Preparation and Wound Repair of Injectable and Self-Healing Benzaldehyde-Modified Konjac Glucomannan Oligosaccharide/Polyglutamic Acid/ε-Polylysine Hydrogel

Feng Huang,Jiajie Chen,Xuan Tang,Yujian Li,Haixin Bao,Xuan Mao,Shunqing Tang

Oligosaccharides always have better water solubility, higher possibilities for modification, and unique biofunctions compared with polysaccharides, but they are rarely used as the matrix of a hydrogel. Here, we prepared a composite BKOS/HPGA/PL hydrogel (BKPP hydrogel) constructed by hydrazone/imine bonds between the aldehyde groups of benzaldehyde-modified konjac glucomannan oligosaccharide (BKOS) and the primary amino groups of both hydrazide-modified polyglutamic acid (HPGA) and ε-polylysine (ε-PL). The hydrogels had both injectable and self-healing properties. The gelation time reached 23 s when 2% of BKOS (DS = 21.7%), 10% HPGA (DS = 11.5%), and 10% ε-PL solutions were mixed in a volume ratio of 5:4.5:0.5. Besides high water-retention capability and good cytocompatibility, the hydrogel also maintained both the immunoactivities of BKOS and the antibacterial performances of ε-PL and HPGA, and thus exhibited good wound healing performance in the whole cortex wound repair process of rats, which might have potential for its biomedical application.

Robust construction of konjac glucomannan/polylactic acid nanofibrous films incorporated with carvacrol via microfluidic blow spinning for food packaging

Lizhen Zhou,Di Zhang,Nitong Bu,Liying Huang,Huanglong Lin,Wei Liu,Guoyu Cao,Ruojun Mu,Jie Pang,Lin Wang

In recent years, the application of biopolymer-based nanofibers prepared via microfluidic blow spinning (MBS) for food packaging has continuously increased due to their advantages of biocompatibility, biodegradability, and safety. However, the poor spinnability, undesirable water barrier capacity, and loss of antibacterial and antioxidant properties of biopolymer-based nanofibers strictly restrict their real-world applications. In this work, carvacrol (CV) incorporated konjac glucomannan (KGM)/polylactic acid (PLA) nanofibrous films (KP-CV) were produced by MBS. The FTIR spectra and XRD analysis revealed the hydrogen bonding interactions among CV, PLA, and KGM, thus significantly improving the TS of KP-CV nanofibrous films from 0.23 to 1.27 MPa with increased content of CV from 0 % to 5 %. Besides, KP-CV nanofibrous films showed improved thermal stability, excellent hydrophobicity (WCA: 128.19°, WVP: 1.02 g mm/m 2  h kPa), and sustained release of CV combined with good antioxidant activities (DPPH radical scavenging activity: 77.51 ± 1.57 %), and antibacterial properties against S. aureus (inhibition zone: 26.33 mm) and E. coli (inhibition zone: 22.67 mm). Therefore, as prepared KP-CV nanofibrous films can be potentially applied as packaging materials for the extended shelf life of cherry tomatoes.

Quercetin-loaded melanin nanoparticle mediated konjac glucomannan/polycaprolactone bilayer film with dual-mode synergistic bactericidal activity for food packaging

Di Zhang,Nitong Bu,Lizhen Zhou,Lihong Lin,Yifan Wen,Xianrui Chen,Liying Huang,Huanglong Lin,Ruojun Mu,Lin Wang,Jie Pang

It is still difficult for a single antibacterial modality to realize satisfactory management of bacterial breeding in food preservation. To solve this problem, we developed a photothermal-derived dual-mode synergistic bactericidal konjac glucomannan (KGM)/polycaprolactone (PCL) bilayer film incorporated with quercetin-loaded melanin-like nanoparticles (Q@MNPs). The results showed that the mechanical properties (TS: 29.8 MPa, EAB: 43.1 %), UV shielding properties, and water resistance (WCA: 124.1°, WVP: 3.92 g mm/m 2  day kPa) of KGM-Q@MNPs/PCL bilayer films were significantly improved. More importantly, KGM-Q@MNPs/PCL bilayer film presented outstanding photothermal inversion and controlled release behavior of Q triggered by near infrared (NIR) radiation, thus contributing to excellent dual-mode synergistic antibacterial properties against E. coli and S. aureus . Meanwhile, the KGM-Q@MNPs/PCL bilayer film possessed good biocompatibility and low toxicity. As a proof-of-concept application, we further verified the significant value of film for the preservation of cherry tomatoes. Since KGM-Q@MNPs/PCL bilayer film showed excellent biodegradability, this work will aid the development of sustainable antibacterial food packaging materials.

Bionic porous konjac glucomannan/polylactic acid - based composite film: Synergistic functional design for antibacterial preservation and passive radiative cooling

Yueguang Wang,Zifeng Huang,Oujun Dai,Xin Lei,Meining Li,Hanlin Zhou,Jie Pang

Inspired by the pore structure of plant leaves, this study fabricates a konjac glucomannan/polylactic acid (KGM/PLA)-based composite film that mimics the passive thermal regulation mechanism of natural stomata, enabling highly efficient cooling. By integrating xanthan gum (XG)-stabilized KGM/PLA matrices and hexadecyltrimethoxysilane-carvacrol@zinc oxide/cellulose nanocrystal nanosheets (HDTMS-CRV@ZnO/CNC NSs), the composite film exhibits potent antibacterial activity (25.22 ± 1.5 mm inhibition zone against Staphylococcus aureus ) and extends produce shelf life by over 7 days. The film also demonstrates outstanding optical properties, achieving 91.06 % solar reflectance (0.3–2.5 μm) and 91.40 % infrared emissivity (8–13 μm), which enables sub-ambient cooling of ∼10 °C under sunlight. Practical tests show the film maintains an internal temperature of 27.18 °C at 35 °C ambient conditions, effectively preserving the freshness of strawberries and tomatoes compared to deteriorated control samples. This work presents an innovative approach to intelligent food packaging, combining preservation functionality and environmental adaptability for sustainable food storage applications.

Konjac glucomannan-based emulsion films with lysozyme-amyloid fibrils-chitosan complexes as dual-function antimicrobial emulsifiers for salmon preservation

Yangyang Guo,Xingru Yan,Jie Pang,Bo Liu,Yihua Bai,Di Wu,Yukun Song,Mingqian Tan,Wentao Su

Salmon is highly susceptible to spoilage due to microbial contamination and lipid oxidation, whereas active packaging can effectively delay quality deterioration during storage. This study employed chitosan-modified lysozyme amyloid fibrils (LC) as both antimicrobial agents and emulsifiers to prepare stable turmeric essential oil (TEO) Pickering emulsions with different LC-to-TEO volume ratios (1:1, 2:1, 3:1, 4:1), designated as TEOP1, TEOP2, TEOP3, and TEOP4 respectively. These emulsions were then incorporated into a konjac glucomannan (KGM) matrix to develop composite films. This study systematically evaluated the structural properties and freshness-preserving effects of composite films, using pure KGM film as the primary control and commercial polyethylene terephthalate (PET) film as the reference control. The results showed that strong interactions among components enhanced film properties: TEOP4/KGM reduced water vapor and oxygen permeability by 44 % (6.79 ± 0.59 g·mm/ (m 2 ·day·kPa)) and 31 % (6.55 ± 0.36 g/ (m·s·kPa)). TEOP3/KGM exhibited optimal tensile strength (26.63 ± 1.45 MPa) and antioxidant activity (DPPH: 47.5 ± 2.6 %, ABTS: 96.3 ± 0.1 %), while TEOP4/KGM had highest elongation at break (68.35 ± 0.81 %). This suggests a trade-off, where a higher oil content (TEOP4) plasticizes the film, increasing flexibility but reducing strength. TEOP/KGM films inhibited E. coli , S. aureus , and L. monocytogenes , with the TEOP3/KGM and TEOP4/KGM formulations achieving 100 % inhibition rates. In salmon preservation, TEOP3/KGM best maintained appearance, pH stability, and reduced microbial growth/TVB-N, confirming its potential in aquatic product preservation.

Effects of konjac glucomannan with different molecular weights on gut microflora with antibiotic perturbance in in vitro fecal fermentation

Yu-Heng Mao,Yi-Xuan Xu,Yan-Heng Li,Jing Cao,Feng-Ling Song,Dan Zhao,Yimin Zhao,Zhao-Mei Wang,Yan Yang

This study investigated the effect of konjac glucomannan (KGM) of different molecular weight on fecal microflora against antibiotic disturbance. KGM (~1.8 × 10 7  Da) was partially hydrolysed with trifluoroacetic acid (TFA) for 10 and 60 min to KGM1 (~2.1 × 10 4  Da) and KGM2 (7413 Da), respectively. The acid treatment caused significant reduction of intrinsic viscosity , average molecular weight (MW) and particle size of KGM, but brought limited change to the molecular structure. Low-MW KGM2 showed the most significant effect on fecal microflora in the presence of two common antibiotics (ampicillin and clindamycin), by increasing the relative abundance of Bifidobacteriaceae while decreasing the proportion of Enterobacteriaceae . Additionally, both the native and acid-treated KGM counteracted the adverse influence of antibiotics on the production of short chain fatty acids . The results have demonstrated the effect of KGM on gut microbiota with antibiotic disturbance.

A novel biocompatible double network hydrogel consisting of konjac glucomannan with high mechanical strength and ability to be freely shaped

Zhiyong Li,Yunlan Su,Baoquan Xie,Xianggui Liu,Xia Gao,Dujin Wang

A novel physically linked double-network (DN) hydrogel based on natural polymer konjac glucomannan (KGM) and synthetic polymer polyacrylamide (PAAm) has been successfully developed. Polyvinyl alcohol (PVA) was used as a macro-crosslinker to prepare the PVA–KGM first network hydrogel by a cycle freezing and thawing method for the first time. Subsequent introduction of a secondary PAAm network resulted in super-tough DN hydrogels. The resulting PVA–KGM/PAAm DN hydrogels exhibited unique ability to be freely shaped, cell adhesion properties and excellent mechanical properties, which do not fracture upon loading up to 65 MPa and a strain above 0.98. The mechanical strength and microstructure of the DN hydrogels were investigated as functions of acrylamide (AAm) content and freezing and thawing times. A unique embedded micro-network structure was observed in the PVA–KGM/PAAm DN gels and accounted for the significant improvement in toughness. The fracture mechanism is discussed based on the yielding behaviour of these physically linked hydrogels.

Photothermal antibacterial bilayer films from konjac glucomannan/shellac: A Prussian blue-engineered strategy for preservation of fresh-cut Benincasa hispida

Xin Lei,Yueguang Wang,Xianrui Chen,Jing Zhang,Xuan Lin,Wenjun Guo,Xiuqing Guan,Chengrong Wen,Di Zhang,Lin Wang,Jie Pang

PB@MSN-CIN nanoparticles enabled CIN release with NIR-triggered photothermal conversion. • S/P-HK bilayer films combined PB@MSN-CIN-loaded P-HK inner layer with a hydrophobic SHL-QCT outer layer. • Regulating the SHL-QCT layer thickness improved moisture barrier, UV shielding, and photothermal retention of the bilayer films. • Under NIR treatment, the optimized S/P-HK film suppressed microbial growth, browning, and water loss of fresh-cut Benincasa hispida . Fresh-cut vegetables are highly susceptible to microbial growth, enzymatic browning, and moisture loss, creating a need for environmentally sustainable active packaging with effective barrier and antimicrobial functions. In this study, a multifunctional bio-based bilayer film (S/P-HK) was developed using the thickness of the hydrophobic outer layer as a key design variable. The inner active layer (P-HK) comprised a konjac glucomannan/hydroxyethyl cellulose matrix containing cinnamaldehyde (CIN)-loaded mesoporous silica nanoparticles coated with Prussian blue (PB@MSN-CIN), whereas the outer layer was composed of quercetin-containing shellac (SHL-QCT). SEM revealed a compact bilayer structure, while FT-IR, XRD, and TGA indicated intermolecular interactions and good thermal stability. Increasing the thickness of SHL-QCT layer enhanced moisture-barrier performance and heat retention under near-infrared (NIR) irradiation. Among which, the S20/P-HK film exhibited nearly complete UV shielding, strong antioxidant activity (DPPH and ABTS radical scavenging rates >80%), excellent antibacterial activity (antibacterial rates >99.99% against Escherichia coli and Staphylococcus aureus ), and controlled release of CIN. During preservation of fresh-cut Benincasa hispida , the S20/P-HK film with NIR irradiation significantly inhibited microbial growth, suppressed the browning index to 0.90 ΔOD₄₂₀·mL/g, and restricted weight loss to 0.26% over 10 days. These findings demonstrate a promising thickness-regulated bilayer design that integrates moisture-barrier protection, NIR-responsive CIN release, and photothermal antibacterial activity for active packaging. Future studies should validate the applicability of S/P-HK films to other perishable foods and evaluate their antifungal efficacy. EGCG-functionalized deacetylated konjac glucomannan/carboxylated cellulose nanofiber films: Development, characterization, and application in antibacterial preservation of fresh-cut fruits

Yang Cao,Yile Zhao,Zhixin Li,Di Wu,Chen Chen,Aili Jiang,Xiangzheng Yang,Da Wang,Xian Li,Kunsong Chen

A bio-based antibacterial film was fabricated by combining EGCG with DKGM and CCNF. • EGCG exhibited good antibacterial activity when acting on fresh-cut fruit flesh. • EGCG interacted with the film via H-bond, thereby increasing its structural ordering. • The incorporation of EGCG also enhanced the thermal stability of the DKGM/CCNF films. • The films inhibited >80% S. aureus after being covered on fresh-cut apples for 1 h. Fresh-cut fruits are susceptible to microbial contamination that leads to quality deterioration. In short-term distribution scenarios oriented toward consumers, there is a need for active packaging materials that are contact-safe and capable of exerting rapid antibacterial effects. The present study incorporated epigallocatechin gallate (EGCG) into a deacetylated konjac glucomannan/carboxylated cellulose nanofibers (DKGM/CCNF) matrix to prepare bio-based antibacterial films loaded with low and high EGCG concentrations (LEG-DKGM/CCNF and HEG-DKGM/CCNF). The effects of EGCG incorporation on the structure, mechanical properties, and barrier performance of the composite films were systematically studied, and the films were applied to fresh-cut fruits. Results indicated that EGCG formed hydrogen bond interactions with the DKGM/CCNF network and altered the film's microstructure, enhancing thermal stability while reducing the tensile strength and elongation at break of the composite films. In vitro antibacterial tests demonstrated that EGCG-loaded composite films inhibited foodborne pathogens associated with fresh-cut produce contamination, including Staphylococcus aureus , Listeria monocytogenes , and enterohemorrhagic Escherichia coli , with more pronounced inhibition against S. aureus . Further using S. aureus -inoculated fresh-cut apple as a model, after short-term (1 h ) coverage with the films, LEG-DKGM/CCNF and HEG-DKGM/CCNF films achieved an inhibition rate of over 80% against S. aureus . In addition, fresh-cut apple storage tests showed that EGCG-loaded films reduced browning index and helped maintain the appearance and firmness of fresh-cut apples during storage. Overall, the EGCG-functionalized DKGM/CCNF films provide a feasible solution for developing bio-based antibacterial packaging with rapid antibacterial and preservation functions for fresh-cut fruits. Formation and properties of β-conglycinin particle-Konjac Glucomannan co-stabilized Pickering emulsion: Dynamic adsorption behavior and complex interface microstructure

Yanan Guo,Tianfu Cheng,Shuo Zhang,Haotian Liu,Ramnarain Ramakrishna,Lianzhou Jiang,Zhongjiang Wang,Zengwang Guo

The interaction between polysaccharides and proteins in co-adsorption played a critical role in determining interfacial properties and emulsion stability. This study focused on the regulation mechanism of different concentrations of polysaccharide on the dynamic adsorption behavior of protein, and how the interfacial film microstructure affected the stability of emulsion. The research demonstrated that Konjac Glucomannan (KGM) and β-conglycinin (7S) formed stable composite through hydrogen bonding and hydrophobic interactions. The results from interfacial adsorption kinetics and Quartz Crystal Microbalance with Dissipation Monitoring (QCM-D) indicated that the structure of 7S stable interface layer is loose and disordered, which was not conducive to the formation of emulsion. KGM could synergistically facilitate the initial diffusion, penetration, and rearrangement of 7S at the oil-water interface thus forming a dense and viscoelastic three-dimensional gel-like multilayer interfacial structure. High-density adsorption of the 7S-KGM composite significantly increased the thickness and the protein content of the interfacial layer, enhancing emulsion stability. Dissipative particle dynamics simulations further demonstrated that the synergistic adsorption of KGM enhanced the interfacial adsorption efficiency of 7S, thereby reducing droplet size. This provided insights into the mechanisms by which polysaccharides enhanced protein emulsification performance, offering critical guidance for optimizing protein interfacial properties and enhancing emulsion stability.

Rheological properties of konjac glucomannan composite colloids in strong shear flow affected by mesoscopic structures: Multi-scale simulation and experiment

Lu Liu,Ning Zhou,Ying Yang,Xin Huang,Renhui Qiu,Jie Pang,Shuyi Wu

The rheological properties of biomacromolecule colloids under large shear rates (> 10 3 s -1 ) have attracted considerable interest due to the fundamental role in spraying of macromolecular solutions, degradation of macromolecular by atomization , and high-speed transportation of colloids. However, it is difficult for common instruments to characterize the rheological property under large shear rates and investigate the effect of the dynamical cluster structure on the rheological properties. In this study, multi-scale simulations and experiments were implemented to investigate the rheological property of biomacromolecule colloids composed of konjac glucomannan and hydroxypropyl methylcellulose. The multi-scale method combined with molecular dynamics simulation and Brownian dynamics simulation was effective and accurate to analyze the dynamical behavior of clusters (~ 500 nm) for a long time (~ 2 ms). Moreover, for the first time, the cluster structure and rheological properties in the strong shear flows were predicted by multi-scale simulations, such as the breakage of polymers and decreased storage modulus at large shear rates (10 4 ~ 10 5 s −1 ) where the test accuracy of common instruments is limited. This study provided more understanding of the rheological property of biomacromolecule colloids, which will shed light on the application of the biomacromolecule colloids in strong shear flows.

Preparationof Konjac Glucomannan-Based Composite Films Loaded with Hydroxypropyl-β-Cyclodextrin/Hesperetin Inclusion Complex and Their Preservation Effect on Strawberries

Guangsen Li,Junwei Zhang,Youxiang Wu,Xiang Wang,Xin Lei,Hanyu Wei,Zhiwen Hu,Qibiao Weng,Qinhua Zhang,Chengrong Wen,Yilan Sun,Qian Ning

A KGM/CMC/PVA film loaded with a hydroxypropy-β-cyclodextrin/hesperetin (HP-β-CD/HES) inclusion complex was developed for strawberry preservation. The inclusion complex showed an encapsulation efficiency of 70.41 ± 4.01%. Loading the inclusion complex enhanced the barrier and bioactive properties of the films. At the better loading level (KCP-HH3, 1.5%), the film provided nearly complete UV shielding at 200–350 nm and showed a transmittance of approximately 21% at 800 nm. Relative to KCP, KCP-HH3 exhibited lower water vapor permeability (0.69 ± 0.06 g·mm/(m2·h·kPa)) and oxygen permeability (0.066 ± 0.006 g·mm/(m2·h·kPa)). The cumulative HES release from KCP-HH3 reached 82.43 ± 2.19% after 7 days at 98% relative humidity. KCP-HH3 scavenged 78.19 ± 3.71% of DPPH radicals and 88.89 ± 4.30% of ABTS radicals. It also inhibited Escherichia coli (91.18 ± 2.99%), Staphylococcus aureus (79.21 ± 3.45%), and Aspergillus niger (92.85 ± 2.75%). This highest-loading formulation delivered the best overall strawberry-preservation performance. After 12 days of storage, strawberries packaged with KCP-HH3 showed higher firmness (0.89 ± 0.04 N) and total soluble solids (7.96 ± 0.21%), as well as lower weight loss (14.30 ± 0.40%), than the polyethylene control, which showed a firmness of 0.42 ± 0.05 N, total soluble solids of 6.22 ± 0.20%, and weight loss of 16.36 ± 0.66%. These results support the potential of HP-β-CD/HES-loaded KCP films as biodegradable active packaging for fruit preservation.
