Small-molecule intercalation induces defective generation of bromine-doped bismuth oxychloride to enhance photocatalytic degradation and detoxification of tetracycline

Jianchao Liu,Yihao Qin,Guanghua Lu,Runren Jiang

Photocatalysts are one of the effective methods to degrade antibiotic contamination, but the efficiency is low and the toxicity is not well recognized. Deep lattice doping to induce defect generation is an effective way to improve the performance of photocatalysts. Here, defect-rich bromine-doped BiOCl-XBr photocatalysts were constructed with the help of small molecules inserted into the interlayer. The photocatalytic degradation performance of BiOCl-XBr was significantly enhanced, and its degradation rate was up to about 12 times that of BiOCl monomer. The main reasons for the stronger photocatalytic performance of BiOCl-XBr include Br doping to enhance visible light absorption, surface defects, and Bi valence changes to improve charge transport. The degradation of tetracycline (TC) produced more toxic intermediates, and the biotoxicity experiments also confirmed that the toxicity showed a trend of increasing and then decreasing, indicating that the more toxic intermediates were also mineralized during the degradation process. However, the mortality and hatching rate of zebrafish in the exposed group after degradation recovered but changed their activity pattern under light and dark conditions. This further warns us to focus on the toxicity changes after antibiotic degradation. Finally, based on the free radical analysis, the mechanism of photocatalytic degradation and detoxification of TC by BiOCl-XBr was proposed.

Oxidative degradation and detoxification of multiple mycotoxins using a dye-decolorizing peroxidase from the white-rot fungus Bjerkandera adusta

Huimin Shao,Xiaoyun Su,Yaru Wang,Jie Zhang,Tao Tu,Xiaolu Wang,Huoqing Huang,Bin Yao,Huiying Luo,Xing Qin

Mycotoxins are widely present from cereal crops to food products, posing a significant risk to food safety and causing serious health and economic losses. Therefore, it is imperative to develop a sustainable and efficient detoxification strategy to address mycotoxin contamination. Herein, a dye-decolorizing peroxidase, Ba DyP, from the white-rot fungus Bjerkandera adusta was successfully expressed as a soluble form in Escherichia coli with the help of molecular chaperones. Purified recombinant Ba DyP could efficiently degrade multiple mycotoxins including aflatoxin B 1 , zearalenone, and deoxynivalenol in the presence of mediators such as Mn 2+ or 1-HBT. Meanwhile, their degradation products were identified using UPLC-MS/MS, including AFB 1 -diol, AFQ 1 , 15-OH-ZEN, HZEN, and C 15 H 18 O 8 . Furthermore, cell survival tests exhibited a significant reduction in the biological toxicity of these degradation products. These findings indicate that the use of dye-decolorizing peroxidase could be a promising approach for the effective removal of multiple mycotoxins in cereal crops and food products.

Far-UVC (UV222) based photolysis, photooxidation, and photoreduction of chlorophenols using a KrCl-excimer lamp: Degradation, dechlorination, and detoxification

Yu-Jia Nong,Qian-Yuan Wu,Yun-Peng Wu,Ju-Won Lee,Min-Yong Lee,Wen-Long Wang

The KrCl-excimer lamp, emitting far-UVC light at 222 nm (UV222), offers a promising alternative to conventional UVC light at 254 nm (UV254) for the photolysis of organic pollutants and the activation of radical sensitizers. This study was aimed to investigate the efficiencies of UV222 in the treatment of halogenated aromatics, focusing on its performance in degradation, dechlorination and detoxification. Chlorophenols, representative recalcitrant and toxic halogenated aromatics, were used as target pollutants. The pathways of direct photolysis, photooxidation and photoreduction under UV222 illumination were compared. UV222 outperformed UV254 in photolyzing chlorophenols (1.4−34.1 times faster), especially protonated chlorophenols, due to substantially higher UV absorption (17.1−108.0 times) and quantum yields (2.1−3.4 times). The quantum yields of chlorophenols were influenced by the inducive electron-withdrawing effect of Cl-substitutes. Moreover, UV222 improved the dechlorination of chlorophenols to 95 % compared to 60 % by UV254. The introduction of radical sensitizer (e.g., H 2 O 2 , nitrate, and sulfite) reduced 4-chlorophenol photolysis by competing for UV222 absorption, though the sensitizers partially increased radical oxidation via generating • OH or e aq − . UV222 photolysis of 4-chlorophenol increased the toxicity by 88.6 times through forming toxic intermediates (e.g., hydroquinone and resorcinol). Notably, • OH and e aq − (i.e., UV222/H 2 O 2 and UV222/sulfite) increased the dechlorination and • OH (i.e., UV222/H 2 O 2 ) detoxified the mixture solution. Moreover, UV222 photolysis remained effective for 4-chlorophenol removal in real paper-mill wastewater, indicating the potential application of KrCl* lamp UV222.

Glyoxal oxidase-mediated detoxification of reactive carbonyl species contributes to virulence, stress tolerance, and development in a pathogenic fungus

Xiaoyu Liu,Nemat O. Keyhani,Hong Liu,Yue Zhang,Yuxian Xia,Yueqing Cao

Reactive carbonyl and oxygen species (RCS/ROS), often generated as metabolic byproducts, particularly under conditions of pathology, can cause direct damage to proteins, lipids, and nucleic acids. Glyoxal oxidases (Gloxs) oxidize aldehydes to carboxylic acids, generating hydrogen peroxide (H 2 O 2 ). Although best characterized for their roles in lignin degradation, Glox in plant fungal pathogens are known to contribute to virulence, however, the mechanism underlying such effects are unclear. Here, we show that Glox in the insect pathogenic fungus, Metarhizium acridum , is highly expressed in mycelia and during formation of infection structures (appressoria), with the enzyme localizing to the cell membrane. MaGlox targeted gene disruption mutants showed RCS and ROS accumulation, resulting in cell toxicity, induction of apoptosis and increased autophagy, inhibiting normal fungal growth and development. The ability of the MaGlox mutant to scavenge RCS was significantly reduced, and the mutant exhibited increased susceptibility to aldehydes, oxidative and cell wall perturbing agents but not toward osmotic stress, with altered cell wall contents. The Δ MaGlox mutant was impaired in its ability to penetrate the host cuticle and evade host immune defense resulting in attenuated pathogenicity. Overexpression of MaGlox promoted fungal growth and conidial germination, increased tolerance to H 2 O 2 , but had little to other phenotypic effects. Transcriptomic analyses revealed downregulation of genes related to cell wall synthesis, conidiation, stress tolerance, and host cuticle penetration in the Δ MaGlox mutant. These findings demonstrate that MaGlox -mediated scavenging of RCS is required for virulence, and contributes to normal fungal growth and development, stress resistance. The ability of organisms to detoxify reactive metabolic byproducts is crucial for normal development, stress resistance, and where applicable, pathogenesis. Here, using the insect pathogenic fungus, Metarhizium acridum , we show that a glyoxal oxidase acts to detoxify cellular reactive carbonyl species (RCS) as well as leading to decreased reactive oxygen species (ROS) levels by alleviating aldehydes accumulation which can trigger ROS production. Mutants deficient in MaGlox showed increased levels of RCS/ROS leading to increased autophagy and apoptosis, resulting in impaired fungal growth, development, and reduced virulence. These findings link Glox mediated RCS/ROS detoxification to cell degradation/turnover processes and cell death, impacting fungal pathogenicity and development.

Non-cytochrome P450 enzyme aldehyde oxidase is involved in the oxidative metabolic pathway of diquat and its detoxification effect

Zhengsheng Mao,Youjia Yu,Gen Ba,Hongmei Zhao,Qifang Shi,Yue Cao,Weiran Xie,Jinsong Zhang,Hao Sun,Feng Chen

Diquat (DQ) poisoning has garnered attention in recent years, primarily due to the rising incidence of cases worldwide, coupled with the absence of a viable antidote for its treatment. Despite the fact that diquat monopyridone (DQ-M) has been identified as a significant metabolite of DQ, the enzyme responsible for its formation remains unknown. In this study, we have identified aldehyde oxidase (AOX) as a vital enzyme involved in DQ oxidative metabolism . The metabolism of DQ to DQ-M was significantly inhibited by AOX inhibitors including raloxifene and hydralazine . The source of oxygen incorporated into DQ-M was proved to be from water through a H2 18 O incubation experiment which further corroborated DQ-M formation via AOX metabolism. The product of DQ-M in vitro generated by fresh rat tissues co-incubation was consistent with its AOX expression. The result of the molecular docking analysis of DQ and AOX protein showed that DQ is capable of binding to AOX. Furthermore, the cytotoxicity of DQ was significantly higher than DQ-M at the same concentration tested in six cell types. This work is the first to uncover the involvement of aldehyde oxidase, a non-cytochrome P450 enzyme, in the oxidative metabolic pathway of diquat, thus providing a potential target for the development of detoxification treatment.

Boosted chloramphenicol mineralization and detoxification of UV/S(IV) processes with straightforward aeration: The critical contribution of post-reoxygenation

Shangding Yang,Xiaoke Yan,Shitai Shen,Li Wu,Bolin Li,Liuyang He,Lijuan Wu,Zulin Zhang,Lie Yang

Although advanced reduction processes (ARPs) based on UV/S(IV) (UV/sulfite) processes could achieve higher dechlorination and degradation efficiencies for organic pollutants , the mineralization and detoxification efficiencies still need to be further improved. In this study, post-reoxygenation was employed to improve the mineralization and detoxification of chloramphenicol (CAP) in UV/S(IV)-ARPs. The effects of initial pH and aeration rate on the dechlorination and mineralization efficiency of CAP were investigated. At the optimized conditions (pH 3.0, aeration rate of 0.3 L min −1 ), 92.75 % of dechlorination efficiency and 29.3 % of COD removal were obtained after 2 h of reoxygenation. Meanwhile, the effects of inorganic anions (Cl - , SO 4 2- , HCO 3 – , NO 3 – , and HPO 4 2- ) on the removal of COD and dechlorination efficiency were also evaluated. Furthermore, the electron paramagnetic resonance analysis disclosed that SO 4 •- and HO • mainly contributed to the mineralization of CAP. Further degradation and mineralization of CAP were confirmed in the post-reoxygenation UV/S(IV)-ARPs (PRO-UV/S(IV)-ARPs) and the intermediates were identified. The ecotoxicity prediction and residual toxicity assessment revealed that the ecotoxicity of CAP solution was significantly reduced after the treatment of post-reoxygenation. Overall, the PRO-UV/S(IV)-ARPs provides a promising choice with straightforward operation for the purification and detoxification of CAP wastewater.

Plant-Derived Isoferulic Acid and RNAi Synergize to Overcome Detoxification Defenses in the Bird Cherry–Oat Aphid (Rhopalosiphum padi)

Wenhua Hou,Xiaoxiao Yuan,Linhai Xia,Ni Wang,Zhenyin Zhu,Hongcheng Tang,Jaime C. Piñero,Maohua Chen,Xiong Peng

Sustainable management of insect pests is threatened by insecticide resistance, driving demand for effective, environmentally safe tools. The bird cherry–oat aphid (Rhopalosiphum padi), a major cereal pest, shows resistance to pyrethroids, neonicotinoids, and other insecticide classes. We identified isoferulic acid, a phenolic compound from Prunus padus, as a potent aphicide with lethal and sublethal impacts. Bioassays showed dose-dependent lethality (LC50 = 1.74 mg/L at 48 h), while sublethal exposure shortened adult longevity (17.7 to 10.2 d) and reduced fecundity (81 to 53 nymphs/female). Transcriptomic and enzymatic analyses revealed broad induction of detoxification pathways, with RpLipase consistently upregulated. RNAi-mediated knockdown of RpLipase enhanced susceptibility, lowering the LC50 to 0.85 mg/L and confirming its role in tolerance. Nontarget assays with the predator Harmonia axyridis revealed no acute toxicity. Collectively, isoferulic acid is a promising plant-derived insecticide, and silencing induced detoxification genes enhances its potency, providing a foundation for developing novel resistance-breaking strategies.

Polyethylenimine grafted H2O2-oxidized cellulose membrane as a novel biosorbent for Cr(VI) adsorption and detoxification from aqueous solution

Zhang Shengli,Chen Haoyu,Zhang Silue,Kai Chengcheng,Jiang Man,Wang Qun,Zhou Zuowan

Abstract A novel biosorbent PEI-HOCM was successfully prepared by grafting polyethyleneimine (PEI) onto H 2 O 2 -oxidized cellulose membrane (HOCM). The whole systhesis process occurred in aqueous solution and was environmentally friendly without any toxic cross-linkers involved. ATR-FTIR, XPS and titration analysis confirmed that the hydroxyl groups of cellulose were oxidized to carbonyl groups by H 2 O 2 and PEI was successfully introduced to the HOCM. The PEI-HOCM had positively charged surface (+ 20.6 to + 86.6 mV) at the measured pH range 3–9 according to the zeta potential analysis. The adsorption of Cr(VI) on the PEI-HOCM obeyed the pseudo-second-order kinetic model and Langmuir isotherm model. Its maximum equilibrium adsorption capacity was 147.06 mg/g at 25 °C, which was much higher than that of cellulose membrane (CM, 30.61 mg/g). According to the obtained results from the intraparticle diffusion, Body kinetic and diffusion-chemisorption models, film diffusion was the rate-limiting step and chemical adsorption was the main mechanism of Cr(VI) adsorption. The Cr(VI) adsorbed on the PEI-HOCM was completely reduced to Cr(III), suggesting that PEI-HOCM was not only useful for Cr(VI) removal but also good for its detoxification. The competitive uptakes from coexisting components (Na + , Mg 2+ , Ca 2+ , phenol, Cl − and NO 3 − ) were insignificant except H 2 PO 4 − and SO 4 2− which reduced the Cr(VI) uptake with a efficiency loss of 22–41%. Ionic strength had little effect on the adsorption in the range of 0–1000 mg/L NaCl. After five adsorption/desorption cycles, the PEI-HOCM still retained good adsorption capacity. More importantly, it can be separated easily after adsorption, which will greatly benefit the wastewater treatment by simplifying the separation procedure and reducing costs. Graphical abstract

Detoxification of coal-tar pitch by alkylation method and toxicity evaluation by chemical index, fumes and water-soluble substances

Wen-shuo Zhang,Li-peng Tan,Jun Shen,Yu-gao Wang,Gang Liu,Qing-bai Xu

Coal-tar pitch (CTP) output in China has consistently exceeded 7 million tons annually since 2011. CTP contains 16 toxic polycyclic aromatic hydrocarbons (PAHs) that are preferentially monitored by the American Environmental Protection Agency (EPA). As a result, the widespread and extensive use of CTP is restricted. Consequently, finding ways to reduce the toxicity of CTP has become an urgent concern. In the paper, the reaction conditions were optimized using model compounds (naphthalene, fluorene, anthracene, phenanthrene, fluoranthene, pyrene, benzo [a]pyrene) of CTP. Subsequently, the CTP was modified through alkylation reaction using 1-dodecene, and the detoxification effect was evaluated based on chemical index, fumes, and water-soluble substances. The total content of 16 toxic PAHs in the modified CTP (MCTP) decreased by 75.56% compared to the original CTP. The toxicity equivalent quotient (TEQ) of the MCTP was reduced by more than 65% according to different environmental standards. The content of the 16 PAHs detected in MCTP fumes released at 140 °C decreased by 80.89%, while in water-soluble substances it decreased by 64.04% and 76.67% at 30 °C and 60 °C, respectively. These results strongly indicate the effectiveness of the alkylation method in reducing the toxicity of CTP. However, in order to develop more environmentally-friendly materials, it is necessary to explore more efficient catalysts and cheaper modifiers, which will be the focus of our future research. Novelty Statement This study investigated the toxicity inhibition of CTP to reduce the content of PAHs by alkylation method. The modification process was analyzed using model compounds to understand the potential reaction pathways. The objective of toxicity inhibition was achieved by creating low-toxicity and long-chain alkyl substitutes. Additionally, the toxicity of CTP and MCTP was assessed by introducing fumes and water-soluble substances.

Identification of 14 glutathione S-transferase genes from Lasioderma serricorne and characterization of LsGSTe1 involved in lambda-cyhalothrin detoxification

Xi-Ya Liu,Hui-Mei Sun,Yi-Hong Luo,Mao-Ye Li,Hai-Bing Liu,Su Liu

In insects, glutathione S -transferases (GSTs) play a pivotal role in the detoxification of a wide range of pesticides. The cigarette beetle, Lasioderma serricorne , is an economically important pest insect of stored products. Recently, pyrethroid insecticides have been used to control this pest. However, little is known concerning the responses and functions of GSTs in L. serricorne under pyrethroid exposure. In this study, transcriptome sequencing was performed on L. serricorne , and a total of 14 GSTs were identified by retrieving the unigene dataset. Of these, 13 predicted GSTs fell into six cytosolic classes, namely, delta, epsilon, omega, sigma, theta, and zeta, and one was assigned to an “unclassified” group. The GST genes were differentially expressed in various larval tissues and at different developmental stages. Exposure to the pyrethroid insecticide lambda-cyhalothrin (LCT) caused oxidative stress in L. serricorne larvae and led to significantly elevated expression levels of six genes, among which LsGSTe1 was the most upregulated. Recombinant LsGSTE1 protein displayed LCT-metabolizing activity. Furthermore, LsGSTE1 protects cells against oxidative stress. Moreover, knockdown of LsGSTe1 by RNA interference dramatically increased the susceptibility of L. serricorne larvae to LCT treatment. The results from this study provide sequence resources and expression data for GST genes in L. serricorne . Our findings indicate that LsGSTE1 plays a dual role in LCT detoxification by metabolizing the pesticide and by preventing LCT-induced oxidative stress. Thus, the LsGSTe1 gene could be used as a potential target for sustainable management of the cigarette beetle.

Rational Design of a Zr-MOF@Curli-Polyelectrolyte Hybrid Membrane toward Efficient Chemical Protection, Moisture Permeation, and Catalytic Detoxification

Jing Liu,Heguo Li,Bin Yan,Chao Zhong,Yue Zhao,Xuan Guo,Jinyi Zhong

Developing high-performance protective materials is important for soldiers and civilians who are exposed to the atmosphere of highly toxic chemical warfare agents (CWAs). Polyelectrolyte membranes are promising candidates with excellent chemical resistance and moisture permeability, but they cannot efficiently degrade CWAs. Here, we design and prepare a hybrid membrane through in situ growth of catalytically active zirconium-based metal–organic frameworks (Zr-MOFs) on a polyelectrolyte membrane mediated by biofilm-inspired curli nanofibers (CNFs). Superior to the bare polyelectrolyte membrane, the prepared MOF-808@CNF-PQ hybrid membrane exhibits improved rejection of the nerve agent simulant dimethyl methyl phosphonate (DMMP) vapor and permeation of the water vapor by 113 and 45%, respectively. The water/DMMP selectivity of the hybrid membrane reaches 498.6, approximately 13 times that of the commercial polyelectrolyte membrane Nafion 117. In addition, the hybrid membrane possesses appreciable catalytic activity for the hydrolysis of the nerve agent simulant dimethyl 4-nitrophenyl phosphate (DMNP) with a half-life of ∼38 min. Nanomechanical characterization results based on atomic force microscopy (AFM) techniques demonstrate the critical role of CNFs in mediating Zr-MOF nucleation and the dominant effect of electrostatic interactions on self-assembly of CNFs on polyelectrolyte base. It is also confirmed that the Zr-MOF toppings serve as the key components in physically adsorbing and chemically degrading the DMNP molecules through multiple strong intermolecular interactions. Our work offers a rational strategy to develop advanced membranes toward efficient chemical protection, moisture permeation, and catalytic detoxification against CWAs.

Ethnopharmacological relevance Aconitum carmichaelii Debeaux, a famous traditional medicinal herb for collapse, rheumatic fever , and painful joints, always raises global concerns about its fatal toxicity from toxic alkaloids when improperly processed. Therefore, it is urgent to clarify the internal molecular mechanism of processing detoxification on Aconitum and develop simple and reliable approaches for clinical application, which is also of great significance to the rational medicinal use of Aconitum . Aim of the study The study aimed at developing a complete molecular mechanism exploration strategy in complex medicinal herb decocting system, clarifying the internal molecular mechanism of processing detoxification on Aconitum, and exploring valid approaches for detoxification. Materials and methods Aconiti Lateralis Radix Praeparata (Fuzi) was selected as the model for exploring the complex Aconitum detoxification mechanism using an advanced online real-time platform based on extractive electrospray ionization mass spectrometry. The methods realized the sensitive capture of dynamic trace intermediates, accurate qualitative and quantitative analysis, and real-time and long-term monitoring of multi-components with satisfactory accuracy and resistance to complex matrices. Results Components in the complex Aconitum decocting system were real-timely characterized and fat meat was discovered and verified to directionally detoxify Aconitum while reserving the therapy effect. More importantly, the dynamic detoxification mechanism in the chemically complex Aconitum decoction was molecularly profiled. A novel reaction pathway based on nucleophilic substitution reaction mechanism was proposed. As confirmed by the theoretic calculations at DFT B3LYP/6-31G (d) levels, fatty acids ( e.g. , palmitic acid) acted as a green, cheap, and high-performance catalyst and promote the decomposition of toxic diester alkaloids to non-toxic and active benzoyl-monoester alkaloids through the discovered mechanism. Conclusion The study exposed a novel detoxification molecular mechanism of Aconitum and provided an effective method for the safe use of Aconitum , which could effectively guide the development of traditional processing technology and compatibility regulation of the toxic herb and had great value to the modernization and standardization development of traditional medicine.
