Anti-skin-aging effect of epigallocatechin gallate by regulating epidermal growth factor receptor pathway on aging mouse model induced by d-Galactose

Jiming Chen,Yifan Li,Qiangqiang Zhu,Tong Li,Hao Lu,Nan Wei,Yewei Huang,Ruoyu Shi,Xiao Ma,Xuanjun Wang,Jun Sheng

Epigallocatechin gallate(EGCG) is a monomer separated from tea catechins, as an well-known antioxidant, which helps fight wrinkles and rejuvenate skin cells. In this study, we investigated the anti-aging effect of EGCG, and to clarify underlying mechanism of skin aging in a d -galactose-induced aging mouse model. Forty-five male mice were divided into 5 groups and treated with different dose of EGCG, Vitamin C (Vit   C) to mice as a positive control. All groups except vehicle were established aging model induced by d -galactose (200   mg/kg/day) that was subcutaneously injected to mice for 8 weeks. Two weeks after injection of d -galactose, EGCG and Vit C groups were simultaneously administered once a day by subcutaneously inject after 5   h for injecting d -galactose. The results show that EGCG can be absorbed by the skin. Overall, the conditions of the skin of EGCG-treatment groups were improved, the whole structure of skin were better than control groups, and the levels of oxidative stress and the expression of relate with EGFR proteins were significantly higher than control group after EGCG treatment. All these findings suggest that EGCG can resist skin senility effectively. And the EGFR with relate of downstream proteins are implicated in the skin aging.

Dual effects of ascorbic acid on the stability of EGCG by the oxidation product dehydroascorbic acid promoting the oxidation and inhibiting the hydrolysis pathway

Lin Chen,Weiwei Wang,Jianyong Zhang,Hongchun Cui,Dejiang Ni,Heyuan Jiang

A series of incubation systems of pure (−)-Epigallocatechin gallate (EGCG), ascorbic acid (AA) and dehydroascorbic acid (DHAA) at 80 °C were performed to investigated the effect and mechanism of AA on the stability of EGCG. Results shows the dual function of AA, protect action at low concentration and promoting degradation at high concentration, and the critical concentration is about 10 mmol/L. The protective properties of AA due to the reversible reaction from AA to DHAA inhibiting oxidation pathway of EGCG to EGCG quinone or other activated intermediates, and both AA and DHAA can inhibit the hydrolysis of EGCG. The properties of promoting degradation is mainly due to the fact that DHAA, the oxidation product of AA, can react with EGCG to generate some ascorbyl adducts of EGCG. This result is helpful to control the stability of catechins and further clarify the complex interaction on healthy between EGCG and AA.

Antioxidant-Targeting Synergistic Probiotic Delivery System with Improved Probiotics Viability and Intestinal Retention for Ulcerative Colitis Therapy

Xinyi Tang,Abiao Li,Xiaohong Cheng,Ruyi Ren,Tiange Zhao,Xiaoli Zhang,Tianqi Qin,Zhiyong Cui,Yuanyuan Ding,Chenguang Liu

Oral probiotics are a multitarget regulatory strategy for treating ulcerative colitis (UC), but their viability is mainly affected by gastric acid and reactive oxygen species (ROS). Herein, a pH and ROS dual-responsive alginate-based hydrogel facilitating the delivery of epigallocatechin gallate (EGCG)-Mn nanoparticle-modified Lacticaseibacillus rhamnosus GG (LGG@EGCG-Mn) to the colon is constructed to treat UC. EGCG-Mn layer enhances LGG’s oxidative stress resistance by 6.28-fold. The dual-responsive alginate/thioketal hydrogel (SA-tK-Ca2+) protects LGG@EGCG-Mn from gastric acid with a 71.6% survival rate and releases it at colonic lesions, resulting in improved LGG@EGCG-Mn colon retention. In vivo, LGG@EGCG-Mn@SA-tK-Ca2+ effectively prevents and treats UC by increasing the expression of ZO-1 and Occludin to restore the intestinal barrier, mitigating colon inflammation, maintaining intestinal homeostasis, and facilitating the synthesis of short-chain fatty acids. This study overcomes the challenges of probiotic delivery across the barriers of harsh gastric pH and intestinal ROS, offering a more effective probiotic therapy for UC.

Revealing the influence of top-brewing and bottom-brewing methods on the flavor of green tea based on the interaction between EGCG and aroma compounds

Mengke Zheng,Zhongyi Mao,Yinan Yang,Guihong Qi,Peng Li,Yufeng Luo,Donghao Zhang,Jianping Xie,Shihao Sun

The brewing process is a crucial step in determining the flavor profile of green tea infusions. However, the flavor differences arising from top-brewing and bottom-brewing methods remain poorly understood, and the underlying mechanisms are largely unexplored. In this study, GC-MS and sensory analysis were employed to investigate the flavor differences in Shucheng Little Cymbidium tea infusions under varying water temperatures and brewing methods. Subsequently, the variations in major polyphenolic compounds were analyzed, and the potential causes of flavor differences were explained from the perspective of the interactions between EGCG and key aroma compounds. Multivariate statistical analysis revealed that compounds such as geraniol, nonanal, and methyl salicylate serve as important markers for discriminating between different brewing conditions. Moreover, EGCG exhibited a significant binding effect with compounds such as geraniol and nonanal, driven by hydrogen bonding and hydrophobic interactions. This binding may regulate the transfer of aroma compounds from tea leaves to the infusion during brewing, thereby shaping distinct sensory characteristics. These findings provide a molecular-level rationale for empirical techniques in traditional tea art. More importantly, it inspires researchers to pay attention to the crucial role of the tea infusion matrix in determining flavor.

Synergistic therapy for melanoma via bioactive compounds derived metal-phenolic networks

Zhenghao Zhou,Cong Li,Shuai Cui,Shian Sun,Shaowei Zhu,Qing Yu,Ning Ding,Hao Hong,Dongzhi Yang

Chemodynamic therapy (CDT) and photothermal therapy (PTT) are important adjunctive therapeutic approaches for tumors, complementing conventional methods like chemotherapy and radiotherapy. They play a crucial role in inhibiting tumor proliferation and metastasis. In this study, we established a combinational therapy strategy involving PTT and CDT for in situ and metastatic melanoma in experimental murine models. As-prepared S@M−EF system incorporated EGCG, a bioactive natural compound, known for its anti-tumor properties, which coordinated with Fe III to form an acidity-sensitive metal–organic framework (MPN). Sorafenib (SRF) was loaded in the system and released in the tumor microenvironment to work synergistically with Fe III and enhance the ferroptosis effect. Mesoporous polydopamine (MPDA) components inside S@M−EF exhibited photothermal characteristics. We conducted systematic in vitro and in vivo studies to assess the specificity and therapeutic effect of various S@M conjugates. Cellular examination confirmed that tumor cell ferroptosis could be induced and temperature could be increased due to photothermal conversion, and these two effects reinforced with each other. S@M−EF induced higher cell apoptosis compared to single functionalized conjugates (S@M, SRF, or M−EF). Enhanced therapeutic efficacy in melanoma and its metastasis was confirmed in murine models. This tumor microenvironment-responsive nanoplatform, based on natural molecules, combined CDT and PTT, offering a mutually enhanced treatment method for inhibiting tumor proliferation and metastasis.

Mitigation of Cisplatin-Induced Nephrotoxicity and Augmentation of Anticancer Potency via Tea Polyphenol Nanoparticles’ Codelivery of siRNA from CRISPR/Cas9 Screened Targets

Lingjiao Li,Chengyao Feng,Wenchao Zhang,Lin Qi,Binfeng Liu,Hua Wang,Chenbei Li,Zhihong Li,Chao Tu,Wenhu Zhou

Cisplatin, a frontline chemotherapeutic agent against cancer, faces challenges in clinical application due to significant toxicities and suboptimal efficacy. Renal toxicity, a dose-limiting factor of cisplatin, results from multifactorial processes including cisplatin-induced cellular pyroptosis, oxidative damage, and inflammatory responses. Our findings reveal that Tea Polyphenols Nanoparticles (TPNs) derived from Epigallocatechin gallate (EGCG) effectively could address these diverse mechanisms, comprehensively alleviating cisplatin-induced nephrotoxicity. Leveraging TPNs as carriers, chemical conjugation enables the encapsulation of tetravalent cisplatin prodrug, extending its systemic half-life, enhancing tumor tissue accumulation, while simultaneously mitigating renal toxicity. Concurrently, employing a CRISPR/Cas9 kinase library, we identified CSNK2A1 as a target sensitizing tumor cells to cisplatin, enabling specific siRNA sequences to augment cisplatin susceptibility, thereby minimizing the dosage requirement. Benefiting from the versatile carrier properties of TPNs to codeliver cisplatin prodrug and anti-CSNK2A1 siRNA, we developed a codelivery system, Pt-TPNs/siRNA. Pt-TPNs/siRNA not only enhances the anticancer effects but also mitigates cisplatin-induced renal toxicity, achieving efficacy while reducing toxicity. Mechanistic and safety assessments of these nanoparticles were conducted at both cellular and animal levels, opening new avenues for improved clinical utilization of cisplatin.

The Inhibition of LPS-Induced Oxidative Stress and Inflammatory Responses Is Associated with the Protective Effect of (-)-Epigallocatechin-3-Gallate on Bovine Hepatocytes and Murine Liver

Tianle Xu,Run Liu,Hao Zhu,Yu Zhou,Tianxu Pei,Zhangping Yang

This study aimed to evaluate whether (-)-epigallocatechin-3-gallate (EGCG) alleviates hepatic responses to lipopolysaccharide (LPS)-induced inflammation and oxidation. Isolated bovine hepatocytes and BALB/c mice were used for LPS challenge and EGCG pretreatment experiments in vitro and in vivo. LPS-challenged (6 μg/mL) hepatocytes exhibited increased levels of NF-κB (p65 and IκBα) and MAPK (p38, ERK, JNK) phosphorylation as well as increased binding activity of p65 to target pro-inflammatory gene promoters, and these effects were suppressed by pretreatment with 50 μM EGCG. Moreover, the reduction in Nrf2 signaling and antioxidant enzyme activities induced by LPS stimulation were reversed upon EGCG treatment. In vivo experiments demonstrated the protective role of EGCG in response to GalN/LPS-induced mortality and oxidative damage. Together, our results suggest that EGCG is hepatoprotective via inhibition of MAPK/NF-κB signaling and activation of the Nrf2 cascade. This information might help design strategies for counteracting hepatitis in ruminants and monogastric animals. Keywords:EGCG;bovine hepatocytes;hepatoprotective;MAPK/NF-κB signaling;antioxidant activity

A Multifunctional Coating Strategy for Promotion of Immunomodulatory and Osteo/Angio-Genic Activity

Shiqi Xiao,Jiawei Wei,Shue Jin,Xue Xia,Li Yuan,Qin Zou,Yi Zuo,Jidong Li,Yubao Li

Building of multifunctional coatings in a more effective way is crucial for meeting the multilevel requirements of regenerative medicine. Herein, inspired by diatom and mussel, a specific but universal approach is proposed for building multifunctional coatings on slow-degradable and fast-degradable scaffolds or various substrates by using epigallocatechin gallate (EGCG) and polyethyleneimine (PEI) as bridges of silicon coupling. The results reveal that the polyphenol EGCG facilitates silica precipitation and coating topological morphology in synergy with PEI, and realizes antioxidant and immunomodulatory effects. The introduction of EGCG and the release of silicon ions present effective modulation of the immune microenvironment and remarkable promotion of angiogenesis and osteogenesis. The EGCG/silica coating strategy demonstrates a promising perspective for designing multifunctional coatings and optimizing tissue regeneration and reconstruction.

Effects of ultrasound-assisted modification during freeze-thaw cycle on gel properties and conformation of myosin in EGCG-modified systems: Roles of ionic strength and ultrasound power

Xi Zhou,Yatong Wang,Jianmei Zheng,Jun Qi,Zhichao Xiao,Lin Chen,Guoqiang Zhang,Xianchao Feng

This study investigated effects of ultrasound power and ionic strength on conformation and gel properties of chicken myosin under freeze-thaw (FT) cycle in EGCG-mediated system. Ultrasound significantly increased solubility, α -helix content, and surface hydrophobicity, while reduced turbidity, particle size, sulfhydryl content, and intrinsic fluorescence ( P  < 0.05), indicating that mechanical forces induced by ultrasound promoted structural unfolding, which increased disulfide bonding and hydrophobic interactions, thereby enhancing myosin-myosin and myosin-EGCG interactions. Ultrasound significantly increased β -sheet, viscosity, gel strength, and ratio of immobile water, while reduced α-helix content and thawing loss of frozen myosin gel ( P  < 0.05). SEM, LF-NMR, and MRI analyses indicated that ultrasound facilitated formation of a compact network structure of myosin gel after FT cycle, significantly increasing water-holding capacity (WHC) of frozen myosin gel. WHC of myosin gel at 0.6 M NaCI was better than that at 0.3 M NaCI, resulting in higher gel strength and thawing loss.

Nanogels of dual inhibitor-modified hyaluronic acid function as a potent inhibitor of amyloid β-protein aggregation and cytotoxicity

Jiang Zhiqiang,Dong Xiaoyan,Yan Xin,Liu Yang,Zhang Lin,Sun Yan

Inhibition of amyloid β-protein (Aβ) aggregation is considered as a promising strategy for the prevention and treatment of Alzheimer’s disease. Epigallocatechin-3-gallate (EGCG) and curcumin have been recognized as effective inhibitors of Aβ aggregation. Herein, we proposed dual-inhibitor modification of hyaluronic acid (HA) to explore the synergistic effect of the two inhibitors. EGCG-modified HA (EHA) formed dispersed hydrogel structures, while EGCG-curcumin bi-modified HA (CEHA) self-assembled into nanogels like curcumin-modified HA (CHA). Thioflavin T fluorescent assays revealed that the inhibitory effect of CEHA was 69% and 55% higher than EHA and CHA, respectively, and cytotoxicity assays showed that the viability of SH-SY5Y cells incubated with Aβ and CEHA was 28% higher than that with Aβ and the mixture of EHA and CHA. These results clearly indicate the synergism of the two inhibitors. It is considered that the difference in the hydrophobicities of the two inhibitors made the bi-modification of HA provide a favorable CEHA nanostructure that coordinated different inhibition effects of the two inhibitors. This research indicates that fabrication of dual-inhibitor nanosystem is promising for the development of potent agents against Aβ aggregation and cytotoxicity.

Interactions between β-cyclodextrin and tea catechins, and potential anti-osteoclastogenesis activity of the (−)-epigallocatechin-3-gallate–β-cyclodextrin complex

Huanhuan Xu,Titi Liu,Jing Xu,Jin Li,Fei Chen,Zemin Xiang,Yewei Huang,Dongying Zhang,Lihong Hu,Banglei Zhang,Chengting Zi,Xuanjun Wang,Jun Sheng

Galloylated catechins, the most important secondary metabolites in green tea including (−)-epigallocatechin-3-gallate (EGCG) and (−)-epicatechin-3-gallate, constitute nearly 75% of all tea catechins and have stronger health effects than non-galloylated catechins such as (−)-epigallocatechin and (−)-epicatechin. EGCG is the most abundant, active, and thoroughly investigated compound in green tea, and its bioactivity might be improved by complexing with β-cyclodextrin (β-CD). We investigated interactions between four catechins and β-CD in a PBS buffer solution of pH 6.5 at 25 °C using biolayer interferometry and isothermal titration calorimetry, and to determine whether β-CD could enhance the anti-osteoclastogenesis effect of EGCG. β-CD could directly bind galloylated catechins at a stoichiometric ratio close to 1 : 1, with high specificities and affinities, and these inclusion interactions were primarily enthalpy-driven processes. We synthesized the EGCG–β-CD complex and identified it using infrared radiation and nuclear magnetic resonance spectra. Interestingly, we revealed that the EGCG–β-CD complex could inhibit osteoclastogenesis significantly more than EGCG.

Photothermal nanoagent for anti-inflammation through macrophage repolarization following antibacterial therapy

Yueying Xu,Yanjun Cai,Yu Xia,Quanxin Wu,Mingen Li,Ning Guo,Yingfeng Tu,Bin Yang,Yun Liu

Owing to the ignorable bacterial resistance, photothermal therapy (PTT) has been widely studied for bacterial infection. However, few research efforts have focused primarily on the simultaneous utilization of photothermal agents (PTAs) for anti-inflammatory therapy following PTT. Here, we developed a nano-sized PTA ( [email protected] ) through the self-assembly of Fe 3+ and epigallocatechin-3-gallate (EGCG) by using BSA as the nanoreactor and colloidal stabilizer. Next, we assessed its photothermal and reactive oxygen species (ROS) scavenging properties. In addition, we evaluated its antibacterial and anti-inflammatory abilities through in vitro antibacterial and cellular assays, respectively. Finally, we tested its in vivo sterilization and anti-inflammatory effects. Firstly, [email protected] displayed good photothermal capacity due to the inherent photothermal ability of metal-phenolic networks (MPNs). Consequently, [email protected] showed excellent both in vitro and in vivo antibacterial capacities. Moreover, [email protected] exhibited outstanding anti-oxidative property owing to ROS scavenging effect of EGCG. As a result, it presented extraordinary both in vitro and in vivo anti-inflammatory efficiencies through promoting the repolarization of pro-inflammatory (M1) macrophages to anti-inflammatory (M2) macrophages with the released EGCG. The research findings indicated that the prepared nanoagent has great potential to be used for combined antibacterial and anti-inflammatory treatments in future clinical applications.
