Hydrogen sulfide (H 2 S) is a vital gaseous signaling molecule that plays a central role in various physiological and pathological processes. Given the complementary advantages of fluorescence (FL) and photoacoustic (PA) imaging, there is a growing demand for dual-ratiometric probes that enable precise in vivo monitoring of H 2 S levels. In this study, the use of 2-mercapto-1,3,4-thiadiazole (MTD) as a novel recognition group of H 2 S is presented for the first time, following conjugation with cyanine dyes to obtain a new PA probe Cy-MTD. To achieve dual-ratiometric imaging, Cy-MTD is incorporated into down-conversion nanoparticle (DCNP), resulting in the creation of a pioneering NIR-II FL/NIR-I PA dual-ratiometric nanoprobe DCNP@Cy-MTD. Cy-MTD undergoes the blueshift in absorption from 840 to 670 nm after reaction with H 2 S, enabling NIR-I ratiometric PA imaging of H 2 S by measuring the ratio of PA signal at 670 and 840 nm (PA 670 /PA 840 ). In addition, due to the strong absorption of Cy-MTD ≈840 nm and the overlapping between the absorption spectrum of Cy-MTD and 808 nm excitation band of DCNP, the 808 nm-excited FL emission (F 1550 nm,808Ex ) of DCNP in DCNP@Cy-MTD nanoprobe is quenched through the competitive absorption, while it is restored upon the interaction with H 2 S because of the blueshift in absorption of Cy-MTD. Using the stable FL emission of DCNP under 980 nm excitation (F 1550 nm,980Ex ) as the reference signal, NIR-II ratiometric FL imaging (F 1550 nm,808Ex /F 1550 nm,980Ex ) of H 2 S is achieved. The dual-ratiometric response features of the DCNP@Cy-MTD nanoprobe offer a significant advancement over traditional single-signal or single-modality imaging techniques. By providing enhanced accuracy and reliability, this probe allows for the diagnosis of hepatitis by monitoring the H 2 S, surpassing the capabilities of conventional histopathological methods, which provides a new way for more effective diagnostic strategies for liver diseases.

Near-infrared (NIR) light-triggered and separable segmented microneedles (MNs), consisting of lauric acid and polycaprolactone (LA/PCL) arrowheads and poly(vinyl alcohol) and polycaprolactone (PVA/PVP) supporting bases, have been fabricated. A hypoglycemic drug (metformin) and photothermal conversion factor (Cu7S4 nanoparticles) are encapsulated into LA/PCL arrowheads. Due to the dissolution of soluble supporting bases after the absorption of tissue fluid, the separable MNs arrowheads can be embedded into skin after insertion. Under the NIR-light irradiation, the LA/PCL arrowheads exhibit an excellent thermal-ablation change with a low amount of Cu7S4 nanoparticles (0.1 wt %) due to the low melting point of LA and PCL, thus enabling the release behavior of the encapsulated model drug to be photothermally triggered. Compared to the hypodermic injection of metformin, the thermal ablation of separable MNs triggered by NIR irradiation in the current research exhibit an excellent hypoglycemic effect in vivo. It suggests that the NIR-induced thermal-ablation MNs comprise a prospective transdermal drug-delivery system for the precise control of the timing and dosage of a drug that is dependent on NIR administration.

Bismuth selenide (Bi2Se3) attracts a lot of attention nowadays due to its unique electronic and thermoelectric properties. In this study, fabrication of Bi2Se3 nanosheets by selenite-reducing bacterium (SeRB) was first reported. Morphology, size, and location of the biogenic Bi2Se3 are bacteria-dependent. It is difficult to separate Bi2Se3 generated by Bacillus cereus CC-1 (Bi2Se3-C) from the biomass because of strong interaction with the cell membrane. However, Bi2Se3 produced by Lysinibacillus sp. ZYM-1 (Bi2Se3-Z), is highly dispersed in extracellular space with high stability. Further characterization by X-ray diffraction (XRD), scanning electron microscopy (SEM), transmission electron microscopy (TEM), and atomic force microscopy (AFM) on Bi2Se3-Z indicates that the product is a rhombohedral-phase, ultrathin nanosheet-like structure with an average size of ∼100 nm. Subsequently, the photothermal performance of Bi2Se3-Z with the irradiation of 808 nm near-infrared (NIR) laser was determined. When the Bi2Se3-Z concentration was 26 mg L–1, and irradiation power was 2 W, the photothermal conversion efficiency was calculated as 30.7%. At the same condition, 100% of the MCF7 and A549 cancer cells were killed within 10 min of irradiation in vitro. Moreover, using 1% (v/v) PVP as surfactant, a novel nanodumbbell structure of Bi2Se3 was obtained. Overall, this bacteria-driven Bi2Se3 fabrication paves a new way for biocompatible photothermal nanomaterials.

Presently, there is a need to expand the near-infrared (NIR) emission range, especially in the bio-transparent window, to improve the internal quantum efficiency and external quantum efficiency, and to minimize the shortcomings of luminescence properties. In this work, an ultra-broadband NIR emission Cr3+-doped Cs2NaLuCl6 all-inorganic metal halide perovskite phosphor was synthesized by a grinding–sintering method. Under 300 nm excitation, Cs2NaLuCl6:Cr3+ phosphors exhibited ultra-broadband NIR emission of 800–1400 nm, which covers nearly the entire NIR-I (700–1000 nm) and NIR-II (1000–1400 nm) regions; the emission peaked at around 960 nm, with a full width at half maximum of ∼182 nm. Noticeably, the internal- and external-quantum efficiencies of the optimized Cs2NaLuCl6:Cr3+ phosphor were as high as ∼92.9% and ∼60.75%, respectively. We showed through analysis and simulation that the weak crystal field strength is conducive to increasing the odd term of the crystal field, and therefore it partially released the restriction imposed by the parity prohibition law and improved the energy absorption efficiency of Cr3+. As the size of the phosphor approaches the wavelength of NIR luminescence, the apparent Mie scattering effect starts to take effect, which enhances the light extraction of NIR luminescence in the radial direction.

Metal nanoparticles have been found to exhibit a broad spectrum of antibacterial activities, yet not all metals possess the captivating plasmonic properties necessary for effective interaction with incident light. In this study, we demonstrate the feasibility of synergistically enhancing the Cu-induced antibacterial activity through the incorporation of plasmonic photothermal therapy by hybridlike anchoring the Cu component on a gold (Au) nanoplate that can effectively absorb near-infrared light. To fabricate the Janus structure, plate-like Au seeds were mixed with copper(ii) nitrate, cetyltrimethylammonium bromide, ascorbic acid, and hexamethylenetetramine, resulting in the formation of Au–Cu Janus nanostructures (JNSs). Notably, the plasmonic absorption of Au was preserved without being hindered by the Cu coating. Photothermal measurements revealed that the temperature rise of Au–Cu JNSs aqueous suspensions could reach up to 28.3 °C in just 10 min when exposed to 1064 nm laser light at a power density of 1 W·cm–2, with a photothermal conversion efficiency of 42.14%. Additionally, they show broad-spectrum antibacterial properties, making them potential contenders for addressing a wide range of bacterial infections. Our findings demonstrate that the antibacterial performance of metal nanoparticles composed of less plasmon-active components but with exceptional antibacterial properties can be significantly enhanced by coupling them with a plasmon-active nano-antenna through a Janus architecture. The Au–Cu JNSs possess potential application prospects in photothermal therapy, broad-spectrum antibacterial treatments, biomedical imaging, targeted drug delivery, sensitive diagnostics, and the development of advanced materials.

Broadband near-infrared (NIR) phosphor-converted light-emitting diodes (pc-LEDs) have become important as new NIR light sources in NIR spectroscopy and NIR imaging techniques . However, currently developed broadband NIR phosphors commonly suffer from weak emission and poor thermal stability. In this work, a series of Gd 3 Lu 2 Ga 3 O 12 : Cr 3+ phosphors were synthesized via high-temperature solid-state reaction. Under 460 nm excitation, the phosphors exhibit an intense emission spanning from 650 to 1150 nm with a full width at half maximum (FWHM) of 122 nm. After introducing Yb 3+ as a co-dopant, the emission of the phosphor has been broadened greatly. Interestingly, the studied phosphors demonstrate super high thermal stability in a wide temperature range of 35–560 K, with the integrated emission intensity fluctuating within 10 %. At 435 K, the emission intensity of Gd 3 Lu 2 Ga 3 O 12 : 0.03Cr 3+ remains 99.5 % and Gd 3 Lu 2 Ga 3 O 12 : 0.03Cr 3+ , 0.01 Yb 3+ keeps 102.7 % of the initial intensity at room temperature. These NIR phosphors have been encapsulated with a common commercial 460 nm blue LED chip to fabricate a pc-LED device. The NIR output power of the device reaches 23.93 mW@100 mA, with a photoelectric conversion efficiency of 9.81 % at 20 mA current. The applications of the prepared NIR light source in night vision, and bioimaging have been demonstrated. The results show that these Cr 3+ /Yb 3+ doped garnet phosphors can be effectively used in NIR light sources .

The second near-infrared (NIR-II, 1000–1700 nm) window is advantageous for bioimaging due to its deep tissue penetration and low autofluorescence. Despite these benefits, creating highly sensitive surface-enhanced Raman scattering (SERS) nanoprobes in this range is challenging. This study introduces an ultra-sensitive SERS nanoprobe developed using plasmonic engineering. We selected the commercial Raman dye IR1061, with an absorption peak at 1061 nm, and synthesized gold nanotriangles (AuNTs) to absorb visible light and transfer energy to NIR-II phonons. The combination of IR1061 and AuNTs significantly improved the nanoprobe's sensitivity to 116 fM in the NIR-II window, due to enhanced electromagnetic fields and SERS effects. Theoretical analysis shows AuNTs have stronger electromagnetic hotspots than gold nanorods, enhancing SERS sensitivity. The nanoprobe penetrates pig tissue over 4 mm and agarose gels up to 5 cm. This work presents a novel approach to ultra-sensitive NIR-II SERS nanoprobes for bioimaging and diagnostics. It can not only overcome the bottleneck of traditional imaging technology, but also provide strong support for high-resolution imaging of deep tissues and provide more accurate information for clinical diagnosis and treatment. Ferroptosis, a unique iron-dependent mode of cell death characterized by lipid peroxide accumulation, holds significant potential for the treatment of glioblastoma (GBM). However, the effectiveness of ferroptosis is hindered by the limited intracellular ferrous ions (Fe 2+ ) and hydrogen peroxide (H 2 O 2 ). In this study, a novel near-infrared (NIR)-light-responsive nanoplatform (ApoE-UMSNs-GOx/SRF) based on upconversion nanoparticles (UCNPs) was developed. A layer of mesoporous silica and a lipid bilayer were coated on UCNPs sequentially and loaded with glucose oxidase (GOx) and sorafenib, respectively. Further attachment of the ApoE peptide endowed the nanoplatform with BBB penetration and GBM targeting capabilities. Our results revealed that ApoE-UMSNs-GOx/SRF could efficiently accumulated in the orthotopic GBM and induce amplified ferroptosis when combining with NIR irradiation. The UCNPs mediated the photoreduction of Fe 3+ to Fe 2+ by converting NIR to UV light, and excess H 2 O 2 was produced by the reaction of glucose with the loaded GOx. These processes greatly promoted the production of ROS, which together with inhibition of system Xc − by the loaded sorafenib, leading to enhanced accumulation of lipid peroxides and significantly improved the antiglioma effect both in vitro and in vivo . Our strategy has the potential to enhance the effectiveness of ferroptosis as a therapeutic approach for GBM.

Peritumoral subcutaneous injection has been highly envisioned as an efficient yet low-risk administration of photothermal agents for superficial tumor photothermal therapy. However, obstructed by complex subcutaneous tissue, the delivery of injected photothermal agents to the specific tumor remains a critical issue. Herein, the study reports a polydopamine (PDA)-encapsulated spherical core/shell nanomotor with fluorescent indocyanine green (ICG) immobilized on its PDA shell. Upon the first near-infrared (NIR-I) irradiation, this motor can generate favorable photothermal heat, and meantime, emit a robust ICG fluorescence in the second near-infrared window (NIR-II). The heat turns the motor into an active photothermal agent able to perform thermophoretic propulsion along the irradiation direction in subcutaneous tissue, while the ICG fluorescence can direct the subcutaneous propulsion of motors toward specific tumor through real-time NIR-II imaging. These functions endow the motor with the ability of moving to tumor after being injected at peritumoral site, enabling an enhanced photothermal therapy (PTT). The results demonstrated herein suggest an integrated nanorobotic tool for the superficial PTT using peritumoral administration, highlighting an NIR-II imaging-directed subcutaneous propulsion.

The integration of photoacoustic imaging (PAI) and near-infrared II (NIR-II) fluorescence imaging holds transformative potential for cancer diagnostics but faces challenges in developing high-performance dual-modal contrast agents. We report tH7@CT8, an organic nanoprobe engineered through fluorophore trimerization to address the photothermal conversion efficiency limitations of small-molecule agents. Trimerization of the [1,2,5]thiadiazolo[3,4-g]quinoxaline fluorophore derivative H7 induced aggregation, suppressing fluorescence quantum yield (0.11% vs 0.42% monomer) while amplifying PTCE to 43.1%. Encapsulation within an osteosarcoma-targeting amphiphilic matrix yielded nanoparticles (72 ± 19 nm) with 4-fold enhanced tumor uptake versus nontargeted controls. In orthotopic osteosarcoma models, tH7@CT8 enabled PAI detection and real-time NIR-II guidance with signal (tumor)-to-background ratios >12:1. This work establishes a molecular design strategy to reconcile depth-resolution trade-offs in oncology while advancing organic contrast agents toward clinical translation.

Remote modulation of specific neurons is critical for neural circuit dissection and neurological disease therapy. Traditional neuromodulation strategies applying electrodes and external stimuli may suffer from invasive injuries and insufficient spatiotemporal resolution, respectively. Herein, transcranial optical neuromodulation with precise spatiotemporal control was realized by establishing a nanophotonic neural interface, where the polyethylene glycol-modified two-dimensional Ti 3 C 2 T x (MXene) served as near-infrared (NIR) photoactive nanosheets (PANS). Polymer modification of the nanosheets contributed to a robust PANS-neuron interface, which allowed efficient modulation of non-transgenic neurons with subcellular resolution and millisecond precision. The PANS-enabled transcranial optical neuromodulation not only evoked neuronal action potentials in brain slices with a high synchronization rate of 98.33%, but also facilitated neuronal firing in vivo. With PANS as stimulus targets, the transcranial NIR light can excite neurons in desired brain regions and activate defined neural circuits, thus regulating mouse behaviors with minimal invasiveness. Such nanosheet-enabled transcranial and precise optical neuromodulation, eliminating invasive implants and genetic manipulation, may open opportunities for neurophotonics and neuroscience research. Ischemic stroke, an acute cerebral local blood circulation disorder caused by neurological deficit syndrome, has emerged as a prominent ailment that can affect the well-being and quality of life of the elderly. The difficulty in early assessment and narrow therapeutic time window can increase the risks of mortality and disability. Monitoring the temperature of brain microregions is a promising strategy for assessing the onset of an early stroke, which is positively correlated with the intricate immune response. In this study, an innovative hybrid nanothermometer combining lanthanides and quantum dots was established for contactless monitoring of cerebral lesion temperatures in mice with ischemic stroke. The output of ratio optical signals in the near-infrared-II window (1000–1700 nm) guaranteed deep penetration, high accuracy, and reduced tissue damage. An obvious temperature increase was detected in the minor middle cerebral artery occlusion model mice (4 h after reperfusion), indicating the occurrence of a minor stroke even in the absence of abnormal behaviors. Moreover, a further increase in the brain temperature of the ischemic region resulted in a more severe cerebral lesion, suggesting the potential advancement of the treatment window for early ischemic stroke. The proposed approach can enable precise assessment of ischemic stroke, with significant implications for the incidence and progression of the illness, ultimately enhancing the quality of life of affected patients.
