Synthesis and characterization of D-/L-methionine grafted PEDOTs for selective recognition of 3,4-dihydroxyphenylalanine enantiomers

Dong Li-qi,Hu Du-fen,Duan Xue-min,Wang Zhi-peng,Zhang Kai-xin,Zhu Xiao-fei,Sun Hui,Zhang You-shan,Xu Jing-kun

Two pairs of amino-acid functionalized poly(3,4-ethylenedioxythiophene) (PEDOT) derivatives, namely, poly( N -(tert-butoxycarbonyl)- L -methionyl (3,4-ethylenedioxythiophene-2’-yl)methylamide) ( L -PEDOT-Boc-Met) and poly( N -(tert-butoxycarbonyl)- D -methionyl (3,4-ethylenedioxythiophene-2’-yl)methylamide) ( D -PEDOT-Boc-Met); poly( L -methionyl (3,4-ethylenedioxythiophene-2’-yl)methylamide) ( L -PEDOT-Met) and poly( D -methionyl (3,4-ethylenedioxythiophene-2’-yl)methylamide) ( D -PEDOT-Met) were synthesized via chemical oxidative polymerization of corresponding monomers. The structural characterization, spectroscopic properties and thermal stability of these monomers and polymers were systematically explored by FTIR spectra, Raman spectra, XRD spectra, UV-Vis spectra and thermogravimetric analysis. As chiral electrode materials, these polymers were employed to successfully recognize 3,4-dihydroxyphenylalanine (DOPA) enantiomers by cyclic voltammetry (CV) in sulphuric acid solution. The measurement results reveal that the tendency was hetero-chiral interaction between L -PEDOT-Met/PVA/GCE and D -DOPA, D -PEDOT-Met/PVA/GCE and L -DOPA, respectively. Also, the mechanism of chiral discrimination was discussed. All the results implied that the combination of electrochemical molecular recognition technology and chiral PEDOT materials can be a promising approach for chiral recognition and may open new opportunities for facile, biocompatible, sensitive and robust chiral assays in biochemical applications.

Enantiospecific profiling of D-amino acid for gastric cancer diagnosis by using a biocatalytic MHOF nanoreactor

Qianxi Zhou,Hao Zhang,Tianyu Zeng,Jiahua Yang,Qizhi Liang,Hai Shi,Yongmei Yin,Genxi Li

The dynamic variation of D-amino acids (D-AAs) in human saliva has great potential as the indicator for individual physiological states and disease progression; however, enantiospecific profiling of D-AAs is a great challenge. In this work, a nanoreactor based on mesoporous hydrogen-bonded organic framework (MHOF) has been fabricated for the enantiospecific profiling of D-AAs. The nanoreactor is prepared by using MHOFs as the scaffold to organize the biocatalytic cascade of D-AA oxidase (DAAO) and horseradish peroxidase (HRP). In the meanwhile, the large, stable, and robust transport channels of MHOFs allow the unimpeded transport of the involved biocatalytic substrates. Once D-AA substrates efficiently transport into MHOFs, the chromogenic reactions of 3,3′,5,5′-tetramethylbenzidine (TMB) and H 2 O 2 can be activated. So, using this MHOF nanoreactor, enantiospecific and visual detection of D-AAs can be achieved. Moreover, the stable nanoreactor can be operated in complex solution and utilized for the profiling of D-AA levels in saliva samples collected from gastric cancer patients, suggesting that D-AAs may be a powerful fingerprint for the diagnosis of gastric cancer. Therefore, the fabricated MHOF-based nanoreactor may provide an emerging tool for the enantiospecific profiling of D-AAs and supports the D-AA fingerprint as the indicator of gastric cancer diagnosis.

Impact of the key residues of 1-Deoxy-D-Xylulose-5-Phosphate synthase on its catalysis

Yan-Fei Liang,Shuai Feng,Dan Wu,Meng Ma,Jin-Tao Ren,Qian Zhao,Wen-Yun Gao,Heng Li

1-Deoxy- D- xylulose-5-phosphate synthase (DXS) is the first rate-limiting enzyme in the 2-methyl- D- erythritol-4-phosphate (MEP) terpenoid biosynthetic pathway. To explore the active sites of the protein, we made a series of mutations to the key amino acid residues of Escherichia coli DXS including H49, R420, D427, and R478. The results show that several mutants including H49Q lose almost all their catalytic activity in DXP synthesis, but the double-point mutants H49QD427H and H49QD427 N recover 100 % activity of the wild type enzyme. The kinetic characterization displays that the affinity of the double-point mutants for pyruvate and D- glyceraldehyde-3-phosphate only slightly decreases. We also assessed impact of the mutation on the oxidative decarboxylation activity of all the mutants. Despite the fact that H49QD427H and H49QD427 N get back the full activity with regard to DXP synthesis, their oxidase activity remains unrestored. Furthermore, we assayed the acceptor substrate spectrum of the mutants and the results show that a large portion of them catalyzes ligation between pyruvate and nitrosobenzene to form N-phenyl-N-hydroxyacetamide, whereas R420D, R420F, and D427H exhibit activity in connection of pyruvate and benzaldehyde to afford phenylacetocarbinol (PAC), with the first two mutants yielding ( S )-PAC (yields 21 %-25 %, ee values 71–75 %) while D427H furnishing ( R )-PAC (yield 41 %, ee value 91 %), implying the key role of D427 for the acceptor substrate recognition and the stereoselectivity of the enzyme. It is worth noting that this is the first report of DXS mutants that can recognize benzaldehyde as an acceptor substrate and generate both enantiomers of PAC.

Competitive Chiral Cocrystallization Inspired Enantioseparation: Mechanistic Insights into R/S-Mandelic Acid and d/l-Prolinamide

Detao Li,Zichen Ning,Feiqiang He,Zhi Gao,Limin Zhou,Li Xu,Zhijian Zheng,Jerry Heng,Shichao Du,Jinbo Ouyang

To enable efficient separation of chiral drugs and enhance their practical utility, this study investigates cocrystallization as a novel separation and purification strategy. Using R/S-mandelic acid and d/l-prolinamide as model compounds, we screened potential cocrystal/salt coformers and successfully synthesized seven novel chiral salts. The proton transfer was elucidated by analyzing molecular electrostatic potential differences at the hydrogen-bonding sites. Moreover, the formation mechanism of salts was explored through structural analyses, such as Hirshfeld surface calculations, energy framework assessments, and crystal void analysis. Considering the distinct properties of various chiral salts, we conducted competitive cocrystallization experiments, establishing a complementary chiral pairing principle that provides a theoretical foundation for cocrystallization-based chiral resolution. Based on these findings, we developed multiple processes for the separation of R- and S-mandelic acid as well as l- and d-prolinamide, achieving product purities exceeding 80%, which strongly demonstrate the feasibility of cocrystallization separation technology for chiral resolution. This work not only introduces a groundbreaking approach to chiral separation but also highlights the vast potential of cocrystallization technology in pharmaceutical purification and beyond.

Saikosaponin D Exacerbates Acetaminophen-induced Liver Injury by Sabotaging GABARAP-SNARE Complex Assembly in Protective Autophagy

Guifang Fan,Xiaojiaoyang Li,Fanghong Li,Ranyun Chen,Xiaoyong Xue,Le Wang,Qi Zheng,Shuni Duan,Ranyi Luo,Rong Sun,Runping Liu

Background : Radix Bupleuri (RB) and acetaminophen (APAP) are two popular medications having potential hepatotoxicity and substantial risks of irrational co-administration and excessive use, posing an overlooked danger of drug-induced liver injury (DILI). Autophagy is a protective mechanism against APAP-induced DILI, yet, saikosaponin d (SSd) in RB has been characterized to regulate autophagy, although the current findings are controversial. Purpose : We aim to elucidate whether SSd promoted APAP-induced liver injury by regulating autophagy. Methods : UPLC-MS analysis was employed to measure the hepatic abundance of APAP-cysteine protein adducts. Multiple techniques such as fluorescence probe, proteinase K protection assay, immunoprecipitation-coupled proteomic analysis, surface plasmon resonance, molecular docking and et.al were applied to evaluate the SSd on autophagy flux. Results : We discovered that, by inhibiting autophagy, SSd impaired the removal of APAP-cysteine protein adducts and delayed the compensation of damaged mitochondria. This ultimately potentiated the development of severe liver toxicity induced by subtoxic APAP. The use of autophagy probes, transmission electron microscopy, membrane curative assay, and protein K assay collectively revealed that SSd predominately disrupted autophagosome-lysosome fusion, without affecting other stages of autophagic flux. Immunoprecipitation-coupled proteomic analysis and surface plasmon resonance further found that SSd directly bound to GABARAP, thus preventing the recruitment and autoactivation of STX17 and the following assembly of STX17-SNAP29-VAMP8 complex. Conclusion : In conclusion, our findings not only highlight the significant risk of drug-induced liver injury associated with the co-administration of RB and APAP in clinical practice but also unveils that GABARAP-SNARE complex is a novel druggable target for the treatment of autophagy-related diseases.

Adiponectin inhibits D‑gal‑induced cardiomyocyte senescence via AdipoR1/APPL1

Ruiying Liu,Jing Meng,Danfei Lou

The aim of the present study was to examine whether adiponectin could inhibit cardiomyocyte senescence induced by D‑galactose (D‑gal), and whether it functioned via the adiponectin receptor 1 (AdipoR1)/adaptor protein phosphotyrosine interacting with PH domain and leucine zipper 1 (APPL1) signaling pathway. For this purpose, the expression levels of adiponectin, AdipoR1 and APPL1 in mouse plasma and myocardial tissues were detected via reverse transcription‑quantitative PCR (RT‑qPCR) and western blotting. An adiponectin‑overexpression plasmid was transfected into D‑gal‑treated H9c2 cells prior to the detection of AdipoR1 and APPL1 expression by RT‑qPCR. Senescence‑associated β‑galactose staining was then performed to observe cellular senescence following the transfection of small interfering RNAs (si) targeting AdipoR1 and APPL1 into D‑gal‑treated H9c2 cells overexpressing adiponectin. Commercial kits were used to detect reactive oxygen species (ROS) production and malondialdehyde (MDA) content in the different groups. The expression levels of heme oxygenase (HO)‑1 and high mobility group box 1 (HMGB1) were examined by western blot analysis. The results revealed that the expression levels of adiponectin, AdipoR1 and APPL1 were downregulated in aged mouse plasma, myocardial tissues and D‑gal‑treated cardiomyocytes. It was also observed that AdipoR1 and APPL1 expression levels were significantly upregulated following the overexpression of adiponectin into D‑gal‑treated cardiomyocytes. Moreover, adiponectin overexpression reduced cellular senescence induced by D‑gal and the expression of p16 and p21; these effects were reversed following transfection with si‑AdipoR1 and si‑APPL1. Adiponectin also downregulated the levels of ROS and MDA in D‑gal‑treated H9c2 cells via AdipoR1/APPL1. Additionally, the release of HO‑11/HMGB1 was affected by adiponectin via AdipoR1/APPL1, and adiponectin/AdipoR1/APPL1 suppressed ROS production via HO‑1/HMGB1. On the whole, the present study demonstrated that adiponectin played an inhibitory role in cardiomyocyte senescence via the AdioR1/APPL1 signaling pathway and inhibited the levels of oxidative stress in senescent cardiomyocytes via the HO‑1/HMGB1 signaling pathway.

Antibacterial activity and mechanisms of D-3263 against Staphylococcus aureus

Liu Xiaoju,Xiong Yanpeng,Peng Renhai,Zhang Yufang,Cai Shuyu,Deng Qiwen,Yu Zhijian,Wen Zewen,Chen Zhong,Hou Tieying

Multi-drug-resistant Staphylococcus aureus infections necessitate novel antibiotic development. D-3263, a transient receptor potential melastatin member 8 (TRPM8) agonist, has potential antineoplastic properties. Here, we reported the antibacterial and antibiofilm activities of D-3263. Minimum inhibitory concentrations (MICs) against S. aureus , Enterococcus faecalis and E. faecium were ≤ 50 µM. D-3263 exhibited bactericidal effects against clinical methicillin-resistant S. aureus (MRSA) and E. faecalis strains at 4× MIC. Subinhibitory D-3263 concentrations effectively inhibited S. aureus and E. faecalis biofilms, with higher concentrations also clearing mature biofilms. Proteomic analysis revealed differential expression of 29 proteins under 1/2 × MIC D-3263, influencing amino acid biosynthesis and carbohydrate metabolism. Additionally, D-3263 enhanced membrane permeability of S. aureus and E. faecalis . Bacterial membrane phospholipids phosphatidylethanolamine (PE), phosphatidylglycerol (PG), and cardiolipin (CL) dose-dependently increased D-3263 MICs. Overall, our data suggested that D-3263 exhibited potent antibacterial and antibiofilm activities against S. aureus by targeting the cell membrane.

Methane Hydrate Formation on Hydrophilic Zr4+/D-Gel-Coated Surfaces

Cuiping Tang,Xinqi Liu,Yong Chen,Xiaoya Zang,Jinan Guan,Deqing Liang

During the production and transportation of natural gas, hydrate formation and accumulation in pipelines can lead to blockages, resulting in gas leakage, economic losses, and safety hazards. Recently, the development of superhydrophobic antigas hydrate coatings for pipelines has been identified as an effective strategy to mitigate pipeline plugging. Although superhydrophobic surfaces can effectively reduce the adhesion strength of hydrates to the wall, they tend to promote surface gas enrichment and water molecule ordering, thereby accelerating hydrate nucleation and growth. Furthermore, the microstructure of superhydrophobic surfaces is relatively fragile and susceptible to damage, which limits their long-term stability and reliability. This study proposes a hydrophilic gel coating, Zr4+/DMSO-Gel, which can delay the nucleation induction period of methane hydrates on solid surfaces and reduce their growth rate. Microscopic observations revealed that the coating induced partial decomposition of methane hydrate, resulting in the formation of a large water layer, which can reduce wall adhesion. Furthermore, Raman spectroscopy was employed to analyze the structure of surface water molecules, revealing the mechanism by which the coating inhibits hydrate formation. The results showed that strong hydrogen bonding interactions between the coating surface and water molecules resulted in the predominance of bound water (BW) on the surface, while the proportion of tetra-coordinated hydrogen-bonded water (4-HBW) was reduced, thereby inhibiting the formation of hydrate cage structures. These findings not only provide a novel approach for the development of hydrate-resistant coatings but also offer new insights into antiplugging strategies for natural gas pipelines.

Removal of boron from aqueous solutions by N-methyl-D-glucamine@γ-glycidoxypropyl-trimethoxysilane with nonwoven basalt fibers support

Yuanyang Zhang,Xiaoping Liang,Tao Jiang,Zishang Chen,Xiujie Ji

Waste water with boron is a widespread environmental problem. This work presents a kind of novel organic-inorganic composite boron adsorbent of NGN, N-methyl- D -glucamine (NMDG) @γ-glycidoxypropyl-trimethoxysilane (GPTMS) on the nonwoven basalt fibers (NBF), which is environmentally friendly, water resistance, non-toxicity and low cost. The effect of various parameters on the adsorption performance of NGN, such as pH value, boron concentration, contact time and different ion concentrations, was investigated. The optimum adsorption capacity of 1.64 mmol/g occurred in the vicinity of pH 9.0, and NGN possessed the fast adsorption equilibrium of boron within 40 min. In the presence of competing ions, Na + and Li + had little effect on the adsorption of boron, while Mg 2+ promoted the adsorption of boron. The features of NGN adsorption process were assessed by Langmuir and Freundlich adsorption isotherms . The experimental data of NGN boron adsorbent was well described by using the pseudo-second-order kinetic model. In addition, after eight times adsorption-desorption cycles, NGN still featured excellent regeneration performance. All the experimental results illustrate that NGN could be efficiently utilized for separating and recycling boron from aqueous solutions.

Total flavonoid– and nerve growth factor–loaded gelatin–genipin hydrogel improves repair after spinal cord injury

Liang Yu,Xi Deshuang,Teng Yilin,Liu Pan,Feng Yanbing,Huang Qiumei,Gao Ming,Zong Shaohui

Export Rat nerve growth factor and total flavonoids from hawthorn leaf contribute to the recovery of neurological function after spinal cord injury, including traumatic, non-traumatic spinal cord injuries. However, it remains challenging to efficiently deliver nerve growth factor and total flavonoids from hawthorn leaf to spinal cord injury sites, ensure their sustained release, and minimize further damage. In the present study, we chose a biocompatible and biodegradable gelatin as the substrate, which was crosslinked with the natural biological crosslinker genipin to form a gelatin–genipin hydrogel carrier for the slow release of nerve growth factor and total flavonoids from hawthorn leaf in spinal cord injury sites. The prepared gelatin–genipin hydrogel had good injectable properties and photothermal effects. Furthermore, when the hydrogel with 2% genipin, 200 ng/mL nerve growth factor, and 320 μg/mL total flavonoids from hawthorn leaf was combined with near infrared irradiation, there was a slow release of total flavonoids from hawthorn leaf and nerve growth factor, reduced oxidative stress, an improved inflammatory microenvironment, and accelerated angiogenesis and axonal regeneration via inhibition of the nuclear factor kappa-B signaling pathway, thereby promoting recovery from spinal cord injury. Collectively, our results indicate that this new hydrogel may improve the prognosis of spinal cord injury, and may represent a new strategy for treating spinal cord injury.

Biomaterial gels drive endogenous neurogenesis to support functional recovery after adult traumatic cortical injury

Wu Jinting,Hao Peng,Duan Hongmei,Zhao Wen,Gao Yudan,Cui Zhiqiang,Yang Zhaoyang,Li Xiaoguang

Activation and neuronal differentiation of endogenous neural stem/progenitor cells within the hippocampal neurogenic niche (a neurogenic region) of the adult mammalian brain are considered potential bases for restoration of cognitive and memory functions. However, following traumatic cortical brain injury (i.e., injuries in a non-neurogenic region), the local microenvironment becomes hostile due to the presence of inhibitory factors such as inflammation, edema, and hypoxic–ischemic stress. These conditions hinder the migration and subsequent neuronal differentiation of activated neural stem/progenitor cells into the injured cortex. Numerous recent studies have confirmed that biomaterials can improve the microenvironment of the injured area and promote nerve regeneration. In this study, we report the findings obtained by injecting a biodegradable chitosan–gelatin–collagen gel into the cortical lesion area. We observed that the chitosan–gelatin–collagen gel established a permissive microenvironment for neural repair by suppressing inflammation and scar infiltration while promoting neovascularization. Thus, it enabled activated endogenous “seed” cells, i.e., neural stem/progenitor cells, to migrate into the cortical lesion area and generate new neurons and thereby facilitate functional recovery. These findings represent a novel biomaterial-based therapeutic strategy for the treatment of traumatic cortical brain injury, stroke, and other neurological disorders.

Icariside II Suppresses Ferroptosis to Protect Against MPP+-Induced Parkinson's Disease through Keap1/Nrf2/GPX4 Signaling

Fan Wenbo,Zhou Jianwu

Parkinson's disease (PD) is recognized as a degenerative and debilitating neurodegenerative disorder. The novel protective role of icariside II (ICS II) as a plant-derived flavonoid compound in neurodegenerative diseases has aroused much attention. Herein, the definite impacts of ICS II on the process of PD and the relevant action mechanism were studied. Human neuroblastoma SK-N-SH cells were challenged with 1-methyl-4-phenylpyridinium ion (MPP + ) to construct the PD cell model. MTT assay and flow cytometry analysis, respectively, appraised cell viability and apoptosis. Caspase 3 Activity Assay examined caspase 3 activity. Corresponding kits examined oxidative stress levels. BODIPY 581/591 C11 assay evaluated lipid reactive oxygen species. Iron Assay Kit assessed iron content. Western blot tested the expression of apoptosis-, ferroptosis- and Kelch-like ECH-associated protein 1 (Keap1)/nuclear factor erythroid 2-related factor 2 (Nrf2)/glutathione peroxidase 4 (GPX4) signaling-associated proteins. Molecular docking verified the binding of ICS II with Keap1. The existing experimental results unveiled that ICS II elevated the viability whereas reduced the apoptosis, oxidative stress, and ferroptosis in MPP + -treated SK-N-SH cells in a concentration-dependent manner. Furthermore, ICS II declined Keap1 expression while raised Nrf2, heme oxygenase 1, and GPX4 expression. In addition, ICS II had a strong binding with Keap1 and Nrf2 inhibitor ML385 partially abolished the suppressive role of ICS II in MPP + -triggered apoptosis, oxidative stress, and ferroptosis in SK-N-SH cells. To summarize, ICS II might inhibit apoptosis, oxidative stress, and ferroptosis in the MPP + -stimulated PD cell model, which might be due to the activation of Keap1/Nrf2/GPX4 signaling.
