In this study, the interface-modified ternary blends based on polylactic acid (PLLA)-polycaprolactone (PCL)-polycarbonate diols (PCDL) were prepared via suspension flame spray. Morphological analysis showed that the interface modification of PCDL significantly improves the compatibility of PLLA and PCL. Mechanical analysis revealed that the addition of PCDL resulted in great enhancement of the toughness of the PLLA/PCL composite coatings with a dramatic 326 % increase of the fracture strain for 10PLLA/90PCL/10PCDL compared with 10PLLA/90PCL. Further physicochemical and antibacterial test revealed no significant difference among PLLA/PCL and PLLA/PCL/CAM composite coatings with and without PCDL. Time-course release analysis showed that the release mechanism of the composite coating generally remains the same when the PCDL added is lower than the main components in PLLA/PCL, and even changed from the original abnormal diffusion to Fick diffusion. Overall, the ternary blending approach of PLLA-PCL-PCDL using suspension flame spray is a promising technique with great potential drug-delivery applications with different mechanical requirements.

Introduction: Hierarchical nanofibrous scaffolds are emerging as a promising bone repair material due to their high cell adhesion activity and nutrient permeability. However, the existing method for hierarchical nanofibrous scaffolds fabrication is complicated and not perfectly suitable for further biomedical application in view of both structure and function. In this study, we constructed a hierarchical nanofibrous poly (l-lactic acid)/poly(ε-caprolactone) (PLLA/PCL) scaffold and further evaluated its bone healing ability. Methods: The hierarchical PLLA/PCL nanofibrous scaffold (PLLA/PCL) was prepared by one-pot TIPS and then rapidly mineralized at room temperature by an electrochemical deposition technique. After electrode-positioning at 2 V for 2 hrs, a scaffold coated with hydroxyapatite (M-PLLA/PCL) could be obtained. Results: The pore size of the M-PLLA/PCL scaffold was hierarchically distributed so as to match the biophysical structure for osteoblast growth. The M-PLLA/PCL scaffold showed better cell proliferation and osteogenesis activity compared to the PLLA/PCL scaffold. Further in vivo bone repair studies indicated that the M-PLLA/PCL scaffold could accelerate defect healing in 12 weeks. Conclusion: The results of this study implied that the as-prepared hydroxyapatite coated hierarchical PLLA/PCL nanofibrous scaffolds could be developed as a promising material for efficient bone tissue repair after carefully tuning the TIPS and electrodeposition parameters.

Epoxy thermosets have been widely used in engineering applications due to their excellent thermal stability and chemical resistance. However, their inherent brittleness causes low bonding strength. Therefore, improvement of toughness is required. By using a variety of modifiers, epoxy thermosets can be toughened via the generation of fine phase-separated morphologies. In this work, we reported the toughening of epoxy thermosets with polyhedral oligomeric silsesquioxane (POSS)-capped poly(ε-caprolactone)s (PCL) . By taking control of types of R groups ( i.e., isobutyl and/or isooctyl) of POSS cages and the lengths of PCL chains, the epoxy thermosets were regulated with the morphologies from nano-to macro-scales. It was found that the improvement of toughness was quite dependent on types of R groups of POSS cages. Under the identical condition, the thermosets contain isooctyl POSS-PCL n had the fracture toughened larger than those containing isobutyl POSS groups. For the thermoset containing 15 wt% of isooctyl POSS-PCL 1.00 , the critical stress intensity factor ( K IC ) was enhanced from 0.70 to 1.69 MPa m 1/2 at room temperature and from 1.11 to 2.51 MPa m 1/2 at low temperature and the thermal stability remained invariant. Furthermore, the bonding strength on stainless steel and anodized aluminum substrates were significantly improved with the incorporation of 15 wt% of isooctyl POSS-PCL 1.00 . These findings provide a novel approach to regulate phase structure in epoxy thermosets, offering the insights into structure-performance relationships, thus opening up a new way for designing epoxy resins with high toughness and adhesive strength.

Vascular basement membrane (VBM) is a thin layer of fibrous extracellular matrix linking endothelium, and collagen type IV (COL IV) is its main composition. VBM plays a crucial role in anchoring down the endothelium to its loose connective tissue underneath. For vascular grafts, constructing biomimetic VBMs on the luminal surface is thus an effective approach to improve endothelialization in situ . In the present work, three types of polycaprolactone (PCL) membranes were produced and characterized through cell counting kit-8 (CCK-8) assay, adhesion force and elastic modulus test to examine the influence of fiber diameter and membrane composition on vascular endothelial cell (EC) behaviors. The PCL membranes with finer fibers of 54.77 nm (PCL-54) could biomimic the nanotopography of VBMs more efficiently than 544.64 nm (PCL-544), and they were more suitable for Pig iliac endothelium cells (PIECs) adhesion and proliferation, meanwhile, inducing higher elastic modulus and adhesion force of PIECs. On this foundation, we further immobilized COL IV onto PCL-54 (PCL-COL IV) to biomimic VBMs compositionally. Results showed that PIECs on PCL-COL IV exhibited the highest viability and proliferation. Besides, quantitative data indicated that the elastic modulus of the PIECs on PCL-COL IV (4441.00 Pa) was as two times higher than that on PCL-54 (2312.26 Pa), and the adhesion force grew to 1120.99 pN from 673.58 pN of PIECs on PCL-54. In summary, the PCL-COL IV membranes show high similarity with the native VBMs in terms of structure and composition, suggesting a promising potential for surface modification to vascular grafts for improved endothelialization.

The purpose of this paper is to fabricate novel nanoparticles (NPs) from a single disulfide bond-bridged block copolymer poly(hydroxyethyl methacrylate)-S-S-polycaprolactone (PHEMA-S-S-PCL). The novel biomaterial was synthesized by ring-opening polymerization and reversible addition–fragmentation chain transfer polymerization. The cargo-free NPs were fabricated with the solvent evaporation method, and studies on NPs' characterizations were carried out. The hydrogen nuclear magnetic resonance ( 1 H NMR) and Fourier transform infrared spectroscopy spectra confirmed the synthesis of PHEMA-S-S-PCL copolymer. Thermo-gravimetric analysis curves indicated that the obtained PHEMA-S-S-PCL copolymer had good thermostability. Transmission electron microscopy and dynamic light scatter results suggested that the cargo-free NPs were in round shapes with an average diameter of 103.6 ± 0.12 nm. The low critical micelle concentration of cargo-free NPs (7.9 × 10 −4 mg/ml) indicated that these NPs would keep their spherical shapes after being attenuated by abundant liquid (e.g., blood or body fluid). Furthermore, these NPs showed high stability at the presence of bovine serum albumin. Therefore, it could be speculated that these NPs would not be absorbed by proteins in blood, and they could be used as a candidate carrier for drug delivery.

Tissue-engineered vascular grafts (TEVGs) have been proposed as a promising approach to fulfill the need for small-diameter blood vessel substitutes. However, common failure caused by thrombosis and neointimal proliferation after implantation has restricted their use in the clinic. Herein, a NO-generating scaffold for vascular tissue engineering was developed by coelectrospinning poly(ε-caprolactone) (PCL) with keratin. The morphology and surface chemical composition were characterized via SEM, ATR-FTIR spectroscopy and XPS. The biocomposite scaffold selectively enhanced the adhesion and growth of endothelial cells (ECs) while suppressing the proliferation of smooth muscle cells (SMCs) in the presence of GSH and GSNO due to the catalytic generation of NO. In addition, these mats displayed excellent blood compatibility by prolonging the blood-clotting time. In summary, these NO-generating PCL/keratin scaffolds have potential applications in vascular tissue engineering with rapid endothelialization and reduced SMC proliferation.

The differences in micro-environment between cancer cells and the normal ones offer the possibility to develop stimuli-responsive drug-delivery systems for overcoming the drawbacks in the clinical use of anticancer drugs, such as paclitaxel, doxorubicin, and etc. Hence, we developed a novel endosomal pH-sensitive paclitaxel (PTX) prodrug micelles based on functionalized poly(ethylene glycol)-poly(ε-caprolactone) (mPEG-PCL) diblock polymer with an acid-cleavable acetal (Ace) linkage (mPEG-PCL-Ace-PTX). The mPEG-PCL-Ace-PTX5with a high drug content of 23.5 wt % was self-assembled in phosphate buffer (pH 7.4, 10 mM) into nanosized micelles with an average diameter of 68.5 nm. The in vitro release studies demonstrated that mPEG-PCL-Ace-PTX5micelles was highly pH-sensitive, in which 16.8%, 32.8%, and 48.2% of parent free PTX was released from mPEG-PCL-Ace-PTX5micelles in 48 h at pH 7.4, 6.0, and 5.0, respectively. Thiazolyl Blue Tetrazolium Bromide (MTT) assays suggested that the pH-sensitive PTX prodrug micelles displayed higher therapeutic efficacy against MCF-7 cells compared with free PTX. Therefore, the PTX prodrug micelles with acetal bond may offer a promising strategy for cancer therapy. Keywords:prodrug;polymer micelles;pH-sensitive;acetal;paclitaxel

Bioactive synthetic scaffolds with 3D porous structure are the most attractive materials among various guided bone regeneration membranes. However, its osteogenic potential is still insufficient. This study was aimed to develop a Calcium surface-anchored Collagen I-PLGA/PCL scaffolds (PP/COL I-pDA-Ca) with enhanced osteogenicity. PP/COL I-pDA-Ca was electrospun from a collagen I-blended PLGA/PCL matrix and then modified by the chelation of Ca-ions via mussel-inspired polydopamine coating. PLGA/PCL, PLGA/PCL-polydopamine, PLGA/PCL-pDA chelated by Ca-ions were used as controls. Osteogenic effects of the scaffolds were examined using MC3T3-E1 cell culture. PP/COL I-pDA-Ca maintained 3D porous architecture with interconnected pores formed by randomly-oriented filamentous fibers and MC3T3-E1 cells cultured on it for 12 h or 24 h were more stretched and spread than those on the controls. PP/COL I-pDA-Ca significantly upregulated α10, α11 and β1 integrin expression after 48 h culture. ALP activity, OCN, OSX, BMP2 and RUNX2 expression of MC3T3-E1 cells cultured on PP/COL I-pDA-Ca scaffolds for 7 and 14 days were substantially enhanced when compared to controls ( p < 0.05). The pDA-based Ca chelation, COL I incorporation and 3D bionic structure promoted cell adhesion and osteogenic differentiation of MC3T3-E1 cells. This novel PP/COL I-pDA-Ca scaffolds is an attractive alternative for guided bone regeneration.

Multimodal optimization improved scaffold structure, mechanics, and bioactivity. • Low-dose BT generated a detectable but mild local piezoelectric response. • PTB4 achieved the best overall balance of printability and biological performance. • PTB4 enhanced osteogenic differentiation and VEGF-related endothelial responses. • PTB4 promoted osteogenesis–angiogenesis coupling and calvarial bone regeneration. Critical-size bone defects (CSDs) remain a major clinical challenge. Although three-dimensional (3D) printing enables precise structural control and defect-specific scaffold fabrication, conventional polymer-based scaffolds often exhibit insufficient mechanical strength and limited osteogenic activity. In this study, poly(ε-caprolactone)/β-tricalcium phosphate (PCL/β-TCP) scaffolds containing 0, 1, 2, 4, or 8 wt% barium titanate (BaTiO 3 , BT) were fabricated by direct ink writing. Their rheology, microstructure, mechanical properties, local electromechanical response, cytocompatibility, angiogenesis-related activity, osteogenic differentiation, and bone regeneration in vivo were evaluated. All formulations exhibited suitable printability and regular porous structures. Moderate BT incorporation increased surface roughness and mechanical strength, with compressive strength remaining within the range of human cancellous bone. Piezoresponse force microscopy confirmed a detectable but mild local piezoelectric response. However, its direct osteogenic contribution was not isolated from other BT-related material effects and requires further verification. All scaffolds exhibited good cytocompatibility. PTB4 showed the most favorable biological performance, including enhanced MC3T3-E1 cell proliferation, increased ALP-positive area, greater mineralized deposition, and more pronounced VEGF-related fluorescence in HUVECs. After 12 weeks in a rat calvarial critical-size defect model, PTB4 exhibited significantly higher bone volume fraction and bone mineral density than the blank and PT groups. Histological and immunohistochemical analyses showed more extensive new bone formation, collagen deposition, and stronger osteogenesis–angiogenesis coupling. Overall, PTB4 achieved the best balance among printability, mechanical compatibility, cytocompatibility, angiogenesis-related activity, and osteogenic performance, supporting low-dose BT modification as a promising strategy for safe, printable scaffolds for critical-size craniofacial and oral bone defect repair. This study introduces a new bioactive Janus membrane for guided bone regeneration (GBR) to treat periodontal bone defects. • Unlike conventional barrier membranes that lack long-term stability or biological function, our bilayer membrane, composed of poly(ε-caprolactone) (PCL), chitosan (CS), and the natural polyphenol proanthocyanidin (PA), combines mechanical strength with antioxidant, antibacterial, and anti-inflammatory properties. • Its asymmetric design, with in situ biomineralized hydroxyapatite particles (HAp) on a chitosan layer, supports bone growth on one side while preventing bacterial invasion on the PA-incorporated side. • This innovative approach addresses major challenges in periodontal tissue engineering and offers a promising platform for more effective and biologically intelligent therapies in clinical bone regeneration. The achievement of periodontal bone regeneration using a barrier membrane in guided bone regeneration (GBR) technology is often limited by either its inferior mechanical strength or bioactivity. To address this limitation, we developed a novel Janus bi-layer membrane by sequentially electrospinning poly(ε-caprolactone) (PCL) mixed with proanthocyanidin (PCL-PA) and chitosan (CS) in acetic acid, then biomineralized it in simulated body fluid (BSF) to form hydroxyapatite particles (HAp) in situ on a CS nanofiber layer (CS-HAp): PCL-PA/CS-HAp. This bi-layer membrane exhibited superior mechanical performance compared to single-layer membranes, with a peeling strength of 58 J/m 2 and tensile strength exceeding 5.0 MPa under hydrated conditions. In vitro antioxidant assays demonstrated the strong free-radical-scavenging ability of PCL-PA/CS-HAp. In addition, the PA-incorporated membranes exhibited greater than 95% bacterial inhibition against both Gram-negative E. coli and Gram-positive S. aureus . The bilayer membrane also alleviated oxidative stress induced by hydrogen peroxide (H 2 O 2 ), thereby promoting the proliferation of MC3T3-E1 cells, as monitored by cytoskeletal staining. Flow cytometry analysis showed that the PA-incorporated membrane can stabilize immunomodulatory effects by about 10% in lipopolysaccharides (LPS)-stimulated RAW264.7 cells. When used in a rat periodontal bone defect model, significant bone regeneration was observed in the PCL-PA/CS-HAp group, with a bone volume to total volume ratio of 36.2% and a significant reduction in bone defect size after 8 weeks, outperforming other treatment groups ( p < 0.001). The PCL-PA/CS-HAp bi-layer membrane represents a promising strategy for guided bone regeneration, effectively combining mechanical stability and bioactive properties. A bi-layer barrier membrane, PCL-PA/CS-HAp, prepared via electrospinning, was effective for periodontal bone regeneration in a rat model. Chemo-immunotherapy aims to integrate chemotherapy-induced antigen release with innate immune activation to achieve enhanced antitumor efficacy. However, the pharmacokinetic mismatch and systemic toxicity of free small molecules often compromise the synergistic effects. Herein, we propose a reactive oxygen species (ROS)-responsive nanoplatform based on mPEG–PCL for the co-delivery of a paclitaxel (PTX) prodrug and the TLR7/8 agonist R848. The nanoparticles remain stable under physiological conditions while releasing drug in oxidative tumor microenvironments, thereby achieving synchronous intratumoral delivery and activation. In vitro, the system exhibited efficient intracellular uptake and potent cytotoxicity against tumor cells while maintaining low toxicity toward normal cells due to prodrug design. Moreover, the platform effectively induced immunogenic cell death and promoted dendritic cell maturation, highlighting its capacity for chemo-immunotherapeutic synergy. This co-delivery strategy provides a versatile approach to realize controllable and safe chemo-immunotherapy through stimuli-responsive nanomedicine design.

Bone regeneration in compromised environments, such as infected or inflammatory defects, remains a significant clinical challenge. This study addresses the limitations of curcumin (Cur), including poor solubility, low bioavailability, and limited antibacterial activity, by developing a series of curcumin-metal complexes through chelation reactions. These complexes were encapsulated into nanoliposomes and subsequently incorporated into poly(lactic-co-glycolic acid)/polycaprolactone (PLGA/PCL) electrospun scaffolds to construct multifunctional bone regeneration platforms. The fabricated scaffolds exhibited uniform nanofibrous morphology, enhanced mechanical properties, and improved surface hydrophilicity, which collectively promoted cell adhesion and proliferation. Among the complexes, Cur-Cu demonstrated the highest antioxidant activity and superior osteogenic performance, significantly enhancing alkaline phosphatase (ALP) activity and upregulating integrin β1/α1 gene expression in MC3T3-E1 cells. In contrast, Cur-Zn exhibited broad-spectrum antibacterial efficacy against both Escherichia coli and Staphylococcus aureus , while Cur-Ca showed limited functional benefits due to rapid ion release and potential cellular interference. These findings highlight the tailored functionality of Cur-metal complexes. Cur-Cu scaffolds are promising for repairing bone defects under oxidative stress, whereas Cur-Zn scaffolds are effective in preventing implant-associated infections. This work provides a rational strategy for designing microenvironment-responsive biomaterials with integrated antioxidant, osteogenic, and antibacterial properties for advanced bone tissue engineering.
