IL-1/IL-1R signaling induced by all-trans-retinal contributes to complement alternative pathway activation in retinal pigment epithelium

Xinxuan Cheng,Danxue He,Chunyan Liao,Sijie Lin,Liying Tang,Yuan-Liang Wang,Jiaoyue Hu,Wei Li,Zuguo Liu,Yalin Wu,Yi Liao

The underlying mechanisms of complement activation in Stargardt disease type 1 (STGD1) and age-related macular degeneration (AMD) are not fully understood. Overaccumulation of all- trans -retinal (atRAL) has been proposed as the pathogenic factor in both diseases. By incubating retinal pigment epithelium (RPE) cells with atRAL, we showed that C5b-9 membrane attack complexes (MACs) were generated mainly through complement alternative pathway. An increase in complement factor B (CFB) expression as well as downregulation of complement regulatory proteins CD46, CD55, CD59, and CFH were observed in RPE cells after atRAL treatment. Furthermore, interleukin-1β production was provoked in both atRAL-treated RPE cells and microglia/macrophages. Coincubation of RPE cells with interleukin-1 receptor antagonist (IL1Ra) and atRAL ameliorated complement activation and downregulated CFB expression by attenuating both p38 and c-Jun N-terminal kinase (JNK) signaling pathways. Our findings demonstrate that atRAL induces an autocrine/paracrine IL-1/IL-1R signaling to promote complement alternative pathway activation in RPE cells and provide a novel perspective on the pathomechanism of macular degeneration.

Sodium alginate hydrogel-encapsulated trans-anethole based polymer: Synthesis and applications as an eradicator of metals and dyes from wastewater

Saleem Raza,Ehsan Ghasali,Asif Hayat,Pengfei Zhang,Yasin Orooji,Hongjun Lin

Clean and safe water resources are essential for environmental safety and human health. Hydrogels and biomass polymers have attracted considerable attention in recent years, considering their nontoxicity, controllable performance, and high adsorption capacity. The interpenetrating network described here is a combination of a biomass polymer and a hydrogel adsorbent was established, the biomass polymer microspheres were first prepared with the combination of biomass monomer trans-anethole and maleic anhydride copolymer. A simple, environmentally friendly, and facile method of incorporating biomass polymer into sodium alginate biopolymer was developed by introducing the cross-linking agents calcium chloride and glutaraldehyde into the biomass polymer. Furthermore, the biomass polymer sodium alginate hydrogel (BP@SA/H) was characterized by FTIR, XPS, SEM, and XRD. In order to test materials' performance, the removal of pollutants and the adsorption study were also investigated after and before adsorption toward metals and dyes in water. We examined the factors influencing the materials, adsorption capability, such as initial concentration, time, absorbent amount, and pH. Moreover, the maximum adsorption values for Pb +2 and Cd +2 were 734.9 and 722 mg/g. While the adsorption toward RhB dye are 745 mg/g. In addition, the adsorption results were investigated using kinetic and isothermal models, demonstrating that biomass polymer hydrogel adsorption is chemisorption. Therefore, the as-developed biomass polymer sodium alginate hydrogel (BP@SA/H) is an exceptional multifunctional material that can be used to remove hazardous pollutants from wastewater.

Evidences of ultraviolet visible spectra of hydrogen sulfide scavenging trans-crotonaldehyde induced by hydrogen peroxide through mitochondria of rat heart

Yanbin Su,Yanwen Su,Tongxing Mu,Ning Jiang,Qingsong Zhang,Mengjie Li,Yuan Li,Shuxin Li

Evidences of ultraviolet visible spectra of hydrogen sulfide scavenging trans-crotonaldehyde (TCA) induced by hydrogen peroxide through mitochondria were probed for the first time. TCA was induced by hydrogen peroxide through mitochondria. When sodium hydrosulfide as hydrogen sulfide donor, comparison with control, ultraviolet visible spectra of TCA decrease were clear. Hydrogen sulfide released by garlic was detected by ultraviolet visible spectroscopy. When garlic as hydrogen sulfide donors, the ultraviolet visible spectra of TCA decrease were very obvious. The findings are strongly recommended that garlic as safe, stable, and controllable hydrogen sulfide donor become a promising drug which is worthy of further study.

Synthesis of isotactic polystyrene-block-polyethylene by the combination of sequential monomer addition and hydrogenation of 1,4-trans-polybutadiene block

Liang Hua-qing,Zhou Qi-hua,Long Yong-jiang,Wei Wan-chu,Liang Guo-dong,Wu Qing,Gao Hai-yang,Zhu Fang-ming

Herein, we demonstrate the synthesis of a well-defined diblock copolymer consisting of isotactic polystyrene ( i PS) and linear polyethylene, isotactic polystyrene-block-polyethylene ( i PS- b -PE), by the combination of sequential monomer addition and hydrogenation. Isospecific living polymerization of styrene and living trans -1,4-polymerization of 1,3-butadiene were catalyzed by 1,4-dithiabutandiyl-2,2′-bis(6-cumenyl-4-methylphenoxy) titanium dichloride (complex 1) activated by triisobutyl aluminum modified methylaluminoxane (MMAO) at room temperature to provide highly isotactic polystyrene ( i PS) and 1,4- trans -polybutadiene (1,4- trans -PBD) with narrow molecular weight distribution. Furthermore, the iPS-b-1,4- trans -PBD was synthesized via sequential monomer addition in the presence of complex 1 and MMAO. The hydrogenation of the 1,4- trans -PBD block was promoted by RuCl 2 (PPh 3 ) 3 used as a catalyst to produce i PS- b -PE.

Effects of the plant volatile trans‑2-hexenal on the dispersal ability, nutrient metabolism and enzymatic activities of Bursaphelenchus xylophilus

Yunhe Zhao,Shuangyu Xu,Hongbao Lu,Daxia Zhang,Feng Liu,Jin Lin,Chenggang Zhou,Wei Mu

Bursaphelenchus xylophilus causes pine wilt disease (PWD), which severely damages pine species. The plant volatile trans ‑2-hexenal has strong activity against nematodes, although the precise mechanism of this inhibitory action remains unclear. In this paper, the fumigant effects of the LC 10 and LC 30 of trans ‑2-hexenal on B. xylophilus were demonstrated. The trans ‑2-hexenal treatments significantly inhibited the dispersal ability of nematodes. The results also indicated that trans ‑2-hexenal affects the metabolism of nutrients and the activity of digestive enzymes . Among detoxifying enzymes , after treatment with trans ‑2-hexenal, glutathione S-transferase activity increased significantly and general esterase activity decreased significantly. Based on these results, trans ‑2-hexenal disturbs the normal physiological and biochemical activities of this nematode. These results provide valuable insight into the nematicidal mechanisms of trans ‑2-hexenal.

Engineered Zea mays phenylalanine ammonia-lyase for improve the catalytic efficiency of biosynthesis trans-cinnamic acid and p-coumaric acid

Jiangmei Zheng,Ruobin Sun,Dan Wu,Pengcheng Chen,Pu Zheng

Phenylalanine ammonia-lyase (PAL) plays a pivotal role in the biosynthesis of phenylalanine. PAL from Zea mays (ZmPAL2) exhibits a bi-function of direct deamination of L-phenylalanine (L-Phe) or L-tyrosine(-L-Tyr) to form trans -cinnamic acid or p -coumaric acid. trans -Cinnamic acid and p -coumaric acid are mainly used in flavors and fragrances, food additives, pharmaceutical and other fields. Here, the Activity of ZmPAL2 toward L-Phe or L-Tyr was improved by using semi-rational and rational designs. The catalytic efficiency ( k cat / K m ) of mutant PT10 (V258I/I459V/Q484N) against L-Phe was 30.8 μM −1 s −1 , a 4.5-fold increase compared to the parent, and the catalytic efficiency of mutant PA1 (F135H/I459L) to L-tyrosine exhibited 8.6 μM −1 s −1 , which was 1.6-fold of the parent. The yield of trans -cinnamic acid in PT10 reached 30.75 g/L with a conversion rate of 98%. Meanwhile, PA1 converted L-Tyr to yield 3.12 g/L of p -coumaric acid with a conversion rate of 95%. Suggesting these two engineered ZmPAL2 to be valuable biocatalysts for the synthesis of trans -cinnamic acid and p -coumaric acid. In addition, MD simulations revealed that the underlying mechanisms of the increased catalytic efficiency of both mutant PT10 and PA1 are attributed to the substrate remaining stable within the pocket and closer to the catalytically active site. This also provides a new perspective on engineered PAL.

Fabrication of antimicrobial PCL/EC nanofibrous films containing natamycin and trans-cinnamic acid by microfluidic blow spinning for fruit preservation

Menglu Wu,Zi-An Deng,Chaoyi Shen,Zhichao Yang,Zihan Cai,Di Wu,Kunsong Chen

Fruit is susceptible to various postharvest pathogens; thus, the development of multifunctional preservation materials that can achieve the broad-spectrum inhibition of different pathogens is a current research hotspot. Here, microfluidic blow spinning was used to create a biodegradable polycaprolactone/ethyl cellulose (PCL/EC) nanofibrous film that incorporated two naturally-sourced compounds, natamycin and trans -cinnamic acid, resulting in multi-microbial inhibition. The PCL/EC-based film had a smooth and even morphology, indicating the favorable integration of PCL and EC. After the incorporation of ingredients, the film exhibited good inhibitory activity against Escherichia coli , Staphylococcus aureus , and Botrytis cinerea , and it had finer fiber diameters, higher permeability, and antioxidant properties. We further demonstrated that strawberries that were padded with the film had good resistance to Botrytis cinerea . Also, the film did not interference with the qualities of the strawberries during storage. The study demonstrates a promising application for multi-antimicrobial and bio-friendly packaging materials in postharvest fruit preservation.

Investigation into the effects of the refining steps before deodorization on the formation of trans fatty acids in linseed oils

Mengru Kong,Yanli Du,Xiaodie Chen,Ruixin Cai,Jianhua Xie,Mingyue Shen

Trans fatty acids (TFAs) are formed in refined linseed oil during the deodorization step. Effects of deodorization on TFA formation in vegetable oils have been intensively investigated. However, the role of refining process (degumming, neutralization, bleaching) before deodorization on TFA formation in the deodorization step remains unclear. In this study, TFA generation potential in linseed oils refined by different ways of degumming, neutralization, bleaching was evaluated, and changes in their quality indexes were also measured. Results showed that adequate refining could improve the quality of linseed oil, while inadequate neutralization or bleaching induced more TFA formation during the deodorization step, suggesting that pigments or free fatty acids in linseed oils might promote TFA formation. Meanwhile, spiked experiments, in which partly refined linseed oils were spiked with pigments or α-linolenic acid, confirmed that pigments or free fatty acids in linseed oils induced the isomerization of unsaturated fatty acids. This study demonstrated that TFA formation in refined linseed oils could be mitigated by optimizing refining processes before deodorization, and also provided theoretical basis for oil refining industry to control the oil quality.

Novel nano-C@BaTiO3 fibers/polar trans-cyclohexane modified polyimide composite films with enhanced dielectric properties and energy storage density

Xingwu Jiang,Xiang Wang,Peifeng Liu,Jinzheng Huang

Polyimide (PI), a kind of polymer with excellent heat resistance, processing, mechanical and electrical insulation performance, stands out as a prime candidate for advanced dielectric energy storage materials. However, the low dielectric constant of PI has hindered its broader development, requiring dielectric modification. In order to increase the dielectric constant of PI, PI with a polar trans-cyclohexane structure was synthesized and characterized and then reinforced with nano-C@BT-fibers to produce a novel nanofiber reinforced PI film. Experimental results demonstrate that polar trans-cyclohexane enables pure PI films to achieve a dielectric constant of 8.89 at 100 Hz. Meanwhile, the dielectric constant of C@BT-fibers/modified PI composite films was indeed increased up to 39.48 at 100 Hz and the energy storage density increases to 1.028 J cm−3 with only 1.0 wt% addition content of C@BT-fibers, increased by 117.3% compared with that of pure PI (0.473 J cm−3).

Precision pore engineering in coconut shell-derived carbon molecular sieves enables high-efficiency separation of cis/trans-1,3-dimethylcyclohexane isomers

Jinhui He,Jun Yang,Jingdong Gou,Xue Ma,Yaning Zhao,Huiyong Chen,Xianjie Wang,Louwei Cui,Hong Hao,Liuyi Pan,Dong Li

In chemical engineering, biomedical, and reagent applications, cis/ trans -1,3-dimethylcyclohexane (cis/ trans -1,3-DMCH) isomers play critical roles due to their distinct structural configurations. However, their nearly identical boiling points and similar molecular structures make separation particularly challenging. This study developed a coconut shell-derived carbon molecular sieve (CCMS) using a stepwise activation strategy to precisely control pore structure and surface chemistry, creating a series of CCMS-x-y adsorbents. The CCMS-900-0.5 variant demonstrated superior performance in static adsorption tests, achieving maximum cis -1,3-DMCH uptake (62.73 mg/g) and selectivity (14.87). Dynamic breakthrough experiments confirmed exceptional separation efficiency with stable separation intervals of 275 ± 12.5 min, attributed to its optimal pore size distribution (5.37–5.99 Å). Grand canonical Monte Carlo (GCMC) simulations revealed that pure graphene slit models with 6 Å pores achieved maximum cis -isomer selectivity (2.76). Oxygen-containing functional groups were found to reduce separation efficiency by introducing steric hindrance. Advanced disordered mesoporous carbon models demonstrated faster diffusion rates for trans -1,3-DMCH compared to its cis counterpart. Interaction energy analysis indicated that adsorption primarily occurs through van der Waals forces, with stronger combined van der Waals and electrostatic interactions observed between the adsorbent and cis -1,3-DMCH. This work establishes both a practical framework for designing high-performance adsorbents for cyclic alkane isomer separation and a theoretical foundation for developing energy-efficient, low-carbon cis/ trans -1,3-DMCH separation technologies.

Ir-catalyzed tandem hydroformylation-transfer hydrogenation of olefins with (trans-/cis-)formic acid as hydrogen source in presence of 1,10-phenanthroline

Lei Liu,Han Gao,Shu-Qing Yang,Xiao-Chao Chen,Yong Lu,Ye Liu,Fei Xia

The one-pot tandem hydroformylation-reduction to synthesize alcohols from olefins is in great demand but suffering from low yields, poor selectivity and harsh condition. Herein, 1,10-phenanthroline ( L1 ) modified Ir-catalyst proved to exhibit multiple catalysis in terms of FA dehydrogenation, hydroformylation of olefins, and transfer hydrogenation of aldehydes to accomplish the tandem hydroformylation-transfer hydrogenation of olefins with formic acid (FA) as hydrogen source, as the result of 52–70% yields for the target alcohols and the corresponding formate esters (obtained upon esterification of the alcohol with FA). In this sequence, only the use of FA with trans - and cis -conformers could fulfill the reduction of aldehydes to the alcohols through transfer hydrogenation as well as greatly depress hydrogenation of the olefin, which universally occurred in the high pressured gaseous H 2 . Both the in situ FT-IR spectroscopic analysis and the DFT-calculations verified that, over L1 -[Ir(COD)Cl] 2 catalyst, cis -FA serving as the hydride-ligand was responsible for the efficient dehydrogenation of FA to release H 2 along with CO 2 , and trans- FA serving as the carbonyl O-containing ligand corresponded to the transfer hydrogenation of the aldehydes to the alcohols under the same catalytic conditions.

Fumigant Activity of 6 Selected Essential Oil Compounds and Combined Effect of Methyl Salicylate And Trans-Cinnamaldehyde Against Culex pipiens pallens

Wei-Bin Ma,Jun-Tao Feng,Zhi-Li Jiang,Xing Zhang

We studied the knockdown activity and lethal toxicity of 6 essential oil compounds—methyl salicylate, linalool, 2-phenethyl alcohol, eugenol, β-citronellol, and trans-cinnamaldehyde—as fumigants against adult female Culex pipiens pallens in the laboratory. Of the 6 products tested, trans-cinnamaldehyde was the most toxic (LC50  =  0.26 µl/l air, 24 h) with a slow knockdown time (KT95  =  176.5 min at 0.5 µl/l air). Methyl salicylate displayed a lower toxicity (LC50  =  1.17 µl/l air, 24 h) but the fastest knockdown activity (KT95  =  16.8 min) at the sublethal concentration 0.5 µl/l air. Furthermore, the binary mixture of methyl salicylate and trans-cinnamaldehyde exhibited a combined effect of fast knockdown activity and high toxicity against Cx. p. pallens adults, showing potential for development as natural fumigants for mosquito control.
