Modified chitosan/oxidized carrageenan composite hydrogels incorporated with sea cucumber protein peptides as a potential novel biomedical material

Hongrui Dong,Jinhong Huang,Chengpeng Li,Mengyuan Pang,Zhang Hu

CS-TMG/O-Car/SPH hydrogel was constructed via dynamic imine bonds and physical interactions. • CS-TMG/O-Car/SPH exhibits excellent physical properties and strong antibacterial activity. • The hydrogel shows excellent biocompatibility and is a promising antibacterial biomaterial. Facing the challenges of bacterial infection and antibiotic resistance, the development of biomaterials with both excellent antibacterial properties and biocompatibility is of great significance. Using chitosan (CS) and κ-carrageenan (Car) as raw materials, tris(hydroxymethyl)methylglycine-modified chitosan (CS-TMG) and oxidized carrageenan (O-Car) were respectively synthesized. And then by incorporating sea cucumber protein peptides (SPH), a multi-network CS-TMG/O-Car/SPH hydrogel was successfully prepared. Characterization by UV, FTIR, 1 H NMR, XPS and XRD confirmed the successful grafting of TMG onto CS, the formation of aldehyde groups in Car, and the network structure stabilized by dynamic imine bonds and multiple physical interactions. CS-TMG/O-Car/SPH hydrogel exhibited a lamellar porous morphology and a high swelling ratio (2199%), coupled with good self-healing properties, injectability, and sustained-release characteristics (73.12% of SPH cumulative release within 72 h). Antibacterial tests revealed that CS-TMG/O-Car/SPH possessed significant antibacterial activities against both Staphylococcus aureus and Escherichia coli . Assessment of the biosafety revealed that CS-TMG/O-Car/SPH had a low hemolysis rate and negligible cytotoxicity, showing excellent biocompatibility. Consequently, CS-TMG/O-Car/SPH hydrogel is a promising candidate as an antibacterial biomaterial in the fields of wound healing and tissue engineering.

Sea cucumber-inspired dual-responsive black phosphorus/amorphous calcium-based bone-targeting nanoplatform for synergistic therapy of osteoporosis

Shen Xiang,Lan Zixun,Zhu Yaqiong,Zhong Tingting,Li Bozhang,Chen Jiaheng,Zhu Yanglong,Ye Jing,Wang Xiaolei

The core pathological mechanism of osteoporosis (OP) resides in the imbalance between osteoblast-mediated bone formation and osteoclast-mediated bone resorption. Furthermore, the accumulation of reactive oxygen species (ROS) and pro-inflammatory factors in bone tissue induces disorders in the immune microenvironment, which substantially elevates the risk of fracture. Inspired by the biological behaviors of sea cucumbers, we designed and constructed a bone-targeting platform (BA@PDA-PA) based on black phosphorus (BP) and amorphous calcium carbonate (ACC). This platform enables precise targeted therapy for diseased bone tissue while improving drug bioavailability. Upon reaching the target site, BA@PDA-PA can respond to the acidic microenvironment of OP lesions, and then release BP and calcium ions (Ca 2+ ) on demand. Specifically, BP efficiently scavenges local excessive ROS and improves the immune microenvironment. Meanwhile, the Ca 2+ released from ACC degradation, along with the low phosphorus oxides derived from BP breakdown, work together to promote osteoblast proliferation and differentiation while inhibiting osteoclast activity. This establishes a synergistic therapeutic mechanism involving “targeted delivery, microenvironment regulation, and bone metabolism remodeling.” In vivo experimental results demonstrate that this platform significantly improves the pathological bone microstructure of OP mice, thereby providing a promising therapeutic strategy for OP treatment. Household non-thermal processing for the dissolution of sea buckthorn flavonoids and the effects on its bioactive characteristics

Jie Sheng,Lanlan Yao,Qingying Dong,Bin Zhou,Bin Li,Guoqiang Zhang,Hongshan Liang

Sea buckthorn flavonoids exhibit high utilization value in the processing of functional foods and non-thermal processing (NTP) has emerged as a sustainable alternative for flavonoid extraction. Among total flavonoid content evaluation by juicing, shearing, ultrasound, colloid mill, and high-pressure homogenization, juicing demonstrated comparable efficiency. Systematic characterization of sea buckthorn flavonoids/flavonols dissolution (SBF) revealed that its color, turbidity, and particle size progressively changed during processing, flavonol releasement exhibited time-dependent patterns, with cumulative dissolution rates reaching 14.15 % (quercetin), 12.79 % (kaempferol), 6.75 % (isorhamnetin) within 180 min. Fitting with first-order kinetics and Weibull model suggested that the dissolution mechanisms involved both diffusion-controlled and erosion-mediated. Besides, sea buckthorn dissolution products (SBD) showed strong α-glucosidase inhibition (IC50: 5.18 mg/mL) and oxygen radical absorbance capacity (ORAC: 428.76 μmol TE/mL). This study elucidated the dissolution behavior of SBF under NTP conditions, with particular focus on the kinetics of flavonol release, providing guidance for future value-maximizing utilization of SBF.

Ultrasensitive and self-alarm pressure sensor based on laser-induced graphene and sea urchin-shaped Fe2O3 sandwiched structure

Xiu-man Wang,Yujun Chai,Congcong Zhu,Jiabing Yu,Xianping Chen

Flexible pressure sensors are widely used in wearable electronics. In recent years, it is still a challenge to prepare high-sensitivity and high-performance pressure sensors, though flexible pressure sensors have made significant progress. Therefore, a new sensor structure is designed in this study. This structure is composed of sea urchin-like Fe 2 O 3 as a spacer layer, which is sandwiched between two laser-induced graphene (LIG)/polyimide (PI) films. Additionally, a flexible multifunctional sensing device with both health monitoring and alarm functions is prepared based on the excellent mechanical properties and thermal conductivity of LIG. Multifunctional sensing devices have high sensitivity, integrated monitoring, and alarm functions. Specifically, the sensitivity of the pressure sensor is 603 kPa −1 for the pressure ranges of 0–10 kPa. The pressure sensor possesses a working range of up to 200 kPa and good cycle stability (for over 3000 loading–unloading cycles). Furthermore, LIG is employed as a heater to realize the alarm function. In this work, the combination of high performance and simple methods provides an effective method for the development of the integration of multifunctional pressure sensors for wearable electronic devices.

The Sustained-Release Oxygen Granule Preparation and Its Application in the Transportation of Live Sea Cucumber Apostichopus Japonicus

Yimeng Li,Jinghe Sun,Yan Zhou,Liming Sun,Jingfeng Yang

This study developed a sustained-release oxygen granule (SOG) to improve the transport of live sea cucumbers. During 8-day storage, SOG provided continuous oxygen while keeping seawater pH stable at 7.10 without affecting salinity. Compared to controls with added air, SOG significantly better preserved sea cucumber quality, delaying tissue autolysis and maintaining superior appearance and texture for up to 7 days. Furthermore, SOG prevented hypoxic stress, as evidenced by stable activities of key enzymes (AKP, ACP, SDH, LDH), supporting the organisms’ immune response throughout the storage period.

Aqueous Extract of Sea Squirt (Halocynthia roretzi) with Potent Activity against Human Cancer Cells Acts Synergistically with Doxorubicin

Yuting Zhu,Shanhao Han,Jianhui Li,Hongwei Gao,Bo Dong

Marine ascidian is becoming one of the main sources of an antitumor drug that has shown high bioactivity and extensive application in cancer treatment.Halocynthia roretzi, an edible marine sea squirt, has been demonstrated to have various kinds of biological activities, such as anti-diabetic, anti-hypertension, and enhancing immunity. In this study, we reported that aqueous extracts from the edible parts ofH. roretzipresented significantly inhibiting the efficiency on HepG-2 cell viability. The separate mixed compound exhibited strong effects of inhibitory proliferation and induced apoptosis via the generation of ROS along with the concurrent loss of mitochondrial membrane potential on tumor cells. Furthermore, we found that there existed a significantly synergistic effect of the ascidian-extracted compound mixture with the anti-cancer drug doxorubicin. In the presence of the extracts fromH. roretzi, the dose of doxorubicin at the cellular level could be reduced by a half dose. The extracts were further divided by semipreparative-HPLC and the active ingredients were identified as a mixture of fatty amide, which was composed of hexadecanamide, stearamide, and erucamide by UHPLC-MS/MS. Our results suggest that the potential toxicity of ascidianH. roretziin tumor cells, and the compounds extracted fromH. roretzicould be potentially utilized on functional nutraceuticals or as an adjunct in combination with chemotherapy. Keywords:ascidian;Halocynthia roretzi;anti-tumor;drug combination;doxorubicin

Sea Cucumber-Derived Peptide Attenuates Scopolamine-Induced Cognitive Impairment by Preventing Hippocampal Cholinergic Dysfunction and Neuronal Cell Death

Yue Zhao,Zhiqiang Lu,Xiaomeng Xu,Na Sun,Songyi Lin

The incidence of neurodegenerative diseases related to cognitive decline and memory loss is on the rise as the global elderly population increases. In this study, we evaluated the effect of the sea cucumber-derived peptide Phe–Tyr–Asp–Trp–Pro–Lys (FYDWPK) on scopolamine-induced neurotoxicity in an animal model. The Morris water maze, passive avoidance apparatus, and shuttle box test were used to assess learning and memory abilities. In behavioral tests, FYDWPK effectively alleviated learning and memory impairment. FYDWPK also alleviated cholinergic dysfunction in mice with dementia. Furthermore, FYDWPK significantly improved oxidative imbalance by increasing superoxide dismutase activity and decreasing malondialdehyde levels (P < 0.05). The pathological results showed that FYDWPK alleviated neuronal loss, blurred caryotheca, and pyknotic nuclei in the hippocampus, and a high dose of FYDWPK had the best effect. In conclusion, FYDWPK alleviated cognitive and memory impairments by regulating oxidative imbalance, reducing cholinergic dysfunction, and relieving pathological alterations.

In-situ construction of direct Z-scheme sea-urchin-like ZnS/SnO2 heterojunctions for boosted photocatalytic hydrogen production

Shaodong Sun,Dong Ren,Man Yang,Jie Cui,Qing Yang,Shuhua Liang

Constructing direct Z-scheme heterostructure is an effective way to promote the separation of photogenerated carriers and optimize the redox ability of the photocatalytic system. This work reports the in-situ synthesis of sea-urchin-like ZnS/SnO 2 Z-scheme heterojunctions via a one-step hydrothermal method . Both experimental results and density functional theory (DFT) calculations indicate that the tight interfaces derived from in-situ precursor dissociation can ensure a fast transfer for photogenerated carriers, meanwhile, the Z-scheme type of heterojunctions can increase the carrier separation efficiency and maintain the high reduction ability of photogenerated electrons. As expected, the photocatalytic hydrogen evolution rate of the as-optimized ZnS/SnO 2 sample can reach 2.17 mmol g −1  h −1 , which is 15.5 times higher than that of the commercial ZnS. This work can offer a novel strategy for designing Z-scheme heterojunction as well as controlling the contact interface for boosted photocatalytic activity .

Fabrication of sea urchin-like Au@SiO2 nanoparticles SERS substrate for the determination of malachite green in tilapia

Yuqian Liu,Lei Lei,Yeyu Wu,Yuyun Chen,Jun Yan,Weiwei Zhu,Xuecai Tan,Qi Wang

The quantitative determination of malachite green (MG) is of great importance in aquatic product safety. Although it has been testifying that sea urchin-like gold nanoparticles (SGNPs) have good enhancement effect, the practical application remains challenging for surface-enhanced Raman spectroscopy (SERS) due to poor stability. Here, sea urchin-like Au@SiO 2 nanoparticles (SG@SiO 2 NPs) is designed and synthesized through a controllable and surfactant-free hydrothermal method to improve its stability. The morphology of SG@SiO 2 NPs and the SiO 2 shell thickness were adjusted to obtain good SERS performance. The SERS signal intensity was linear for MG, crystal violet (CV) and methylene blue (MB) at 1619 cm −1 , 1623 cm −1 and 1627 cm −1 over the concentration range of 10 −5 -10 −9 M, 10 −5 -10 −9 M and 10 −5 -10 −8 M, respectively. The substrate was easily used for MG detection with limit of detection (LOD) as low as 1.5 × 10 −9 M. The recovery of MG added to several tilapia fillets samples at different concentrations ranged from 90 % to 110 %. Results indicated that SERS substrate can be used for the determination of trace amounts of MG in tilapia and demonstrated the potential of SG@SiO 2 NPs SERS substrate in food analysis fields.

Potential geographical distribution of harmful algal blooms caused by the toxic dinoflagellate Karenia mikimotoi in the China Sea

Changyou Wang,Yiwen Xu,Haifeng Gu,Zhaohe Luo,Zhuhua Luo,Rongguo Su

The fish-killing dinoflagellate Karenia mikimotoi frequently blooms in China and poses a threat to food safety and human health. To better understand harmful algal blooms (HABs) caused by K. mikimotoi and predict the risk of HABs under climate change, the combined effect of nitrate and norfloxacin (NOR) on the growth of K. mikimotoi was tested. A growth model was used to test the effects of nutrients and pollutants on the carrying capacity of the unicellular algae. The carrying capacity increased with increasing concentrations of nitrate and NOR, reaching a maximum at 62.2 μmol L −1 of nitrate and 9.03 mg L −1 of NOR. The calculated carrying capacity of K. mikimotoi in the China Sea showed a declining trend from nearshore to offshore, with a value >30 × 10 6 cells L −1 in the estuary of the Changjiang River and Hangzhou Bay. The HAB index proposed in this study as a measurement of HAB risk was constructed using the carrying capacity and relative abundance from the MaxEnt (maximum entropy) model. The index showed that HABs caused by K. mikimotoi consecutively occurred in Zhejiang and Fujian coastal waters and predicted that they will continue until 2100, regardless of the greenhouse gas emission scenario. The center of the integrated area moved northward, with a range of 120–900 km. The HAB index integrates the characteristics of the carrying capacity and suitability of habitats, and expresses the information contained in the intensive and extensive variables that affect HAB occurrence. This index is a promising predictor of HAB risk in coastal waters.

Non-targeted Profiling of Sea Buckthorn Fruit Oil Fingerprints from 3 Regions and Study on Its Antioxidant Activity

Sun Ziyi,Zhang Gaoning,Li Xiaoru,Cui Qingqi,Gong Lili,Tian Zhenhua

This study investigated the chemical and volatile characteristics of sea buckthorn fruits from three different regions in China. The chemical composition of the volatile oil was determined by using a non-targeted gas chromatography and mass spectrometry (GC/MS) method and the differences in chemical composition among the three producing areas were compared by heatmap providing a visual basis for researchers. A total of 93 compounds were identified, including 52 compounds from the Northeast China, 51 from the Xinjiang region, and 37 from Inner Mongolia region. Then, the in vitro antioxidant activity of sea buckthorn fruit oil was measured using DPPH, ABTS, and SOD inhibition tests, and the results showed that sea buckthorn fruit oil in northeast China was the strongest antioxidant, followed by Inner Mongolia and Xinjiang. The results of the CCK-8 experiment indicated that within the tested concentration, there is no cell cytotoxicity of the essential oil in human umbilical vein endothelial cells (HUVECs) cells. The results could supply reference to distinguish sea buckthorn fruit from different production areas and, meanwhile, clarify the activity and safety of sea buckthorn oil.

Multiple light scattering and nanotip effect of hierarchical sea urchin-like W18O49 boosting photocatalytic hydrolysis of ammonia borane

Mei Li,Shengbo Zhang,Mohammed Ahmed Abdulraheem Alwafi,Jinyu Han,Xinli Zhu,Qingfeng Ge,Hua Wang

The morphology of catalysts is a key factor influencing their photocatalytic performance. Herein, we prepared three W 18 O 49 photocatalysts with different morphologies of sea urchin (W 18 O 49 SU), nanorod (W 18 O 49 NR) and hollow sphere (W 18 O 49 HS), and evaluate their performances in photocatalytic hydrolysis of ammonia borane (AB). Remarkably, W 18 O 49 SU exhibited an impressive photocatalytic H 2 production rate with the highest TOF of 53.1 min −1 under visible light, which is 10.4 and 7.5 times higher than those of W 18 O 49 HS and W 18 O 49 NR, respectively. The comprehensive characterizations and control experiments revealed that the unique sea urchin-like morphology enabled the strong light harvesting by multiple light scattering, fast separation of photogenerated charges and directional electron enrichment by nanotip effect, which collaboratively boost the photocatalytic activity . Moreover, the enhancement effect of sea urchin-like morphology is again approved well on Nb 2 O 5 photocatalysts. This work opens a paradigm to synergistically improve light harvesting and charge separation efficiency through rational design of photocatalyst nanostructure .
