In Vivo Selective Capture and Rapid Identification of Luteolin and Its Metabolites in Rat Livers by Molecularly Imprinted Solid-Phase Microextraction

Die Gao,Dan-Dan Wang,Qian Zhang,Feng-Qing Yang,Zhi-Ning Xia,Qi-Hui Zhang,Chun-Su Yuan

A method based on molecularly imprinted solid-phase microextraction (MIP-SPME) coupled with liquid chromatography-quadrupole time-of-flight tandem mass spectrometry (QTOF-MS/MS) was developed for the detection of luteolin and its metabolites in vivo. The MIP-SPME fibers were first fabricated by dopamine and silane, and then luteolin MIPs-coated fibers were successfully prepared using luteolin, acrylamide (AM), and ethylene glycol dimethacrylate (EGDMA) as the template, functional monomer and cross-linker, respectively. The characterizations of polymers were analyzed by scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FT-IR), and the Brunauer–Emmett–Teller method (BET). The properties involving adsorption and selective experiments were evaluated, and these results revealed that MIP fibers presented high adsorption capacity and selectivity to luteolin. Furthermore, the developed MIP-SPME coupled with the LC-QTOF-MS/MS method was adopted to capture and identify luteolin and its metabolites in rat livers in vivo, and eventually, apigenin, chrysoeriol, and diosmetin were rapidly identified as metabolites.

Luteolin improves myocardial cell glucolipid metabolism by inhibiting hypoxia inducible factor-1α expression in angiotensin II/hypoxia-induced hypertrophic H9c2 cells

Jia Wang,Tian Gao,Feng Wang,Jie Xue,Hua Ye,Meilin Xie

Luteolin , a natural flavonoid , can attenuate hepatic lipid accumulation and insulin resistance in obese mice . Therefore, we hypothesized that luteolin may also improve the abnormal glucolipid metabolism of hypertrophic myocardial cells. This study aimed to investigate the effect and possible molecular mechanisms of luteolin. Hypertrophic H9c2 cells were induced by angiotensin II/hypoxia and simultaneously treated with 2 to 8 μg/mL luteolin for 24 h. Luteolin might dose-dependently decrease intracellular total protein, atrial natriuretic peptide , and free fatty acid levels , and increase supernatant glucose levels . Western blot assay showed that luteolin could inhibit the expressions of intracellular hypoxia inducible factor-1α (HIF-1α) and glucose transporter-4 (GLUT-4) proteins, and increase the expressions of intracellular peroxisome proliferator-activated receptor α (PPARα), carnitine palmitoyltransferase-1A (CPT-1A), and pyruvate dehydrogenase kinase-4 (PDK-4) proteins. These findings demonstrate that luteolin can improve abnormal glucolipid metabolism in angiotensin II/hypoxia-induced hypertrophic H9c2 cells, and its mechanisms are related to the inhibition of HIF-1α expression and subsequent modulation of PPARα-mediated target genes, including CPT-1A, PDK-4, and GLUT-4.

A disposable and sensitive sensor based on a ZIF-8@graphene modified carbon paper electrode for the quantitative determination of luteolin

Wanyu Pang,Yali Gao,Tuoping Hu,Xuemei Ma

Rapid and accurate determination of luteolin is of great significance for pharmaceutical quality control. Herein, a disposable and sensitive luteolin sensor was fabricated by a hydrothermal method with carbon paper as substrate where ZIF-8 grew on GR in situ. Notably, the large specific surface area of ZIF-8 provided active sites on the electrode surface and the ability of GR to promote electron transfer greatly improved the sensitivity towards the oxidation of luteolin. Under the optimum conditions, the ZIF-8@GR/CP showed excellent detection performance for luteolin with a linear detection range of 0.04–3.2 μM and 3.2–120 μM, with LOD of 12 nM (S/N = 3). Furthermore, this disposable and sensitive sensor was successfully applied for the quantitative detection of luteolin in a capsule of Lamiophlomis rotata.

Rapid quantitative detection of luteolin using an electrochemical sensor based on electrospinning of carbon nanofibers doped with single-walled carbon nanoangles

Liwen Zhang,Danfeng Qin,Jun Feng,Tingfan Tang,Hao Cheng

In this study, single-walled carbon nanoangles/carbon nanofibers (SWCNHs/CNFs) were synthesized by electrospinning, followed by annealing in a N2 atmosphere. The synthesized composite was structurally characterized by scanning electron microscopy, transmission electron microscopy, and X-ray photoelectron spectroscopy. The electrochemical sensor was fabricated by modifying a glassy carbon electrode (GCE) for luteolin detection, and its electrochemical characteristics were investigated using differential pulse voltammetry, cyclic voltammetry, and chronocoulometry. Under optimized conditions, the response range of the electrochemical sensor to luteolin was 0.01–50 μM, and the detection limit was 3.714 nM (S/N = 3). The SWCNHs/CNFs/GCE sensor showed excellent selectivity, repeatability, and reproducibility, thus enabling the development of an economical and practical electrochemical method for the detection of luteolin.

Enhancement of oral bioavailability and anti-colitis effect of luteolin-loaded polymer micelles with RA (rosmarinic acid)-SS-mPEG as carrier

Zhaomin Lu,Juan Liu,Liangjian Zhao,Chenli Wang,Feng Shi,Zhengqi Li,Xuesong Liu,Zhiwei Miao

Objective Polymer micelles were prepared (L-RSPMs) with luteolin and synthetic RA-SS-mPEG polymeric material before evaluation of their anti-inflammatory effect on 2, 4, 6-trinitro-benzene-sulfonic acid (TNBS)-induced ulcerative colitis (UC) model in rats. Methods The synthetic RA-SS-mPEG was characterized with NMR spectroscopy, before preparation of luteolin-coated RA-SS-mPEG polymer micelles. The in vitro characterization and evaluation of the formulation were accomplished, couple with its pharmacokinetic parameters. The levels of PEG2, MDA, CRP and GSH, as well as concentrations of TNF-α, IL1-β, IL-6 and IL-10 in serum and colon tissue were detected via ELISA kit. The degree of colon injury and inflammation was evaluated via histopathologic examination. Results L-RSPMs displayed small average droplet size (133.40 ± 4.52 nm), uniformly dispersed (PDI: 0.163 ± 0.011), good stability, slow release and enhanced solubility. We observed 353.28% increase in the relative bioavailability of L-RSPMs compared to free luteolin, while the half-life of the micelle was extended by 6.16h. Compared to model (M) group, luteolin (low and high doses) and L-RSPMs (low and high doses) significantly reduced levels of MDA, PEG2, CRP, TNF-α, IL-6 and IL-1β in colon tissue and serum of colitic rats but dose dependently increased IL-10 and SOD levels (p < 0.01). Histopathologic examination of colon showed that luteolin (low and high doses) and L-RSPMs (low and high doses) improved colonic inflammation in colitic rats to varying degrees compared to M group. Conclusion L-RSPMs could improve TNBS-induced colon inflammation by enhancing bioavailability, promoting antioxidant effects and regulating cytokine release, which may become a potential agent for UC treatment in clinical settings.

Ameliorative Effects of Luteolin and Activated Charcoal on Growth Performance, Immunity Function, and Antioxidant Capacity in Broiler Chickens Exposed to Deoxynivalenol

Mubashar Hassan,Yanan Wang,Shahid Ali Rajput,Aftab Shaukat,Ping Yang,Muhammad Zahid Farooq,Qianhui Cheng,Mehboob Ali,Xiaomei Mi,Yu An,Desheng Qi

Deoxynivalenol (DON, Vomitoxin) is a threatening mycotoxin that mainly produces oxidative stress and leads to hepatotoxicity in poultry. Antioxidant dietary supplements dramatically boost immunity, safeguarding animals from DON poisoning. Luteolin (LUT) is an active plant-derived compound that poses influential antioxidants. This study explored the effectiveness of LUT in combination with activated charcoal (AC) in detoxifying DON in broilers. The 180 one-day broiler chickens were allocated into five different groups having six replicates in each group, provided with ad libitum feed during the trial period (28 days) as follows: in the control group, basal diet (feed with no supplementation of LUT, AC or DON); in group 2, a basal diet added with 10 mg/kg DON from contaminated culture (DON); in group 3, a basal diet augmented by 350 mg/kg LUT and DON 10 mg/kg (DON + LUT); in group 4, a basal diet supplemented by DON 10 mg/kg + AC 200 mg/kg (DON + AC); and in group 5, a basal diet supplemented by 10 mg/kg DON + 350 mg/kg LUT + 200 mg/kg AC (DON + LUT + AC). Concerning the control group, the DON-treated broilers demonstrated a significant decrease in growth performance (p< 0.05) and serum immunoglobulin (p< 0.05) contents, negatively changing the serum biochemical contents and enzymatic activities and an increase in histopathological liver lesions. Furthermore, DON substantially increased (p< 0.05) malondialdehyde (MDA) concentration and decreased total superoxide dismutase (T-SOD), catalase (CAT), and glutathione peroxidase (GSH-Px) levels in the serum and liver. The intake of AC and LUT to the DON-contaminated diet decreased DON residue in the liver and potentially reduced the adverse effects of DON. Considering the results, supplementation of LUT with mycotoxin adsorbent has protective effects against mycotoxicosis caused by DON. It could be helpful for the development of novel treatments to combat liver diseases in poultry birds. Our findings may provide important information for applying LUT and AC in poultry production. Keywords:deoxynivalenol;luteolin;activated charcoal;growth performance;antioxidant capacity;immunity;broilersKey Contributions:Supplementation of luteolin and activated charcoal for four weeks improve the immune system and antioxidant capacity and protects liver injury in broiler chickens against DON-induced toxicity.

Overoxidized polypyrrole/overoxidized poly(3,4-ethylenedioxythiophene)/graphene quantum dots-chitosan as enhanced electrochemical sensing platform for luteolin determination in peanut shells

Rongqian Meng,Yanfeng Xue,Lijun Guo,Hong Yang,Yongbo Song,Qiaoling Li,Yulan Niu

A novel electrochemical sensor was constructed based on an overoxidized polypyrrole/overoxidized poly(3,4-ethylenedioxythiophene)/graphene quantum dots-chitosan composite film modified glass carbon electrode (oPPy/oPEDOT/GQDs-CS/GCE), for the accurate determination of luteolin in peanut shells. The oPPy/oPEDOT/GQDs-CS/GCE was prepared using layer-by-layer assembly methods. After incubation in NaOH–EtOH solution, the current of luteolin on the composite electrode increased from 0 to ∼30 μA, the Rct value decreased by ∼80 fold, and the specific surface area (A) increased by ∼7 fold. Based on a three-level four-factor Box–Behnken design (BBD), the optimal preparation conditions for the composite electrode were as follows: a mass ratio of CS to GQDs of 5, a polymerization cycle number for EDOT of 4, a polymerization cycle number for Py of 4, with the peak current increased to 43.21 μA (RSD = 3.51%, n = 3). A morphology with multi-channels and grooves was observed on the surface of the incubated composite film via SEM analysis. The additional –CN– and –SO/–SO2 groups formed were observed by XPS and FTIR analysis. The electrochemical sensor constructed exhibited good anti-interference ability, repeatability, and stability. The linear detection range recorded by the current responses of the composite electrode to luteolin was 0.01 to 10.0 μM (R2 = 0.991) and the limit of detection was 7.5 nM (S/N = 3). The observed content of luteolin in peanut shells was 0.769%, which was close to the results obtained from High Performance Liquid Chromatography (HPLC) analysis (0.808%). This agreement indicated that the constructed electrochemical sensor based on oPPy/oPEDOT/GQDS-CS/GCE was capable of accurate detection of luteolin in peanut shells.

Single-walled carbon nanohorns and cobalt@nitrogen-doped carbon nanotube polyhedrons modified electrode as sensitive electrochemical sensing platform for luteolin detection

Ai Yijing,Luo Guiling,Yan Lijun,Yao Yucen,Zhang Zejun,Wang Bei,Sun Wei

In this work, ZIF-8@ZIF-67 was prepared and pyrolyzed to obtain Co nanoparticles-embedded N-doped carbon nanotube hollow polyhedron (Co@NCNHP). Then, single-walled carbon nanohorns (SWCNHs) were mixed with Co@NCNHP to obtain the SWCNHs-Co@NCNHP composite. The morphological structure of the prepared material was characterized by scanning electron microscopy, and the as-prepared SWCNHs-Co@NCNHP was modified on the surface of glassy carbon electrode (GCE). The modified electrode was used as electrochemical sensor for analysis of luteolin. Because of the excellent synergistic effects between SWCNHs and Co@NCNHP, the sensor based on SWCNHs-Co@NCNHP/GCE could be used to detect luteolin with the detection range from 0.1 to 4.0 μM and the low limit of detection of 10.0 nM (S/N = 3). Besides, the fabricated sensor showed favorable selectivity and stability, and was used in the analysis of luteolin in Duyiwei capsules with satisfactory results.

Ultrasound Improved the Non-Covalent Interaction of β-Lactoglobulin with Luteolin: Regulating Human Intestinal Microbiota and Conformational Epitopes Reduced Allergy Risks

Titi Wang,Wenmei Chen,Yanhong Shao,Jun Liu,Zongcai Tu

The present study aims to investigate the effects of ultrasound on the non-covalent interaction of β-lactoglobulin (β-LG) and luteolin (LUT) and to investigate the relationship between allergenicity and human intestinal microbiota. After treatment, the conformational structures of β-LG were changed, which reflected by the decrease in α-helix content, intrinsic fluorescence intensity and surface hydrophobicity, whereas the β-sheet content increased. Molecular docking studies revealed the non-covalent interaction of β-LG and LUT by hydrogen bond, van der Walls bond and hydrophobic bond. β-LG-LUT complex treated by ultrasound has a lower IgG/IgE binding ability and inhibits the allergic reaction of KU812 cells, depending on the changes in the conformational epitopes of β-LG. Meanwhile, the β-LG-LUT complex affected the composition of human intestinal microbiota, such as the relative abundance ofBifidobacteriumandPrevotella. Therefore, ultrasound improved the non-covalent interaction of β-LG with LUT, and the reduction in allergenicity of β-LG depends on conformational epitopes and human intestinal microbiota changes.Keywords:ultrasound;β-LG;LUT;allergenicity;human intestinal microbiota

Binary-amplifying electrochemiluminescence sensor for sensitive assay of catechol and luteolin based on HKUST-1 derived CuO nanoneedles as a novel luminophore

Beibei Wang,Wenrui Zhao,Lin Wang,Kai Kang,Xianrui Li,Duo Zhang,Jujie Ren,Xueping Ji

Herein, a highly novel and effective electrochemiluminescence (ECL) sensor based on metal-organic framework (MOF, HKUST-1) derived CuO nanoneedles (HKUST-1 derived CuO NNs), gold nanoparticles (AuNPs) and TiO 2 was developed for ultrasensitive detection of catechol and luteolin. The HKUST-1 derived CuO NNs were employed as luminophore for the first time, which were successfully fabricated by using HKUST-1 as precursor. The results revealed that the HKUST-1 derived CuO NNs exhibit excellent ECL activity ascribed to its abundant active site and the high specific surface area, thus obviously promoting the separation and transfer of charge and further improving the current density of ECL sensor. To binary-amplify the signal of the ECL sensor, the AuNPs and TiO 2 nano-materials with good biocompatibility, great electron transport efficiency and high catalytic activity were used as co-reaction accelerators in the ECL process. Dependent on the above brilliant strategy, the proposed ECL sensor achieved wide linear ranges from 3 × 10 −9 – 1 × 10 −4  M for catechol and 1 × 10 −8 – 2 × 10 −4  M for luteolin, with the detection limits of 1.5 × 10 −9  M for catechol and 5.3 × 10 −9  M for luteolin, respectively. Furthermore, the ECL sensor exhibited outstanding selectivity, repeatability, stability and obtained great feedback on determination of catechol and luteolin in actual samples. The method not only filled a gap in the ECL application of MOF-derived materials but also provided a novel sight for design other highly efficient luminescent materials.

Fabrication and Characterization of Gelatin-Finger Citron Polysaccharide Nanoparticles for Enhanced Solubility and Bioavailability of Luteolin in Treating Acute Alcoholic Liver Disease

Feilin Wang,Yi Liu,Peijian Cai,Xin Zhong,Jinhong Zhong,Yufu Li,Huizhong Hu,Yuanhao Sheng,Haihui Pan,Fansheng Kong

Luteolin (Lut) is a natural flavonoid that has been widely used in the treatment of liver and glycolipid metabolic diseases. However, poor water solubility and bioavailability limit its efficacy and application. To overcome these challenges, a protein–polysaccharide composite nanoparticle (L-GF NPs) for Lut delivery was prepared by a self-assembly method based on amphiphilic gelatin and active finger citron polysaccharide (FCP). The L-GF NPs were approximately spherical in shape and were formed by hydrogen bonding, electrostatic interaction, and hydrophobic interaction. Lut was encapsulated in the nanoparticles in an amorphous state, and the encapsulating efficiency was 79.49%. The efficacy of L-GF NPs was evaluated by intragastric administration of an alcoholic liver disease (ALD) mouse model. L-GF NPs have been demonstrated to significantly ameliorate the symptoms of ALD mice, which are mainly achieved by reducing lipid accumulation and oxidative stress and inhibiting the NF-κB pathway. In conclusion, the aqueous solubility and bioavailability of Lut were markedly enhanced by FCP encapsulation. Meanwhile, the combined effect of Lut and FCP significantly ameliorated ALD and attenuated alcohol-induced lipid metabolic disorders and oxidative damage. This provides a new strategy to promote the prevention and treatment of ALD.

Experimental Assessment, Quantum Computation (DFT, ELF, and LOL), and Integrative Nutriology Insights on Lactoferrin/Luteolin Nanocomplex Self-Assembly via pH-Driven Method

Guan Tianzhu,Ren Chenxi,Feng Yining,Gao Ya,Li Ning,Liang Li,Rao Shengqi,Yang Zhenquan,Liu Qiaoquan

As the limited water solubility and stability severely restrict the food application of luteolin, a deep knowledge of luteolin is required to understand the reactivity in complex formation. In the present study, the formation mechanism between lactoferrin and luteolin via a pH-driven method was systematically explored to confirm the application in hydrophilic nutraceutical delivery. The physicochemical properties of lactoferrin-luteolin nanocomplexation were investigated through particle size, PDI, ζ-potential, encapsulation efficiency, and loading amount assays. Observation of complexation behaviors revealed the pH-driven method effectively incorporated luteolin into the cores of lactoferrin with pH-dependent solubility characteristics. The improved thermal stability resulted in a stable nanocomplex formation with a zeta potential near − 15 mV. Furthermore, multi-spectroscopy (FTIR and fluorescence spectroscopy) demonstrated that luteolin-lactoferrin nanocomplexation was a spontaneous process together with secondary structure alterations during acidification. Fascinating biological applications of luteolin inspire to do quantum chemical calculations in order to confirm its bioactivities. In-depth investigations of electronic structure (MEP), topology characteristic (ELF, and LOL), and frontier molecular orbital (HOMO and LUMO, energy gap) help confirm the nucleophilic and electrophilic regions of luteolin, which play a crucial role in recognition between luteolin and lactoferrin. Consistent with the observed behavior in vitro, molecular docking further elucidated that hydrophobic contact and hydrogen bonding dominated the location of luteolin into the hydrophobic and deprotonated pocket of lactoferrin, especially at pH 4. From the integrative nutriology perspective, the synergistically biological mechanism of lactoferrin-luteolin nanocomplexation possessed multi-target and multi-pathway properties in preventing oxidative stress-related diseases. In conclusion, experimental and theoretical studies demonstrate great potential in using the pH-driven method for manufacturing luteolin-lactoferrin nanocomplexation with better physical and biological characteristics for delivery of hydrophobic nutraceuticals in the food sector.
