Redox-triggered controlled release systems-based bi-layered nanocomposite coating with synergistic self-healing property

Ting Wang,LingHua Tan,ChenDi Ding,MingDong Wang,JianHua Xu,JiaJun Fu

Redox-triggered smart nanocontainers, RTSNs 1, assembled by installing supramolecular switches onto the exterior surface of mesoporous silica nanoparticles, were successfully synthesized. The reversible transition upon redox stimuli from self-complexation to self-dissociation of supramolecular switches, which are in the form of β-cyclodextrin monofunctionalized with a ferrocene moiety, realizes switch behavior and regulates the release of p-coumaric acid (CA) encapsulated in mesoporous silica nanoparticles. Based on these facts, a bi-layered nanocomposite coating was designed and coated on the surface of AA2024 by successive deposition of a Ce(IV)-doped ZrO2–SiO2 sol–gel coating and an RTSNs 1 incorporated ZrO2–SiO2 coating. The bi-layered nanocomposite coating shows high barrier performance, effectively delaying the corrosion process occurring on the metallic surface in 0.5 M NaCl solution for 20 days. Once the bi-layered nanocomposite coating suffers local damage, Ce(IV) salt meets with RTSNs 1 in the scratched area and thus open the supramolecular switches, resulting in the release of the organic corrosion inhibitors, CA molecules. In the meantime, Ce(IV) salt is reduced to the inorganic corrosion inhibitor, Ce(III) salt. The combination of CA and Ce(III) produces a synergistic inhibition effect, which rapidly repairs the damaged surface and provides durable self-healing functionality.

Superhydrophobic controlled-release fertilizers coated with bio-based polymers with organosilicon and nano-silica modifications

Shugang Zhang,Yuechao Yang,Bin Gao,Yuncong C. Li,Zhiguang Liu

Although bio-materials for controlled-release fertilizers (CRFs) have been developed rapidly because of their low-cost and green and sustainable properties, the release characteristics of these CRFs still face problems because the coating is often hydrophilic with many micro-holes. In this paper, a novel coating material was developed to coat fertilizers using bio-based polyurethane (BPU) derived from liquefied wheat straw (LWS). Organosilicon (OS) and nano-silica (NS) were also used to modify the BPU to increase the nanoscale surface roughness and reduce the surface energy, and thus to make it superhydrophobic. The nutrient release characteristics of the superhydrophobic BPU coated fertilizer (SBPCF) were greatly enhanced, compared with the unmodified BPU coated fertilizer (BPCF). This was because the nanoscale surface roughness of the SBPCF prevented water from contacting the coating material. In addition, the nanoparticles on the SBPCF surface also block the micro-holes on the coating and thus reduced the entrance of water. The results suggest that the new coating technology can create a superhydrophobic surface on CRFs and thus improve their controlled-release characteristics.

Dual-Cross-Linked Gelatin-Polysaccharide Hydrogel Beads for Efficient Protection and Controlled Release of Probiotics

Xiangxiang Lv,Yong Yu,Jinyao Yu,Qin Zheng,Wei Jiang,Ranran Wang,Jihong Wu,Hongxia Wang,Hai Chen,Yuhao Zhang,Hongjie Dai

This study fabricated gelatin–sodium alginate-carboxylated cellulose nanofibers (GSC) composite hydrogel beads using a dual-cross-linking strategy based on Ca2+ and transglutaminase (TG) for probiotic encapsulation. The influences of TG cross-linking time on the properties of hydrogel beads and probiotic incorporation were investigated. The results indicated that probiotics were effectively immobilized through hydrophobic interactions with gelatin and electrostatic interactions with Ca2+. The internal structure of the hydrogel beads became denser as the TG cross-linking time increased. The survival rates reached 87.9%, 70.9%, and 34.4% after low-temperature storage, freeze-drying, and heat treatment at 65 °C for 10 min, respectively. Furthermore, a high viable count was maintained after 2 weeks of storage in beverage matrices including orange juice and yogurt. Following in vitro simulated gastrointestinal digestion, the hydrogel beads released the probiotics via a swelling mechanism, increasing the survival rate up to 85%, while the release duration was extended to 4–6 h. GSC hydrogel beads significantly improved the stability and delivery of probiotics, among which GSC-TG30-LP showed the best overall performance. These findings demonstrate that the GSC hydrogel beads can be applied as an effective encapsulation carrier for probiotics, showing potential applications in the food industry.

Macroscopic Phase-Change Drug Carrier Ball with Photothermally Controlled Release and Magnetically Controlled Transport Properties

Hyunji Lee,Jihyeok Park,Minkyeong Kang,Woochan Kim,Sang Kug Chung,Joohyung Lee

Phase-change materials (PCMs) are widely regarded as effective thermo-responsive gating materials for on-demand drug release. Most previous studies utilizing PCMs for this purpose focus on small colloidal micro- and nanoparticles. In this study, macroscopic phase-change drug carriers are demonstrated in the form of millimeter-sized balls, which offer enhanced visibility. These wax-based phase-change macroballs, composed of paraffin, fatty alcohols, or fatty acids, can be easily fabricated with customizable volumes by depositing melted wax droplets containing functional components–such as drugs, photothermal agents, surfactants, and magnetites–onto omniphobic surfaces, where they spontaneously dewet and solidify upon natural cooling. These macroballs can be rapidly heated and melted upon near-infrared irradiation due to the incorporation of low-melting-point liquid metal (LM) eutectic Ga–In microdroplets as photothermal agents, enabling efficient on-demand drug release. Furthermore, by adsorbing magnetically susceptible Fe₃O₄ nanoparticles onto the LM microdroplets, the transport of the macroballs becomes magnetically controllable, as demonstrated by the use of a permanent magnet or electromagnet. The strategies demonstrated in this study provide a simple and scalable approach to fabricating high-visibility, trackable macroscopic phase-change balls with on-demand photothermal drug release and magnetically controlled transport properties.

Limonene-loaded nanostructured lipid carriers incorporated in hydrogels: Enhanced acid stability and controlled release properties

Simin Feng,Yitong Tian,Jiahao Yu,Yang Lin,Alan Giovanini de Oliveira Sartori,Ping Shao

Improving the acid stability of nanostructured lipid carriers (NLCs) is critical for broadening their engineering applications. The integration of NLCs with hydrogels results in a more stable, multifunctional system that facilitates the effective and sustained release of bioactive ingredients. In this study, The chitosan/alginate hydrogel (CAH) exhibited minimal limonene release under combined high-temperature and acidic conditions when the degree of carboxyl substitution (DS) of chitosan reached 17.68 ± 0.14 % (NLC-CAH1). It exhibited the lowest release rate and superior stability under high-temperature and acidic conditions. The cumulative release rates of limonene in NLC and NLC-CAH1 within 3 h of acid solution were 35.24 ± 0.97 % and 22.41 ± 2.09 %, respectively. The hydrogels effectively delayed the release of NLCs, reducing the release rate of limonene and facilitating a prolonged release of bioactive ingredients. Consequently, the NLCs-hydrogel system represents a novel delivery mechanism, and offers innovative strategies for the administration of lipophilic bioactive compounds.

pH-responsive controlled-release antimicrobial active film based on gelatin/chitosan incorporated with HNT nanocomposite

Yuemei Zhang,Xueyu Cheng,Chenghao Guo,Lijing Lu,Lixin Lu,Yongjin Qiao

Stimuli-responsive active packaging materials represent a promising frontier in food preservation by combining controlled-release functionality with environmental adaptability. Herein, a novel pH-responsive antibacterial bioactive packaging film based on a chitosan/gelatin (CS/GA) composite was developed by incorporating nanocomposites (NP) prepared through layer-by-layer assembly of natural polyelectrolytes onto modified halloysite nanotube (HNT), with carvacrol (Car) as the encapsulated active agent. The results showed that the developed composite films exhibited pH-responsive behavior under different pH conditions (4.0, 7.0, and 9.0), with a preferential release under alkaline conditions. The release rate was governed by the synergistic effects of carrier structural relaxation/dissolution within the NP, film diffusion, and structural relaxation/dissolution. In addition, hydrogen bonding and electrostatic interactions among CS, GA, and the NP improved the dispersion of the NP within the film matrix, thereby enhancing the film's mechanical strength, gas barrier performance, and thermal stability. The resulting active film effectively inhibited the growth of E. coli and S. aureus , and markedly prolonged the shelf life of pork. Overall, the developed pH-responsive active packaging films have great potential for application in future smart food preservation systems and lay a foundation for designing stimuli-responsive materials for different food environments.

Dual modules-molecularly imprinted patch-enabled enantioselectively controlled release of racemic drugs for transdermal delivery

Yang Zhang,Xiaowen Liu,Jiaxu Wu,Peng Quan,Chao Liu,Jie Liu,Mingzhe Liu,Liang Fang

Over 90 % of chiral drugs applied in transdermal drug delivery system (TDDS) are racemates, significantly increasing risks of side effects. Herein, we designed a chiral molecularly imprinted patch (CMIP) that achieved enantioselectively controlled release of S-enantiomers (eutomers) and inhibited the release of R-enantiomers (distomers) for transdermal drug delivery. It is composed of chiral pressure sensitive adhesive (PSA) and molecularly imprinted polymers (MIP), showing better transdermal delivery of S-enantiomers than that of R-enantiomers in vitro (1.86-fold) and in vivo (3.74-fold), significantly decreasing the intake of distomers. Additionally, synthesized fluorescent probe enantiomers visualized enantioselective process of CMIP. Furthermore, investigations of molecular mechanism indicated that dependence on spatial conformation was dominant. On one hand, imprinted cavity of MIP with D-isomer and stronger chiral interaction with R-enantiomers led to more specific adsorption. On the other hand, L-isomer of PSA controlled the release of S-enantiomers by multiple interaction including chiral H-bond, π-π interaction and Van der Waals force. Tthus, the innovatively designed transdermal patch with enantioselective ability released eutomers of racemate and simultaneously inhibited release of distomers, significantly improving therapeutical efficiency and avoiding overdose.

Controlled Release of Cold Atmospheric Plasma by Gelatin Scaffold Enhances Wound Healing via Macrophage Modulation

Nian Zhang,Guang Yang,Yan Wu,Liru Hu,Chengzhi Zhao,Hang-Hang Liu,Li Wu,Jian Pan,Xian Liu

Cold atmospheric plasma (CAP) has emerged as a promising therapeutic tool for wound healing and tissue regeneration. However, the clinical application of CAP faces challenges, particularly the need for repeated treatments to maintain therapeutic efficacy and the difficulty in controlling the release of reactive oxygen and nitrogen species (ROS/RNS), which can cause tissue damage at high concentrations. In this study, we developed a controlled-release system by integrating CAP with a skin bionics gelatin scaffold (CAP-GS), enabling the sustained, localized release of CAP without the need for repeated applications. Our results demonstrate that CAP-GS significantly accelerates skin wound healing by modulating the immune microenvironment and reducing inflammation. The gelatin scaffold effectively regulates the release of ROS/RNS, maintaining levels conducive to M2 macrophage polarization and minimizing oxidative damage. Furthermore, CAP-GS enhances macrophage metabolism, including oxidative phosphorylation (OXPHOS) and fatty acid oxidation (FAO), metabolic pathways characteristic of M2 polarization. The increased secretion of growth factors by CAP-GS-treated macrophages contributes to cell proliferation, migration, and tissue regeneration. These findings highlight CAP-GS as a promising, clinically applicable system for wound healing and tissue repair, overcoming the limitations of traditional CAP treatments and offering an approach to regenerative medicine.

Controlled Release of Ceria and Ferric Oxide Nanoparticles via Collagen Hydrogel for Enhanced Osteoarthritis Therapy

Xian Chen,Lili Wang,Jingting Zhang,Huiyu Yan,Shenghong Wang,Jianxi Xiao

Osteoarthritis (OA), characterized by chronic inflammation and cartilage degeneration, significantly affects over 500 million people globally. Nanoparticles have emerged as promising treatments for OA; however, current strategies often employ a single type of nanoparticle targeting specific disease stages, limiting sustained therapeutic efficacy. In this study, a novel collagen hydrogel is introduced, thiol crosslinked collagen-cerium oxide-poly(D,L-lactic-co-glycolic acid) microspheres encapsulating nanoparticles (CSH-CeO 2 -pFe 2 O 3 ), designed for the controlled release of cerium oxide (CeO 2 ) and ferric oxide (Fe 2 O 3 ) nanoparticles for comprehensive OA management. The sulfhydryl cross-linked collagen matrix embeds CeO 2 nanoparticles and poly(D,L-lactic-co-glycolic acid) (PLGA) microspheres encapsulating Fe 2 O 3 nanoparticles. The CSH-CeO 2 -pFe 2 O 3 hydrogel exhibits enhanced mechanical strength and remarkable injectability, along with a significant promotion of cell adhesion, proliferation, and chondrogenic differentiation. Notably, the hydrogel demonstrates intelligent responsiveness to high levels of reactive oxygen species, initiating the rapid release of CeO 2 nanoparticles to address the intense inflammatory responses of early-stage OA, followed by the sustained release of Fe 2 O 3 nanoparticles to facilitate cartilage regeneration during the proliferative phase. In a rat model with cartilage defects, the hydrogel significantly alleviates inflammation and enhances cartilage regeneration, holding substantial potential for effectively managing the pathologically complex OA.

Controlled release active films prepared from temperature responded emulsions and their application in grape preservation

Yucong Shi,Bin Li,Jianhua Yuan,Yong Qin,Lei Zhou,Junzhen Zhong,Wei Liu,Chengmei Liu

This study constructed a sodium alginate (SA)-based temperature-responsive emulsion film. The addition of 6 % of whey isolate protein fibre (WPF) and glycyrrhizic acid (3:1) prepared stable emulsions and endowed it temperature-responsive release capability at 4–45℃. The incorporation of prepared emulsions (10–50 % v/v) into SA-based films (E-SA) improved the UV-blocking and antioxidant properties, while it decreased the breaking strength and water solubility. Especially, 30 %E-SA film exhibited best elongation at break water vapor barrier performance, and temperature-responsive release capability. In preservation experiments, the 30 %E-SA film had excellent effect on maintaining pH, soluble solids and V C content of grapes. Especially, the weight loss of grape treated with 30 %E-SA film (19.88 %) was reduced by 44.50 % compared to control grapes at 45°C after 6 d. Therefore, the emulsion-based films prepared are expected to provide an attractive method for preserving grape freshness in different temperature environments .

An Injectable Magnesium-Based Cement Stimulated with NIR for Drug-Controlled Release and Osteogenic Potential

Yanbin Zhao,Yangyang Li,Bin Wang,Junyan Yao,Yue Fan,Peng He,Jing Bai,Cheng Wang,Feng Xue,Chenglin Chu

Magnesium phosphate bone cement (MPC) has gained widespread usage in orthopedic implantation due to its fast-setting and high initial strength benefits. However, the simultaneous attainment of drug-controlled release and osteogenic potential in MPC remains a significant challenge. Herein, a strategy to create a smart injectable cement system using nanocontainers and chondroitin sulfate is proposed. It employs nanocontainers containing alendronate-loaded mesoporous silica nanoparticles, which are surface-modified with polypyrrole to control drug release in response to near-infrared (NIR) stimulation. The alendronate-incorporated cement (ACMPC) exhibits improved compressive strength (70.6 ± 5.9 MPa), prolonged setting time (913 s), and exceptional injectability (96.5% of injection rate and 242 s of injection time). It also shows the capability to prevent degradation, thus preserving mechanical properties. Under NIR irradiation, the cement shows good antibacterial properties due to the combined impact of hyperthermia, reactive oxygen species, and alendronate. Furthermore, the ACMPC (NIR) group displays good biocompatibility and osteogenesis capabilities, which also lead to an increase in alkaline phosphatase activity, extracellular matrix mineralization, and the upregulation of osteogenic genes. This research has significant implications for developing multifunctional biomaterials and clinical applications.

An injectable hydrogel dressing for controlled release of hydrogen sulfide pleiotropically mediates the wound microenvironment

Junwei Yang,Xianzhen Dong,Wenying Wei,Kun Liu,Xiaopei Wu,Honglian Dai

The healing of scalded wounds faces many challenges such as chronic inflammation, oxidative stress, wound infection, and difficulties in vascular and nerve regeneration. Treating a single problem cannot effectively coordinate the complex regenerative microenvironment of scalded wounds, limiting the healing and functional recovery of the skin. Therefore, there is a need to develop a multi-effect treatment plan that can adaptively address the issues at each stage of wound healing. In this study, we propose a scheme for on-demand release of hydrogen sulfide (H2S) based on the concentration of reactive oxygen species (ROS) in the wound microenvironment. This is achieved by encapsulating peroxythiocarbamate (PTCM) in the ROS-responsive polymer poly(ethylene glycol)–poly(L-methionine) (PMet) to form nanoparticles, which are loaded into a thermosensitive injectable hydrogel, F127-poly(L-aspartic acid-N-hydroxysuccinimide) (F127-P(Asp-NHS)), to create a scald dressing. The H2S released by the hydrogel dressing on demand regulates the wound microenvironment by alleviating infection, reducing oxidative stress, and remodeling inflammation, thereby accelerating the healing of full-thickness scalded wounds. This hydrogel dressing for the adaptive release of H2S has great potential in addressing complex scalded wounds associated with infection and chronic inflammation.
