Apoptosis and Paraptosis Induced by Disulfiram-Loaded Ca2+/Cu2+ Dual-Ions Nano Trap for Breast Cancer Treatment

Zhenhu Guo,Xiaohan Gao,Jingsong Lu,Ying Li,Zeping Jin,Abdul Fahad,Neema Ufurahi Pambe,Hirotaka Ejima,Xiaodan Sun,Xiumei Wang,Wensheng Xie,Guifeng Zhang,Lingyun Zhao

Regarded as one of the hallmarks of tumorigenesis and tumor progression, the evasion of apoptotic cell death would also account for treatment resistance or failure during cancer therapy. In this study, a Ca2+/Cu2+ dual-ion “nano trap” to effectively avoid cell apoptosis evasion by synchronously inducing paraptosis together with apoptosis was successfully designed and fabricated for breast cancer treatment. In brief, disulfiram (DSF)-loaded amorphous calcium carbonate nanoparticles (NPs) were fabricated via a gas diffusion method. Further on, the Cu2+-tannic acid metal phenolic network was embedded onto the NPs surface by self-assembling, followed by mDSPE-PEG/lipid capping to form the DSF-loaded Ca2+/Cu2+ dual-ions “nano trap”. The as-prepared nanotrap would undergo acid-triggered biodegradation upon being endocytosed by tumor cells within the lysosome for Ca2+, Cu2+, and DSF releasing simultaneously. The released Ca2+ could cause mitochondrial calcium overload and lead to hydrogen peroxide (H2O2) overexpression. Meanwhile, Ca2+/reactive oxygen species-associated mitochondrial dysfunction would lead to paraptosis cell death. Most importantly, cell paraptosis could be further induced and strengthened by the toxic dithiocarbamate (DTC)–copper complexes formed by the Cu2+ combined with the DTC, the metabolic products of DSF. Additionally, the released Cu2+ will be reduced by intracellular glutathione to generate Cu+, which can catalyze the H2O2 to produce a toxic hydroxyl radical by a Cu+-mediated Fenton-like reaction for inducing cell apoptosis. Both in vitro cellular assays and in vivo antitumor evaluations confirmed the cancer therapeutic efficiency by the dual ion nano trap. This study can broaden the cognition scope of dual-ion-mediated paraptosis together with apoptosis via a multifunctional nanoplatform.

The depressing role of sodium alginate in the flotation of Ca2+-activated quartz using fatty acid collector

Long Wang,Hanyu Gao,Shimin Song,Weiguang Zhou,Na Xue,Yimiao Nie,Bo Feng

The ions activation of quartz is a thorny problem that deteriorates the flotation of metal ore. Polymer inhibitors have exhibited excellent depression performance in the selective flotation of metal-ion activated minerals. In this study, the inhibitory role of sodium alginate (NaAl) in the flotation of Ca 2+ -activated quartz was thoroughly explored by zeta potential, dynamic light scattering (DLS), Fourier transform infrared (FTIR) analysis, X-ray photoelectron spectroscopy (XPS) analysis and adsorption test. The flotation results suggested that NaAl could inhibit the flotation of Ca 2+ -activated quartz in sodium oleate (NaOl) system to different degrees, depending on the addition sequence of NaOl and NaAl. More significant depression of Ca 2+ -activated quartz could be achieved when NaAl was added prior to NaOl. It was found that prominent mutual interaction existed among Ca 2+ , NaOl and NaAl in the bulk solution, probably forming calcium alginate–calcium oleate crosslink product, which was not affected by the addition sequence of these two reagents. However, the interfacial behaviors of these two reagents on Ca 2+ -activated quartz surfaces differed under different reagent conditions. The addition of NaAl after NaOl could not only partially desorb the pre-adsorbed NaOl from mineral surfaces, but also decrease the mineral surface hydrophobicity by polymer coating, which therefore deteriorated the flotation of Ca 2+ -activated quartz. In contrast, when NaAl was added prior to NaOl, the pre-adsorption of NaAl made the further adsorption of NaOl rather difficult, which was likely attributed to the reticulated structure of NaAl and the associated steric hindrance effect. As a result, the flotation of Ca 2+ -activated quartz was tightly depressed.

Influence of Ca2+ on phosphate removal from water using a non-core-shell Fe3O4@ZIF-67 composites

Yinghao Xue,Ping Xiang,Yuzhu Jiang,Huilan Lian,Jingyu Mo,Mengying Li

Excessive phosphate in surface water can spur the growth of organism, which put a threat on the bio-safety of drinking water. Different from previous study, sodium acetate (NaAc) was used as modification agent of Fe 3 O 4 , then the magnetic Fe 3 O 4 @ZIF-67 composites with a non-core-shell sturture were synthesized. According to ZIF-67 loading and phosphate adsorption of synthesized Fe 3 O 4 @ZIF-67, the Fe 3 O 4 @ZIF-67 composites with a Co 2+ /Fe 3 O 4 molar ratio of 1:2 was taken into further evaluation. Fe 3 O 4 @ZIF-67 composites exhibited satisfactory adsorption performance and easy magnetic recovery, and the sorption capacity reached 116.59 mg P/g within 180 min (C 0 = 20 mg P/L, 298 K). The maximum adsorption capacity of phosphate was obtained at the neutral pH, and not significantly interfered with ions of K + , Mg 2+ , NO 3 − and SO 4 2- . Remarkably, Fe 3 O 4 @ZIF-67 described enhanced phosphate adsorption ability in the presence of Ca 2+ . To investigate the role of Ca 2+ , two modes (preloading and addition) of Ca 2+ dosing were examined and the improved adsorptive performance could be owing to the synergistic eff ;ect of adsorption and Ca-P micro-precipitation on the surface of Fe 3 O 4 @ZIF-67. During the regeneration process, the performance of desorbent was evaluated by phosphate releasing and secondary adsorption, and the absence or presence of Ca 2+ would have a significant impact on the selection of desorption solution.

Nerol-induced apoptosis associated with the generation of ROS and Ca2+ overload in saprotrophic fungus Aspergillus flavus

Tian Jun,Gan Yeyun,Pan Chao,Zhang Man,Wang Xueyan,Tang Xudong,Peng Xue

The contamination of food with Aspergillus flavus and subsequent aflatoxins is one of the most serious safety problems in the world. In this study of nerol (NEL)’s antifungal mechanism of action, we observed morphological and physiological changes in Aspergillus flavus. We found that NEL resulted in elevated levels of reactive oxygen species (ROS) and calcium ions (Ca 2+ ). On ROS assays, compared with the controls, the proportion of fluorescent cells treated with concentrations of 0.25, 0.5, 1, and 2 μL/mL NEL increased to 8.4 ± 1.07%, 10.2 ± 1.72%, 13.4 ± 0.50%, and 26.2 ± 4.21%, respectively. Increased mitochondrial dysfunction and oxidative stress induced by the interactions between Ca 2+ and ROS subsequently activate the release of cytochrome c and caspase activity. Characteristic changes of apoptosis were also observed via various detection methods, including phosphatidylserine externalization, nuclear condensation, and DNA fragmentation. Meanwhile, we found that the expression of CaMKs increased significantly in NEL-treated cells. In conclusion, our findings indicate that NEL has great potential as an eco-friendly antifungal agent for food preservation.

Gray mold , caused by Botrytis cinerea is an intractable fungal disease that causes extensive damage to agricultural products. In the search for novel antifungal active ingredients, we discovered a linear pyranocoumarin Pd-D-V was effective against B. cinerea in both in vitro and in vivo assays. Furthermore, this study investigated the effects of Ca 2+ and the Ca 2+ -calcineurin signaling pathway on its antifungal activity against B. cinerea . The results indicated that Pd-D-V reduced the concentration of Ca 2+ in the mycelia of B. cinerea ; CaCl 2 , the Ca 2+ channel blocker verapamil , or the calcineurin inhibitor cyclosporin A could affect the sensitivity of Pd-D-V against B. cinerea ; the expression of genes ( Bccch1, Bcmid1, BccnA, Bccnb1, Bcpmc1, and Bcpmr1 ) of the Ca 2+ -calcineurin signaling pathway decreased after Pd-D-V treatment. In summary, Pd-D-V is compound for developing fungicides against B. cinerea . Pd-D-V can reduce intracellular Ca 2+ concentration and disturb Ca 2+ homeostasis . The Ca 2+ -calcineurin signaling pathway is important in the antifungal activity of Pd-D-V against B. cinerea .

Enhanced performance of solid oxide fuel cells using highly conductive Ca2+ and F− co-doped Gd0.2Ce0.8O1.9 buffer layer

Shuting Li,Yuanyuan Liu,Changyang Liu,Ke Xue,Liuzhen Bian,Shengli An

Gadolinium-doped ceria (GDC) is the most widely used buffer layer to prevent the reaction between YSZ electrolyte and La 0.6 Sr 0.4 Co 0.2 Fe 0.8 O 3-δ (LSCF) cathode for commercial solid oxide fuel cell (SOFC). However, conductivity and sintering activity are vital to cell performance. In this paper, Ca 2+ and F − co-doped GDC (Ca 0.03 Gd 0.17 Ce 0.8 F 0.06 O 1.84-δ , CGDCF) is used as a buffer layer to improve the performance and stability of SOFC. The co-doped F − and Ca 2+ significantly improve the sintering properties and oxygen ion transport of traditional GDC material. Meanwhile, the optimized sintering temperature is 1300 °C, with the lowest ohmic resistance . The peak power density of a cell with a CGDCF buffer layer reaches 1.809 W cm −2 at 850 °C, which is 37 % higher than that of GDC. In addition, the cell with the CGDCF buffer layer exhibits excellent stability during the 150 h test owing to the lower migration of La and Sr elements. Thus, the Ca 2+ and F − co-doped GDC oxide emerges as a promising material for buffer layer applications.

Self-plasticized Ca2+-selective electrode with polyaniline and copolymer of aniline and 2,5-dimethoxyaniline as solid contact layers

Wang Youwei,Wang Xiushan,Xu Huan,Wu Ang,Chen Ying,Li Guiqiang,Dong Chuanxin,Li Yahui,Mei Chao,Li Shuailin,Zhang Hehu

To address solubility, plasticizer leakage, and ionic carrier issues in PVC-based solid ion-selective electrodes, a novel plasticizer-free photocurable solid Ca 2+ -selective electrode (GC/PANI/CPANI/Ca 2+ (PBA)-ISE) was developed using a novel electrode structure. A new Ca 2+ -sensitive membrane based on poly(butyl acrylate) (PBA) was synthesized by ultraviolet polymerization, with conductive polyaniline (PANI) as the solid contact layer and a copolymer of aniline and 2,5-dimethoxyaniline (CPANI) as the coating. All potential electrochemical tests were carried out on the prepared Ca 2+ -selective electrode. The response slope of the electrode was 28.33 mV decade −1 , the detection limit was 6.9 × 10 −6  M, the stable linear range was 1 × 10 −6 to 1 × 10 −2  M, and it had a good Nernst response. The chronopotentiometric analysis and water layer tests showed that the potential drift of GC/PANI/CPANI/Ca 2+ (PBA)-ISE was as low as 0.0125 mV s −1 and 1.247 mV h −1 . In addition, the interference tests of environmental factors were carried out. When the electrode was in different interference light and gas environments, the potential changes of 1.6 × 10 −4  mV s −1 and 1.2 × 10 −3  mV s −1 were observed. Compared with previous reports and commercial electrodes, the prepared GC/PANI/CPANI/Ca 2+ (PBA)-ISE showed good electrochemical performance, and the recovery rate in Ca 2+ test solutions was higher (97.2–99.9%). This study provided theoretical insights to address solubility, plasticizer leakage, and ion carrier leakage issues in solid-state contact ion-selective electrodes. At the same time, this study provided a new structure and preparation method for the preparation of all-solid-state Ca 2+ -selective electrode. Graphical abstract

Conductive MXene/adECM hydrogel promotes skeletal muscle regeneration and innervation through Ca2+ influx modulation and neuromuscular junction formation

Jin Mengying,Zhang Yahui,Liang Wei,Ao Rigele,Zhou Yuchen,Dang Wanwen,Wu Hongxu,Han Meng,Zhen Yonghuan,An Yang

Background Volumetric muscle loss (VML) leads to severe skeletal muscle dysfunction. While muscle tissue engineering offers a promising strategy, challenges persist due to insufficient neuromuscular innervation and poor reconstruction of neuromuscular junctions (NMJs). Conductive hydrogels can mimic the electrophysiological microenvironment and thus promote structural and functional regeneration, yet commonly used conductive materials still suffer from poor hydrophilicity, non-degradability, and potential cytotoxicity, while their underlying mechanisms remain unclear. Ti 3 C 2 T x MXene, a class of two-dimensional nanomaterials with high conductivity and biocompatibility, shows potential for repairing electroactive tissues. In this study, we developed a novel biomimetic electroactive hydrogel by incorporating Ti 3 C 2 T x MXene nanosheets into adipose-derived decellularized extracellular matrix (adECM). This study aimed to investigate the effects and mechanisms of MXene/adECM hydrogel on muscle regeneration and innervation. Results MXene/adECM hydrogel demonstrated excellent biocompatibility, biodegradability, and conductivity. Compared to the adECM hydrogel, the incorporation of MXene promoted myogenesis, along with increased expression of Desmin, MyoD1, and Myf5. Furthermore, the MXene/adECM hydrogel at the optimal concentration increased the average neurite length by 47.29 μm ( p  < 0.05) relative to the adECM group. Transcriptomic analysis combined with a neuromuscular co-culture system indicated that the MXene/adECM hydrogel promoted the formation of neuromuscular junctions (NMJs). The incorporation of MXene upregulated the expression of specific voltage-gated calcium channels at the motor endplate, with transcript levels of Cacna1a and Cacna1s increased to 2.1-fold and 3.1-fold, respectively. It was further observed that calcium signaling was enhanced in the MXene/adECM group, with the peak calcium signal intensity being 2.40 times that of the adECM group. In vivo rat VML model confirmed that, compared to the adECM hydrogel, the MXene/adECM hydrogel promoted an increase in regenerated muscle fiber area, reduced collagen deposition, and elevated the fluorescence intensity of CD31 and Tuj. The co-localization percentage of presynaptic and postsynaptic NMJ markers increased from 27.85 ± 8.69% to 42.21 ± 15.52%. Gait analysis showed significant improvements in print area, swing/stance ratio, and movement velocity. In the MXene/adECM group, the isometric tetanic force (ITF) upon sciatic nerve stimulation was significantly higher than that of the adECM group (0.082 ± 0.012 N vs. 0.057 ± 0.014 N, p  < 0.05), approaching the level of the uninjured group. Conclusion Together, these findings demonstrate that the incorporation of MXenes into adECM provides a promising strategy that integrates microenvironmental support with endogenous electrical cues to modulate calcium influx and promote NMJ formation, offering a new paradigm for the treatment of VML. Graphical Study on the therapeutic strategy and underlying mechanism of conductive MXene/adECM hydrogel for volumetric muscle loss (VML) repair. Ti3C2Tx MXene nanosheets were integrated into an adipose-derived decellularized extracellular matrix (adECM) to fabricate a conductive composite hydrogel. Upon implantation, the MXene/adECM hydrogel promoted structural regeneration and reinnervation of the injured skeletal muscle. The bioactive microenvironment and endogenous electrical conductivity facilitated neuromuscular junction (NMJ) formation, accompanied by enhanced calcium influx

Achieving high-performance far-red emission in Cr3+ activated garnet phosphor via Ca2+/Zr4+ doping toward plant lighting

Qinghong Meng,Jiaming Yuan,Xiangming Li,Wanjun Yu,Ji-Guang Li

Equimolar Ca2+ and Zr4+ doping was implemented to establish novel garnet-type Gd3‒xCaxAl2.95Ga2‒xZrxO12:0.05Cr3+ (x=0‒2.0) far-red (FR) phosphors. The quantum efficiency and temperature-dependent luminescence analysis successfully produced the optimal x=0.5 phosphor, which exhibited highly efficient (IQE=92.8%, EQE=31.6%) broadband FR-emission peaking at ~714 nm (full-width at half-maximum ~81 nm) and excellent thermal stability (95.5%@423 K) under 450 nm light excitation. Its performance is far superior to that of most reported Cr3+ activated FR-emitting garnet phosphors, and the emission spectrum can well superpose with the absorption band of the far-red phytochrome, making this phosphor have huge potential application in the construction of high-quality FR pc-LED used for plant lighting.

Sustainable and selective capture of Ca2+/Mg2+ from seawater desalination brines via hierarchical chitosan-based biochar aerogel adsorption

Wenjie Wang,Xin Li,Jiawang Qin,Chunyan Yang,Quanxun Liang,Shun Tang,Zhaoying Li,Jiale Wang,Shengchi Bai,Xiaoqi Wang,Wen Wen,Baicang Liu

To address the challenge of removing high-concentration calcium and magnesium ions (Ca 2+ /Mg 2+ ) from seawater desalination brines (SDB), this study introduces a novel chitosan-based biochar aerogel (CBA) system specifically engineered for hypersaline water treatment. The innovative adsorbent material utilizes high-temperature pyrolysis chitosan (85 % and 95 % deacetylation degree (DD)) generate biochar and then crosslinked into three-dimensional porous aerogels (CBA85 and CBA95), which were compared with conventional activated carbon aerogel (ACA). Comprehensive characterization revealed that CBA95 exhibited a high density of free amino groups and a well-developed mesoporous structure, with a specific surface area of 107.04 m 2 /g, approximately 3.14 times that of ACA. The maximum adsorption capacities of CBA95 for Ca 2+ and Mg 2+ were 5.15 mg/g and 9.37 mg/g, respectively. Selectivity adsorption experiments demonstrated CBA95 exhibited high selectivity for Ca 2+ and Mg 2+ , with distribution coefficient ( K d ) of 164.25 mL/g and 42.73 mL/g, respectively. The equilibrium data were aligned with the Langmuir isotherm model, suggesting a monolayer chemical adsorption mechanism, and further the electrostatic and complexation adsorption mechanism was identified and proposed. Most notably, fixed-bed reactor of CBA95 maintains over 87 % of Ca 2+ and 91 % of Mg 2+ removal efficiency after 9 adsorption-desorption cycles. The pilot-scale experiment of CBA95 in Qingdao SDB achieved 72.3 % Ca 2+ and 74.6 % Mg 2+ removal, confirming high industrial potential. Thus, this research not only establishes a theoretical foundation for applying chitosan-based materials in SDB softening but also provides a solution for large-scale engineering-scale applications due to their environmental sustainability and potential.

Ca2+-induced whey protein emulgels for the encapsulation of crystalline nobiletin: Effect of nobiletin crystals on the viscoelasticity

Yan Hu,Qiao Shu,Fei Liu,Lingling Lei,Bin Li,Yanping Cao,Yan Li

Due to the high crystallization, nobiletin was encapsulated by Ca 2+ -induced whey protein concentrate (WPC)-stabilized emulgels. Hydroxypropyl methylcellulose (HPMC) was used to regulate the interfacial properties of emulsions and viscoelasticity of emulgels. The formation of nobiletin crystals were monitored in emulgels and after digestion. Micro-rheometer was used to characterize the viscoelasticity of emulgels. Bioaccessibility of nobiletin was determined by an in vitro digestion model. Ca 2+ crosslinking increased the particle size of WPC-stabilized emulsion, while HPMC/WPC-stabilized emulsions kept smaller particle size. WPC-stabilized emulgels were more apt to syneresis, which could be prevented in the presence of HPMC. Nobiletin crystallization process in emulgels was delayed by HPMC. In the gelation process, HPMC addition increased the viscosity, but decreased the elasticity of emulgels. HPMC interrupted the crosslinking process of WPC and Ca 2+ . After destruction, emulgels had lower viscosity and elasticity, which became lower in the presence of HPMC. The fluctuation of mean square displacement profiles could indicate the formation of nobiletin crystals, which reduced the viscosity and elasticity of emulgels. HPMC increased the bioaccessibility of nobiletin. Higher Ca 2+ concentration was undesirable to enhance the nobiletin bioaccessibility, depending on the structure of emulgels.

Janus-structured nanocomposite films constructed with Ca2+ bridging TOCNF/CNTs networks for enhancing electromagnetic shielding and mechanical properties

Xinxin Cai,Jiayi Li,Xinqi Wang,Ruixiang Zhou,Bin Wang,Shudong Lin

With the advancement of 5G communications, the Internet of Things (IoT), and wearable electronics, there has been an increasing demand for lightweight, flexible, and high-performance electromagnetic interference (EMI) shielding materials. In this study, AgNWs/TOCNF/CNTs* nanocomposite films with Janus structure were successfully prepared by vacuum-assisted filtration followed by hot pressing. The film consisted of a conductive silver nanowire (AgNWs) layer and a supporting TOCNF/CNTs* layer. Specifically, the electromagnetic shielding and mechanical properties were effectively enhanced by constructing a three-dimensional (3D) network within the TOCNF/CNTs* layer through ionic crosslinking between Ca 2+ ions and carboxylate anions (−COO − ). Comprehensive characterization demonstrated that the film possessed excellent mechanical strength (tensile strength of 93.92 ± 3.20 MPa), superior electrical conductivity (67,114 S/m), outstanding EMI shielding effectiveness (EMI SET of 63.70 dB, EMI SEE/t exceeding 10,284 dB·cm 2 /g, and a shielding efficiency of 99.999957 %), as well as rapid Joule heating response under low voltage (2.0 V, reaching 115.60°C within less than 6s). These properties underscore the film’s strong potential for applications in portable electronics, aerospace, and military technologies.
