Bioinspired amino acid-functionalized carbon dots nanozymes with enhanced peroxidase-like activity for sensitive detection of GSH in food

Xiaoqing Song,Xiaoye Wen,Yongfei Huang,Zhefeng Fan

The development of nanozymes that exhibit high catalytic activity remains challenging. Herein, carbon dots (CDs) functionalized by different acidic, neutral and basic amino acids were synthesized. The CDs modified with the L-aspartic acid (Asp-Fe-CDs) showed bright blue fluorescence and remarkable peroxidase-like activity. The specific activity of Asp-Fe-CDs was greatly enhanced around 4 times compared to Fe-CDs without the L-aspartic acid. The excellent enzymatic activity of Asp-Fe-CDs was ascribed to carboxyl group of L-aspartic acid in the side chain. Given the advantages of Asp-Fe-CDs, a dual-mode sensor was developed for GSH detection. The limits of detection were 0.189 μM and 0.296 μM for fluorimetry and colorimetry, respectively. Moreover, the sensor was used to detect GSH in food with satisfactory results. In summary, this research not only proposes a new strategy for the design of carbon dots nanozymes but also develops a multimode sensing platform for GSH detection.

Multifunctional Fluorescent Probe for Simultaneous Detection of ATP, Cys, Hcy, and GSH: Advancing Insights into Epilepsy and Liver Injury

Ting Yu,Yang Li,Jing Li,Yabing Gan,Zhengze Long,Yun Deng,Youyu Zhang,Haitao Li,Peng Yin,Shouzhuo Yao

Adenosine triphosphate (ATP) is a critical intracellular energy currency that plays a key role in various cellular processes and is closely associated with numerous diseases. Similarly, biothiols such as glutathione (GSH), cysteine (Cys), and homocysteine (Hcy) are integral to many physiological and pathological processes due to their strong redox properties. Simultaneous discrimination and detection of ATP and biothiols offer valuable insights into the pathogenesis of conditions such as epilepsy and liver injury. This study introduces the first fluorescent probe, BCR , designed for multifunctional detection of ATP, GSH, Hcy, and Cys. With outstanding optical properties, excellent biocompatibility, high selectivity, and superior sensitivity, probe BCR enables effective imaging of ATP and biothiol dynamics in vivo. Moreover, probe BCR successfully visualizes changes in ATP, GSH, Hcy, and Cys levels in a PTZ-induced epileptic zebrafish model and an APAP-induced mouse liver injury tissue section model. These findings underscore the significant potential of probe BCR for early disease diagnosis and therapeutic applications.

Visualizing GSH-ONOO− Redox and Tracking Lesion-Remedy of Acute Kidney Oxidative Injury Based on a Dual-Site Chemosensor

Meili Yang,Zuzhe Kang,Xinpei Zhong,Sirui Bai,Dong-En Wang,Qin Tu,Sheng Chen,Jinyi Wang,Mao-Sen Yuan

Drug-induced acute kidney injury (DIAKI) is strongly associated with the accumulation of peroxynitrite (ONOO − ) and the depletion of glutathione (GSH), both of which contribute to increased morbidity and mortality. Owing to the lack of an effective method for the real-time monitoring of nephrotoxicity, it remains a diagnostic and therapeutic challenge and is considered a significant issue in clinical practice. Herein, a dual-site fluorescent chemosensor, BPS is developed , which enables the simultaneous real-time monitoring of ONOO − and GSH through distinct channels (ONOO − : λ ex = 410 nm, λ em = 548 nm; GSH: λ ex = 490 nm, λ em = 660 nm) without spectral crosstalk. The sensor displays remarkable sensitivity toward ONOO − and GSH, with low detection limits (181 n m and 2.42 µ m , respectively), exceptional specificity, and anti-interference effects. BPS is used to monitor ONOO − /GSH in real time to track the lesion of cisplatin-stimulated DIAKI and the following remedy process in mice and HK-2 cells for the first time. BPS provides a feasible tool for investigating oxidative stress-related DIAKI disease processes and further exploring some physiological mechanisms. Moreover, this study also offers a strategy for visualizing, modulating, and controlling the intracellular GSH/ONOO − redox.

Facile synthesis of NS-doped carbon dots as sensitive “ON-OFF-ON” fluorescent sensor for Cu2+ and GSH detection

Huan Wang,Wencheng Mu,Siying Wang,Lin Shi,Tianfeng Ma,Yongchang Lu

In this paper, a novel nitrogen and sulfur co-doped carbon quantum dots (NS-CQDs) were successfully prepared by a dehydration exothermic carbonization method. The NS-CQDs exhibited uniform size distribution, splendid photostability, and bright fluorescence emission with a fluorescence quantum yield of 24.1 %. It was found that Cu 2+ could quench the fluorescence at 467 nm based on the static quenching effect when Cu 2+ was added to the NS-CQDs. At this time, the fluorescence sensor changed from the “ON” state to the “OFF” state. When glutathione (GSH) was further introduced into the NS-CQDs/Cu 2+ system, the fluorescence intensity of NS-CQDs was amazingly restored through the coordination reaction between GSH and Cu 2+ . The fluorescence sensor changed from the “OFF” state to the “ON” state. Therefore, NS-CQDs as an “ON-OFF-ON” fluorescence sensor was designed for sequential detection of Cu 2+ and GSH. Furthermore, this study successfully demonstrated the sensor’s ability to selectively detect Cu 2+ and GSH within a wide concentration range. Specifically, the detection range for Cu 2+ was 0.1 μM−200.0 μM with a detection limit of 0.07 μM, while the range for GSH was 0.6 μM−180.0 μM with a detection limit of 0.1 μM. Most importantly, the NS-CQDs nanosensor could reliably monitor Cu 2+ and GSH levels in human serum samples, with significant potential for practical applications.

Enzyme/GSH/pH-responsive hyaluronic acid grafted porous silica nanocarriers bearing Ag2S QDs for fluorescence imaging and combined therapy

Jun Hou,Yanbao Zhao,Lei Sun,Xueyan Zou

Hyaluronic acid (HA) is a naturally polysaccharide that has been used for drug delivery, but is limited by low drug loading capacity and drug leakage in circulation. To improve drug delivery efficient, HA modified porous silica (pSiO 2 ) nanocarriers were successfully prepared for drug delivery and combining therapy. pSiO 2 nanocarriers have stable porous structure and high loading capacity, and pSiO 2 /HA nanocarriers would possess advantages of HA-based carriers and pSiO 2 nanoparticles . Herein, pSiO 2 nanocarriers were prepared by two-phase process, followed by embedding Ag 2 S QDs in the pore walls of pSiO 2 carriers, which render the carriers photothermal effect. pSiO 2 nanocarriers have size of 30 nm, large channels, and high loading capacity (29.3 %). To graft HA, a sensitive linker with alkyl amine and disulfide bond was conjugated on the surface of Ag 2 S/pSiO 2 nanocarriers by three-step reaction. After loading doxorubicin (DOX), HA was grafted via sensitive linker onto the surface of Ag 2 S/pSiO 2 carriers via the formation of amide bonds to seal the loaded drugs. The interaction between HA and CD44 confers the carrier targeting ability to cancer cells. HA coating can be degraded by hyaluronidase resulting in the release of internal cargo. The Ag 2 S/pSiO 2 /HA nanocarriers performs responsive drug release and combining photothermal chemotherapy.

Watson–Crick Base Pairing-Inspired Laser/GSH Activatable miRNA-Coordination Polymer Nanoplexes for Combined Cancer Chemo-Immuno-Photothermal Therapy

Jiulong Zhang,Xiaoyan Sun,Xiufeng Zhao,Lin Liu,Xin Cheng,Chunrong Yang,Haiyang Hu,Mingxi Qiao,Dawei Chen,Xiuli Zhao

The tumor immunosuppressive microenvironment (TIM) greatly hindered the efficacy of cancer immunotherapy. Overexpressed indoleamine 2,3-dioxygenase-1 (IDO1) in tumor tissues plays a vital role in TIM generation, and downregulation of IDO1 expression may reverse TIM. Inspired by the Watson–Crick base-pairing rule, a versatile noncationic miRNA vector (miDAC@PDA) is developed for cancer immunotherapy. Doxorubicin (DOX), adenosine triphosphate (ATP), and copper ions (Cu2+) are coassembled into coordination polymer nanoparticles (DAC) and bind miRNA via the hydrogen bond interaction (miDAC) between adenine residues (ATP) and uracil residues (miRNA). Polydopamine (PDA) is deposited onto the surface of miDAC for photothermal therapy. miDAC@PDA can efficiently accumulate into tumor tissues for cellular uptake. Under laser irradiation and high intracellular GSH levels, the PDA shell of miDAC@PDA can dissociate from miDAC for miRNA release due to local hyperthermia. Cu2+-mediated GSH consumption and intracellular ATP release can amplify the DOX-based immunogenic cell death (ICD) cascade, together with miR-448-mediated IDO1 inhibition, and these versatile nanoplexes will not only restrain primary tumor growth but also display a remarkable abscopal effect on distant tumors. Collectively, our study provides a unique strategy for intracellular gene delivery and an inspirational approach for multimechanism cancer management.

Patulin disrupts SLC7A11-cystine-cysteine-GSH antioxidant system and promotes renal cell ferroptosis both in vitro and in vivo

Hui Chen,Lixing Cao,Kai Han,Han Zhang,Jinling Cui,Xuan Ma,Shuang Zhao,Chong Zhao,Shutao Yin,Lihong Fan,Hongbo Hu

Patulin (PAT) is a common food-borne mycotoxin with diverse toxic effects including nephrotoxicity . The induction of oxidative stress is suggested to be a key mechanism contributed to toxicities of PAT. Reduced glutathione (GSH), a sulfhydryl-containing tripeptide , is a key reason for PAT-mediated oxidative stress. Cystine/glutamate antiporter (system x c − )-mediated cystine uptake plays a critical role in maintaining redox balance via promoting GSH biosynthesis. In this study, we addressed if GSH reduction by PAT was associated with inhibition of system x c − -mediated GSH biosynthesis. Results showed that PAT significantly decreased activity of SLC7A11, a core subunit of system x c − , through activating AMPK-mediated formation of beclin1-SLC7A11 complex. Furthermore, PAT promoted ferroptosis induced by a known ferroptosis inducer RSL3 in normal renal cells, and exacerbated folic acid-induced nephrotoxicity in a mouse model of acute kidney injury . The findings of the present study provide new insights into PAT-induced kidney toxicity, and implicate that patients with ferroptosis-associated diseases maybe more susceptible to PAT.

Microwave-assisted sonochemical synthesis of Cu and Mn doped GSH–ZnS polypeptide quantum dots and their enhanced photoelectrochemical properties

Jian-Jun Shi,Lei Gong,Yan-Hong Zhang,Ping Yang,Jie He

A series of Cu and Mn doped glutathione (GSH)–ZnS polypeptide quantum dots (QDs) were successfully prepared by a facile one-pot microwave-assisted sonochemical synthesis approach. Herein, GSH acted as a sulfide source and dispersant, as well as a reducing agent to form polypeptide QDs. The products were characterized by high-resolution transmission electron microscope (HRTEM) and energy dispersive spectroscopy (EDS). Results show that the size of the polypeptides QDs was 2–3 nm. In Fourier transform infrared (FT-IR) spectra, the splitting of S–H bonds and substitution by S–Zn can be noticed. The bonding interaction between Cu, Mn and Zn ions toward S was further confirmed from X-ray photoelectron spectroscopy (XPS) analysis. The results confirmed the partial Cu(Mn) substitution of Zn. Fluorescence (FL) spectra and UV-vis absorption spectra were used to investigate the optical properties. It was found that the doping of Cu and Mn could decrease the band gap of GSH–ZnS from 3.67 eV to 2.86 eV. The photoelectrochemical (PEC) properties of GSH–ZnS were enhanced due to the doping of Cu and Mn which has excellent chemical stability. The possible photoelectrochemical reaction mechanism is discussed.

GSH-Depleting and H2O2 Self-Supplying Calcium Peroxide-Based Nanoplatforms for Efficient Bacterial Eradication via Photothermal-Enhanced Chemodynamic Therapy

Fuqiang Shi,Jie Chen,Lesan Yan,Jing Tu

Chemodynamic therapy (CDT), an innovative approach for treating bacterial infections, has garnered significant attention due to its ability to generate hydroxyl radicals (•OH) via Fenton/Fenton-like reactions. However, the effectiveness of CDT is considerably hindered by the limited availability of endogenous hydrogen peroxide (H2O2) and the overexpression of glutathione (GSH) within the infection microenvironment. To address these limitations, a multifunctional nanoplatform with self-supplying H2O2, GSH-depletion properties, and photothermal properties was developed through a straightforward and mild strategy. This platform employs calcium peroxide (CaO2) as the core, coated with silica (SiO2) to enhance stability and further modified with a Cu(II)-doped polydopamine (PDA) layer, forming a core–shell structured CaO2@SiO2@PDA-Cu (CSPC). The Cu(II) released by CSPC, combined with the H2O2 produced from CaO2 degradation, participates in a Fenton-like reaction to generate toxic •OH radicals. Additionally, Cu(II)-mediated redox reactions deplete overexpressed GSH, thereby enhancing CDT efficacy. Upon coordination with Cu(II), the photothermal properties of PDA are significantly enhanced, achieving a photothermal conversion efficiency of up to 43%. The hyperthermia induced by photothermal therapy (PTT) further increases •OH production, augmenting CDT. The CSPC nanomaterials demonstrated outstanding synergistic photothermal bactericidal activity against Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) at 60 μg/mL, achieving complete eradication. Moreover, CSPC eliminated 65.90 ± 3.46% of the S. aureus biofilm under near-infrared (NIR) irradiation. In vivo experiments demonstrated that CSPC treatment effectively eradicated bacteria, with a bacterial survival rate of 6.56 ± 3.28%, and accelerated wound healing, reducing the relative wound size to 7.0 ± 2.6%. Therefore, this study successfully developed versatile nanomaterials that significantly enhance the PTT/CDT dual-mode antibacterial performance.

Trimetazidine attenuates Ischemia/Reperfusion-Induced myocardial ferroptosis by modulating the Sirt3/Nrf2-GSH system and reducing Oxidative/Nitrative stress

Mingyue Tan,Yunfei Yin,Weixiang Chen,Jun Zhang,Yifeng Jin,Yue Zhang,Lei Zhang,Tingbo Jiang,Bin Jiang,Hongxia Li

Ferroptosis is a newly defined mode of cellular demise. The increasing investigation supports that ferroptosis is a crucial factor in the complex mechanisms of myocardial ischemia–reperfusion (I/R) injury. Hence, targeting ferroptosis is a novel strategy for treating myocardial injury. Although evidence suggests that trimetazidine (TMZ) is potentially efficacious against myocardial injury, the exact mechanism of this efficacy is yet to be fully elucidated. This study aimed to determine whether TMZ can act as a ferroptosis resistor and affect I/R-mediated myocardial injury. To this end, researchers have constructed in vitro and in vivo models of I/R using H9C2 cardiomyocytes, primary cardiomyocytes, and SD rats. Here, I/R mediated the onset of ferroptosis in vitro and in vivo, as reflected by excessive iron aggregation, GSH depletion, and the increase in lipid peroxidation. TMZ largely reversed this alteration and attenuated cardiomyocyte injury. Mechanistically, we found that TMZ upregulated the expression of Sirt3. Therefore, we used si-Sirt3 and 3-TYP to interfere with Sirt3 action in vitro and in vivo, respectively. Both si-Sirt3 and 3-TYP partly mitigated the inhibitory effect of TMZ on I/R-mediated ferroptosis and upregulated the expression of Nrf2 and its downstream target, GPX4-SLC7A11. These results indicate that TMZ attenuates I/R-mediated ferroptosis by activating the Sirt3-Nrf2/GPX4/SLC7A11 signaling pathway. Our study offers insights into the mechanism underlying the cardioprotective benefits of TMZ and establishes a groundwork for expanding its potential applications.

Virus-Inspired Biodegradable Tetrasulfide-Bridged Mesoporous Organosilica with GSH Depletion for Fluorescence Imaging-Guided Sonodynamic Chemotherapy of Glioblastoma Multiforme

Feng Wei,Yanling Lin,Rujun Pan,Yilong Peng,E Chen,Junlong Kang,Jiang Zhu,Jiayin Wang,Baofang Wu,Wenwen Shen,Jinyan Lin,Hongzhi Gao,Xinhua Tian

Glioblastoma multiforme (GBM), a highly prevalent and lethal form of malignant tumor, is typically treated with Temozolomide (TMZ), a chemotherapeutic agent. Nevertheless, the effectiveness of TMZ is hampered by inadequate cell entry, systemic adverse effects, and monotherapy constraints. Previous clinical studies have demonstrated that combination therapy can significantly enhance the therapeutic efficacy. Herein, we developed ultrasmall virus-inspired biodegradable tetrasulfide-bridged mesoporous organosilica coloaded with TMZ and indocyanine green (ICG) (designated as vMSTI) for fluorescence imaging-guided sonodynamic chemotherapy and glutathione (GSH) depletion, aiming to enhance the therapeutic efficiency of GBM. Once accumulated within the tumors, the vMSTI nanosystem efficiently entered tumor cells via “spike surface”-assisted endocytosis. Subsequently, intracellular overproduction of GSH within tumor cells triggered the degradation of vMSTI, resulting in the release of both TMZ and ICG, while simultaneously depleting intracellular GSH levels. Upon ultrasound (US) irradiation, the released ICG generated abundant reactive oxygen species (ROS) for sonodynamic therapy, which could be further potentiated by GSH depletion. Furthermore, released TMZ effectively elicited DNA damage to enable chemotherapy. Consequently, the vMSTI effectively triggered apoptosis, suppressing GBM growth under the guidance of fluorescence imaging. Our nanosystems offered a promising strategy for imaging-guided combination therapy for GBM.

Targeted Reduction-Responsive Nanovehicles for Photodynamic Therapy-Primed Immunotherapy in Melanoma

Chenqian Feng,Lingfeng Zhou,Bo Chen,Hui Li,Min Mu,Rangrang Fan,Haifeng Chen,Gang Guo

Melanoma, a common malignant skin tumor, faces challenges with multidrug resistance and high recurrence rates. Combining photodynamic therapy (PDT) and immunotherapy offers a promising personalized treatment approach. However, poor water solubility and significant side effects of photosensitizers and immune checkpoint inhibitors (ICIs) limit their application. Enhancing delivery efficiency while reducing adverse effects is crucial. Herein, we formulate BM@HSSC nanoparticles (NPs), which consist of a reduction-responsive hyaluronic acid (HA) backbone modified with photosensitizer Chlorin e6 (Ce6) and loaded with the programmed cell death-ligand 1 (PD-L1) inhibitor BMS-1. This system synergistically integrates PDT, immunogenic cell death (ICD), and immunotherapy for melanoma treatment. BM@HSSC NPs target and accumulate at the tumor site via the CD44 receptor. The disulfide bonds in the nanoparticles react with high GSH concentrations in tumor cells, rapidly releasing Ce6 and BMS-1. Under 660 nm laser irradiation, BM@HSSC NPs generate cytotoxic reactive oxygen species (ROS), inducing cell apoptosis and triggering ICD via. PDT Damage-associated molecular patterns (DAMPs) and tumor-associated antigens (TAAs) released from ICD promote dendritic cell (DC) maturation, enhancing antigen presentation and activating cytotoxic T lymphocytes (CTLs). Meanwhile, BMS-1 blocks the programmed cell death-1 (PD-1)/PD-L1 pathway, countering the immunosuppressive tumor microenvironment and inhibiting tumor cell immune escape. This strategy amplifies antitumor immune responses by enhancing immunogenicity and synergizing with ICIs, resulting in robust antitumor efficacy.
