Lecithins-Zein nanoparticles for antifungal treatment: Enhancement and prolongation of drug retention in skin with reduced toxicity

Shuang Zhang,Wenting Song,Hangyi Wu,Jiao Wang,Yuling Wang,Zhenhai Zhang,Huixia Lv

Fungal infections are one of the major skin healthcare issues and cause significant morbidity. Ketoconazole (KC) as a broad-spectrum antifungal drug is widely used to treat skin fungal diseases. However, its therapeutic effects are limited by low concentration, short duration of drug efficacy in the skin and severe systemic toxicity. Here, the ketoconazole loaded Lecithins-Zein nanoparticles (KLZ-NPs) with core-shell structure were designed to resolve above problems. In vitro penetration test confirmed that the ketoconazole concentration of the KLZ-NPs group in the stratum corneum and deeper layers increased significantly (2.98-fold, 1.51-fold higher to free ketoconazole, respectively). Meanwhile, follicular closing technique showed the formed nanoparticles via follicle pathway into the skin had been significantly enhanced, and the results of the visual fluorescent images also confirmed it. Additionally, in the in vivo imaging experiment, the fluorescence intensity of the single applying of the DiR-LZ-NPs was higher than that of the thrice usage of the free DiR. More importantly, the results also indicated that the accumulation of nanoparticles in the liver and spleen was significantly reduced. Hence, Lecithins-Zein nanoparticles are a promising strategy to enhance the drug concentration, prolong efficacy and reduce systemic toxicity in the topical administration for antifungal treatment.

Modulation of interfacial phenolic antioxidant distribution in Pickering emulsions via interactions between zein nanoparticles and gallic acid

Zijun Zhao,Muwen Lu,Zhu Mao,Jie Xiao,Qingrong Huang,Xuechun Lin,Yong Cao

The impacts of protein nanoparticles on the interfacial distribution of antioxidants and the oxidative stability in Pickering emulsions are attracting increasing research interests. In the present work, the distribution of gallic acid (GA) in zein nanoparticles-stabilized Pickering emulsions (ZPE) was determined by employing a pseudophase kinetic model. The interfacial distribution of GA was found to be favored in ZPEs with higher zein nanoparticle concentration (C zein ). Upon increasing C zein , the interfacial loading of nanoparticles (Γ) dominated the modulation of %GA I via hydrogen bonding between zein nanoparticles and GA. The interfacial percentage of GA (%GA I ) increased from 28% to 39% as Γ increased from 0.48 to 1.12 mg/m 2 . In the presence of GA, a direct correlation between C zein or Γ and oxidation stability was recognized, whereas the oxidative stability showed a non-linear dependence on either C zein or Γ in the absence of GA. By excluding antioxidant effects of zein nanoparticles, we found that the %GA I , which was regulated by Γ, took the leading role over the physical barrier effect on the oxidative stability of emulsions. The present work extends our current knowledge on how protein based nanoparticles manipulate the interfacial distribution of antioxidant and then affect the oxidative stability of emulsions.

Development of Antibacterial and Hemostatic PCL/Zein/ZnO-Quaternary Ammonium Salts NPs Composite Mats as Wound Dressings

Yang Wang,Maoli Ying,Meng Zhang,Xuehong Ren,Ick Soo Kim

Wound dressings are usually employed to maintain suitable conditions around the injured skin to accelerate wound healing. This paper aims to report functional composite dressings combined with polycaprolactone (PCL) nanofibers, self-assembly zein coating and modified ZnO nanoparticles (ZnO NPs) for wound management. The synthetic compounds and prepared composite dressings are characterized by Fourier infrared spectroscopy (FTIR), scanning electron microscopy (SEM) and energy-dispersive X-ray spectroscopy. Meanwhile, the water vapor transmission rate, hemostatic performance, antibacterial activity, and cytocompatibility of the composite dressings are systematically tested to evaluate their applicability in wound treatment. The results show that the multilayer dressings can retain moisture and prevent excessive dehydration of wound. In vitro hemostatic test is conducted, and the enhanced blood clotting capacity and the activation of platelets indicate that the desired dressings are able to control hemorrhaging from wound. Meanwhile, the composite dressings with excellent biocidal efficacy against Gram-positive Staphylococcus aureus (6.01 Log within 30 min) and Gram-negative Escherichia coli O157:H7 (6.04 Log within 30 min) can effectively prevent wound infection. Furthermore, the dressings show no toxicity which are evidenced in hemolysis and cytocompatibility evaluation, and have potential application for wound healing.

Zein/gelatin/nanohydroxyapatite nanofibrous scaffolds are biocompatible and promote osteogenic differentiation of human periodontal ligament stem cells

Qianmin Ou,Yingling Miao,Fanqiao Yang,Xuefeng Lin,Li-Ming Zhang,Yan Wang

In bone tissue engineering, it is important for biomaterials to promote the osteogenic differentiation of stem cells to achieve tissue regeneration. Therefore, it is critical to develop biomaterials with excellent cytocompatibility and osteoinductive ability. In our previous study, we found a zein/gelatin electrospinning scaffold with good biocompatibility, but low osteoinductive ability for human periodontal ligament stem cells (hPDLSCs). Therefore, herein, we fabricated novel zein/gelatin/nanohydroxyapatite (zein/gelatin/nHAp) nanofibrous membranes to overcome the drawbacks of the zein/gelatin scaffold. The results showed that the surface wettability of the zein/gelatin/nHAp nanofiber membranes was increased. Moreover, the inclusion of nHAp facilitated the attachment, proliferation, and osteogenic differentiation of hPDLSCs. Overall, the zein/gelatin/nHAp nanofiber membranes showed good biocompatibility and osteoinductive activity for hPDLSCs in vitro and in vivo; this suggested potential applications of these membranes in bone tissue engineering.

Preparation of zein nanofibers with cinnamaldehyde encapsulated in surfactants at critical micelle concentration for active food packaging

Ping Shao,Yong Liu,Christos Ritzoulis,Ben Niu

This work studied the effect of nonionic surfactant (Tween 80), anionic surfactants sodium dodecyl sulfate (SDS), cationic surfactants sodium cetyltrimethyl ammonium bromide (CTAB), amphoteric surfactant (Lecithin) on the characteristics of electrospun zein nanofibers and on the release of cinnamaldehyde essential oil (CEO) in zein nanofibers. The effect of applying nanofibers as an antibacterial package was also investigated. Scanning electron micrographs demonstrated that the flat shape of zein nanofibers could shift to round when surfactants were added. The interaction between zein and lecithin caused many granules while the addition of other surfactants produced a smooth surface. Fourier transform infrared indicated that the addition of surfactants could change the secondary structure of zein. The addition of CTAB to zein nanofibers decreased the release of CEO while addition of Tween80, lecithin and SDS accelerated release in ethanol aqueous solution. The nanofibers inhibited the growth of microorganisms and prolonged the shelf life of mushrooms, which confirmed that it can be applied in active antibacterial package.

Fabrication of zein-coated brush-like silica nanocarriers for high foliage deposition and responsive release of pesticide

Yuting Zhang,Tao Hong,Menghui Wan,Yanbao Zhao,Lichao Peng

Responsive release systems have received extensive attention to enhance pesticide utilization efficiency and reduce environmental pollution. In this study, pH/GSH dual responsive release system based on brush-like silica (bSiO 2 ) carriers was constructed to enhance the utilization of pesticides. The bSiO 2 carriers present core-shell structure, length of 550 nm, diameter of 350 nm and shell thickness of 100 nm. The carrier had a high pesticide loading (20.0 %, w/w) for dinotefuran (Din). After loading Din, zein was covalently linked with cysteine-bridge to seal the loaded pesticides (namely Din@bSiO 2 @Zein). The Din@bSiO 2 @Zein exhibited superior foliar affinity, retention and photostability, and retention rate still remain above 95 % with 220 min UV irradiation. Din@bSiO 2 @Zein displayed pH/GSH responsive release and the cumulative release within 92 h was up to 81 % under pH=9/C GSH =6 mM, mimicking the microenvironment of lepidopteran. The Din@bSiO 2 @Zein possessed good control efficacy against Plutella xylostella . Appreciably, Din@bSiO 2 @Zein could be transported bi-directionally to various regions of tobacco plants within 24 h, which had potential to promote pesticide efficacy. This work offers a strategy to minimize the pesticide dosage and encourage sustainable agricultural development.

High internal phase Pickering emulsions co-stabilized by zein nanoparticles and cellulose nanocrystals: Fabrication, characterization, and application

Song Cui,Yue Yu,David Julian McClements,Chaoran Liu,Xingfeng Xu,Qingjie Sun,Pengjie Wang,Lei Dai

There has been great interest in the development of edible high internal phase Pickering emulsions (HIPPEs) for a variety of applications in the food industry, including enhancing shelf life and creating novel textures. However, it is often challenging to create HIPPEs with the required stability, texture, and processability. Herein, we prepared HIPPEs using Pickering emulsifiers (2%) consisting of different mass ratios of zein nanoparticles and cellulose nanocrystals (CNC). The hydrophobic interaction and hydrogen bond between zein nanoparticles and CNC were the main ways to stabilize HIPPEs. Meanwhile, the compact three-dimensional network structure formed by these nanoparticles and nanocrystals increased the stability and rheological properties of the HIPPEs. As the mass ratio of zein nanoparticles to CNC increases from 1:2 to 2:1, the storage modulus increases from about 545 Pa to 2790 Pa, which indicated that the texture of HIPPEs can be modulated by adjusting the mass ratio of zein nanoparticles to CNC. Moreover, rheological properties analysis displayed that the HIPPEs were viscoelastic materials with predominantly solid-like characteristics and good texture recoverability after shearing. Cryogenic scanning electron microscopy results observed that zein nanoparticles and CNC formed a coating around the oil droplets, and they formed filamentous network connections between different oil droplets. Finally, the incorporation of HIPPEs into cookies proved to be an effective fat replacer, and the HIPPEs replaced butter could improve their overall appearance and color. HIPPEs may therefore be used as butter substitutes in some food products, but further research is required to optimize their application.

Effect of Cu2+ on the binding of catechins to zein through multi-spectral and in silico analyses

Huihui Li,Zhuangwei Zhang,Jin Chen,Mengchu Sun,Hongjin Tang

This study aimed to elucidate how Cu 2+ influences the interaction between catechins and zein using multi-spectral and molecular modeling techniques. As a result, UV–vis spectra revealed characteristic changes, indicating chelation between catechins and Cu 2+ at a 1:1 M ratio. Fluorescence spectra further confirmed interaction through a static quenching mechanism between catechins/catechin-Cu 2+ complexes and zein. Catechins induced changes in the microenvironment and hydrophobicity surrounding the binding site of zein, whereas Cu 2+ had minimal impact on these aspects. CD spectra underscored catechins' role in altering zein's secondary structure conformation, alongside Cu 2+ . Various types of interactions (hydrophobic, hydrogen bonding, electrostatic, and van der Waals) contributed to the binding of catechins/catechin-Cu 2+ complexes with zein. Molecular modeling elucidated key residues and binding conformations, highlighting the significance of hydrophobic interactions and hydrogen bonding in their association. These findings not only deepen our understanding of catechin-Cu 2+ -zein interactions but also underscore their potential applications in the food industry.

Characterization of zein / hyaluronic acid complex colloidal nanoparticles stabilized Pickering emulsions as a delivery vehicle of curcumin

Di Wu,Yang Lu

In this work complex nanoparticles composed of zein and hyaluronic acid (HA), referred to as ZHPs, were fabricated through the combined effects of hydrogen bonding and electrostatic attractions. Compared to zein particles, ZHPs exhibited a larger mean particle size and a reversal in zeta potential from positive to negative. The three-phase contact angle ( θ o/w ) of ZHPs with a 10:3 ratio was measured at 88.9°, indicating their suitability for stabilizing Pickering emulsions. Both droplet diameter and the emulsified phase volume fraction of the emulsions were significantly affected by the concentration of ZHPs and the oil phase ratio. Confocal laser scanning microscopy revealed that ZHPs adsorbed on the oil-water interface formed a compact interfacial layer. Rheological analysis showed that emulsions displayed shear-thinning behavior and exhibited characteristics of an elastic gel network. Moreover, the Pickering emulsions stabilized by ZHPs markedly suppressed curcumin degradation under UV exposure compared to both bulk oil and conventional emulsions. Results from vitro digestion indicated that the release of free fatty acids (FFAs) and the bioaccessibility of curcumin were enhanced in ZHP-stabilized emulsions relative to bulk oil. Overall, the findings support the potential of ZHP-stabilized Pickering emulsions as efficient delivery vehicles for food and pharmaceutical applications.

Covalent grafting of corn stover cellulose nanofibers with zein for pickering emulsion stabilization and controlled quercetin delivery

Xinling Wu,Qiang Zhang,Deyi Zhou,Jianpeng Dou,Qing Liang,Li Zhang,Haigen Zhou,Xiaodi Gao,Xuelin Zhao,Jinsong Zhang

Enhancing hydrophobic bioactives' bioaccessibility remains challenging in functional foods due to instability and insufficient controlled-release ability in conventional protein-polysaccharide carriers. We pioneer a new interaction model by covalently grafting corn stover cellulose nanofibers (CNF) with Zein using N-(3-dimethylaminopropyl)-N′-ethyl carbodiimide hydrochloride (EDC) and N-hydroxysuccinimide (NHS), creating conjugates with gradient grafting degrees (CNF/Zein 0.5, CNF/Zein 1, and CNF/Zein 2). Excitingly, compared to the non-covalent complex (CNF-Zein), conjugates exhibited a stable network-like microstructure, lower crystallinity, and enhanced thermal stability. Furthermore, the conjugates exhibited excellent amphiphilicity, forming denser and more stable emulsions. Confocal laser scanning microscope (CLSM) revealed a dual stabilization mechanism of “interface adsorption-network encapsulation”. Additionally, CNF/Zein-stabilized emulsions demonstrated controlled quercetin release and improved bioaccessibility during in vitro digestion tests, with the highest grafted CNF/Zein 2 showing 43.65 % bioaccessibility. This study first demonstrates the effectiveness of the EDC/NHS technique in alcohol-solution systems, providing novel strategies for conjugate design and lipophilic nutraceutical delivery.

Preparation of gastrointestinal pH-responsive zein coated tea polyphenol-heparin hydrocolloids using for inflammatory bowel disease therapy

Hui Li,Shuangshuang Chen,Yumei Li,Zelin Sang,Zhenhua Chen,Xifan Mei,Xiuli Ren

In the pathogenesis of inflammatory bowel disease (IBD), overstimulation of inflammatory factors can trigger a coagulation cascade, increase the risk of intestinal micro-thrombosis and lead to microcirculation disorders. However, prevention of microcirculation pathways has not received enough attention. Heparin is commonly used in anticoagulant therapy, but oral delivery does not have an excellent anticoagulant effect. To improve the stability of heparin (HEP) in the gastrointestinal tract, zein/tea polyphenol nanospheres with a core-shell structure (EGNs@Z) were developed for oral administration of heparin (HEGNs@Z). The Zein shell has pH-responsive properties and is effective in preventing premature dissolution of heparin. At the same time, EGCG nanospheres (EGNs) play an anti-inflammatory role, jointly improve the vicious cycle between inflammation and microthrombosis. The results of SEM, TEM and FTIR showed that EGNs successfully encapsulated heparin and formed zein shells on the surface of microspheres with a thickness of 50-100 nm. In vitro simulated digestion experiments showed that zein shells prevented the breakdown of microspheres and heparin in a simulated gastric environment, whereas EGNs and HEP were slowly degraded and released in a simulated intestinal environment. Coagulation analysis showed that HEGNs@Z was effective in delaying clotting time. A mouse model of acute colitis has also shown that HEGNs@Z robust promotes colonic epithelial regeneration, inhibits malignant microcirculation, and reduces bleeding risk. These findings reveal that this orally bioavailable multifunctional material may provide a novel, effective and convenient treatment for inflammatory bowel disease.
