In vivo mechanism of action of matrix metalloprotease (MMP) in the autolysis of sea cucumber (Stichopus japonicus)

Zi-qiang Liu,Da-yong Zhou,Yu-xin Liu,Xiao-yang Liu,Yan Liu,Bing Liu,Liang Song,Fereidoon Shahidi

Matrix metalloprotease (MMP) is the key endogenous protease that boosts the autolysis of sea cucumber, while its action and mechanism in vivo are still unclear. MMP inhibitor (1,10-phenanthroline) was injected into coelomic cavity of sea cucumber ( Stichopus japonicus ) and its effects on autolysis and mechanism of action were investigated for the first time by using scanning electron microscopy, multi-functional in vivo imaging system, and confocal laser scanning microscopy. In response to external stimuli, the level of Ca 2+ in live sea cucumber dermis increased, which may activate MMP activity. MMP inhibitor could diffuse from coelomic cavity into body wall and significantly prevented the activation of MMP by chelating Ca 2+ , and thus, retarded the autolysis of fresh sea cucumber by preventing degradation of interfibrillar proteoglycan bridges between adjacent collagen fibrils. This work provides information toward understanding the mechanism behind the sea cucumber autolysis and demonstrate a potential approach to prevent the autolysis. Practical applications Autolysis occurs in sea cucumber in response to external stimulation. Matrix metalloproteinase (MMP) has been found to be the key endogenous protease that boosts the autolysis of sea cucumber body wall (SCBW). However, the role of MMP in the autolysis of SCBW in vivo and the mechanisms involved remain unclear. In this study, MMP inhibitor (1,10-phenanthroline) was injected into the body cavity of live sea cucumber, and its effects on in situ changes in SCBW were studied. Our results indicated that the activity of endogenous MMP in live SCBW was activated in response to external stimulation, which may be responsible for the autolysis. After injection into coelomic cavity, MMP inhibitor could diffuse into SCBW and significantly reduced the progress of typical morphological and microstructural changes of autolytic sea cucumber. Through chelating Ca 2+ , the MMP inhibitor could prevent the activation of MMP, and thus reduced the autolysis of fresh sea cucumber.

Determination of newly synthesized dihydroxylated polybrominated diphenyl ethers in sea fish by gas chromatography-tandem mass spectrometry

Mengtao Zhang,Wei Guo,Juntong Wei,Jianghong Shi,Jiawei Zhang,Hui Ge,Huanyu Tao,Xiaowei Liu,Qing Hu,Zongwei Cai

Dihydroxylated polybrominated diphenyl ethers (diOH-PBDEs) can be natural products of marine organisms or the metabolites of PBDEs. The optimal determination method and concentration of diOH-PBDEs in seafood are unknown due to a lack of commercially available standards. In the present study, diOH-PBDEs were synthesized, and an efficient measurement method for OH-PBDEs and diOH-PBDEs in sea fish muscle samples, including extraction, clean-up and gas chromatography-tandem mass spectrometry (GC-MS/MS) analysis, was established. Pressurized liquid extraction (PLE) followed by partitioning with a KOH solution and florisil cartridge clean-up proved to be a reliable and robust method for detecting all OH-PBDEs/diOH-PBDEs. GC-MS/MS with an electron ionization (EI) source analysis was a sensitive analytical instrument for OH-PBDEs/diOH-PBDEs. The recovery using this method ranged from 19% to 101%, 28%–88% and 42%–90% for 10 ng, 20 ng and 40 ng spiking levels, respectively. The equipment detection limits (EDLs) were in the range of 0.31–2.78 pg/μL, and the limits of detection (LOD) for the method were in the range of 5.07–38.74 pg/g wet weight. Concentrations of diOH-PBDEs in the marine fish muscle samples were in the range of 32.43–1528.63 pg/g wet weight. Similar compositions of OH-PBDEs/diOH-PBDEs were found within the same family of marine fish.

Sea-island nanostructured polyvinylidene fluoride/zeolitic imidazolate framework-8 polyelectrolyte for high-performance all-solid-state supercapacitors

Chao Lu,Xi Chen

All-solid-state supercapacitors with high power density, lightweight and durability have shown enormous potential for sustainable energy applications . However, the insufficient mechanical and electrochemical properties of polyelectrolyte membranes restrict their energy storage performances. Here, we report a sea-island nanostructured polyvinylidene fluoride/zeolitic imidazolate framework-8 (PVDF/ZIF-8) polyelectrolyte with superior mechanical and ionic properties for boosting energy storage performances of the supercapacitors. ZIF-8 nanoparticles with 8% content were dispersed uniformly into PVDF substrate and improved ionic liquid uptake of the membrane (from 42% to 74%) because of its high surface area (764 m 2  g −1 ) and porous structure (pore width of 2.1 nm). Meanwhile, plastication effect of ZIF-8 materials improved mechanical properties of the membrane with tensile modulus of 2.88 MPa and break elongation of 11.6%. The PVDF/ZIF-8 based supercapacitors delivered a high specific capacitance of 156 F g −1 at current density of 1 A g −1 and showed good rate capability at current densities from 1 to 10 A g −1 . The device could keep working over 20000 cycles at 1 A g −1 with capacitance retention of 91.7%, showing excellent cycling stability and enormous potential for applications. This work sheds light on design of novel polyelectrolyte membranes with high electrochemical and mechanical properties for long life energy storage devices.

Insights into the Headgroup and Chain Length Dependence of Surface Characteristics of Organic-Coated Sea Spray Aerosols

Shumin Cheng,Siyang Li,Narcisse T. Tsona,Christian George,Lin Du

The structure of sea spray aerosols (SSAs) has been described as a saline core coated by organic surfactants. The presence of surface-active compounds at the interface can exert significant effect on physical, chemical, and optical properties of SSAs. The surfactant molecules chosen for this study, palmitic acid (PA), stearic acid (SA), arachidic acid (AA), methyl palmitate (MP), methyl stearate (MS), and methyl arachidate (MA), were used to explore the effect of alkyl chain length, headgroups, and sea salts on the surface properties of these monolayers. Surface pressure–area (π–A) isotherms obtained with a Langmuir trough was used for revealing macroscopic phase behavior of the monolayers. Moreover, infrared reflection absorption spectroscopy (IRRAS) was employed to investigate the interfacial organization at the molecular level. The π–A isotherms indicated that sea salts, present in the subphase, have an intense condensing effect on fatty acid monolayers while exerting an expanding effect on fatty acid methyl ester monolayers, which was confirmed by results from IRRAS experiments. IRRAS further uncovered the all-trans conformation of the hydrocarbon chains, which can be evidenced by the relatively low νa(CH2) and νs(CH2) stretching vibrations. The conformational order changes in the alkyl chains of different film-forming species (C16 < C18 < C20) were directly revealed by analyzing the intensity ratios of the νa(CH2) and νs(CH2) bands. Thus, all three factors alter the phase behavior and molecular packing of the monolayers at the air–aqueous interface.

Enantioselective resolution of (±)-1-phenylethyl acetate by extracellular proteases from deep-sea bacterium Bacillus sp. DL-2

Lu Dong,Yongkai Xu,Yun Zhang,Aijun Sun,Yunfeng Hu

Chiral 1-phenylethanol and its ester derivative are important chiral chemicals in diverse industries and the preparation of those optically pure enantiomers is of great importance. One bacterium, Bacillus sp. DL-2, whose extracellular proteases could efficiently asymmetrically hydrolyse (±)-1-phenylethyl acetate, was isolated from the deep sea of the Western Pacific. After the immobilization of extracellular proteases and the optimization of enzymatic reactions, (R)-1-phenylethanol was prepared with the enantiomeric excess (e.e.) being 97% and the yield being 41%, respectively. The optimal resolution reaction condition for the preparation of (R)-1-phenylethanol using immobilized extracellular proteases was found to be 5-mM (±)-1-phenylethyl acetate, 360 mg/mL immobilized extracellular proteases, pH 6.5, and 20 °C for 2 h. (S)-1-phenylethyl acetate was generated through enzymatic kinetic resolution with the e.e. being as high as 99% and the yield being 71%, respectively. The optimal resolution reaction condition for the preparation of (S)-1-phenylethyl acetate was found to be 2.5-mM (±)-1-phenylethyl acetate, 440 mg/mL immobilized extracellular proteases, pH 7.5, and 35 °C for 10 h. Our report is the first report about the kinetic resolution of (±)-1-phenylethyl acetate using proteases and the enantio-preference of the proteases was found to be the same as those of most other esterases/lipases. Also notably, the optical purity of (S)-1-phenylethyl acetate generated through kinetic resolution using the proteases of Bacillus sp. DL-2 was the highest report so far. Proteases from deep-sea Bacillus sp. DL-2 are new contributions to the biocatalyst library for the preparation of valuable chiral alcohols and chiral esters through kinetic resolution.

Effects of chitosan-gentianic acid derivatives on myofibrillar proteins in sea bass (Lateolabrax maculatus) during refrigerated storage

Weiqing Lan,Mingxing Zhou,Bingjie Zhang,Shucheng Liu,Jing Xie

Phenolic acid-chitosan derivatives have received extensive attention due to their greatly enhanced mechanical, antibacterial and antioxidant properties, especially in food preservation. The chitosan-gentianic acid (CS-g-GA) was prepared and its impact on myofibrillar proteins (MPs) in sea bass ( Lateolabrax maculatus ) during refrigerated storage was investigated in this study. Fish fillets were immersed in distilled water, CS, GA and CS-g-GA solutions, respectively, followed by an 18-day refrigerated storage. MPs were extracted from fish fillets every 3 days, and then the degree of oxidation and conformational changes of MPs were evaluated. The results of myofibril fragmentation index (MFI) and surface hydrophobicity confirmed that CS-g-GA can retard the fragmentation and degradation of myofibrils. Additionally, the total sulfhydryl content of CS-g-GA-treated samples in the later storage was significantly higher than those of other groups, confirming that CS-g-GA treatment protected the structure and function of protein. Results of Fourier transform infrared spectroscopy (FT-IR) and inner fluorescence intensity (IFI) showed that CS-g-GA maintained the secondary and tertiary structure of MPs. The results from scanning electron microscope (SEM) further clarified that CS-g-GA can maintain the tight arrangement of muscle fibers and protect the microstructure of myofibrils. Furthermore, the observation of confocal laser scanning microscopy (CLSM) demonstrated that CS-g-GA can prevent protein cross-linking and aggregation. Therefore, CS-g-GA treatment can effectively protect the structure and delay the oxidation of MPs.

Construction of a Biomimetic Tubular Scaffold Inspired by Sea Sponge Structure: Sponge-Like Framework and Cell Guidance

Si Meng,Nihuan Wu,Jie Fang,Yidan Yu,Xin Tang,Yihan Wang,Xiaokang Deng,Cheng Qi,Tiantian Kong,Tengda Ding,Zhou Liu

Engineering hollow fibers with precise surface microstructures is challenging; yet, essential for guiding cells alignment and ensuring proper vascular tissue function. Inspired by Euplectella sponges, a novel strategy to engineer biomimetic hollow fibers with spiral surface microstructures is developed. Using oxidized bacterial cellulose, bacterial cellulose, and polydopamine, a “brick-and-mortar” scaffold is created through precise shear control during microfluidic coaxial spinning. The scaffold mimics natural extracellular matrices, providing mechanical stability and supporting cell growth. In vitro studies show successful co-culture of endothelial cells (ECs) and smooth muscle cells (SMCs), with SMCs aligning along spiral surface microstructures and ECs forming a confluent inner layer. In vivo implantation confirms biocompatibility, biodegradability, and low immunogenicity. This Euplectella -inspired scaffold presents a promising approach for vascular tissue engineering and regenerative medicine.

Deep-Sea-Derived Isobisvertinol Targets TLR4 to Exhibit Neuroprotective Activity via Anti-Inflammatory and Ferroptosis-Inhibitory Effects

Zi-Han Xu,Ming-Min Xie,Chun-Lan Xie,Xian-Wen Yang,Jun-Song Wang

Neuroinflammation and neuronal cell death are leading causes of death in the elderly and underlie various neurodegenerative diseases. These diseases involve complex pathophysiological mechanisms, including inflammatory responses, oxidative stress, and ferroptosis. Compounds derived from deep-sea fungi exhibit low toxicity and potent neuroprotective effects, offering a promising source for drug development. In this study, we isolated 44 natural products from deep-sea-derived fungi and identified isobisvertinol (17) as a compound with anti-inflammatory and ferroptosis-inhibiting effects. Using LPS-induced microglial inflammation and RSL3-induced neuronal ferroptosis models, we found that17targets TLR4 to provide neuroprotection. Molecular docking studies revealed that17inhibits TLR4 activation by occupying the hydrophobic pocket at the TLR4-MD2 binding site. Additionally,17suppresses TLR4, reducing p38 MAPK phosphorylation, and inhibits ferroptosis by decreasing lipid peroxidation and modulating mitochondrial membrane potential. Metabolomic analysis showed that17rescues alterations in multiple metabolic pathways induced by RSL3 and increases levels of antioxidant metabolites, including glutamine, glutamate, and glutathione. In summary, our results indicate that isobisvertinol (17) targets TLR4 in neural cells to reduce inflammation and inhibit p38 MAPK phosphorylation, while regulating metabolic pathways, mainly GSH synthesis, to provide antioxidant effects and prevent ferroptosis in neurons.

A Highly Sensitive 3D-Printed Flexible Sensor for Sensing Small Pressures in Deep-Sea High-Pressure Environment

Siqi Hu,Wuxu Zhang,Shengbin Li,Yuwei Wang,Zhiyi Gao,Xiangling Xia,Huiyun Xiao,Qi Zhang,Dan Xu,Feng Xu,Jinyun Liu,Baoru Bian,Yuanzhao Wu,Yiwei Liu,Jie Shang,Run-Wei Li

The origin of life on Earth is believed to be from the ocean, which offers abundant resources in its depths. However, deep-sea operations are limited due to the lack of underwater robots and rigid grippers with sensitive force sensors. Therefore, it is crucial for deep-sea pressure sensors to be integrated with mechanical hands for manipulation. Here, a flexible stress sensor is presented that can function effectively under high water pressure in the deep ocean. Inspired by biological structures found in the abyssal zone, our sensor is designed with an internal and external pressure balance structure (hollow interlocking spherical structure). The digital light processing (DLP) three-dimensional (3D) printing technology is utilized to construct this complex structure after obtaining optimized structural parameters using finite element simulation. The sensor exhibits linear sensitivity of 0.114 kPa–1 within the range of 0–15 kPa and has an extremely short response time of 32 ms, good dynamic-static load response capability, and excellent resistance cycling stability. It shows high sensitivity of 1.74 kPa–1 even under 30 MPa static water pressures and the theoretical working pressure can exceed 110 MPa, which provides a new solution for deep-sea robots.

Sea-Island Micelle Structured Hydrogel Scaffold: A Dual-Action Approach to Combat Cartilage Damage under RA Conditions

Abudureheman Bahatibieke,Jianming Zhao,Danping Fan,Zixiang Zhou,Junfei Li,Xing Wang,Hongyan Zhao,Tianyang Wang,Ziyuan Fang,Yajie Xie,Cheng Huang,Cheng Xiao,Yudong Zheng

Rheumatoid arthritis (RA) is a common autoimmune joint disease characterized by persistent synovial inflammation and cartilage damage. The current clinical treatments primarily utilize drugs such as triptolide (TP) to address inflammation, yet they are unable to directly repair damaged cartilage. Furthermore, the persistent inflammation often undermines the effectiveness of traditional cartilage repair strategies, preventing them from achieving optimal outcomes. To tackle this challenge, this study successfully developed a drug-loaded polyurethane hydrogel-oriented porous scaffold, designed to address persistent inflammation and facilitate cartilage repair under RA conditions. A drug-loaded hydrogel was formed via solvent-induced polyurethane-gelatin, resulting in the scaffold TP@GSPU. The sea-island micelle structure of TP@GSPU enables efficient loading of TP. The release of TP in the in vivo environment regulates the expression of inflammatory factors in macrophages, thereby improving the inflammatory microenvironment within the joint cavity. Additionally, the gelatin component of the scaffold provides robust support for cartilage regeneration. The efficacy of the TP@GSPU in regulating the inflammatory microenvironment and facilitating cartilage repair under RA conditions, which was demonstrated through cartilage damage repair experiments conducted in a rat collagen-induced arthritis (CIA) model. The design scheme of this material offers a potential approach to cartilage repair in the conditions of RA.

In silico assessment and structural characterization of antioxidant peptides from major yolk protein of sea urchin Strongylocentrotus nudus

Wen-Hui Shang,Yue Tang,Sheng-Yi Su,Jia-Run Han,Jia-Nan Yan,Hai-Tao Wu,Bei-Wei Zhu

Sea urchin gonads have been demonstrated to contain major yolk protein (MYP), which can be hydrolyzed by enzymes to release biologically active peptides. The in silico analysis of the MYP sequence in the BIOPEP database showed the presence of fragments with antioxidant activity. The sequence was hydrolyzed by 21 kinds of proteases and 23 antioxidant peptides were obtained. Eight peptides, including Leu-Trp (LW), Arg-Trp (RW), Ala-Trp (AW), Thr-Trp (TW), Ala-Asp-Phe (ADF), Leu-Trp-Lys (LWK), Ser-Asp-Phe (SDF) and Leu-Tyr (LY), were screened and a score over 0.5 was obtained using PeptideRanker. The peptides LW, TW and LWK showed a stronger antioxidant capacity with IC50 values of 8.85, 9.59 and 9.62 mmol L−1, respectively, compared to that of glutathione (10.81 mmol L−1). Furthermore, AW, LW and LY showed Trolox equivalent antioxidant capacity (TEAC) values of 3.07, 1.87 and 1.52 mmol TE per mmol peptide, respectively. These results suggest that the MYP from sea urchin (S. nudus) gonads is a good source of antioxidant peptides with abundant tryptophan.

Sea Anemone-Inspired Fluorosilicone Polyurethane Coating with Synergistic Low-Surface-Energy and Cationic Antibacterial Action for Static Antifouling

Shuiwang Jiang,Yuyi Zhu,Xiangfeng Chen,Hongyi Liu,Xuezhi Jiang,Yahao Zhang,Hui Gong,Ting Huang,Dengfeng Zeng,Quan Liu

Conventional polydimethylsiloxane (PDMS)-based antifouling coatings, despite their inherent fouling-release capability, exhibit critically insufficient antifouling performance under static seawater conditions. Inspired by the synergistic physical–chemical defense strategy of sessile marine organisms, specifically sea anemones, which combine a physical mucus barrier with antimicrobial peptide secretion, the present work develops a multi-mechanism hybrid coating—designated as sea anemone-inspired fluorosilicone polyurethane—that integrates low-surface-energy physical antifouling and cationic antibacterial chemical antifouling. This coating system is constructed from silicone polyurethane (PDMS-PU), a cationic antibacterial moiety (PDMS-N+), and fluorinated functional monomers. Through systematic compositional optimization, an optimal formulation (P-4) is identified, which achieves a fracture elongation of 78.19%, a normal adhesion strength of approximately 2.5 MPa, a water contact angle of 120°, and a surface energy of 12.86 mN/m. Notably, its antibacterial rates against both Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) exceed 95%. The resultant coating uniquely synergizes low surface energy, potent antibacterial activity, excellent mechanical properties, and thermal stability, thereby enabling long-term and stable antifouling performance in static seawater environments. This work provides a crucial technological foundation for the engineering application and industrialization of green, durable marine antifouling coatings.
