Electrospun PCL/keratin/AuNPs mats with the catalytic generation of nitric oxide for potential of vascular tissue engineering

Xiuzhen Wan,Pengcheng Liu,Xingxing Jin,Xuanxuan Xin,Pengfei Li,Jiang Yuan,Jian Shen

Nitric oxide (NO)-generating materials are beneficial for vascular tissue engineering (VTE) scaffold because the produced NO would enhance endothelial cells viability while inhibit smooth muscle cell (SMC) proliferation and reduce platelet adhesion, resulting in ideal hemocompatibility and endothelialization. Herein, poly(ε-caprolactone) (PCL)/keratin biocomposite mats were first fabricated, followed by in situ gold (Au) nanoparticles loading to afford PCL/keratin/AuNPs mats. These mats were characterized using field emission scanning electron microscopy (FE-SEM), X-ray photoelectron spectroscopy (XPS), X-ray diffraction (XRD), and thermogravimetric analysis (TGA). The PCL/keratin/AuNPs mats were demonstrated to be capable of catalyzing NO release in the mimicked blood microenvironments. The generated NO could enhance human umbilical vein endothelial cell growth and inhibit human umbilical arterial SMC viability. In addition, these mats maintained the antibacterial activity of Au nanoparticles with good blood compatibility. Taken together, these keratin-based composite mats have potential usage in the VTE. © 2018 Wiley Periodicals, Inc. J Biomed Mater Res Part A: 106A: 3239–3247, 2018.

Enhanced osteogenesis and angiogenesis by PCL/chitosan/Sr-doped calcium phosphate electrospun nanocomposite membrane for guided bone regeneration

Huilin Ye,Junjin Zhu,Dan Deng,Shue Jin,Jidong Li,Yi Man

Membranes play pivotal role in guided bone regeneration (GBR) technique for reconstruction alveolar bone. GBR membrane that is able to stimulate both osteogenic and angiogenic differentiation of cells may be more effective in clinic practice. Herein, we fabricated the Sr-doped calcium phosphate/polycaprolactone/chitosan (Sr-CaP/PCL/CS) nanohybrid fibrous membrane by incorporating 20 wt% bioactive Sr-CaP nanoparticles into PCL/CS matrix via one-step electrospinning method, in order to endow the membrane with stimulation of osteogenesis and angiogenesis. The physicochemical properties, mechanical properties, Sr2+ release behavior, and the membrane stimulate bone mesenchymal stem cell (BMSCs) differentiation were evaluated in comparison with PCL/CS and CaP/PCL/CS membranes. The SEM images revealed that the nanocomposite membrane mimicked the extracellular matrix structure. The release curve presented a 28-day long continuous release of Sr2+ and concentration which was certified in an optimal range for positive biological effects at each timepoint. The in vitro cell culture experiments certified that the Sr-CaP/PCL/CS membrane enjoyed excellent biocompatibility and remarkably promoted rat bone mesenchymal stem cell (BMSCs) adhesion and proliferation. In terms of osteogenic differentiation, BMSCs seeded on the Sr-CaP/PCL/CS membrane showed a higher ALP activity level and a better matrix mineralization. What’s more, the synergism of the Sr2+ and CaP from the Sr-CaP/PCL/CS membrane enhanced BMSCs angiogenic differentiation, herein resulting in the largest VEGF secretion amount. Consequently, the Sr-CaP/PCL/CS nanohybrid electrospun membrane has promising applications in GBR.

An injectable thermosensitive hydrogel self-supported by nanoparticles of PEGylated amino-modified PCL for enhanced local tumor chemotherapy

Jinxuan Guo,Zujian Feng,Xiang Liu,Changrong Wang,Pingsheng Huang,Jianhua Zhang,Liandong Deng,Weiwei Wang,Anjie Dong

We synthesized amino-modified poly(ε-caprolactone) PCN-b-PEG-b-PCN (PECN) triblock copolymers and studied the contribution of the introduced amino groups to the drug delivery efficiency of PECN nanoparticles (NPs) and their injectable thermosensitive hydrogels. PECN15 with an optimal amino group content was obtained. Firstly, the hydrophobic drug paclitaxel (PTX) was loaded into PECN15 up to 5.91% and formed PTX/PECN NPs 90 nm in size and with a slightly positive charge (7.3 mV). Furthermore, the injectable PTX/PECN NPs aqueous solution (25 wt%) at ambient temperature could undergo fast gelation at 37 °C and sustainedly release PTX/PECN NPs in 10 days. More importantly, compared with our previously reported PECT NPs, the PECN NPs without an increase in toxicity could improve the cell uptake and enhance intracellular drug release by responding to the acidic environment of the endosome. Thus, the PTX/PECN NPs presented a lower IC50 of 3.14 μg mL−1 than that of the PTX/PECT NPs (7.67 μg mL−1) and free PTX (4.65 μg mL−1). Moreover, through peritumoral injection, the PTX/PECNGel showed 94.27% inhibition rate of tumor growth on day 19, higher than PTX/PECTGel (72.28%) and Taxol® (47.03%). Therefore, the PECN NPs hydrogel provided a more effective injectable platform to enhance local cancer chemotherapy, and also provided the possibility of further functionalization by the reactive amino groups.

Electrospun PCL-based polyurethane/HA microfibers as drug carrier of dexamethasone with enhanced biodegradability and shape memory performances

Lv Haitao,Tang Dongyan,Sun Zhaojie,Gao Jingru,Yang Xu,Jia Shuyue,Peng Jing

Shape memory polymers (SMP) with better biodegradability and better stability have great potential applications in biomedical fields, such as the drug carriers or the tissue engineering scaffolds. In this study, poly(ε-caprolactone)(PCL)-based polyurethane(PU) microfibers were fabricated with the containing of hydroxyapatite(HA) to enhance the biodegradability and to exhibit excellent shape memory performance. The composition, the morphology, the thermal stability, and the mechanical properties of the microfibers were characterized and detected using Fourier transform infrared spectroscopy (FTIR), 1 H nuclear magnetic resonance ( 1 HNMR), scanning electron microscopy (SEM), differential scanning calorimetry (DSC). and dynamic mechanical analysis (DMA), etc. And dexamethasone was selected as drug model to investigate the delivery and release behaviors of the carrier of the microfibers. It was revealed that HA enhanced the degradation rate of the shape memory polyurethane (SMPU) fibers, and the fibers could guarantee a sustained long time drug release. The detection on the shape memory performance found that, with the different addition amounts of HA, the composite microfibers of (SMPU) and HA exhibited the different shape memory transition temperature ( T trans ) values. And with the addition of 3 wt% of HA, the excellent shape recovery ratios of R r (> 97%) and the shortest recovery time of ~ 6 s could be obtained. With further increase of the amounts of HA, the recovery force and the recovery time were reduced and prolonged, respectively. The obtained results proved that the biodegradable SMPU/HA composite microfibers have more valuable application prospects in biomedical fields.

Targeted delivery of ginsenoside compound K using TPGS/PEG-PCL mixed micelles for effective treatment of lung cancer

Lei Yang,Zhenghai Zhang,Jian Hou,Xin Jin,Zhongcheng Ke,Dan Liu,Mei Du,Xiaobing Jia,Huixia Lv

Ginsenoside compound K (CK) is one of the effective ingredients in antitumor composition of ginsenoside. However, the poor water solubility and significant efflux have limited the widespread clinical use of CK. In this study, preparation of novel CK-loaded d -alpha-tocopheryl polyethylene glycol 1,000 succinate/poly(ethylene glycol)-poly(ε-caprolactone) mixed micelles (CK-M) is discussed to solve the above problems. Particle size, zeta potential, and morphology were characterized using dynamic light scattering and transmission electron microscopy. CK-M are spherical shaped with an average particle size of 53.07±1.31 nm with high drug loading of 11.19%±0.87% and entrapment efficiency of 94.60%±1.45%. Water solubility of CK was improved to 3.78±0.09 mg/mL, which was ~107.35 times higher than free CK. A549 and PC-9 cells were used to evaluate in vitro cytotoxicity and cellular uptake. IC 50 values of CK-M in A549 and PC-9 cells (24 h) were 25.43±2.18 and 18.35±1.90 μg/mL, respectively. Enhanced cellular uptake of CK-M was observed in both cells. Moreover, CK-M promoted tumor cell apoptosis, inhibited tumor cell invasion, metastasis, and efflux through regulation of Bax, Bcl-2, matrix metalloproteinase-2, Caspase-3, and P-glycoprotein. In vivo imaging indicated that CK-M has excellent tumor targeting effect within 24 h, and the relative tumor inhibition rate of CK-M was 52.04%±4.62% compared with control group ( P <0.01). Thus, CK-M could be an appropriate delivery agent for enhanced solubility and antitumor effect of CK.

Electrospun PLGA/PCL Nanofiber Film Loaded with LPA Promotes Full-Layer Wound Healing by Regulating the Keratinocyte Pyroptosis

Xinqi Huang,Jianghuiwen Lu,Yumei An,Mingyuan Xu,Xueshi Chen,Chao Liu,Xuefeng Zhou,Haiyan Shan,Yunzhu Qian,Mingyang Zhang

Electrospun nanofibers have a number of qualities that make them a suitable choice for skin wound healing. Lysophosphatidic acid (LPA) stimulates the keratinocytes and fibroblasts to proliferate, differentiate, and migrate and enhances skin wound healing. Here, we developed the electrospun scaffolds contained in polycaprolactone (PCL) and polylactic-co-glycolic acid (PLGA). The scaffolds loaded with LPA nanoparticles retained a porous nanofiber structure and showed better physicochemical properties and biocompatibility. The scaffold continuously releases LPA to quickly initiate cell signaling and maintain long-term anti-inflammatory activity. In this study, we found that PP scaffold with LPA reduces the disordered collagen deposition and the thickness of the newborn epidermis, improves skin healing, and reduces scar formation. Explaining the mechanism of LPA mineralized tissue regeneration in skin wound healing, LPA inhibited the pyroptosis of keratinocyte, a cell death process that induces inflammation and scar formation by inhibiting the expression of TNF-α and β-catenin proteins. Thus, the electrospun PP scaffold with LPA can be potentially developed as a therapeutic avenue to target skin wound healing.

Development of a colorimetric nanofibrous film sensor based on polycaprolactone (PCL) incorporated with alizarin for tracking shrimp freshness

Yue Zou,Siyang Chen,Hanzheng Dou,Wenxiu Zhu,Dongyun Zhao,Ying Wang,Haisong Wang,Xiaodong Xia

In this study, a colorimetric nanofibrous film was developed using polycaprolactone (PCL) and alizarin through electrospinning, and the film was subsequently employed for tracking shrimp freshness. Scanning electron microscopy (SEM) showed that the film was composed of beaded, continuous and smooth nanofibers. In the pH range of 4–13, the nanofibrous films displayed discernible color changes from yellow green to bluish-purple. Furthermore, when used together with shrimps, the films underwent a color change from yellow to purple, demonstrating a robust association with TVB-N and the pH value. Thus, the alizarin-based colorimetric nanofibrous films developed here could be potentially used as an indicator for tracking the freshness of shrimps.

3D-printed PCL@BG scaffold integrated with SDF-1α-loaded hydrogel for enhancing local treatment of bone defects

Wang Chenglong,Dong Jinlei,Liu Fanxiao,Liu Nan,Li Lianxin

Background The long-term nonunion of bone defects is always a difficult problem in orthopaedics treatment. Artificial bone implants made of polymeric materials are expected to solve this problem due to their suitable degradation rate and good biocompatibility. However, the lack of mechanical strength, low osteogenic induction ability and poor hydrophilicity of these synthetic polymeric materials limit their large-scale clinical application. Results In this study, we used bioactive glass (BG) (20%, W/W) and polycaprolactone (PCL, 80%, W/W) as raw materials to prepare a bone repair scaffold (PCL@BG20) using fused deposition modelling (FDM) three-dimensional (3D) printing technology. Subsequently, stromal cell-derived factor-1α (SDF-1α) chemokines were loaded into the PCL@BG20 scaffold pores with gelatine methacryloyl (GelMA) hydrogel. The experimental results showed that the prepared scaffold had a porous biomimetic structure mimicking that of cancellous bone, and the compressive strength (44.89 ± 3.45 MPa) of the scaffold was similar to that of cancellous bone. Transwell experiments showed that scaffolds loaded with SDF-1α could promote the recruitment of bone marrow stromal cells (BMSCs). In vivo data showed that treatment with scaffolds containing SDF-1α and BG (PCL@BG-GelMA/SDF-1α) had the best effect on bone defect repair compared to the other groups, with a large amount of new bone and mature collagen forming at the bone defect site. No significant organ toxicity or inflammatory reactions were observed in any of the experimental groups. Conclusions The results show that this kind of scaffold containing BG and SDF-1α serves the dual functions of recruiting stem cell migration in vivo and promoting bone repair in situ. We envision that this scaffold may become a new strategy for the clinical treatment of bone defects.

Structurally sophisticated 3D-printed PCL-fibrin hydrogel meniscal scaffold promotes in situ regeneration in the rabbit knee meniscus

Hebin Ma,Bowen Xie,Hongguang Chen,Lifang Hao,Haigang Jia,Dengjie Yu,Yuanbo Zhou,Puzhen Song,Yajing Li,Jing Liu,Kaitao Yu,Yantao Zhao,Yadong Zhang

A meniscus injury is a common cartilage disease of the knee joint. Despite the availability of various methods for the treatment of meniscal injuries, the poor regenerative capacity of the meniscus often necessitates resection, leading to the accelerated progression of osteoarthritis. Advances in tissue engineering have introduced meniscal tissue engineering as a potential treatment option. In this study, we established the size of a standardized meniscal scaffold using knee Magnetic Resonance Imaging (MRI) data and created a precise Polycaprolactone (PCL) scaffold utilizing 3-Dimensional (3D) printing technology, which was then combined with Fibrin (Fib) hydrogel to form a PCL-Fib scaffold. The PCL scaffold offers superior biomechanical properties, while the Fib hydrogel creates a conducive microenvironment for cell growth, supporting chondrocyte proliferation and extracellular matrix (ECM) production. Physical and chemical characterization, biocompatibility testing, and in vivo animal experiments revealed the excellent biomechanical properties and biocompatibility of the scaffold, which enhanced in situ meniscal regeneration and reduced osteoarthritis progression. In conclusion, the integration of 3D printing technology and the Fib hydrogel provided a supportive microenvironment for chondrocyte proliferation and ECM secretion, facilitating the in situ regeneration and repair of the meniscal defect. This innovative approach presents a promising avenue for meniscal injury treatment and advances the clinical utilization of artificial meniscal grafts.

A flexible piezoresistive pressure sensor with high sensitivity and wide sensing range based on TPU/PCL/MXene@PEDOT:PSS nanofibers

Chuang Wu,Manlin Jiang,Jiacheng Wu,Jiuxin Zhang

To overcome the difficulty of forming stable and continuous conductive pathways of MXene in conventional electrospinning systems, we develop a high-performance flexible pressure sensor featuring a synergistic embedded–coated dual conductive architecture. A TPU/PCL nanofiber membrane uniformly incorporating MXene is fabricated via one-step electrospinning, followed by PEDOT:PSS dip-coating to construct an external conductive layer. Subsequent Ecoflex 00–30 encapsulation ensures mechanical integrity and maintains continuous multilevel electron-transport networks. Structural characterizations confirm the homogeneous incorporation of MXene within the nanofibers and the continuous surface coverage of PEDOT:PSS. The optimized sensor exhibits a maximum sensitivity of 7.05 kPa⁻¹ in the low-pressure region, together with a broad sensing range up to 100 kPa, enabling reliable detection across both subtle and high-pressure regimes. It further demonstrates fast dynamic response (0.11 s/0.31 s for response/recovery), a low detection limit of 50 Pa, and excellent durability over 2000 loading cycles. Demonstration tests confirm its ability to detect subtle physiological motions at different body locations, such as wrist movements and throat motions during swallowing, as well as various joint motions. Overall, this synergistic dual conductive strategy offers a promising platform for wearable health monitoring, electronic skins, and human–machine interaction.

HE@PCL/PCE Gel-Nanofiber Dressing with Robust Self-Adhesion toward High Wound-Healing Rate via Microfluidic Electrospinning Technology

Rong Chen,Zhiting Liu,Tingting Cui,Xiaoying Zhang,Cai-Feng Wang,Guo-Xing Li,Gefei Wang,Su Chen

Hydrogels have attracted increasing attention in the biomedical field due to their similarity in structure and composition to natural extracellular matrices. However, they have been greatly limited by their low mechanical strength and self-adhesion for further application. Here, a gel-nanofiber material is designed for wound healing, which synergistically combines the benefits of hydrogels and nanofibers and can overcome the bottleneck of poor mechanical strength and self-adhesion in hydrogels and inadequate healing environment created by nanofibers. First, a nanofiber scaffold composed of polycaprolactone/poly(citric acid)-ε-lysine (PCL/PCE) nanofibers is fabricated via a new strategy of microfluidic electrospinning, which could provide a base for hyaluronic acid-polylysine (HE) gel growth on nanofibers. The prepared HE@PCL/PCE gel-nanofiber possesses high tensile strength (24.15 ± 1.67 MPa), excellent air permeability (656 m3/m2 h kPa), outstanding self-adhesion property, and positive hydrophilicity. More importantly, the prepared gel-nanofiber dressing shows good cytocompatibility and antibacterial properties, achieving a high wound-healing rate (92.48%) and 4.685 mm granulation growth thickness within 12 days. This material may open a promising avenue for accelerating wound healing and tissue regeneration, providing potential applications in clinical medicine.

Functional β-TCP/MnO2/PCL artificial periosteum promoting osteogenic differentiation of BMSCs by reducing locally reactive oxygen species level

Feng Qian,Zongwang Huang,Wenbin Liu,Yanling Liu,Xi He

Segmental bone defects caused by trauma, tumor resection or congenital malformations are often reconstructed with autologous, allogeneic bone grafts or artificial bone materials, of which, about 5% ~ 10% will have delayed healing or even nonunion of fractures. The loss of periosteum and excessive accumulation of ROS in fracture site leads to the aging of osteoblasts and the decline of their proliferation and differentiation, thus affecting the fracture healing process. In this study, we prepared a functional modified artificial periosteum β-TCP/MnO 2 /PCL(β-TMP) by electrospinning with a function of catalyzing decomposition of H 2 O 2 . We examined the surface morphology of β-TMP, the concentration of Ca, P and Mn of β-TMP, as well as the diameter distribution range of nanofibers on β-TMP. Through X-ray diffraction patterns and Fourier transform infrared spectra, β-TMP was characterized and its hydrophilicity was tested. The release of Mn 2+ and Ca 2+ of 0.1 and 0.05% β-TMP in different pH values (7.4 and 5.5) determined by ICP. We also identified that β-TMP could reduce the level of ROS in cells by lowering the level of H 2 O 2 . 0%, 0.05% and 0.1% β-TMP displayed good cell compatibility, cell adhesion and cellular morphology in the condition with or without H 2 O 2 . 0.5% β-TMP showed compromised cell compatibility in normal condition, however, the compromised phenotypes could be partially rescued in the present of H 2 O 2 . Compared with 0%, 0.05% and 0.1% β-TMP displayed higher osteoblastic differentiation with or without H 2 O 2 in BMSCs as well as in MG-63. In sum, β-TMP helped osteogenesis and promoted repair of bone defects.
