l-tryptophan (l-Trp) plays an important role in human and animal metabolism, and an imbalance or deficiency can cause some chronic diseases. While d-tryptophan (d-Trp) is mainly present in microorganisms and green plants and is rarely found in animals. In this work, a simple and efficient electrochemical sensing platform for the simultaneous detection of tryptophan enantiomers based on an l-lysine-functionalized nickel–zinc bis(dithiolene) metal–organic framework and ferrocene-modified multiwalled carbon nanotubes (Fc-MWCNTs) is presented. The identification effect of l-Lys-Ni Zn-MOF/Fc-MWCNTs/GCE on tryptophan (Trp) enantiomers was studied by differential pulse voltammetry. The synthesized materials have been successfully applied to the detection and analysis of Trp enantiomers, and the enantiomer selection coefficient (IL/ID) detected by the sensor is 1.866, indicating that l-Lys-Ni Zn-MOF/Fc-MWCNT composites have good application potential.

Efficient discrimination of amino acids (AAs) isomers is of significant importance for life science and analytical chemistry. Here, a dual-mode chiral discrimination strategy is proposed for visual and electrochemical chiral discrimination of tryptophan (Trp) isomers. Shikimic acid chiral ionic liquids (SCIL) is coordinated with copper ions (Cu 2+ ), and the obtained SCIL-Cu 2+ can form ternary complexes with the Trp isomers. Owing to the inherent chirality of SCIL and the reverse homochirality of L-Trp and D-Trp, the ternary complex of SCIL-Cu-D-Trp has higher stability than SCIL-Cu-L-Trp, as revealed by the calculated stability constants ( K ) and changes in Gibbs free energy (Δ G ). The difference in the stability can be utilized for the chiral discrimination of L-Trp and D-Trp, resulting in discernible differences in colors and the electrochemical signals of the Trp isomers. Besides Trp, the isomers of phenylalanine (Phe) can also be discriminated by the proposed dual-mode chiral discrimination strategy with the SCIL-Cu 2+ complex.

Hollow polymer microspheres (HPMs) were synthesized, which were then hydrolyzed in aqueous ammonia to produce carboxyl (–COOH) groups on their surface. L-phenylalanine (L-Phe) was grafted to the hydrolyzed HPMs (H-HPMs) through amidation reactions, endowing the H-HPMs with chirality. The resultant chiral HPMs (C-HPMs) were used for the chiral discrimination of tryptophan (Trp) isomers. Due to the same rotatory direction of L-Phe and L-Trp, the C-HPMs showed greatly higher selectivity toward L-Trp than its isomer. After being adsorbed by the C-HPMs, the absorbance of the residual L-Trp is significantly lower than that of the residual D-Trp, and thus spectroscopic chiral discrimination of the Trp isomers was successfully achieved. The Trp isomers were also discriminated by the chiral solid polymer microspheres (C-SPMs), while the difference in the absorbance of the residual L-Trp and D-Trp is remarkably smaller than that obtained by the C-HPMs. The outstanding discrimination capability of the C-HPMs might be ascribed to their high surface permeability resulted from their unique hollow structure.

A highly sensitive surface-enhanced Raman spectroscopy (SERS) approach was established for the detection of monosultap using the nanosubstrate of β -CD/Trp/MgO/AgNPs which was MgO-protected silver nanoparticles (AgNPs) assembled tryptophan (Trp) and β -cyclodextrin ( β -CD). Six nanosubstrates including AgNPs, MgO/AgNPs, Trp/AgNPs, β -CD/MgO/AgNPs, Trp/MgO/AgNPs and β -CD/Trp/MgO/AgNPs were compared for their enhancement effect on monosultap. Among them, β -CD/Trp/MgO/AgNPs had the greatest enhancement effect, with an enhancement factor (EF) reaching up to 1.95 × 10 6 . The nanosubstrate of β -CD/Trp/MgO/AgNPs was characterized by various instrumental techniques, such as transmission electron microscopy (TEM), high-angle annular dark-field scanning transmission electron microscopy (HAADF-STEM), zeta potential, X-ray diffraction (XRD), thermogravimetric analysis (TGA), ultraviolet-visible (UV–vis) and Fourier transform infrared (FT-IR). The binding of β -CD/Trp/MgO/AgNPs with monosultap was studied in detail. The optimum experimental conditions were confirmed through single-factor experiments and response surface analysis. The interferences from the coexisting substances (common metal ions, glucose and congeneric drugs) were investigated to evaluate the method selectivity. Under the optimal conditions, the standard curve equation of monosultap was established within the concentration range of 2.00 × 10 −8 –1.00 × 10 −6 mol L −1 , with detection limit (LOD) of 0.970 nmol L −1 (3 σ / S ). The SERS approach was utilized for detecting monosultap in several vegetables and fruits with the recoveries from 96.3 % to 105 %.

A chiral sensor is described for the enantioselective recognition of L- and D-tryptophan (Trp). The sensor is based on the use of (a) Cu(II) ions coordinated with β-cyclodextrin (Cu-β-CD) that was self-assembled with carboxymethyl cellulose (CD-Cu-CMC) as a chiral selector, (b) of N-doped reduced graphene oxide (N-rGO) as substrate materials, and (c) of differential pulse voltammetry that was used for enantiorecognition. The 3D N-rGO was prepared by using reduced graphene oxide and pyrrole as the starting materials. Electrostatic interaction occurs between the carboxy groups of CMC and Cu(II) ions in Cu-β-CD. The FT-IR, SEM, XRD and XPS techniques showed that 3D N-rGO and the CD-Cu-CMC composite were successfully synthesized. The 3D N-rGO enabled the immobilization of the chiral selector (CD-Cu-CMC) and improves the active areas. A glassy carbon electrode was modified with N-rGO/CD-Cu-CMC and then showed a stronger electrochemical signal for L-Trp than for D-Trp, typically at a working potential of around 0.78 V (vs. SCE). UV-vis spectroscopy proved that CD-Cu-CMC has a higher affinity for D-Trp. The enantioselectivity for D-Trp over L-Trp is 4.72. The modified electrode had a limit of detection of 0.063 μM and 0.0035 μM for L-Trp and D-Trp, respectively, with a linear response range of 0.01 mM to 5 mM. The sensor was used to detect Trp (D- or L-Trp) in spiked real human urine and human serum protein samples.
Schematic representation of an electrochemical chirality sensor based on the merits of N-rGO and CD-Cu-CMC. It involves the preparation of a chiral selector and a substrate material. N-Doped graphene oxide (N-rGO) was prepared by using graphene oxide (GO) and pyrrole as precursor. The combination of carboxymethyl cellulose (CMC) with Cu-β-CD leads to a novel self-assembly framework.

Mitoxantrone (MX) can induce the immunogenic-cell death (ICD) of tumor cells and activate anti-tumor immune responses. However, it can also cause high expression of indole amine 2, 3-dioxygenase (IDO) during ICD, leading to T-cell apoptosis and a weakened immune response. An IDO inhibitor, 1-methyl tryptophan (1-MT), can inhibit the activity of IDO caused by MX, resulting in enhanced chemo-immunotherapy. Here, MX-1-MT was connected by ester bond which could be broken in an acidic tumor microenvironment. MX-1-MT was combined with polyethylene glycol (PEG) via a disulfide bond which could be reduced by glutathione overexpressed in tumors, thereby accelerating drug release at target sites. Folic acid-modified distearoyl phosphoethanolamine-polyethylene glycol (DSPE-PEG-FA) was introduced to form targeting micelles. The micelles were of uniform particle size, high stability, and high responsiveness. They could be taken-up by drug-resistant MCF-7/ADR cells, displayed high targeting ability, and induced enhanced cytotoxicity and ICD. Due to 1-MT addition, micelles could inhibit IDO. In vivo studies demonstrated that micelles could accumulate in the tumor tissues of nude mice, resulting in an enhanced antitumor effect and few side-effects.

An electrochemical chiral multilayer nanocomposite was prepared by modifying a glassy carbon electrode (GCE) via opposite-charge adsorption of amino-modified β-cyclodextrin (NH 2 -β-CD), gold-platinum core-shell microspheres (Au@Pts), polyethyleneimine (PEI), and multi-walled carbon nanotubes (MWCNTs). The modified GCE was applied to the enantioselective voltammetric determination of tryprophan (Trp). The Au@Pts enable an effective immobilization of the chiral selector (NH 2 -β-CD) and enhance the electrochemical performance. Scanning electron microscopy, transmission electron microscopy, UV-vis spectroscopy, FTIR and electrochemical methods were used to characterize the nanocomposite. Trp enantiomers were then determined by differential pulse voltammetry (DPV) (with a peak potential of +0.7 V vs. Ag/AgCl). The recognition efficiency was expressed by an increase in peak height by about 32% for DPV determinations of L-Trp compared to D-Trp in case of a 5 mM Trp solution of pH 7.0. Response was linear in the 10 μM to 5.0 mM concentration range, and the limits of detection were 4.3 μM and 5.6 μM with electrochemical sensitivity of 43.5 μA·μM −1 ·cm −2 and 34.6 μA·μM −1 ·cm −2 for L-Trp and D-Trp, respectively (at S/ N = 3).

Background: Indocyanine2,3-dioxygenase (IDO) is an enzyme that can catalyze the metabolism of tryptophan (Trp) into kynurenine (Kyn), thus inhibiting the tumor immune microenvironment. Method: Based on its inhibitor, NLG919 (NLG), the authors developed a new immunomodulatory polymer micelle and established and verified an ultrahigh performance liquid chromatography–mass spectrometry method for the simultaneous determination of NLG, Trp and Kyn in mouse tumors through the ratio determination of Trp/Kyn tissue distribution and pharmacokinetics. The linear range of the method was 0.001–10 μg/ml. Results: Compared with NLG solution, the immunomodulatory polymeric drug-loaded micelles based on polystyrene-arginine showed higher Trp/Kyn ratio, more tumor aggregation and good pharmacokinetics. Conclusion: This method has been successfully applied to the simultaneous determination of Trp/Kyn and NLG in tumor tissues of mice.

A pair of stereoisomers of nickel Schiff base complexes, namely [Ni(C 17 H 15 N 4 O 2 ) 2 ·2CH 3 OH] n ( 1 ) (C 17 H 15 N 4 O 2 = 2-acetylpyrazine- l -tryptophan) and [Ni(C 17 H 15 N 4 O 2 ) 2 ·2CH 3 OH] n ( 2 ) (C 17 H 15 N 4 O 2 = 2-acetylpyrazine- d -tryptophan), were synthesized and characterized by physico-chemical and spectroscopic methods. The X-ray diffraction single-crystal analyses indicate that the structure of 2 is essentially 0D monomeric, while a 2D layer structure is formed through N–H…O intermolecular hydrogen bonds for 1 . The interactions between complex 1 and calf thymus DNA were investigated by spectroscopic and viscometric methods. The results indicate that complex 1 interacts with DNA very strongly ( K
b = 1.01 × 10 7 mol −1 L and K
sq = 1.11). The nature of the binding seems to be mainly an electrostatic interaction between DNA and the complex. However, other binding modes, such as hydrogen bonding, may also be present in this system.

An electrochemical chiral sensor based on reduced graphene oxide (RGO) non-covalently functionalized with 1,10-phenanthroline copper(II) (PhenCu) complex has been developed for electrochemical discrimination of tryptophan (Trp) enantiomers. The formation and morphology of reduced graphene oxide/1,10-phenanthroline copper(II) (RGO/PhenCu) composites were confirmed by Fourier-transform infrared spectroscopy (FT-IR) and scanning electron microscopy (SEM). Cyclic voltammetry (CV) was employed to monitor the electrochemical behavior of RGO/PhenCu immobilized on glassy carbon electrode (RGO/PhenCu/GCE). The reduction in peak current was significantly different when the chiral sensor interacted with L-Trp and D-Trp. This suggested that RGO/PhenCu/GCE could be used as an electrochemical chiral sensor for the discrimination of Trp enantiomers. Further studies showed that the peak current decreased linearly along with an increasing percentage of L-Trp in the Trp mixture. The RGO/PhenCu/GCE electrochemical chiral sensor, with rapid recognition, good sensitivity and high stability, provided an efficient method to recognize and discriminate Trp enantiomers.

For the first time, a familiar metal–organic framework MIL-101(Fe) and silver nanoparticles composite (AgNPs/MIL-101) acted as a novel electrode modified material for the detection of tryptophan (Trp). The AgNPs/MIL-101 modified glassy carbon electrode (AgNPs/MIL-101/GCE) produces an increase in the oxidation current of Trp compared with the bare electrode. Additionally, the electrochemical behaviours of Trp on AgNPs/MIL-101/GCE were investigated by cyclic voltammetry (CV) and differential pulse voltammetry (DPV). Under the optimum experimental conditions, the oxidation peak currents are proportional to the concentrations of tryptophan over the ranges of 1 μM to 50 μM and 50 μM to 150 μM, respectively. The detection limit is 0.14 μM (S/N = 3). Moreover, the presented method was successfully applied to the determination of Trp in urine samples with good recovery.

A luminescent anthracene-based Zn-MOF with a robust 3D framework was successfully constructed. • The Zn-MOF exhibits highly selective and sensitive dual fluorescence sensing toward Fe³⁺ and L-tryptophan in aqueous media. • The sensor achieves low detection limits of 1.51 μM (Fe 3+ ) and 2.08 μM (L-Trp), with fluorescence quenching dominated by the inner filter effect. • A blue-emitting LED device based on Zn-MOF was fabricated, demonstrating its optoelectronic application potential. In this study, a new three-dimensional Zn-MOF was designed and synthesized. The complex [Zn 2 (DCPA) 2 (tib)(H 2 O)] was successfully prepared using the linear, highly conjugated anthryl derivative ligand 9,10-di(4′-carboxyphenyl)anthracene (DCPA) and the nitrogen-based ligand 1,3,5-tri(1H-imidazol-1-yl) benzene (tib). Single-crystal X-ray diffraction, thermogravimetric analysis, powder X-ray diffraction and X-ray photoelectron spectroscopy (XPS) confirmed that the Zn-MOF not only possessed excellent water and thermal stability but could also serve as a highly selective fluorescent sensor for specific recognition of L-tryptophan (L-Trp) among diverse amino acids and Fe 3+ in cationic aqueous solutions. In addition, this material also showed potential application value in the field of light-emitting devices.
