Due to the very important role in physiological process, a simple and sensitive hemin detection method is necessarily required. Biomass-based carbonized polymer dots (CPDs) have been widely studied especially as fluorescence probe owing to the advantages of low toxicity and the variety of fluorescence color, yet there are still challenges in developing their multi-color emission property from the same raw materials. In this work, red, white and blue emissive CPDs derived from chlorophyll have been synthesized via hydrothermal method. Then white-emitted CPDs (white-CPDs) with the Commission International d’Eclairage (CIE) coordinates at (0.34, 0.32) were used to develop a fluorescence quenched sensing system for hemin determination. There is a good linear relationship between (F 0 -F)/F 0 and concentration of hemin in the range of 0.1–0.95 μM with a detection limit of 0.043 μM, and the quenching mechanism was considered to be caused by inner filter effect (IFE). Moreover, it has been successfully used for hemin detection in serum and also for visual determination, which indicating great potential in applications of disease diagnoses and trace identification.

Overuse and abuse of morphine (MOP), one of the main components of pericarpium papaveris, have attracted increasing attention in the medical field owing to its pharmacological and toxicological activity. Herein, we proposed a new electrochemical nano-biosensor for MOP detection based on surface-confined building of Au@Pt-centered and multi-G-quadruplex/hemin wire-surrounded electroactive super-nanostructures. The center Au@Pt was flower-shaped and irregularly protruded, allowing substantial loading of multiple G-quadruplex wire/hemin complexes on its surface to accomplish the assembly of electroactive super-nanostructures. Interestingly, as the super-nanostructures were closely confined on the electrode surface, a significantly amplified electrochemical signal was thus obtained in the absence of MOP. In contrast, the introduction of target MOP can induce an intense competitive effect and strongly destroy the assembly process, resulting in the reduction of the electrochemical response that is correlated with the logarithmic concentration of MOP. Under optimal conditions, the electrochemical nano-biosensor is capable of highly sensitive detection of MOP in a dynamic concentration range from 1 ppt to 500 ppb. The limit of detection is achieved as low as 0.69 ppt, and the practical application was confirmed by examining MOP from chafing dish condiments. We expect the electrochemical platform utilizing this unique nanoarchitecture to provide rational guidelines to design high-performance analytical tools.

When photoelectrocatalysis (PEC) couples with electroenzymatic catalysis (EEC), effective utilization of electrons from PEC process for yield of H 2 O 2 plays an important role in the enzymatic catalysis. In the present work, we designed a novel dual-cathode PEC-EEC system for pollutant removal. In this system, BiVO 4 photoanode was prepared by electrodeposition for visible light-driven PEC process while carbon cloth was used as the first cathode for electrogeneration of H 2 O 2 and hemin/Cu prepared by electrophoretic deposition acted as the second cathode for enzymatic catalysis. The performances of the constructed dual-cathode PEC-EEC system were evaluated by decoloring Rhodamine B (RhB), which showed high decoloration percentage with acceptable stability and reusability. Further, the system was applied to degradation of tetracycline (TC), indicating that about 93.6% of TC was removed after 2-h treatment. A pathway for TC degradation in such a dual-cathode PEC-EEC system was proposed based on the intermediates determined by liquid chromatography coupled to mass spectrometry in tandem (LC-ESI-MS/MS).

Multi-emissive red fluorescence carbon dots (R-CDs) were synthesized using Stellaria pallida and urea precursors through a solvothermal method, based on which a ratiometric fluorescent sensor was constructed and integrated into a portable dual-mode sensing platform for determining the residual levels of Chlortetracycline Hydrochloride (CTC) and Hemin Chloride (Hemin) in food products. The ratiometric fluorescent sensor achieved limits of detection (LODs) were 13.00 × 10 −3 μM for CTC and 0.74 × 10 −3 μM for Hemin, while the visual detection platform provided LODs of 1.50 × 10 −3 μM and 0.53 × 10 −3 μM, respectively. The relative standard deviation of the spike recovery tests ranged from 0.69% to 1.12%. This work presents a green, low-cost, and highly sensitive dual-analyte sensing strategy combined with a visual readout, offering a reliable platform for on-site food safety monitoring.

Here, taking the advantage of single-stranded (ss) DNA specific nuclease (S1) and peptide nucleic acid (PNA), we demonstrated a novel, rapid, and label-free colorimetric nanosensor for the sensitive and accurate detection of SNPs based on the intrinsic peroxidase-like activity of hemin-functionalized single-walled carbon nanotubes (hemin-SWCNTs). PNA, a man-made mimic of DNA with extraordinary stability toward enzymatic degradation, can effectively protect DNA in the fully matched DNA/PNA duplexes from nuclease digestion. While the DNA in DNA/PNA duplexes containing a mismatch can be cleaved into small fragments. This difference can be visually monitored from the specific color change of TMB/H 2 O 2 system by employing the peroxidase activity of hemin-SWCNTs because of its different aggregation states responding to ssPNA or DNA/PNA duplex. Under optimized conditions, the SNPs in the human tumor suppressor gene TP53 have been successfully genotyped in a linear range of 50–1000 nM with a detection limit of 0.11 nM. Moreover, this platform can effectively discriminate a series of single-base mismatches. This assay avoids the assistance of sophisticated instruments and complicated modifications of probes or nanomaterials , and function well for both cell lysate samples and PCR amplicons from standard cell lines, implying its potential practical applications for bioanalysis and biosensors.

In this work, a novel Pb 2+ electrochemical DNAzyme sensor was developed for ultrasensitive detection of lead ions (Pb 2+ ) in water environment by coupling with the MoS 2 –AuPt nanomaterials and hemin/G-quadruplex DNAzyme, which acting as the electrocatalytic signal tag. Streptavidin (SA) modified tin dioxide-functionalized reduced graphene oxide (rGO-SnO 2 ) /gold nanoparticles (AuNPs) served as a sensor platform for enhancing conductivity and immobilizing more Pb 2+ -specific DNAzyme. In the presence of Pb 2+ , the Pb 2+ -dependent DNAzyme specifically reacted with Pb 2+ , cleaving the substrate strand (SS) into two free fragment and releasing the biotin-modified enzyme strand (Bio-ES) on the electrode. Connecting MoS 2 –AuPt nanocomposites labeled with G-rich DNA (G-DNA) strand and exposure of Bio-ES through the Helper DNA, as well as adding hemin to form a hemin/G-quadruplex, the biosensor achieved signal amplification . Chronoamperometry was used to record the current signal, which was primarily derived from the cocatalysis reduction of H 2 O 2 by MoS 2 –AuPt nanocomposites and the hemin/G-quadruplex. Under optimal conditions, the designed biosensor exhibited sensitive detection of Pb 2+ from 0.1 pg mL −1 to 1000 ng mL −1 , with a lower detection limit of 38 fg mL −1 (based on 3σ). This proposed biosensor is ultrasensitive and specific, representing a potential application for the detection of Pb 2+ in a water environment.

A novel multiple amplified enzyme-free immunosensor was developed for competitive immunoassay of microcystin-LR (MC-LR). Classical electrochemical immunosensors usually employ enzymes as biocatalysts to afford amplified signals, but the proteolytic degradation and poor stability are still crucial problem. Herein, monodisperse core-shell mesoporous silica (SiO 2 @MSN)-functionalized DNAzyme concatamers were synthesized to load hemin and methylene blue (MB) as the mimic enzyme . Firstly, the surface of SiO 2 @MSN was conjugated with secondary antibody as the recognition of MC-LR antibody and with a DNA strand as the initiator. Two auxiliary DNA strands were then selected for the in-situ propagation to form a double-helix DNA through hybridization chain reaction (HCR), forming numerous DNAzymes (G-quadruplex/hemin) after the addition of hemin. Secondly, MB was inserted into the formed double-helix DNA, and also loaded in the brush-like structure of mesoporous SiO 2 @MSN. The molecular docking study showed that electrons can transfer more effectively with π-π stack of hemin/G-quadruplex and intercalation of MB/DNA, thus the catalytic ability of DNAzymes can be greatly improved. With the aid of MB, DNAzymes can catalyze the reduction of H 2 O 2 to produce the electrochemical signal. This enzyme-free electrochemical immunosensor can successfully detect MC-LR in a range of 0.5 ng/L and 25 μg/L with a detection limit of 0.3 ng/L. This stable, sensitive and selective nonenzymatic electrochemical immunoassay shows promise for applications in food and environmental monitoring.

A toehold-aided DNA recycling amplification technology was developed based on the combination of toehold-aided DNA recycling and the hemin/G-quadruplex label. The dsDNA formed between aptamer and DNA1 was first immobilized on magnetic beads. On addition of target analyte (exemplified here for riboflavin), the aptamer-riboflavin complex is formed and DNA1 is released by the beads. After magnetic separation, the supernatant containing the released DNA1 is added to a solution containing the hairpin capture DNA on magnetic beads. DNA1 will hybridize with the hairpin capture DNA via toehold binding and branch migration. This process will open the hairpin structure, and an external toehold is formed in the newly formed dsDNA. On addition of reporter DNA containing the G-quadruplex, it will interact with the formed dsDNA via toehold binding and branch migration, resulting in the releasing of DNA1 and capturing of reporter DNA on the magnetic beads. The released DNA1 will bind to another hairpin capture DNA which can start another round of DNA1 recycling. Chemiluminescence (CL) is generated by the G-quadruplex-hemin-luminol CL reaction system. Under optimal conditions, the calibration plot is linear in the 0.1 to 700 nM riboflavin concentration range, with a 30 pM detection limit (at a signal-to-noise ratio of 3). The method was successfully applied to the quantitation of riboflavin in spiked urine samples.

A new signal-amplification strategy based on copper(II) (Cu 2+ )-dependent DNAzyme was developed for sensitive impedimetric biosensing of Cu 2+ in aqueous solution by coupling with target-induced formation of hemin/G-quadruplex-based DNAzyme and enzymatic catalytic precipitation technique. Initially, the target analyte cleaved the Cu 2+ -specific DNAzyme to generate an initiator strand on the sensor. Thereafter, the initiator strand underwent an unbiased strand-displacement reaction between hairpin probes in turn to construct a nicked double-helix, accompanying the formation of hemin/G-quadruplex DNAzyme on the long duplex DNA . The newly formed DNAzyme could oxidize the 4-chloro-1-naphthol (4-CN) to produce an insoluble precipitation on the sensor, thus resulting in a local alteration of the conductivity. Under the optimal conditions, the resistance increased with the increasing Cu 2+ in the sample, and exhibited a wide dynamic working range from 0.1 pM to 5.0 nM with a detection limit of 60 fM. The methodology also displayed a high selectivity for Cu 2+ relative to other potentially interfering ions owing to the highly specific Cu 2+ -dependent DNAzyme, and was applicable for monitoring Cu 2+ in real river samples. Thus, our strategy has a good potential in the environment surveys.

Monitoring beta-amyloid 1 – 42 (Aβ 42 ) is vital and challenging, which is a typical biomarker of Alzheimer's disease. Here, a novel electrochemical microbiosensor is developed to detect Aβ 42 on an acupuncture needle. Hemin is well known for its characteristics, including its ability to self-assemble on single-walled carbon nanotube (SWCNT), the molecular interaction with Aβ 42 , and the intrinsic electroactive signal. These properties are exploited to anchor and respond to Aβ 42 after integrating a molecularly imprinted surface polymer (SMIP). The SMIP layer of polydopamine/poly (ionic liquid) can be prepared by electropolymerization on an acupuncture needle microelectrode (ANME), which undergoes growth and formation of a polymeric structure around the anchored Aβ 42 . Interestingly, the imprinted cavities express a fluent signal of built-in hemin after eluting the templates, and show a highly selective and sensitive hindrance response for the recombined Aβ 42 . Under optimized conditions, the microbiosensor displays a linear range of 100 to 1 × 10 10 fM with a limit of detection of 0.05 fM. There are development and advances for the discipline of electroanalysis after comparing the technique and important indicators with the electrochemical biosensors reported of Aβ 42 . The microbiosensor also exhibited excellent selectivity, good stability, and reproducibility, which was effectively used to detect Aβ 42 in real spiked samples. The improved behavior of the developed microbiosensor can be attributed to its superficial highly matched imprinted cavities, built-in hemin label, and electronic barrier without signal of the nonimprinted surface to outside molecules. This microbiosensor has a scientific and reference value for directly sensing non-electroactive biomarkers, functionalizing microelectrodes, and electron transport cavities. It would also be amazing if this new microbiosensor could combine with the unclear and magical property of acupuncture in the treatment of neurological disorders.

G-quadruplex/hemin DNAzyme (G-DNAzyme) is widely utilized in biosensors due to its ability to catalyze peroxidase-like reactions. However, its relatively low catalytic efficiency can limit detection sensitivity. To address this limitation and enable trace detection, this study presents a G-DNAzyme-mediated biosensor that combines entropy-driven catalytic reactions (EDC) and catalytic hairpin assembly (CHA) to enhance sensitivity and broaden its applicability for detecting trace levels of aflatoxin B1 (AFB1). In this biosensor, AFB1 specifically binds to aptamer, triggering the release of a complementary initiator chain, which activates both EDC and CHA amplification processes. This dual amplification results in the generation of numerous free G-quadruplex sequences, which then bind to hemin, catalyzing a color change in the chromogenic substrate and significantly amplifying the signal. The biosensor demonstrated excellent linearity between the absorbance intensity and logarithm of AFB1 concentrations, with a detection limit (LOD) of 0.47 pM. Furthermore, spiked recovery tests of AFB1 in actual samples showed recovery rates between 96.12 % and 108.50 %, indicating strong accuracy. The method is simple, rapid, and does not require complex instrumentation, making it an efficient tool for detecting AFB1 in food safety applications.

Hemin as the organic linker ligand and Cu (II) as the metal center were applied to prepare a copper-metal-organic framework (Cu-hemin-MOF) via one-step hydrothermal method. Characterization using scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FT-IR), X-ray photoelectron spectroscopy (XPS), and X-ray powder diffraction (XRD) demonstrate that the acquired Cu-hemin-MOF possesses the appearance of 3D ball-flower shape with the existence of C, N, O, Fe, and Cu on the surface. Further study found that this 3D ball-flower shaped Cu-hemin-MOF exhibits peroxidase-like activity, which can catalyze the peroxidase substrate of o-phenylenediamine (OPD) to generate 2,3-diaminophenazine (DAP) in the presence of H 2 O 2 . DAP has a yellow color and also emits a strong fluorescence when excited by ultraviolet light. Interestingly, Cu-hemin-MOF’s peroxidase-like activity can be strongly inhibited by glutathione (GSH). In view of this, a dual readout (fluorescence and colorimetry) was proposed to detect GSH for the first time. Under optimal conditions, the proposed method exhibits good linear relationship between the signal response (fluorescence and colorimetry) and the concentration of GSH, and low limits of detection (LOD) of 2.3 and 26.6 nM, respectively.
