Osteoporosis (OP) is typically brought on by disruption of bone homeostasis. Excessive oxidative stress and mitochondrial dysfunction are believed to be the primary mechanisms underlying this disorder. Therefore, in order to restore bone homeostasis effectively, targeted treatment of oxidative stress and mitochondrial dysfunction is necessary. Cinnamaldehyde (CIN), a small molecule that acts as an agonist for the nuclear factor erythroid 2-related factor (Nrf2), has been found to possess antiapoptotic, anti-inflammatory, and antioxidant properties. We found that CIN, while rescuing apoptosis, can also reduce the accumulation of reactive oxygen species (ROS) to improve mitochondrial dysfunction and thus restore the osteogenic differentiation potential of BMSCs disrupted by hydrogen peroxide (H2O2) exposure. The role of CIN was preliminarily considered to be a consequence of Nrf2/HO-1 axis activation. The ovariectomized mice model further demonstrated that CIN treatment ameliorated oxidative stress in vivo, partially reversing OVX-induced bone loss. This improvement was seen in the trabecular microarchitecture and bone biochemical indices. However, when ML385 was concurrently injected with CIN, the positive effects of CIN were largely blocked. In conclusion, this study sheds light on the intrinsic mechanisms by which CIN regulates BMSCs and highlights the potential therapeutic applications of these findings in the treatment of osteoporosis.

The low-usage of solar energy and the sluggish separation efficiency of the photogenerated electrons/holes pairs are the obstacles in the practical application of photocatalysts. The integration of upconversion and Z-scheme heterojunction is expected to break the barriers to achieve the efficient charge separation and broaden near-infrared light absorption . Herein, an effective indirect Z scheme AgInS 2 /In 2 S 3 heterostructure with carbon quantum dots (CQDs, as the electron conduction medium) and Lu 3 NbO 7 :Yb, Ho (as upconversion function) has been successfully synthesized. Consequently, the Lu 3 NbO 7 : Yb, Ho/CQDs/AgInS 2 /In 2 S 3 heterostructure exhibited superior photocatalytic activities for Cr(VI) reduction and H 2 O 2 production, reducing 99.9% of Cr(VI)(20 ppm, 15 min) and 78.5% of Cr(VI) (40 ppm, 30 min) with visible light irradiation as well as 94.0% of Cr(VI) (20 ppm, 39 min) under NIR light irradiation. Simultaneously, the heterostructure could generate 902.9 μM H 2 O 2 for 5 h under visible light irradiation. The intensive photocatalytic properties could primarily be attributed to the boosted light absorption capacity, the improved solar-to-energy conversion by the remarkable upconversion capacity of Lu 3 NbO 7 : Yb, Ho/CQDs and the faster charge transfer through a Z-schematic pathway. This work is anticipated to open a novel “window” for designing the efficient photocatalysts by coupling of Lu 3 NbO 7 : Yb, Ho and CQDs.

The MFeO 3 : R 3+ (M = La, Gd; R = Eu, Er, Ho) nanocrystalline powders were successfully synthesized by the sol–gel combustion method using glycine-based gel at a low ignition temperature. The as-burnt products present honeycomb-like structure with many irregular continuous pores due to the releasing of a large amount of gas during the combustion reaction process. The lattice parameters and cell volumes of MFeO 3 :R 3+ (M = La, Gd; R = Eu, Er, Ho) nanocrystalline powders are compared in detail. The magnetic properties of all the samples were studied systematically. The results show that the doping of rare-earth ions enhanced greatly the coercivity of GdFeO 3 and LaFeO 3 as a result of the enhancement of magnetic anisotropy . The influence of doped rare-earth ions on the magnetic properties was summarized. The distinction of magnetism of MFeO 3 :R 3+ (M = La, Gd; R = Eu, Er, Ho) were explained in detail by the intrinsic magnetism of doped rare-earth ions. This work is significant for the application of rare-earth elements in other magnetic materials.

Cadmium (Cd) is a nonessential metal that is a contaminant in aquatic ecosystems . Cd can accumulate in aquatic animals, leading to detrimental effects in tissues, and Cd exposure can induce immunotoxicity in fish. MicroRNAs (miRNAs) play critical roles in immune responses, yet the participation of miRNAs in Cd-induced immunotoxicity remains poorly understood. The present study evaluated the effects of Cd exposure on the immune responses and the mRNAs and miRNAs expressions of immune-related genes in Cyprinus carpio ( C. carpio ). Then, microRNA-155 (miR-155) was overexpressed and microRNA-181a (miR-181a) was knocked down to determine which miRNA plays a key role in the immune response to Cd. The results showed that 0.5 mg/L Cd 2+ significantly decreased the activity of alkaline phosphatase (AKP) and acid phosphatase (ACP) in the kidneys of C. carpio . Cd exposure upregulated the mRNA expressions of interleukin (IL)-1β, IL-8, nuclear factor-kappa B (NF-κB), tumour necrosis factor-α (TNF-α), and Toll-like receptor 4(TLR-4) and downregulated those of IL-10 and heme oxygenase-1 (HO-1) in C. carpio kidneys. Cd exposure also led to upregulation of miR-155 and miR-181a expressions. Furthermore, AKP and ACP activity in the kidneys was markedly changed after intraperitoneal injection of C. carpio with miR-155 agomir and miR-181a antagomir . All detected mRNA expressions were significantly decreased after injection of miR-155 agomir, and IL-10, NF-κB, TNF-α, and HO-1 mRNA expressions were markedly increased after injection of miR-181a antagomir. The results of this study demonstrate that Cd exposure can immunocompromise C. carpio by targeting HO-1 through miR-155 and miR-181a. This is the first study to reveal that Cd exposure induces immunotoxicity through miR-155 and miR-181a in the kidneys of C. carpio .

Rare earth-doped upconversion nanoparticles have promising potential in the field of pesticide detection because of their unique frequency upconverting capability and high detection sensitivity. This paper reports a novel aptamer-based nanosensor for acetamiprid detection using fluorescence resonance energy transfer (FRET) between NH 2 -NaYF 4 : Yb, [email protected] 2 (UCNPs) and gold nanoparticles (GNPs). Herein, GNPs as acceptors efficiently quench the fluorescence of UCNPs and acetamiprid specifically interacts with acetamiprid binding aptamer (ABA), causing the conformation changes of ABA from random coil to hairpin structure. Accordingly, ABA no longer stabilizes the GNPs in salt solution, leading to the varying aggregation extent of GNPs. Thus, the fluorescence of UCNPs are proportionally recovered. Under the optimized conditions, the enhancement efficiency was observed to increase linearly with the concentration of acetamiprid from 50 nM to 1000 nM, resulting in a relatively low limit of 3.2 nM. Additionally, the aptasensor demonstrated high selectivity to similar structure pesticides such as imidacloprid and chlorpyrifos , and further confirmed its application capacity in adulterated tea samples.

This study synthesized La0.9X0.1AlO3 (X = Eu, Gd, Ho, Tm) and LaAlO3 ceramics by a solid-state method. The characterization of the ceramics revealed that the high degree of lattice distortion induced by variations in the size, mass, and electronegativity of rare-earth elements enhances high-temperature stability, leading to aging drift rates between 0.4% and 2.3% after 500 h at 1000 °C. Furthermore, the emergence of intrinsic defects and dislocations alters the charge conduction mode in LaAlO3, resulting in significant linearity at temperatures above 1000 K. This study further elucidates the transition in the charge conduction mechanism from thermally activated conduction to hopping conduction, which is driven primarily by oxygen vacancies and dislocations. DFT calculations support these findings, demonstrating a reduction in the band gap and increased carrier concentration. This work paves the way for the development of advanced NTC thermistors for reliable temperature monitoring in challenging environments.

Pulmonary fibrosis (PF) is a chronic and progressive lung disease, and oxidative stress plays a critical role in its pathogenesis. Rosa roxburghii Tratt, known for its anti-inflammatory and antioxidant properties, has been shown to alleviate fibrosis. This study aimed to explore whether two Rosa roxburghii fermentation broths (RRFBs) (with different proportions) could attenuate bleomycin (BLM)-induced PF in mice and to elucidate the molecular mechanisms. The results revealed that RRFBs reduced structural lung damage, collagen deposition, and lung inflammation. RRFBs also suppressed fibrotic markers (Collagen I, Vimentin, and α-SMA) while enhancing epithelial marker E-cadherin expression. Additionally, RRFBs alleviated BLM-induced oxidative stress and apoptosis by activating the Nrf2/HO-1/NQO1 signaling pathway and facilitating Nrf2 nuclear translocation. Furthermore, RRFBs attenuated the BLM-induced changes in the gut microbiota; in particular, they decreased the abundance of the pathogenic bacterium Proteus and increased the abundance of the probiotics Ileibacterium and Dubosiella . Spearman correlation analysis revealed a strong association between oxidative stress inhibition and gut microbiota composition. These results indicated that RRFBs could exert lung-protective effects by inhibiting oxidative stress and alleviating intestinal disturbances.

Current therapeutic options for Parkinson's disease (PD) are limited in efficacy and burdened with severe side-effects. Inflammation, intertwined with oxidative stress and influenced by gut microbiota-related changes making the gut-brain axis the main target of innovative therapies. This study reveals the multiple protective effects of polysaccharides extracted from Ulva lactuca (UPE) on PD models. In vitro, UPE was shown to have direct cytoprotective effects, including mitigating 1-methyl-4-phenylpyridinium-induced cell apoptosis, reducing reactive oxygen species levels, and maintaining mitochondrial membrane potential. In vivo, oral UPE significantly restored motor deficits and dopaminergic neuron loss caused by MPTP. These in vivo benefits were associated with the inhibition of oxidative stress and inflammation. Moreover, oral UPE inhibits the excessive activation of glial cells and the excessive production of pro-inflammatory factors and oxidative stress markers in serum. Furthermore, 16S rRNA analysis of gut microbiota revealed that UPE reshaped the gut microbiota of PD mice by increasing the abundance of norank_f_Murbaculaceae and decreasing the abundance of Desulfovibrio . Mechanically, the Nrf2/HO-1 and TLR4/NF-κB signaling pathways are involved in the anti-PD effect of UPE. In general, UPE offers new perspectives on PD treatment and demonstrates the potential of natural polysaccharides in treating neurodegenerative diseases.

Non-small cell lung cancer (NSCLC) is the most common type of lung cancer. (-)-Epigallocatechin-3-gallate (EGCG), a major polyphenol in green tea, is widely studied as a cancer chemopreventive agent with potential anti-cancer effects. The NF-E2-related factor 2 (Nrf2)/heme oxygenase-1 (HO-1) signaling pathway is considered to mediate cellular resistance to EGCG. Metformin, a classical antidiabetic drug, has been shown to prevent cancer progression. Researchers have not reported whether metformin potentiates the anti-cancer efficacy of EGCG. In this study, metformin inhibited HO-1 expression and augmented the anti-tumor effect of EGCG. Metformin also enhanced ROS (reactive oxygen species) generation induced by EGCG (100 μM), subsequently resulting in apoptosis. Based on the results of the in vivo study, size of xenografts treated with the combination of metformin and EGCG was smaller than other groups. Mechanistically, metformin modulated the EGCG-activated Nrf2/HO-1 pathway through Sirtuin 1 (SIRT1)-dependent deacetylation of Nrf2. Moreover, metformin upregulated SIRT1 expression partially through the NF-kB pathway. Comparatively, the combination of EGCG and metformin showed little impact on normal lung epithelial BEAS-2B cells. Based on our findings, metformin sensitized NSCLC cells to the EGCG treatment by suppressing the Nrf2/HO-1 signaling pathway.

Ethnopharmacological relevance ShengHui decoction (SHD), a traditional Chinese herbal formula, has long been used clinically to mitigate Alzheimer’s disease (AD), demonstrating significant efficacy in alleviating cognitive impairment. However, its molecular mechanisms remain insufficiently understood. Aim of the study To clarify the effects of SHD on oxidative stress–related markers and inflammatory cytokines in an aluminum chloride (AlCl 3 )-induced zebrafish model of AD and as well as uncover its underlying mechanisms in alleviating oxidative stress and neuroinflammation.. Materials and methods Ultra-performance liquid chromatography–tandem mass spectrometry (UPLC-MS/MS) was employed to identify the bioactive constituents of SHD. Behavioral tests and histopathological analyses were then conducted to evaluate the effects of SHD (0.74, 1.48, and 2.96 mg/mL) on apoptosis and amyloid-beta (Aβ) deposition in an AlCl 3 -induced AD zebrafish model. Biochemical assays and enzyme-linked immunosorbent assay were used to quantify superoxide dismutase (SOD), malondialdehyde (MDA), glutathione peroxidase (GSH-Px), tumor necrosis factor-alpha (TNF-α), interleukin-6 (IL-6), IL-1 beta (IL-1β), and IL-18.. Western blot analysis was performed to determine the protein expression of nuclear factor erythroid 2–related factor 2 (Nrf2), heme oxygenase-1 (HO-1), NAD(P)H quinone dehydrogenase 1 (NQO1), nuclear factor-kappa B p65 (NF-κB p65), phosphorylated NF-κB p65 (p–NF-κB p65), and NOD-like receptor family pyrin domain containing 3 (NLRP3). Results UPLC–MS/MS profiling identified phenolic acids, saponins, lignans/coumarins, flavonoids, and aliphatic alcohols as the main constituents of SHD. SHD significantly ameliorated cognitive deficits and histopathological abnormalities in AlCl 3 -induced zebrafish. Moreover, SHD attenuated oxidative stress by upregulating Nrf2, HO-1, and NQO1 expression, while downregulating AlCl 3 -induced levels of NF-κB p65, p–NF-κB p65, NLRP3 and pro-inflammatory factors. Conclusions SHD effectively improved cognitive performance and reduced AD-like pathological lesions in AlCl 3 -induced zebrafish. Importantly, SHD exhibits both antioxidant and anti-inflammatory activities by activating the Nrf2/HO-1 axis and inhibiting NF-κB signaling, thereby alleviating oxidative stress and neuroinflammatory responses in AD. These findings provide experimental evidence supporting SHD as a potential therapeutic agent for AD, and substantiate the traditional Chinese medicine principle of tonifying the kidney and replenishing essence in the treatment of AD.

In this study, a series of high-entropy rare-earth zirconate (La 0.2 Nd 0.2 Sm 0.2 Eu 0.2 A 0.2 ) 2 Zr 2 O 7 (A = Dy, Ho, or Er) ceramics containing five principal elements were synthesized via the high-temperature solid-state method, and their potential as high-temperature thermosensitive ceramics was investigated. X-ray diffraction and Raman spectroscopy confirmed the single-phase pyrochlore structural characteristics of these ceramics, while scanning electron microscopy revealed a dense microstructure. Electrical measurements demonstrated that these ceramics are capable of operating at temperatures up to 1500 °C and exhibiting exceptional sensitivity within the 400–1500 °C range, both of which are attributed to their high resistivity at elevated temperatures and a large material constant B value (≥11,241 K) across the entire operating temperature span. Notably, (La 0.2 Nd 0.2 Sm 0.2 Eu 0.2 A 0.2 ) 2 Zr 2 O 7 (A = Dy, Ho, or Er) ceramics also maintained a resistivity drift rate of ≤0.53 % after 500 h of continuous exposure at 1500 °C, a result ascribed to the high-entropy effect suppressing lattice distortion, as evidenced by ab initio molecular dynamics simulations. These findings validate the high-entropy strategy as a promising approach for developing durable, high-performance NTC thermistors for extreme-temperature applications.

Given that the role of Gelsemine in neuroinflammation has been demonstrated, this research aimed to investigate the effect of Gelsemine on neonatal hypoxic-ischemic (HI) brain injury. An in vivo HI brain injury neonatal mouse model and an in vitro oxygen–glucose deprivation (OGD) cell model were established and pretreated with Gelsemine. The brain infarct volume, neuronal loss and apoptosis, as well as spatial learning and memory were examined by TTC staining, Nissl’s staining, TUNEL staining and Morris water maze test. Immunohistochemical staining was applied to detect the microglia cells and astrocytes in the mouse brain tissue. The cell viability was analyzed by CCK-8 assay. The levels of malondialdehyde (MDA), superoxide dismutase (SOD), TNF-α, IL-1β, and IL-6 were determined via ELISA. The lactate dehydrogenase (LDH) release and reactive oxygen species (ROS) level in OGD-treated cells were detected by colorimetry and DCFH-DA staining. Nrf2, HO-1, and inflammation-related factors were analyzed by immunofluorescence, qRT-PCR, or western blot. Gelsemine reduced the infarct volume and neuronal loss and apoptosis, yet improved spatial learning and memory impairment of HI-injured mice. Gelsemine inhibited the elevated MDA, TNF-α, IL-1β, IL-6, LDH and ROS levels, promoted the reduced SOD level and viability, and strengthened the up-regulation of HO-1 and Nrf2 in brain tissues and OGD-treated cells. However, Nrf2 silencing reversed the effects of Gelsemine on the Nrf2/HO-1 pathway, inflammation, and oxidative stress in OGD-treated cells. Gelsemine produces neuroprotective effects on neonatal mice with HI brain injury by suppressing inflammation and oxidative stress via Nrf2/HO-1 pathway.
