Luteolin inhibits cell proliferation and induces cell apoptosis via down-regulation of mitochondrial membrane potential in esophageal carcinoma cells EC1 and KYSE450

Ping Chen,Jing-Yang Zhang,Bei-Bei Sha,Yan-Er Ma,Tao Hu,Yang-Cheng Ma,Hao Sun,Jian-Xiang Shi,Zi-Ming Dong,Pei Li

In current study, we investigated the anti-tumor effect of luteolin in human ESCC cell lines in vitro and in vivo and tried to explore the potential mechanisms. Results from flow cytometry showed that luteolin could induce apoptosis and caspase-3 activation and induce cell cycle arrest at G2/M phase in a dose- and time-dependent manner in EC1 and KYSE450 cells. JC-1 test results showed that membrane potential of mitochondria after luteolin treatment was down-regulated and this was an indicator for intrinsic apoptosis. Western Blot results showed the expression of cell cycle regulatory protein p21 and p53 increased and three apoptosis related proteins that participate in mitochondrial apoptotic pathway, namely, Bim, CYT-c and cPARP, also increased in luteolin treated cells compared with control groups. We further confirmed that luteolin could significantly inhibit the growth of ESCC tumors in xenograft mouse models and no evidence of systemic toxicity was observed. Our results suggest that luteolin can induce cell apoptosis and cell cycle arrest in G2/M phase through mitochondrial pathway in EC1 and KYSE450 cell lines and proper utilization of luteolin might be a practical approach in ESCC chemotherapy.

Luteolin ameliorates chronic stress-induced depressive-like behaviors in mice by promoting the Arginase-1+ microglial phenotype via a PPARγ-dependent mechanism

Yuan Nai-jun,Zhu Wen-jun,Ma Qing-yu,Huang Min-yi,Huo Rou-rou,She Kai-jie,Pan Jun-ping,Wang Ji-gang,Chen Jia-xu

Accumulating evidence shows that neuroinflammation substantially contributes to the pathology of depression, a severe psychiatric disease with an increasing prevalence worldwide. Although modulating microglial phenotypes is recognized as a promising therapeutic strategy, effective treatments are still lacking. Previous studies have shown that luteolin (LUT) has anti-inflammatory effects and confers benefits on chronic stress-induced depression. In this study, we investigated the molecular mechanisms by which LUT regulates the functional phenotypes of microglia in mice with depressive-like behaviors. Mice were exposed to chronic restraint stress (CRS) for 7 weeks, and were administered LUT (10, 30, 40 mg· kg −1 ·day −1 , i.g.) in the last 4 weeks. We showed that LUT administration significantly ameliorated depressive-like behaviors and decreased hippocampal inflammation. LUT administration induced pro-inflammatory microglia to undergo anti-inflammatory arginase (Arg)-1 + phenotypic polarization, which was associated with its antidepressant effects. Furthermore, we showed that LUT concentration-dependently increased the expression of PPARγ in LPS + ATP-treated microglia and the hippocampus of CRS-exposed mice, promoting the subsequent inhibition of the NLRP3 inflammasome. Molecular dynamics (MD) simulation and microscale thermophoresis (MST) analysis confirmed a direct interaction between LUT and peroxisome proliferator-activated receptor gamma (PPARγ). By using the PPARγ antagonist GW9662, we demonstrated that LUT-driven protection, both in vivo and in vitro, resulted from targeting PPARγ. First, LUT-induced Arg-1 + microglia were no longer detected when PPARγ was blocked. Next, LUT-mediated inhibition of the NLRP3 inflammasome and downregulation of pro-inflammatory cytokine production were reversed by the inhibition of PPARγ. Finally, the protective effects of LUT, which attenuated the microglial engulfment of synapses and prevented apparent synapse loss in the hippocampus of CRS-exposed mice, were eliminated by blocking PPARγ. In conclusion, this study showed that LUT ameliorates CRS-induced depressive-like behaviors by promoting the Arg-1 + microglial phenotype through a PPARγ-dependent mechanism, thereby alleviating microglial pro-inflammatory responses and reversing microglial phagocytosis-mediated synapse loss.

Luteolin-bisphenol A epoxy double-walled self-healing microcapsules: Preparation and performance for deep formation wellbore stabilization in water-based drilling fluids

Han Yan,Jin-Sheng Sun,Kai-He Lv,Jing-Ping Liu,Ying Yang,Yuan-Wei Sun,Tai-Feng Zhang,Yu-Fan Zheng,Lu Zhang

Deep hydrocarbon resources are recognized as a critical replacement domain for global oil and gas reserves. However, wellbore instability during the application of water-based drilling fluids represents a critical risk for cores, which severely constrains drilling efficiency and operational safety. Herein, a double-walled self-healing microparticles (MPSD) encapsulating epoxy monomer of bisphenol A and luteolin diglycidyl ether composite (BL-DGE) for wellbore stabilization was innovatively proposed and developed. MPSD exhibited a regular spherical morphology with a compact, dense surface, coupled with excellent thermal resistance and mechanical performance. After being immersed in an aqueous environment at 160–200 °C for 16 h, the shale slices treated with MPSD exhibit a shear strength of 634–707 kPa. Furthermore, under high-temperature immersion conditions, the shale cuttings are bonded into an integral structure by MPSD. The permeability of the artificial core treated with MPSD was reduced from 0.95 to 0.162 mD, with a plugging rate of 82.95%. The shell of the microparticles fracture under high-temperature conditions, releasing BL-DGE. By plugging the pores of the rock debris, the loose and fragmented rock is bonded into a dense and stable monolithic structure, significantly improving its mechanical properties and stability. This work provides an important theoretical foundation and technical solution for wellbore stabilization using water-based drilling fluids in deep fractured formations.

Amelioration of type 2 diabetes by the novel 6, 8-guanidyl luteolin quinone-chromium coordination via biochemical mechanisms and gut microbiota interaction

Xiaodong Ge,Xiaoyu He,Junwei Liu,Feng Zeng,Ligen Chen,Wei Xu,Rong Shao,Ying Huang,Mohamed A. Farag,Esra Capanoglu,Hesham R. El-Seedi,Chao Zhao,Bin Liu

Introduction Luteolin is a plant-derived flavonoid that exhibits a broad range of pharmacological activities. Studies on luteolin have mainly focused on its use for hyperlipidaemia prevention, whereas the capacity of the flavonoid to hinder hyperglycaemia development remains underexplored. Objectives To probe the anti-hyperglycemic mechanism of 6,8-guanidyl luteolin quinone-chromium coordination (GLQ.Cr), and to assess its regulatory effect on intestinal microbiota in type 2 diabetes mellitus (T2DM) mice. Methods High-sucrose/high-fat diet-induced and intraperitoneal injection of streptozotocin was used to develop a T2DM model. Glycometabolism related indicators, histopathology, and gut microbiota composition in caecum samples were evaluated, and RNA sequencing (RNA-seq) of liver samples was conducted. Faecal microbiota transplantation (FMT) was further used to verify the anti-hyperglycemic activity of intestinal microbiota. Results The administration of GLQ.Cr alleviated hyperglycaemia symptoms by improving liver and pancreatic functions and modulating gut microbe communities ( Lactobacillus, Alistipes , Parabacteroides , Lachnoclostridium , and Desulfovibrio ). RNA-seq analysis showed that GLQ.Cr mainly affected the peroxisome proliferative activated receptor (PPAR) signalling pathway in order to regulate abnormal glucose metabolism. FMT significantly modulated the abundance of Lactobacillus , Alloprevotella , Alistipes , Bacteroides, Ruminiclostridium, Brevundimonas and Pseudomonas in the caecum to balance blood glucose levels and counteract T2DM mice inflammation. Conclusion GLQ.Cr improved the abnormal glucose metabolism in T2DM mice by regulating the PPAR signalling pathway and modulating intestinal microbial composition. FMT can improve the intestinal microecology of the recipient and in turn ameliorate the symptoms of T2DM-induced hyperglycaemia.

Exploring the molecular mechanisms of the inhibition of acrolein-induced BEAS-2B cytotoxicity by luteolin using network pharmacology and cell biology technology

Dan Liu,Ye Cheng,Junliang Chen,Xueying Mei,Zhipeng Tang,Xiangyu Cao,Jianli Liu

Acrolein is a highly reactive unsaturated hazardous air pollutant , which is extremely irritating to the respiratory tract. Luteolin , an active flavonoid compound, possesses multiple biological activities . The purpose of this study was to evaluate the mechanism of the inhibition of acrolein-induced human bronchial epithelial (BEAS-2B) cells cytotoxicity by luteolin using network pharmacology and cell biology technology . Firstly, network pharmacology results indicated that oxidative stress processes might play an important role in luteolin inhibiting lung injury. Next, it was verified at the cellular level. Reactive oxygen species (ROS) generation increased, glutathione (GSH) level decreased after exposure to acrolein. MAPK signaling pathways were activated, which activated downstream IκBα/NF-κB signaling pathways. Meanwhile, acrolein caused oxidative DNA damage and double-strand breaks, induced DNA damage response (DDR) and apoptosis . These adverse effects were significantly reversed by luteolin, which inhibited the activation of MAPK/IκBα/NF-κB and DDR pathways, and reduced the ratio of Bax/Bcl-2. Moreover, luteolin also had a similar effect to antioxidant N-acetyl cysteine (NAC) in the regulation of signaling transduction mechanisms, which indicated that the regulation of oxidative stress played an important role in the process. These results provide an experimental basis for elucidating the molecular mechanisms of the inhibition of acrolein-induced BEAS-2B cytotoxicity with luteolin.

Detection of luteolin in food using a novel electrochemical sensor based on cobalt-doped microporous/mesoporous carbon encapsulated peanut-like FeOx composite

Jiejun Li,Yuhong Li,Yaqi Yang,Pengcheng Zhao,Junjie Fei,Yixi Xie

Luteolin (Lu) is a dietary flavonoid that has attracted much attention due to its multiple health benefis effects. Herein, an ultrasensitive electrochemical sensor for Lu was constracted based on cobalt-doped microporous/mesoporous carbon (MMC) encapsulated peanut-like Fe 2 O 3 composite. The FeOx-Co-MMC composite was obtained by pyrolyzing a precursor named Fe 2 O 3 -Co-microporous/mesoporous dopamine (Fe 2 O 3 -Co-MMPDA) which was synthesized by a soft template method. Under optimized conditions, the sensor exhibited good detection of Lu with a low limit of detection (LOD) of 0.031 nM and a wide linear range from 0.05 to 1000 nM in detecting Lu. It also demonstrated good reproducibility (RSD = 3.16%), stability (RSD = 2.34%), and anti-interference properties. The sensor successfully detected Lu in real food samples such as honeysuckle with recoveries ranging from 96.1% to 104.8% (RSD <3%, n = 3). The present study provides an alternative method for Lu detection in food.

Gallic acid-affinity molecularly imprinted polymer adsorbent for capture of cis-diol containing Luteolin prior to determination by high performance liquid chromatography

Yukui Tong,Baoyue Zhang,Bailin Guo,Wenjie Wu,Yingxue Jin,Fang Geng,Miaomiao Tian

A gallic acid-affinity molecularly imprinted polymer (G-MIP) was first used as an adsorbent for selective identification and capture of luteolin (LTL) in herbal medicine samples. The G-MIP was prepared by using LTL as the template, gallic acid (GA) as the functional monomer, ethylene glycol dimethacrylate (EGDMA) as the crosslinking agent, and 2,2′-azobis(2-methylpropionitrie) (AIBN) as the initiator. The properties of G-MIP were characterized by FT-IR, transmission electron microscope, scanning electron microscope, dynamic light scattering, specific surface area, and X-ray photoelectron spectrum. The adsorption conditions were optimized, and the adsorption equilibrium model and adsorption kinetics model of the adsorbent were investigated under the best experimental conditions. The saturated adsorption capacity is 1.24 mg g −1 , which is not only higher than the adsorption capacity of 4-carboxyphenylboronic acid-affinity MIP adsorbent but also superior to those of many reported adsorbents for enriching of LTL. The LTL was quantified by HPLC. The linear range is 0.05–100 mg L −1 , the detection limit is 0.020 mg L −1 . This method was successfully applied in the selective recognition of LTL in herbal medicines with recoveries of 93.9–114.2%, and the relative standards deviations (RSDs) are 0.4–5.6%. Thus, this work provides a potential possibility and practical platform for the determination of LTL in complex matrices.

Role of luteolin extracted from Clerodendrum cyrtophyllum Turcz leaves in protecting HepG2 cells from TBHP-induced oxidative stress and its cytotoxicity, genotoxicity

Gang Li,Jing Zhou,Mengyu Sun,Juren Cen,Jing Xu

Luteolin derived from Clerodendrum cyrtophyllum Turcz leaves exhibited antioxidant activity. The IC 50 values of luteolin for ABTS· + and DPPH· free radical scavenging was 17.3 ± 0.82 μM and 13.2 ± 0.18 μM, respectively, and the compound showed better antioxidant activities than BHT (Butylated hydroxytoluene) and VC (Vitamin C). Luteolin exhibited neither cytotoxic nor promotional activities and no genotoxic effect toward HepG2 cells. The cell viability was significantly increased in a dose-dependent manner after preincubation of the cells with 1–40 μM luteolin followed by direct exposure to TBHP ( tert -Butyl hydroperoxide). At the concentration of 30 μM, luteolin can reduce intercellular reactive oxygen species accumulation by 43.7 ± 3.98%, lactate dehydrogenase release by 38.7 ± 3.07% malondialdehyde levels by 24.5 ± 1.14% and increased 73.7 ± 6.04% superoxide dismutase activity and 72.3 ± 1.75% glutathione activity. A western blot assay showed that luteolin upregulated procaspase-3 and downregulated cleaved caspase-3 to counteract TBHP-triggered oxidative stress. Our study lays a theoretical foundation for the utilization and further development of luteolin as a promising natural antioxidant.

Luteolin combined with low-dose paclitaxel synergistically inhibits epithelial–mesenchymal transition and induces cell apoptosis on esophageal carcinoma in vitro and in vivo

Tiantian Qin,Jinzhu Zhao,Xiaojie Liu,Leilei Li,Xueyan Zhang,Xiaoli Shi,Yu Ke,Weihua Liu,Junfeng Huo,Yalong Dong,Yiwei Shen,Yanyu Li,Mingjing He,Shuhua Han,Linlin Li,Chengxue Pan,Cong Wang

Although paclitaxel is a promising frontline chemotherapy agent for various malignancies, the clinical applications have been restricted by side effects, drug resistance, and cancer metastasis. The combination of paclitaxel and other agents could be the promising strategies against malignant tumor, which enhances the antitumor effect through synergistic effects, reduces required drug concentrations, and also suppresses tumorigenesis in multiple ways. In this study, we found that luteolin, a natural flavonoid compound, combined with low-dose paclitaxel synergistically regulated the proliferation, migration, epithelial–mesenchymal transition (EMT), and apoptosis of esophageal cancer cells in vitro, as well as synergistically inhibited tumor growth without obvious toxicity in vivo. The molecular mechanism of inhibiting cell migration and EMT processes may be related to the inhibition of SIRT1, and the mechanism of apoptosis induction is associated with the reactive oxygen species (ROS)/c-Jun N-terminal kinase (JNK) pathway–mediated activation of mitochondrial apoptotic pathway.

Application of bifunctional monomer surface MIP with MOFs nanocomposite for efficient trapping and analysis of luteolin in compound Anoectochilus roxburghii (Wall.) Lindl. oral liquid

Qiuhua Zhang,Youjia Wu,Pingping Wu,Liying Huang,Lingyi Huang

Luteolin is one of the bioactive components from the compound Anoectochilus roxburghii (Wall.) Lindl. oral liquid (CAROL), which was reported to have excellent hepatoprotective and anti-inflammatory activities. However, the enrichment and quantitation of luteolin from CAROL is challenging due to the low content and complex aqueous matrix. In this study, a bifunctional monomer surface molecularly imprinted polymer (MIP) with metal–organic frameworks (MOFs) as cores was prepared for the selective adsorption of luteolin from the aqueous system CAROL. Compared with conventional MIPs, this unique nanocomposite adsorbent (MOF@MIPs) has the advantages of short kinetic equilibrium time, good selectivity, and high adsorption capacity in aqueous solution. The theoretical maximum adsorption capacity of MOF@MIPs for luteolin was 36.99   mg/g. After adsorption enrichment of luteolin from CAROL using MOF@MIPs, liquid chromatography–tandem mass spectrometry was applied to analyze the target. The corresponding linearity range for analyte was 10–6000   ng/mL with good linearity (R 2 =0.9992), and the added recoveries varied from 85.70% to 99.25%. The present method has been successfully employed for the analysis of luteolin in five different batches of CAROL. Notably, we found no significant difference in the content of luteolin between these batches, which proved that the composition was stable between batches. The novel structure MIPs are suitable for the specific recognition of template molecules in aqueous solution. Therefore, this study provides a technical reference for the special identification and determination of trace components in complex samples, while the novel MOF@MIP nanocomposite can also provide valuable references for the extraction and purification methods of specific substances in traditional Chinese medicine and expand the application environment of MIPs material.

Luteolin alleviates non-alcoholic fatty liver disease in rats via restoration of intestinal mucosal barrier damage and microbiota imbalance involving in gut-liver axis

Xia Liu,Runzhou Sun,Zhaozhen Li,Ruixin Xiao,Pengfei Lv,Xiangrong Sun,Mark A. Olson,Yanling Gong

Nonalcoholic fatty liver disease (NAFLD) is demonstrated to be closely related to the disorder of gut microbiota and the intestinal mucosal barrier. Luteolin is a natural flavonoid with various activities. We aimed to investigate whether Luteolin can alleviate NAFLD and its possible mechanism involving the gut-liver axis. A rat NAFLD model was established by feeding a high-fat diet (HFD), and Luteolin was administered intragastrically. The effects of Luteolin on liver biochemical parameters, intestinal histopathology and integrity, gut microbiota , lipopolysaccharides (LPS), inflammatory cytokines, and the Toll-like receptor 4 (TLR4) signaling pathway were evaluated. We found that Luteolin restored the expression of the tight junction proteins in the intestine and ameliorated the increase permeability of the intestinal mucosa to Fluorescein isothiocyanate-dextran (FD4) caused by a high-fat diet, thus enhancing the function of the intestinal barrier. In addition, Luteolin inhibited the TLR4 signaling pathway in the liver, thereby reducing the secretion of pro-inflammatory factors and alleviating NAFLD. 16S rRNA gene sequencing revealed that Luteolin intervention significantly altered the composition of the gut microbiota in NAFLD rats and increased the richness of gut microbiota. Luteolin alleviates NAFLD in rats via restoration and repair of the damaged intestinal mucosal barrier and microbiota imbalance.

Mitigation of Allergic Asthma in Mice: A Compound Mixture Comprising Luteolin, Arbutin, and Marmesin from Gerbera Piloselloides Herba by Suppression of PI3K/Akt Pathway

Chunhua Liu,Yu He,Kun Zhou,Hong Wang,Meng Zhou,Jia Sun,Yuan Lu,Yong Huang,Yonglin Wang,Ting Liu,Yongjun Li

Background Gerberae Piloselloidis Herba (GPH) exhibits notable efficacy in alleviating allergic asthma. Previous studies in our research have identified a mixture of luteolin, arbutin, and marmesin as effective components of GPH in treating allergic asthma. However, the underlying mechanism remains unclear. This study aims to elucidate the molecular mechanism of these active components. Method Using an ovalbumin (OVA)-induced allergic asthma mouse model, various treatment groups were administered, including GPH, the active component mixture (termed "Mixture") containing luteolin, arbutin, and marmesin, and a positive drug (dexamethasone, DEX). Relevant indices were assessed, including behavioral characteristics, inflammatory cell counts, cytokine levels, histopathological examination of lung tissue, apoptosis, and expression of key proteins such as Caspase-3, Bax, Bcl-2, PI3K, p-PI3K, Akt, and p-Akt. The effect of the Mixture on the PI3K/Akt signaling pathway was further verified using the PI3K inhibitor LY294002. Results The Mixture significantly alleviated asthma symptoms, decreased IgE levels, cytokine levels (IL-4, IL-5, IL-13 and TNF-α), and the number of inflammatory cells in serum or bronchoalveolar lavage fluid (BALF), leading to the alleviation of lung pathological lesions. Additionally, the Mixture reduced the expression of Bax and Caspase-3 while increasing Bcl-2 expression, resulting in mitigated apoptosis in lung tissue. Furthermore, there appeared a decrease in the levels of PI3K and p-PI3K, as well as the ratio of p-Akt to Akt in the Mixture group, indicating the suppression of PI3K and Akt phosphorylation. Interestingly, the effects of the Mixture were comparable to those of GPH, LY294002, or the combination of LY294002 with the Mixture. Conclusion The study confirms that the Mixture containing luteolin, arbutin, and marmesin indeed alleviates allergic asthma induced by OVA in mice by suppressing the PI3K/Akt signaling pathway. These findings highlight the potential of the GPH-derived Mixture as a novel therapeutic for the treatment of allergic asthma.
