Solid chip-based detection of trace morphine in solutions via portable surface-enhanced Raman spectroscopy

Lingyi Zhao,Yi Wei,Hao Fu,Ruiqin Yang,Qian Zhao,Hongwen Zhang,Weiping Cai

The accurate, sensitive and portable detection of morphine is important to handle judicial cases, but remains to be a great challenge. In this work, a flexible route is presented for the accurate identification and efficient detection of trace morphine in solutions based on surface-enhanced Raman spectroscopy (SERS) and a solid substrate/chip. A gold-coated jagged silicon nanoarray (Au-JSiNA) is designed and prepared via Si-based polystyrene colloidal template-reactive ion etching and sputtering deposition of Au. Such Au-JSiNA has three-dimensional nanostructure with good structural uniformity, high SERS activity and hydrophobic surface . Adopting this Au-JSiNA as SERS chip, trace morphine in solutions could be detected and identified in both dropping and soaking ways, and the limit of detection is below 10 -4 mg/mL. Importantly, such chip is especially suitable for the detection of trace morphine in aqueous solutions and even domestic sewage. The good SERS performance is attributed to the high-density nanotips and nanogaps on this chip as well as its hydrophobic surface. Additionally, the appropriate surface modification of this Au-JSiNA chip with 3-mercapto-1-propanol or 3-mercaptopropionic acid/1-(3-dimethylaminopropyl)-3-ethylcarbodiimide can further increase its SERS performances to morphine. This work provides a facile route and practical solid chip for SERS detection of trace morphine in solutions, which is significant to develop the portable and reliable instruments for on-site analysis of drugs in solutions.

Direct count of fluorescent microspheres in a microfluidic chip based on the capillary electrophoresis method

Jing Yang,Zhenqing Li,Dawei Zhang,Yoshinori Yamaguchi,Wen Xiao

Fluorescent microspheres (FMs) are tiny particles with special functions that are widely employed in biological research. Counting of microscale FMs is a great challenge by capillary electrophoresis. Herein we developed a method to count 2 μm FMs based on a microfluidic chip with a gradual change in inner size. Such a microfluidic chip can inhibit sample blocking at the inlet of the capillary. The results showed that FMs migrated in the wide part of the microchannel side by side, and then passed through the narrow part one by one. There was a linear relationship between the number of peaks in the electropherogram and concentration of FMs if they were running in the microchannel for more than 20 min. A high separation voltage may lead to aggregation of FMs in the microchannels, and about 2 × 104 FMs can be counted within 30 min by this microfluidic chip.

Microfluidic paper-based chip for parathion-methyl detection based on a double catalytic amplification strategy

Li Shuhuai,Pang Chaohai,Ma Xionghui,Zhang Yanling,Xu Zhi,Li Jianping,Zhang Meng,Wang Mingyue

The rapid detection of insecticides such as parathion-methyl (PM) requires methods with high sensitivities and selectivities. Herein, a dual catalytic amplification strategy was developed using Fe 3 O 4 nanozyme-supported carbon quantum dots and silver terephthalate metal–organic frameworks (Fe 3 O 4 /C-dots@Ag-MOFs) as current amplification elements. Based on this strategy, a novel electrochemical microfluidic paper-based chip was designed to detect PM. Fe 3 O 4 /C-dots@Ag-MOFs were synthesised by a hydrothermal method, and a molecularly imprinted polymer (MIP) was then synthesised on the surface of Fe 3 O 4 /C-dots@Ag-MOFs using PM as a template molecule. Finally, the reaction zone of a chip was modified with MIP/Fe 3 O 4 /C-dots@Ag-MOFs. PM from a sample introduced into the reaction zone was captured by the MIP, which generated a reduction current response at − 0.53 V in a three-electrode system embedded in the chip. Simultaneous catalysis by Fe 3 O 4 /C-dots and Ag-MOFs significantly enhanced the signal. The chip had a detection limit of 1.16 × 10 −11  mol L −1 and was successfully applied to the determination of PM in agricultural products and environmental samples with recovery rates ranging from 82.7 to 109%, with a relative standard deviation (RSD) of less than 5.0%. This approach of combining a dual catalytic amplification strategy with an MIP significantly increased the sensitivity as well as selectivity of chips and can potentially be used to detect a wide variety of target analytes using microfluidic paper-based chips. Design and preparation of a recyclable microfluidic SERS chip with integrated Au@Ag/TiO2 NTs

Chunyan Wang,Yi Xu,Conghui Deng,Zhixu Liu,Rong Wang,Huazhou Zhao

In the present paper, we report on the design and fabrication of a recyclable microfluidic SERS chip with an integrated Au@Ag/TiO2 NTs (gold core silver shell nanoparticles (NPs) with decorated TiO2 nanotube arrays (NTs)) composite SERS substrate. The stripped TiO2 NTs with regularly arranged pore structures were prepared on a Ti foil by easy tape lithography followed by anodic oxidation. The Au@Ag NPs were fabricated on the mouth surface and the inner walls of the TiO2 NTs within a microchannel without clogging via a facile self-assembly chemical plating composite method. Under optimized conditions, the microfluidic SERS chip provided excellent sensitivity to R6G molecules with a detection limit of 10−10 M and a SERS enhancement factor (EF) of 1.15 × 108. Versus single Au NPs, Au@Ag NPs and the Au@Ag/TiO2 plate, the optimized Au@Ag/TiO2 NTs showed improved SERS peak intensities for the C–C–C bending (613 cm−1) of 20, 2.3 and 1.6 times enhancement, respectively. The enhancement effect was attributed to both electromagnetic and chemical enhancement. The photocatalytic performance of the Au@Ag/TiO2 NTs was exploited to recycle the microfluidic SERS chip through UV light photocatalytic purification. The experimental result shows that the microfluidic SERS chip featured high reproducibility and could be used as an efficient SERS detector for many biochemical molecules.

Rapid Determination of Phenylalanine by Micro-chip Based Field Asymmetric Waveform Ion Mobility Spectrometry Technology

Da-Peng GUO,Yong-Huan WANG,Tian-Bai XU,Yuan ZHANG,Ling-Feng Li,Jin-Kai CHEN,Xiao-Zhi WANG,Peng LI,Ji-Kui LUO

The microchip field asymmetric ion mobility spectrometry (FAIMS) technology was used for rapid analysis of phenylalanine (PHE) using the metal diffusion tube-microchip μ-FAIMS system. The test pressure was set at 0.25 MPa, and the temperature of the gas generator for the diffusion tube was at 190 °C. The ion characteristic spectra were acquired under the optimized conditions with a gas flow rate of 2000 mL min −1 , a dispersion voltage of 152.8 V and a compensation voltage of −0.62 V for phenylalanine in positive mode. Phenylalanine gas samples with different concentrations were analyzed in detail by FAIMS. The results showed that the ion intensity was linearly correlated with phenylalanine concentration in the range from 6 mg L −1 to 20 mg L −1 , with the detection limit of 5.93 mg L −1 for phenylalanine. This investigation shows the feasibility of FAIMS technique for rapid detection of phenylalanine. The FAIMS instrument is simple, and has high sensitivity and fast response, and shows great potential for rapid detection of phenylalanine.

A novel three-dimensional microfluidic platform for on chip multicellular tumor spheroid formation and culture

Sun Duanping,Lu Jing,Chen Zuanguang,Yu Yanyan,Li Yinbao

The formation of three-dimensional (3D) multicellular cell spheroids such as microspheres and embryoid bodies has recently gained much attention as a useful cell culture technique, but few studies have investigated the suitability of glass for spheroids formation and culture. In this work, we present a novel three-dimensional microfluidic device made of poly(dimethylsiloxane) (PDMS) and glass for the easy and rapid synthesis and culture of tumor spheroid. The cell culture unit is composed of an array of microwells on the bottom of a glass plate, bigger microwells and elastomeric microchannels on the top of a PDMS plate. Cell suspension can be easily introduced into the cell culture unit and exchange with the external liquid environment by the microfluidic channels. A single tumor spheroid can be formed and cultured in each glass cell culture chamber, the surface of which was modified with poly(vinyl alcohol) to render it to be resistant to cell adhesion. As the cell culture medium could be replaced, spheroids of the human breast cancer (MCF-7) cells were cultured on the chip for 3 days, reaching the diameters of about 150 μm. Furthermore, the MCF-7 cells were successfully cultured on the chip in 2D and 3D culture modes. Results have shown that glass is well suitable for multicellular tumor spheroids culture. The established platform provides a convenient and rapid method for tumor spheroid culture, which is also adaptable for anticancer drug screening and fundamental biomedical research in cell biology.

Discriminative detection of soman or VX exposure using europium chelated microparticle-based immunofluorescence microfluidic chip

Qiao Lv,Pei-Yu Jiang,Jun-Mei Xia,Long-Hui Liang,Bo Chen,Yang Yang,Long Yan,Hui-Lan Yu,Chang-Cai Liu

The retrospective detection of organophosphorus nerve agents (OPNAs) exposure has been achieved by the off-site analysis of OPNA-human serum albumin (HSA) adducts using mass spectrometry-based detection approaches. However, few specific methods are accessible for on-site detection. To address this, a novel immunofluorescence microfluidic chip (IFMC) testing system combining europium chelated microparticle (EuCM) with self-driven microfluidic chip assay has been established to unambiguously determine soman (GD) and VX exposure within 20 min, respectively. The detection system was based on the principle of indirect competitive enzyme-linked immunosorbent assay. The specific monoclonal antibodies that respectively recognized the phosphonylated tyrosine 411 of GD-HSA and VX-HSA adducts were labeled by EuCM to capture corresponding adducts in the exposed samples. The phosphonylated peptides in the test line and goat- anti -rabbit antibody in the control line were utilized to bind the EuCM-labeled antibodies for signal exhibition. The developed IFMC chip could discriminatively detect exposed HSA adducts with high specificity, demonstrating a low limit of detection at exposure concentrations of 0.5 × 10 −6  mol/L VX and 1.0 × 10 −6  mol/L GD. The exposed serum samples can be qualitatively detected following an additional pretreatment procedure. This is a novel rapid detection system capable of discriminating GD and VX exposure, providing an alternative method for rapidly identifying OPNA exposure.

Machine learning assisted discrimination and detection of antibiotics by using multicolor microfluidic chemiluminescence detection chip

Fang Li,Min Zhu,Zimu Li,Nuotong Shen,Hao Peng,Bing Li,Jianbo He

The fabrication of multicolor chemiluminescence (CL) sensing chip for the discrimination and detection of multianalytes remains a great challenge. Herein, machine learning assisted multicolor microfluidic CL detection chip for the identification and concentration prediction of antibiotics was presented. Firstly, a three-channel microfluidic CL detection chip was fabricated. The three detection zones of the microfluidic detection chip were modified with CL catalyst Co(II) and different CL reagents including luminol, luminol mixed with fluorescein , and luminol mixed with phloxine B , respectively. Strong blue, green and pink-purple colored light emissions can be generated from the three detection zones in the presence of H 2 O 2 solution. The three multicolor CL emissions show different degrees of reduce in intensity and change in color in the presence of different antibiotics, including diethylstilbestro (DES), metronidazole (MNZ), kanamycin (KAN), isoniazide (INH), and ceftiofur sodium (CS), resulting in distinct fingerprint-like response patterns. The red (R), green (G), blue (B) and gray scale values of the three multicolor light emissions were extracted and ten characteristic sensing parameters were chosen to obtain multicolor CL response database. Then, machine learning assisted data analysis were carried out. The five antibiotics can be facilely classified by using principal component analysis (PCA) and hierarchical clustering analysis (HCA), and further quantified by using deep neural networks (DNN) algorithm. Good results were obtained for identification of binary antibiotic mixtures, spiked antibiotics in water samples, and unknown antibiotic samples. Satisfied results were obtained for concentration prediction of antibiotics. This work provides a simple machine learning assisted and multicolor microfluidic CL detection chip based CL sensing strategy for discrimination and quantitative detection of multiple analytes.

Molecular imprinting-based indirect fluorescence detection strategy implemented on paper chip for non-fluorescent microcystin

Li Bowei,Qi Ji,Liu Feng,Zhao Rongfang,Arabi Maryam,Ostovan Abbas,Song Jinming,Wang Xiaoyan,Zhang Zhiyang,Chen Lingxin

Fluorescence analysis is a fast and sensitive method, and has great potential application in trace detection of environmental toxins. However, many important environmental toxins are non-fluorescent substances, and it is still a challenge to construct a fluorescence detection method for non-fluorescent substances. Here, by means of charge transfer effect and smart molecular imprinting technology, we report a sensitive indirect fluorescent sensing mechanism (IFSM) and microcystin (MC-RR) is selected as a model target. A molecular imprinted thin film is immobilized on the surface of zinc ferrite nanoparticles (ZnFe 2 O 4 NPs) by using arginine, a dummy fragment of MC-RR. By implementation of IFSM on the paper-based microfluidic chip, a versatile platform for the quantitative assay of MC-RR is developed at trace level (the limit of detection of 0.43 μg/L and time of 20 min) in real water samples without any pretreatment. Importantly, the proposed IFSM can be easily modified and extended for the wide variety of species which lack direct interaction with the fluorescent substrate. This work offers the potential possibility to meet the requirements for the on-site analysis and may explore potential applications of molecularly imprinted fluorescent sensors.

Enhanced on-Chip modification and intracellular hydrogen peroxide detection via gigahertz acoustic streaming microfluidic platform

Feng Zhu,Zeyu Liu,Xiaoyu Wu,Die Xu,Quanning Li,Xuejiao Chen,Wei Pang,Xuexin Duan,Yanyan Wang

Developing effective strategies for the flexible control of fluid is vital for microfluidic electrochemical biosensing. In this study, a gigahertz (GHz) acoustic streaming (AS) based sonoelectrochemical system was developed to realize an on-chip surface modification and sensitive hydrogen peroxide (H 2 O 2 ) detection from living cells. The flexible and controlled fluid surrounding the electrochemical chip was optimized theoretically and applied in the sonoelectrochemical deposition of Au nanoparticles (AuNPs) first. Under the steady and fast flow stimulus of AS, AuNPs could be synthesized with a smaller and evener size distribution than the normal condition, allowing AuNPs to show an excellent peroxidase-like activity. Moreover, the AS also accelerated the mass transport of target molecules and improved the catalytic rate, leading to the enhancement of H 2 O 2 detection, with an extremely low detection limit of 32 nM and a high sensitivity of 4.34 μA/ (mM·mm 2 ). Finally, this system was successfully applied in tracking H 2 O 2 release from different cell lines to distinguish the cancer cells from normal cells. This study innovatively integrated the surface modification and molecules detection process on a chip, and also proposed a simple but sensitive platform for microfluidic biosensing application.

Rapid Evaluation of Antibacterial Carbohydrates on a Microfluidic Chip Integrated with the Impedimetric Neoglycoprotein Biosensor

Haijie Ji,Xueqiong Yang,Hang Zhou,Feiyun Cui,Qin Zhou

The colonization of some bacteria to their host cell is mediated by selective adhesion between adhesin and glycan. The evaluation of antiadhesive carbohydrates in vitro has great significance in discovering new antibacterial drugs. In this paper, a microfluidic chip integrated with impedimetric neoglycoprotein biosensors was developed to evaluate the antibacterial effect of carbohydrates. Mannosylated bovine serum albumin (Man-BSA) was taken as the neoglycoprotein and immobilized on the microelectrode-modified gold nanoparticles (Au NPs) to form a bionic glycoprotein nanosensing surface (Man-BSA/Au NPs).Salmonella typhimurium(S. typhimurium) was selected as a bacteria model owing to its selective adhesion to the mannose. Electrochemical impedance spectroscopy (EIS) was used to characterize the adhesion capacity ofS. typhimuriumto the Man-BSA/Au NPs and evaluate the antiadhesive efficacy of nine different carbohydrates. It was illustrated that the 4-methoxyphenyl-α-D-pyran mannoside (Phenyl-Man) and mannan peptide (Mannatide) showed excellent antiadhesive efficacy, with IC50values of 0.086 mM and 0.094 mM, respectively. The microfluidic device developed in this study can be tested in multiple channels. Compared with traditional methods for evaluating the antibacterial drug in vitro, it has the advantages of being fast, convenient, and cost-effective.Keywords:impedimetric biosensor;microfluidic chip;glycoprotein nano-sensing surface;Man-BSA/Au NPs;antiadhesive carbohydrates

A Disposable Multiplexed Chip for the Simultaneous Quantification of Key Parameters in Water Quality Monitoring

Huiyun Jiang,Bing Sun,Yan Jin,Junjie Feng,Hongwei Zhu,Lin Wang,Shucai Zhang,Zhe Yang

On-site simultaneous quantification of multiple contaminants in a water body is challenging, especially for parameters requiring complicated chemical reactions to measure such as Chemical Oxygen Demand (COD), ammonia nitrogen, and phosphate. A novel disposable multiplexed microfluidic device has been developed herein that allows the quantitative detection of up to five parameters at once. Solid reagent rather than commonly used liquid reagent was used to ensure long shelf life, and a “flow to dissolve” mechanism was provided accordingly for the thorough dissolution and mixing of a solid reagent on chip. Samples from river water and industrial wastewater were tested using the microfluidic chip, showing less than 15% deviation from results acquired with the traditional standard method. The test time though was only 1/6 of that required by the traditional method. In addition, the feasibility of using a smartphone to collect the colorimetric signal was discussed, and a data analysis method was provided for quantification purposes. The combination of the multiplexed chip and smartphone imaging provides a convenient and practical way to obtain accurate information on the water quality within a short period of time without the use of any sophisticated instruments.
