Biofilms formed by Bacillus licheniformis usually serve as a continuous bacterial contamination in the dairy industry. The purpose of this study was to explore how 1% organic acids (citric acid, phenyllactic acid, malic acid , and butyric acid) and their 1% organic peroxyacids (peroxycitric acid, peroxyphenyllactic acid, peroxymalic acid, and peroxybutyric acid) can inactive the planktonic and biofilm cells of B. licheniformis . Results demonstrated that the number of B. licheniformis planktonic cells was increased from 4 to 6.46 Log CFU mL −1 when treated with water. Organic peroxyacids, especially peroxyphenyllactic acid (PPA), obviously inactivated planktonic cells of B. licheniformis by a range of 3.32–4.14 Log CFU mL −1 . Moreover, organic peroxyacids could also decrease the cell surface hydrophobicity of bacteria, and effectively control the Ca 2+ -mediated biofilm formation by B. licheniformis in 96-well plates (reduction of OD values by 0.14–0.33) and on stainless steel (reduction of cell counts by 1.20–2.49 Log CFU cm −2 ). The low minimum biofilm eradication concentration of PPA (0.125%) also suggests its strong biofilm inactivation activity, which could serve as a novel and environmentally friendly biofilm control agent in the dairy industry.

Aptamer-based sensors (aptasensors) have been extensively employed in biosensing and diagnostics. However, the broader application of aptasensors fabricated with DNA and RNA aptamers has been limited due to the intolerance of DNA and RNA to nucleases in biological fluids. A majority of sugar-modified nucleic acids, in particular 2’-F and 2’-OMe-modified nucleic acids, demonstrate a remarkable resistance to nuclease degradation and thus hold great potential in the development of more stable aptamers and aptasensors. In this work, employing enzymes including a laboratory-evolved polymerase for 2’-modified nucleic acids, we carried out systematic evolution of ligands by exponential enrichment (SELEX) experiments to obtain 2’-F/OMe-RNA aptamers for Bacillus cereus 5/B/6 metallo-β-lactamase (B. cereus 5/B/6 MBL), a crucial marker of many penicillin-resistant strains. Three aptamers composed of 2’-OMe-A, 2’-F-U, 2’-F-C, and 2’-OMe-G nucleotides (Apt-FUC1, Apt-FUC2, and Apt-FUC3) have been obtained and systematically characterized. All of them exhibited excellent target binding affinities, with equilibrium dissociation constant (Kd ) values of 95.9, 70.0, and 81.3 nM, and outstanding biological stability. Combining FAM-labeled Apt-FUC1, Apt-FUC2, and Apt-FUC3 with graphene oxide (GO), which could quench the fluorescence of FAM, we successfully constructed three sensitive and biologically stable 2’-F/OMe-RNA aptasensors for B. cereus 5/B/6 MBL, with the limit of detection (LOD) values of 0.38, 0.27, and 0.29 nM. Moreover, these three aptasensors all demonstrated good specificity for target detection in biological samples. This work has established a streamlined strategy for generating stable 2’-F/OMe-RNA aptamers and aptasensors, which should broaden the application of sugar-modified nucleic acids in biosensing and diagnostics.

The recovery of precious metals is a project with both economic and environmental significance. In this paper, how to use bacterial mineralization to selectively recover gold from multi-ionic aqueous systems is presented. TheBacillus licheniformisFZUL-63, isolated from a landscape lake in Fuzhou University, was shown to selectively mineralize and precipitate gold from coexisting ions in aqueous solution. The removal of Au(III) almost happened in the first hour. Scanning electron microscope with X-ray energy dispersive spectroscopy (SEM/EDS-mapping) results and fourier transform infrared spectroscopy (FTIR) data show that the amino, carboxyl, and phosphate groups on the surface of the bacteria are related to the adsorption of gold ions. X-ray photoelectron spectroscopy (XPS) results implied that Au(III) ions were reduced to those that were monovalent, and the Au(I) was then adsorbed on the bacterial surface at the beginning stage (in the first hour). X-ray diffraction (XRD) results showed that the gold biomineralization began about 10 h after the interaction between Au(III) ions and bacteria. Au(III) mineralization has rarely been influenced by other co-existing metal ions. Transmission electron microscope (TEM) analysis shows that the gold nanoparticles have a polyhedral structure with a particle size of ~20 nm. TheBacillus licheniformisFZUL-63 could selectively mineralize and recover 478 mg/g (dry biomass) gold from aqua regia-based metal wastewater through four cycles. This could be of great potential in practical applications. Keywords:selective biomineralization;recovery of Au(III);AuNP;Bacillus licheniformisFZUL-63;aqua regia-based metal wastewater

Glutaminase plays a crucial role in nitrogen metabolism and is of vital significance for soy sauce production. In this study, the high glutaminase-producing strains, Bacillus pumilus -T1-11 and Bacillus safensis -T3-20, were isolated from fermented moromi through a two-step screening process. T1-11 displayed intracellular glutaminase activity of 146.39 ± 8.14 U/g, while T3-20 presented 139.26 ± 15.62 U/g, and both strains maintaining activity in Secondary Screening medium(SRM). Culture conditions were optimized by adjusting nitrogen/carbon sources, glutamine, NaCl, and pH. T1-11 achieved the maximum activity of 5716.95 ± 168.18 U/g in optimized medium and T3-20 reached peak activity of 4927.72 ± 62.79 U/g. The application of T1-11 and its enzyme extracts in soy sauce fermentation was evaluated. The moromi fermented with 7.0 log CFU/mL inoculum for 2 d showed the peak glutaminase activity (98.99 ± 15.13 U/g), significantly enhancing umami flavor, as confirmed by sensory and biochemical analyses. In extracted soy sauce, a 6.0 log CFU/mL inoculum cultured for 14 d total glutamic acid content to 82.78 ± 0.53 mmol/L. These modifications significantly enhanced the umami characteristics of soy sauce, demonstrating the potential application of this strain in improving the quality of traditional fermented food .

Tobacco bacterial wilt caused by Ralstonia solanacearum is one of the most devastating diseases. Microbial keystone taxa were proposed as promising targets in plant disease control. In this study, we obtained an antagonistic Bacillus isolate EM-1 from bacterial wilt-suppressive soil, and it was considered rhizosphere-resident bacteria based on high (100%) 16S rRNA gene similarity to sequences derived from high-throughput amplicon sequencing. According to 16S rRNA gene sequencing and MLSA, strain EM-1 was identified as Bacillus velezensis . This strain could inhibit the growth of R. solanacearum , reduce the colonization of R. solanacearum in tobacco roots, and decrease the incidence of bacterial wilt disease. In addition, strain EM-1 also showed a strong inhibitory effect on other phytopathogens, such as Alternaria alternata and Phytophthora nicotianae , indicating a wide antagonistic spectrum. The antimicrobial ability of EM-1 can be attributed to its volatile, lipopeptide and polyketide metabolites. Iturin A (C14, C15, and C16) was the main lipopeptide, and macrolactin A and macrolactin W were the main polyketides in the fermentation broth of EM-1, while heptanone and its derivatives were dominant among the volatile organic compounds. Among them, heptanones and macrolactins, but not iturins, might be the main potential antibacterial substances. Complete genome sequencing was performed, and the biosynthetic gene clusters responsible for iturin A and macrolactin were identified. Moreover, strain EM-1 can also induce plant resistance by increasing the activity of CAT and PPO in tobacco. These results indicated that EM-1 can serve as a biocontrol Bacillus strain for tobacco bacterial wilt control. This study provides a better insight into the strategy of exploring biocontrol agent based on rhizosphere microbiome.

The aim of the study was to analyze the correlation between the key aroma compounds and gDNA copies of Bacillus natto during fermentation and maturation of natto. Solvent-assisted flavor evaporation (SAFE) techniques were employed to extract volatile organic compounds. Aroma compounds were separated and analyzed through gas chromatography-olfactometry-mass spectrometry (GC-MS/O) using DB-5 and DB-Wax capillary columns. Seventy-nine compounds were extracted and identified, including 17 ketones, 17 alcohols, 9 aldehydes, 6 acids, 7 esters, 8 pyrazines, 8 aromatics, and 7 others. Twenty-two compounds were identified as the key aroma compounds of natto by aroma extraction dilution analysis (AEDA). Pyrazines played a key role in the flavor of natto both during the fermentation and maturation. 2,5-Dimethylpyrazine, 2,3,5-trimethylpyrazine, 2,3,5,6-tetramethylpyrazine, and 2,3,5-methyl-6-ethylpyrazine contributed substantially to natto flavor. They presented significant positive and negative correlations with the gDNA copies of Bacillus natto during fermentation and maturation, respectively.

A meticulously engineered bacillus-shaped CoNi ZIF-MXene@MWCNT carbon cloth (CC) composite was synthesized through a one-step immersion method, which was employed in the development of a novel electrochemical (EC) sensor for the sensitive detection of salidroside (SAL) in Rhodiola. Characterization of the CoNi ZIF-MXene@MWCNT CC was conducted using scanning electron microscopy (SEM), X-ray photoelectron spectroscopy (XPS), energy-dispersive X-ray spectroscopy (EDS), and EC methods. The bacillus-shaped composite showcased an exceptionally high rate of active site exposure, superior conductivity, and electrocatalytic efficiency, attributed to the synergistic effects of the constituent materials. The remarkable electrocatalytic capability facilitated the redox reaction of SAL, marked by a significant enhancement in the current signal. This led to a concentration-dependent response enabling the quantitative detection of SAL. The mechanism of the redox reaction might be the oxidation of the phenolic hydroxyl group of SAL. Demonstrating high sensitivity, selectivity, and stability, this novel bacillus-shaped composite-based EC sensor achieved a low detection limit (LOD) of 0.0958 µ g/mL for SAL, within an effective linear range of 0.5–500 µg/mL. Importantly, the EC sensor proved to be adept at practical detection of SAL in Rhodiola samples with complex matrices, and the salidroside content of Rhodiola crenulata was much higher than that of Rhodiola rosea , indicating that Rhodiola crenulata may have a higher medicinal value, offering a promising avenue for rapid and sensitive detection of SAL.

BACKGROUND Biological control agents are promising alternatives to conventional fungicides for managing postharvest diseases but their practical use is hindered by variable effectiveness and by application challenges. One promising yet underexplored approach involves utilizing bacteria that produce volatile organic compounds (VOCs) with antifungal properties. RESULTS In this study, Bacillus velezensis BF01, a strain with significant antifungal activity, was identified and isolated. Volatile organic compounds produced by this strain inhibited the growth and conidial germination of Botrytis cinerea , Fusarium oxysporum , and Colletotrichum gloeosporioides . Gas chromatography-mass spectrometry identified 90 VOCs, with ketones, esters, nitrogen compounds, aldehydes, and alcohols as the dominant components, many of which exhibited antifungal or plant resistance-inducing properties. To enhance practical application, B. velezensis BF01 was incorporated into a kaolin-based composite, which maintained long-term viability under refrigerated conditions and inhibited fungal growth through the controlled release of volatile organic compounds. This composite significantly extended the shelf life of strawberries by reducing postharvest decay. CONCLUSION The development of a kaolin-based B. velezensis BF01 composite provides an effective and sustainable biocontrol strategy for postharvest disease management. By improving bacterial stability and facilitating the controlled release of antifungal volatile organic compounds, this approach offers a practical alternative to chemical fungicides, highlighting its potential in extending fruit shelf life and improving food safety. © 2025 Society of Chemical Industry.

Background Millet bran (MB), a byproduct of millet production, is rich in functional components, but has a low utilization rate. In recent years, researchers have shown that fermentation can improve the biological activities of cereals and their byproducts. This study used Bacillus natto to ferment millet bran to improve its added value and broaden the application areas of MB. The bioactive component content, physicochemical properties, and functional activities of millet bran extract (MBE) from fermented millet bran were determined. Results After fermentation, the contents of soluble dietary fiber (SDF), β-glucan, polypeptides, polyphenols, flavones, and total amino acids increased by 92.0%, 164.4%, 111.4%, 32.5%, 16.4%, and 95.4%, respectively. Scanning electron microscopy revealed that the microscopic morphology of MBE changed from complete and dense blocks to loosely porous shapes after fermentation. After fermentation, the solubility, water holding capacity, and viscosity significantly increased, and the particle size decreased. Moreover, the glucose adsorption capacity (2.1 mmol/g), glucose dialysis retardation index (75.3%), and α-glucosidase inhibitory (71.4%, mixed reversible inhibition) activity of the fermented MBE (FMBE) were greater than those of the unfermented MBE (0.99 mmol/g, 32.1%, and 35.1%, respectively). FMBE presented better cholesterol and sodium cholate (SC) adsorption properties, and the adsorption was considered inhomogeneous surface adsorption. Conclusion Fermentation increased the bioactive component content and improved the physicochemical properties of MBE, thereby improving its hypoglycemic and hypolipidemic properties. This study will not only resolve the problem of millet bran waste but also promote the development of a higher value-added application method for millet bran. This article is protected by copyright. Grapes are susceptible to fungal infections before, during transport, or after harvest, causing significant economic losses. Traditional methods have high equipment costs and are prone to the development of drug resistance. The antifungal potential of volatile organic compounds (VOCs) produced by endophytic bacteria is beginning to be understood. VOCs are ideal for post-harvest disease control in fruits and vegetables, as they can work without direct contact with the fruit. In this study, we isolated pathogenic fungi ( Penicillium crustosum G-4, Alternaria alternata G-5, Arthrinium phaeospermum G-8) from post-harvest rotting green grape and used G-8 as a test organism, screened the endophytic bacterium Bacillus toyonensis D-6 from Typhonium giganteum Engl. that could produce strong antagonistic active VOCs, analyzed the components of its VOCs and screened for the highly effective inhibitory compound 3-methylvaleric acid (3-MA). The inhibitory activities and mechanisms of action of the VOCs and antifungal compounds were investigated. The results showed that the VOCs and 3-MA produced by D-6 inhibited the growth and reproduction of the pathogenic fungi, which mainly exerted antagonistic effects by destroying the normal morphological structure of mycelium and spores and damaging the integrity of cell membranes. In addition, appropriate concentrations of VOCs and 3-MA inhibited the spread of pathogenic fungi on green grapes and effectively delayed the decline in fruit quality. This opens up new possibilities for post-harvest preservation of green grapes and provides a theoretical basis for related applications.

Cottonseed meal (CSM) is a cost-effective protein source, but its application is limited by the toxicity of free gossypol. Traditional physical and chemical detoxification methods are costly, energy-intensive, and cause nutrient loss, while microbial fermentation-based biological detoxification is considered more sustainable than chemical or physical approaches. This study reports an alkaline protease from the marine strainBacillus safensisDL12 isolated from Yellow Sea sediments. Following cloning of its encoding gene and heterologous expression, enzymatic characterization of the purified enzyme revealed optimal activity at pH 8.0 and 50 °C, with Fe2+, Cu2+, Ni2+, and dithiothreitol (DTT) significantly enhancing its activity. Substrate hydrolysis analysis using the purified enzyme on soybean meal, peanut meal, rapeseed meal, and cottonseed meal demonstrated that, compared to the control group, cottonseed meal hydrolysates exhibited a 55.6% relative increase in peptide content and a 41.5% relative improvement in the degree of hydrolysis (DH), indicating higher hydrolysis efficiency among the four substrates. Notably, when hydrolyzing cottonseed meal with purified enzyme versus crude enzyme preparation at equivalent activity, the purified enzyme effectively reduced free gossypol content by 70% compared to the control, achieving more efficient detoxification than the crude enzyme preparation and most reported microbial treatments. These results highlight the potential ofB. safensisDL12 protease as a marine-derived enzyme, offering promising prospects for enhancing protein digestibility and addressing the long-standing challenge of gossypol toxicity in cottonseed meal utilization.

To investigate the dynamic changes during millet bran fermentation by Bacillus natto , we systematically monitored microbial growth, key enzyme activities, and the contents of major bioactive components over time. The changes in viable bacterial count, spore count, and the activities of protease, amylase, cellulase, and nattokinase (NK) fibrinolytic activity were measured throughout the 0–84 h fermentation process. Concurrently, variations in the contents of total sugars, reducing sugars, soluble dietary fiber (SDF), β-glucan, arabinoxylan, peptides, and polyphenols were analyzed. The results indicated that the viable bacterial count in the fermentation broth peaked at 48 h (9.3 log CFU/mL) and subsequently declined, while the spore count significantly increased to 7.6 log CFU/mL by 84 h. The activities of protease, amylase, cellulase, and NK fibrinolytic activity all exhibited a trend of initial increase followed by a decrease, reaching their respective maximum levels at 48 h. The contents of SDF, peptides, and polyphenols attained their highest values at 60 h, corresponding to 2.4 times, 2.17 times, and 1.5 times those of the unfermented control, respectively. The β-glucan content peaked at 24 h (31.31 mg/g millet bran), whereas the arabinoxylan content reached its maximum at 60 h, which was 19.4 times higher than that of the unfermented sample. Based on a comprehensive evaluation of all indicators, 48–60 h was determined to be the optimal fermentation duration for millet bran using B. natto . This research elucidates the relationship between enzyme activities and the accumulation of active components during fermentation, providing a theoretical foundation for the high-value utilization of millet bran and the development of functional products.
