Construction of AgNPs-loaded oriented hydrogel based on Periostracum Cicadae chitosan by electro-assembly for rapid hemostasis and wound healing

Yujie Lei,Shilei Wang,Qian Liu,Duan Zeng,Kexing Wang,Cong Chang,Baohui Zhang,Lin Zhang,Zhaohua Shi,Yan Meng

Oriented hydrogels have been recognized for their ability to promote cell proliferation and enhance wound repair. Chitosan, valued for its biocompatibility and biodegradability, is widely used in hydrogel preparation. However, chitosan derived from Periostracum Cicadae (“Chantui”) has not been explored for creating antibacterial-oriented hydrogels. In this study, we successfully developed a Periostracum Cicadae chitosan-based oriented hydrogel (O-CH) using an electro-assembly method. The O-CH hydrogel exhibited superior hemostatic performance and mechanical strength, with stress levels 7.6 times higher than those of traditional alkali-neutralized hydrogels. Subsequently, silver nanoparticles (AgNPs) were formed in situ on the O-CH hydrogel without the need for additional reducing agents or stabilizers, yielding O-CH@Ag20, which exhibited excellent biocompatibility and enhanced antibacterial properties. In an S. aureus -infected full-thickness rat skin defect model, O-CH@Ag20 reduced bacterial counts at the wound site by 6-fold compared to the gauze group on day 3 and achieved a 97 % wound healing rate by day 10, versus 75 % in the gauze group. Histological analysis revealed that O-CH@Ag20 regulated angiogenesis, collagen deposition, and fibroblast differentiation into myofibroblasts, thereby accelerating wound contraction during remodeling. This study presents a novel approach to addressing clinical challenges in infected wound healing using Periostracum Cicadae chitosan-based antibacterial hydrogels.

Conduction of AgNPs with different surface charges in pine trees and their prevention and control of pine wood nematode disease

Shouhe Pan,Pengkun Yan,Rui Zhao,Fengyu Li,Lei Wang,Yinmin Wang,Ze Lv,Yingjian Ma,Meng Yu,Xinyu Guo,Yonggang Xia,Yong Xu,Xuemin Wu

Pine wood nematode disease is caused by pine wood nematodes and is difficult to prevent and control because the nematodes are located inside the trees. In this study, three types of silver nanoparticles (AgNPs) with different surface charges were prepared using Cetyl trimethyl ammonium chloride (CTAC), Polyvinyl pyrrolidone (PVP), and Acacia senegal (GA), and the ion and element concentrations in tree sap were measured after the introduction of AgNPs. Additionally, the effects of the introduction of AgNPs by foliar and soil applications and by trunk injection on the delivery of the AgNPs in the trees were compared. It was found that the LC 50 values of AgNP-GA, AgNP-PVP, and AgNP-CTAC against pine wood nematodes were 0.59, 1.15, and 1.25 mg∙L − 1 , respectively. Our toxicity mechanism studies showed that AgNP can inhibit the activity of acetylcholinesterase and catalase in pine wood nematodes, which is the main origin of the inhibition of pine wood nematode activity by AgNPs. The ion concentration of the sap has a significant impact on the delivery of AgNPs within the tree, with negatively charged AgNP-GA displaying better delivery within pine trees. In addition to conduits and sieve tubes, resin channels and intercellular spaces are likely to be the key sites for AgNP delivery. It was found that trunk injection facilitates the delivery of AgNPs within trees. This study provides an effective strategy for the prevention and control of pine wilt disease, tree trunk pests, and other diseases.

Berberine@AgNPs@Carboxylated chitosan hydrogel dressing with immunomodulatory and anti-biofilm properties promotes wound repair in drug-resistant bacterial infections

Xiaoqi Wang,Jue Hu,Chunjing Chen,Jun Lu,Chang Liu,Yi Ning,Fangguo Lu

Methicillin-resistant Staphylococcus aureus (MRSA) is a bacterial strain resistant to multiple antibiotics frequently encountered in clinical settings. Excessive antibiotic use has increased bacterial resistance, leaving a lack of effective treatments for MRSA infections. MRSA often colonizes the surface of skin wounds, resulting in chronic inflammation and protracted wound healing. The biofilm formation hinders the complete eradication of the bacteria, exacerbating the local inflammatory response and impeding wound healing. This study presents an innovative methodology for managing MRSA-infected skin wounds. The novel immunomodulatory hydrogel composed of Berberine, silver nanoparticles (AgNPs), and carboxylated chitosan (designated as Ber@AgNPs@CHI hydrogel) demonstrates enhanced therapeutic efficacy in a murine model of MRSA skin infection. This hydrogel is effective in eradicating MRSA and preventing biofilm formation. Furthermore, it modulates the local immune microenvironment by facilitating the transition of macrophages from the M1 to M2 phenotype and increasing the production of vascular endothelial growth factor (VEGF). These actions collectively facilitate the progression of the wound from the inflammatory to the proliferative phase, enhancing the early stages of wound healing. Hence, this safe and effective hydrogel mediates wound healing from multiple perspectives and targets, providing a new potential avenue for treating persistent infected wounds caused by clinical MRSA.

Facile Grafting of Silver Nanoparticles into Copper and Guanosine 5′-Monophosphate Metal Organic Frameworks (AgNPs@Cu/GMP): Characterization and Antimicrobial Activity

Soomro Najaf Ali,Amur Safdar Ali,Wei Yun,Shah Aamir Hassan,Jiao Mengzhao,Liang Hao,Yuan Qeping

Abstract Silver nanoparticles (AgNPs) are effective antibacterial agent but lack of stability and rapid aggregation limits their antibacterial efficiency in biomedical applications. To overcome these shortcomings, various strategies have been adopted so far but those strategies are not much effective at room temperature and in absence of external stimuli. Thus, herein, we have successfully grafted AgNPs into copper and guanosine 5′-monophosphate (Cu/GMP) metal organic frameworks at room temperature without using any external stimuli. The loading encapsulation ratio of AgNPs into Cu/GMP was approx. 99.83% at neutral pH. The AgNPs@Cu/GMP was characterized by various techniques, such as energy-dispersive x-ray spectroscopy, transmission electron microscopy, x-ray diffraction and Fourier transforms infrared spectroscopy. All these techniques have confirmed the loading of AgNPs into Cu/GMP (MOFs). Furthermore, antimicrobial activities of AgNPs, Cu/GMP and AgNPs@Cu/GMP were assessed against Escherichia coli (Gram-negative) and Staphylococcus aureus (Gram-positive) bacterium. As compared to pure AgNPs, the composite material AgNPs@Cu/GMP had shown significant growth of inhibition zones against tested bacteria. The stability of AgNPs@Cu/GMP was remarkable, i.e., six times higher than free AgNPs even after 15 days. These results make AgNPs encapsulated into Cu/GMP a promising antimicrobial candidate and is expected to deliver an advance platform to pharmaceuticals and biomedical applications. Graphic abstract

A room temperature synthesized SERS substrate of AgNPs/PVA hydrogel for highly sensitive detection of enrofloxacin using a portable Raman system

Qingli Xie,Xueyang Wang,Boxue Chang,Yinlan Ruan

Enrofloxacin (EnFX), an antibiotic widely used in animal husbandry, can accumulate in the food chain due to excessive or long-term residues, posing a potential threat to human health. Surface-enhanced Raman scattering (SERS) technology, with its advantages of high sensitivity and rapid label-free analysis, offers a new approach for antibiotic residue detection. However, SERS substrate preparation at high temperature, poor signal reproducibility, and dependence on Raman spectroscopy still limit its practical application. To address these issues, this study proposed a flexible SERS substrate preparation scheme based on polyvinyl alcohol (PVA) hydrogel and silver nanoparticles (AgNPs). The substrate is prepared by a room-temperature seed growth method, which is simple, highly reproducible, and can be combined with portable Raman devices for rapid and flexible on-site detection. Research showed that the AgNPs/PVA hydrogel substrate exhibited excellent sensitivity and signal consistency in the detection of enrofloxacin and Rhodamine 6G (R6G), with a relative standard deviation (RSD) as low as 6%, and can be stably stored in air for more than one month. This method combines low cost, high scalability, and excellent environmental adaptability, providing an efficient, economical, and sustainable solution for the portable detection of antibiotic residues.

Mengni Fan,Jingxing Si,Xiaogang Xu,Linfu Chen,Junpeng Chen,Chao Yang,Jingwu Zhu,Lihuang Wu,Jiang Tian,Xiaoyi Chen,Xiaozhou Mou,Xiaojun Cai

Development of multifunctional antibacterial agent with long-lasting antibacterial activity and biofilm ablation performance is significant for the effective treatment of bacterial infections. Here, by utilizing the electrostatic interaction between sulfonated chitosan (SCS) and Ag + and chitosan (CS), and the sodium borohydride reduction method, a versatile antibacterial agent (AgNPs@CS/SCS) capable of generating silver nanoparticles (AgNPs) in-situ and long-acting slow-release Ag + was developed. AgNPs@CS/SCS has a good physiological stability and can long-acting slow-release of Ag + due to the pH-dependent Ag + release behavior of AgNPs. Noteworthy, AgNPs@CS/SCS can exert both excellent short- and long-term antibacterial and biofilm ablation activity. Importantly, it also exhibits superior antibacterial activity in the treatment of implant infections, accompanied by good biocompatibility. Together, this study suggest that AgNPs@CS/CSC is indeed a versatile antibacterial agent, and is expected to provide an effective treatment modality for implant infections in the clinic settings.

Transcriptomics-based investigation of molecular mechanisms underlying synergistic antimicrobial effects of AgNPs and Domiphen on the human fungal pathogen Aspergillus fumigatus

Du Wenlong,Xu Ruolin,He Zhiqiang,Yang Huan,Gu Yufan,Liu Yi

Critically ill patients have higher risk of serious fungal infections, such as invasive aspergillosis (IA) which is mainly caused by the human fungal pathogen Aspergillus fumigatus. Triazole drugs are the primary therapeutic agents for the first-line treatment of IA, which could easily cause drug resistance problems. Here, we assess the potential of AgNPs synthesized with Artemisia argyi leaf extract and domiphen as new antifungal agents to produce synergistic antimicrobial effects on Aspergillus fumigatus, and dissect possible molecular mechanisms of action. Plate inoculation assays combined with drug susceptibility test and cytotoxicity test showed that the combination of AgNPs and domiphen has synergistic antimicrobial effects on A. fumigatus with low cytotoxicity. Gene Ontology (GO) enrichment analysis showed that AgNPs and domiphen inhibit the growth of A. fumigatus by suppressing nitrate assimilation, and purine nucleobase metabolic process and amino acid transmembrane transport, respectively. When the two drugs are combined, AgNPs has epistatic effects on domiphen. Moreover, the combination of AgNPs and domiphen primarily influence secondary metabolites biosynthesis, steroid biosynthesis and nucleotide sugar metabolism of A. fumigatus via Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis. Furthermore, protein–protein interactions (PPI) analysis combined with validation experiments showed that the combination of AgNPs and domiphen could enhance the expression of copper transporter and inhibit nitrogen source metabolism. In addition, the synergistic antimicrobial effects could be enhanced or eliminated depending on exogenous addition of copper and nitrogen source, respectively. Taken together, the results of this study provide a theoretical basis and a new strategy for the treatment of IA.

Enhancing K2S2O8 electrochemiluminescence based on silver nanoparticles and zinc metal–organic framework composite (AgNPs@ZnMOF) for the determination of L-cysteine

Lina Wang,Qi Wu,Ru Yu,Hongge Zhang,Fei Nie,Wenyan Zhang

A silver nanoparticle-doped Zn(II) metal–organic framework composite (AgNPs@ZnMOF) was investigated as an electrochemiluminescence (ECL) signal enhancer for potassium persulfate. First, ZnMOF was prepared by a one-step hydrothermal method, and then AgNPs@ZnMOF composite was obtained by depositing AgNPs on the surface and interior of ZnMOF. After the AgNPs@ZnMOF composite was modified on the glass carbon electrode (GCE), the cathode luminescence of potassium persulfate on bare GCE was enhanced by 8 times. A dual amplification mechanism provided by Zn(II) and Ag nanoparticles in the AgNPs@ZnMOF composite has been validated by ECL spectra, fluorescence spectra, and electrochemical methods. The interaction between the sulfhydryl groups in L-cysteine (L-Cys) and AgNPs significantly affects the catalytic luminescence of the AgNPs@ZnMOF composite. Thus, a sensitive ECL method for the determination of L-Cys was developed based on the inhibition effect of L-Cys on the ECL signal within the linear range from 5.0 nM to 1.0 μM and the limit of detection was found to be 2 nM (S/N = 3). The established method has been successfully applied to the determination of L-Cys in human urine.

Natural glycyrrhizic acid-tailored hydrogel with in-situ gradient reduction of AgNPs layer as high-performance, multi-functional, sustainable flexible sensors

Hao Zhang,Nan Tang,Xia Yu,Zhongkai Guo,Zhen Liu,Xiaoming Sun,Min-Hui Li,Jun Hu

To date, flexible sensors derived from hydrogel materials have been popularly adopted in the field of wearable electronic devices. It is urgent and prospective to create a high-performance and multi-functional hydrogel by simple and green methods to satisfy the various demands for long-term direct human contact. Herein, a sustainable physical double network hydrogel is prepared by natural glycyrrhizic acid (GL) and biocompatible polyvinyl alcohol (PVA) in water/glycerol solvent, which exhibits high mechanical properties, good biocompatible, anti-freezing and anti-drying ability. Inspired by the reducing abilities of GL, a gradient silver nanoparticles layer can be constructed in situ on the hydrogel surface without additional reducing agents. It not only affords the UV-light shielding and antibacterial effect for the hydrogel, but also effectively improves the sensing performance. The simultaneous realization of desired properties enables the hydrogel to be assembled into a sensor to detect all-dimensional human motion with high sensitivity, stability and durability. Benefiting from the renewable raw material, non-toxic reagent and facile preparation process, this work provides new insight and inspiration for the design of flexible sensors with high performance, multi-functionality and sustainability.

Electrically Conductive AgNPs/TPU-Electrospun Fibers with Hierarchical Microstructures by a One-Step Method for a High-Performance Flexible Pressure Sensor

Chaoying Wang,Chuanguo Ma,Xiaoyu Wu,Mengnan Li,Shaoning Lu,Peibang Dai

To obtain flexible pressure sensors with simple preparation process, high sensitivity, fast response time, wide detection range, and good stability at the same time is still a great challenge. Herein, silver trifluoroacetate (C2AgF3O2) was introduced into thermoplastic polyurethane (TPU)-electrospun solution, and a C2AgF3O2/TPU nanofiber film with hierarchical microstructure was prepared by “one-step” electrostatic spinning. Then, the conductive silver nanoparticle/TPU composite nanofiber film was obtained after the reduction of the C2AgF3O2/TPU nanofiber film. The hierarchical microstructure of the nanofiber film consists of tiny bumps with varying heights on the surface and holes with different sizes inside, which achieves a wide detection range of 0–100 kPa and a high sensitivity of 7.51 kPa–1. In addition, the response time and recovery time are 100 and 70 ms, respectively, and the stability of the sensor is still high even after 1000 test cycles of testing. The developed sensors show good sensing performance in the monitoring of pulse, finger movement, Morse code, and other signals, proving that they have a broad application prospect in human motion detection and wearable devices.

Cascade amplifying aptasensor for positively correlated detecting OTA: Based on DNase I-assisted cyclic enzyme digestion and AgNPs@gel-enhanced fluorescence

Chenfei Han,Yu Xiao,Zhenjiang Liu,Daolin Du,Ming Li

To improve the performance of aptasensor, the coupling of novel material and strategy had been the hottest research with the promising prospects. In this study, a cascade amplifying aptasensor (CA-aptasensor), which had coupled the DNase I-assisted cyclic enzyme digestion and AgNPs@gel-enhanced fluorescence, had reflected the desired performance for positively correlated detecting OTA . For this purpose, the GO@PVP was used to adsorb and quench the free FAM-Apt, while the complex of OTA and FAM-Apt was digested by the DNase I. The released OTA allowed the free FAM-Apt to separate from the GO@PVP cyclically, thus the OTA level was amplified preliminarily. The AgNPs@gel could combine with the released FAM to form the metal enhanced fluorescence (MEF) effect, resulting in the cascade signal amplification. The OTA was detected at pg mL −1 level with an ultrasensitive LOD of 3.33 pg mL −1 and IC 50 of 13.6 pg mL −1 , which improved 90-fold and 728-fold than that of ELASA for same aptamer. The CA-aptasensor indicated high specificity, reliability and practicability. Meanwhile, the CA-aptasensor exhibited more highlights, including the efficient homogeneous system, low background signal and standardized aptamer recognizer. This study might provide a new approach for improving the performance of aptasensor, which could also guide the signaling cascade and positively correlated detection for other risk factors.
