Purpose The aim of this study was to develop an “all-in-one” nanoplatform that integrates at the second near-infrared (NIR-II) region dye IR1061 and anticancer drug paclitaxel (PTX) into an apoferritin (AFN) nanocage (IR-AFN@PTX). Simultaneously, folic acid (FA), tumor target molecule, was conjugated onto IR-AFN@PTX to be IR-AFN@PTX-FA for tumor-targeted and pH/NIR-II-triggered synergistic photothermal-chemotherapy.Methods IR1061 was firstly reacted with PEG and then conjugated with AFN to be IR-AFN. Then, FA was conjugated onto the surface of IR-AFN to be IR-AFN-FA. At last, PTX was incorporated into IR-AFN-FA to fabricate a nanoplatform IR-AFN@PTX-FA. The NIR-II photothermal properties and pH/NIR-II triggered drug release were evaluated. The ability of IR-AFN@PTX-FA to target tumors was estimated using optical bioluminescence. In vitro and in vivo synergistic therapeutic effects of pH/NIR-II-triggered and tumor-targeted photothermal-chemotherapy were investigated in 4T1 tumor model. Results IR-AFN@PTX-FA showed excellent water solubility and physiological stability, which significantly enhanced the solubility of both IR1061 and PTX. After 5 min of laser irradiation at 1064 nm, IR-AFN@PTX-FA exhibited an effective photothermal effect compared with laser irradiation at 808 nm, even when blocked with 0.6 cm thick chicken breast. Cellular uptake experiments showed IR-AFN@PTX-FA utilized clathrin-mediated and caveolae-mediated endocytosis pathways to enter 4T1 cells, and was then delivered by the endosome to the lysosome. NIR-II laser irradiation and pH could synergistically trigger PTX release, inducing significant tumor inhibition in vitro and in vivo. Conclusion As a novel “all-in-one” nanoplatform, IR-AFN@PTX-FA was found to selectively target tumors and showed very efficient NIR-II photothermal effects and pH/NIR-II triggered drug release effects, showing a remarkable, synergistic photothermal-chemotherapy effect.

Near-infrared-II (NIR-II) fluorescence imaging-guided photothermal therapy (PTT) represents a cutting-edge approach for precise tumor diagnosis and treatment, providing real-time therapeutic efficacy evaluation. However, a significant challenge lies in the creation of phototheranostic agents that provide robust imaging and efficient photothermal conversion. To address this, a donor- π -acceptor (D- π -A) structure heptamethine cyanine derivative, IR1116, that confers a strong intramolecular charge transfer effect is developed. To enhance its applicability, IR1116 is formulated with DSPE-PEG to create a water-dispersible NIR-II phototheranostic nanoheater, NP IR1116 . This nanoheater benefits from the incorporation of an electron-withdrawing group, leading to reduced photooxidation activity and significantly improved photostability. NP IR1116 exhibits strong NIR-II absorption and fluorescence, peaking at 1004 and 1116 nm, respectively, as well as a photothermal conversion efficiency of 79% under 1064 nm lasers. In vitro and in vivo studies showed the efficacy of NP IR1116 in tumor imaging via NIR-II fluorescence and its ability to effectively induce tumor cell apoptosis under 1064 nm laser irradiation. These findings underscore the potential of NP IR1116 in NIR-II fluorescence imaging-guided PTT for tumor treatment, paving the way for further advancements in NIR-II dye development and bioimaging technologies.

The integration of chemodynamic therapy (CDT) and photothermal therapy (PTT) has played a huge role in improved anticancer treatments. Here, a novel multifunctional nanoplatform based on Cu2−xS conjugated NaYF4:Yb/Er@NaYF4:Yb upconversion nanoparticles (UCNPs) was proposed and designed. In the UCNPs–Cu2−xS nanocomposites, UCNPs with excellent luminescent properties and a high X-ray attenuation coefficient can serve as an upconversion luminescence (UCL) and computer tomography (CT) imaging contrast agent; meanwhile, Cu(II) in the Cu2−xS nanodots enables the nanocomposites to have a magnetic resonance imaging (MRI) ability owing to the presence of unpaired electrons. Moreover, the Cu2−xS nanodots with a strong absorbance in the NIR II biowindow not only could be employed as a stable photothermal agent under NIR laser irradiation, but also could be used as a photothermal-enhanced Fenton nanocatalyst to respond to over-expressed H2O2 in the tumor microenvironment (TME) and generate toxic hydroxyl radicals (˙OH) to effectively kill cancer cells. Furthermore, the UCNPs–Cu2−xS nanocomposites possess negligible cytotoxicity and a high photothermal conversion efficiency (43.8%) in the NIR-II biowindow (1064 nm), indicating that they possess great potential for the UCL/CT/MR multi-modal imaging guided synergistic enhanced CDT/PTT of cancer.

In this research, a fluorescent probe of 7-(diethylamine) coumarin derivatives with multiple binding sites to detect biothiols in tumor cell with strong NIR II luminescence in vivo was synthesized. The biothiols include cysteine (Cys) and glutathione (GSH) in tumor cells, and the tumor-response luminescence was proved by the cell experiment. Importantly, the monolayer functional phospholipid (DSPE-PEG) coating and aggregation induced emission (AIE) dye of TPE modification made the probe have good stability and biocompatibility with little luminescence quenching in aqueous phase, which was proved by in vitro and in vivo experiments. The final aqueous NIR II probe combined with bevacizumab (for VEGF recognition in the cancer cells) and Capmatinib (for Met protein recognition in the cancer cells) has stronger targeted imaging on head and neck squamous cell carcinoma (HNSCC) cancer with intravenous injection. This GSH/Cys detection in the tumor cell and strong dual-molecular NIR II bioimaging in vivo may provide new strategy to tumor detection.

Narrow bandgap Ag 2 Te quantum dot (QDs) hold great promise in the fields of bioimaging, chemo-/bio-detection, and thermoelectric materials, but their biosafety has not been well estimated yet. In this study, we studied the biodistribution and toxicity of polyethylene glycol–coated Ag 2 Te QDs (Ag 2 Te QDs-PEG) in mice after a single intravenous injection. After entering the mice, Ag 2 Te QDs-PEG displayed a short circulation time in the blood (half-life was less than 2 h), and mainly accumulated in the liver and spleen. The stability of Ag 2 Te QDs-PEG in the mice depended on accumulation organ and time. Ag 2 Te QDs-PEG were stable in the spleen within 28 days but decomposed in the liver after 7 days. The decomposed Ag 2 Te QDs-PEG were eliminated from the body via different routes, as Ag was detected only in feces while Te was found only in urine. Meanwhile, Ag 2 Te QDs-PEG did not show obvious adverse effects on the body weights, organ indices, biochemical parameters, the levels of essential metal Cu and Zn in organs, and organ pathological investigations during 28 days. In addition, the important effects of anion to the fates of silver chalcogenide QDs in vivo was demonstrated by comparing the distributions of the Ag 2 Te QDs and other two silver chalcogenide QDs (Ag 2 S and Ag 2 Se). Our findings indicate Ag 2 Te QDs have good biocompatibility in mice, which is a guarantee for their safe applications in the future.

Extensive efforts have been devoted to the design of organic photothermal agents (PTAs) that absorb in the second near-infrared (NIR-II) bio-window, which can provide deeper tissue penetration that is significant for phototheranostics of lethal brain tumors. Herein, the first example of NIR-II-absorbing small organic molecule (N1) derived from perylene monoamide (PMI) and its bio-application after nano-encapsulation of N1 to function as a nano-agent for phototheranostics of deep orthotopic glioblastoma (GBM) is reported. By adopting a dual modification strategy of introducing a donor-acceptor unit and extending π-conjugation, the obtained N1 can absorb in 1000–1400 nm region and exhibit high photothermal conversation due to the apparent intramolecular charge transfer (ICT). A choline analogue, 2-methacryloyloxyethyl phosphorylcholine, capable of interacting specifically with receptors on the surface of the blood-brain barrier (BBB), is used to fabricate the amphiphilic copolymer for the nano-encapsulation of N1. The obtained nanoparticles demonstrate efficient BBB-crossing due to the receptor-mediated transcytosis as well as the small nanoparticle size of approximately 26 nm. The prepared nanoparticles exhibit excellent photoacoustic imaging and significant growth inhibition of deep orthotopic GBM. The current study demonstrates the enormous potential of PMI-based NIR-II PTAs and provides an efficient phototheranostic paradigm for deep orthotopic GBM.

Lanthanide-doped upconversion nanoparticles (UCNPs) are promising bioimaging nanoprobes due to their excellent photostability. As one of the most commonly used lanthanide activators, Tm3+ions have perfect ladder-type electron configuration and can be directly excited by bio-friendly near-infrared-II (NIR-II) wavelengths. Here, the emission characteristics of Tm3+-doped nanoparticles under laser excitations of different near-infrared-II wavelengths were systematically investigated. The 1064 nm, 1150 nm, and 1208 nm lasers are proposed to be three excitation strategies with different response spectra of Tm3+ions. In particular, we found that 1150 nm laser excitation enables intense three-photon 475 nm emission, which is nearly 100 times stronger than that excited by 1064 nm excitation. We further optimized the luminescence brightness after investigating the luminescence quenching mechanism of bare NaYF4: Tm (1.75%) core. After growing an inert shell, a ten-fold increase of emission intensity was achieved. Combining the advantages of NIR-II wavelength and the higher-order nonlinear excitation, a promising facile excitation strategy was developed for the application of thulium-doped upconversion nanoparticles in nanoparticles imaging and cancer cell microscopic imaging. Keywords:upconversion nanoparticles;near-infrared-II;excitation mechanisms;luminescence quenching;microscopic imaging

The second near-infrared window (NIR-II, 1000-1700 nm) phosphor-converted LEDs (pc-LEDs) show great potential for portable spectroscopic detection. However, high-performance long-wavelength (>1350 nm) NIR-II phosphors that combine broadband emission, high thermal stability, and efficient blue-light excitation, are still relatively scarce. Herein, a Lu 3 Ga 5 O 12 :Cr 3+ ,Ni 2+ phosphor was developed via a Cr 3+ sensitization strategy. Cr 3+ and Ni 2+ stably occupy the [GaO 6 ] octahedral sites, enabling efficient energy-transfer from Cr 3+ to Ni 2+ . The optimal sample exhibits broadband NIR-II emission at 1427 nm with an FWHM of 277 nm, an EQE of 23.1%, and excellent thermal stability (I 373K /I 293K = 81.5%). The pc-LED based on this NIR-II phosphor achieves penetration imaging and non-destructive identification of daily necessities. More notably, rapid spectral differentiation of common plastics (PA, PE, PP, PVC and PET) was successfully realized using the NIR-II luminescence of the as-prepared phosphor. This work provides guidance for constructing high-performance NIR-II phosphors and promotes the practical deployment of NIR spectroscopy in resource recycling.

Luminescent thermometry in the second near-infrared (NIR-II) window holds significant promise for non-invasive. However, its practical application is severely hindered by thermal quenching at elevated temperatures, which limits the development of highly sensitive NIR-II thermometers capable of thermally enhanced emission. Here, we present a rationally designed integer phonon-matching strategy that enables NIR thermal enhancement specifically in singly Er 3+ -doped Ca 2 YNbO 6 double perovskite hosts. Through combined experimental investigations and rate equation modeling, we reveal that efficient multi-phonon relaxation (MPR) between the Er 3+ : 4 I 11/2 and 4 I 13/2 levels is activated when the electronic energy gap corresponds to an integer multiple ( n ≤ 5) of the dominant phonon energy. This resonant phonon-matching condition facilitates highly efficient MPR with increasing temperature, leading to a pronounced 1.66-fold enhancement in the Er 3+ NIR-II emission. Furthermore, we demonstrate the simultaneous occurrence of NIR thermal enhancement and ratiometric temperature sensing in Ca 2 YNbO 6 : Yb 3+ /Er 3+ /Ho 3+ /Pr 3+ crystals, achieving an exceptional relative sensitivity of 0.87% K −1 at 398 K. These findings not only circumvent the thermal quenching limitations of NIR-II phosphors but also establish phonon engineering as a versatile strategy for designing thermally enhanced luminescence, opening new avenues for flexible monitoring of pipeline integrity and organic compound leakage based on differential NIR-II luminescence absorption.

Recurrence of solid tumors after surgical resection is a major barrier to tissue regeneration. As an emerging treatment strategy, photo-thermo-electric therapy ablates tumor cells via photothermal effects and generates reactive oxygen species (ROS) via thermoelectric effects to disrupt heat shock proteins, thereby suppressing their protective function in tumor cells. However, conventional materials suffer from low thermoelectric efficiency and weak tissue penetration ability. In this study, we fabricated iodine-doped bismuth sulfide (I-Bi 2 S 3 ) nanorods with bonding heterostructures to improve thermoelectric performance. The approach employed iodine doping to introduce additional electrons, thereby regulating the band structure of Bi 2 S 3 and exploiting the dual low-energy vibration effect of the heterostructures to reduce thermal conductivity. More importantly, controlling the type of heterostructure modulated the bandgap width, thereby expanding the light absorption range to the higher-penetration near-infrared (NIR)-II region for deep tissue treatment. The I-Bi 2 S 3 nanorods were incorporated into poly- l -lactic acid (PLLA) scaffolds to confer antitumor functionality. According to the results, the bonding heterostructures enhanced the conductivity of Bi 2 S 3 and reduced its thermal conductivity, significantly enhancing thermoelectric efficacy. The heterostructures reduced the bandgap of Bi 2 S 3 from 1.23 to 0.88 eV, enabling optical absorption in the NIR-II region. The ROS tests showed that the PLLA/I-Bi 2 S 3 scaffold exhibited good photothermal effects and ROS generation under 1064-nm laser irradiation. The antitumor efficacy of the PLLA/I-Bi 2 S 3 scaffold reached 84.6% against MG-63 cells, demonstrating its exceptional potential in cancer treatment.

Purpose Cervical cancer (CC) is still the fourth most common cause of cancer deaths in women. However, current biomedical imaging techniques exhibit inherent limitations in the diagnosis and treatment of CC. This study aims to develop a biomimetic nanoplatform based on tumor cell membranes, loaded with a palladium (Pd)-based computed tomography (CT) contrast agent and the near-infrared (NIR) fluorescent probe indocyanine green (ICG). This multifunctional nanoplatform is designed to integrate multimodal imaging with photothermal therapy (PTT), thereby improving the diagnostic accuracy and therapeutic efficacy against CC.Methods In this study, biomimetic nanoparticles (NPs), designated as M@Pd-ICG NPs, were synthesized by encapsulating Pd and ICG within HeLa cell membranes derived from cell-derived xenograft (CDX) models. Subsequently, the toxicity, biocompatibility, and tumor suppression capability of the M@Pd-ICG NPs were evaluated in vitro. In vivo, the multimodal imaging performance of the M@Pd-ICG NPs and their photothermal therapeutic efficacy under 808-nm laser irradiation were investigated in mouse model bearing subcutaneous cervical tumor. Results The M@Pd-ICG NPs were successfully prepared and exhibited favorable stability, excellent photothermal conversion efficiency (34.04%), and good biocompatibility, enabling homologous targeting and prolonged circulation time. The M@Pd-ICG NPs integrated the complementary advantages of NIR-II fluorescence imaging (900–1700 nm, NIR-II FI), photothermal imaging (PTI), and CT imaging. Both in vitro and in vivo studies demonstrated that, under 808-nm laser irradiation, M@Pd-ICG NPs induced significant photothermal effects and tumor ablation. Conclusion M@Pd-ICG NPs successfully integrate multimodal imaging and PTT, owing to their excellent targeting capability and good biocompatibility, demonstrating potential for further biomedical applications.

Near-infrared II (NIR-II, 1000–1700 nm) fluorescence
imaging
enables deep-tissue analysis, but conventional “always-on”
probes suffer from high background that compromises the signal-to-background
ratio for low-abundance targets. Activatable probe strategies could
mitigate this issue, yet most existing designs rely on fluorescence
resonance energy transfer with acceptors confined to the visible region,
hampering efficient signal quenching in the NIR-II window. Here, we
present a generalizable strategy based on morphology-engineered nanometal
surface energy transfer (NSET) to construct a NIR-II core–satellite
nanoprobe for intracellular miR-21 detection. By systematically tuning
the branch aspect ratio of gold nanostars (AuNSs), their localized
surface plasmon resonance (LSPR) absorption can be red-shifted to
achieve precise spectral overlap with the NIR-II emission of Ag 2 S quantum dots. Guided by DNA self-assembly, an AuNS@Ag 2 S core–satellite structure with controllable interparticle
spacing is established, enabling an initial fluorescence quenching
efficiency of up to 88%. Upon target recognition, miR-21-triggered
strand displacement reaction induces the dissociation of Ag 2 S quantum dots from the AuNS surface, resulting in a pronounced NIR-II
fluorescence “turn-on” response. The nanoprobe exhibits
exceptional performance, including a low detection limit of 75.8 pM,
robust linearity, single-base mismatch discrimination capability,
and enables visualization of differential miR-21 expression in living
cells. Notably, high-contrast NIR-II signals are retained at penetration
depths of up to 15 mm in tissue-mimicking experiments. This work establishes
an NSET-based analytical platform through precise spectral matching
and rational structural engineering, providing a robust strategy for
high signal-to-background biosensing and in situ analysis of low-abundance
miR-21 in deep tissues.
