Self-Assembled Polymeric Nanoparticles for Redox- and Gene-Regulated Osteoarthritis Therapy

Bing Wu,Ye Yang,Xiancheng Huang,Yuanchao Shi,Chongzhou Fang,Fei Liu,Yang-Chi-Dung Lin,Jiaxin Wang,Jianjing Lin,Jing Mu,Xintao Zhang

Osteoarthritis (OA) is a progressive degenerative joint disease characterized by synovial inflammation and cartilage degradation, and its progression is closely related to excessive production of reactive oxygen species (ROS) and upregulation of catabolic enzymes such as matrix metalloproteinases. Here, we developed a self-assembled multifunctional polymeric nanoparticle (NP) system for the co-delivery the ROS scavenger 2,2,6,6-Tetramethylpiperidoxyl (TEMPO) and small interfering RNA (siRNA) against MMP-13 (siMMP-13). The nanoplatform is constructed from antioxidant TEMPO-PEG-PLGA and cationic PEG-PLGA-OA9 to enable efficient siRNA encapsulation and intra-articular delivery. TEMPO mimics superoxide dismutase to neutralize ROS, while siMMP-13 silences catabolic gene expression to suppress cartilage degradation. This redox- and gene-regulatory NP system demonstrated potent anti-inflammatory and cartilage-protective effects in vitro and in vivo , effectively attenuating cartilage damage and inflammation in Osteoarthritis. This work presents a promising and translational approach to Osteoarthritis treatment via integrated redox and gene regulation strategies. Statement of Significance Osteoarthritis (OA) remains a major clinical challenge due to its complex and multifactorial pathogenesis, which current palliative therapies fail to adequately address. This study presents a novel, multifunctional therapeutic strategy that concurrently targets two critical pathological features of OA—oxidative stress and extracellular matrix degradation—through the co-delivery of a reactive oxygen species (ROS) scavenger (TEMPO) and MMP-13–targeting siRNA via a self-assembled polymeric nanoparticle system. By integrating redox modulation and gene silencing within a single nanoplatform, this approach enables precise regulation of inflammatory and catabolic pathways, thereby enhancing therapeutic efficacy and cartilage preservation. The findings offer a promising foundation for the development of next-generation disease-modifying OA therapies with strong potential for clinical translation.

Surface indium vacancies promote photocatalytic H2O2 production over In2S3

Zhu Qiong,Su Jingjing,Lin Guoan,Li Guisheng,Zhuo Zhiwen,Wang Weiyi,Li Jialin,Xu Xiaoxiang

Photocatalytic oxygen reduction reaction (ORR) represents a green and cost-effective means to produce the high-value industrial compound H 2 O 2 . However, the efficient H 2 O 2 synthesis awaits in-depth knowledge of the ORR mechanisms for the design of highly active photocatalysts. In this work, surface indium vacancies (denote hereafter as V In ) have been introduced into In 2 S 3 , which significantly boosts the photocatalytic activity for H 2 O 2 production, with an optimal H 2 O 2 generation rate of 4.77 ± 0.05 mmol·h −1 ·g cat. −1 under visible light illumination (λ ≥ 420 nm) and an apparent quantum efficiency (AQE) of 7.49 ± 0.01% at 420 ± 20 nm. Mechanistic analysis reveals the multifunctionality of V In , such as enlarging the chemisorption capacity of O 2 , enriching photo-generated electrons for ORR, endorsing high reducing power to photo-generated electrons, and promoting H 2 O 2 desorption. These findings justify that surface cation vacancies open up new possibilities for H 2 O 2 photosynthesis by leveraging their reaction dimensions in ORR.

Synergistic effect of cell structure and surface engineering: High-Aspect-Ratio nanofiber production and applications from bamboo pulp waste

Zebing Xiao,Yutong Xiong,Yonghua Zhang,Xiaoyun Li,Yixiang Wang,Xiaoying Wang

Bamboo pulp fines (BPF) are defined as bamboo pulp waste. BPF, including fine fibers and abundant non-fibrous cells, are detrimental to the performance of paper and are usually regarded as waste and discarded. In this work, inspired by the loose structure of non-fibrous cell walls in bamboo, a mild method for converting BPF into CNFs was developed. By leveraging the loose structure of BPF, high-density surface carboxyl groups were introduced via low-toxic (relatively less hazardous than nitro-based oxidants) maleic anhydride (MAH) esterification reactions, significantly enhancing dispersibility and interfacial compatibility. Combined with gentle mechanical disintegration, successfully resulting in CNFs (M−CNFs) with a diameter of approximately 2.6 nm, an aspect ratio exceeding 1000, a carboxyl content of 1.45 mmol/g, and a yield of approximately 98%. Furthermore, M−CNF−based nanofilms exhibited exceptional mechanical properties, achieving 107 MPa tensile strength with unique stress-dispersion mechanisms. The resulting M−CNFs serve as sustainable building blocks for multidimensional advanced cellulose materials and emulsion stabilizers. This method-produced M−CNFs offer advantages including ultrahigh aspect ratios, colloidal stability, high yield, and significant application potential, establishing a high-value valorization route for bamboo pulp waste.

Band-aid based piezoresistive sensor with hierarchical multiscale architectures for high-sensitivity and wide-range pressure monitoring

Nan Li,Le Yang,Xingxiang Ji,Wei Chen

Flexible piezoresistive sensors are crucial for wearable electronics. Conventional approaches predominantly enhance sensitivity and detection range through innovations in conductive materials, sensor substrates, or sophisticated microstructure engineering. This study presents a novel approach using commercial band-aid as multifunctional piezoresistive sensor substrates, utilizing their inherent structural properties. This substrate is functionalized through immersion into a multi-walled carbon nanotubes (MWCNT) and cellulose nanofibers (CNFs) composite dispersion, forming a nanoscale conductive film while preserves the native microscale patterns. These patterns amplify localized pressure responses, thereby enhancing sensitivity. Simultaneously, the conductive band-aid is assembled into a millimeter-scale arched architecture that optimizes the sensitivity-range trade off: under low stress, the minimal initial contact area optimizes sensitivity, while the arch design stabilizes large deformations, extending the measurable pressure range. This cross-scale integration synergizes stress redistribution and dynamic conductive pathway adjustments, enhancing contact area homogeneity under pressure. The resultant sensor demonstrates exceptional performance including high sensitivity (66.8 kPa −1 ), broad detection range (0–219.2 kPa), rapid response/recovery times (1 ms/4 ms), exceptional durability (10,000 cycles), and ultra-low detection limit (7 Pa). Moreover, the methodology addresses scalability and cost barriers by utilizing mass-produced, biocompatible band-aids as off-the-shelf substrates, bypassing complex and costly nanofabrication processes. By harmonizing material accessibility, structural ingenuity, and multi-functional sensing performance, this approach not only resolves enduring trade-offs between sensitivity, range, and durability in wearable sensor design but also establishes a scalable platform for next-generation epidermal electronics.

Enzyme-free multicolor sensor for visual detection of malathion based on a bimetallic magnetic nanozyme-mediated gold nanorod etching

Yuanying Chen,Tianran Lin,Shuangshuang He,Qi Jiao,Shulin Zhao,Li Hou

Malathion plays an important role in agricultural production, and the accurate detection of malathion residues is essential to ensure food safety. A novel enzyme-free sensing method was proposed by employing bimetallic magnetic nanozymes for both colorimetric quantification and multicolor semi-quantitative detection of malathion. The nanozyme exhibits peroxidase-like activity, catalyzing H 2 O 2 -mediated oxidation of 3,3′,5,5′-Tetramethylbenzidine dihydrochloride (TMB). Malathion inhibits this activity via Zr O P coordination, reducing oxidized TMB (oxTMB) generation. Under acidic conditions, oxTMB is further converted to TMB 2+ , which etches gold nanorods, inducing aspect-ratio variations accompanied by distinct multicolor changes. This method demonstrates low detection limits of 0.68 ng/mL. It does not require biological enzymes and can semi-quantify the malathion concentration through color changes. Thus, it can achieve convenient and accurate quantitative analysis without instruments, meeting on-site detection requirements. This work contributes to food safety and the development of efficient and reliable methods for malathion detection.

Regulation of early hydration and microstructure of cement slurry by cellulose nanofibers

Kaiqiang Liu,Wenli Tang,Lieqiang Luo,Guihai Li,Haiping Wang,Xingguo Zhang

In this study, cellulose nanofibres (CNFs) with different structures were prepared using ball-milling and 2,2,6,6-tetramethylpiperidine-1-oxyl-mediated (TEMPO) oxidation. Then, microcalorimeter, scanning electron microscope, electrical conductivity, and micro-CT were used to investigate the interaction mechanisms between the CNFs and cement slurry. The results showed that the combination of ball-milling and TEMPO oxidation produced CNFs with carboxyl functional groups. When the NaClO concentration was 2 mmol/g and oxidation time was 2 h, the carboxyl content on the surface of the nanofibres was 0.0856 mmol/g. Based on the hydration rate and electrical conductivity results, the carboxylated CNFs reduced the hydration rate of the cement slurry and delayed the exothermic peak by 0.5–2.3 h. The decrease rate of the electrical conductivity of the cement slurry during transition stage increased from 2.805 to 3.543–5.142 mS/(cm·h). The electrical conductivity of the solid–liquid two-phase cement slurry was mainly provided by the pore solution. The decrease in the electrical conductivity of the cement slurry during the transition stage could mainly be attributed to the formation of hydration products that changed the microstructure of the cement slurry from ‘dispersed particles’ to a ‘gel grid’ during the hydration acceleration stage, the porosity and pore connectivity of cement slurry are reduced. The zeta potential results indicate that CNF in the cement slurry provides a location for the nucleation and growth of early hydration products through adsorption and bridging effects, changes the microstructure of the cement slurry, and accelerates the transition from liquid to solid state.

Oil–water interfaces drive gold precipitation via microdroplet chemistry in thermal geological systems

Guanghui Yuan,Jixuan Wang,Yingchang Cao,Zihao Jin,Fang Hao,Keyu Liu,Yang Li,Hans-Martin Schulz,Jon Gluyas,Richard N. Zare

Sedimentary basins host high-grade gold mineralization at intersections of auriferous hydrothermal fluids and hydrocarbons. However, the precise mechanism of native gold formation associated with organic matter remains poorly understood. Here, we investigate gold precipitation at oil–water interfaces through in situ thermal experiments using various combinations of oil and HAuCl4-bearing solutions. Our results reveal that gold particles form spontaneously following the extensive generation and evolution of water microdroplets at oil–water interfaces at temperatures of 140 to 400 °C. We propose that electrons (e−), released from the conversion of hydroxide ions (OH−) to hydroxyl radicals (·OH) in water microdroplets, together with H atoms (·H) formed through electron transfer involving H3O+, and spontaneously generated H2O2 from·OH recombination, drive the reduction of Au3+ to Au0. The atomic gold progressively aggregates into Au0 clusters and Au nanoparticles (AuNPs), ultimately forming micrometer-scale gold particles and wires. This precipitation process occurs within minutes at temperatures above 350 °C and within hours below 200 °C. The experimentally produced gold particles exhibit textures like those in natural ore deposits. This interfacial microdroplet–induced mechanism provides a unique perspective on native gold formation in hydrocarbon-rich geosystems. Beyond its geological significance, this mechanism offers a potentially simplified approach for gold recovery from electronic waste without the need to introduce complex adsorbents or reducing agents into the waste stream.

Quantum tunneling of homogeneous catalyst altering CO2 reduction reaction pathway for stable Mg-CO2 batteries

Wenbo Liu,Lu Li,Menggang Li,Zongqiang Sun,Youxing Liu,Shaojun Guo

Mg-CO 2 battery has emerged as a promising battery technology by harnessing greenhouse gas as an active material. However, its development is greatly hindered by sluggish CO 2 conversion kinetics, resulting in high overpotentials and poor reversibility. Herein, we report a class of 2,2,6,6-tetramethylpiperidoxyl (TEMPO) homogeneous catalyst to regulate CO 2 adsorption and optimize reaction pathways through a quantum tunneling effect induced by electron transfer from the TEMPO free radical to CO 2 that classical electron transfer mechanisms cannot overcome. This quantum tunneling effect not only enables CO 2 reduction at lower voltage but also regulates the CO 2 adsorption environment, leading to the alternated reaction pathway for the formation of flower-like MgC 2 O 4 as the discharge product, rather than the dense MgCO 3 typically formed in traditional models. The TEMPO-based Mg-CO 2 batteries achieve an exceptional discharge voltage of 1.1 volts and a charge voltage of 1.3 volts, with stable cycling performance for over 450 hours, representing the best-reported performance among Mg-CO 2 battery systems to date.

A mutually beneficial waste-to-biochar strategy: Mn-mediated dual enhancement of 5-HMF production and catalytic performance

Yan Sun,Ranxin Li,Qiubo Wang,Wen-da Oh,Mingjie Huang,Xiaohui Wu,Tao Zhou

The concept of converting biowaste to energy is popular worldwide; however, overcoming the trade-off between bio-oil recovery and obtaining functional biochar remains challenging. This study investigated the effects of three transition metal ions (Mn 2+ , Cu 2+ and Fe 2+ ) throughout the entire process of recovering 5-hydroxymethylfurfural (5-HMF) from forestry waste and on the catalytic abilities of biochar derived from the oil-extracted residue. The Cu 2+ , Fe 2+ , and Mn 2+ ions significantly increased 5-HMF yields by 93- to 158-fold compared to the metal-free control. However, only marginal improvements in the catalytic ability of the residue-derived biochar for persulfate activation were observed for Cu- or Fe-doped biochar, despite the general benefit of transition metal doping for biochar functionalization. Interestingly, a dual Mn-mediated promotion was achieved beyond enhancing 5-HMF production. The Mn-doped biochar (Mn-Biochar) exhibited excellent peroxydisulfate (PDS) activation capability, enhancing the degradation of the antibiotic sulfamethoxazole (SMX) by 6.6- to 15.0-fold. Characterizations revealed that incorporated nitrogen was responsible for this enhancement. N-doping transformed the Mn species into MnO, which tripled the Mn loading capacity and enhanced the electrical conductivity of the biochar. The formed Mn-N x structure in the biochar (Mn-N-FBBC) facilitated polarization of the O O bond and cleavage of the S O bond in PDS, generating 1 O 2 for SMX degradation. Life cycle assessment (LCA) indicated that this dual Mn-mediated process for 5-HMF recovery and biochar production offers synergistic benefits, realizing the “waste-resource-material-treatment” paradigm in biomass conversion.

Fabrication of flexible and sensitive resistance strain sensor by flexible TPU decorated with CQDs/TNC composite

Wei Chen,Xiuchuan Jing,Zhuang Li,Yongxi Ma,Hongda Gu,Haiqing Miao,Zhong Wang,Guohe Wang

Attaching flexible resistive strain sensors to human skin enables response detection of behaviours towards real-time physiological monitoring. However, current flexible resistive strain sensors hinder themselves from practical applications due to their low sensitivity, small linear working range, and poor wearing comfort. In this research, a conductive thermoplastic polyurethane (TPU) membrane is fabricated using TEMPO modified nanocellulose (TNC) decorated with carbon quantum dots (CQDs) to prepare flexible highly sensitive, and breathable strain sensors. Results show that the mechanical property and strain response range of the TPU membrane is significantly enhanced by CQDs-TNC. The TPU membrane also exhibits the highest sensitivity (GF= 49.4 in the 30∼80 % strain range) and a wide response range (0–80 %). At 5 %wt of CQDs-TNC, the membrane displays the highest elongation at break (389.8 %) with maximum breaking stress approaching 9.7 MPa. The CQDs-TNC/TPU membrane exhibits fast response and recovery times (85 ms and 93 ms), low hysteresis (8.9 %), and excellent dynamic cyclic strain performance (1000 stretch/release cycles). The CQDs/TMC/TPU-resistive strain sensor (CTT-RSS) sensor well conforms to human joints, and accurately produces real-time regular resistance strain responses based on human movements. In all, this study contributes a novel research design for fabricating flexible and sensitive sensor to the development of smart textiles.

A FRET-based ratiometric fluorescent chemosensor for mitochondrial ONOO⁻ detection with application to Rheumatoid Arthritis

Jianhong Liu,Xinyue Zhao,Li Zhang,Haowen Zhou,Xudong Li,Guorui Zheng,Xibin Zhou,Yongjin Peng,Jie Zhang

Rheumatoid arthritis (RA) is a chronic autoimmune disorder characterized by progressive joint destruction, where early diagnosis is crucial for impeding disease progression. Peroxynitrite (ONOO⁻), a key reactive nitrogen species, is critically involved in the pathogenesis of RA and represents a promising diagnostic target for early detection. Herein, we developed MRN, a mitochondria-targeting ratiometric fluorescent probe based on a Förster resonance energy transfer (FRET) mechanism for the dynamic monitoring of ONOO⁻ in RA inflammatory models. The probe integrates a rhodamine-naphthalimide FRET pair and a hydrazide group as the specific recognition unit, which facilitates a rapid, ONOO⁻-triggered ratiometric response within 30   seconds and achieves a detection limit of 7.5   nM. Furthermore, the conjugation of a triphenylphosphonium group ensures precise mitochondrial localization. Cellular assays demonstrated excellent biocompatibility and specific mitochondrial targeting. We successfully applied MRN for real-time ratiometric imaging of both endogenous and exogenous ONOO⁻ fluctuations in live BV-2 cells. Importantly, in a Complete Freund's Adjuvant (CFA)-induced rheumatoid arthritis mouse model, MRN enabled in situ ratiometric fluorescence imaging of ONOO⁻ at articular sites, revealing a strong positive correlation between the fluorescence signal intensity and the degree of inflammatory severity. This work establishes MRN as a novel and robust molecular tool for the early diagnosis of RA.

Directional selection mechanism of single atom catalyst Co-S-C/CB regulating peroxymonosulfate radical pathway

Bin Ou,Hongwei Rong,Baihui Cui,Jie Zhang,Fang Zhang,Dabin Guo

The coordination calcination method is used to form a coordination complex between Co 2 + and thiophene, which is adsorbed on carbon black (CB) and then calcined to prepare Co-S-C/CB single atom catalyst. Co-S-C/CB could efficiently activate peroxymonosulfate (PMS) by forming Co-S 3 coordination structure and Co-S 3 is the active site for activating PMS. Electron rich region was formed on the Co atom and the Co atom could keep in a low valence state during the catalytic process and donate electrons to PMS to produce radicals (•OH and SO 4 •- ) through a single-electron-reduction. Co-S-C/CB could not only promote PMS to generate •OH and SO 4 •- , but also inhibit the generation of 1 O 2 and O 2 •- . The radical pathway has the lowest Gibbs free energy (ΔG) compared to the 1 O 2 pathway and the Co(IV)=O pathway. Therefore, Co-S-C/CB could regulate the radical pathway of PMS. •OH and SO 4 •- are mainly responsible for the removal of Rhodamine B (RhB). Co-S-C/CB could be applied to efficiently degrade various organic pollutants and has good stability. Macro preparation of Co-S-C/CB could be achieved from both the preparation process and raw material aspects.
