Background N-Acetyl-seryl-aspartyl-lysyl-proline (Ac-SDKP) is a short peptide with an anti-silicosis effect. However, the short biological half-life and low plasma concentration of Ac-SDKP hamper discovery of specific targets in organisms and reduce the anti-silicosis effect. A novel peptide, Ac-SDK (biotin) proline, termed “Ac-B”, with anti-fibrotic properties was synthesized. Methods Ac-B was detected quantitatively by high-performance liquid chromatography. Phagocytosis of Ac-B by the alveolar epithelial cell line A549 was investigated by confocal laser scanning microscopy and flow cytometry. To further elucidate the cellular-uptake mechanism of Ac-B, chemical inhibitors of specific uptake pathways were used. After stimulation with transforming growth factor-β1, the effects of Ac-B on expression of the myofibroblast marker vimentin and accumulation of collagen type I in A549 cells were analyzed by Western blotting. Sirius Red staining and immunohistochemical analyses of the effect of Ac-B on expression of α-smooth muscle actin (SMA) in a rat model of silicosis were undertaken. Results Ac-B had good traceability during the uptake, entry, and distribution in cells. Ac-B treatment prevented an increase in α-SMA expression in vivo and in vitro and was superior to that of Ac-SDKP. Caveolae-mediated uptake of Ac-B by A549 cells led to achieving anti-epithelial–mesenchymal transformation (EMT) effects. Conclusion Ac-B had an anti-fibrotic effect and could be a promising agent for the fibrosis observed in silicosis in the future.

Nanoscale phase-variant ultrasound contrast agents have attracted the interest of many researchers. However, it is a challenge to design nanobubbles that are activated by low-frequency medical ultrasound that do not cause damage to normal tissues. In this study, we designed a new type
of nanoparticle consisting of perfluoropentane and iron polyphthalocyanine loaded into liposomes. These nanoparticles can be activated by a low-frequency medical ultrasound imager at a frequency of 5 MHz for ultrasound imaging and clear reactive oxygen species at a rate of more than 50%. This
ability to scavenge excess reactive oxygen species can alleviate the damage these species cause and protect macrophages. Moreover, these nanoparticles can effectively enhance ultrasound contrast imaging for real-time visualization in the diagnosis and treatment of diseases.

Cathode membranes can provide innovative solutions for membrane-fouling mitigation and filtration performance enhancement, in combination with electric field in a new electrochemical system. In this research, a novel polyvinylidene fluoride/stainless steel/polypyrrole (PVDF/SS/PPy) cathode membrane was prepared via in situ flow-through electro-polymerization method. The PVDF/SS/PPy membrane exhibited excellent electric catalytic activity for 2-electron ORR (oxygen reduction reaction). A battery system with PVDF/SS/PPy membrane cathode and Fe sacrificial anode was developed for in situ •OH generation. A pH range of 3.5–4.0 is preferred and the area ratio of iron anode to PVDF/SS/PPy cathode membrane was ~1:6 according to the •OH production and iron consumption results. The phenol filtration tests suggested that the PVDF/SS/PPy cathode membrane has improved selectivity for removing small refractory organic pollutant such as phenol, the removal rate increased from 54.7% to 79.3%, while flux decreased from 152 L/(m 2 ·h) to 19 L/(m 2 ·h). The results of filtrating river water and BSA indicated that the PVDF/SS/PPy cathode membrane had excellent anti-fouling performance when the foulants load matches the •OH production.

Benzotriazole ultraviolet stabilizers (BUVSs) and phthalate esters (PAEs), widely employed industrial additives, have raised ecological concerns due to their potential environmental risks. However, their co-occurrence patterns in aquatic systems remain understudied, with their potential correlations poorly characterized. This study investigated the seasonal and spatial distribution patterns of BUVSs and PAEs in the Huangpu River, while evaluating the potential of PAEs as tracers for BUVSs. The results indicated that total BUVSs (∑BUVSs) and PAEs (∑PAEs) concentrations were higher in summer than in winter, ranging from 3.80 to 8.63 and 1702.67–8130.31 ng/L, respectively. Additionally, ∑BUVSs and ∑PAEs concentrations showed an increasing and then decreasing trend from upstream to downstream of the Huangpu River due to upstream industrial areas and shipping activities. Principal component analysis (PCA) identified common contamination sources associated with personal care products and plastic manufacturing. Pearson correlation analysis established correlations between specific BUVSs — 2-(benzotriazol-2-yl)-4-methylphenol (UV-P), 2-(benzotriazol-2-yl)-4-tert-butylphenol (UV-PS), 2-tert-butyl-6-(5-chlorobenzotriazol-2-yl)-4-methylphenol (UV-326), and 2-(benzotriazol-2-yl)-4-(2,4,4-trimethylpentan-2-yl) phenol (UV-329) — and PAEs including dimethyl phthalate (DMP), diethyl phthalate (DEP), and dibutyl phthalate (DBP) within this watershed. While the meta-analysis of published data revealed a significant correlation between BUVSs and PAEs, the experimental results of this study demonstrate a divergent trend. The observed specificity in correlations between individual PAEs and BUVSs suggests that the utility of PAEs as tracers for BUVSs warrants further systematic investigation. This study provides a foundation for understanding the distribution and potential association of BUVSs and PAEs in the Huangpu River.

Traditional hydrogel sensors can easily freeze or evaporate under extreme conditions, resulting in their performance degradation. At the same time, the low strength/toughness and high resistance of hydrogels hinder their practical applications in different fields. In this work, we report a simple and effective strategy to prepare mechanically strong and conductive nanocomposite hydrogels by covalent interaction of acidified carbon fibers and various ions. In this paper, a wearable strain sensor with high sensitivity and flexibility and self-healing, anti-freezing and anti-drying properties was prepared using polyacrylic acid, sodium alginate and acidified carbon fibers as raw materials. The hydrogel has an interpenetrating cross-linked network structure. The interpenetrating cross-linking structure makes the hydrogel sensor exhibit excellent ductility, remarkable repeatability and stable sensing performance. The hydrogel can be used as a human strain sensor for the real-time detection of human movement, including joint movement and speech. The hydrogel strain sensor shows low strain detection limit (3% strain), fast response time (∼100 ms), high sensitivity factor and excellent stability. The hydrogel has good self-adhesion with various substrates (such as copper, glass, plastic and skin). The hydrogel has good strain sensing performance at different strains (1–200%). In addition, the hydrogel can maintain excellent flexibility, extensibility and conductivity in a wide temperature range from −20 °C to 50 °C, effectively improving the practicability and durability of the hydrogel in practical applications. Therefore, the hydrogel has a good application prospect in wearable devices, soft robot systems and health monitoring applications.

Copper-related reactive oxygen species (ROS) formation can lead to neuropathologic degradation associated with Alzheimer's disease (AD) according to amyloid cascade hypothesis. A complexing agent that can selectively chelate with copper ions and capture copper ions from the complex formed by copper ions and amyloid−β (Cu − Aβ complex) may be available in reducing ROS formation. Herein, we described applications of guluronic acid (GA), a natural oligosaccharide complexing agent obtained from enzymatic hydrolysis of brown algae , in reducing copper-related ROS formation. UV–vis absorption spectra demonstrated the coordination between GA and Cu(II). Ascorbic acid consumption and coumarin–3–carboxylic acid fluorescence assays confirmed the viability of GA in reducing ROS formation in solutions containing other metal ions and Aβ. Fluorescence kinetics, DPPH radical clearance and high resolution X − ray photoelectron spectroscopy results revealed the reductivity of GA. Human liver hepatocellular carcinoma (HepG2) cell viability demonstrated the biocompatibility of GA at concentrations lower than 320 μM. Cytotoxic results of human neuroblastoma (SH–SY5Y) cells verified that GA can inhibit copper-related ROS damage in neuronal cells. Our findings, combined with the advantages of marine drugs, make GA a promising candidate in reducing copper-related ROS formation associated with AD therapy.

Fumonisin B1(FB1) is the most common food-borne mycotoxin produced by theFusariumspecies, posing a potential threat to human and animal health. Pigs are more sensitive to FB1ingested from feed compared to other farmed livestock. Enzymatic degradation is an ideal detoxification method that has attracted much attention. This study aimed to explore the functional characteristics of the carboxylesterase FumDSB in growing pigs from the perspective of brain–gut regulation. A total of 24 growing pigs were divided into three groups. The control group was fed a basal diet, the FB1group was supplemented with FB1at 5 mg/kg feed, and the FumDSB group received added FumDSB based on the diet of the FB1group. After 35 days of animal trials, samples from the hypothalamus and jejunum were analyzed through HE staining, qRT-PCR and immunohistochemistry. The results demonstrated that the ingestion of FB1can reduce the feed intake and weight gain of growing pigs, indicating that several appetite-related brain-gut peptides (including NPY, PYY, ghrelin and obestatin, etc.) play important roles in the anorexia response induced by FB1. After adding FumDSB as detoxifying enzymes, however, the anorexia effects of FB1were alleviated, and the expression and distribution of the corresponding brain-gut peptides exhibited a certain degree of regulation. In conclusion, the addition of FumDSB can reduce the anorexia effects of FB1by regulating several brain-gut peptides in both the hypothalamus and the jejunum of growing pigs. Keywords:fumonisin B1;carboxylesterase FumDSB;hypothalamus;jejunum;brain-gut peptide;ghrelin;obestatin;growing pigsKey Contribution:In this study, the effects of carboxylesterase FumDSB on appetite-related brain-gut peptides in both the hypothalamus and the jejunum of growing pigs were evaluated; next, the mechanism and safety of FumDSB was explored from the perspective of brain–gut regulation, which provided a research basis for the application of FumDSB in actual production.

β tricalcium phosphate (β-TCP) is a common biological ceramic in bone tissue engineering due to its excellent biocompatibility and biodegradability. However, owing to its inherent properties of poor injectability and forming ability, the application of TCP in extrusion 3D printing is quite limited. To solve this problem, we innovatively printed gelatin and PVA (Polyvinyl alcohol) binary systems by taking advantage of the ability of gelatin to solidify rapidly at low temperatures and the ability of PVA to effectively improve phase separation during extrusion. Here, we fabricated a series of novel Gel-PVA-TCP scaffolds by incorporating different concentrations of PVA (5%, 10%, 15%) through 3D printing. Collectively, the addition of PVA made differences in three ways. First, compared to the PVA-free system, the novel printing system significantly improved the printability of TCP paste. The printing temperature was decreased to 35–45 °C by doping different concentrations of PVA. Next, the novel printing system maintained the excellent forming ability and printing precision of the scaffolds. Compared to the macropores of scaffolds without doping PVA (456.1 ± 11.2 μm), the macropores of Gel-PVA-TCP were 486.1 ± 26.5, 446.1 ± 15.2, and 443.7 ± 26.3 μm. Last, after sintering, the scaffolds with different concentrations of PVA exhibited similar compressive strength (3.82 ± 0.22, 3.34 ± 0.23, 3.74 ± 0.38, 3.47 ± 0.48 Mpa) and distinct micropores (4.44 ± 0.7, 1.54 ± 0.3, 2.59 ± 0.6, 3.59 ± 1.0 μm) on the surfaces of the scaffolds. Moreover, it was found that differences in the microstructure significantly facilitate the adhesion of bone marrow mesenchymal stem cells to scaffolds. Hence, the methods we provided hold a great promise for application in 3D printing techniques.

Adding thermal conductive filler is an effective method to improve the thermal conductivity of polymer matrix . In this research, we demonstrated that the polymer composites with much improved thermal conductivity while maintaining low electrical conductivity , which could be achieved via using hybrid 2D stacked filler and controlling the alignment of the filler in polymer matrix. In order to do this, the graphene oxide (GO) was prepared and simultaneously reduced/functionalized by diethylenetriamine (DETA) to obtain NH 2 -functionalized graphene (NfG) which designed to be immobilized on the surface of large-sized insulating hexagonal boron nitride (h-BN) via π-π stacking interaction. In this situation, since the NfG sheets were fixed on the surface of h-BN, the NfG sheets were well separated from each other and participated in the resin curing process. Hence, not only significantly enhanced thermal conductivity (∼3.409 W/m·K, in-plane direction) was obtained, but also a very low electrical conductivity was achieved. The low electrical conductivity was believed to be ascribed to both embedded insulating network of h-BN to inhibit the mobility of charge carrier and well-separated NfG sheets via π-π stacking interaction. In addition, the nanocomposites also exhibited good thermal stability and adhesive properties . We believed that this special structure will provide a new thought for fabricating thermal interface materials (TIMs) with much high thermal conductivity as well as low electrical conductivity.

Evaluating the plant risk of soil pollution by plant physiological indices usually requires a long cycle and has significant uncertainty. In this study, oxidative potential (OP) of the in situ heavy metal contaminated soils was measured by the dithiothreitol method. The oxidative stress response of the model plant ryegrass ( Lolium perenne L. ) induced by heavy metal contaminated soil was evaluated by the biomarkers, including superoxide dismutase and total antioxidant capacity. The comprehensive biomarker response index has a significant exponential correlation with the OP of soil ( r = 0.923, p < 0.01) in ryegrass. Metabolomics analysis also showed a significant relationship of the metabolic effect level index of amino acids and sugars with OP. Random forest was selected from four machine learning models to screen the metabolites most relevant to OP, and Shapley additive explanations analysis was used to explain the contribution and the influence direction of the features on the model. Based on the selected 20 metabolites, the metabolic pathways most related to OP in plants, including alkaloid synthesis and amino acids metabolism, were identified. Compared to the plant physiological indices, OP is a more stable and faster indicator for the plant risk assessment of heavy metals contaminated soil.

This study aimed to determine how antibiotic-driven intestinal dysbiosis impairs the development and differentiation of the digestive tract and immune organs of host animals. BALB/C neonatal mice were orally administered ceftriaxone or vancomycin from postnatal day 1 to day 21 and sacrificed on day 21. The diversity and abundance of the intestinal bacteria, morphological changes and barrier function of intestinal tract, and the splenic CD4+CD25+Foxp3+ T cells were investigated. The gut microbiota and intestinal tissue were damaged, and the numbers of Ki67-, Muc2- and ZO-1-positive cells were significantly decreased in the antibiotic treatment groups. Furthermore, the administration of ceftriaxone, but not vancomycin, led to a significant reduction in the abundance of splenic CD4+CD25+Foxp3+ T cells. Each antibiotic caused intestinal dysbiosis and characteristically influenced the regeneration of intestinal epithelial cells, formation of the intestinal mucus layer and tight junctions, and differentiation of splenic Foxp3+ Treg cells of the neonatal mice before any clinical side effects were observed. The potent ability of each antibiotic to affect the makeup of intestinal commensal microbiota may be a key determinant of the spectrum of antibiotics and influence the health of the host animal, at least partly.

Micronutrient intake during early life is crucial for supporting healthy intestinal development and function. However, the impact of the timing of vitamin B6 (VB6) intake on intestinal development in young organisms has not been investigated. The study investigated the effects of VB6 (45 mg/kg) intake during the active phase versus the rest phase on intestinal digestion, intestinal barrier, and ileal and colonic microbiota, and used fecal microbiota transplantation to explore the role of the microbiota in VB6-mediated intestinal health. VB6 intake during the active phase increased digestive enzyme activity, intestinal villus height, and mRNA expression of tight junction proteins ( Occludin and ZO-1 ), thereby enhancing digestive function and epithelial barrier integrity. Microbiota analysis revealed that active phase VB6 intake predominantly modulated the ileum microbial community, promoting colonization by beneficial genera such as Ligilactobacillus and Bifidobacterium , and increasing ileum short-chain fatty acid levels. Fecal microbiota transplantation confirmed that VB6 acts through the gut microbiota. Therefore, this study demonstrates that VB6 modulates intestinal development via the gut microbiota, with active phase intake conferring greater benefits on intestinal development than rest phase intake .
