A novel colorimetric strategy based on canine IgM for ultrasensitive and selective detection of Staphylococcus aureus

Yun Zhang,Yibing Zhang,Ruihua Lun,Qingshan Fu,Yuqiao Chang,Jiansen Du,Yi Zhang,Junping Yu

Conventional methods for Staphylococcus aureus ( S. aureus ) detection using mammalian immunoglobulins could yield false positive results, owing to the presence of the protein G-producing Streptococcus . In this study, because canine IgM can bind to protein A but not to protein G, we firstly designed a colorimetric platform based on canine IgM to identify S. aureus . Magnetic beads (MBs) functionalized with canine IgM acted as captors to enrich S. aureus , relying on the binding interaction between the Fc region of canine IgM and protein A in the bacterial cell wall, while horseradish peroxidase (HRP)-labeled rabbit anti- S. aureus IgG served as a tracer in the sandwich-type immunoassay, catalyzing the colorimetric reaction. Under optimal conditions, the proposed strategy achieved a satisfactory analytical performance with a broad linear range from 1.6 × 10 3  CFU/mL to 1.0 × 10 5  CFU/mL, a colorimetric minimum resolution of as low as 1.4 × 10 2  CFU/mL in 100 μL phosphate-buffered saline (PBS), and good selectivity in differentiating target from protein G-producing Streptococcus . Moreover, the proposed approach successfully identified S. aureus in various application scenarios (except for samples containing IgG), can be completed in less than 90 min, and was not dependent on any specialized equipment.

A high electron mobility transistor biosensor-based GaN for facile and sensitive detection of Staphylococcus aureus

Shengjie Hu,Yin Zou,Xuecheng Jiang,Yuyan Weng,Yu Liu,Xue Tang,Guofeng Yang,Naiyan Lu

Staphylococcus aureus is a common foodborne pathogen known to cause infections and poisoning. Hence, there is a pressing need to establish a rapid and sensitive detection method to mitigate the risk of bacterial contamination. Gallium nitride-based high electron mobility transistors (GaN HEMTs), representing emerging semiconductor sensors, have garnered significant research attention due to their advantages in integration, miniaturization, and rapid sensitivity. In recent years, the device has demonstrated excellent performance in detecting gases, heavy metals, and biomarkers. This paper prepares a novel GaN HEMT sensor with aptamer modification that can be applied to the detection of Staphylococcus aureus , which has great potential for application in food safety detection. The linear detection range of the functionalized device is 10 2  CFU/mL–10 7  CFU/mL, and the detection limit was 19 CFU/mL. In terms of specificity, interfering bacteria such as E. coli produced only 9.6 %–16.7 % of the current signal generated by S. aureus . When tested with actual samples, the device showed consistent detection results compared to those in the buffer system (95.2 %–97.2 %). This work demonstrates performance that is comparable to or even superior to other methods in terms of detection limits and response speed. Additionally, the HEMT sensor stands out by eliminating the need for supplementary reference electrodes or complex systems. With its rapid detection time, user-friendly operation, and exceptional selectivity, this sensor shows great promise for practical applications, particularly in field testing scenarios.

Simultaneous detection of Staphylococcus aureus and Salmonella typhimurium using multicolor time-resolved fluorescence nanoparticles as labels

Xiaole Wang,Yukun Huang,Shijia Wu,Nuo Duan,Baocai Xu,Zhouping Wang

Foodborne illnesses caused by Staphylococcus aureus and Salmonella typhimurium are common public health issues worldwide, affecting both developing and developed countries. In this study, aptamers labeled with multicolor lanthanide-doped time-resolved fluorescence (TRFL) nanoparticles were used as signal probes, and immobilized by Fe 3 O 4 magnetic nanoparticles were used as the capture probes. The signal probes were bonded onto the captured bacteria by the recognition of aptamer to form the sandwich-type complex. Under the optimal conditions, TRFL intensity at 544   nm was used to quantify S. typhimurium (y   =   10,213   ×   −   12,208.92, R 2   =   0.9922) and TRFL intensity at 615   nm for S. aureus (y   =   4803.20   ×   −   1933.87, R 2   =   0.9982) in the range of 10 2 –10 5   CFU/ml. Due to the magnetic separation and concentration of Fe 3 O 4 nanoparticles, detection limits of the developed method were found to be 15, 20   CFU/ml for S. typhimurium and S. aureus , respectively. The application of this bioassay in milk was also investigated, and results were consistent with those of plate-counting method. Therefore, this simple and rapid method owns a great potential in the application for the multiplex analysis in food safety.

Synergistic antibacterial effect of Bi2S3 nanospheres combined with ineffective antibiotic gentamicin against methicillin-resistant Staphylococcus aureus

Lulu Ma,Jie Wu,Shilei Wang,Hao Yang,Donghui Liang,Zhong Lu

In this paper, Bi 2 S 3 nanospheres with size of 212   nm were prepared by a simple hydrothermal process. The selectively enhanced antibacterial effects of Bi 2 S 3 nanospheres with three classes of ineffective antibiotics, β-lactam (cefuroxime, CXM; cefotaxime , CTX and piperacillin , PIP), quinolone (ciprofloxacin, CIP) and aminoglycoside (gentamicin, GEN) against clinical isolated methicillin-resistant Staphylococcus aureus (MRSA) were investigated for the first time. GEN shows significantly synergistic growth inhibition against MRSA when combined with Bi 2 S 3 nanospheres, while CXM, CTX, PIP and CIP do not. Raman spectroscopy and Z potential studies reveal that Bi 2 S 3 could interact with GEN and the combination showed small electronegativity , which probably induced the increase of GEN content in cytoplasm of bacteria. Furthermore, the combination of Bi 2 S 3 nanospheres and GEN can destroy the bacterial membrane function and induce more bactericidal reactive oxygen generation than that of Bi 2 S 3 or GEN alone. The cytotoxicity test indicates that the combination of Bi 2 S 3 and GEN presented low toxicity to human normal hepatocyte L02. This work shows that Bi 2 S 3 nanospheres can be used to enhance the action of ineffective antibiotic GEN against MRSA, thus strengthening the antibiotic capacity for fighting MRSA infections.

Highly sensitive and selective colorimetric determination of Staphylococcus aureus via chicken anti-protein A IgY antibody

Yun Zhang,Wenqing Tan,Lin Zhang,Shuyou Shi,Yuna Niu,Xue Yang,Jinjuan Qiao,Hui Wang

Staphylococcus aureus (S. aureus) is a food-borne pathogenic organism that causes an array of infectious diseases ranging from superficial skin infections to potentially fatal diseases. Thus quantitative detection of S. aureus with high selectivity and sensitivity is of great significance. In this study, Staphylococcus protein A (SPA), which is specifically secreted on the surface of S. aureus, was selected as an epitope. On the basis of the specific recognition of SPA by chicken anti-protein A immunoglobulin Y (IgY), a facile colorimetric immunoassay for the detection of S. aureus was established. To the best of our knowledge, this is the first time chicken anti-protein A IgY has been utilized as an antibody pair for the quantitative detection of S. aureus. Chicken anti-protein A IgY was conjugated with magnetic particles for bacterial capture. Then horseradish peroxidase (HRP) labeled chicken anti-protein A IgY was employed as the revealing partner to perform a sandwich format assay, followed by an HRP catalyzed colorimetric reaction. Optimized detection conditions produced a calibration curve for S. aureus ranging from 7.8 × 102 CFU mL−1 to 2.0 × 105 CFU mL−1 with a detection limit of 2.3 × 102 CFU mL−1 (3σ) in phosphate-buffered saline (PBS). The detection limits (3σ) in spiked samples were very close to those in PBS. The entire assay was accomplished in less than 60 min and no cross-reactivity with the other tested bacterial species was observed. These results demonstrate that the proposed method is effective for rapid and selective screening of S. aureus in food samples.

Nontoxic Carbon Quantum Dots/g-C3N4 for Efficient Photocatalytic Inactivation of Staphylococcus aureus under Visible Light

Chenyi Tang,Chao Liu,Yu Han,Qiaoqi Guo,Wei Ouyang,Huajun Feng,Meizhen Wang,Feng Xu

The widespread use of antibiotics has caused the rapid emergence of antibiotic-resistant bacterial strains and antibiotic resistance genes in the past few decades. Photocatalytic inactivation, a promising approach for the killing of pathogens, efficiently avoids the problems induced by antimicrobial drugs. However, traditional photocatalysts usually have some disadvantages, such as high costs of raw materials, ultraviolet ray excitation, and potential leaching of toxic metals. Here, a metal-free heterojunction photocatalyst, denoted as CQDs/g-C 3 N 4 , is synthesized through incorporating carbon quantum dots (CQDs) on graphitic carbon nitride (g-C 3 N 4 ), which significantly enhances photocatalytic inactivation of Staphylococcus aureus ( S. aureus ) compared with pure g-C 3 N 4 in vitro. CQDs/g-C 3 N 4 causes a rapid increase of intracellular reactive oxygen species levels and destruction of cell membranes under visible light, eventually leading to death of bacteria. The efficacy of CQDs/g-C 3 N 4 is further examined by a mouse cutaneous infection model of S. aureus . CQDs/g-C 3 N 4 markedly reduces the bacterial loads and prompts lesion recovery in mice, as compared with g-C 3 N 4 -treated group. In vivo and in vitro toxicity analyses show that the side effects of CQDs/g-C 3 N 4 are negligible. Considering the efficient photocatalytic inactivation and nontoxicity of CQDs/g-C 3 N 4 , this visible-light-driven photocatalyst paves a brand new avenue for the treatment of S. aureus infection.

An antioxidant and antibacterial polydopamine-modified thermo-sensitive hydrogel dressing for Staphylococcus aureus-infected wound healing

Pengjin Ge,Shuhua Chang,Ting Wang,Quan Zhao,Gang Wang,Bin He

Bacteria-infected wound healing is a complex and chronic process that poses a great threat to human health. A thermo-sensitive hydrogel that undergoes a sol–gel transition at body temperature is an attractive wound dressing for healing acceleration and infection prevention. In this paper, we present a thermo-sensitive and reactive oxygen species (ROS)-scavenging hydrogel based on polydopamine modified poly(ε-caprolactone-co-glycolide)-b-poly(ethylene glycol)-b-poly(ε-caprolactone-co-glycolide) (PDA/P2) triblock copolymer. The PDA/P2 solution at a concentration of 30 wt% could form a gel at 34–38 °C. The ROS-scavenging ability of PDA/P2 was demonstrated by DPPH and ABTS assays and intracellular ROS downregulation in RAW264.7 cells. Furthermore, silver nanoparticles were encapsulated in the hydrogel (PDA/P2–4@Ag gel) to provide antibacterial activity against E. coli and S. aureus. An in vivo S. aureus-infected rat model demonstrated that the PDA/P2–4@Ag hydrogel dressing could promote wound healing via inhibiting bacterial growth, alleviating the inflammatory response, and inducing angiogenesis and collagen deposition. This study provides a new strategy to prepare temperature-sensitive hydrogel-based multifunctional wound dressings.

Enzyme activated photodynamic therapy for methicillin-resistant Staphylococcus aureus infection both inv itro and in vivo

Xiu-Jun Fu,Yun-Qing Zhu,Yin-Bo Peng,You-Shuang Chen,Yi-Ping Hu,Hua-Xiang Lu,Wei-Rong Yu,Yong Fang,Jian-Zhong Du,Min Yao

In recent years, methicillin-resistant Staphylococcus aureus (MRSA) has become one of the most common multi-drug resistant bacteria in both hospital and community. The aim of this study is to investigate the selective inhibition of MRSA by a modified photosensitizer (LAEtNBS) in vitro and the efficacy of MRSA infection treatment by photodynamic therapy (PDT) with LAEtNBS in vivo . LAEtNBS was synthesized by adding a cationic photosensitizer molecule (EtNBS-COOH) and a quencher molecule to two side chains of cephalosporin , which was then shown to have similar absorption and emission wavelengths with EtNBS-COOH, but suppressed yields of fluorescence quantum and singlet oxygen . The selective inactivation and less phototoxicity of LAEtNBS, compared to that of EtNBS-COOH, were assessed and confirmed by conducting PDT to two Staphylococcus aureus strains and human skin cells at a fluence of 15   J/cm 2 with 640   ±   10   nm LED light. Furthermore, using mouse skin wound model infected with 10 8 CFU of MRSA, we found that both LAEtNBS and EtNBS-COOH were able to treat MRSA infection and enhance wound repair. However, there was no significant difference in the two photosensitizers that might be due to the environment in vivo . Modification of the photosensitizer will be very beneficial for developing new strategies to treat drug resistant bacterial infection with less harm to host tissue.

NIR-II Photo-Amplified Sonodynamic Therapy Using Sodium Molybdenum Bronze Nanoplatform against Subcutaneous Staphylococcus Aureus Infection

Xiaojun He,Ji-Ting Hou,Xiaoshuai Sun,Paramesh Jangili,Jusung An,Yuna Qian,Jong Seung Kim,Jianliang Shen

Ultrasound (US)-mediated sonodynamic therapy (SDT) has the advantages of non-invasiveness and deep tissue penetration. Nanosystems are prominently used in sonosensitization; however, most nano-sonosensitizers have a low reactive oxygen species (ROS) yield, thus restraining the application of SDT. Sodium molybdenum bronze nanoparticles (SMB NPs) with rich oxygen vacancies are developed and interlayer gaps of molybdenum trioxide nanobelts are expanded. Owing to the increased oxygen vacancy density and wide interlayer gap-induced narrower band gap of SMB NPs, the electrons (e – ) and holes (h + ) generated by US are separated more rapidly, and oxygen vacancies prevent electrons–holes recombination under US irradiation. SMB NPs exhibit a second near-infrared (NIR-II) photothermal effect to promote the generation of ROS by the sonosensitizer. The SMB NPs system is successfully realized to eliminate Staphylococcus aureus ( S. aureus ) and dissipate biofilm. Therefore, multimodal therapy using SMB NPs serves as an effective and promising regimen for deep-seated bacterial infections. The newly developed Mo-based sonosensitizer is presented for the first time to demonstrate excellent antimicrobial activity through hyperthermia-promoting SDT therapeutics. This work proposes a novel strategy in the field of NIR-II photo-amplified SDT with Mo-based materials for bacterial eradication and other important biomedical applications.

Bactericidal and antibiofilm properties of Rumex japonicus Houtt. on multidrug-resistant Staphylococcus aureus isolated from milk

G. Kim,Y.J. Xu,A.K. Farha,Z.Q. Sui,H. Corke

ABSTRACT Multidrug-resistant (MDR) Staphylococcus aureus and its biofilm formation have been challenging to control in milk and dairy industries. Biofilms formed by Staph. aureus may result in the failure of antibacterial agents and disinfectants to penetrate the biofilm in an attempt to control contamination. Novel natural antibacterial agents are required to combat MDR bacteria and biofilms. In this study, we evaluated the bactericidal, antibiofilm, and antimotility effects of Rumex japonicus Houtt. (RJH) extract on MDR Staph. aureus isolated from milk. The RJH extract exhibited good antibacterial activity against MDR strains with minimum inhibitory concentrations (MIC) ranging from 0.78 to 6.25 mg/mL and minimum bactericidal concentrations ranging from 3.125 to 12.5 mg/mL. The extract showed strong inhibition of biofilm formation (81.9%) at sub-MIC value and eradication of biofilm at higher concentrations. The motility of Staph. aureus was effectively blocked by the extract. Major compounds emodin, chrysophanol, and physcion were identified in RJH extract using HPLC-linear trap quadrupole (LTQ)/Orbitrap-mass spectrometry. The extract was nontoxic to human epithelial cell lines such as Caco-2 and HT-29 cell lines at concentrations ranging from 0.1 to 0.5 mg/mL, and from 0.1 to 0.75 mg/mL, respectively. These findings suggest that RJH extract could be an alternative to synthetic preservatives in milk and dairy products.

Integrated label-free erbium-doped fiber laser biosensing system for detection of single cell Staphylococcus aureus

Jiandong Liu,Bin Liu,Juan Liu,Xing-Dao He,Jinhui Yuan,Zabih Ghassemlooy,Hamdi Torun,Yong-Qing Fu,Xuewu Dai,Wai Pang Ng,Richard Binns,Qiang Wu

A critical challenge to realize ultra-high sensitivity with optical fiber interferometers for label free biosensing is to achieve high quality factors (Q-factor) in liquid. In this work a high Q-factor of 10 5 , which significantly improves the detection resolution is described based on a structure of single mode -core-only -single mode fiber (SCS) with its multimode (or Mach-Zehnder) interference effect as a filter that is integrated into an erbium-doped fiber laser (EDFL) system for excitation. In the case study, the section of core-only fiber is functionalized with porcine immunoglobulin G (IgG) antibodies, which could selectively bind to bacterial pathogen of Staphylococcus aureus ( S. aureus ). The developed microfiber-based biosensing platform called SCS-based EDFL biosensors can effectively detect concentrations of S. aureus from 10 to 10 5  CFU/mL, with a responsivity of 0.426 nm wavelength shift in the measured spectrum for  S. aureus  concentration of 10 CFU/mL. The limit of detection (LoD) is estimated as 7.3 CFU/mL based on the measurement of S. aureus with minimum concentration of 10 CFU/mL. In addition, when a lower concentration of 1 CFU/mL is applied to the biosensor, a wavelength shift of 0.12 nm is observed in 10% of samples (1/10), indicating actual LoD of 1 CFU/mL for the proposed biosensor. Attributed to its good sensitivity, stability, reproducibility and specificity, the proposed EDFL based biosensing platform has great potentials for diagnostics.

Antibacterial effect of protease-responsive cationic eugenol liposomes modified by gamma-polyglutamic acid against Staphylococcus aureus

Xiaochen Chen,Yiwei Wang,Changzhu Li,Zichun Hua,Haiying Cui,Lin Lin

Eugenol, as a natural antibacterial agent, has been widely studied for its inhibitory effect on the common food-borne pathogen Staphylococcus aureus (S. aureus). However, the widespread application of eugenol is still limited by its instability and volatility. Herein, γ-polyglutamic acid coated eugenol cationic liposomes (pGA-ECLPs) were successfully constructed by self-assembly with an average particle size of 170.7 nm and an encapsulation efficiency of 36.2%. The formation of pGA shell significantly improved the stability of liposomes, and the encapsulation efficiency of eugenol only decreased by 20.7% after 30 days of storage at 4 °C. On the other hand, the pGA layer can be hydrolyzed by S. aureus, achieving effective control of release through response to bacterial stimuli. The application experiments further confirmed that pGA-ECLPs effectively prolonged the antibacterial effect of eugenol in fresh chicken without causing obvious sensory effects on the food. The above results of this study provide an important reference for extending the action time of natural antibacterial substances and developing new stimuli-responsive antibacterial systems.
