A novel cellulose/TiO2/β-CD hydrogel with high photocatalytic antibacterial activity and sustained release of drug was prepared. TiO2 sol with a diameter of about 9 nm in this hydrogel was the photoantibacterial agent. β-CD can significantly enhance photoantibacterial activity and the sustained-release effect of this hydrogel. The structure and property of this hydrogel composite were characterized by Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), scanning electron microscopy (SEM), and transmission electron microscopy (TEM). Escherichia coli and Staphylococcus aureus were selected as model bacteria to evaluate the antibacterial activity of the composite hydrogel under natural light irradiation and darkness, respectively. The in vitro release experiment used curcumin as a model drug in PBS buffer solution. The influence of β-CD on the antibacterial activity and sustained-release effect of the composite hydrogel was investigated. The results showed that the hydrogel composite presented a good photoantibacterial property, while the antibacterial activity can be neglected under the condition of no light. The complete release of curcumin from the composite was achieved after 120 h. Therefore, the hydrogel system has the characteristics of low cost and high effect, which could be used for an antibacterial sustained-release material.

Atropine sulfate (ATS) eye drops at low concentrations constitute a limited selection for myopia treatment, with challenges such as low ophthalmic bioavailability and inadequate stability. This study proposes a novel strategy by synthesizing ophthalmic sodium polystyrene sulfonate resin (SPSR) characterized by a spherical shape and uniform size for cationic exchange with ATS. The formulation of ATS@SPSR suspension eye drops incorporates xanthan gum and hydroxypropyl methylcellulose (HPMC) as suspending agents. In vitro studies demonstrated that ATS@SPSR suspension eye drops exhibited sustained release characteristics, and tropic acid, its degradation product, remained undetected for 30 days at 40 °C. The ATS levels in the tear fluids and aqueous humor of New Zealand rabbits indicated a significant increase in mean residence time (MRT) and area under the drug concentration–time curve (AUC 0-12 h ) for ATS@SPSR suspension eye drops compared to conventional ATS eye drops. Moreover, safety assessment confirmed the non-irritating nature of ATS@SPSR suspension eye drops in rabbit eyes. In conclusion, the cation-responsive sustained-release ATS@SPSR suspension eye drops enhanced the bioavailability and stability of ATS, offering a promising avenue for myopia treatment.

BACKGROUND:Osteoarthritis (OA) is a chronic and degenerative joint disease that remains a great challenge in treatment due to the lack of effective therapies. 4-octyl itaconate (4-OI) is a novel and potent modulator of inflammation for the treatment of inflammatory disease. However, the clinical usage of 4-OI is limited due to its poor solubility and low bioavailability. As a promising drug delivery strategy, injectable hydrogels offers an effective approach to address these limitations of 4-OI. OBJECTIVE:The aim of the study was to verify that the composite 4-OI/SA hydrogels could achieve a controlled release of 4-OI and reduce damage to articular cartilage in the group of osteoarthritic rats treated with the system. METHODS:In this study, an injectable composite hydrogel containing sodium alginate (SA) and 4-octyl itaconate (4-OI) has been developed for continuous intra-articular administration in the treatment of OA. RESULTS:After intra-articular injection in arthritic rats, the as-prepared 4-OI/SA hydrogel containing of 62.5 μM 4-OI effectively significantly reduced the expression of TNF-α, IL-1β, IL-6 and MMP3 in the ankle fluid. Most importantly, the as-prepared 4-OI/SA hydrogel system restored the morphological parameters of the ankle joints close to normal. CONCLUSION:4-OI/SA hydrogel shows a good anti-inflammatory activity and reverse cartilage disruption, which provide a new strategy for the clinical treatment of OA.

Rhamnogalacturonan-I (RG-I) was considered to be incompatible with gel formation. Due to its important health benefits, our previous study reported synergistic gelation of RG-I enriched pectin from citrus membranes with sequential acidic and alkaline treatment. In the present study, we further fabricated RG-I enriched pectin-based emulsion gels with the synergistic gelation for hydrophobic bioactive encapsulation. The effect of oil fraction and interfacial compositions including pectin itself and Tween 20 on the physical properties and delivery characteristics of emulsion gels was investigated. As to interfacial compositions, pectin reduced syneresis and swelling ratio and increased water holding capacity . Oil fraction had no marked influence on syneresis and swelling ratio, but significantly increased water holding capacity. Particularly, all the emulsion gels showed an elastic behavior and a full structure recovery after shearing. Increase in pectin instead of Tween 20 onto the surface of oil droplets and oil fraction markedly improved their gel strength. Curcumin was encapsulated within the oil phase of emulsion gels. Improvement in gel strength provided enhanced protection of curcumin against heat-induced degradation. In vitro digestion revealed that interfacial compositions decelerated curcumin release by increasing the proportion of pectin onto the surface, but didn't change its bioaccessibility eventually. Increase in oil fraction accelerated curcumin release, but reduced its bioaccessibility. Overall, fabrication of RG-I enriched pectin-based emulsion gel with pectin itself as interfacial composition and low oil fraction would contribute to improving curcumin bioavailability for its sustained release, which should be further validated in vivo .

Pork releases a large amount of exudate during storage, leading to microbial contamination, thereby shortening its shelf life. In this work, we designed an aerogel constructed of kenaf cellulose nanofiber (KCNF) and polyvinyl alcohol (PVA) as a support for cinnamon essential oil (CEO). This aerogel features low density (27.5 mg/cm 3 ), high porosity (96.6%), and strong water absorption (3067.82 wt%), while simultaneously possessing the functions of adsorbing pork exudate and sustaining the release of (CEO). The CEO-loaded aerogel, serving as a meat preservation pad, can simultaneously achieve water absorption, antibacterial, and antioxidant functions during the storage of chilled pork. Aerogel preservation experiments on chilled pork showed that it can prolong the preservation period of chilled pork by 6 days. Release kinetics studies revealed that the CEO within the aerogel was released continuously and evenly into the fatty food simulant, with this process primarily governed by Fickian diffusion. The designed aerogel shows great potential in meat preservation, providing a innovative idea for natural fibers used in sustainable packaging.

In order to develop an eco-friendly superabsorbent composite, flax yarn waste (FYW) was used as raw material to synthesize a novel flax yarn waste-g-poly(acrylic acid-co-acrylamide) (FYW/PAA) superabsorbent composite. Acrylic acid (AA) and acrylamide (AM) were grafted onto the pretreated flax yarn waste (PFYW) by free-radical graft copolymerization in homogeneous aqueous solution. The properties and synthesis conditions of the FYW/PAA superabsorbent composite were investigated. As a result, the prepared FYW/PAA attained the best water absorbency of 875 g/g in distilled water, 490 g/g in rainwater and 90 g/g in 0.9 wt% NaCl solution. The urea loading percentage of FYW/PAA could be modulated by the concentration of urea. The release of urea from FYW/PAA in water showed a typical three-stages sustained release behavior. Meanwhile, a weight residue of 53.6 wt% was attained after being buried in soil for 90 d.

Oridonin (ORI) is a natural active ingredient with strong anticancer activity. But its clinical use is restricted due to its poor water solubility, short half-life, and low bioavailability. The aim of this study is to utilize the metal organic framework material MOF-5 to load ORI in order to improve its release characteristics and bioavailability. Herein, MOF-5 was synthesized by the solvothermal method and direct addition method, and characterized by Scanning Electron Microscopy (SEM), X-Ray Diffraction (XRD), Fourier Transform Infrared Spectrometer (FTIR), Thermogravimetric Analysis (TG), Brunauer–Emmett–Teller (BET), and Dynamic Light Scattering (DLS), respectively. MOF-5 prepared by the optimal synthesis method was selected for drug-loading and in vitro release experiments. HepG2 cells were model cells. MTT assay, 4′,6-diamidino-2-phenylindole (DAPI) staining and Annexin V/PI assay were used to detect the biological safety of blank carriers and the anticancer activity of drug-loaded materials. The fabrication of scaffolds capable of the sustained release of the vascular endothelial growth factor (VEGF) to promote angiogenesis in long time remains a challenge in tissue engineering. Here, we report a facile approach for effectively fabricating a bioactive scaffold that gradually releases VEGF to promote angiogenesis. The scaffold was fabricated by coating polydopamine (PDA) on a konjac glucomannan (KGM) scaffold, followed by the surface immobilization of VEGF with PDA. The resulting VEGF-PDA/KGM scaffold, with a porous and interconnected microstructure (392 μm pore size with 84.80 porosity), combined the features of long-term biodegradability (10 weeks with 51 % degradation rate), excellent biocompatibility, and sustained VEGF release for up to 21 days. The bioactive VEGF-PDA/KGM scaffold exhibited multiple angiogenic activities over time, as confirmed by in vivo and in vitro experiments. For example, the scaffold significantly promoted the attachment and proliferation of human umbilical vein endothelial cells and the formation of vascular tubes in vitro . Moreover, the in vivo results demonstrated the formation and maturation of blood vessels after subcutaneous implantation in rats for four weeks. This promising strategy is a feasible approach for producing bioactive materials that can induce angiogenesis in vivo . These findings provide a new avenue for designing and fabricating biocompatible and long-term biodegradable scaffolds for sustained VEGF release to facilitate angiogenesis.

Overexpression of epidermal growth factor receptor (EGFR) in cancer is a key cause of recurrence of cervical cancer (CC). Although the EGF-EGFR pathway has been studied for decades, preventing tumor growth and recurrence caused by peripheral EGF remains a great challenge. In this work, a strategy is proposed to reduce the stimulation of high concentration EGF on tumor growth by using a thermo-sensitive hydrogel. The hydrogel is a triblock copolymer composed of polyethylene glycol (PEG) and poly (lactide glycolide) (PLGA). Based on the excellent temperature sensitivity, carrier capacity, swelling property and biocompatibility, the hydrogel can absorb the liquid around the tumor by injection and release EGF continuously at low concentration. The inhibitory effect of hydrogel on tumor growth is fully confirmed by an implanted tumor mouse model with human cervical cancer cell lines (HeLa) using triple-immunodeficient NCG mice. Compared with free EGF, the EGF-loaded hydrogel can hardly induce surface plasmon resonance (SPR) response, which proves that hydrogel can effectively weaken cytoskeleton rearrangement and inhibit cell migration by continuously releasing low concentration EGF. In addition, the EGF-loaded hydrogel can reduce cell proliferation by delaying the progress of cell cycle progression. Taken together, the hydrogel can effectively protect tumor microenvironment from the stimulation of high concentration EGF, delay cancer cellular processes and tumor growth, and thus providing an approach for inhibiting tumor recurrence of CC.

An anode-preloaded NaBF 4 reservoir enables long-lasting SEI regulation. • Solubility contrast allows NaBF 4 incorporation via aqueous slurry processing. • BF 4 − -derived species promote a B/F-rich, low-impedance SEI. • Ah-level Si-graphite pouch cells retain 89.2% capacity after 160 cycles. • The SRE strategy is extendable to graphite and SiO x /C anodes. Persistent active lithium (Li) loss caused by repeated rupture and reconstruction of the solid electrolyte interphase (SEI) remains a major barrier to long-life Li-ion batteries, especially for silicon (Si)-containing anodes with pronounced interfacial instability. Here, we report a sustained-release-enhanced (SRE) interphase strategy that enables long-lasting SEI regulation through an anode-preloaded NaBF 4 reservoir. By exploiting the high solubility of NaBF 4 in the aqueous slurry and its limited solubility in the carbonate electrolyte, NaBF 4 can be uniformly implanted during electrode fabrication and maintain the prolonged availability of BF 4 − -derived species near the anode/electrolyte interface during cycling. This solubility-governed process enhances anion participation in interfacial solvation and promotes the formation of a B/F-rich SEI with improved mechanical robustness and Li + transport kinetics. Importantly, the strategy is successfully demonstrated in a practically relevant micron-sized Si-graphite composite anode, where long-term interfacial stabilization is particularly challenging, delivering reduced polarization, improved rate capability, and markedly enhanced cycling stability. The strategy is further applicable to graphite and SiO x /C anodes. In practical full cells paired with a LiNi 0.8 Co 0.1 Mn 0.1 O 2 cathode, the SRE anode increases the initial Coulombic efficiency from 73.86% to 81.58%, and an Ah-level pouch cell retains 89.2% of its capacity after 160 cycles. This work provides a scalable route to regulate long-term SEI evolution in practical micron-sized Si-containing anodes and extend the lifetime of high-energy Li-ion batteries. A low-cost NaBF 4 salt is preloaded into the anode coating via aqueous slurry processing to construct a local reservoir. The prolonged availability of BF 4 − -derived species promotes a B/F-rich, low-impedance SEI, reducing lithium loss and interfacial resistance. As a result, silicon-graphite cells and Ah-level pouch cells achieve improved cycling stability, retaining 89.2% capacity after 160 cycles. Temperature is one of the most important factors driving cyanobacterial harmful algal blooms (CHABs). However, existing algicide designs rarely consider the influence of temperature on algal growth. Novel temperature-responsive sustained-release microspheres (T-SRMs) of anti-algal 5,4′-dihydroxyflavone (DHF) were designed using n -heptadecane as the temperature-responsive material, which can adjust DHF release behavior in response to temperature. Compared to DHF direct application, T-SRMs showed higher inhibition rates against Microcystis aeruginosa and sustained inhibitory activity at various temperatures (15-25 °C). The theoretical inhibition time of T-SRMs substantially exceeds that of conventional SRMs, with the potential to cover the entire CHABs outbreak period and extend the duration of validity. T-SRMs can accurately regulate DHF release rate based on water temperature. When the temperature is higher than the melting point of n -heptadecane (22 °C), the phase transition of heptadecane significantly promotes the DHF release rate. Moreover, salinity had no significant effect on the DHF release behavior, and the temperature-responsive materials can be adjusted to linearly regulate the release of DHF, which is favorable for the wide application of T-SRMs in freshwater, brackish and salt waters with flexible temperature windows. In addition, the T-SRMs had no apparent toxic effects on Chlorella vulgaris, Daphnia magna and Danio rerio . Our study has an important scientific significance to achieve a more intelligent, accurate and efficient control of CHABs.

Poly (lactic-co-glycolic acid) (PLGA) microspheres are widely used for controlled drug delivery, but the drug existing state in the microspheres significantly influences the characteristics of microspheres. The focus of the present study was to formulate meloxicam injectable sustained release microspheres (MLX-MS) and to investigate the influence of the drug existing state within the microspheres on the release properties, degradation and to evaluate the intra-articular (IA) treatment efficacy of osteoarthritis (OA). Different MLX-MS were prepared by changing drug existing forms (microcrystal, particle and molecular state) by the method of cosolvent combination or micronized drug and they were characterized by scanning electron microscopy-energy dispersive spectroscopy (SEM-EDS), X-ray diffraction (XRD) and in vitro release. The therapeutic effect of MLX-MS was investigated using OA rabbits after IA injection. The degradation, drug retention, and pharmacodynamics were also assessed. The results highlighted that the release and degradation of MLX-MS in the joint cavity were directly correlated to the drug existing state. This study offers crucial insights for developing sustained-release injectable MLX-MS, the microspheres with drug existing in molecular state (Mol-MS) were the optimum formulation which can provide sustained release and more effective therapy of joint inflammation.
