As a major public health issue, early cancer detection is of great significance. A field-effect transistor (FET) based on an MoS2/C60composite nanolayer as the channel material enhances device performance by adding a light source, allowing the ultrasensitive detection of cancer-related miRNA. In this work, atomic layer deposition (ALD) was used to deposit MoS2layer by layer, and C60was deposited by an evaporation coater to obtain a composite nanolayer with good surface morphology as the channel material of the FET. Based on the good absorption of C60by blue-violet light, a 405 nm laser was selected to irradiate the channel material, improving the function of FET biosensors. A linear detection window from 10 pM to 1 fM with an ultralow detection limit of 5.16 aM for miRNA-155 was achieved. Keywords:ultrasensitive detection;FET;MoS2/C60composite nanolayer

Background The complex process by which lactation is initiated upon neonate delivery remains incompletely understood. Microvesicles (MVs) can transmit microRNAs (miRNAs) into recipient cells to influence cell function, and recent studies have identified miRNAs essential for mammary gland development and lactation. This study aimed to investigate the expression of lactation-related miRNAs in MVs isolated from human umbilical cord blood immediately after delivery. Material/Methods Umbilical cord blood samples were collected from 70 healthy pregnant women, and MVs were isolated through differential centrifugation and characterized by transmission electron microscopy, Western blotting, and nanoparticle tracking analysis. Lactation-related miRNAs were screened using bioinformatics tools for miRNA target prediction, gene ontology, and signaling pathway analyses. miRNA PCR arrays were used for miRNA expression analysis, and the results were validated by real-time PCR. Upon exposure of HBL-100 human mammary epithelial cells to MVs, MV uptake was examined by fluorescence confocal microscopy and β-casein secretion was detected by ELISA. Results Spherical MVs extracted from umbilical cord blood expressed CD63 and had an average diameter of 167.0±77.1 nm. We profiled 337 miRNAs in human umbilical cord blood MVs and found that 85 were related to lactation by bioinformatics analysis. The 25 most differentially expressed lactation-related miRNAs were validated by real-time PCR. MV uptake by HBL-100 cells was after 4 h in culture, and significantly increased secretion of β-casein was observed after 96 h from cells exposed to MVs (P<0.05). Conclusions Umbilical cord blood MVs contain many lactation-related miRNAs and can induce β-casein production by HBL-100 cells in vitro . Thus, umbilical cord blood MVs may mediate secretion of β-casein through miRNAs, thereby playing an important role in fetal-maternal crosstalk.

The simultaneous imaging of the dynamic expression variations of regulatory RNAs in cells, which remains a major challenge, has important applications in precise disease diagnosis, treatment and prognosis. Here, we describe the establishment of a biodegradable ZnO nanoparticle (NP)-assisted asymmetric amplification approach for the simultaneous imaging of microRNA-21 (miRNA-21) and programmed cell death 4 (PDCD4) mRNA at distinct expression levels in live cells. The DNA signal probe complexes are immobilized on the ZnO NPs and readily delivered into the target cancer cells via the endocytosis pathway. The acidic microenvironment in cancer cells leads to the dissolution of the ZnO NPs to release Zn2+ ions and the intracellular miRNA-21 activates the Zn2+-dependent DNAzyme to cleave the substrate signal probes with the assistance of the Zn2+ cofactor to show green fluorescence for imaging miRNA-21. Meanwhile, the PDCD4 mRNA can displace the other quenched signal probes to generate red fluorescence. Importantly, the PDCD4 mRNA sequences can be recycled and reused by using the DNAzyme-cleaved sequences as the fuel strands through two strand displacement reactions to yield amplified red fluorescence for detecting low levels of PDCD4 mRNA. Moreover, our approach can be used to evaluate the varied expression levels of miRNA-21 and PDCD4 mRNA responsive to different drugs in cells, reflecting its usefulness for precise cancer diagnosis and prognosis upon anticancer drug treatment.

Early cancer detection is crucial for improving patient survival rates. However, current single-biomarker detection methods often face challenges, such as insufficient sensitivity, poor accuracy, and false positives. To address these issues, we report a dual-functionalized glass micropipet sensor (DFMS) capable of simultaneously detecting two cancer biomarkers, nucleic acids and proteins. The inner surface of the sensor is functionalized with amino-modified silicon nanowires (SiNWs) to capture disease-related miRNAs, enabling ionic-current-based detection, while the outer surface is decorated with gold nanoparticles to anchor specific protein aptamers for Raman-based detection. This dual-functionalization significantly enhances the sensitivity and selectivity by combining ionic current amplification with plasmonic Raman signal enhancement. The sensor achieves detection limits of 1 aM for miRNAs and 0.001 ng/mL for proteins, with minimal mutual interference between the two detection modes, ensuring accurate and independent detection. Validation with prostate cancer biomarkers miRNA-1246 and PSA, as well as gastric cancer biomarkers miRNA-106a and CD44, demonstrates its outstanding sensitivity, selectivity, stability, and broad applicability, providing a novel approach for early cancer detection with significant clinical implications.

A sensitive electrochemiluminescence (ECL) assay for microRNAs (miRNAs) based on a semiconductor nanomaterial sensitized with noble-metal Au nanoparticles (NPs) is successfully developed. TiO 2 nanotubes (NTs) were equipped with Au NPs to obtain an enhanced ECL emitter. Then, an ECL assay for miRNA-21 was fabricated, which was based on the use of probe 2 DNA–functionalized Pt/PAMAM nanocomposites (NCs) assembled on the surface of Au/TiO 2 NT conjugate via DNA hybridization between probe 1 DNA and capture DNA. The Pt/PAMAM NCs act as an ECL quencher of Au/TiO 2 NTs via resonance energy transfer. After the binding of target miRNA-21 and the capture DNA, the Pt/PAMAM NCs were released and the ECL signal was recovered. An “off–on” ECL assay was achieved with a linear response from 0.01 to 10,000 pM. Finally, this method has been validated to be sensitive and specific for miRNAs in human serum samples. The ECL enhancement strategy opens a new way for fabricating various sensitive biosensors.
A sensitive “off–on” electrochemiluminescence analysis method was developed, which combined Au NP–enhanced ECL emission of TiO 2 nanotubes and an efficient energy-transfer system between Au/TiO 2 nanotubes and Pt/PAMAM nanocomposites

Tea is a widely consumed beverage globally, but the tea industry faces a significant waste management challenge. In this study, we developed tea leaf exosome-like nanoparticles (TELNs) with an average size of 274 ± 24.7 nm and a zeta potential of -20.6 ± 0.78 mV, using polyethylene glycol (PEG) 6000 precipitation followed by ultracentrifugation. Structural analysis confirmed that TELNs are composed of lipids, proteins, and RNAs. In vitro assays on HepG-2 cells revealed that TELNs are non-toxic at concentrations up to 300 µg/mL and can be efficiently internalized. TELNs exhibited significant antioxidant capacity and were able to significantly ameliorate H 2 O 2 -induced oxidative stress, increase the viability and reduce the accumulation of ROS in Hepg-2 cells. Notably, TELNs significantly alleviated OA-induced lipid metabolic disorders and hepatocellular injury. Further molecular analysis revealed that TELNs downregulated the expression of miR-21-5p, miR-17-3p, and miR-107, leading to the upregulation of their target genes PPARα, CYP7A1, and CPT-1A, which contributed to the improvement of lipid metabolism. This study is the first to demonstrate the lipid metabolism regulation potential of TELNs, providing new insights into their underlying mechanisms and helping to develop new therapeutic strategies for lipid metabolism-related diseases. Furthermore, it expands the scope of tea use and helps to reuse tea residues. Graphical

MicroRNAs (miRNAs) are key regulators of gene expression and play a crucial role in various biological processes. However, existing methods for miRNA detection face challenges in terms of sensitivity, specificity and ease of use. These limitations highlight the need for innovative and highly sensitive detection platforms capable of accurately quantifying miRNAs. In this work, an integrated biochip based on a self-powered sensor system developed for the ultra-sensitive detection of miRNAs-21 and miRNA-20a. The system uses a bilirubin oxidase (BOD)-supported bio-cathode and a glucose oxidase (GOD)-supported bio-anode to form a closed loop that converts chemical energy into electrical energy. The complementary chains of target miRNAs are modified on the bio-anode using a bio-conjugate amplification strategy, enabling the system to generate an electrical signal proportional to the concentration of miRNAs. The sensor has a wide linear range of 5 fmol/L to 10 nmol/L for miRNA-20a and 1 fmol/L to 10 fmol/L for miRNA-21, with low detection limits of 0.176 fmol/L and 0.138 fmol/L, respectively. These detection limits are significantly lower than those achieved by existing technologies, making our biosensor a significant advancement in the field. In addition, the sensor system is integrated into a portable chip that transmits electrical signals in real time via Bluetooth to a mobile phone interface, enabling convenient and rapid analysis.

Herein, we present a colorimetric sensing strategy for the identification and quantification of tumor-associated miRNAs based on dual DNAzyme amplification. In this sensing ensemble, the substrate portion of the Pb 2+ -dependent 8−17 DNAzyme combines with the G-quadruplex portion to form a hairpin substrate strand. The two split 8−17 DNAzyme strands are partially complementary to the substrate strand and serve as a recognition unit for binding the target miRNA. In the presence of the target miRNA, the activated DNAzyme cleaves the substrate strand, releasing the G-quadruplex. This G-quadruplex binds to hemin to form a G-quadruplex/hemin complex with horseradish peroxidase (HRP)-like properties, which catalyzes the oxidation of ABTS 2− by H 2 O 2 . This oxidation reaction produces a colorimetric signal output, enabling the detection of the target miRNA. Under the optimal reaction conditions explored in this study, the constructed sensing ensembles tailored for each of the specific target miRNAs successfully identified and quantified the four target miRNAs—miR-122, miR-21, miR-335, and miR-155—in both buffer solutions and cell extracts. This colorimetric sensing strategy offers significant advantages in terms of simplicity, cost, and versatility and holds great potential for wide application in biomedical research and clinical diagnostics.

Diabetic wound healing is critically impaired by dysregulated macrophage polarization, compromised endothelial angiogenic function, and diminished fibroblast proliferation/migration under persistent hyperglycemia. Current therapies, predominantly focused on single-cell targeting, lack coordinated modulation across these key cellular components. We developed a novel triple-targeting core-shell nanoparticle (miR-RPC) leveraging the shared integrin αvβ3 receptor on macrophages, endothelial cells, and fibroblasts to address this limitation. miR-RPC features an RGD/phosphatidylserine (PS)-modified lipid shell encapsulating a chitosan/miR-146a-5p core. This miRNA was selected as a model RNA because of its widely recognized beneficial role in three key cell types in wound healing. The RGD peptide enables specific αvβ3-mediated triple-targeting. The anionic lipid PS facilitates core-shell assembly via electrostatic interaction with the cationic chitosan/RNA core and mimics apoptotic signals to enhance macrophage phagocytosis and phenotypic transition. miR-RPC effectively reprogrammed macrophages towards the M2 phenotype, restored endothelial angiogenic capacity under high glucose, and stimulated fibroblast proliferation, migration, and collagen secretion. Incorporated into a gelatin methacrylate (GelMA)/oxidized hyaluronic acid (OHA) double cross-linked hydrogel (GelO), miR-RPC@GelO significantly accelerated diabetic wound healing in rat models, demonstrating reduced inflammation, increased vascular density, and enhanced collagen deposition. This innovative triple-targeting system achieves coordinated diabetic wound repair through synergistic “immunomodulation-angiogenesis-collagen deposition” mechanisms, offering a promising therapeutic approach. Furthermore, the successful preparation of miR-RPC expands the application of anionic lipids in RNA delivery systems and highlights its potential as a versatile gene delivery vector.

Remote activation of biomarker sensing holds a great promise of shifting the success of in vitro diagnostics to spatiotemporally controlled in vivo visualization of tumor, and in turn, imaging guided therapy. Herein, a “dual-key-one-lock” nanodevice was designed and built by assembling thermo-activatable probe of trimeric DNA hybrids into a mesoporous polydopamine nanoparticle-based multifunctional nanotransducer (probe host, fluorescence quencher, and photothermal conversion agent), enabling precisely switchable theranostic operations under the co-activation of exo/endogenous stimulations (near-infrared (NIR) light and microRNA (miRNA)). By this design, the NIR irradiation-induced local heat through the porous nanotransducer can be transferred to the DNA nanothermometer for triggering the exposure of the miRNA recognition segment, as well as the subsequent fluorescence activation by strand displacement reactions (SDR). A programmable application of short- (3 min) and long-duration (10 min) NIR irradiation was administered sequentially to induce a milder and a stronger hyperthermia, respectively, to activate the localized miRNA imaging, and in turn, tumor thermoablation under the fluorescence guidance in vivo . By reducing nonspecific activation, dual factor co-activatable nanodevices exhibited a high tumor-to-background ratio (TBR) value of 8.9, as well as a significantly lower (6–9-fold) normal tissue fluorescence as compared with those sensing miRNA solely. The in vivo results show that the tumors were significantly suppressed after the photothermal therapy with the assistance of the accurate miRNA diagnosis. This rationally integrated nanoplatform may pave a new avenue for advanced theranostic systems with high spatiotemporal precision by activatable designs.

Therapeutic efficacy of solid tumor is often severely hampered by poor penetration of therapeutics into diseased tissues and lack of tumor targeting. In this study, the functionalized upconversion nanoparticles (UCNP)-based delivery vector targeting cancer cells was developed. Firstly,
NaYF4:Yb/Tm (UCNP) was prepared with the solvothermal method for the uniform nanoparticle size and brilliant lattice structure. The SiO2 coated UCNP was demonstrated a high upconversion emission and good monodispersity, which was coupled with polyetherimide (PEI) and
miR-145 vector. Then, it was further functionalized via hyaluronic acid (HA) (UCNP/PEI/HA Nanocomplex, UCNPs) coating for the targeted delivery and improved biocompatibility. The UCNPs/miR-145 displays an excellent biocompatibility, a high level of cellular uptake and miR-145 expression, which
results in a significant cell cycle arrest in G1, and induces CCND1, CDK6 and CCNE2 proteins downregulation. In vivo, the HA-coated UCNPs were enriched at the tumor site by targeting and retention effects, which resulted in a significant inhibition of tumor growth. Histological experiments
demonstrated that UCNPs did not show significant toxicity in mice colon cancer model. Taken together, a UCNPs-based delivery platform was successfully constructed and used for miRNA target delivery, which provided a new method and idea for bioengineering and nanotechnology-based tumor therapy.

Tumor-derived exosomal miRNAs serve as crucial biomarkers for clinical diagnosis and disease treatment monitoring. However, the need to lyse exosomes and extract miRNAs renders most methods expensive, laborious, and time-consuming. The simple, rapid, and sensitive analysis of exosomal miRNA still poses a significant challenge. Herein, we present a multifunctional biomimetic interface biosensor (MBIB) for exosome capture, miRNA release and amplified detection in one step. Catalytical hairpin assembly (CHA) probes functionalized MCF-7 cell membrane was firstly coated onto the glass substrate to fabricate the MBIB. Due to the homologous recognition ability, the MBIB could specifically capture MCF-7 cell-derived exosomes from samples and then release miRNA through membrane fusion. Finally, the released miRNA could be detected by the CHA probes on the cell membrane. Owing to the fluidity of the cell membrane, the CHA probes could collide effectively, resulting in an enhanced signal. The developed biosensor exhibited high sensitivity, reproducibility and stability. It has been successfully applied to the real samples and could accurately distinguish the tumor-bearing mice and healthy mice, showing great potential in clinical application.
