Rapid differentiation of D/L-homocysteine, D/L-homocystine, and S-allyl-D/L-cysteine using ion mobility-mass spectrometry

Li Xiang,Ling Ling,Chi Chaoxian,Ding Chuan-Fan

The development of a rapid analytical method for distinguishing chiral sulfur-containing compounds is essential due to the significant differences in biological activity exhibited by different enantiomers. This work presents a straightforward and rapid method for the chiral discrimination of D/L-homocysteine (D/L-Hcy), D/L-homocystine (D/L-Hcy 2 ), and S-allyl-D/L-cysteine (D/L-SAC) using cyclodextrin (CD) and metal adducts in conjunction with ion mobility-mass spectrometry (IM-MS). Through the formation of ternary complexes, separation was achieved with IM peak resolutions of 1.41 for [α-CD+D/L-Hcy+Co−H] + , 1.24 for [α-CD+D/L-Hcy 2 +Ca−H] + , and 1.65 for [β-CD+D/L-SAC+Fe−H] + . Theoretical calculations were performed to identify the optimal conformations of these ternary complexes. Conformational analysis elucidated distinct structural differences at the molecular level, providing insight into the chiral separation mechanism. Owing to its superior resolving power, this method enables the direct quantification of D/L enantiomers in mixtures. It was successfully applied to the determination of D/L-SAC in garlic samples, obviating the need for laborious sample pretreatment or derivatization steps. Overall, this work establishes a robust analytical platform for the chiral analysis of sulfur-containing amino acids and their derivatives. Graphical abstract

Microbial production of D-arabitol from D-xylose by engineered Trichosporonoides oedocephalis: Fermentation performance and salt-stress transcriptional responses

Yue Tang,Xin Ju,Yuqi Peng,Zhicheng Xie,Wei Liu,Jiaolong Fu,Jiajia Chen,Liangzhi Li

D-Arabitol is a naturally occurring pentitol widely used as a low-calorie sweetener and functional ingredient in food formulations. However, microbial production from biomass-derived D-xylose remains constrained by low product titers, limited conversion yields, and the lack of efficient engineered xylose-utilizing strains. In this study, the osmotolerant yeast Trichosporonoides oedocephalis was engineered to produce D-arabitol from D-xylose through the introduction of a heterologous XI (xylose isomerase)-ArDH (arabitol dehydrogenase) pathway. Fermentation conditions relevant to high-sugar food systems were systematically optimized. Compared with the addition of 0.5 g/L KCl (yielding 17.69 g/L D-arabitol), supplementation with 30 g/L KCl significantly increased D-arabitol production, reaching 35.13 g/L in shake flasks and 43.91 g/L in a 5-L bioreactor. Semi-batch fermentation further increased the final titer to 55.21 g/L with a yield of 0.280 g/g. Kinetic analysis indicated predominantly growth-associated D-arabitol formation under these conditions (Luedeking-Piret coefficient, α = 0.61). Comparative transcriptomic analysis revealed that KCl supplementation induced phase-dependent physiological responses, including enrichment of nucleotide metabolism and coordinated transcriptional regulation of central carbon metabolism during exponential growth, as well as increased expression of sugar transport and protein maintenance pathways during the stationary phase. These changes were accompanied by elevated activities of xylose isomerase and arabitol dehydrogenase, suggesting that salt stress is associated with efficient carbon utilization and the biosynthesis of polyols. Importantly, KCl represents a food-compatible and physiologically relevant process regulator, highlighting the potential of salt-mediated metabolic regulation for food-related polyol fermentations. These findings provide an effective strategy for food-related D-arabitol production from xylose and offer insights into salt-associated metabolic regulation in yeast.

Detecting N-Phenyl-2-Naphthylamine, L-Arabinose, D-Mannose, L-Phenylalanine, L-Methionine, and D-Trehalose via Photocurrent Measurement

Feng Li,Ruoxi Yang,Jian Xu,Guohai Xu,Ye Wu

The concentration of small molecules reflects the normality of physiological processes in the human body, making the development of simple and efficient detection equipment essential. In this work, inspired by a facile strategy in point-of-care detection, two devices were fabricated to detect small molecules via photocurrent measurement. A linear response of the photocurrent against the concentration of the small molecules was found. In the first device, metal ions were introduced into gel substrates made by xanthan gum to enhance photocurrent response. N-phenyl-2-naphthylamine was measured when iron or manganese ions were used. L-Phenylalanine was measured when the gel was modified by samarium, iron, cerium, or ytterbium ions. L-(+)-Arabinose was detected via the gels modified by iron or holmium ions. D-(+)-Mannose was detected when the gel was modified by ytterbium, manganese, chromium, or sodium ions. L-Methionine was detected in the gels modified by samarium, zinc, or chromium ions. The second device was based on a paper sheet. A sugar-like molecule was first synthesized, which was then used to modify the paper. The detection was possible since the photocurrent showed a linear trend against the concentration of D-Trehalose. A linear fit was conducted to derive the sensitivity, whose value was found to be 5542.4. This work offers a novel, simple, and environmentally sustainable platform that is potentially useful in remote areas lacking medical devices.

Fluorescent DNA-templated silver nanoclusters for highly sensitive detection of D-penicillamine

Lingyuan Liu,Qianyi Zhang,Feng Li,Mei Wang,Jing Sun,Shuyun Zhu

Herein, fluorescent DNA-templated silver nanoclusters (DNA-AgNCs) with red emission were synthesized and utilized as novel probe to detect D-penicillamine (D-Pen) for the first time. D-Pen molecules contain a thiol which can combine with Ag to form a non-fluorescent ground state complex, inducing the aggregation of DNA-AgNCs followed by the fluorescence quenching. The quenching mechanism is well-studied and found to be a static quenching process. This method can detect D-Pen in the range of 0.025–0.7 μM with the detection limit as low as 8 nM, which is 1–3 orders of magnitude more sensitive than those based on other fluorescent nanoprobes. More importantly, the preparation procedure for DNA-AgNCs is fast and without the requirement of heavy metal ions. Thus, this detection strategy is time-saving and eco-friendly. Satisfactory recoveries have been acquired for monitoring D-Pen in human serum samples and pharmaceutical samples owing to the high sensitivity.

Chiral Enantioselective Assemblies Induced from Achiral Porphyrin by l- and d-Lysine

Shanshan Wu,Zheng-Zhi Yin,Datong Wu,Yongxin Tao,Yong Kong

π-Conjugated porphyrins have aroused particular attention for nanofabrication and biomimics; however, little attention has been paid to porphyrins-based chiral analysis owing to the achiral feature of porphyrins. Here, we demonstrated a chiral self-assembly of achiral porphyrin induced by l- and d-lysine (l- and d-Lys), and the resultant porphyrin self-assembly exhibited alterable morphologies depending on the inducer used (l- or d-Lys). The supramolecular chirality of the self-assembly was characterized by circular dichroism (CD) spectra, confirming successful transfer of molecular chirality from l- and d-Lys to the self-assembly. The enantioselective property of the chiral self-assembly was also investigated by using tryptophan (Trp) isomers as the model, and the results indicated that the developed chiral self-assembly showed significantly higher affinity toward l-Trp than d-Trp. Also in this work, the l-/d-Lys-induced chiral self-assembly of porphyrin and the supramolecular interaction between the self-assembly and l-/d-Trp were also studied by density functional theory (DFT).

A non-enzymatic urine glucose sensor with 2-D photonic crystal hydrogel

Yan Zequn,Xue Min,He Qian,Lu Wei,Meng Zihui,Yan Dan,Qiu Lili,Zhou Lijun,Yu Yingjie

A novel polymerized crystalline colloidal array (PCCA) sensing material for the detection of urine glucose was developed by embedding a two-dimensional (2-D) polystyrene crystalline colloidal array (CCA) in 3-acrylamidophenylboronic acid (3-APBA)-functionalized hydrogel. After adjusting the cross-linker concentration, this material showed significant sensitivity for glucose under lab conditions, the particle spacing of the PCCA changed from 917 to 824 nm (93 nm) within 3 min as the glucose concentration increased from 0 to 10 mM, and the structural color of the PCCA changed from red through orange, to green, and finally, to cyan. In further experiments, this material was used to semi-quantitatively detect glucose in 20 human urine (HU) samples. Compared with the traditional dry-chemistry method, which was applied widely in clinical diagnosis, the PCCA method was more accurate and cost-effective. Moreover, this method can efficiently avoid the errors induced by most of the urine-interfering elements like vitamin C and ketone body. With a homemade portable optical detector, this low-cost intelligent sensing material can provide a more convenient and efficient strategy for the urine glucose detection in clinical diagnosis and point-of-care monitoring.

Ultrasensitive colorimetric and fluorescent dual-readout assay for D-penicillamine based on the chloride boosted oxidative ability of copper ions

Dan Zhao,Jingjing Huang,Juan Li,Xiao Ma,Fang Wang,Honglei Zhang,Jiaze Xie,Jian Sun,Chuanxia Chen

Dual-readout protocols with two signal transduction channels can significantly improve the detection precision and diversity. In contrast to the previous tedious and time-consuming preparation of two response materials, a convenient and commercially available small molecule o -Phenylenediamine (OPD)-based dual-mode system was developed for the ultrasensitive detection of D-penicillamine (D-PA). Fortunately and interestingly, it is demonstrated that chloride ions (Cl − ) could largely enhance the oxidative ability of copper ions (Cu 2+ ) and improve the reaction rate and efficiency of the Cu 2+ -OPD system, leading to the boosted colorimetric and fluorescent signals and shorter reaction time. Meanwhile, D-PA could strongly bind with Cu 2+ to form a stable Cu 2+ -D-PA complex, thereby inhibiting the aforementioned oxidation process and exhibiting decrease of absorbance values and fluorescence intensity. Inspired by the excellent analytical performance, an “INH” molecular logic gate was devised by employing Cl − and D-PA as the inputs, and the dual-mode strategy has been successfully employed to probe D-PA in tablets and human serum samples. Such cost-effective yet efficacious analytical tool not only improve the sensing performance of the Cu 2+ -OPD reaction system, but also enhance the detection sensitivity for D-PA and promote the application and development of OPD-based reactions.

Measurement and Correlation of Liquid–Liquid Equilibria for the Ternary Systems of Water + d-Fructose + 1-Butanol, Water + d-Glucose + 1-Butanol, and Water + d-Galactose + 1-Butanol at (288.2, 303.2 and 318.2) K

Tiantian Ye,Haibin Qu,Xingchu Gong

The liquid–liquid equilibrium (LLE) data of ternary systems of water + d-fructose + 1-butanol, water + d-glucose + 1-butanol, and water + d-galactose + 1-butanol were determined at three temperatures (288.2, 303.2, and 318.2 K) and atmospheric condition. The distribution coefficients of the monosaccharides were lower than 0.05. For all the three saccharides, their content in the organic-rich phase increased as their content in the aqueous phase increased. The experimental tie-line data were fitted using the Othmer–Tobias equation. The saccharide universal quasichemical functional-group activity coefficient (S-UNIFAC) model was used to predict the LLE data with all the average absolute deviations (AAD) values higher than 0.013. The modified universal quasichemical model was then used to correlate the LLE data. The correlation results agreed better with experimental data than the prediction results obtained with S-UNIFAC. In the production of traditional Chinese medicines, 1-butanol is a promising extractant used to separate active compounds and saccharides in aqueous herbal extracts.

Protective effect of thymoquinone on glycation of human myoglobin induced by d-ribose

Jing-Jing Liu,Zhan-Yi Wang,Bin-Bin Jiang,Shu-Qin Gao,Ying-Wu Lin

Nonenzymatic glycation and the subsequent accumulation of advanced glycation end-products (AGEs) in proteins are factors underlying long-term pathogenesis in diabetes. The study of protein glycation is crucial for elucidating their relationship with diabetes mellitus and related disorders. This study explores the interaction between d -ribose and human myoglobin (HMb), as well as the protective effect of thymoquinone (TQ) on glycation. A time-dependent in-vitro glycation study was performed to investigate the mechanism of d -ribose-induced structural interference of HMb in the absence and presence of TQ. Spectroscopic and proteomic analysis indicated that the presence of TQ significantly reduced the total amount of AGEs while maintaining structural characteristics of HMb. 14 glycated sites on HMb were further identified via liquid chromatography-tandem mass spectrometry (LC-MS/MS) after incubation with d -ribose for 12 h, predominantly interacting with lysine residues. TQ was found to disrupt this interaction, reducing the glycated sites from 14 to 12 sites and the percentage of glycated peptides from 26.50 % to 12.97 %. Additionally, there was a significant decrease in the degree of glycation at the same sites. In summary, our findings suggest that TQ has the potential to act as an anti-glycation agent and provide a comprehensive understanding underlying the inhibition mechanism of glycation.

Surface Activity of Alkoxy Ethoxyethyl β-d-Glucopyranosides

Yulin Fan,Fang Fu,Langqiu Chen,Jiping Li,Jing Zhang

Dioxyethene fragment (−(OCH2CH2)2−) was introduced into traditional alkyl β-d-glucopyranosides to ameliorate the water solubility, and eight nonionic surfactants, that is, alkoxy ethoxyethyl β-d-glucopyranosides with alkyl chain lengths (n = 6–16), were synthesized and characterized. Their hydrophilic and lipophilic balance number, water solubility, critical micelle concentration (cmc), γcmc, Γmax, and hygroscopic rate decreased with an increase in the alkyl chain length. Hexadecoxy ethoxyethyl β-d-glucopyranoside had no water solubility at 25 °C. Decoxy ethoxyethyl β-d-glucopyranoside had the best emulsifying property in the toluene/water and n-octane/water systems and the strongest foaming property, whereas dodecoxy ethoxyethyl β-d-glucopyranoside had the best emulsifying property in the rapeseed oil/water system. Such β-d-glucopyranosides (n = 6–12) exhibited excellent surface activity. In addition, for the binary mixture of alkoxy ethoxyethyl β-d-glucopyranosides (n = 8, 10, 12) and sodium dodecyl sulfate or cetyl trimethyl ammonium chloride, their cmc values were lower than the pure β-d-glucopyranosides, indicating that they had synergistic interactions. The fan focal conic textures of alkoxy ethoxyethyl β-d-glucopyranosides (n = 7–16) were observed during the cooling process under a polarizing optical microscope. Alkoxy ethoxyethyl β-d-glucopyranosides (n = 14, 16) had the related melting points and the clear points with differential scanning calorimetry. With β-d-glucopyranosides (n = 6–16) and n-butanol as the surfactant and cosurfactant, respectively, and with cyclohexane as the oil phase, the related microemulsion areas in their pseudoternary phase diagram system were investigated with the visual observation at 25 °C. Along with the slashing requirements of petroleum consumption, environmental protection, and green and sustainable development, nonionic sugar-based alkoxy ethoxyethyl β-d-glucopyranosides should be expected to have their potential practical application because of their strengthened hydrophilicity, improved water solubility, and enhanced surface activity.

D-limonene ameliorates metabolic dysfunction-associated steatotic liver disease by inhibiting the PPARγ/SCD-1 pathway and improving lipid metabolism disorders

Fang Wang,Yan Qing Liu,Jun Nan Wei,Xiao Fen Wang,Feng Qin Li,Xiu Jia Tian,Dan Li,Yu Xuan Gao,Xin Zhi Guo,Xue Yan Li,Ping Luo

Introduction Metabolic dysfunction-associated steatotic liver disease (MASLD) is a highly prevalent chronic liver disorder characterized by intrahepatic lipid accumulation, oxidative stress, and inflammatory responses, underscoring the urgent need for effective therapeutic strategies. D-limonene, a cyclic monoterpenoid primarily derived from citrus fruits, possesses well-documented antioxidant, anti-fibrotic, and wound-healing properties. However, its therapeutic potential in MASLD remains largely unexplored. Methods To evaluate the therapeutic efficacy of D-limonene, a rat model of MASLD was established using a high-fat diet (HFD). An integrated approach combining metabolomics, transcriptomics, and network analysis was employed to identify key signaling pathways and molecular targets. The expression levels of candidate targets were validated by RT-qPCR and Western blotting. Molecular docking simulations were performed to predict the binding affinity between D-limonene and the target proteins. Furthermore, a cellular thermal shift assay (CETSA) was conducted to confirm direct binding and protein stabilization. In parallel, cellular experiments using BRL-3A and HepG2 cells were carried out to assess the effect of D-limonene on lipid accumulation. Results D-limonene treatment significantly ameliorated liver injury, reduced excessive lipid deposition, attenuated inflammation, and suppressed oxidative stress in HFD-induced rats. Integrated multi-omics and network analyses identified the PPAR signaling pathway as the core pathway mediating the regulatory effects of D-limonene on lipid metabolism, with PPARγ and SCD-1 pinpointed as key downstream targets. RT-qPCR and Western blot analyses confirmed that D-limonene significantly downregulated the expression of both PPARγ and SCD-1. Molecular docking simulations revealed stable binding of D-limonene to PPARγ and SCD-1. Importantly, CETSA further demonstrated that D-limonene directly binds to and stabilizes these two proteins. Consistently, cellular assays showed that D-limonene significantly reduced lipid accumulation in BRL-3A and HepG2 cells by inhibiting the PPARγ/SCD-1 axis. Conclusion Collectively, these findings suggest that D-limonene alleviates HFD-induced MASLD, and its protective effects may be mediated, at least in part, through inhibition of the PPARγ/SCD-1 axis. This study provides preliminary evidence supporting the potential development of D-limonene as a targeted therapeutic agent for MASLD. Further studies are warranted to validate these findings and elucidate additional underlying mechanisms.

Nitrogen-doped Ti3C2 MXene quantum dots as an effective FRET ratio fluorometric probe for sensitive detection of Cu2+ and D-PA

Xun Zhou,Juerui Zhang,Dongyan Huang,Yinhui Yi,Kechen Wu,Gangbing Zhu

In this work, a ratiometric fluorescence sensing platform was established to detect Cu 2+ and D-PA ( d -penicillamine) based on nitrogen-doped Ti 3 C 2 MXene quantum dots ( N -MODs) that was prepared via a simple hydrothermal method and exhibited strong fluorescent and photoluminescence performance as well as excellent stability. Since the oxidation reaction between o -phenylenediamine (OPD) and Cu 2+ induced the formation of 2,3-diaminophenazine (ox-OPD) which not only can emerge an emission peak at 570 nm, but also inhibit the fluorescence intensity of N -MQDs at 450 nm, a ratiometric reverse fluorescence sensor via fluorescence resonance energy transfer (FRET) was designed to sensitively detect Cu 2+ , where N -MQDs acted as energy donor and ox-OPD as energy acceptor. More importantly, another considerably interesting phenomenon was that their catalytic oxidation reaction can be restrained in the presence of D-PA because of the coordination of Cu 2+ with D-PA, further triggering the obvious changes in ratio fluorescent signal and color, thus a ratiometric fluorescent sensor of determining D-PA was proposed also in this work. After optimizing various conditions, the ratiometric sensing platform showed rather low detection limits for Cu 2+ (3.0 nM) and D-PA (0.115 μM), coupled with excellent sensitivity and stability.
