Self-assembly of pH-responsive dextran-g-poly(lactide-co-glycolide)-g-histidine copolymer micelles for intracellular delivery of paclitaxel and its antitumor activity

Jiulong Zhang,Kang Chen,Ying Ding,Xiu Xin,Wenpan Li,Mengjun Zhang,Haiyang Hu,Mingxi Qiao,Xiuli Zhao,Dawei Chen

pH-Sensitive copolymers have been widely used in drug delivery systems to selectively enhance drug release at tumor sites. Herein, dextran (DX) was conjugated with poly(lactide-co-glycolide) (PLGA) and histidine (His) to prepare a pH-responsive nanocarrier, dextran-g-poly(lactide-co-glycolide)-g-histidine (HDP) micelles, for the delivery of antitumor drugs. Different grafting ratio HDP micelles were synthesized and prepared successfully and confirmed by the critical micelle concentration (CMC) and particle size distribution (PSD). In vitro drug release showed that the release behaviour of paclitaxel (PTX) loaded HDP micelles was pH-dependent. All blank micelles were nontoxic in the in vitro cytotoxicity assay. In an MTT assay, PTX-loaded HDP micelles with medium grafting ratios (MHDP) showed the highest cytotoxicity against MCF-7 cells. Cellular uptake experiments revealed that these pH-sensitive micelles could be taken up effectively and PTX could be delivered into the cytoplasm to reach the antitumor effect. NIR fluorescence imaging experiments demonstrated that HDP micelles could specifically accumulate in tumor sites via enhancing the permeability and retention (EPR) effect to reduce their systematic toxicity. In vivo antitumor activities showed that HDP micelles could effectively inhibit tumor growth and prolong survival time compared with free PTX solution. These results confirm that the biocompatible pH-responsive HDP micelles are a novel nanocarrier for the intracellular delivery of PTX.

Direct preparation of porous cellulose microspheres via a self-growth process on bamboo fibers and their functionalization for specific adsorption of histidine-rich proteins

Chao Liang,Liangshen Zhao,Shasha Li,Liangzhi Qiao,Tao Wang,Kaifeng Du

The traditional preparation of cellulose microspheres always involves tedious synthetic procedures (e.g., dissolution, emulsification and regeneration) and inevitable organic solvents, which undergoes both high production cost and environmental contamination. To overcome these issues, a feasible and green synthesis strategy is proposed to construct porous cellulose microspheres (PCMs) via one-step spontaneous formation relying on sodium periodate oxidation of pure bamboo fibers. By this strategy, a cluster of robust cellulose microspheres grow up on the surface of bamboo fibers in aqueous phase through amorphous oxidized cellulose self-assembly accumulation and then drop out when their sizes increase to about 15 µm. After being immobilized with Cu(II), the prepared cellulose microspheres serve as metal affinity adsorbent for proteins adsorption, showing high adsorption capacity, good selectivity and excellent reusability for bovine hemoglobin (BHb). Together with green and easy synthesis, the novel cellulose microspheres show a promising alternative to commercially available adsorbent support.

An array of poly-l-histidine functionalized multi-walled carbon nanotubes on 4-aminothiophenol self-assembled monolayer and the application for sensitively glucose sensing

Yuan Chen,Xiao-Mi Liu,Xi Wu,Xue-Chen Liu,Wen-Hao Dong,Bing-Kai Han,Xin Du,Cong Zhang,Yan-Yan Zhang,Hong-Tao Wang,Qiang Chen

Novel approachs to functionalize carboxylated multi-walled carbon nanotubes (CMWCNTs) array with poly- l -histidine (PLH) on a self-assembled monolayer (SAM) of 4-aminothiophenol (4ATP) were developed for the first time. With help of π–π interaction and chelation of the side imidazole groups in poly- l -histidine, the array showed efficient electron transfer and mild biocompatibility. By immobilizing glucose oxidase (GOx) to AuNPs electrodeposited on the array, a high-powered glucose biosensor with excellent electrocatalytic performance was fabricated and applied for glucose determination in human serum samples. The prepared biosensor exhibited highly sensitive response to glucose under the operating voltage of 600 mV with sensitivity of 96.04 μA mM −1  cm −2 , linear range of 0.005–6.5 mM, lowest detection limit (LOD) of 0.7 μΜ (S/N = 3) and Michaelis constant ( K M ) of 3.92 mM. Compared with other glucose sensors previously reported, this work showed higher sensitivity, lower detection limit and wider linear range. Furthermore, the developed array may provide a promising platform for various biosensors fabrication.

A Convenient and Label-Free Colorimetric Detection for L-Histidine Based on Inhibition of Oxidation of 3,3′,5,5′-Tetramethylbenzidine-H2O2 System Triggered by Copper Ions

Zhang Zhikun,Zhao Wenmeng,Hu Cuixia,Cao Yapeng,Liu Yumin,Liu Qingju

L-Histidine (L-His) is an essential amino acid, which is used to synthesize proteins and enzymes. The concentration of L-His in the body is controlled to regulate tissue growth and repair of tissues. In this study, a rapid and sensitive method was developed for colorimetric L-his detection using Cu2+ ions to inhibit the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB)–H2O2 system. H2O2 can oxidize TMB to oxTMB in the presence of copper, and the change in color from colorless (TMB) to blue (oxTMB) is similar to that observed in the presence of peroxidase. However, because the imidazole ring and carboxyl group of L-His can coordinate with Cu2+ ions to form stable L-His–Cu2+ complexes, the color of the TMB–H2O2 solution remains unchanged after the addition of L-His. Therefore, because L-His effectively hinders the colorimetric reaction of TMB with H2O2, this assay can be used to quantitatively determine the concentration of L-His in samples. Under optimized conditions, our colorimetric sensor exhibited two linear ranges of 60 nM to 1 μM and 1 μM to 1 mM for L-His detection and a detection limit of 50 nM (S/N = 3); furthermore, the assay can be performed within 20 min. Moreover, the proposed assay was used to determine the concentration of L-His in urine samples, suggesting that this convenient and label-free colorimetric method presents promising applications in bioanalytical chemistry and clinical diagnosis.

Effect of l-histidine and l-lysine on the properties of oil-in-water emulsions stabilized by porcine myofibrillar proteins at low/high ionic strength

Xiuyun Guo,Feng Gao,Yawei Zhang,Zengqi Peng,Muneer Ahmed Jamali

The effects of l -histidine (His) and l -lysine (Lys) on the emulsifying and interfacial properties of porcine myofibrillar proteins (MPs) at low/high ionic strength were investigated to improve physical stability of MPs emulsion at low ionic strength. Results showed that His and Lys increased emulsion stability index, and decreased creaming index at low ionic strength, but caused no significant changes in both indexes at high ionic strength. Additionaly, His and Lys increase d the emulsifying activity and apparent viscosity, but decreased the droplet size at both ionic strength. His and Lys also promoted the diffusion of MPs and increase d the interfacial pressure. Meanwhile, the interfacial loading of MPs was increased (p < 0.05) by 16.07% and 12.79% at low ionic strength, and by 8.85% and 8.24% at high ionic strength in the presence of Lys and His, respectively. Raman spectra revealed that the α-helix content of MPs was decreased (p < 0.05) by 5.05% and 11.79% at low ionic strength, and by 12.14% and 7.23% at high ionic strength in the presence of Lys and His, respectively. In summary, His and Lys enhanced the stability of MPs emulsion at low ionic strength via changing the structure and interfacial behaviour of MPs.

Design and synthesis of a multifunctional rhodamine-based chemosensor for simultaneous detection of Cu2+, Zn2+ and endogenous histidine (His) and its application in living HeLa cells and zebrafishes

Shi Zhoutong,Li Jun,Wei Gang,Cheng Biao

In organisms, the over ingesting of Cu 2+ and Zn 2+ will result in the poisoning of living cells, which severely damages the human body. Meanwhile, endogenous Histidine (His) can cause metabolic disorders like Histidinemia (His). Therefore, it is a pressing matter to design an effective method that accurately detects the amount of Cu 2+ , Zn 2+ , and His. In our study, through the nucleophilic addition and substitution reactions synthesis methods, a novel multifunctional probe, Rhodamine derivatives with vinyl functional groups at the end (RHDA) was designed and synthesized. The probe RHDA can recognize Cu 2+ and Zn 2+ separately, and the experiment further substantiates that the RHDA probe can demonstrate various turn-on colorimetric and fluorescent signals, i.e ., selectively detecting Cu 2+ in DMSO/H 2 O solution (v/v=4/6) with the “naked-eye” show evident and significant color change from colorless to pink and Zn 2+ with strong green fluorescence in DMF/H 2 O (v/v=1/1). Under optimal conditions, they could be identified under the low detection limit of 0.10 μmol·L −1 and 16.0 nmol·L −1 for Cu 2+ and Zn 2+ , respectively. Furthermore, Histidine (His) was recognized by the formed RHDA-Zn 2+ with a fluorescence “turn-off” range of 0.03–10.0 μM and a detection limit of 230.0 nmol L −1 . This article performed an extensive study of the sensing mechanisms of RHDA toward two metal ions and the ensemble RHDA-Zn 2+ toward His. The practical application experiments in DMF/H 2 O (v/v=1/1) further established that the chemosensor RHDA exhibits good cell permeability and could be employed to monitor free Zn 2+ . Finally, the complex RHDA-Zn 2+ could be applied to assay His in the living HeLa cells and zebrafishes, respectively.

Rapidly and accurately screening histidine decarboxylase inhibitors from Radix Paeoniae alba using ultrafiltration–high performance liquid chromatography/mass spectrometry combined with enzyme channel blocking and directional enrichment technique

Guizhou Hu,Lu Wang,Xinqi Li,Jin Qi

Histidine Decarboxylase (HDC), an unique enzyme responsible for the synthesis of histamine, which is an important mediator in allergy. Inhibition of HDC activity to decrease histamine production is one way to alleviate allergic symptoms. Traditional Chinese medicines (TCMs) with reported anti-allergy effect is one of important source to search for natural HDC inhibitor. Ultrafiltration combined with high–performance liquid chromatography/mass spectrometry (UF–HPLC/MS) is an effective method for screening HDC inhibitor from TCMs. Nevertheless, false-positive and false-negative results caused by the non-specific binding and the neglection of the trace active compounds are major problems in this method. In this study, an integrated strategy that combined UF–HPLC/MS with enzyme channel blocking (ECB) technique and directional enrichment (DE) technique was developed to seek natural HDC inhibitors from Radix Paeoniae alba (RPA), and at the same time, to reduce false-positive and false-negative results. HDC activity was detected to determine the validity of the screened compounds by RP–HPLC–FD in vitro . Molecular docking was applied to assay the binding affinity and binding sites. As a result, three compounds were screened from low content components of RPA after the DE. Among them, two non-specific compounds were eliminated by ECB, and the specific compound was identified as catechin, which has obvious HDC inhibition activity with IC 50 0.52 mM. Furthermore, gallic acid (IC 50 1.8 mM) and paeoniflorin (IC 50 >2 mM) from high content components of RPA were determined having HDC inhibitory activity. In conclusion, the integrated strategy of UF–HPLC/MS combined with ECB and DE technique is an effective mode for rapid and accurate screening and identification of natural HDC inhibitors from TCMs.

Oxidative characteristics and gel properties of porcine myofibrillar proteins affected by l-lysine and l-histidine in a dose-dependent manner at a low and high salt concentration

Xiuyun Guo,Junjun Wu,Xiangren Meng,Yawei Zhang,Zengqi Peng

Summary This study investigated the effects of l -lysine (Lys) and l -histidine (His) on the oxidative characteristics and gel properties of porcine myofibrillar proteins (MP). Results showed that Lys and His had a strong ferrous ion-chelating ability and hydroxyl radical-scavenging activity. Moreover, Lys and His inhibited the protein carbonyl formation and MP aggregation at 0.2 M and 0.6 M NaCl, respectively, in a dose-dependent manner. Furthermore, 2 and 4 mg mL −1 Lys and His decreased the oxidation-induced loss of the tertiary structure of MP accompanied by the lower surface hydrophobicity. The water-holding capacity and gel strength of MP gels increased with increasing Lys and His concentrations due to more regular and lamellar structures with smaller and homogeneous pores at 0.6 M NaCl and more orderly crosslinking via fibrous filament at 0.2 M NaCl. In summary, Lys and His chelated the ferrous ions and scavenged hydroxyl radicals, decreased the oxidation-induced physicochemical changes, thus preventing oxidative damage during the formation of a three-dimensional gel network, which resulted in better gel quality.

Copper nanoclusters-modified with papaya juice for fluorescence turn-on detection of serum l-histidine Today four files of proofs was sent to m.saksena@elsevier.com Please check the files

Qiqi Tan,Juan Qiao,Rongyue Zhang,Li Qi

Papaya juice protected copper nanoclusters was depicted for detection of serum l -histidine. The fluorescent probe was synthesized by using a simple “one pot” hydrothermal preparation strategy with papaya juice playing dual roles both as a green reductant and stabilizer. The excitation and emission peaks of the copper nanoclusters were at 400 nm and 490 nm, respectively. The fluorescence was turned-on in addition of l -histidine due to the enhanced surface electron density. The fluorescent intensity increased linearly with l -histidine concentration ranging from 2.0 to 50.0 µM. The proposed copper nanoclusters was further applied to determine l -histidine in rat serum after intraperitoneal injection.

An“on-off-on” reversible fluorescent probe based on nitrogen-doped carbon dots derived from Mahonia fortunei: Sequential detection of Pb2+ and L-histidine and construction of an implication molecular logic gate

Meiling Long,Yue Liu,Jun Yang,Shaogui Wu

Biomass-derived B-CDs were synthesized from Mahonia fortunei fruits. • The B-CDs showed bright blue emission with a quantum yield of 31.1%. • Pb 2+ selectively quenched B-CD fluorescence with a low LOD of 26 nM. • L-Histidine restored fluorescence via competitive Pb 2+ coordination. • An IMPLICATION logic gate was constructed from the on–off–on response. Nitrogen-doped carbon dots (B-CDs) with deep-blue fluorescence were synthesized through a one-step hydrothermal route using the fruits of the medicinal plant Mahonia fortunei as the biomass carbon source and 1-naphthylamine as the nitrogen dopant. The resulting B-CDs showed excitation-independent emission, a high fluorescence quantum yield of 31.1%, and excellent resistance to salt, photobleaching, and long-term storage. Pb 2+ efficiently quenched B-CD fluorescence by forming non-emissive ground-state complexes through bidentate coordination with oxygen- and nitrogen-containing surface groups, switching the probe to the “off” state. By contrast, L-histidine (L-His), whose imidazole group has stronger competitive affinity for Pb 2+ , dissociated the carbon dot‑lead complexes and markedly restored fluorescence, returning the system to the “on” state. The platform afforded a detection limit of 26 nM for Pb 2+ , while the L-His-induced fluorescence recovery response was linear over 40–80 μM, with an average fluorescence recovery efficiency of 88.2%. On the basis of this reversible fluorescence response, an IMPLICATION molecular logic gate was constructed using Pb 2+ and L-His as chemical inputs and fluorescence intensity as the output, enabling logic discrimination in a two-component system. UV–vis absorption spectroscopy, time-resolved fluorescence lifetime analysis, and FT-IR spectroscopy jointly supported the reversible “on-off-on” response mechanism. This work provides a practical strategy for developing natural plant-derived carbon-dot platforms for reversible fluorescence sensing and molecular logic devices. A ratiometric and far-red fluorescence “off-on” sensor for sequential determination of copper(II) and L-histidine based on FRET system between N-acetyl-L-cysteine-capped AuNCs and N,S,P co-doped carbon dots

Dong Wenjuan,Wang Ruiping,Gong Xiaojuan,Liang Wenting,Fan Li,Song Shengmei,Dong Chuan

A far-red fluorescence “off-on” sensing strategy is described for sequential ratiometric determination of Cu 2+ and L-histidine (L-His) based on fluorescence resonance energy transfer (FRET) system. N,S,P co-doped carbon dots (N,S,P-CDs) and N-acetyl-L-cysteine functionalized gold nanoclusters (NAC-AuNCs) are used in the FRET system, which serve as energy donor and acceptor, respectively. After adding NAC-AuNCs into the solution of N,S,P-CDs, the fluorescence of N,S,P-CDs is effectively quenched, while the far-red fluorescence of NAC-AuNCs appears. Cu 2+ can decrease fluorescence of NAC-AuNCs, and then L-His can effectively recover the fluorescence of NAC-AuNCs. The possible reason is that the stronger affinity between Cu 2+ and L-His can pull Cu 2+ away from the surface of NAC-AuNCs. Through it all, the emission intensity of N,S,P-CDs remains nearly constant, so the ratio of fluorescence intensities at 485 and 625 nm exhibits a linear correlation to the Cu 2+ and L-His concentration, respectively. The sensing platform shows good selectivity towards Cu 2+ and L-His with a linear range of 0.65–26.58 μM and 3.13–56.25 μM and determination limits of 0.50 μM and 0.374 μM, respectively. The proposed method has been successfully used for Cu 2+ and L-His determination in real samples with satisfying results. Graphical abstract
