This study aimed to evaluate the application of encapsulated L . plantarum and eugenol as potential biocontrol agents in sliced apples. The combined encapsulated L . plantarum and eugenol treatment was more effective than separate encapsulated L . plantarum and eugenol treatments, with regards to browning inhibition and consumers panel test. The application of encapsulated L . plantarum and eugenol reduced the decline of the physicochemical qualities of the samples, and improved the ability of antioxidant enzymes to scavenge reactive oxygen species. Furthermore, reductions in the growth of L . plantarum of only 1.72 log CFU/g were observed after 15 days of storage at 4 °C for samples treated with encapsulated L . plantarum and eugenol. Results suggest the combined encapsulated L . plantarum and eugenol appears to be a promising method to protect fresh-cut apples from food-borne pathogens while maintaining the visual appearance.

Polygonatum odoratum (Yu-Zhu) can be utilized to treat the digestive and respiratory illness . Previous studies have revealed that the underlying therapeutic mechanism of P. odoratum polysaccharides (POPs) is associated with remodeling the gut microbiota . However, POPs in terms of the chemical composition and fermentation activities have been understudied. Here we developed the three-level fingerprinting approaches to characterize the structures of POPs and probed into the beneficial effects on promoting the growth and fermentation of Lactobacillus johnsonii . POPs were prepared by water decoction followed by alcohol sedimentation, while trifluoroacetic acid under different conditions to prepare the hydrolyzed oligosaccharides and monosaccharides . POPs exhibited three main molecular distribution of 601–620 kDa, 4.12–6.09 kDa, and 3.57–6.02 kDa. Hydrolyzed oligosaccharides with degree of polymerization (DP) 2–13 got primarily characterized by analyzing the rich fragmentation information obtained by hydrophilic interaction chromatography/ion mobility-quadrupole time-of-flight mass spectrometry (HILIC/IM-QTOF-MS). Amongst them, the DP5 oligosaccharide was characterized as 1,6,6-kestopentaose. The molecular ratio of Fru: Ara : Glc: Gal : Xyl was 87.72: 0.30: 11.56: 0.19: 0.23. In vitro fermentation demonstrated that 4.5 mg/mL of POPs could significantly promote the growth of L. johnsonii . Co-cultivated with 4.5 mg/mL of POPs, L. johnsonii exhibited stronger antimicrobial activity against Klebsiella pneumoniae . The concentrations of short-chain fatty acids in the POPs-lactobacilli fermented products, including acetic acid, isobutyric acid , and isovaleric acid , were increased. Conclusively, POPs represent the promising prebiotic candidate to facilitate lactobacilli , which is associated with exerting the health benefits.

The microecological stability of the gut microbiota plays a pivotal role in both preventing and treating colorectal cancer (CRC). This study investigated whether Lactobacillus plantarum CBT (LP-CBT) prevents CRC by inducing alterations in the gut microbiota composition and associated metabolites. The results showed that the LP-CBT inhibited colorectal tumorigenesis in azoxymethane/dextran sulfate sodium (AOM/DSS) treated mice by repairing intestinal barrier function. Furthermore, LP-CBT decreased pro-inflammatory cytokines and anti-inflammatory cytokines. Importantly, LP-CBT remodeled intestinal homeostasis by increasing probiotics (Coprococcus, Mucispirillum, and Lactobacillus) and reducing harmful bacteria (Dorea, Shigella, Alistipes, Paraprevotella, Bacteroides, Sutterella, Turicibacter, Bifidobacterium, Clostridium, and Allobaculum), significantly influencing arginine biosynthesis. Therefore, LP-CBT-treatment regulated invertase and metabolites associated with the arginine pathway (carbamoyl phosphate, carboxymethyl proline, L-lysine, 10,11 -Epoxy- 3-geranylgeranylindole, n-(6)- [(Indol-3-yl) acetyl]-L-lysine, citrulline, N2 -succinyl-L-ornithine, and (5 -L-Glutamyl)-L-glutamate). Furthermore, the inhibitory effect of LP-CBT on colorectal cancer was further confirmed by the MC38 subcutaneous tumor model. Collectively, these findings offer compelling evidence to support the potential of LP-CBT as a viable preventive strategy against CRC.

Food allergy is a serious public health problem because of its high incidence and risk. Probiotics can induce immune regulation in patients with allergic diseases, but its mechanism is not fully clear. In this paper, β-lactoglobulin (β-LG)-sensitized mice were used as models to explore the mechanism of Bifidobacterium animalis KV9 (KV9) and Lactobacillus vaginalis FN3 (FN3) on reducing allergic reactions and regulating immune cell function. The results showed that oral administration of KV9 and FN3 significantly reduced the scores of allergic symptoms, hypothermia symptoms, and serum levels of β-LG-specific immunoglobulins E (β-LG-sIgE), histamine, and mast cell protease in allergic mice. Flow cytometry analysis of intestinal dendritic cells (DCs) showed that the proportion of CD11c+major histocompatibility complex (MHC)-II+DCs, CD11c+CD80+DCs, and CD11c+ CD86+DCs increased after KV9 and FN3 intervention, indicating that the strains induced immature DCs and decreased the antigen-presenting capacity of DCs. Meanwhile, the toll-like receptor 4 (TLR4)-NF-κB signaling pathway was activated in DCs. The secretion of interleukin-12 (IL-12) was significantly increased, while interleukin-4 (IL-4) was decreased by DCs after KV9 and FN3 intervention, indicating that DCs have the potential to promote T-cell differentiation into T helper type 1 (Th1) cells. Furthermore, the proportion of CD3+CD8−IFN-γ+ T cells in the spleen increased, while CD3+CD8−IL-4+T cells decreased after oral administration of KV9 and FN3, correcting the T helper type 2 (Th2)-skewed immune responses. These results indicate that KV9 and FN3 reduce β-LG-induced allergic symptoms in mice, and suggest that the two potential probiotics might be used as an alternative therapeutic agent for mitigating food allergy.

Medical applications of nanotechnology are promising in creating efficient and targeted therapies. However, so far, nanodrug design has not taken into consideration possible effects on human microbiota. The beneficial functions of bacteria could be stimulated by nanodrugs while negative effects on beneficial bacteria could cause risks to human health. Here, simulated intestinal fluid (IF) was optimized for culturing a human commensal and probiotic bacterial strain,Lactobacillus casei, to study the effects of medically relevant NPs—Ag and hyaluronic acid-coated Au NPs (HA-Au NPs)—in conditions pertinent to the gastrointestinal tract. When cultivated either aerobically or anaerobically, the specific growth rates ofL. caseiwere ~0.2 h−1in IF and ~0.4 h−1in the standard medium of lactobacilli (MRS). Ag NPs inhibited the growth ofL. caseiin IF at lower concentrations (EC50~ 65 and 15 mg/L in aerobic and anaerobic conditions, respectively) than in MRS (EC50> 100 mg/L), likely caused by differences in the composition of the two media and different intrinsic growth rates of bacteria in IF and MRS. Ag NP dissolution in IF and MRS did not explain the differences in growth inhibition, implying NP-specific effects. HA-Au NPs were not growth-inhibitory toL. caseiup to 250 mg/L. Still, both NPs at sub-growth-inhibitory concentrations suppressed the expression of bacteriocin genes inL. casei, suggesting an inhibitory effect of NPs on the probiotic properties ofL. casei, i.e., its competitiveness in microbial communities. However, HA-Au NPs did not appear to affect or even stimulated the immunomodulatory properties ofL. caseiin human intestinal epithelial cells. Thus, medically relevant NPs at low, sub-bacteriostatic levels can affect the metabolism of beneficial human bacteria and potentially induce changes in the microbiota and immune signaling. Keywords:commensal bacteria;microbiota;bacteriocin;Caco-2;intestinal fluid;antibacterial

Biogenic amines (BAs) are produced by microbial decarboxylation in various foods. Histamine and tyramine are recognized as the most toxic of all BAs. Applying degrading amine enzymes such as multicopper oxidase (MCO) is considered an effective method to reduce BAs in food systems. This study analyzed the characterization of heterologously expressed MCO fromL. sakeiLS. Towards the typical substrate 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), the optimal temperature and pH for recombinant MCO (rMCO) were 25 °C and 3.0, respectively, with the specific enzyme activity of 1.27 U/mg. Then, the effect of different environmental factors on the degrading activity of MCO towards two kinds of BAs was investigated. The degradation activity of rMCO is independent of exogenous copper and mediators. Additionally, the oxidation ability of rMCO was improved for histamine and tyramine with an increased NaCl concentration. Several food matrices could influence the amine-oxidizing activity of rMCO. Although the histamine-degrading activities of rMCO were affected, this enzyme reached a degradation rate of 28.1% in the presence of surimi. Grape juice improved the tyramine degradation activity of rMCO by up to 31.18%. These characteristics of rMCO indicate that this enzyme would be a good candidate for degrading toxic biogenic amines in food systems. Keywords:Lactobacillus sakei;biodegradation;multicopper oxidase;food matrices;biogenic amines

Previous studies have shown that Lactobacillus species play a role in ameliorating colorectal cancer (CRC) in a mouse model. However, the underlying mechanisms remain largely unknown. Here, we found that administration of a probiotic strain, Lactobacillus plantarum L168 and its metabolite, indole-3-lactic acid, ameliorated intestinal inflammation, tumor growth, and gut dysbiosis. Mechanistically, we indicated that indole-3-lactic acid accelerated IL12a production in dendritic cells by enhancing H3K27ac binding at the enhancer regions of IL12a that contributed to priming CD8+ T cell immunity against tumor growth. Furthermore, indole-3-lactic acid was found to transcriptionally inhibit Saa3 expression related to cholesterol metabolism of CD8+ T cells through changing chromatin accessibility and subsequent enhancing function of tumor-infiltrating CD8+ T cells. Together, our findings provide new insights into the epigenetic regulation of probiotics-mediated anti-tumor immunity and suggest the potential of L. plantarum L168 and indole-3-lactic acid to develop therapeutic strategies for patients with CRC.

Immunosuppression is a manifestation imbalance in the immune system, often during unhealthy states. In recent years, lactic acid bacteria (LAB) have been found to be important components of the body's innate immune system, and indispensable to maintaining normal immune function. Lactobacillus plantarum BF_15, a novel strain isolated from the feces of breast-fed infants, which has shown potential as an immunomodulator in vitro. In the present study, with the Polymerase Chain Reaction-Denaturing Gradient Gel Electrophoresis (PCR-DGGE) based on RNA-polymerase beta subunit encoding gene (rpoB) to analyze the colonization of L. plantarum BF_15 in the intestine of mice. In addition, Lactobacillus rhamnosus GG (LGG) as a positive control strain, by measuring immune-related indexes and the diversity of intestinal microbiota, the effects of BF_15 on immunoregulation and intestinal microbiota dysbiosis were investigated to elucidate whether the attenuation of immunosuppression is related to the modulation of intestinal microbiota. Results did indeed support this notion that BF_15 did colonize murine intestines well, in which it could still be detected in mice feces 14 days after stopping the probiotic administration. Moreover, BF_15 found to protect mice against reduction in the levels of several immune-related indicators, including the thymus and spleen indexes, splenic lymphocyte proliferation, toe swelling degree, serum hemolysin-antibody level, and macrophage phagocytosis index, triggered by high-dose (200 mg kg−1) intraperitoneal administration of cyclophosphamide (CTX). In addition, the strain was also found to effectively balance intestinal microbiota dysbiosis in the mice. Collectively, these results indicated that L. plantarum BF_15 can not only successfully colonize murine intestines, but also can effectively alleviate CTX-induced immunosuppression, once established, by rebalancing the intestinal microbiota. This, therefore, provides strong evidence for the view that BF_15 has the potential to become a highly effective immunomodulating probiotic in human microbiota as well.

Dicaffeoylquinic acids (DiCQAs), the main components of kudingcha made from the leaves of Ilex kudingcha, could be transformed by gut microbiota. However, the information about the related microorganisms and enzymes involved in the biotransformation of DiCQAs in the human gut is limited. Therefore, a strain of bacteria that could hydrolyze DiCQAs, belonging to Lactobacillus fermentum named L. fermentum LF-12, was isolated from human feces in the present study. Furthermore, an esterase for the hydrolysis of DiCQAs was purified from L. fermentum LF-12 and heterogeneously expressed in Escherichia coli. The esterase could be induced to exert superior hydrolytic activity in the presence of lactose as the carbon source. The molecular weight of the purified esterase was determined to be 31.9 kDa, and the isoelectric point, optimal pH and temperature for the esterase were 4.71, 6.5 and 45 °C, respectively. The enzyme activity was improved by Mg2+ and Ca2+, and reduced by Co2+, Cu2+, EDTA and some kinds of organic solvents. The present results provide new insights into the metabolism of DiCQAs by the human gut.

Enzymatic NADP + regeneration is a promising approach to produce valuable chemicals under economic conditions. Among all the enzymatic routes, using water-forming NADH oxidase is an ideal one because there is no by-product. However, most NADH oxidases have a low specific activity to NADPH. In this work, a thermostable NADH oxidase from Lactobacillus rhamnosus (LrNox) was rationally engineered to switch its specificity from NADH to NADPH. The results show that mutants D177A, G178R, D177A/G178R, D177A/G178R/L179S improved the NADPH activity by a factor of 4–6. The highest NADPH catalytic efficiency ( K cat /K m 223.71 S −1 μm −1 , 47.6-fold higher than wild-type LrNox) and 51% of NADH activity retention were achieved by replacing the single amino acid Leu179 for serine (L179S) in LrNox. Modeling of L179S-NADPH complex reveals that the phosphate group of NADPH interacts with the hydroxyl of Ser179 with a strong hydrogen bond and several shorter hydrogen bonds with the amino group of Lys185 could stabilize the binding of NADPH in the L179S mutant. This work provides an efficient method for converting NAD(P)H specificity and shows that L179S mutant is a potential and efficient auxiliary enzyme for NADP + regeneration.

This study investigated the physicochemical characteristics and antioxidant activities of cell-bound exopolysaccharides (c-EPS) from Lactobacillus fermentum S1 extracted using either phenol (CFP) or ethylenediaminetetraacetic acid (CFE). The extraction products, CFP and CFE, were homogeneous polysaccharides with molecular weights of 2.89 × 10 5 and 2.44 × 10 5 Da, respectively. Both CFP and CFE were composed of five kinds of monosaccharides. They had a main chain of β-D-Glc p -(1→2), α-L-Rha p -(1→2), β-D-Gal p -(1→3), α-D-Glc p- (1→3), α-D-Gal p -(1→3) and β-L-Rha p- (1→3,4) residues, and a tail of α-D-Man p -(1→) residue, but with different molar ratios. Trace amounts of D-Gal p- (1→) and L-Ara f -(1→2,5) residues were randomly distributed in the sugar chains. Although both CFP and CFE were in triple-helical conformation, they differed from each other in relation to surface morphology and thermal properties. The two c-EPS had different performances in radical scavenging and ferric reducing antioxidant power assays in vitro . The antioxidant capacity of both c-EPS extracts was confirmed in vivo using Caenorhabditis elegans , whose total antioxidant capacity and total superoxide dismutase activity increased after c-EPS treatment. Overall, the extraction method significantly affected the structure and biological activity of c-EPS. Moreover, the potent antioxidant activities of CFP and CFE support their use as natural antioxidants in dietary supplements or food additives.

Probiotics are believed to provide benefits to human health; however, a good storage stability is the prerequisite for the probiotic products making function. Herein, we reported a pathway to fabricate the Ca-alginate/cryoprotectants/cellulose composite (ACFP) capsules to protect L. plantarum cells, which showed the minimal loss of viability during the vacuum freeze-drying process. The trehalose and whey protein isolate (WPI) ingredients in the cryoprotectants prolonged the dissolution time of Ca-alginate shell, which contributed to controlling the release of cells in the desired region. The dry ACFP capsules exhibited gradual release of L. plantarum cells in simulated intestinal fluid (SIF), 2.6 × 10 6 cfu/mL at 210 min. In addition, 0.1 g of the dry ACFP capsules showed the viable release amount of 3.3 × 10 6 cfu/mL after the storage of 160 days at 4 °C. The promising results provided a strategy of encapsulating probiotic cells to achieve long-term storage stability and enhanced controlled release behavior in simulated intestinal fluid in the meantime.
