Photothermal-enhanced chemodynamic therapy (CDT) based on the Fenton reaction has been widely studied as a safe, tumor-specific, and highly effective nanomedicine therapy. Herein, a collective hyperthermia-facilitated glutathione (GSH) inhibition strategy for boosted oxidative stress treatment was presented using mesoporous Prussian blue (MPB NPs) as a multifunctional carrier and loading β-lapachone (Lap) as an oxidative mediator. A phase transfer catalyst (lauric acid, La) was co-packed for preventing drug leakage and achieving temperature-controlled drug release. After biocompatible decoration by external encapsulation with polyethylene glycol (PEG), the MPB-Lap/La@PEG nanocatalysts (Mlalp NCs) were rationally constructed. Upon 808 nm light irradiation, the Mlalp NCs showed a good photothermal effect with temperature increases for directly damaging tumor cells and accelerating the release of Lap. Significantly, the expression of NAD(P)H: quinone oxidoreductase-1 (NQO1) in cells could increase and generate more hydrogen peroxide (H 2 O 2 ) with Lap and reduce the level of GSH concurrently, forming a hyperthermia-facilitated glutathione inhibition strategy. Most importantly, the hyperthermia also accelerated the Fenton reaction rate, resulting in an enhanced CDT effect. In addition, the Mlalp NCs also equipped with excellent T 1 -weighted magnetic resonance imaging properties. To sum up, this work displays a collective hyperthermia-facilitated glutathione inhibition strategy for boosted oxidative stress treatment.

Medical adhesives play an important role in clinical medicine because of their flexibility and convenient operation. However, they are still limited to laparoscopic surgeries, which have demonstrated urgent demand for liver retraction with minimal damage to the human body. Here, inspired by the suction cup structure of octopus, an adhesive patch with excellent mechanical properties, robust and switchable adhesiveness, and biocompatibility is proposed. The adhesive patch is combined by the attachment body mainly made of poly(acrylic acid) grafted with N -hydroxysuccinimide ester, crosslinked biodegradable gelatin methacrylate and biodegradable biopolymer gelatin to mimic the adhesive sucker rim, and the temperature-sensitive telescopic layer of microgel-crosslinked poly( N -isopropylacrylamide- co -2-hydroxyethyl methacrylate) to shrink and form internal cavity with reduced pressure. Through mechanical tests, adhesion evaluation, and biocompatibility analysis, the bioinspired adhesive patch has demonstrated its capacity not only in adhesion to tissues but also in potential treatment for medical applications, especially laparoscopic technology. The bioinspired adhesive patch can break through the limitations of traditional retraction methods, and become an ideal candidate for liver retraction in laparoscopic surgery and related clinical medicine.

Inspired by natural organisms, a four-dimensional (4D)-printed starfish-like bionic soft robot (SBSR) was effectively prepared by integrating three-dimensional (3D) printing with smart hydrogels. The body of the SBSR is composed of a reduced graphene oxide-poly(N-isopropylacrylamide) hydrogel (rGO-PNH) with superior mechanical properties. In addition, the enhanced photothermal conversion effect was obtained by the reduction of graphene oxide nanosheets after the 3D printing process. Cylindrical actuators prepared using rGO-PNH exhibited bending and orientation toward the light source within 20 s of exposure to near-infrared light, thus demonstrating the rapid photoresponsivity of rGO-PNH. Furthermore, the 4D-printed SBSR showcased effective grasping, lifting, and releasing of objects by mimicking the predatory behavior of starfish. This study would provide insights into the development of responsive materials in 4D printable bionic soft robots and their applications in areas such as biomimetic devices and artificial muscles.

The critical-sized bone defect resulting from trauma, tumor resection, and congenital deformity fails to undergo spontaneous healing due to its substantial size, while the ensuing inflammatory process and hypoxic environment further impede the regenerative process. Therefore, it has consistently presented a significant clinical challenge. In the present study, we incorporate a glycyrrhizic acid (GA)-functionalized hydrogel onto the surface of a Hydroxyapatite (Hap)-modified Polycaprolactone (PCL) scaffold to fabricate a composite scaffold. The composed scaffold showed favorable anti-inflammatory and antioxidative capabilities by modulating macrophage polarization and scavenging reactive oxygen species (ROS); the modification of Hap enhanced its osteogenic ability. An in vivo rat skull defect model confirmed that the composed scaffold efficiently promotes bone regeneration. In general, the composed scaffold with the ability of osteoimmune microenvironment regulation can effectively repair critical-sized bone defects. This strategy provides a promising method for the reconstruction of large segmental bone defects.

Effective thermal management materials are crucial for modern electronics, aerospace, and energy applications. This study investigates a novel approach by constructing a Si₃N₄ skeleton with a triply periodic minimal surface (TPMS) structure and reinforcing phase change materials (PCM) with boron nitride (BN) to develop Si₃N₄@BN/PCM composites that exhibit enhanced thermal conductivity, thermal stability and electrical insulation. Utilizing digital light processing 3D printing, a Si₃N₄ TPMS structure was prepared and subsequently reinforced with BN via vacuum-assisted infiltration into PCM. The incorporation of 3D Si₃N₄ skeleton and 2D BN flakes created a dual-scale network within the PCM, significantly increasing thermal conductivity from 0.24 W/m·K to 2.04 W/m·K. The presence of Si₃N₄ and BN notably improved thermal stability and reduced the coefficient of thermal expansion. Additionally, electrical insulation properties improved with an increase in volume resistivity and breakdown strength to 5.1 × 10 1 ⁴ Ω·cm and 62.4 kV/mm, respectively. This work demonstrates that Si₃N₄@BN/PCM composites effectively integrate high thermal conductivity, stability, and electrical insulation, thereby meeting the stringent demands of thermal management applications.

Ionic conductive hydrogels (ICHs) have emerged as a landmark soft material for a wide range of applications, such as flexible wearable sensors and electronic skins. However, to achieve a super-stretchability, high strength, and self-adhesion simultaneously by 3D printing remains a significant challenge. In the construction of a hydrogel, ionic liquids (ILs) and tannic acid (TA) have been successfully introduced in the copolymerization of acrylamide (AAm) and poly(ethylene glycol) (diol) diacrylate (PEGDA) to form a p(AAm-co-PEGDA) hydrogel (PAP) system. The PAP hydrogel showed super-stretching (4300%), high strength, and self-adhesion properties. More specifically, the 3D printing of the ICHs provided an effective and flexible way to manufacture flexible wearable sensors, thus greatly simplifying the device fabrication process. In addition, the sensors could be specified by a customized production and were, thus, adapted to a wider range of applications. It is believed that the here presented hydrogel integration with 3D printing will inspire new ideas on how to prepare novel flexible sensors, thus promoting further research on the construction of electronic skins, human-computer interactions, and advanced materials.

As a superior alternative to sutures, tissue adhesives have been developed significantly in recent years. However, existing tissue adhesives struggle to form fast and stable adhesion between tissue interfaces, bond weakly in wet environments, and lack bioactivity. In this study, a degradable and bioactive citrate-based polyurethane adhesive is constructed to achieve rapid and strong tissue adhesion. The hydrophobic layer was created with polycaprolactone to overcome the bonding failure between tissue and adhesion layer in wet environments, which can effectively improve the wet bonding strength. This citrate-based polyurethane adhesive provides rapid, non-invasive, liquid-tight, and seamless closure of skin incisions, overcoming the limitations of sutures and commercial tissue adhesives. In addition, it exhibits biocompatibility, biodegradability, and hemostatic properties. The degradation product citrate could promote the process of angiogenesis and accelerate wound healing. This study provides a novel approach to the development of a fast-adhering wet tissue adhesive and provides a valuable contribution to the development of polyurethane-based tissue adhesives.

Designing a smart hydrogel to accelerate skin tissue regeneration at wound sites and restore the tissue function is highly desirable in clinical applications. In this study, a series of hydrogels with promising antioxidative and antibacterial traits based on recombinant human collagen type III (rhCol III), which is an emerging biomaterial, and chitosan (CS) were fabricated. The rhCol III-CS hydrogel could realize rapid gelation at wound locations and completely cover irregular wounds. Additionally, the hydrogel facilitated the proliferation and migration of cells and showed potent antibacterial efficacy against both strains, Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli) in vitro. Significantly, the rhCol III-CS2 hydrogel increased the deposition of collagen, thereby accelerating full-thickness wound healing. Collectively, this bioinspired hydrogel was a promising multifunctional dressing to reconfigure the damaged tissue without additional drugs, exogenous cytokines, or cells, providing an effective strategy for the repair and regeneration of skin wounds.

Intervertebral disc degeneration (IVDD) is a leading cause of low back pain, but existing therapies fail to halt its progression. Here, we develop an exosome-loaded methacrylated silk fibroin hydrogel (SM@ME) that synergistically activates the mitochondrial unfolded protein response (UPRmt) to combat IVDD. SilkMA hydrogel, optimized for mechanical compatibility with nucleus pulposus (NP) tissue, enables sustained release of Cavin-2-engineered exosomes (M−EXO). RNA sequencing revealed that SM@ME upregulates DKK2, which suppresses the Wnt/β-catenin pathway while activating UPRmt, thereby reducing apoptosis, and senescence in rat and human NP cells. In vivo , SM@ME injection into puncture-induced IVDD rats attenuated histological degeneration. Notably, lentiviral-mediated overexpression of DKK2 in NP tissues resulted in therapeutic effects similar to those of SM@ME treatment. This work suggests that hydrogel-delivered exosomes can modulate UPRmt for IVDD therapy, offering a novel biomaterial strategy for disc regeneration.

It is often difficult to close and repair irregular wounds such as gums with traditional dressings that have a fixed shape, while injectable hydrogels that can be molded in situ offer tremendous advantages in treating complex, large wounds and can better seal or fill a variety of irregular wounds. While the majority of injectable hydrogels developed in current research have poor mechanical properties and biocompatibility and obstacles to clinical applications, this paper reports an injectable hydrogel that can be molded in situ with appropriate mechanical properties, excellent biocompatibility, and antibacterial activity. Under UV irradiation, a methacrylate-gelatin/poly(vinyl alcohol) loaded with copper ion cross-linked tannic acid nanoparticle (GelMA/PVA-TA/Cu2+NPs) precursor solution could rapidly gel within 45 s and adhere stably to the wound tissue by topological adhesion and hydrogen bonding. Through hydrogen bonding and physical entanglement, TA/Cu2+NPs are anchored in the GelMA/PVA interpenetrating network to form the GelMA/PVA-TA/Cu2+NPs composite hydrogel. This injectable hydrogel, which can quickly achieve photocurable in situ molding, can not only achieve closure and repair of irregular wounds but also has appropriate swelling characteristics, absorbing tissue exudate and blocking bacteria, while providing a good moist environment for wounds and continuously promoting wound healing. Cytotoxicity tests and wound healing experiments have shown that GelMA/PVA-TA/Cu2+NPs injectable hydrogels have good biocompatibility and wound closure and repair ability and have promising applications in the treatment of irregularly wounded orifices.

Chronic wound infection often leads to irregular tissue closure and accompanies delayed healing and economy issues. Developing an ideal wound dressing that can control the occurrence of antibacterial infections and biological responses is highly desirable. In this study, a multifunctional hybrid hydrogel (GS@EG–Cu–CA NPs) containing synthesized thiolated gelatin, methacrylated silk fibroin, and (−)-epigallocatechin gallate–copper ionic–carrageenan nanoparticles (EG–Cu–CA NPs) was engineered by a thio–ene click reaction. The metal–polyphenol EG–Cu–CA NPs were encapsulated with kappa-carrageenan to enhance its aqueous-soluble, mechanical, and bioactive properties and endowed the hydrogel dressing with fascinating antibacterial, antioxidation, and nitric oxide (NO) generation by catalyzing. The hybrid hydrogels also illustrated a favorable cytocompatibility. Benefiting from the thio–ene click reaction, the hybrid hydrogels were injected and photocured rapidly in situ to cover an irregular wound. In an SD rat full-thickness skin-wound-infected model, the methicillin-resistant Staphylococcus aureus-infected wound covered with GS@EG–Cu–CA NPs was almost completely healed after 10 days. This study presents a facile design of hydrogel dressing incorporating metal–polyphenol nanoparticles, which demonstrates a promising potential way for dealing with effective wound infection management and other complicated wound healings.

Prosthesis implantation and fat transplantation are common breast reconstruction methods. In general, prosthesis implantation alone does not achieve a realistic enough appearance, and fat transplantation alone is difficult to achieve in the correct capacity. To date, no reports have focused on methods of combining fat with implanted prostheses for breast reconstruction. Using a newly designed bionic ink (i.e., polyether F127 diacrylate (F127DA) & poly(ethylene glycol) diacrylate (PEGDA)) and projection-based three-dimensional bioprinting (PBP), we report the development of a new method for printing porous prostheses. PEGDA was used to improve the printing precision of the prosthesis by increasing the gel point of F127DA and reducing the impact of external temperature. The compression modulus of the printed prosthesis was very close to that of prostheses currently used in clinical practice and to that of natural breasts. Finally, stromal vascular fraction gel (SVF-gel), a human fat extract, was injected into the pores of the synthesized prostheses to prepare a prosthesis mixed with adipose tissue. These were implanted subcutaneously in nude mice to observe their biological performance. After 14 and 28 days of observation, the prosthesis showed good biocompatibility, and adipose tissues grew well in and around the prosthesis. This result shows that a porous prosthesis containing pre-placed adipose tissues is a promising breast reconstruction material. Graphic abstract
