A three-layer small-diameter vascular graft (SDVG) of polycaprolactone (PCL) and l -polylactic acid (PLLA) with pore diameter gradient was prepared through combining variable-extrusion speed iterative electrospinning and ultrasonic reaming. The SEM results showed that the three-layer PCL/PLLA SDVG has distinct pore diameter gradient. The Young’s modulus, ultimate tensile strength (UTS), burst pressure (BP) and compliance of the vascular graft were high as 20.116 ± 0.465 MPa, 2.259 ± 0.141 MPa, 2050 ± 249 mmHg and 4.203 ± 0.330 %/100 mmHg, respectively. The UTS of the graft is similar to human coronary arteries. The BP meets the basic requirements to prevent aneurysmal dilation, and the compliance is higher than that of sheep carotid artery. The fluid–solid coupling simulation results showed that the graft had high mechanical properties to withstand the same blood flow velocity as human coronary arteries, and there was no stress concentration in the wall. Besides, it had good biocompatibility, promoting the adhesion and proliferation of rat vascular endothelial cells (ECs). The successful preparation of the small-diameter vascular graft with high mechanical properties for preventing itself from rupturing and ensuring the overall function, provides more possibilities for the clinical transplantation of small-diameter vascular grafts.

Asymmetrical dressings, which are composed of a compact top layer and a porous bottom layer, are commonly used to mimic the characteristics and structure of the epidermis and dermis layers, and overcome the flaws of traditional dressings such as wound dryness and bacterial penetration. Herein, a bio-inspired double-layer asymmetric wettable wound dressing was prepared by low-temperature 3D printing coupled with electrospinning technology. The hydrophobic top layer of poly(caprolactone)(PCL) film produced by electrospinning was used to simulate the compact and air-permeable epidermis. The hydrophilic bottom layer of the dressing, a scaffold composed of chitosan and copper ions doped Laponite (Cu@CS-Lap) was used to kill bacteria and speed up wound healing. Additionally, the composite dressings also showed excellent cytocompatibility and antibacterial properties in vitro experiments. The migratory area of Cu-doped group human umbilical vein endothelial cells increased by about 48.19% compared to the control group, as revealed by the results of the cell scratch experiment. Furthermore, in vivo experiments in rats showed that wound closure at the 0.5Cu@CS5-PCL dressing reached 98.24% after 12 days, indicating the enormous potential of asymmetric double dressings in boosting wound healing.

A series of polymer brushes with degradable poly(caprolactone) (PCL) backbones were synthesized via “grafting onto” approach by combination of the ring-opening polymerization (ROP) and “click” reaction. First, P(αClCL) was synthesized by ROP and then the chloride was substituted by azide group to obtain P(N 3 CL) backbone; then polymer brushes were synthesized by “click” reaction of alkynyl-PCL (ay-PCL) and alkynyl-PEO (ay-PEO) onto P(N 3 CL). The products were characterized by gel permeation chromatography (GPC), FT-IR and 1 H NMR. Results indicated that the ay-PCL can be grafted onto P(N 3 CL) backbone quantitatively at 1 : 1 ratio of alkynyl group to azide group and the branch was attached onto each repeating unit because of lower steric resistance and graft density. Also the amphiphilic polymer brushes with various hetero-branches of PEO/PCL ratios were synthesized by one-pot two-step process. The amphiphilic polymer brushes self-assembled into micelles with various PEO/PCL ratios and have potential application in drug loading and delivery.

Development of efficient ocular drug delivery systems was still a challenging task. The objective of this article was to develop a thermosensitive PEG–PCL–PEG (PECE) hydrogel and investigate its potential application for ocular drug delivery of diclofenac sodium (DIC). PECE block polymers were synthesized by coupling MPEG-PCL co-polymer using IPDI reagent, and then its sol–gel transition as a function with temperature was investigated by a rheometer. The results showed that 30% (w/v) PECE aqueous solution exhibited sol–gel transition at approximately 35 °C. In vitro release profiles showed the entrapped DIC was sustained release from PECE hydrogels up to 7 days and the initial drug loading greatly effect on release behavior of DIC from PECE hydrogels. MTT assay results indicated that no matter PECE or 0.1% (w/v) DIC-loaded PECE hydrogels were nontoxic to HCEC and L929 cells after 24 h culturing. In vivo eye irritation test showed that the instillation of either 30% (w/v) PECE hydrogels or 0.1% (w/v) DIC-loaded PECE hydrogels to rabbit eye did not result in eye irritation within 72 h. In vivo results showed that the AUC0–48 h of 0.1% (w/v) DIC-loaded PECE hydrogels exhibited 1.6-fold increment as compared with that of commercial 0.1% (w/v) DIC eye drops, suggesting the better ophthalmic bioavailability could be obtained by the instillation of 0.1% (w/v) DIC-loaded PECE hydrogels.

Repairing critical alveolar bone defects in inflammatory microenvironments is challenging. To address this, dual-functional nanofibrous scaffolds with anti-inflammatory and osteogenic properties were developed. These scaffolds incorporate polycaprolactone (PCL) short fibers and mesoporous bioactive glass nanoparticles (MBGNs), fabricated through electrospinning and sol-gel processes, respectively. 3D Fibrous scaffolds were achieved through sequential dispersion, homogenization, thermal self-assembly, and surface modification. During this process, gelatin facilitated the self-assembly of the scaffold, while carboxymethyl chitosan was used to integrate MBGN into the PCL scaffolds. Characterization confirmed a porous structure of scaffolds and enhanced mechanical and in vitro acellular properties after MBGN incorporation. Non-cytotoxicity was confirmed by CCK-8 and Live/Dead assays. Flow cytometry showed scaffold-induced macrophage polarization from M1 (pro-inflammatory) to M2 (anti-inflammatory), supported by qRT-PCR and ELISA, which showed reduced pro-inflammatory cytokines and increased anti-inflammatory cytokines. In co-culture with human bone marrow mesenchymal stem cells, the scaffolds promoted osteogenic differentiation and mineralization, as shown by ALP/ARS staining and increased expression of osteogenic markers (RUNX2, OCN). These findings highlight the dual immunomodulatory and osteogenic functions of the scaffolds, suggesting their potential as a promising strategy for inflammatory alveolar bone regeneration.

Combining multiple drugs or biologically active substances for wound healing could not only resist the formation of multidrug resistant pathogens, but also achieve better therapeutic effects. Herein, the hydrophobic fluoroquinolone antibiotic ciprofloxacin (CIP) and the hydrophilic broad-spectrum antibiotic tetracycline hydrochloride (TH) were introduced into the coaxial polycaprolactone/gelatin (PCL/GEL) nanofiber mat with CIP loaded into the PCL (core layer) and TH loaded into the GEL (shell layer), developing antibacterial wound dressing with the co-delivering of the two antibiotics (PCL-CIP/GEL-TH). The nanostructure, physical properties, drug release, antibacterial property, and in vitro cytotoxicity were investigated accordingly. The results revealed that the CIP shows a long-lasting release of five days, reaching the releasing rate of 80.71%, while the cumulative drug release of TH reached 83.51% with a rapid release behavior of 12 h. The in vitro antibacterial activity demonstrated that the coaxial nanofiber mesh possesses strong antibacterial activity againstE. coliandS. aureus. In addition, the coaxial mats showed superior biocompatibility toward human skin fibroblast cells (hSFCs). This study indicates that the developed PCL-CIP/GEL-TH nanofiber membranes hold enormous potential as wound dressing materials. Keywords:coaxial nanofiber mesh;drug co-delivery;wound dressing;polycaprolactone;gelatin

Water-based polyurethane (WPU) provides a number of benefits over typical solvent-based polyurethane. WPU is more environmentally friendly, safe, and reliable. The WPU with polycaprolactone diol/polycarbonate diol (PCL/PCDL) complex soft segment was developed using a prepolymer technique to enhance the color depth of cotton knitted fabrics dyed with reactive dyes. The appearance, particle size, zeta potential, and viscosity of WPU emulsions were analyzed. The chemical structure, thermal stability, phase change performance, and topography of WPU films were characterized by Fourier-transform infrared (FTIR), thermogravimetric analysis (TGA), differential scanning calorimetry (DSC), and atomic force microscopy (AFM), respectively. The tensile and hydrophilic properties of the WPU films were examined by an electronic universal testing machine and contact angle goniometer, respectively. The K / S value, rubbing fastness, washing fastness, and wearability of finished and unfinished dark cotton knitted fabrics were measured. Simultaneously, the chemical structure and morphology of fabrics were investigated using FTIR and scanning electron microscopy (SEM). The results show the WPU-2 emulsion demonstrated excellent stability, and the WPU-2 film showed superior thermal stability. The K / S value of the dark cotton knitted fabric finished with WPU-2 emulsion (40 g/ l ) increased by 18.58 %, and the wet rubbing color fastness of the resulting fabric was enhanced. The SEM results show a layer of film covered the surface of the finished fabric. In summary, the WPU can provide a new technique for enhancing the color depth of fabrics.

This study develops a PCL-HAP/CS bilayer membrane to address limitations in current bone regeneration materials. Traditional single-component materials often exhibit poor biocompatibility, mechanical mismatch, and insufficient osteogenic capacity. To overcome these issues, a composite structure was engineered by combining hydroxyapatite (HAP) synthesized via co-precipitation and silane-treated for enhanced compatibility, polycaprolactone (PCL) for mechanical strength, and chitosan (CS) for bioactivity. The bilayer design features a PCL-HAP layer fused with a CS layer, mimicking natural bone’s stratified architecture. Material characterization confirmed the composite’s structural integrity and functional properties. In vitro tests demonstrated selective cell compatibility: osteoblasts thrived on the mineral-rich PCL-HAP side, while fibroblasts showed enhanced adhesion on the CS layer. Rat skull defect experiments revealed dual functionality—the membrane provided mechanical support for bone regeneration while delivering antibacterial effects to surrounding soft tissues. The synergistic combination leverages PCL’s durability, HAP’s osteoinduction, and CS’s antimicrobial properties, effectively compensating for individual material weaknesses. This asymmetric membrane design shows significant potential for clinical bone repair applications, particularly in oral surgery, by simultaneously addressing hard tissue regeneration and soft tissue protection requirements.

In this study, astragalus polysaccharide / hydroxypropyl-β-cyclodextrin (APS@HP-β-CD) was efficiently loaded into polycaprolactone/chitosan (PCL/CS) by electrospinning and encapsulation techniques. Then, using Span80 as an emulsifier, PCL/CS/APS@HP-β-CD nanofibers were produced through electrospinning of water-in-oil emulsions. The physicochemical and antimicrobial properties of the nanofibrous materials were subsequently evaluated in vitro. Compared with the PCL/CS nanofiber membranes (PC membrane) without added active substances, the tensile test showed that the flexibility of the PCL/CS/APS@HP-β-CD nanofiber membrane (PCA membrane) was significantly enhanced. The thermodynamic test results show that the thermal stability of the PCA membrane is enhanced. The results of 1 H NMR, FTIR, and XRD showed that APS was successfully encapsulated in HP-β-CD. The biodegradation test results showed that the PC and PCA nanofiber membranes had no pollution to the environment and could biodegrade. In addition, PCA membranes exhibited strong antibacterial activity against both Gram-positive and Gram-negative bacteria as well as outstanding free radical scavenging capacity. The APS release mechanism was determined through the Higuchi and Korsmeyer-Peppas kinetic models. Among all the PCA nanofiber membranes, the APS release was more consistent with the Korsmeyer-Peppas kinetic model, and the transport mechanism was Fickian diffusion. Finally, this study also optimized the experimental protocol by introducing carboxymethyl groups to modify PCL/CS nanofiber membranes by chemical grafting. In conclusion, the PCA nanofibrous membranes prepared in this study have a strong potential to become wound dressings.

PCL nanofibers were prepared by needle-free electrostatic spinning using atmospheric atmospheric pressure plasma to pretreat Polycaprolactone (PCL) electrostatic spinning solution. The results show that the conductivity and acidity of the plasma-treated polymer solution increase significantly, and the viscosity increases and then decreases. The untreated and treated solutions were then subjected to electrostatic spinning, and it was found by scanning electron microscopy that the solutions obtained after plasma treatment had fewer beads on the surface of the fibers, and the nanofibers were more uniformly distributed in diameter and finer. It was also found that the plasma treatment affected the size of the molecular chain segments of the PCL, and the crystal structure of the fibers was altered. The XPS results showed that the surface elements of the nanofibers do not change before and after the plasma treatment, and the chemical bonding between the carbon and oxygen elements is consistent.

The combination of shape memory polymer composites and 3D printing technology provides novel opportunities for customized manufacturing and intelligentization of products. Here, we developed magneto-responsive shape memory polymer composites with well-balanced strengths and toughness by adding Fe 3 O 4 and cellulose nanofibers (CNFs) to a poly-hydroxybutyrate/poly(ε-caprolactone) blend as functional particles and reinforcing component. The influences of Fe 3 O 4 and CNFs on the properties including micromorphology, and rheological, melting-crystallizational, thermomechanical, static mechanical, and magneto-responsive shape-memory behaviors were investigated. The results displayed that the PHB/PCL (80:20) composites with 10 wt% Fe 3 O 4 and 0.5 wt% CNFs had the optimal comprehensive mechanical and magneto-responsive shape-memory properties. Moreover, 3D printing tests found that the addition of CNFs and printing parameters were crucial for the mechanical and magneto-responsive shape-memory properties of the resulting products. Finally, the fabrication of a scaffold with excellent load-carrying performance and a magneto-responsive self-recovery snowflake demonstrated the application feasibility of our work.

Aiming at achieving the concurrent performances of high loading, well controlled release and active targeted delivery, folate (FA) modified dual pH/reduction-responsive mixed polymeric micelles were rationally assembled using FA-PEG-PDEAEMA and PEG-SS-PCL by dissipative particle dynamics (DPD) simulations. The optimized polymers PEG112-PDEAEMA40, FA-PEG112-PDEAEMA40, and PEG112-SS-PCL70 were synthesized and characterized using 1H NMR, FT-IR and GPC, and their mixed micelles were applied for doxorubicin (DOX) delivery. The drug loading capacity (LC) and encapsulation efficiency (EE) values of the MIX1 (FA-PEG112-PDEAEMA40/PEG112-SS-PCL70) at a DOX/polymer feeding ratio of 15 mg/30 mg were 20.22% and 50.69%, which were higher than those of single polymer micelles and MIX2 (PEG112-PDEAEMA40/PEG112-SS-PCL70). Particle size distributions, mesoscopic morphologies, DPD simulations and in vitro drug release profiles all confirmed the well-controlled release performance of the DOX-loaded micelles formed by MIX1: slow DOX release with a cumulative release of 20.46% in the neutral environment and accelerated release with a cumulative release of 74.20% at pH 5.0 + 10 mM DTT within 120 h, which were similar to those of MIX2. Cytotoxicity assay found that both MIX1 and MIX2 blank micelles were biocompatible, and a superior inhibitory effect of the FA-modified DOX-loaded micelles MIX1 on HepG2 cells was found compared to that of free DOX and non-FA-modified DOX-loaded micelles MIX2. All of these confirmed the superiority of MIX1 micelles with high loading capacity, well controlled release, and enhanced inhibitory effects on HepG2 cells, which might be a prospective candidate for anticancer drug delivery.
