Fluorescent hydrogel kit based on photonic crystal–enhanced DNA-templated silver nanoclusters for miRNA detection

Hu Shengjie,Ma Yuchen,Fan Boyue,Miao Wenjun,Huang Rongrong

MicroRNAs (miRNAs) are post-transcriptional gene regulators and promising tumor biomarkers. Traditional detection methods are limited by high cost and specialized equipment. DNA-templated silver nanoclusters (DNA-AgNCs) are attractive fluorescent materials for biosensing but suffer from suboptimal brightness, with existing fluorescence enhancement strategies lacking universality. This study introduces a novel hydrogel kit integrating photonic crystals (PCs) and DNA-AgNCs for portable detection of miRNA-21. The formation of AgNCs within hybridization chain reaction (HCR) monomers generates a fluorescence signal, which is further amplified by PCs. Upon addition of miRNA-21, the conformational changes induced by miRNA-21 in the monomers trigger a cascade of hybridization events. Meanwhile, the formation of silver nanoclusters (AgNCs) is inhibited, thereby leading to an obvious decrease in fluorescence signal that can be distinguished by the naked eye. Hence, through the synergy of HCR-mediated target recognition, fluorogenic AgNCs-based transduction units, and PCs-triggered signal amplification, the hydrogel kit enhances the convenience and specificity of miRNA detection. In conclusion, this kit enables high-throughput fluorescence detection with a range of 1.56 to 200 nM. Moreover, it exhibits robust anti-interference capability in serum analysis, with recovery rates ranging from 91.84 to 102.74%. Owing to its strong matrix tolerance, the kit facilitates facile miRNA-21 detection, overcomes the drawbacks of conventional approaches, and holds considerable promise for clinical tumor diagnostic applications.

Molybdenum-catalyzed ATRP polymerization electrochemical biosensor for precise detection of esophageal cancer-associated MiRNA-320b

Shijin Zhou,Nan Ma,Baolin Huang,Qian Yang,Jinming Kong,Xueji Zhang

An ultrasensitive electrochemical biosensor has been developed for the detection of miRNA-320b, an exosomal RNA biomarker associated with lymph node metastasis in esophageal squamous cell carcinoma (ESCC). The platform employs a molybdenum-catalyzed atom transfer radical polymerization (Mo-ATRP) signal amplification strategy, eliminating the need for enzymatic or thermal amplification and enabling cost-effective, early-stage cancer diagnostics. A thiolated peptide nucleic acid (PNA) probe is immobilized on a gold electrode to specifically capture miRNA-320b. Upon hybridization, a sodium molybdate (Na 2 MoO 4 )-catalyzed ATRP reaction, initiated by tetrahydrocurcumin (THC), drives the in situ polymerization of 4-methacryloyloxy-2,2,6,6-tetramethylpiperidine-1-oxyl (TEMPO) monomers, forming redox-active polymer brushes directly on the electrode surface. The electrochemical response is exclusively derived from the surface-confined poly(TEMPO) layer, which forms only upon successful target recognition, ensuring high signal-to-noise performance. The biosensor achieves a detection limit of 0.417 fM with a linear range spanning 1 × 10 −15 to 1 × 10 −10 M ( R 2 = 0.992) and demonstrates excellent selectivity and robustness in complex biological matrices. Its resistance to common interferents and consistent performance under real-sample conditions underscore its suitability for clinical translation. This Mo-ATRP-based highly sensitive approach provides a promising foundation for non-invasive liquid biopsy and early detection of aggressive cancers such as ESCC.

A framework nucleic acid -based cascade-amplified self-sustaining electrochemical ratiometric sensor for miRNA detection

Hexiang Li,Yao Yao,Ziyi Gao,Yinghui Feng,Qi Liu,Xiaoqing Chen

Electrochemical sensors are often limited in stability and reliability due to influences such as probe modification density and electrode surface degradation. This study constructed an electrochemical ratiometric biosensor based on framework nucleic acid (FNA), cascade catalytic hairpin assembly (CHA), and hybridization chain reaction (HCR), achieving efficient and accurate detection of miRNA-21. Compared to traditional one-dimensional DNA electrochemical ratiometric sensors, the strong rigidity of FNA prevents probe entanglement and enhances the stability of reference probes. Notably, the introduction of a bulge loop structure provides the framework with better functional versatility. Furthermore, the application of cascade amplification endows the ratiometric sensor with enhanced adaptability for detecting low-abundance targets. By employing this sensing approach, miRNA-21 can be quantified across an extensive linear concentration range (100 aM to 100 pM), with a detection limit of 45 aM. Compared to conventional ratiometric sensors, the proposed self-sustaining FNA-based electrochemical ratiometric sensor demonstrates superior stability, accuracy, and sensitivity, showing great potential for applications in bioanalysis and medical diagnostics.

Triple-targeting miRNA-loaded core-shell nanoparticles in injectable hydrogel enable coordinated diabetic wound repair

Ziyan Guo,Lele Wu,Yu Niu,Yan Dong,Guangsheng Wu,Pengyu Zhao,Yichen Li,Jiayi Li,Yimin Zhao,Liang Kong,Zhongshan Wang

Diabetic wound healing is critically impaired by dysregulated macrophage polarization, compromised endothelial angiogenic function, and diminished fibroblast proliferation/migration under persistent hyperglycemia. Current therapies, predominantly focused on single-cell targeting, lack coordinated modulation across these key cellular components. We developed a novel triple-targeting core-shell nanoparticle (miR-RPC) leveraging the shared integrin αvβ3 receptor on macrophages, endothelial cells, and fibroblasts to address this limitation. miR-RPC features an RGD/phosphatidylserine (PS)-modified lipid shell encapsulating a chitosan/miR-146a-5p core. This miRNA was selected as a model RNA because of its widely recognized beneficial role in three key cell types in wound healing. The RGD peptide enables specific αvβ3-mediated triple-targeting. The anionic lipid PS facilitates core-shell assembly via electrostatic interaction with the cationic chitosan/RNA core and mimics apoptotic signals to enhance macrophage phagocytosis and phenotypic transition. miR-RPC effectively reprogrammed macrophages towards the M2 phenotype, restored endothelial angiogenic capacity under high glucose, and stimulated fibroblast proliferation, migration, and collagen secretion. Incorporated into a gelatin methacrylate (GelMA)/oxidized hyaluronic acid (OHA) double cross-linked hydrogel (GelO), miR-RPC@GelO significantly accelerated diabetic wound healing in rat models, demonstrating reduced inflammation, increased vascular density, and enhanced collagen deposition. This innovative triple-targeting system achieves coordinated diabetic wound repair through synergistic “immunomodulation-angiogenesis-collagen deposition” mechanisms, offering a promising therapeutic approach. Furthermore, the successful preparation of miR-RPC expands the application of anionic lipids in RNA delivery systems and highlights its potential as a versatile gene delivery vector.

A multifunctional biomimetic interface biosensor for one-step amplified detection of tumor-derived exosomal miRNA

Lu Tian,Baiqiang Chen,Jingxue Huang,Yu Cao,Tingting Wu

Tumor-derived exosomal miRNAs serve as crucial biomarkers for clinical diagnosis and disease treatment monitoring. However, the need to lyse exosomes and extract miRNAs renders most methods expensive, laborious, and time-consuming. The simple, rapid, and sensitive analysis of exosomal miRNA still poses a significant challenge. Herein, we present a multifunctional biomimetic interface biosensor (MBIB) for exosome capture, miRNA release and amplified detection in one step. Catalytical hairpin assembly (CHA) probes functionalized MCF-7 cell membrane was firstly coated onto the glass substrate to fabricate the MBIB. Due to the homologous recognition ability, the MBIB could specifically capture MCF-7 cell-derived exosomes from samples and then release miRNA through membrane fusion. Finally, the released miRNA could be detected by the CHA probes on the cell membrane. Owing to the fluidity of the cell membrane, the CHA probes could collide effectively, resulting in an enhanced signal. The developed biosensor exhibited high sensitivity, reproducibility and stability. It has been successfully applied to the real samples and could accurately distinguish the tumor-bearing mice and healthy mice, showing great potential in clinical application.

Yam-Derived Vesicles Alleviate Osteoarthritis via miRNA-Mediated Suppression of the TLR4–TRAF6 Signaling Axis

Junyan Liu,He Ke,Lingyu Su,Zhirou Lin,Chong Liu,Shilong Jiang,Hong Huang,Chengxian Guo,Qinghua Zhang,Huifang Tang,Zhaoqian Liu,Wenhu Zhou,Qi Pei,Qianfei Cui

Osteoarthritis (OA) is a degenerative joint disease, for which effective disease-modifying therapies remain lacking. Here, we report yam-derived nanovesicles (YDNVs) as a plant-derived exosome-like nanoplatform with intrinsic therapeutic activity for OA treatment. YDNVs exhibited typical nanoscale vesicular characteristics and were efficiently internalized by chondrocytes via clathrin-mediated endocytosis. Through enzymatic pretreatment and multi-omics analyses, RNA-particularly microRNAs (miRNAs)-was identified as the primary bioactive component mediating their anti-inflammatory effects. Mechanistic studies further confirmed that aof-miR168a directly binds to the 3′ untranslated regions of TLR4 and TRAF6, thereby suppressing downstream inflammatory signaling. To enhance targeting and therapeutic efficacy, a cartilage-targeting peptide-modified system (YDNVs-CAP) was engineered via membrane insertion of a cholesterol-anchored conjugate. This modification significantly improved cellular uptake, cartilage targeting, and intra-articular retention. Functionally, YDNVs-CAP effectively alleviated inflammation, oxidative stress, and extracellular matrix degradation in vitro, and markedly attenuated OA progression in a destabilization of the medial meniscus (DMM) mouse model. Collectively, this study establishes an RNA-mediated therapeutic mechanism for plant-derived nanovesicles and highlights YDNVs-CAP as a promising biomimetic nanoplatform for disease-modifying OA therapy.

Shimeles Addisu Kitte,Fuad Abduro Bushira,Chen Xu,Yong Wang,Haijuan Li,Yongdong Jin

The development of biosensors for biologically important substances with ultralow content such as microRNA is of great significance. Herein, a novel surface plasmon-enhanced electrogenerated chemiluminescence-based aptasensor was developed for ultrasensitive sensing of microRNA by using nitrogen vacancy-rich carbon nitride nanosheets as effective luminophores and gold nanoparticles as plasmonic sources. The introduction of nitrogen vacancies improved the electrochemiluminescence behavior due to improved conductance and electrogenerated chemiluminescence activity. The introduction of plasmonic gold nanoparticles increased the electrochemiluminescence signal intensity by more than eightfold. The developed surface plasmon-enhanced electrogenerated chemiluminescence aptasensor exhibited good selectivity, ultrasensitivity, excellent stability, and reproducibility for the determination of microRNA-133a, with a dynamic linear range of 1 aM to 100 pM and a limit of detection about 0.87 aM. Moreover, the surface plasmon-enhanced electrogenerated chemiluminescence sensor obtained a good recovery when detecting the content of microRNA in actual serum.

An APE1-mediated isothermal target cycling amplification for label-free and rapid detection of miRNA

Mengxu Sun,Yanxin Zhang,Juan Xie,Ya Zhang,Ting Huang,Minmin Li,Jin-Xiang Chen,Zong Dai,Jun Chen

Developing simple, rapid and sensitive strategies for miRNA analysis is extremely important for diseases early diagnosis. Herein, an APE1-mediated isothermal target cycling amplification system combined with magnetic separation was established for label-free and rapid detection of miRNA. As a proof-of-concept, miR-1246 was selected as research model. In the detection system, when the miR-1246 is present, it hybridizes with AP probes of AP-MBs to form a double-stranded, in which the APE1 recognizes specific AP site of double-stranded and cleaves the AP probes, releasing a sequence containing a G-quadruplex (G4). After the cleavage, target miR-1246 can be released to hybridize with another AP of AP-MBs, triggering a continues cleavage reaction. After magnetic separation, label-free analysis of miRNA was achieved by measuring the fluorescence of G4/ThT. Due to the high efficiency and specificity of APE1 toward AP sites in dsDNAs, this method exhibits high specificity and sensitivity with the capability of distinguishing miRNAs with single-base mismatch. The system could detect miRNA with high sensitivity within 2 h, and the limit of detection (LOD) was calculated as 25.6 fM. Furthermore, high accuracy has been achieved through recovery experiments and successful attempts has been made in applying the approach to detect miR-1246 in serum samples from breast cancer patients and normal people. This method is expected to be an effective tool for miRNA-related research and clinical early diagnosis.

Entropy-driven catalysis-based lateral flow assay for sensitive detection of Alzheimer 's-associated MicroRNA

Jing Wang,Lu Shi,Xinyu Zhu,Qiaorong Tang,Mengmeng Wu,Baoxin Li,Wei Liu,Yan Jin

Alzheimer's disease (AD) is a degenerative disease of the brain worldwide. Currently, there is no effective cure. But accurate and early diagnosis of AD is critical to the development of patient care and future treatments. MiRNA-16 has been considered as an effective diagnostic biomarker for AD because of its regulatory effect on key proteins of AD. Herein, a colorimetric lateral flow assay (LFA) was developed for sensitive detection of miRNA-16 based on entropy-driven catalysis (EDC) amplification strategy. MiRNA-16 triggered EDC and released more linker DNAs (LDNA) of sandwich structure. Thus, AuNPs were enriched at the T-line to enhance the colorimetric signal and improve the sensitivity of visual assay. It showed good specificity and sensitivity for detecting miRNA-16 with a detection limit of 1.01 pM. The practical detection of miRNA-16 in human serum obtained satisfactory result. Significantly, EDC achieved signal amplification in homogeneous solution without enzyme and DNA labeling, leading to a cheap and easy detection of miRNA-16. Therefore, it provided a portable and rapid assay for AD-related nucleic acid , which holds a potential for point-of-care testing (POCT) of AD.

CSDR Coupling with Exo III for Ultrasensitive Electrochemistry Determination of miR-145

Moli Zhang,Yang Yang,Lingyi Xin,Hua Zhang,Lun Wu,Jun Zhu,Jing Zhu,Shiyun Liu,Zhaohui Wang,Qinhua Chen,Guangyi Yang

Recently, miRNAs have become a promising biomarker for disease diagnostics. miRNA-145 is closely related to strokes. The accuracy determination of miRNA-145 (miR-145) in stroke patients still remains challenging due to its heterogeneity and low abundance, as well as the complexity of the blood matrix. In this work, we developed a novel electrochemical miRNA-145 biosensor via subtly coupling the cascade strand displacement reaction (CSDR), exonuclease III (Exo III), and magnetic nanoparticles (MNPs). The developed electrochemical biosensor can quantitatively detect miRNA-145 ranging from 1 × 102to 1 × 106aM with a detection limit as low down as 100 aM. This biosensor also exhibits excellent specificity to distinguish similar miRNA sequences even with single-base differences. It has been successfully applied to distinguish healthy people from stroke patients. The results of this biosensor are consistent with the results of the reverse transcription quantitative polymerase chain reaction (RT-qPCR). The proposed electrochemical biosensor has great potential applications for biomedical research on and clinical diagnosis of strokes.Keywords:electrochemistry;miRNA-145;stroke;CSDR;Exo III;MNPs

Intelligent Dual-Lock Deoxyribonucleic Acid Automatons Boosting Precise Tumor Imaging

Liu Wang,Kang Wang,Xiaohui Wang,Ruyan Niu,Xiaohui Chen,Ying Zhu,Zixin Sun,Jichun Yang,Guoxiang Liu,Yang Luo

An early and accurate cancer diagnosis holds the potential to improve treatment and prognosis. Nevertheless, the complexity of the biological system limits the selectivity of existing approaches and makes tumor imaging in vivo particularly challenging. In this study, tumor-specific fluorescence imaging was achieved by building intelligent dual-lock deoxyribonucleic acid automatons (IDEAs) that employed a DNA walking system standing on ZrMOF@MnO2 multifunctional nanocomposites for controllable molecular recognition. The IDEAs exhibited significantly enhanced fluorescence signals only in the coexistence of both miRNA and GSH of tumor cells, enabling accurate distinguishing of tumor cells from healthy ones. Furthermore, the feasibility and specificity of IDEAs were also validated in vivo with tumor bearing mice successfully. This work highlights the potential of the proposed IDEA strategy for tumor-specific imaging, paving the way for successful precision diagnosis and treatment.

Isothermal and protein-free cascade catalytic hairpin assembly induced-DNAzyme sensing strategy for sensitive miRNA analysis

Zhang Yuepeng,Sun Yuhua,Gong Xueshan,Zhao Shuang,Cao Weiwei,Wang Hongbo,Wang Hao,Zhang Xi,Du Changwei,Chen Zhiguo,Deng Lu

Sequence-specific microRNAs (miRNAs) represent promising biomarkers for tumor diagnosis, while current detection methods are often limited by stringent temperature requirements and excessive protease dependency. Herein, we present an isothermal, protein-free cascaded amplification circuit for the sensitive analysis of miRNA-21. This strategy, termed CHA-DNAzyme, integrates a miRNA-activated catalytic hairpin assembly (CHA) reaction with a deoxyribozymes (DNAzyme) component. Specifically, the target miRNA-21 initiates the CHA process via a toehold-mediated strand displacement reaction. This reaction unveils a previously sequestered DNAzyme sequence embedded within a hairpin structure, forming an active double-stranded complex. Subsequently, in the presence of Mg²⁺ cofactors, the catalytically activated DNAzyme drives the continuous cleavage of fluorescent reporter hairpins, releasing a measurable fluorescent signal. Implemented in a one-pot format, the CHA-DNAzyme strategy demonstrates excellent sensitivity (with a limit of detection of 8.70 pM), high specificity (the capability to discern single-base mismatches), short sample-to-answer time (approximately 60 min), and robust performance in analyzing serum samples. This approach holds significant potential to facilitate the advancement of miRNA detection tools and support their application in tumor diagnosis and prognosis assessment.
