Due to the particularity of melanoma location, traditional drug delivery methods face difficulties in penetrating skin tissue and specifically clearing melanoma cells. Here, we designed a hyaluronic acid microneedle drug delivery system loaded with nanomotors which can effectively penetrate the skin tissue and deliver the therapeutic components to melanoma and specifically remove melanoma cells. The loaded nanomotors were prepared by radical polymerization of methacryloyl modified L-arginine (MA) and L-tyrosine (MTyr), named as MA/MTyr nanomotor. The microneedle drug delivery system can use the rigid structure of microneedles to break through the skin barrier and deliver nanomotors to deep tissues. The MA endowed them with the ability to sensitively sense the highly expressed inducible nitric oxide synthase in the tumor microenvironment, so as to realize the effective targeting of melanoma in skin tissue and uptake by tumor cells. Subsequently, the MTyr can produce quinone-enriched intermediates that can induce cell death under the catalysis of tyrosinase that was specifically highly expressed in melanoma cells, and self-assembled to form micron scale fibers through intermolecular hydrogen bonding interactions, thereby inducing melanoma cells to swell. This process synergistically promoted melanoma cell death without occurring in normal cells. The experimental results of cell and animal models confirmed the good therapeutic effect of the nanomotor loaded microneedles, which provided a new therapeutic concept and strategy for the development of therapeutic methods specifically for melanoma.

In recent years, the incidence of inflammatory bowel disease has gradually increased. Traditional drugs can reduce inflammation, but cannot be targeting released and often require the coordination with delivery systems. However, a good targeting performance delivery system is still scarce currently. Inflammation can trigger oxidative stress, producing large amounts of oxides such as nitric oxide (NO). Based on this, the present experiment innovatively designed a hydrogel delivery system with NO response that could be inflammation targeting. The hydrogel is composed of sodium alginate modified with glycerol methacrylate, crosslinked with NO response agent by photo-crosslinking method, which have low swelling (37 %) and good mechanical properties with a stable structure even at 55 °C. The results of in vitro digestion also indicated that the hydrogel had a certain tolerance to gastrointestinal digestion. And in the NO environment, it was interestingly found that the structure and mechanical properties of the hydrogels changed significantly. Moreover, hydrogels have good biocompatibility, which ensures their safe use in vivo. In conclusion, this NO-responsive-based delivery system is feasible and provides a new approach for drugs and active factors targeting delivery in the future.

Since conventional techniques are ineffective for NO removal at low concentrations, photocatalysis has become attractive in this regard, recently. However, in practice, photocatalytic NO removal has drawbacks such as limited light absorption and the proclivity of producing toxic by-products. To address these issues, novel defective Bi/Bi3NbO7structures with good porosity were fabricated by a solvothermal method and used for enhanced photocatalytic NO removal under visible light irradiation. The morphological and structural properties of the prepared materials were comprehensively analyzed. The optimal photocatalytic activity of pore-defective Bi/Bi3NbO7for NO removal was 60.3%, when the molar ratios of urea and Bi(NO)3•5H2O to pristine Bi3NbO7were 1:25 and 1:2, respectively, under the following operational conditions: NO concentration of 700 ppb, catalyst dosage of 50 mg and irradiation time of 14 min. The induced defects and the surface plasmon resonance (SPR) effect of Bi nanodots made remarkable contributions to improving the photocatalytic NO removal as well as inhibiting the toxic byproduct NO2. The photocatalytic NO removal pathway over the prepared photocatalysts was further mechanistically clarified taking advantage of EPR results and scavenging experiments. Considering the increased NO generation in the atmosphere, this work may provide novel insights for designing effective porous photocatalysts to treat gaseous toxic pollutants.Keywords:photocatalysis;Bi3NbO7;surface plasmon resonance;NO removal;defects

Clinical chemotherapy for cancer is limited by the physiological barrier of tumors, resulting in low drug delivery to tumors, poor efficacy of drugs and inability to block tumor metastasis. Here we developed an intelligent switchable nitric oxide (NO)-releasing nanoparticle, IPH-NO, which loads a photosensitizer (IR780) and the chemotherapy drug paclitaxel (PTX) into NO donor-S-nitrosated human serum albumin (HSA-NO). NO exhibits two effects based on its concentration: enhancement of chemotherapy by increasing the enhanced permeability and retention (EPR) effect at low concentrations and direct killing of cancer cells at high concentrations. IPH-NO can slowly release NO in the presence of glutathione to boost tumor vascular permeability and improve drug accumulation. Near-infrared light irradiation was utilized to induce a quick release of NO that can directly kill cancer cells at high concentrations. This combination of phototherapy and NO gas therapy activated by NIR together with chemotherapy showed significant effects in tumor inhibition. Furthermore, IPH-NO blocked tumor metastasis by inhibiting epithelial mesenchymal transition. PH-NO provides a novel strategy to control NO release at tumor site for drug accumulation and combination therapies, consequently potentiating the anticancer efficacy and inhibiting tumor metastasis.

This study was aimed at improving oral bioavailbility and reducing the food effect of cinacalcet hydrochlorde (CINA), a poorly soluble drug for the treatment of chronic kidney disease, by preparing its nanocrystals (NCs) utilizing the precipitation–ultrasonication method. Based on the single factor method and Box–Behnken design, with the particle size and polydispersity index (PDI) as indexes, the optimal formulation was achieved. It was investigated that the particle size and PDI of the NCs prepared on the basis of optimal formulation were 244 ± 2 nm and 0.168 ± 0.001, respectively. The NCs were solidificated by lyophilization. Scanning electron microscopy, differential scanning calorimetry and x-ray powder diffraction were used to characterize the CINA-NCs, and there was no crystalline change during preparation and lyophilization. The CINA-NCs capsules prepared with 30% (w/v) MCC, 8% (w/v) CCNa and 2% (w/v) talcum powder by orthogonal experimental design presented an enhanced in vitro dissolution rate in four media compared with commercial tablets Sensipar® and raw material. The raw material, blank NCs and CINA-NCs were confirmed to be non-toxic to Caco-2 cells when the drug concentration was below 250 μg ml−1. In the in vivo pharmacokinetic study, the Cmax (the peak concentration of CINA in plasma) and AUC0–t (area under curve by trapezoidal area method) of the CINA-NCs capsules were approximately 1.90-fold and 1.64-fold greater than that of Sensipar® in the fasted state. Overall, this nanotechnology is a promising way to optimize the dosage form of CINA oral administration.

In this work, we demonstrate an innovative strategy for efficient and sustainable absorption of NO in a series of ionic liquids (ILs) containing amine groups and azole groups. The results show that the ILs have low viscosities and high performance for NO absorption. For instance, [TEPA][Im] had a high absorption capacity of 6.79 mol NO per mol IL at 30 °C and ambient pressure. The absorbed NO was easily released by heat treatment, and the regenerated IL had significant reversibility. The results of 1H NMR, UV spectra, and FT-IR spectra of ILs before and after NO absorption indicated that the absorption of NO in the [TEPA][Im] mainly resulted from a chemical interaction. NO interacts with not only the secondary amine groups in the cation but also N-imidazole in the anion through the formation of NONOate, indicating [TEPA][Im] is a dual functional IL. By introducing this efficient and reversible absorption of NO by ILs, our work broadens the method of NO capture.

This study reports a tumor-specific ROS-responsive nanoplatform capable of the combination of nitric oxide (NO)-based gas therapy and sensitized photodynamic therapy (PDT). The nanoplatform is constructed on porous coordination network (PCN), which contains NO donor L-Arg and is concurrently coated with cancer cell membrane ( [email protected] @Mem). Under near infrared light (NIR) irradiation, [email protected] @Mem produces plenty of reactive oxygen species (ROS) directly for PDT therapy, while a part of ROS take the role of oxidative to converse L-Arg into NO for combined gas therapy. The results indicate that the transformation of ROS to NO can enhance PDT efficacy in hypoxic tumors owing to the ability of NO in freely diffusing into deep hypoxic tumor site. Moreover, homologous targeting function originated from the coating of cancer cells membrane further improves the tumor treatment effect owing to the biotargeting toward homologous tumors. This [email protected] @Mem nanoplatform provides a new therapy paradigm of overcoming the hypoxia barrier of tumor therapy, and holds great potential for the treatment of tumor and NO-related diseases.

Electrochemical reduction of NO to NH3 is of great significance for mitigating the accumulation of nitrogen oxides and producing valuable NH3. Here, we demonstrate that the CoS nanosheet with sulfur vacancies (CoS1–x) behaves as an efficient catalyst toward electrochemical NO-to-NH3 conversion. In 0.2 M Na2SO4 electrolyte, such CoS1–x displays a large NH3 yield rate (44.67 μmol cm–2 h–1) and a high Faradaic efficiency (53.62%) at −0.4 V versus the reversible hydrogen electrode, outperforming the CoS counterpart (27.02 μmol cm–2 h–1; 36.68%). Moreover, the Zn–NO battery with CoS1–x shows excellent performance with a power density of 2.06 mW cm–2 and a large NH3 yield rate of 1492.41 μg h–1 mgcat.–1. Density functional theory was performed to obtain mechanistic insights into the NO reduction over CoS1–x.

L-arginine (LA) as a semiessential amino acid within human body has shown great promise in cancer therapy since it can continuously generate nitric oxide (NO) in the presence of inducible NO synthase (iNOS) or reactive oxygen species (ROS). However, NO production efficiency in tumor tissue is severely restricted by the hypoxic and H 2 O 2 -deficient tumor microenvironment (TME). Herein, mesoporous calcium peroxide (CaO 2 ) with the ability of supplying O 2 /H 2 O 2 has been prepared to encapsulate and oxidize LA to achieve controllable local release of NO in the acidic TME. Interestingly, the generated NO can aggravate the abnormal retention of Ca 2+ in cells, thus resulting in amplified oxidative stress and immunogenic cell death. To improve the stability of CaO 2 in the body fluid, polydopamine (PDA) is further introduced to coat on the surface of LA-CaO 2 and its high photothermal conversion property will reinforce the immunogenic dying of tumor cells. Such a NO-enhanced phototherapeutic nanoplatform, LA-CaO 2 @PDA, exhibit the most effective treatment in inhibiting primary and distant tumor growth and suppressing lung metastasis of 4T1 tumor by provoking a strong antitumor immune response.

Co-doped MnCeO x /ZrO 2 catalysts were synthesized by impregnation method and their low temperature deNOx performance were evaluated. The physicochemical properties of the catalysts were studied. The results showed that the doped Co could promote the deNOx performance of MnCeO x /ZrO 2 significantly, and the doped catalyst with the Co/Mn molar ratio of 1:2 possessed the best catalytic performance. Compared with pure MnCeO x /ZrO 2 catalyst, the deNOx efficiency of the optimal 1Co 2 MnCeO x /ZrO 2 was higher to 93% at 100 °C, improved nearly by 17%. The complete removal of NO was achieved at the temperature range of 120–250 °C. The promoted catalytic performance of Co-doped MnCeO x /ZrO 2 catalyst was mainly attributed to the improvement of the catalyst support structure and surface acidity by Co. The catalytic reaction of NO with NH 3 over 1Co2MnCeO x /ZrO 2 catalyst follows both Eley–Rideal mechanism and Langmiur–Hinshelwood mechanism.

The conversion of nitrogen oxide (NO) by photocatalysis is promising to solve the increasingly serious air pollution. Herein, raspberry-like TiO2 hollow spheres consisting of small nanocrystals (less than 10 nm) were created using hollow carbon spheres as templates by controlling the synthesis parameters. These raspberry-like hollow spheres were well dispersed and revealed large specific surface area (172.6 m2 g−1) which was about 3 times that of commercial TiO2 powders (P25). The raspberry-like hollow spheres with an anatase phase composition and high crystallinity revealed high NO2 selectivity for NO conversion. Compared with P25, the hollow spheres exhibited excellent removal performance and high stability for NO, in which ∼60% of NO at a concentration of 600 ppb was removed while P25 removed ∼40% of NO. This is ascribed to the large specific surface area and pore volume, exposed (001) facets with high energy, and the reflection and scattering of light in the hollow sphere structure improving the utilization of light. Based on the diffuse reflection spectra, Mott–Schottky curves, and band gap structures, the removal mechanism is discussed.

Drug-coated balloons (DCB) intervention is an important approach for the treatment of atherosclerosis (AS). However, this therapeutic approach has the drawbacks of poor drug retention and penetration at the lesion site. Here, a lipophilic drug-loaded nanomotor as a modified balloon coating for the treatment of AS is reported. First, a lipophilic nanomotor PMA-TPP/PTX loaded with drug PTX and lipophilic triphenylphosphine (TPP) compounds is synthesized. The PMA-TPP/PTX nanomotors use nitric oxide (NO) as the driving force, which is produced from the reaction between arginine on the motor substrate and excess reactive oxygen species (ROS) and inducible nitric oxide synthase (iNOS) in the AS microenvironment. The final in vitro and in vivo experimental results confirm that the introduction of the lipophilic drug-loaded nanomotor technology can greatly enhance the drug retention and permeability in atherosclerotic lesions. In particular, NO can also play an anti-AS role in improving endothelial cell function and reducing oxidative stress. The chemotherapeutic drug PTX loaded onto the nanomotors can inhibit cell division and proliferation, thereby exerting the effect of inhibiting vascular intimal hyperplasia, which is helpful for the multiple therapies of AS. Using nanomotor technology to solve cardiovascular diseases may be a promising research direction.
