Micronutrient intake during early life is crucial for supporting healthy intestinal development and function. However, the impact of the timing of vitamin B6 (VB6) intake on intestinal development in young organisms has not been investigated. The study investigated the effects of VB6 (45 mg/kg) intake during the active phase versus the rest phase on intestinal digestion, intestinal barrier, and ileal and colonic microbiota, and used fecal microbiota transplantation to explore the role of the microbiota in VB6-mediated intestinal health. VB6 intake during the active phase increased digestive enzyme activity, intestinal villus height, and mRNA expression of tight junction proteins ( Occludin and ZO-1 ), thereby enhancing digestive function and epithelial barrier integrity. Microbiota analysis revealed that active phase VB6 intake predominantly modulated the ileum microbial community, promoting colonization by beneficial genera such as Ligilactobacillus and Bifidobacterium , and increasing ileum short-chain fatty acid levels. Fecal microbiota transplantation confirmed that VB6 acts through the gut microbiota. Therefore, this study demonstrates that VB6 modulates intestinal development via the gut microbiota, with active phase intake conferring greater benefits on intestinal development than rest phase intake .

s Purpose The high incidence and mortality rates of lung cancer pose a great challenge for early detection and prognostic assessment. Liquid biopsy biomarkers based on circulating tumor cells (CTC) and circulating tumor DNA (ctDNA), among others, hold promise for non-invasive detection. The aim of this study was to construct a CTC sorting system that can be used to monitor lung cancer progression and compared with ctDNA for downstream analysis. Results The sorting efficiency of the CTC sorting system for lung cancer cells was 95.70 %, and the system demonstrated the ability to monitor CTC changes and tumor progression in animals. The cutoff value of CTC detection in lung cancer was 7.5/5 mL (Area under the curve [AUC]: 0.9976, 95 % CI: 0.9946–1.000), with a sensitivity of 96.70 % and specificity of 98.41 %, effectively indicating progression. Compared with ctDNA, CTC-based analysis detected a broader genomic mutation spectrum with a concordance rate of 19.90 %, but the detection rate of the genomic mutation spectrum could be expanded by the combined test. Conclusion In this study, we successfully developed a novel clinically feasible CTC sorting system capable of monitoring lung cancer progression, which can be applied to early diagnosis, efficacy assessment, and monitoring tumor progression in lung cancer. In addition, CTC showed low concordance with mutations found in ctDNA, and CTC may have specific mutations independent of ctDNA.

This study develops a novel self-monitoring hydrogel via UV-initiated crosslinking of carboxymethyl cellulose(CMC) with silver nanowires(AgNW) and curcumin-β-cyclodextrin (CUR-β-CD), aimed to prolong loquat shelf life. Structural and morphological analysis indicates that hydrogels exhibit stable porous network structures and outstanding mechanical properties when CMC/AgNW/CaCl 2 /cellulose nanofibers (MICCA) are added at concentrations of 0.54 g, 0.8 mL, 1 mL, and 1.9 g, respectively. These hydrogels demonstrate a hardness of 60.67 N (a 338% increase) and a rebound elasticity of 3.943 mm. The hydrogel exhibited pH-dependent swelling(30.12% at pH=5;66.85% at pH=9)and thermal stability(29.46% residue at 800°C). Notably, the material integrates silver nanowires (AgNWs) at an optimal content of 0.016% to construct a piezoresistive sensing unit with a gauge factor of 3.59. It also combines with curcumin-β-cyclodextrin (CUR-β-CD) inclusion complexes at 0.2%, which are activated by blue light to realize efficient photodynamic antimicrobials, achieving inhibition rates of 92.28% and 95.14% against E. coli and Listeria monocytogenes, respectively. This system significantly improves the preservation performance of loquat, extending its shelf-life by 6 days compared to the control group. This dual-function system integrates mechanical-force sensing with on-demand antimicrobial action, offering an active strategy for intelligent fruit preservation.

3D-printed porous titanium–aluminum–vanadium (Ti6Al4V, pTi) scaffolds offer surgeons a good option for the reconstruction of large bone defects, especially at the load-bearing sites. However, poor osteogenesis limits its application in clinic. In this study, a new magnetic coating is successfully fabricated by codepositing of Fe 3 O 4 nanoparticles and polydopamine (PDA) on the surface of 3D-printed pTi scaffolds, which enhances cell attachment, proliferation, and osteogenic differentiation of hBMSCs in vitro and new bone formation of rabbit femoral bone defects in vivo with/without a static magnetic field (SMF). Furthermore, through proteomic analysis, the enhanced osteogenic effect of the magnetic Fe 3 O 4 /PDA coating with the SMF is found to be related to upregulate the TGF β -Smads signaling pathway. Therefore, this work provides a simple protocol to improve the osteogenesis of 3D-printed porous pTi scaffolds, which will help their application in clinic.

Human flap endonuclease 1 (hFEN1) is instrumental in DNA replication and repair. It is able to cleave the 5′ single-stranded protrusion (also known as 5′ flap) resulting from strand displacement reactions. In light of its crucial functions, hFEN1 is now deemed as a nontrivial target in the DNA damage response system for anticancer drug development. Herein, we report that myricetin and some natural flavonoids are able to inhibit hFEN1. Structure–activity relationship, inhibitory mechanisms, molecular docking, and cancer cell-based assays have been performed. Our original findings expand the activity of flavonoids and may pave the way for flavonoid-assisted targeted cancer therapy.

Specifically inhibiting the proliferation of activated macrophages and clearing the high levels of reactive oxygen species (ROS) secreted by macrophages is crucial for osteoarthritis (OA) treatment. Moreover, if the clearance of these high levels of ROS can be simultaneously used to induce oxidation-responsive release of anti-inflammatory drugs, the therapeutic effect of OA may be further improved. Here, a multifunctional anti-inflammatory drug (CPHs) based on a peptide dendrimer nanogel was constructed by physically encapsulating CORM-401 and wrapping its surface with folic acid (FA)-modified hyaluronic acid (HA). CPHs is capable of efficiently entering activated macrophages via FA- and HA-mediated specific targeting effects and then rapidly release large amounts of CO by massive consumption of H 2 O 2 . The generated CO effectively suppresses the secretion of interleukin (IL)-1β, IL-6, and tumor necrosis factor (TNF)-α by inhibiting cell proliferation ; inducing the activation of heme oxygenase (HO-1), and downregulating the expression of p38 MAPK , NF-kB (p50/p65) and TLR-2. In vivo experiments further confirmed that CPHs can massively deplete ROS in OA joints and effectively suppress the degradation of articular cartilage and their extracellular matrix. More importantly, CPHs is non-toxic to normal macrophages, and the high levels of CO generated in the joints do not result in notable changes in the HbCO levels in blood. Together, these results show that CPHs is an effective and safe anti-inflammatory drug and has essential application prospects in OA treatment.

Objective To investigate the potential mechanisms underlying the combined therapeutic effects of sodium propionate and Sishen Pill and to provide experimental evidence supporting their mode of action. Methods The method utilized involved the induction of a mouse model of Diarrhea with Kidney-Yang Deficiency Syndrome by administering adenine combined with Folium sennae. After the successful establishment of the model, the mice in the model group were randomly assigned to one of the following treatment groups: natural recovery group, sodium propionate group, 75% Sishen Pill combined with 60 mg/kg sodium propionate group, or Sishen Pill group. Results The 75% Sishen Pill combined with 60 mg/kg sodium propionate demonstrated significantly better therapeutic effects compared to the Sishen Pill alone and sodium propionate alone groups. The combined treatment significantly improved the behavioral indices of mice (p < 0.05); increased the levels of MUC2 and sIgA (p < 0.01); reduced IL-6 levels (p < 0.05); and improved structural damage in the kidneys and small intestines. Intestinal microbiota analysis showed that 75% Sishen Pill combined with 60 mg/kg sodium propionate significantly increased beneficial bacteria such as Lactobacillus (p < 0.05), while Prevotellamassilia and Maribacter were significantly enriched in this group. Correlation analysis revealed that Lactobacillus and Pediococcus were positively correlated with MUC2 and sIgA, while negatively correlated with IL-6.Conclusion The 75% Sishen Pill combined with 60 mg/kg sodium propionate significantly alleviates symptoms related to Diarrhea with Kidney-Yang Deficiency Syndrome, enhances the efficacy of Sishen Pill by regulating the intestinal microbiota, boosts intestinal immune function, and reduces intestinal inflammation, providing a new approach for treating Diarrhea with Kidney-Yang Deficiency Syndrome.

Herein, we developed a universal “soil for soil remediation” strategy to utilize soil resources for cation-/anion-type heavy-metal-contaminated soil remediation. The representative natural soils fetched from 12 different areas in China have been universally converted into Fe2O3-CAN (hydroxycancrinite) zeolite composite materials. Furthermore, we discover that due to the presence of exchangeable Na+ and OH– in CAN channels together with the intrinsic surface adsorption function of Fe2O3-CAN, Fe2O3-CAN holds distinctive cation/anion dual ion-exchange functions and strong adsorption capacity, which could synergistically remediate cation-/anion-type HM-contaminated soil. When 25 g/kg Fe2O3-CAN is added into Cd/As polluted soil (100–200 ppm), Cd2+ and AsO2– in soil could be immobilized simultaneously and the Cd/As bioaccumulation in the harvested vegetables is dramatically suppressed with reduction ratios of 48.1–67.0%, effectively reducing the HM enrichment in the phytosphere. Overall, this work confirms the high universality of Fe2O3-CAN synthesis using different types of natural soils and demonstrates that the as-synthesized Fe2O3-CAN composite material can be further effectively used for soil remediation. The “soil for soil remediation” strategy developed here could improve the flexibility and economy of soil remediation, which can be extended to a broad range of locations worldwide.

Mercury (II) ion (Hg 2 + ) as highly toxic heavy metal may be accumulated in aquatic ecosystems and animals species so as to enter human body to conduct health harm. To ensure the safety of fishes food, hence, it is of great interest to evaluate the Hg 2+ levels in different kinds of fishes as well as Hg 2+ removal in aquaculture tailwater. In this article, a selective colormetric detection and efficient removal strategy has been developed for Hg 2+ ions by the controlled supermolecular self-assembly of melamine (MA)-platinum (Pt) composites onto mesoporous Fe 3 O 4 carriers. The resulting Fe 3 O 4 @MA-Pt exhibited the largely enhanced catalysis properties, which might rationally decrease in the presence of Hg 2+ ions through the strong Pt-Hg and MA-Hg interactions. A colorimetric analysis technique based on Fe 3 O 4 @MA-Pt catalysis was thereby designed for detecting Hg 2+ ions with the linear concentrations ranging from 0.010 to 1.0 μM. Moreover, Hg 2+ levels in muscles samples of four kinds of carnivorous and omnivorous fishes were evaluated, displaying an obvious difference in order of snakehead > largemouth bass > crucian carp > silver carp. The data indicate that the carnivorous fishes should possess higher Hg 2+ levels than the omnivorous ones, thus providing a recognizable guidance to the safety evaluation of Hg 2+ in different kinds of fishes. Furthermore, the Fe 3 O 4 @MA-Pt composites with high surface-to-volume ratios were utilized to adsorb Hg 2+ ions in aquaculture tailwater, achieving the adsorption efficiencies up to 99.73 %. Such a colorimetric analysis and removal strategy for Hg 2+ ions may promise for aquatic food safety, clinical diagnostics, environmental monitoring, and tailwater treatments.

Carvacrol expresses a wide range of biological activities, but the studies of its mechanisms focused on bacteria, mainly involving the destruction of the plasma membrane. In this study, carvacrol exhibited strong antifungal activities against several phytopathogenic fungi and determined a novel antifungal mechanism against Lasiodiplodia theobromae. RNA sequencing indicated that many genes of L. theobromae hyphae were predominately induced by carvacrol, particularly those involved in replication and transcription. Hyperchromic, hypsochromic, and bathochromic effects in the UV-visible absorption spectrum were observed following titration of calf thymus DNA (ctDNA) and carvacrol, which indicated the formation of a DNA–carvacrol complex. Circular dichroism (CD) spectroscopy indicated the response of DNA to carvacrol was similar to 4’, 6-diamidino-2-phenylindole (DAPI), but different from that of ethidium bromide (EB), implying the ionic bonds between carvacrol and ctDNA. Fluorescence spectrum (FS) analysis indicated that carvacrol quenched the fluorescence of double-stranded DNA more than single-stranded DNA, indicating that carvacrol mainly bound to double-stranded DNA. A displacement assay showed that carvacrol reduced the fluorescence intensity of the DNA–DAPI complex through competing with DAPI, but this did not occur for DNA–EB. FS assay revealed that carvacrol bound to AAA sequence on the minor groove of ds-oligonucleotides. The hydroxyl of carvacrol was verified to bind to ctDNA through a comparative test in which structural analogs of carvacrol, including thymol, 4-ethyl-1,2-dimethyl, etc. were analyzed. The current study indicated carvacrol can destruct plasma membranes and bind to the minor groove of DNA, which inhibited the fungal proliferation by disturbing the stability of dsDNA.
