New class of high-entropy defect fluorite oxides RE2(Ce0.2Zr0.2Hf0.2Sn0.2Ti0.2)2O7 (RE = Y, Ho, Er, or Yb) as promising thermal barrier coatings

He Junjie,He Guo,Liu Jing,Tao Jingchao

High-entropy ceramics exhibit great application potential as thermal barrier coating (TBC) materials. Herein, a series of novel high-entropy ceramics with RE 2 (Ce 0.2 Zr 0.2 Hf 0.2 Sn 0.2 Ti 0.2 ) 2 O 7 (RE 2 HE 2 O 7 , RE = Y, Ho, Er, or Yb) compositions were fabricated via a solid-state reaction. X-ray diffraction (XRD) and energy dispersive spectrometry (EDS) mapping analyses confirmed that RE 2 HE 2 O 7 formed a single defect fluorite structure with uniform elemental distribution. The thermophysical properties of the RE 2 HE 2 O 7 ceramics were investigated systematically. The results show that RE 2 HE 2 O 7 ceramics have excellent high-temperature phase stability, high thermal expansion coefficients (10.3–11.7 × 10 −6 K -1 , 1200 ℃), and low thermal conductivities (1.10-1.37 W m -1 K -1 , 25 ℃). In addition, RE 2 HE 2 O 7 ceramics have a high Vickers hardness (13.7–15.0 GPa) and relatively low fracture toughness (1.14-1.27 MPa m 0.5 ). The outstanding properties of the RE 2 HE 2 O 7 ceramics indicate that they could be candidates for the next generation of TBC materials.

Core constituents and pharmacodynamically equivalent ratios of Polygonum capitatum against urinary tract infection: involvement of Nrf2/HO-1 and NF-κB pathways

Xian Dong Lv,Meiherinisa Maitituersun,Fei Li

This study established HPLC fingerprints of PCAE 70% and RLQ, supported by the 2020 edition of the Chinese Pharmacopeia and relevant literature, and performed network pharmacology prediction for the common constituents. • Multi-index regression analysis of uniform design data was conducted by partial least squares, by which seven active components were optimized and determined as six important active components, and their rationality was experimentally validated. • The entropy weight method was employed to unify heterogeneous data from in vitro and in vivo experiments, and the integrated important active component combination was subjected to in vivo activity validation through a mouse urinary tract infection model. • This study suggests that the active components centered on gallic acid, myricitrin, and quercitrin may constitute the major pharmacodynamic material basis of Polygonum capitatum against UTI, and preliminarily explored the underlying mechanism. Ethnopharmacological relevance As a cornerstone herb in the traditional medicine of the Miao ethnic group in Guizhou Province, Polygonum capitatum has long been used to treat diseases of the urinary system. Documented in Miao medical theory as cold in nature and bitter-pungent in taste, and as acting on the kidney and bladder meridians, it is traditionally used to clear heat, promote diuresis, relieve stranguria, and detoxify, thereby relieving pain. The aqueous extract of this herb is the sole component of Relinqing Granules (RLQ), a clinically widespread formulation. Despite its broad clinical application, the underlying material basis of its pharmacological activity remains to be fully elucidated. Materials and methods The target compounds were identified through literature investigation, and their presence in both the Polygonum capitatum 70% aqueous ethanol extract (PCAE 70%) and RLQ was subsequently verified. The common constituents shared by the two were identified by comparing the fingerprint chromatograms with reference standards, and network pharmacology was employed to predict the potential targets of these components. The constituents were then combined through uniform design to evaluate their antibacterial, anti-inflammatory, and antioxidant activities, optimized via partial least squares regression and the entropy weight method, and ultimately validated in a mouse model of UTI. Results A total of seven common constituents were identified: gallic acid, protocatechuic acid, catechin, myricitrin, quercitrin, quercetin, and kaempferol. Network pharmacology predicted that these constituents might exert anti-UTI effects through modulation of the PI3K-Akt signaling pathway. The optimal ratio combination demonstrated significant antibacterial activity against Escherichia coli and Staphylococcus aureus , while another optimal ratio enhanced catalase activity and modulated the expression of AKT-1/Nrf 2 / HO-1/NF-κB 3 in zebrafish. Following data integration using the entropy weight method, mouse validation results revealed decreased TNF-α/IL-1β levels and increased IL-10 levels, with superior efficacy compared to PCAE 70% and RLQ, without dose-dependent differences. The results implied that gallic acid, myricitrin, and quercitrin represent the potential principal active components of P. capitatum. Conclusion The optimized combination dominated by gallic acid, myricitrin, and quercitrin, identified from P. capitatum , exhibited antibacterial, anti-inflammatory, and antioxidant performance superior or equivalent to existing preparations, holding significant theoretical and practical value. Soy Isoflavones Protects Against Stroke by Inhibiting Keap1/NQO1/Nrf2/HO-1 Signaling Pathway: Network Pharmacology Analysis Combined with the Experimental Validation

Huiming Xue,Zhen Feng,Chang Jin,Yue Zhang,Yongxing Ai,Jing Wang,Meizhu Zheng,Dongfang Shi

Objectives: Ischemic stroke is a severe neurological disorder with high morbidity, mortality, and disability rates, posing a substantial burden on patients, families, and healthcare systems. Soy isoflavone (SI), a naturally occurring phytoestrogen, has demonstrated promising neuroprotective effects. This study aimed to evaluate the anti-stroke efficacy of SI and elucidate its underlying mechanisms through integrated phytochemical profiling, network pharmacology, and both in vitro and in vivo experimental validation. Methods: Active constituents of SI were extracted via reflux and identified using liquid chromatography–mass spectrometry (LC-MS). Network pharmacology was employed to predict therapeutic targets and signaling pathways. The neuroprotective effects of SI were first assessed in PC12 cells subjected to oxygen–glucose deprivation/reoxygenation (OGD/R) injury in vitro. For in vivo evaluation, transient cerebral ischemia–reperfusion injury was induced using the bilateral common carotid artery occlusion (BCCAO) model in adult male ICR rats (27.3 ± 1.8 g; 6–8 weeks old), obtained from the Shanghai Experimental Animal Center, Chinese Academy of Sciences. Forty-eight rats were randomly assigned into four groups (n= 12): sham, model (BCCAO), SI-treated (100 mg/kg, oral gavage for 5 days), and edaravone (EDA)-treated (10 mg/kg, i.p., positive control). All procedures were approved by the Institutional Animal Care and Use Committee of Changchun Normal University (Approval No. 2024003, 13 March 2024) and conducted in accordance with the NIH guidelines and ARRIVE 2.0 reporting standards. Results: In vitro, SI significantly enhanced PC12 cell viability from 57.23 ± 2.88% to 80.76 ± 4.43% following OGD/R. It also reduced intracellular Ca2+by 58.42%, lactate dehydrogenase (LDH) release by 37.67%, caspase-3 activity by 55.05%, and reactive oxygen species (ROS) levels by 74.13% (p< 0.05). A flow cytometry analysis revealed that OGD/R increased the apoptosis rate from 5.34% (control) to 30.85% (model group), which was significantly attenuated by SI treatment, especially in the 560 µg/mL group (20.00%), followed by the 140 and 280 µg/mL groups. In vivo, SI improved neurological scores from 8.3 ± 1.09 to 6.8 ± 1.68, reduced cerebral infarction volume by 18.49%, and alleviated brain edema by 10.42% (p< 0.05). SI also decreased malondialdehyde (MDA) and LDH levels by 31.15% and 39.46%, respectively, while increasing the activity of antioxidant enzymes: superoxide dismutase (SOD) by 11.70%, catalase (CAT) by 26.09%, and glutathione peroxidase (GSH-px) by 27.55% (p< 0.01). Scratch assay results showed that SI restored the impaired migratory ability of the OGD/R-treated PC12 cells, further supporting its role in cellular repair. A Western blot analysis demonstrated the upregulation of nuclear factor erythroid 2–related factor 2 (Nrf2), heme oxygenase-1 (HO-1), and NAD(P)H:quinone oxidoreductase 1 (NQO1) and the downregulation of Kelch-like, ECH-associated protein 1 (Keap1) in the cerebral ischemia–reperfusion model. Conclusions: These findings indicate that soy isoflavone confers significant neuroprotective effects against cerebral ischemia–reperfusion injury by enhancing endogenous antioxidant defense mechanisms, reducing oxidative stress, inhibiting apoptosis, and promoting cell migration. The protective effects are likely mediated through the activation of the Nrf2/Keap1 signaling pathway, supporting the therapeutic potential of SI in ischemic stroke treatment.

Modulation of the microstructure and electrical properties of LaAlO3 ceramics induced by doping with rare earth elements (Eu, Gd, Ho, and Tm)

Kai Ding,Wenye Deng,Jinyang Li,Ni Ai,Yan Xue,Xianghui Chen,Pengjun Zhao,Weiwei Meng,Aimin Chang,Yongxin Xie

This study synthesized La0.9X0.1AlO3 (X = Eu, Gd, Ho, Tm) and LaAlO3 ceramics by a solid-state method. The characterization of the ceramics revealed that the high degree of lattice distortion induced by variations in the size, mass, and electronegativity of rare-earth elements enhances high-temperature stability, leading to aging drift rates between 0.4% and 2.3% after 500 h at 1000 °C. Furthermore, the emergence of intrinsic defects and dislocations alters the charge conduction mode in LaAlO3, resulting in significant linearity at temperatures above 1000 K. This study further elucidates the transition in the charge conduction mechanism from thermally activated conduction to hopping conduction, which is driven primarily by oxygen vacancies and dislocations. DFT calculations support these findings, demonstrating a reduction in the band gap and increased carrier concentration. This work paves the way for the development of advanced NTC thermistors for reliable temperature monitoring in challenging environments.

Cinnamaldehyde Alleviates Bone Loss by Targeting Oxidative Stress and Mitochondrial Damage via the Nrf2/HO-1 Pathway in BMSCs and Ovariectomized Mice

Bing-hao Lin,Run-xun Ma,Jing-tao Wu,Shi-qi Du,Yi-yun Lv,Hao-nan Yu,Wei Zhang,Shu-ming Mao,Guang-yao Liu,Yi-tian Bu,Zi-hao Chen,Chen Jin,Zong-yi Wu,Lei Yang

Osteoporosis (OP) is typically brought on by disruption of bone homeostasis. Excessive oxidative stress and mitochondrial dysfunction are believed to be the primary mechanisms underlying this disorder. Therefore, in order to restore bone homeostasis effectively, targeted treatment of oxidative stress and mitochondrial dysfunction is necessary. Cinnamaldehyde (CIN), a small molecule that acts as an agonist for the nuclear factor erythroid 2-related factor (Nrf2), has been found to possess antiapoptotic, anti-inflammatory, and antioxidant properties. We found that CIN, while rescuing apoptosis, can also reduce the accumulation of reactive oxygen species (ROS) to improve mitochondrial dysfunction and thus restore the osteogenic differentiation potential of BMSCs disrupted by hydrogen peroxide (H2O2) exposure. The role of CIN was preliminarily considered to be a consequence of Nrf2/HO-1 axis activation. The ovariectomized mice model further demonstrated that CIN treatment ameliorated oxidative stress in vivo, partially reversing OVX-induced bone loss. This improvement was seen in the trabecular microarchitecture and bone biochemical indices. However, when ML385 was concurrently injected with CIN, the positive effects of CIN were largely blocked. In conclusion, this study sheds light on the intrinsic mechanisms by which CIN regulates BMSCs and highlights the potential therapeutic applications of these findings in the treatment of osteoporosis.

Ursolic acid ameliorates DNCB-induced atopic dermatitis-like symptoms in mice by regulating TLR4/NF-κB and Nrf2/HO-1 signaling pathways

Zhehuan Wang,Huiru Zhang,Caihong Qi,Hui Guo,Xiangyue Jiao,Jia Yan,Yifei Wang,Qiangsheng Li,Mingming Zhao,Xinhao Guo,Baoluo Wan,Xiaotian Li

Background Ursolic acid (UA) is a triterpenoid compound found in natural plants. It has been reported to have anti-inflammatory, antioxidant, and immunomodulatory properties. However, its role in atopic dermatitis (AD) is unknown. This study aimed to evaluate the therapeutic effect of UA in AD mice and explore the underlying mechanisms. Methods Balb/c mice were treated with 2, 4-dinitrochlorobenzene (DNCB) to induce AD-like lesions. During modeling and medication administration, dermatitis scores and ear thickness were measured. Subsequently, histopathological changes, levels of T helper cytokines, and oxidative stress markers levels were evaluated. Immunohistochemistry staining was used to assess changes in the expression of the nuclear factor of kappa B (NF-κB) and NF erythroid 2-related factor 2 (Nrf2). Furthermore, CCK8 assay, reactive oxygen species (ROS) assay, real-time PCR, and western blotting were employed to evaluate the effects of UA on ROS levels, inflammatory mediator production, and the NF-κB and Nrf2 pathways in TNF-α/IFN-γ-stimulated HaCaT cells. Results The results showed that UA significantly reduced dermatitis score and ear thickness, effectively inhibited skin proliferation and mast cell infiltration in AD mice, and decreased the expression level of T helper cytokines. Meanwhile, UA improved oxidative stress in AD mice by regulating lipid peroxidation and increasing the activity of antioxidant enzymes. In addition, UA inhibited ROS accumulation and chemokine secretion in TNF-α/IFN-γ-stimulated HaCaT cells. It might exert anti-dermatitis effects by inhibiting the TLR4/NF-κB pathway and activating the Nrf2/HO-1 pathway. Conclusion Taken together, our results suggest that UA may have potential therapeutic effects on AD and could be further studied as a promising drug for AD treatment.

Photocatalytic H2O2 production and removal of Cr (VI) via a novel Lu3NbO7: Yb, Ho/CQDs/AgInS2/In2S3 heterostructure with broad spectral response

Kailian Zhang,Man Zhou,Kai Yang,Changlin Yu,Ping Mu,Zhenzhen Yu,Kangqiang Lu,Weiya Huang,Wenxin Dai

The low-usage of solar energy and the sluggish separation efficiency of the photogenerated electrons/holes pairs are the obstacles in the practical application of photocatalysts. The integration of upconversion and Z-scheme heterojunction is expected to break the barriers to achieve the efficient charge separation and broaden near-infrared light absorption . Herein, an effective indirect Z scheme AgInS 2 /In 2 S 3 heterostructure with carbon quantum dots (CQDs, as the electron conduction medium) and Lu 3 NbO 7 :Yb, Ho (as upconversion function) has been successfully synthesized. Consequently, the Lu 3 NbO 7 : Yb, Ho/CQDs/AgInS 2 /In 2 S 3 heterostructure exhibited superior photocatalytic activities for Cr(VI) reduction and H 2 O 2 production, reducing 99.9% of Cr(VI)(20 ppm, 15 min) and 78.5% of Cr(VI) (40 ppm, 30 min) with visible light irradiation as well as 94.0% of Cr(VI) (20 ppm, 39 min) under NIR light irradiation. Simultaneously, the heterostructure could generate 902.9 μM H 2 O 2 for 5 h under visible light irradiation. The intensive photocatalytic properties could primarily be attributed to the boosted light absorption capacity, the improved solar-to-energy conversion by the remarkable upconversion capacity of Lu 3 NbO 7 : Yb, Ho/CQDs and the faster charge transfer through a Z-schematic pathway. This work is anticipated to open a novel “window” for designing the efficient photocatalysts by coupling of Lu 3 NbO 7 : Yb, Ho and CQDs.

Sol–gel combustion synthesis and antiferromagnetic properties of orthorhombic perovskite-type MFeO3:R3+ (M=La, Gd; R=Eu, Er, Ho) nanocrystalline powders

Peixi Hao,Linwen Jiang,Hanlei Zhao,Jiangtao Li,Shiwen Qiu,Hongbing Chen

The MFeO 3 : R 3+ (M = La, Gd; R = Eu, Er, Ho) nanocrystalline powders were successfully synthesized by the sol–gel combustion method using glycine-based gel at a low ignition temperature. The as-burnt products present honeycomb-like structure with many irregular continuous pores due to the releasing of a large amount of gas during the combustion reaction process. The lattice parameters and cell volumes of MFeO 3 :R 3+ (M = La, Gd; R = Eu, Er, Ho) nanocrystalline powders are compared in detail. The magnetic properties of all the samples were studied systematically. The results show that the doping of rare-earth ions enhanced greatly the coercivity of GdFeO 3 and LaFeO 3 as a result of the enhancement of magnetic anisotropy . The influence of doped rare-earth ions on the magnetic properties was summarized. The distinction of magnetism of MFeO 3 :R 3+ (M = La, Gd; R = Eu, Er, Ho) were explained in detail by the intrinsic magnetism of doped rare-earth ions. This work is significant for the application of rare-earth elements in other magnetic materials.

MicroRNA-155 and microRNA-181a, via HO-1, participate in regulating the immunotoxicity of cadmium in the kidneys of exposed Cyprinus carpio

Hui Li,Guilan Di,Yi Zhang,Rongrong Xue,Jing Zhang,Junping Liang

Cadmium (Cd) is a nonessential metal that is a contaminant in aquatic ecosystems . Cd can accumulate in aquatic animals, leading to detrimental effects in tissues, and Cd exposure can induce immunotoxicity in fish. MicroRNAs (miRNAs) play critical roles in immune responses, yet the participation of miRNAs in Cd-induced immunotoxicity remains poorly understood. The present study evaluated the effects of Cd exposure on the immune responses and the mRNAs and miRNAs expressions of immune-related genes in Cyprinus carpio ( C. carpio ). Then, microRNA-155 (miR-155) was overexpressed and microRNA-181a (miR-181a) was knocked down to determine which miRNA plays a key role in the immune response to Cd. The results showed that 0.5 mg/L Cd 2+ significantly decreased the activity of alkaline phosphatase (AKP) and acid phosphatase (ACP) in the kidneys of C. carpio . Cd exposure upregulated the mRNA expressions of interleukin (IL)-1β, IL-8, nuclear factor-kappa B (NF-κB), tumour necrosis factor-α (TNF-α), and Toll-like receptor 4(TLR-4) and downregulated those of IL-10 and heme oxygenase-1 (HO-1) in C. carpio kidneys. Cd exposure also led to upregulation of miR-155 and miR-181a expressions. Furthermore, AKP and ACP activity in the kidneys was markedly changed after intraperitoneal injection of C. carpio with miR-155 agomir and miR-181a antagomir . All detected mRNA expressions were significantly decreased after injection of miR-155 agomir, and IL-10, NF-κB, TNF-α, and HO-1 mRNA expressions were markedly increased after injection of miR-181a antagomir. The results of this study demonstrate that Cd exposure can immunocompromise C. carpio by targeting HO-1 through miR-155 and miR-181a. This is the first study to reveal that Cd exposure induces immunotoxicity through miR-155 and miR-181a in the kidneys of C. carpio .

Fabricating a novel label-free aptasensor for acetamiprid by fluorescence resonance energy transfer between NH2-NaYF4: Yb, Ho@SiO2 and Au nanoparticles

Weiwei Hu,Quansheng Chen,Huanhuan Li,Qin Ouyang,Jiewen Zhao

Rare earth-doped upconversion nanoparticles have promising potential in the field of pesticide detection because of their unique frequency upconverting capability and high detection sensitivity. This paper reports a novel aptamer-based nanosensor for acetamiprid detection using fluorescence resonance energy transfer (FRET) between NH 2 -NaYF 4 : Yb, [email protected] 2 (UCNPs) and gold nanoparticles (GNPs). Herein, GNPs as acceptors efficiently quench the fluorescence of UCNPs and acetamiprid specifically interacts with acetamiprid binding aptamer (ABA), causing the conformation changes of ABA from random coil to hairpin structure. Accordingly, ABA no longer stabilizes the GNPs in salt solution, leading to the varying aggregation extent of GNPs. Thus, the fluorescence of UCNPs are proportionally recovered. Under the optimized conditions, the enhancement efficiency was observed to increase linearly with the concentration of acetamiprid from 50   nM to 1000   nM, resulting in a relatively low limit of 3.2   nM. Additionally, the aptasensor demonstrated high selectivity to similar structure pesticides such as imidacloprid and chlorpyrifos , and further confirmed its application capacity in adulterated tea samples.

Ulva polysaccharide alleviates Parkinson's disease by regulating inflammation, oxidative damage, and gut microbiota

Wenqian Wang,Qingfu Zhang,Tong Zhang,Fuping Lu,Fufeng Liu

Current therapeutic options for Parkinson's disease (PD) are limited in efficacy and burdened with severe side-effects. Inflammation, intertwined with oxidative stress and influenced by gut microbiota-related changes making the gut-brain axis the main target of innovative therapies. This study reveals the multiple protective effects of polysaccharides extracted from Ulva lactuca (UPE) on PD models. In vitro, UPE was shown to have direct cytoprotective effects, including mitigating 1-methyl-4-phenylpyridinium-induced cell apoptosis, reducing reactive oxygen species levels, and maintaining mitochondrial membrane potential. In vivo, oral UPE significantly restored motor deficits and dopaminergic neuron loss caused by MPTP. These in vivo benefits were associated with the inhibition of oxidative stress and inflammation. Moreover, oral UPE inhibits the excessive activation of glial cells and the excessive production of pro-inflammatory factors and oxidative stress markers in serum. Furthermore, 16S rRNA analysis of gut microbiota revealed that UPE reshaped the gut microbiota of PD mice by increasing the abundance of norank_f_Murbaculaceae and decreasing the abundance of Desulfovibrio . Mechanically, the Nrf2/HO-1 and TLR4/NF-κB signaling pathways are involved in the anti-PD effect of UPE. In general, UPE offers new perspectives on PD treatment and demonstrates the potential of natural polysaccharides in treating neurodegenerative diseases.

dECM restores macrophage immune homeostasis and alleviates iron overload to promote DTPI healing: Dysfunctional iron metabolism in pressure injuries is related to aberrant CD163 and homx-1 signal transduction

Zhang Ju,Shan Hui,Guo Jinglin,Wang Xiaoying,Wang Weiwei

Due to its highly insidious and rapid progression, deep tissue pressure injury (DTPI) is a clinical challenge. Our previous study found that DTPI may be a skeletal muscle injury dominated by macrophage immune dysfunction due to excessive iron accumulation. Decellularized skeletal muscle matrix (dECM) hydrogel promotes skeletal muscle injury repair. However, its role in polarizing macrophages and regulating iron metabolism in DTPI remains unclear. Here, porcine dECM hydrogel was prepared, and its therapeutic function and mechanism in repairing DTPI were investigated. The stimulus of dECM hydrogel towards RAW264.7 cells resulted in a significantly higher percentage of CD206+ macrophages and notably decreased intracellular divalent iron levels. In mice DTPI model, dECM hydrogel treatment promoted M1 to M2 macrophage conversion, improved iron metabolism, and reduced oxidative stress in the early stage of DTPI. In the remodeling phase, the dECM hydrogel remarkably enhanced revascularization and accelerated skeletal muscle repair. Furthermore, the immunomodulation of dECM hydrogels in vivo was mainly involved in the P13k/Akt signaling pathway, as revealed by GO and KEGG pathway analysis, which may ameliorate the iron deposition and promote the healing of DTPI. Our findings indicate that dECM hydrogel is promising in skeletal muscle repair, inflammation resolution, and tissue injury healing by effectively restoring macrophage immune homeostasis and normalizing iron metabolism.
