dopamine layer was deposited on the surface of the CNF , which promoted the tight adhesion of the F-ZnO NPs to the aerogel skeleton. The F-ZnO@D-CNF aerogel exhibited a slow release of zinc ions , with the first two days being 0.40 ± 0.16 and 1.01 ± 0.13 mg/mL. The aerogel was light, can stand on the petals without collapsing, has regular and uniform pore structure , good tensile/compressive properties and high antibacterial/anti-fungal properties. Strawberries packaged with F-ZnO@D-CNF aerogel exhibited an extended shelf life of 5 days. Additionally, the strawberries maintained a soluble solid content of 6.9 ± 0.82 % and a Vc content of 44.67 ± 3.51 mg/100 g. The weight loss, color and firmness were also notably superior to the other four groups. The final concentration of zinc ions in strawberries was 3.71 ± 0.28 μg/g, which is far below the recommended dietary intake.

Oral pectin/oligochitosan microspheres for colon-specific controlled release of quercetin to treat inflammatory bowel disease

Shisuo Jing,Huayuan Chen,Ergang Liu,Meng Zhang,Feng Zeng,Huan Shen,Yuefei Fang,Bahtiyor Muhitdinov,Yongzhuo Huang

Inflammatory bowel disease (IBD) is a chronic, life quality-reducing disease with no cures available yet. To develop an effective medication suitable for long-term use is an urgent but unmet need. Quercetin (QT) is a natural dietary flavonoid with good safety and multifaceted pharmacological activities against inflammation. However, orally administrated quercetin yields unproductive outcomes for IBD treatment because of its poor solubility and extensive metabolism in the gastrointestinal tract . In this work, a colon-targeted QT delivery system (termed COS-CaP-QT) was developed, of which the pectin (PEC)/Ca 2+ microspheres were prepared and then crosslinked by oligochitosan (COS). The drug release profile of COS-CaP-QT was pH-dependent and colon microenvironment-responsive, and COS-CaP-QT showed preferential distribution in the colon. The mechanism study showed that QT triggered the Notch pathway to regulate the proliferation of T helper 2 (Th2) cells and group 3 innate lymphoid cells (ILC3s) and the inflammatory microenvironment was remodeled. The in vivo therapeutic results revealed that COS-CaP-QT could relieve the colitis symptoms and maintain the colon length and intestinal barrier integrity.

Homogalacturonan enriched pectin based hydrogel enhances 6-gingerol's colitis alleviation effect via NF-κB/NLRP3 axis

Qun Wang,Zhaomei Wang,Jun Song,Kangjie Xu,Wenni Tian,Xu Cai,Jiamei Mo,Yong Cao,Jie Xiao

A nanolipidcarrier (NLC) loaded homogalacturonan enriched pectin (citrus modified pectin, MCP4) hydrogel was designed as a novel colon inflammation site-specific oral delivery system for 6-gingerol (6G) (6G-NLC/MCP4 hydrogel) administration, and its colitis alleviation effect were investigated. 6G-NLC/MCP4 exhibited typical “cage-like” ultrastructure with 6G-NLC embedded in the hydrogel matrix as observed by cryoscanning electron microscope. And due to the homogalacturonan (HG) domain in MCP4 specifically combined with Galectin-3, which is overexpressed in the inflammatory region, the 6G-NLC/MCP4 hydrogel targeted to severe inflammatory region. Meanwhile, the prolonged-release characteristics of 6G-NLC provided sustained release of 6G in severe inflammatory regions. The matrix of hydrogel MCP4 and 6G achieved synergistic alleviation effects for colitis through NF-κB/NLRP3 axis. Specifically, 6G mainly regulated the NF-κB inflammatory pathway and inhibited the activity of NLRP3 protein, while MCP4 regulated the expression of Galectin-3 and peripheral clock gene Rev-Erbα/β to prevent the activation of inflammasome NLRP3.

Physicochemical properties and Pb2+ adsorption capacity of freeze-dried hawthorn pectin fractions by gradient ethanol precipitation

Zhixin Li,Xiaoyan Zhang,Chuanhe Zhu

Three fractions of FHP20, FHP40 and FHP60 were obtained from freeze-dried hawthorn pectin by gradient ethanol precipitation (20–60 %), and their physicochemical properties and adsorption performance on Pb 2+ were investigated. It was found that the content of galacturonic acid (GalA) and esterification of FHP fractions gradually reduced with the increase of ethanol concentration. FHP60 had the lowest molecular weight (60.69 × 10 3  Da), and the composition and proportion of monosaccharides were significantly different. The experimental results of Pb 2+ adsorption showed that the adsorption process fitted well with the Langmuir monolayer adsorption and the pseudo-second-order models. Our findings suggested that pectin fractions with good homogeneity of molecular weight and chemical construction can be obtained by gradient ethanol precipitation, and hawthorn pectin could be developed as a potential adsorbent for Pb 2+ removal.

Comparison of gelling properties and flow behaviors of microbial transglutaminase (MTGase) and pectin modified fish gelatin

Tao Huang,Huizhu Zhao,Yaoyan Fang,Jinpei Lu,Wenge Yang,Zhaohui Qiao,Qiaoming Lou,Dalun Xu,Jinjie Zhang

The gelling and structural properties of microbial transglutaminase (MTGase) and pectin modified fish gelatin were compared to investigate their performances on altering fish gelatin properties. Our results showed that within a certain concentration, both MTGase and pectin had positive effects on the gelation point, melting point, gel strength, textural, and swelling properties of fish gelatin. Particularly, low pectin content (0.5%, w/v) could give fish gelatin gels the highest values of gel strength, melting temperature, and hardness. Meantime, flow behavior results showed that both MTGase and pectin could increase fish gelatin viscosity without changing its fluid characteristic, but the latter gave fish gelatin higher viscosity. Both MTGase and pectin could increase the lightness of fish gelatin gels but decreases its transparency. More importantly, fluorescence and UV absorbance spectra, particle size distribution, and confocal microscopy results indicated that MTGase and pectin could change the structure of fish gelatin with the formation of large aggregates. Compared with MTGae modified fish gelatin, pectin could endow fish gelatin had similar gel strength, thermal and textural properties to pig skin gelatin.

Engineering of pectin-dopamine nano-conjugates for carrying ruthenium complex: A potential tool for biomedical applications

Jiajing Diao,Feng Bai,Ying Wang,Qianqian Han,Xi Xu,Hongmei Zhang,Qiang Luo,Yanqing Wang

In this work, we designed polysaccharide-metal complex of dopamine-modified pectin linking ruthenium compound, which exhibited a certain inhibition specificity to human renal cell adenocarcinoma cell line 786-O. The chemical structure and physical properties of the polysaccharide-metal complex were well characterized by multiple analysis methods. The multi-spectral results revealed that pectin-dopamine have been successfully coordinated by ruthenium complex to form nano-conjugates, which self-assembled into relatively regular nanospheres of approximately 200 nm. The polysaccharide-metal complex has more amorphous and less viscosity than pectin, and cannot withstand as much strain as the pectin and pectin-dopamine systems. Pectin-dopamine can decrease the toxicity effect of Ru complex in normal cell line such as human normal renal epithelial cell line 293A.
