Cabazitaxel Sustained-Release Nanoparticles Treat Spinal Cord Injury through Inhibiting Fibrin Deposition and Improving the Microenvironment

Yuanli Yang,Chenqian Feng,Xiaoling Li,Min Mu,Hui Li,Bo Chen,Rangrang Fan,Haifeng Chen,Gang Guo

Biomaterial-based modulation of the microenvironment represents a promising neuroprotective strategy for spinal cord injury (SCI). We first fabricated hyaluronic acid (HA)-graft-epigallocatechin gallate (EGCG) nanoparticles (HEN). These nanoparticles can synergistically exert anti-inflammatory and antioxidant effects, while HA can modulate the immune microenvironment. To further enhance therapeutic efficacy, Cabazitaxel (Cab) was incorporated into HEN to generate multifunctional Cabazitaxel-loaded HA-EGCG nanoparticles (Cab-HEN), which inhibited scar formation and modulated microtubule homeostasis, thereby promoting the beneficial regeneration of nerve axons in a rat SCI model. Through in situ injection at the T9 injury site, Cab-HEN significantly downregulated the expression of inflammatory factors, effectively mitigated oxidative damage induced by excessive reactive oxygen species (ROS), and reduced fibrin deposition and scar formation. Furthermore, Cab-HEN regulated microtubules and enhanced the regeneration of nerve fibers. Evaluations based on electrophysiological assessments, Basso, Beattie, and Bresnahan (BBB) scores, and bladder function recovery revealed the nanoparticles’ remarkable therapeutic efficacy in SCI. Therefore, biomaterials can facilitate axonal regeneration, tissue remodeling, and functional restoration following injury.

Phytosterol nanoparticle delivery system based on whey protein-chitosan: enhanced stability, bioavailability, and sustained-release properties

Yang Tingting,Tao Guanhua,Xu Xinye,Yu Chunyu,Shi Jindou,Ma Qingwen

Phytosterol (PS) face challenges of poor stability and rapid release in the gastrointestinal tract. To overcome these limitations, this study developed whey protein-chitosan composite (WP-CS-PS) nanoparticles via reverse solvent precipitation, significantly enhancing PS transport efficiency and functional performance. Through systematic optimization using single-factor and orthogonal tests, the nanoparticles were fabricated under ideal parameters. The optimal conditions included a 1:1 WP/CS mass ratio, 0.8% (w/v) PS concentration, 60 °C hydration, and 15 min stirring, which yielded high encapsulation efficiency (77.43%), uniform particle size (203.41 nm, PDI 0.265), and excellent colloidal stability (zeta potential − 35.42 mV). Structural analyses (SEM, FT-IR) revealed a dense gel-network morphology with PS successfully embedded via hydrogen bonding and electrostatic interactions. The WP-CS matrix markedly improved PS stability against thermal, UV, and storage stresses, while in vitro digestion assays demonstrated sustained intestinal release (17.72% slower than free PS). These results not only validate WP-CS-PS as a robust carrier for lipophilic bioactives but also offer a scalable strategy to enhance the bioavailability of PS in functional foods, potentially addressing global health needs such as cholesterol management and dietary supplementation. By bridging material science and nutritional delivery, this work paves the way for sustainable nutraceutical innovations with broad industrial applicability. Graphical

Time-Programmable Fluorescence in Perovskite Quantum Dots via Reverse Sustained-Release Strategy for Dynamic Information Encryption

Zi-Rong Zhou,Hong-Fei Li,Zi-Hao Liao,Zhe-Xun Zhu,Feng Wang

Smart luminescent materials capable of time-resolved dynamic fluorescence color transitions hold immense potential for advanced information encryption and anti-counterfeiting technologies in the emerging digital era. However, achieving precise control over wide-range fluorescence color modulation remains a significant challenge. In this study, a novel reverse sustained-release strategy is presented to fabricate CsPbBr 3 @PAA perovskite quantum dots (QDs), where ultra-small CsPbBr 3 QDs within quantum confinement size (QCS) are confined within a polyacrylic acid (PAA) matrix. Initially, the CsPbBr 3 @PAA QDs exhibit blue-violet fluorescence in toluene due to the quantum confinement effect. Upon exposure to hydrous alcohol, the PAA matrix enables a controlled, reverse release of CsPbBr 3 QDs into the solvent, triggering a morphological transition from sub-nanometer QDs to nanoblocks exceeding the QCS. This structural evolution induces a dynamic time-dependent fluorescence shift from blue-violet to green, spanning five visually distinct colors (blue-violet, blue, cyan, turquoise, and fluorescent green) across the CIE chromaticity diagram. By leveraging this unique time-resolved color transition mechanism, programmable information encryption and dynamic fluorescent patterning with temporal precision are demonstrated. This work not only advances the design of stimuli-responsive luminescent materials but also provides a versatile platform for next-generation optical security systems

Emulsion Gel Packaging System with Sustained Release and Synergistic Antibacterial for Prolonging Fruit and Vegetable Preservation

Xue Zhang,Pengrui Zhao,Jialin Jia,Junkang Yang,Huajie Liu,Tao He,Tao Zhang,Wenna Wu

Menthol and thymol, as natural antiseptics, have the advantages of nontoxicity and broad-spectrum antimicrobial activity, which solves the problems of antibiotic resistance and toxicity of metal antiseptics. However, their practical applications face the challenges of chemical instability and difficult solubility in water. In this study, menthol and thymol were tactfully physically combined, forming hydrophobic deep eutectic solvent (HDES), which were then encapsulated via an emulsion system to address their instability and hydrophobicity. Then, 2-hydroxyethyl acrylate was added and polymerized into a covalent network to fabricate a HDES-loaded heterogeneous gel, featuring sustained-release characteristics for the preservation of perishable fruits. The combination of emulsion encapsulation and gelation improves the stabilization of menthol and thymol, allowing for delayed release and providing materials with a 34-day antibacterial cycle. Additionally, the emulsion gel presents tensile strain as high as 2250%, making it appropriate as practical packaging materials. Through cherry, tomato, and broccoli preservation tests, it is found that compared with the monomer such as thymol, menthol, and the deep eutectic mixture, the emulsion gel presented the best antibacterial activity, enabling the preservation days to significantly increase. We calculated that it was probably due to the HDES and cross-linking network displaying a synergistic effect. This design will provide a perspective for the preservation of perishable fruits and vegetables.

Development and evaluation of novel zein-based artemisinin sustained-release formulation for treating drug-resistant malaria

Yijie Wang,Xinyu Yu,Xinyu Zhang,Xiaohui He,Yongxin Tang,Ling Fang,Richard Culleton,Qingfeng Zhang,Weifu Dong,Jun Cao

Artemisinin antimalarial drugs initially exhibited remarkable efficacy against Plasmodium falciparum. However, their poor solubility and low bioavailability necessitate high doses and lead to an extremely short in vivo half-life. These limitations not only drive the emergence of drug-resistant Plasmodium strains but also compromise long-term therapeutic outcomes. Herein, we report a zein-based sustained release formulation, wherein zein, a natural maize protein, serves as a biocompatible nanocarrier to effectively encapsulate artemisinin (ART). Notably, this nanocarrier formulation achieves a 200-fold enhancement in ART’s water solubility, addressing a key bottleneck of ART-based therapies. In vitro assays confirm that the zein-based formulation allows for the sustained release of ART, which could help maintain therapeutic concentrations over extended periods and displayed different release rates and good dispersibility in both acid and basic environments. Importantly, in vitro evaluations also demonstrate that the nanoformulation exerts potent inhibitory effects against ART-resistant P. falciparum strains in both ring survival assay and recrudescence assay, attributed to the sustained maintenance of effective ART concentrations. In vivo studies, utilizing both rodent malaria models and humanized erythrocyte mouse models, further validate the nanoformulation’s therapeutic potential. The zein nanocarrier significantly prolongs ART’s in vivo half-life via its sustained-release capability, thereby maintaining effective blood concentrations over an extended duration. Compared to free ART, the nanoformulation exhibits superior efficacy in reducing parasitemia, preventing malaria recrudescence, and, most notably, overcoming ART resistance in drug-resistant Plasmodium infections. Collectively, these findings establish the zein-based nanocarrier as a promising strategy to optimize ART-based therapies by addressing solubility and pharmacokinetic limitations while effectively combating drug-resistant malaria.

Furui He,Qingfeng Zhou,Longzheng Wang,Gaobo Yu,Jiacheng Li,Yuhong Feng

In pest control field, the sustained release of pesticides plays significant roles in effectiveness and environment friendly. In this study, the sodium alginate (SA), polyacrylamide (PAM) and montmorillonite (MMT) were utilized to fabricate the stretchable double-network nanocomposite hydrogel, which apply in a sustained release system of acetamiprid . These nanocomposite hydrogels were fully characterized by X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), thermal gravimetric analysis (TGA), transmission electron microscopy (TEM), and scanning electron microscopy (SEM). The results indicate that adding MMT could significantly improve the sustained release performances of the hydrogels. The swelling behavior of all the hydrogels well fits into the Fickian diffusion model. The hydrogel with 5.0% MMT content shows the maximum pesticides loading rate of 13.32% and minimum pesticides release rate of 76.11%. The nanocomposite hydrogels take advantages of both the gelation property of alginate and the adsorption capacity of clays, has great potential in the application of pesticides carrier to improve the utilization efficiency of water-insoluble pesticides.

Preparation and in vitro release kinetics of ivermectin sustained-release bolus optimized by response surface methodology

Xiangchun Ruan,Xiuge Gao,Ying Gao,Lin Peng,Hui Ji,Dawei Guo,Shanxiang Jiang

Sustained-release formulations of ivermectin (IVM) are useful for controlling parasitic diseases in animals. In this work, an IVM bolus made from microcrystalline cellulose (MCC), starch and low-substituted hydroxypropyl cellulose (LS-HPC) was optimized by response surface methodology. The bolus was dissolved in a cup containing 900 mL of dissolution medium at 39.5 °C, under with stirring at 100 rpm. A quadratic model was formulated using analysis of variance according to the dissolution time. The optimized formulation of the bolus contained 8% MCC, 0.5% starch, and 0.25% LS-HPC. The length, width, and height of the prepared IVM bolus were 28.12 ± 0.14, 16.1 ± 0.13, and 13.03 ± 0.05 mm, respectively. The bolus weighed 11.4842 ± 0.1675 g (with a density of 1.95 g/cm 3 ) and contained 458.26 ± 6.68 mg of IVM. It exhibited in vitro sustained-release for over 60 days, with a cumulative amount and percentage of released IVM of 423.72 ± 5.48 mg and 92.52 ± 1.20%, respectively. The Korsmeyer–Peppas model provided the best fit to the dissolution release kinetics, exhibiting an R 2 value close to 1 and the lowest Akaike Information Criterion among different models. The parameter n (0.5180) of the Korsmeyer–Peppas model was between 0.45 and 0.89. It was demonstrated that the release mechanism of the IVM bolus followed a diffusive erosion style.

Modified chitosan-based hydrogel for sustained release of tetramethylpyrazine and polydatin to promote diabetic wound healing

Changle Lu,Bailei Li,Jiali Liu,Xinyu Gao,Zhen Zhang,Yu Zhou,Qilong Cao,Xiaojun Liu,Rongqing Zhang

Diabetic wound healing remains a clinical challenge due to impaired repair, often leading to complications such as infection and tissue necrosis. Here, we present a novel dual-drug-loaded hydrogel formulated using oxidized hyaluronic acid (OHA) and catechol-modified chitosan (CS-pC). This system is engineered to enhance the local bioavailability of polydatin (PD) and delay the release of tetramethylpyrazine (TMP), achieving superior release performance and synergistic effects compared to single-drug formulations; thus, this system enables synergistic modulation of the target inflammatory microenvironment and promotes angiogenesis, thereby facilitating effective diabetic wound healing. The hydrogel exhibited a porous structure (100–200 μm) and moderate swelling (119.67% ± 6.66% at 24 h). Approximately 80% of TMP and PD were sustainably released within 48–52 h, aligning with clinical dressing intervals. In diabetic mice, it accelerated wound closure to 98.01% within 14 d—20.63% higher than controls. Most notably, the hydrogel significantly promoted wound healing, as evidenced by: (1) upregulation of vascular endothelial growth factor (VEGF) and α-SMA expression (RT-qPCR: 3.2-fold for VEGF and 3.7-fold for α-SMA vs control); (2) enhanced cell migration (3.1-fold vs control) and angiogenesis (1.3-fold vs control); and (3) increased expression of CD31, TGF-β1 and collagen types I and III (IHC). These results demonstrate the therapeutic potential of the OHA/CS-pC/TMP/PD hydrogel for diabetic wound healing.

Microencapsulation of cholic acid via complex coacervation enhances sustained release and antibacterial activity against Escherichia coli

Xiuling He,Ganggang Qu,Danqi Shen,Jing Lu,Haobo Li,Wei Mao,Zhipeng Jia

This study aimed to develop a microencapsulated cholic acid (CA) preparation to improve the stability, sustained-release performance, and antibacterial activity of CA for potential use in antibiotic-free animal production. CA microcapsules were prepared by complex coacervation using gelatin and gum arabic as wall materials, and the formulation was optimized by single-factor and orthogonal experimental designs. The optimal preparation conditions were a wall material concentration of 2.5%, a core-to-wall ratio of 1.8:1, and pH 4.1, yielding a drug loading rate of 49.73% and an encapsulation efficiency of 88.76%. The microcapsules showed spherical morphology, low hygroscopicity, improved thermal and storage stability, and a sustained-release profile over 72 h in simulated gastrointestinal media. Both free CA and microencapsulated CA significantly inhibited Escherichia coli growth (p < 0.001), while the microcapsules provided a more prolonged antibacterial effect consistent with controlled release. These findings indicate that complex coacervation is a practical strategy for improving CA performance and support the potential use of CA microcapsules as antimicrobial feed additives in livestock production.

Preparation and performance of pH-responsive 5-Fu/chitosan sustained-release microspheres based on microfluidic technology

Huidong Fang,Shiai Xu,Siyu Han,Bin Zhao,Chao Zhang,Wanji Zhou

T-shaped droplet microfluidic device was used for preparing 5-Fu-loaded chitosan microspheres. • 5-Fu-loaded chitosan microspheres exhibit pH-responsive release patterns and sustainable release characteristics for up to 60 days. • The average particle size of the microspheres is 3.90 ± 0.57 μm, and the particle size distribution is narrow. • Drug-loaded microspheres of different particle sizes have different drug release rates and cycles. Microsphere formulations are a promising drug delivery system for targeted response and prolonged sustained release. However, conventional emulsification crosslinking methods for microsphere preparation usually result in a broad particle size distribution, large particle size, poor uniformity, and consequently erratic drug release kinetics. In this investigation, chitosan microspheres encapsulating 5-Fluorouracil (5-Fu CS-MS) were fabricated using a T-type droplet microfluidic device, and the formulation was optimized by single-factor experiments. The resulting microspheres had a consistent drug release profile, a smooth surface, high monodispersity, and a uniform particle size distribution with an average particle size of 3.90 ± 0.57 μm. The polymer-drug interaction in 5-Fu CS-MS was analyzed using FTIR and XRD. The XPS and EDS results confirmed the occurrence of a Schiff base reaction between glutaraldehyde (GA) and chitosan (CS), indicating successful encapsulation. TGA results revealed that the cross-linking reaction enhanced the thermal stability of the microspheres. Biocompatibility assessments using Human umbilical vein endothelial cells (HUVEC) line demonstrated the safety and compatibility of the fabricated microspheres. More importantly, the in vitro release results illustrated a pH-responsive drug release profile and sustained release of 5-Fu CS-MS for a period up to 60 days. In summary, the CS sustained-release microspheres produced using the microfluidic technique have the potential for intelligent response, precise drug delivery, and prolonged sustained release of small-molecule drugs. Chitosan–Citral Imine Hydrogels With Self-Ordered Nanostructures as Sustained-Release and Eco-Friendly Agricultural Fungicide

Linfeng Hu,Yunlong Wang,Hongying Niu,Hongxia Zhu

This study developed chitosan-based hydrogels as field-deployable carriers for sustained fungicide delivery to enhance crop disease resistance and yield. Citral and chitosan were reacted at three molar ratios (N1, N1.5, N2) via acid-catalyzed Schiff-base condensation, and the resulting hydrogels were characterized by FTIR, 1 H NMR, SEM and WXRD. Structural analyses confirmed the formation of self-ordered imine clusters with a d -spacing of 20.5 Å stabilized by non-covalent interactions. N1, the highest-cross-linked hydrogel, showed the best thermal stability ( T 10 = 160°C) and the lowest swelling, releasing 7.0% slower than N1.5 and 37.9% slower than N2. Antifungal assays demonstrated that N1 provided the strongest inhibition against five phytopathogens ( Rhizoctonia solani , Fusarium graminearum , Botrytis cinerea , Rhizoctonia cerealis , and Phytophthora capsici ), improving inhibition of B. cinerea , P. capsici , and F. graminearum by 56.8%, 82.9%, and 181.3%, respectively, compared with free citral. These hydrogels combine antifungal activity, water retention, thermal stability and controlled release, providing a promising strategy for green fungicide delivery in agriculture.

Improved Mechanical Properties and Sustained Release Behavior of Cationic Cellulose Nanocrystals Reinforeced Cationic Cellulose Injectable Hydrogels

Jun You,Jinfeng Cao,Yanteng Zhao,Lina Zhang,Jinping Zhou,Yun Chen

Polysaccharide-based injectable hydrogels have several advantages in the context of biomedical use. However, the main obstruction associated with the utilization of these hydrogels in clinical application is their poor mechanical properties. Herein, we describe in situ gelling of nanocomposite hydrogels based on quaternized cellulose (QC) and rigid rod-like cationic cellulose nanocrystals (CCNCs), which can overcome this challenge. In all cases, gelation immediately occurred with an increase of temperature, and the CCNCs were evenly distributed throughout the hydrogels. The nanocomposite hydrogels exhibited increasing orders-of-magnitude in the mechanical strength, high extension in degradation and the sustained release time, because of the strong interaction between CCNCs and QC chains mediated by the cross-linking agent (β-glycerophosphate, β-GP). The results of the in vitro toxicity and in vivo biocompatibility tests revealed that the hydrogels did not show obvious cytotoxicity and inflammatory reaction to cells and tissue. Moreover, DOX-encapsulated hydrogels were injected beside the tumors of mice bearing liver cancer xenografts to assess the potential utility as localized and sustained drug delivery depot systems for anticancer therapy. The results suggested that the QC/CCNC/β-GP nanocomposite hydrogels had great potential for application in subcutaneous and sustained delivery of anticancer drug to increase therapeutic efficacy and improve patient compliance.
