Hypocrellin A (HA), a perihydroxylated polycyclic quinone isolated from the fungus of Shiraia bambusicola, exhibits a wide spectrum of biological activities in pre-clinical studies. However, poor water solubility is a major clinical constraint factor of HA, leading to unpredictable bioavailability. In this study, HA-loaded poly (lactic-co-glycolic acid) (PLGA) nanoparticles (PLGA/HA NPs) were prepared by the single-emulsion solvent-evaporation technique. Characterization of PLGA/HA NPs showed that HA was successfully encapsulated on the PLGA. The drug-loading content and encapsulation efficiency (EE) were 7.0% and 57.5%, respectively. The in vitro release profile demonstrated that HA was released more slowly from nanoparticles in pH 1.5 and pH 6.8 than in pH 7.4. Furthermore, the highest solubility of PLGA/HA NPs in aqueous solution was approximately 35.67-fold that of native HA (n-HA). PLGA/HA NPs had more superior stability compared with n-HA in physiological conditions. In this paper, a sample and sensitive LC-MS/MS method was validated for the first time to quantify HA in rat plasma. The pharmacokinetic parameters and bioavailability of HA between PLGA/HA NPs and n-HA were compared after oral administration in rat. The results implied that the relative bioavailability of PLGA/HA NPs was 2.67-fold that of n-HA, and PLGA/HA NPs had a longer half-life. Therefore, these results suggested that PLGA-blend nanoparticles improved the solubility, stability and bioavailability of HA, and could become a potential and promising carrier for the oral delivery of HA.

Lepidium apetalum is valued for its food and medicinal uses; however, research on its chemistry and bioactivities remains limited. This study investigated volatile oils (VOs) and non-volatile extracts (NVEs) from different plant parts. GC–MS identified 111 volatiles dominated by nitrogen- (1.99–70.38 % across plant parts), sulfur- (5.65–62.07 %), and terpenoid-type compounds (0.28–61.06 %). UPLC-QE-Orbitrap-HRMS characterized 573 metabolites, including fatty acids, amino acids, and phenylpropanoids. PCA and PLS-DA revealed significant metabolic differences, identifying 26 differential compounds from GC–MS and 88 from LC-MS. Bioactivity variations were notable: root and husk VOs showed the strongest NO inhibition (IC 50 1.34–2.24 μg/mL), leaf NVE was most active against HL-60 cells (IC 50 82.10 μg/mL), and seed and husk extracts exhibited the highest antioxidant activity (IC 50 143.69–874.43 μg/mL). Correlation analysis identified key metabolites, including benzyl isothiocyanate and erucin, as strongly linked to bioactivities. These findings expand phytochemical knowledge of L. apetalum and highlight its potential for food and nutraceutical applications.

Hyperlipidemia has been highlighted to be one of the most prominent and global chronic condition nowadays. Daming capsule (DMC), a traditional Chinese medicine (TCM) preparation, has treated hyperlipidemia on clinic in China for decades. Our recent study showed that aloe-emodin (AE) is one of the main bioactive components in DMC. Therefore, the present study aims to further investigate the lipid-lowering effect by serum biochemistry and histopathological examination, and reveal the underlying mechanisms by urinary metabolomics approach. After oral administration of AE for 6 weeks, the total cholesterol (TC) and low-density lipoprotein-cholesterol (LDL-c) levels in 50 and 100 mg/kg AE groups were both decreased significantly ( P < 0.05 and P < 0.001). An ultra-performance liquid chromatography/electrospray ionization quadruple time-of-flight mass spectrometry (UPLC-ESI-QTOF-MS) followed by principal components analysis (PCA), partial least squares- discriminant analysis (PLS-DA), orthogonal partial least squares-discriminant analysis (OPLS-DA), correlation analysis, heat map, and KEGG pathways was employed to identify 26 potential biomarkers. Twenty three among them were restored by AE including L-citrulline, 6-methylaminopurine, imidazoleacetic acid riboside, N-acetylhistamine, 3-methyladenine, 1-methyladenosine, dopamine, N1-methyl-4-pyridone-3-carboxamide, formylanthranilic acid, 4-pyridoxate, cAMP, salsolinol, isethionate, d -ribonic acid, 3-sulfolactic acid, vitamin C, mesaconic acid, sulfosalicylic acid, salicyluric acid, N-acetylanthranilic acid, 4,6- Dihydroxyquinoline, sebacic acid and hyocholic Acid. The related metabolic pathways include TCA cycle, the metabolism of amino acids, taurine, B vitamins, purines and pyrimidines. The results indicate that AE has a favorable therapeutic effect on HFD-induced hyperlipidemia by adjusting the metabolic disorders. Notably, urinary metabolomics combined with pattern recognition analysis provides a powerful and reliable approach into the research and development of TCM and phytochemicals.

Styrax japonicus is a small tree or shrub with flowers of great ornamental value. This experiment was conducted to study the metabolites of S. japonicus petals in two colors to improve their value of ornamental and economic. Two kinds of petals were sampled at the pre-bloom stage (S1), the full-bloom stage (S2), and the post-peak stage (S3), respectively. Extensive non-targeted metabolomic assays were performed on the petals using LC-MS. A total of 552 distinct metabolites were found. Additionally, the relative contents of organooxygen compounds, carboxylic acids and derivatives, and fatty acyls in each developmental stage of both petals were the top 3. Notably, a high relative content of kaempferol-3- O -rutinoside was found in each developmental stage of both petals. The differential metabolites of the two types of petals varied considerably at different developmental stages. A portion of the differential metabolites in the S1 was enriched for amino acid and protein synthesis, while a portion of the differential metabolites in the S2 and S3 were enriched for aromatic compound degradation. Furthermore, compared to white flowers, pink flowers might have more metabolic activity. At the same time, in all developmental stages, there is an enrichment of some differential metabolites into the phenylpropanoid synthesis pathway.

Doxorubicin (DOX) is an effective chemotherapeutic agent, but its use is limited by dose-dependent cardiotoxicity. Palmatine (PAL) has shown potential in preventing doxorubicin-induced cardiotoxicity (DIC); however, the underlying mechanism remains unclear. Since DOX accumulation in cardiomyocytes is a key factor in the development of cardiotoxicity, this study aims to explore the pharmacokinetic mechanisms through which PAL alleviates DIC. First, the effect of PAL on DIC was assessed based on H9c2 cardiomyocytes model using sulforhodamine B (SRB), lactate dehydrogenase (LDH), flow cytometry and Western blot assays, which demonstrated that PAL significantly attenuated DOX-induced cardiac toxicity and apoptosis. Second, a sensitive and efficient LC–MS/MS method was established to quantify DOX in H9c2 cells, with a lower limit of quantification of 10 nM and a total run time of 4 min per sample. Cellular pharmacokinetic study revealed that PAL significantly reduced DOX concentration in H9c2 cells. Finally, molecular docking analysis revealed that PAL exhibited low binding energies and formed hydrogen with uptake transporter proteins involved in DOX transport in cardiomyocytes, including organic cardi transporter (OCT) 1, OCT3, organic cation/carnitine transporter (OCTN) 1, OCTN2, and plasma membrane monoamine transporter (PMAT). This suggests a stable interaction between PAL and these transporters. In conclusion, PAL alleviates DIC, most likely by reducing myocardial DOX accumulation through the inhibition of uptake transporters. This study provides novel pharmacokinetic insights into the mechanism by which PAL mitigates DIC and underscores its potential as a complementary therapeutic agent in combination with DOX in clinical applications.

Background Acute lymphoblastic leukemia (ALL) is a prevalent hematologic malignancy that primarily affects children. The diagnosis and treatment of pediatric ALL remain challenging. This study aimed to identify differential lipids and metabolites that may hold potential for improving ALL treatment. Methods In this retrospective case-control study, serum samples obtained from children with ALL and healthy controls were analyzed. Serum lipidome and metabolome alterations of ALL were analyzed by comparing pediatric patients with ALL with healthy controls based on liquid chromatography high-resolution mass spectrometry analysis of serum lipidomic and metabolomic signatures. Results We identified 2,298 lipid features in the serum. Among them, 72 (3.13%) differed significantly in pediatric patients with ALL compared to healthy controls. Notably, sphingolipids (ceramide and sphingomyelin) and phospholipids exhibited the most pronounced changes. Targeted analysis of ceramides revealed significantly elevated levels of Cer 18:0 and Cer 20:0 in the serum of pediatric patients with ALL. Additionally, gut microbial-related lipids (such as sulfonolipids and fatty acid esters of hydroxy fatty acids) showed significant alterations. Metabolomic analysis identified 15 differential metabolites, indicating disrupted nucleotide and amino acid metabolism. Furthermore, the dysregulated lipids and metabolites correlated with various blood indicators, with ceramide and nucleosides positively associated with white blood cell count but negatively correlated with hemoglobin and platelet. Conclusion These findings shed light on abnormal molecular signatures contributing to pediatric ALL and may serve as potential biomarker panel for therapy of ALL.

Background The quantification of urinary oxalate is essential for the diagnosis of primary hyperoxaluria. This study sought to develop a liquid chromatography-tandem mass spectrometry (LC-MS/MS) method for urinary oxalate determination and to evaluate its concordance with an established in-house high-performance liquid chromatography (HPLC) method and a commercial LC-MS/MS assay. Methods The newly developed LC-MS/MS method invloved the quantification of urinary oxalate through a straightforward single-step derivatization process using o -phenylenediamine at 120 °C for 20 min. A total of 291 urine samples were analyzed using both the in-house LC-MS/MS and HPLC methods, while 94 urine samples were evaluated using both our in-house LC-MS/MS method and a commercial LC-MS/MS assay. Additionally, a reference interval for 24-hour urinary oxalate in Chinese adults was established. Results Oxalate exhibited strong analytical retention in reverse-phase liquid chromatography. The newly developed LC-MS/MS method demonstrated excellent linearity with a correlation coefficient exceeding 0.995, and acheived recovery rates ranging from 92.80 % to 98.73 %. The intra-assay and inter-assay coefficients of variation (CVs) were between 0.79 % to 7.46 %, with a total CV ranging from 2.07 % to 7.50 %. No significant matrix effect was observed. However, discrepancies were identified between the in-house LC-MS/MS method and both the commercial LC-MS/MS assay and the HPLC method. The reference interval was established at 0.12–1.04 mmol/24 h (10.8–93.6 mg/24 h). Conclusions The developed LC-MS/MS method is robust and simple, providing a reliable tool for the clinical assessment of urinary oxalate. An accurate method is essential for screening analyses in primary hyperoxaluria.

Bombyx batryticatusis the dried body of silkworm (Bombyx moriLinnaeus) larvae infected withBeauveria bassiana. It is widely used in traditional Chinese medicine for treating convulsions, epilepsy, and hyperglycemia. In this study,Bombyx batryticatusand its extract were prepared. The total reducing power, hydroxyl radical scavenging and superoxide anion radical scavenging activities, as well as the α-glucosidase inhibitory activity ofBombyx batryticatusextract were superior to those of normal silkworm larvae extract. Among them, the IC50value ofBombyx batryticatusextract for α-glucosidase was 5.76 mg/mL, while that of normal silkworm larvae extract was 7.0 mg/mL. Untargeted metabolomic analysis was employed to compare the material composition of normal silkworm larvae andBombyx batryticatus. The results revealed 101 metabolic differences between the two groups, including a significant increase in fatty acids and their derivatives in theBombyx batryticatusextract. Further separation and purification of theBombyx batryticatusextract were performed using solvents of varying polarity. The chloroform fraction exhibited the highest inhibitory activity against α-glucosidase, with an IC50value of 0.217 mg/mL. LC-MS further identified compounds in the chloroform fraction, suggesting that those alkaloids, fatty acids, and their derivatives may be responsible for its strong α-glucosidase inhibitory activity. This study elucidates the material basis underlying the pharmacological effects ofBombyx batryticatus, particularly its hypoglycemic components, thereby providing critical experimental support for its future development and application in medicine.

Evaluating the quality of embryos and implantation potential is a critical determinant of in vitro fertilization and embryo transfer, and it is also one of the main challenges of assisted reproductive technology. A reliable noninvasive method to choose the best candidate with real implantation potential for transfer from two day 3 embryos with equal morphological quality is still lacking clinically. In this article, a sensitive LC-MS method was developed and metabolomic profiling analysis in 3 days embryo culture medium was performed. Differential metabolites were analysed in two kinds of commercial culture media, and a total of common 66 metabolites were obtained from 106 independent samples in 5 batches. The relationship between changes in key metabolite benzoic acid concentrations and the embryo implantation result was discovered. This work improved coverage through conditional optimization, enhanced the reliability of omics data through multi batch validation, and provided a potential biomarker for evaluating the implantation potential of day 3 embryos.

Aims This study aimed to elucidate the biochemical and metabolic mechanisms underlying glufosinate-ammonium-induced neurotoxicity, with a focus on brain metabolic disturbances. Methods Male Sprague-Dawley rats were administered glufosinate-ammonium by intragastric gavage, with saline-treated animals serving as controls. Neuropathological changes and cognitive performance were assessed at 2 and 6 days post-exposure. Oxidative stress and energy metabolism were evaluated by measuring plasma antioxidant indices, and brain levels of superoxide dismutase (SOD), glutathione (GSH/GSSG), total antioxidant capacity (T-AOC), and adenosine triphosphate (ATP). In addition, untargeted LC-MS/MS-based metabolomics of brain tissue was conducted to characterize exposure-related metabolic perturbations. Results Histopathological examination revealed marked brain injury on day 2 post-exposure, characterized by neuronal necrosis, apoptosis, and severe mitochondrial disruption. Although pathological severity was attenuated by day 6, residual cellular damage remained evident. Behavioral testing revealed significant impairments in spatial learning and memory in the glufosinate-ammonium-exposed group. Biochemical analyses revealed disrupted oxidative stress and energy metabolism, reflected by altered levels of superoxide dismutase (SOD), glutathione (GSH/GSSG), total antioxidant capacity (T-AOC), and adenosine triphosphate (ATP) in brain tissue. Metabolomic analysis revealed pronounced alterations in brain metabolic profiles, involving pathways related to oxidative stress, energy metabolism, and amino acid metabolism. Early-stage disturbances were dominated by alterations in glutamate and tricarboxylic acid (TCA) cycle-related metabolites, whereas later-stage changes primarily involved taurine and hypotaurine metabolism. Conclusion Following glufosinate-ammonium exposure, rats exhibited neurological dysfunction and cognitive impairment, accompanied by metabolic disturbances in brain tissue, particularly in oxidative stress, energy metabolism, and amino acid metabolism. These findings provide mechanistic insight into glufosinate-ammonium-induced neurotoxicity and highlight the critical role of metabolic dysregulation.

Cyclobutane pyrimidine dimers (CPDs) and pyrimidine (6–4) pyrimidone photoproducts (6–4PPs) are genotoxic DNA lesions and mainly generated on thymine–thymine (T–T) dinucleotides upon UV irradiation. Regarding the sensitivity, specificity, and accuracy of analytical methods, it is of first choice to develop a reliable assay for simultaneous detection of these DNA lesions using liquid chromatography-tandem mass spectrometry (LC-MS/MS). However, the dilemma is the low detection sensitivity of the phosphate-containing dimeric photoproducts even using most favorable negative-ion mode for LC-MS/MS analysis. Unexpectedly, we observed that the detection sensitivity of T–T CPD and 6–4PP could be significantly improved using formic acid/acetic acid (∼ppm) as an additive of the mobile phase for reversed-phase LC-MS/MS analysis. This is the first demonstration of the enhancement of LC-MS/MS signals by formic acid/acetic acid in negative-ion mode. Of note, these acidic agents are often used for positive-ion mode in LC-MS assays. Benefited from the developed method, we could quantify both T–T CPD and 6–4PP in mouse embryonic stem cells upon UVC irradiation at low dosage. This sensitive method is applicable to the screening and identification of genes involved in formation, signaling, and repair of UV lesion.

A simple and enantioselective method was developed and validated for the simultaneous determination of (R)- and (S)-trelagliptin in beagle dog plasma by chiral liquid chromatography tandem mass spectrometry. Trelagliptin enantiomers and (R)-rabeprazole (as internal standard, IS) were extracted from plasma samples by liquid–liquid extraction and separated on a CHIRALCEL OX-3R column using acetonitrile-5 ammonium bicarbonate as the mobile phase in gradient elution mode. The multiple reactions monitoring transitions of m/z 358.1→341.2 and 359.9→150.1 were used to quantify trelagliptin enantiomers and IS, respectively. This method was validated for sensitivity, specificity, linearity, precision, accuracy and stability of specific analytes under various conditions. And it was successfully applied to evaluating the pharmacokinetic profile of trelagliptin enantiomers in beagle dogs after single intravenous administration of (R)-trelagliptin injection (at 1 mg/kg) and oral administration (at 6.7 mg/kg). In this study, no chiral bioconversion of (R)-trelagliptin to (S)-trelagliptin in beagle dog plasma was observed. The absolute bioavailability of (R)-trelagliptin was identified to be 128.2%.
