The clinical management of osteosarcoma faces critical challenges, including postoperative recurrence and metastasis of residual tumor cells, chemotherapy resistance, and impaired self-repair capacity due to extensive bone defects following tumor resection. Herein, we develop a novel functionalized 3D-printed scaffold with NIR-II responsiveness (BGS/I-LDH@MgO 2 ), designed to simultaneously address the dual needs of inhibiting osteosarcoma recurrence and promoting bone regeneration. This scaffold consists of 3D-printed bioactive glass scaffold (BGS) and MgO 2 -modified ZnAl-layered double hydroxides (ZnAl-LDHs) intercalated with 5-iodo-isophthalic acid (I-IPA). Under NIR-II irradiation, the scaffold effectively triggers a photodynamic therapy (PDT) effect to eliminate osteosarcoma cells. Notably, the incorporation of MgO 2 enables oxygen release within the tumor microenvironment, alleviating hypoxia and enhancing PDT efficacy for superior antitumor performance. Furthermore, the degradation of ZnAl-LDHs and MgO 2 releases Mg 2+ and Zn 2+ ions and generates a mildly alkaline microenvironment, which collectively facilitate the osteogenic differentiation of bone marrow mesenchymal stem cells and accelerate the process of bone healing. This functionalized 3D-printed scaffold demonstrates excellent anti-tumor and osteogenic properties, showing great promise for the treatment of osteosarcoma-associated bone defects.

The emergence of treatment approaches that integrate conventional phototherapy with additional adjuvant treatments has garnered considerable interest. In this study, we proposed a complex utilizing Fe and polydopamine as a carrier, co-loaded with the nitric oxide initiator L-arginine (L-Arg) and the photosensitizer indocyanine green (ICG), as a potential strategy for the “photothermal/photodynamic/Chemodynamic/nitric oxide gas therapy” of osteosarcoma. Nanoparticles have the ability to undergo degradation within the mildly acidic conditions present in the tumor microenvironment. Consequently, the resulting release of Fe ions facilitates the consumption of hydrogen peroxide through Fenton/Fenton-like reactions, thereby generating hydroxyl radicals (•OH) that possess potent cytotoxic properties. L-Arg can also be catalyzed by reactive oxygen species (ROS) or NO synthase overexpressed in cancer cells to generate NO, which is not only used for gas therapy (GT), but also as a biological messenger to regulate vasodilation to relieve tumor hypoxia. More importantly, the addition of low-dose near-infrared laser can not only promote the efficiency of the above two reactions, but also achieve PTT/PDT and obtain good synergistic tumor treatment effects. The anti-tumor efficacy of nanoparticles was verified in the 143B mouse osteosarcoma model. This “PTT/PDT/CDT/GT” strategy expands bone tumor treatment options through nanoparticle-mediated enhanced therapy.

A dual-drug delivery system (DDDS) based on oxidized alginate (OxAlg) and carboxymethyl chitosan (CMCS) is developed, which can be used for dual-responsive delivery of methotrexate (MTX) and naringin (Nar) for efficient treatment of osteosarcoma. Nar is first loaded onto graphene oxide (GO) through π-π stacking and hydrogen bonding, and then the Nar loaded GO (GO/Nar) is co-encapsulated with MTX by the hydrogel of OxAlg and CMCS generated through Schiff base reaction. The acylhydrazone (–N = C–) bonds between OxAlg and CMCS are prone to be hydrolyzed in an acidic medium, speeding the delivery of the two drugs; meanwhile, GO in this DDDS is an outstanding photothermal agent, leading to near-infrared light (NIR)-responsive delivery of the two drugs. The cumulative release of MTX and Nar can reach 91.09% and 85.69%, respectively, at pH 5.0 with NIR irradiation. The produced hyperthermia under NIR irradiation can result in the ablation of the osteosarcoma cells, and thus the therapeutic efficacy of the DDDS against osteosarcoma is significantly improved via the chemo-photothermal synergistic therapy mode, which can be further confirmed by the cytotoxicity test.

Photothermal therapy based on acceptor–donor–acceptor (A-D-A) type molecules is emerging as a powerful clinical treatment strategy, yet its potential application in malignant osteosarcoma has been scarcely investigated. To resolve this, we constructed novel photothermal reagents from A-D-A molecules, boosting PCE of A-D-A photosensitizers by regulating intramolecular push–pull interactions. Through an optimized push–pull architecture, the IEICO-F molecule achieved an ideal overlap between its absorption spectrum and the 808 nm NIR laser, along with a high absorption coefficient and an increased nonradiative decay rate constant. Accordingly, following 5-min 808 nm laser exposure (0.33 W cm − 2 ), the temperature of IEICO-F nanoparticles climbed to 63.8°C, exceeding IEICO nanoparticles by 9.2°C. These nanoparticles effectively generated heat to kill osteosarcoma cells and exhibited significant tumor suppression in osteosarcoma models. Importantly, our molecular design also ensures good safety profiles for practical medical application. Therefore, this novel A-D-A type molecule-based photothermal therapy strategy holds promise for widespread clinical use in osteosarcoma treatment and may be extendable to other tumor types in the future.

Photothermal therapy (PTT) has been widely used in the treatment of tumors, but its efficacy is greatly limited by the inability of precise drug delivery and the increase of heat shock proteins (HSPs) caused by high temperature. This article describes a therapeutic strategy to enhance PTT with starvation therapy in conjunction with ferroptosis mechanism. A nanoparticle platform ZIF-8@GA was constructed by wrapping together glucose oxidase (GOX) and gold nanospheres (AuNPs) loaded with dihydroartemisinin (DHA) with zeolitic imidazolate framework-8 (ZIF-8). This platform can take advantage of the micro-environment of osteosarcoma (OS) cells, featuring low pH and high reactive oxygen species (ROS), for precision drug delivery. GOX can effectively catalyze glucose to produce gluconic acid and H 2 O 2 , and DHA can also induce ROS production in OS cells. ROS produced by GOX and DHA can further generate lipid peroxidation (LPO) and lead to ferroptosis of OS cells. At the same time, ROS and LPO produced can inhibit the expression of HSPs, thereby increasing the therapeutic effect of PTT. In vitro experiments show that the nanoparticles are pH and ROS responsive. 1 µg/mL GOX combined with 0.2 µg/mL DHA promotes ferroptosis of OS cells, and increases the killing effect of near-infrared (NIR) on OS cells. Further in vivo experiments showed that the nano drug-delivery platform combined with PTT can effectively inhibit the growth of OS cells. Meanwhile, this study provides a new idea for the treatment of OS with biomaterials combined with various treatment methods.

One of the challenges in immunotherapy is the difficulty for immune checkpoint blockers to seek and destruct ‘cold’ tumors. Here we tackled this challenge using a new nanomaterial with the capability of tumor targeting, photoablation, and immune checkpoint blocking, leading to successful eradication of primary and distant metastatic tumors by photothermal immunotherapy. Specifically, the programmed death receptor 1 antibody (aPD-1) was grafted onto novel Au nanodendrites (AuNDs) to prepare a complex ( [email protected] ). The dendritic structure of [email protected] enabled sustained release of aPD-1 for blocking immune checkpoints and effective photoablation upon near-infrared irradiation for making tumors ‘hot’. Using osteosarcoma (OS) as a model ‘cold’ tumor, we found that intravenous injection of [email protected] , followed by near-infrared irradiation only on the primary tumor, destroyed primary and distant metastatic OS via activating T cells and blocking PD-1/programmed death-ligand 1 axis. The photothermal immunotherapy led to tumor eradication with a prolonged survival of mice and exhibited long-term immune memory to prevent tumor reinvasion and recurrence. [email protected] presented good tumor-targeting ability due to a combination of nanomaterials-based passive targeting and antibody-enabled active targeting and thus was excreted in the blood circulation through a special hepatointestinal pathway. Hence, the [email protected] is a promising biomaterial for treating metastatic OS through photothermal immunotherapy.

Background As a broad-spectrum antitumorigenic agent, doxorubicin (DOX) is commonly used as a chemotherapeutic drug for treating osteosarcoma (OS). Still, it is associated with significant cell toxicity and ineffective drug delivery, whereas the zeolite imidazolate framework is extensively applied in the biomedical field as a carrier owing to its favorable biocompatibility, high porosity, and pH-responsiveness. Therefore, we need to develop a drug delivery platform that can effectively increase the antitumorigenic effect of the loaded drug and concurrently minimize drug toxicity. Methods In this study, a Fe3O4@ZIF-8 nanocomposite carrier was prepared with ZIF-8 as the shell and encapsulated with Fe3O4 by loading DOX to form DOX- Fe3O4@ZIF-8 (DFZ) drug-loaded magnetic nanoparticles. Then, we characterized and analyzed the morphology, particle size, and characteristics of Fe3O4@ZIF-8 and DFZ by TEM, SEM, and Malvern. Moreover, we examined the inhibitory effects of DFZ in vitro and in vivo. Meanwhile, we established a tumor-bearing mouse model, evaluating its tumor-targeting by external magnetic field guidance. Results DFZ nanoparticles possessed have a size of ~110 nm, with an encapsulation rate of 21% and pH responsiveness. DFZ exerted a superior cytostatic effect and apoptosis rate on K7M2 cells in vitro compared to DOX(p<0.01). In animal experiments, DFZ offers up to 67% tumor inhibition and has shown a superior ability to induce apoptosis than DOX alone in TUNEL results(p<0.01). Tumor-targeting experiments have validated that DFZ can be effectively accumulated in the tumor tissue and enhance anticancer performance. Conclusion In summary, the DFZ nano-delivery system exhibited a more substantial anti-tumorigenic effect as well as superior active tumor targeting of DOX- Fe3O4@ZIF-8 compared to that of DOX alone in terms of biocompatibility, drug loading capacity, pH-responsiveness, tumor-targeting, and anti-tumorigenic effect, indicating its chemotherapeutic application potential.

A pH, redox and near infrared (NIR) irradiation tri-responsive dual-drug delivery system (DDDS) is constructed for the treatment of osteosarcoma. Methotrexate (MTX) is encapsulated into the mesoporous silica nanoparticles (MSNs) by polydopamine (PDA), and then the core–shell structured MTX/MSNs@PDA is embedded into the graphene oxide (GO) nanosheets to further enhance the photothermal conversion capability of this system. The resultant MTX/MSNs@PDA@GO is co-encapsulated with naringin (Nar) into the hydrogels of carboxymethyl cellulose (CMC) and cystamine (Cys) generated through amidation reaction. The disulfide linkage (–S–S–) in Cys can be reduced to sulphydryl groups (–SH) by glutathione (GSH), resulting in the degradation of the hydrogels; on the other hand, both PDA and the amide linkage between CMC and Cys are pH-sensitive. Therefore, the constructed DDDS can be used for pH-, redox- and NIR irradiation-responsive delivery of MTX and Nar. Finally, the validity of the developed DDDS is evaluated by cytotoxicity test.

Osteosarcoma is a common malignant bone tumor that tends to occur in adolescents, with surgical resection of the tumor tissue as its standard treatment. After surgery, long-term chemotherapy should be performed to prevent patients from recrudescence. However, conventional chemotherapy drugs impose many serious side effects on patients. It is significant to optimize a chemotherapy drug delivery system. Herein, we present an injectable self-healing hydrogel carrying curcumin with pH-responsive drug release and selective toxicity for osteosarcoma therapy. Amino-gelatin and oxidized starch were synthesized and used as substrates to prepare hydrogels based on imine linkages. Polymerized β-CD was synthesized to encapsulate curcumin and incorporate it into hydrogels. As a dynamic covalent bond, imine linkages endowed the hydrogel with injectability and self-healing properties. Under acidic conditions, the hydrogel presented a pH-responsive curcumin release property due to instable imine linkages, showing a higher cumulative release of curcumin at pH 6.5 than at pH 7.4. Moreover, hydrogels could continuously release curcumin for more than 28 days, featuring a sudden early release and a slow late release. Interestingly, differentiated from healthy osteoblasts, the hydrogel showed selective cytotoxicity to osteosarcoma cells and could even accelerate the differentiation of osteoblasts owing to the incorporation of curcumin. Briefly speaking, as a drug delivery system, the composite hydrogel can be widely applied for osteosarcoma treatment.

Osteosarcoma is a prevalent primary malignant bone tumor in young adults and adolescents, characterized by a high recurrence rate despite advancements in chemotherapy and surgical methods. This study investigated the effects of integrating high-dose methotrexate with cisplatin and paclitaxel on survival outcomes in osteosarcoma patients, and to identify prognostic factors influencing these outcomes. A retrospective analysis was conducted on 208 osteosarcoma patients treated between January 2013 and December 2018. Patients were divided into two groups: standard chemotherapy group (SC, n = 104) and cisplatin + paclitaxel + high-dose methotrexate (CPM, n = 104). The primary endpoints were progression-free survival (PFS) and overall survival (OS), while secondary endpoints included efficacy assessments. Kaplan-Meier survival curves were used to assess survival distributions, and statistical analyses were performed using SPSS 29.0. The CPM group demonstrated significantly longer PFS (16.85 ± 3.40 months vs. 15.72 ± 3.21 months, P = 0.015) and higher 5-year OS rates (54.81% vs. 40.38%, P = 0.037) compared to the SC group. Completion of chemotherapy and a response rate greater than 90% were identified as strong positive prognostic indicators. In contrast, pathologic fractures at diagnosis, lung metastases, and elevated lactate dehydrogenase levels were associated with poorer outcomes. Multivariate analysis underscored chemotherapy response and treatment adherence as independent survival predictors. The combination of cisplatin and paclitaxel with high-dose methotrexate significantly improves PFS and OS compared to standard chemotherapy. Moreover, treatment completion and achieving a chemotherapy response greater than 90% are critical factors for favorable prognosis.

An ultrathin micro-patterned MXene/PEDOT:PSS-based organic electrochemical transistor biosensor was constructed, which can significantly amplify the amperometric signal and transistor’s performance. A novel interdigitated OECTs biosensor has been developed for reliable determination of survivin for the following considerations: (1) The synergistic effect of intercalated MXene and ionic PEDOT:PSS enhanced the mobility and volumetric capacitance of OECTs biosensor. (2) Compared with the best previous literatures, our assay demonstrated enhanced detection limit of survivin down to 10 pg mL −1 , as well as satisfactory selectivity, reproducibility, and reliability. (3) Comparison of OECTs against commercial ELISA kit yielded favorable linearity ( Y = 1.0015* X + 0.0039) and correlation coefficient ( R 2 = 0.9717). Those advantages are expected to pave the way to design of an OECTs biosensor with robustness, non-invasiveness, and miniaturization for the point-of-care applications. Graphical abstract

The therapy of tumor recurrence resulting from surgical resection of osteosarcoma is still a significant challenge for clinicians. In this study, we designed curcumin (CM) and indocyanine green (ICG) co-loaded poly(L-lactic acid) (PLLA) (PLLA-CM-ICG) composite microparticles , combining photothermal therapy with chemotherapy to enhance osteosarcoma therapy synergistically. PLLA-CM-ICG microparticles of sizes of (838.3 ± 5.4) nm were prepared by solution-enhanced dispersion by a supercritical CO 2 process. ICG in the PLLA-CM-ICG microparticles was more stable than free ICG. The PLLA-CM-ICG microparticles displayed pH- and near-infrared (NIR)-responsive CM release behavior. Moreover, NIR irradiation improved the anticancer activity of the PLLA-CM-ICG microparticles at a concentration of 250 μg/mL on MG-63 cells. Therefore, PLLA-CM-ICG microparticles could be employed as promising drug delivery systems for synergic photothermo-chemotherapy in osteosarcoma treatment.
