This paper reported a facile method for fabricating CdTe quantum dots decorated fluorescent mesoporous silica nanoparticles containing poly(p-phenylenevinylene) by electrostatic interaction in aqueous solution. The resulted fluorescent hybrid nanomaterials showed the dual-emission centered at 500 nm and 717 nm, which can be used as a fluorescent probe to detect Cu 2+ in water through fluorescence resonance energy transfer. Cu 2+ can be captured by the amino groups of the polyethyleneimine to form an absorbent complex, resulting in a strong quenching of red-emission from CdTe quantum dots on the surface of nanoparticles via energy transfer. However, the green-emission of poly(p-phenylenevinylene) remained. Therefore the addition of Cu 2+ induced the fluorescent evolution of the nanoprobe from red to green. The nanoprobe provided an efficient platform for the sensing of Cu 2+ with a detection limit of 31.2 nM. Compared with pure quantum dots, the nanoprobe can distinguish Cu 2+ from Hg 2+ .

In this work, a versatile cellulose nanocrystal‑carbon dots (CNC-CDs) architecture is prepared and applied in environmental remediation and biological sensing. The citric acid (CA) is employed to extract CNCs and in-situ synthesize blue-emissive CDs. The carboxyl-rich surface of CNC-CDs enables the chelation of Hg 2+ and fluorescence quenching of CDs, realizing the detection and adsorption of Hg 2+ with a film comprised of the architecture. The limit of detection (LOD) is measured as 12.8 nM with satisfying retention efficiency. The versatility of CNC-CDs is demonstrated by the fabrication of a fluorescence resonance energy transfer (FRET) sensor using m-tetrakis(4-sulfonatophenyl)porphine (TSPP) as FRET acceptor and Fe 3+ receptor. Fluorescence of the sensor is responsive to Fe 3+ in a ratiometric manner, which significantly contributes to the visualized and sensitive Fe 3+ detection (6.9 nM). In combination with excellent biocompatibility, the FRET sensor is capable of imaging intracellular Fe 3+ .

A novel far-red fluorescence resonance energy transfer (FRET) fluorescent probe for ratiometric detection of l -cysteine ( l -Cys) has been designed. The system was established a FRET assembly by positively charged carbon dots (CDs) and negatively charged N -acetyl- l -cysteine capped gold nanoparticles (NAC-AuNPs). The fluorescence of CDs at 539 nm could be effectively quenched in the presence of NAC-AuNPs owing to FRET process, while the emission of NAC-AuNPs at 630 nm was appeared. Subsequently, the interactions between l -Cys and NAC-AuNPs resulted in the decreased emission intensity of NAC-AuNPs, but the emission intensity of CDs kept almost constant due to the continuous FRET efficiency. The ratio of emission intensities at 539 and 630 nm ( I 539 / I 630 ) exhibited a linear correlation to the l -Cys concentration in the range of 1.0–110 μM with the detection limit of 0.16 μM. Moreover, this far-red ratiometric sensor also revealed excellent selectivity toward l -Cys over other amino acids , which showed very high potential in the practical application for diagnosing of cysteine-related disease.

Phosphate is not only an important indicator for aquatic ecosystems, but also plays vital roles in biosystems. A new strategy for ultrasensitive and selective detection of phosphate is fabricated based on a new insight found in this paper, in which a lower concentration of surfactant sodium dodecylbenzenesulfonate (SDBS) can greatly induce fluorescence resonance energy transfer (FRET) from ciprofloxacin (CIP) to Eu3+ in the CIP-Eu3+ complex. Surfactant SDBS does not act as a sensitizer for enhancing the fluorescence intensity of the system, but acts as a sensitizer of FRET and makes the native fluorescence of CIP quenched completely (switch off). Eu3+ ions can coordinate with the oxygen-donor atoms of phosphate, which weakens FRET from CIP to Eu3+ and results in the fluorescence recovery of CIP (turn on). The multicomplex of the CIP-Eu3+-phosphate has more sensitive fluorescent response than that of the reported coordination nanoparticle-based fluorescent probes. The LOD (S/N = 3) of this sensing system can attain 4.3 nM. The possible interferential substances existing in environmental samples, such as 17 common metal ions, 11 anions, and fulvic acid investigated, do not interfere with the phosphate detection. This sensing system has been successfully applied for phosphate detection in environmental samples such as wastewater, surface water, and atmospheric particulates. This work not only develops a fluorescent probe for the phosphate detection, but also provides a new strategy for designing fluorescent probes based on FRET or coordination nanoparticles.

Fluorescence resonance energy transfer (FRET) is a kind of energy transfer mechanism depending on the distance between donor and acceptor, which exhibited potential application in biosensors. In this study, an efficient fluorescence “turn-on” strategy for the detection of glutathione (GSH) has been established based on FRET between nitrogen and sulfur dual-doped carbon dots (N,S-CDs) and gold nanoparticles (Au NPs). A novel N,S-CDs was synthesized by a one-pot hydrothermal treatment of 3-aminothiophenol, which possessed excellent fluorescence property with the maximum emission wavelength of 530 nm. Then, the as-prepared N,S-CDs served as energy donor to transfer energy to Au NPs via FRET process, resulting in fluorescence quenching of N,S-CDs. However, the fluorescence of N,S-CDs was recovered efficiently by adding GSH into the mixture solution of N,S-CDs and Au NPs. Therefore, the FRET assembly of N,S-CDs and Au NPs was used as a fluorescence probe for the “turn-on” sensing GSH with the linear range from 3.8 to 415.1 μM and the limit detection of 0.21 μM. This nanosensor platform was employed to monitor GSH in serum samples with satisfying results.

A novel fluorescence resonance energy transfer (FRET)-based ratiometric nanosensor is reported for toxic Hg 2+ ions detection. Poly(p-phenylenevinylene) (PPV) chain was encapsulated into the pores of mesoporous silica nanoparticles (MSNs) by ion-exchange and in situ polymerization , and used as the energy-transfer donor. A spirolactam rhodamine-B derivative (SRhB), as an ion recognition element and energy-transfer acceptor, was embedded in the pores of MSNs to form PPV@MSN@SRhB as the nanosensor . The presence of Hg 2+ in the dispersion solution of nanoparticles can induce the ring-opening reaction of the SRhB moieties and lead to the occurrence of FRET process , affording the nanoparticle system a ratiometric sensor for Hg 2+ ions. The nanoparticle sensor can selectively detect Hg 2+ ions with a detection limit of 200 nM (ca. 40 ppb). It has been found that the fabricated FRET-based scaffold with the conjugated polymer as a donor exhibited high environmental stability and was more preferred for the accurate ratiometric sensing . Moreover, the FRET-based ratiometric nanosensor can be applicable in a relatively wide pH range (pH = 5–8) in solution.

Herein, a universal fluorescent biosensor was developed for detecting Mycobacterium Tuberculosis (MTB) specific insertion sequence IS6110 gene fragment based on Förster resonance energy transfer (FRET) strategy. CdTe quantum dots (QDs), with excellent luminous performance, were used to label single-stranded DNA (ssDNA) as fluorescence donor (QDs-DNA), in which the ssDNA was complementary to the IS6110 gene fragment. A new type of two-dimensional metal-organic framework (Cu-TCPP) was served as an acceptor. The Cu-TCPP exhibited a higher affinity towards ssDNA than double-stranded DNA (dsDNA). In the absence of targets, the fluorescence of QDs-DNA was quenched – due to the π-π stacking interactions between Cu-TCPP and ssDNA. Otherwise, QDs-DNA hybridized with the target to form a double helix and the fluorescence maintained in a target-concentration dependent manner. Excess QDs-DNA would be quenched and produced negligible background signal. The fluorescent sensor possessed a linear range from 0.05 nM to 1.0 nM with a low detection limit of 35 pM. Furthermore, we successfully applied this biosensing system to detect clinical sputum samples. This method displayed high sensitivity, specificity and great potentials in the early diagnosis of Tuberculosis.

Fluorescence resonance energy transfer (FRET) is often applied to construct fluorescent probes for acquiring high selectivity and sensitivity. According to the FRET theory, a homodimer composed of two identical fluorophores with a small Stokes shift has only weak fluorescence due to homo-FRET between fluorophores, and the fluorescence could be recovered after the destruction of the homodimer. In this study, we designed and synthesized a homodimer fluorescent probe, namely 1,3,5,7-tetramethyl-8-(4′-phenylthiophenol)-boron difluoride-dipyrrole methane dimer (D-TMSPB), based on this turn-on strategy. In D-TMSPB, the disulfide moiety was selected as the response moiety of biothiols, and BODIPY fluorophore was chosen as both donor and acceptor in FRET due to the ultra-small Stokes shifts and obvious overlap of its excitation/emission peak. D-TMSPB exhibited only weak fluorescence. After selective reaction with biothiols, FRET was destroyed and the derivative exhibited strong fluorescence at 514 nm with the limit of detection of about 0.15 μM for GSH. Notably, the derivative of biothiols shows remarkable fluorescence only in acidic conditions, which accords with the internal environment of lysosome. Thus, D-TMSPB was applied to image the biothiols of lysosome in living cells. The turn-on fluorescence of D-TMSPB indicated that homo-FRET is a practical strategy to design turn-on fluorescent probes, particularly for the sensing mechanism based on leaving groups.

The development of theragnostic nanosystems integrating FRET (fluorescence resonance energy transfer) imaging and chemodynamic therapy (CDT) for accurate diagnosis and effective treatment of lung tumors is still a big challenge. Herein, a peptide-assembled 3D DNAzyme motor nanodevice is engineered for a self-powered FRET amplifier profiling human neutrophil elastase (HNE) and self-supplied H 2 O 2 enhancing CDT. The nanodevice is prepared by depositing AuNPs on ZIF-8, in which ZIF-8 co-loaded the lysosomal targeting peptide-modified copper peroxides (PCPs) and hairpins (H1, H2, and H3), AuNPs are co-labeled by DNAzyme-peptide (DP) conjugate and H3. In the tumor micro-environment, HNE driven 3D DNAzyme walker followed by an exponential amplification constructed by a synergistic cross-activation between hybridization chain reaction and DNAzyme, generating a self-powered FRET amplifier. The FRET amplifier specifically measures HNE with a sensitivity of 0.026 pM, and successfully images exogenous HNE in living cells and monitors HNE in mouse models. Moreover, the PCPs can target lysosomes, reducing lysosome escape. The self-supplying H 2 O 2 undertaken by PCPs improves the Cu (II)-catalyzed Fenton-like reaction, effectively causing cell apoptosis to inhibit tumor growth. Significantly, the nanodevice successfully screens inhibitors and discriminates the HNE level in normal and lung cancer tissues, suggesting that the nanodevice provides an effective tool for the diagnosis and treatment of lung tumors.

In this paper, we report a novel strategy for highly sensitive, rapid, and selective detection of Salmonella Enteritidis using two-color quantum dots based on fluorescence resonance energy transfer (FRET) and fluoroimmunoassay technology. In this FRET system, two-color QDs were adopted as donors and acceptors simultaneously to improve its sensitivity and accuracy. The fluorescence intensity of QDs receptors decreased linearly with the increasing concentrations of S. Enteritidis from 75 to 5 × 10 5 CFU/mL due to FRET system and fluoroimmunoassay reaction. The limit of detection of S. Enteritidis in this method was 10 CFU/mL without sample enrichment within 1–2 h. This method has been applied to the analysis S. Enteritidis of synthetic samples and eggshell samples. The QDs receptors even can potentially be developed into a sensor for quantitative or qualitative determination of S. Enteritidis in the further.

The development of switchable white-light emitters is extremely challenging but urgently needed for various applications. We describe a class of switchable fluorescent polymeric nanoparticles (SFPNs) that can alternate their emission color among stable white, cyan, or pink. These SFPNs consist of a dye with cyan-light emission and a spiropyran (SP) derivative with photochromism. When the SP derivative is transferred to its merocyanine state, fluorescence resonance energy transfer (FRET) occurs from the cyan dye to the merocyanine one. In a rational design of transfer efficiency, a stable white-light emission is achieved. This white-light emission can be switched to the cyan-light one upon photoirradiation or pink-light one by solvent-induced swelling. Such multi-color fluorescent alternation is reversible and therefore allows for data encryption. Because of the facile synthesis, excellent long-term stability, fast photo-responsibility, high-contrast fluorescence, and outstanding switchability of these nanoparticles, we envision that they should have great potential in various applications such as high-resolution imaging, white luminescence, and data encryption.

A sensitive fluorescence resonance energy transfer (FRET) biosensor is proposed to detect 8-hydroxy-2′-deoxyguanosine (8-OHdG), which is a typical DNA oxidation damage product excreted in human urine. The FRET biosensor was based on carbon dots (CDs)-modified nanoporous alumina membrane with CDs as fluorescence donors. Gold nanoparticles were encapsulated in zeolitic imidazolate framework-8 to form Au@ZIF-8 nanoparticles as signal quenchers. CDs and Au@ZIF-8 nanoparticles were biofunctionalized by 8-OHdG antibody. The capture of 8-OHdG on the membrane substrates can bring Au@ZIF-8 nanoparticles closely to CDs. With 350 nm excitation, the fluorescence of CDs was quenched by Au@ZIF-8 nanoparticles and FRET effect occurred. The quenching efficiency was analyzed. The limit of detection (LOD) was 0.31 nM. Interference experiments of the FRET biosensor showed good specificity for 8-OHdG detection. The biosensor could detect urinary 8-OHdG sensitively and selectively with simple sample pretreatment processes. It shows applicability for detecting biomarkers of DNA damage in urine or other biological fluids. Keywords:nanoporous alumina membrane;fluorescence resonance energy transfer;Au@ZIF-8 nanoparticles;DNA oxidation damage;8-OHdG detection
